The aim of this work was to evaluate the effects of prolonged starvation and refeeding on antioxidant status and some metabolic-related parameters in common dentex (Dentex dentex) liver. Fish deprived of food for 5 weeks showed a significant increase in lipid peroxidation, measured as malondialdehyde (MDA) levels. The activity of the antioxidative enzymes superoxide dismutase (SOD), catalase, and glutathione peroxidase (GPX) in starved fish significantly increased (by 42%, 22%, and 52%, respectively), whereas glutathione reductase (GR) activity was significantly depressed by 53% compared to controls. No qualitative changes in the SOD isoenzymatic pattern were detected by nondenaturing PAGE analysis, but the isoforms corresponding to CuZn-SOD I and II were enhanced in starved fish. The activity of the enzymes indicative of oxidative metabolism, beta-hydroxyacyl CoA dehydrogenase (HOAD) and citrate synthase (CS), significantly increased (by 123% and 28%, respectively), and that of glucose-6-phosphate dehydrogenase (G6PDH) was inhibited by 56%. Oxidative damage under these circumstances is reversible since all biomarkers assayed returned to control values after refeeding. Our results show that prolonged starvation leads to a pro-oxidant situation and oxidative stress despite activation of antioxidant defense mechanisms, and that inhibition of G6PDH activity might be responsible for this failure in cellular antioxidant defenses.  相似文献

In eukaryotes, multienzyme complexes containing five to nine aminoacyl-tRNA synthetase activities have frequently been reported. In this study, we report the existence, in bovine liver cytoplasm, of a multienzyme complex containing at least 16 activities which can be disrupted by homogenization to give rise to smaller complexes and noncomplexed synthetases. Determination of the size and component activity of these complexes and of the molecular weights of all 20 free synthetases suggests that the smaller complexes and free activities normally identified arise from the larger complex by well-defined stages during homogenization. We also show that similar, though not identical, complexes are found in bovine liver mitochondria and give the molecular weights of 16 mitochondrial synthetases.  相似文献

Cytochromes c' have been isolated from six strains of Achromobacter xylosoxidans: NCIB 11015 (formerly Alcaligenes sp. NCIB 11015), GIFU 543, 1048, 1051, 1055 and 1764. They are dimeric proteins with more positive redox potentials than those of cytochromes c' from phototrophic bacteria at neutral pH. The electronic absorption, EPR and MCD spectra on NO-ferrous cytochromes c' at physiological pH showed that the major part of the heme-iron of nitrosylheme was penta-coordinated. The EPR spectral results indicated that the ground state of the heme-iron of ferric cytochromes c' appears to be in an admixed spin states which consists of predominant high-spin with a slight intermediate-spin character at pH 7.2. These spectra were compared with those for cytochromes c' from phototrophic bacteria and the other hemoproteins.  相似文献

The total water content, the amount of non-freezable water, and the Na+ and K+ contents in the gastrocnemius muscle of albino mice with and without a solid tumor were determined. The spin-lattice relaxation time (T1) for the water protons in the two kinds of muscle were measured at six resonance frequencies ranging from 4.5 to 60 MHz over the temperature range +37 to −65°C. Quantitatively calculated T1 values are given. The difference in T1 for the two types of muscle at temperatures above −5°C is attributed to the difference in the distribution ratio of water between hydration and free states, and bears no direct relation to the concentration of Na+.  相似文献

The rate of lipogenesis in the liver was increased by glucose injection at birth, mediated by the insulin secretion. In addition, glucagon decreased the rates of lipogenesis and non-saponifiable-lipid synthesis after birth. These rates decreased after prolonged starvation in the newborn rat. Tri-iodothyronine injection increased the rates of lipogenesis enhanced in response to glucose administration after prolonged starvation in liver and brown adipose tissue. Dexamethasone, however, increased the rates of lipogenesis enhanced in response to glucose in liver and prevented the increase in the rates of lipogenesis in brown adipose tissue.  相似文献

Single injections of rats with hydrocortisone led to the inhibition of regenerating liver cell proliferation and protooncogene++ Ha-ras mRNA synthesis within 48 hours of hormonal induction. Administration of hydrocortisone to rats daily for 10 days resulted in a persistent decrease of the liver cell capacity to proliferate in response to partial hepatectomy. This inhibiting effect was observed for at least 7 days after cessation of hormonal stimulation; the level of Ha-ras mRNA was thereby decreased. A marked inhibition of ascite hepatoma cell growth was demonstrated after injections of those cells to mice induced with hydrocortisone for 10 days. Such a persistent effect of hydrocortisone is thought to be due to the depletion of the hormone-dependent hepatotrophic factors. The effect of the glucocorticoid hormone in vivo can be supposed to involve both the direct and indirect regulation of target cell proliferation. The latter is mediated via the changes in the activity of exogenous factors which control cell growth and proliferation.  相似文献

The aim of this study was to determine whether the effects of hypoxia on aortic contractility reflect a decrease in smooth muscle activation [phosphorylation of the 20-kDa myosin regulatory light chain (LC(20))], the capacity for myofibrillar ATP hydrolysis (mATPase activity), or both. Our results indicate that, in endothelium-denuded aortic rings from rats exposed to hypoxia for 48 h (inspired O(2) concentration = 10%), contractions to phenylephrine and potassium chloride (KCl) are impaired compared with rings from normoxic rats. The proportion of phosphorylated to total LC(20) during aortic contraction induced by 10(-5) M phenylephrine was reduced after hypoxia (51.4 +/- 5.4% in normoxic control rats vs. 32.5 +/- 4.7% in hypoxic rats, P < 0.01). Aortic mATPase activity was also decreased (maximum ATPase rate = 29.6 +/- 3.4 and 20.7 +/- 3.7 nmol. min(-1). mg protein(-1) in control and hypoxic rats, respectively, P < 0.05). Neither proliferation nor dedifferentiation of aortic smooth muscle was evident in this model; immunostaining for smooth muscle expression of the proliferating cell nuclear antigen was negative and smooth muscle-specific isoforms of myosin heavy chains, h-caldesmon, and calponin were increased, not decreased, after hypoxic exposure. Decreased aortic reactivity after hypoxia is associated with both impairment of smooth muscle activation and diminished capacity of the actomyosin complex, once activated, to hydrolyze ATP. These changes cannot be attributed to smooth muscle dedifferentiation or to reduced contractile protein expression.  相似文献
