Effects of long-term mineral fertilization and manuring on the biomass of arbuscular mycorrhizal fungi (AMF) were studied in a field experiment. Mineral fertilization reduced the growth of AMF, as estimated using both measurements of hyphal length and the signature fatty acid 16:1ω5, whereas manuring alone increased the growth of AMF. The results of AMF root colonization followed the same pattern as AMF hyphal length in soil samples, but not AMF spore densities, which increased with increasing mineral and organic fertilization. AMF spore counts and concentration of 16:1ω5 in soil did not correlate positively, suggesting that a significant portion of spores found in soil samples was dead. AMF hyphal length was not correlated with whole cell fatty acid (WCFA) 18:2ω6,9 levels, a biomarker of saprotrophic fungi, indicating that visual measurements of the AMF mycelium were not distorted by erroneous involvement of hyphae of saprotrophs. Our observations indicate that the measurement of WCFAs in soil is a useful research tool for providing information in the characterization of soil microflora.  相似文献

The influence of arbuscular mycorrhizal (AM) fungi on aggregate stability of a semi-arid Indian vertisol was studied in a pot experiment in which Sorghum bicolor (L.) was grown as test plant for 10 weeks. Pasteurized soil inoculated with AM fungi was studied with pasteurized and unpasteurized soils as references. A part of the soil in each pot was placed in nylon mesh bags to separate effects of roots and hyphae. The sorghum plants were planted outside the mesh bags which permitted AM hyphae to enter while excluding roots. Aggregate stability of the soil was determined by wet-sieving and turbidimetric measurements. Development of the AM fungi was quantified as colonized root length and external hyphal length. Soil exposed to growth of roots and hyphae (outside mesh bags) showed aggregates with larger geometric mean diameter (GMD) in pasteurized soil inoculated with AM fungi than in pasteurized uninoculated soil. There was no significant difference in GMD of the inoculated, pasteurized soil and the unpasteurized soil. No significant effects of inoculation or plant growth were found in pasteurized soil exposed to hyphal growth only (inside the mesh bags). However, the unpasteurized soil had significantly higher GMD than the pasteurized soil, irrespective of plants and inoculum. Turbidimetric measurements of soil exposed to roots and hyphae (outside mesh bags) showed the highest aggregate stability for the inoculated pasteurized soil. These results demonstrate that AM fungi contribute to the stabilization of soil aggregates in a vertisol, and that the effect is significant after only one growing season. The effect was associated with both AM hyphae and the stimulation of root growth by AM fungi. The contribution from plant roots and AM hyphae to aggregate stability of different size fractions is discussed. In a wet oligotrophic meadow located in the Czech Republic, a factorial experiment with treatments consisting of fertilization, mowing, and removal of the dominant species (Molinia caerulea) was established in 1994. In 1997 Holcus lanatus, Molinia caerulea, Potentilla erecta, Prunella vulgaris, and Ranunculus auricomus were examined for arbuscular mycorrhizal (AM) hyphae, arbuscles, and vesicles three times over the season. Time had a significant effect on AM in all five species. Except for Molinia arbuscles, a modal effect occurred, with the second sampling having a greater level of AM structures than the first and the third. Fertilization had the greatest effect on AM levels by decreasing the level of Holcus hyphae and vesicles, Potentilla vesicles, Prunella hyphae, and Ranunculus hyphae and vesicles. Mowing significantly increased the number of Potentilla vesicles, and the removal of dominant species had no significant effects. Interactions between time and treatments were common. Significant effects to the arbuscle:vesicle ratio were infrequent, and those that occurred were related to changes over the season. Seasonal effects appear to have a more powerful effect on AM structures and the arbuscle:vesicle ratio than do treatment effects. In a second experiment, Ranunculus auricomus, R. acris, and R. nemorosus, sampled four times over the season, showed significant changes in AM colonization. Overall, AM structures either declined over the season or increased from April to May and then declined. There was no AM colonization response to a spring fertilization in the three species. It is postulated that the patterns observed are due to phosphorus availability and seasonal changes in soil moisture and rates of root growth and turnover.  相似文献

Experimental systems for measuring nutrient transport by arbuscular mycorrhizal (AM) fungi in soil are described. The systems generally include two soil compartments that are separated by fine nylon mesh. Both roots and root-external hyphae grow in one compartment, but only hyphae are fine enough to grow through the mesh into the other compartment. Application of tracer isotopes to the soil of this hyphal compartment can be used to measure nutrient uptake by plants via AM fungal hyphae. Use of compartmented systems is discussed with particular reference to phosphorus, which is the mineral nutrient transported in the largest quantity by AM fungi. Laboratory and field applications of the compartmentation methodology are presented with emphasis on the functioning of native AM fungal communities. Advantages and limitations of the method are considered and future important research directions are discussed in this context. We measured the influences of soil fertility and plant community composition on Glomeromycota, and tested the prediction of the functional equilibrium hypothesis that increased availability of soil resources will reduce the abundance of arbuscular mycorrhizal (AM) fungi. Communities of plants and AM fungi were measured in mixed roots and in Elymus nutans roots across an experimental fertilization gradient in an alpine meadow on the Tibetan Plateau. As predicted, fertilization reduced the abundance of Glomeromycota as well as the species richness of plants and AM fungi. The response of the glomeromycotan community was strongly linked to the plant community shift towards dominance by Elymus nutans. A reduction in the extraradical hyphae of AM fungi was associated with both the changes in soil factors and shifts in the plant community composition that were caused by fertilization. Our findings highlight the importance of soil fertility in regulating both plant and glomeromycotan communities, and emphasize that high fertilizer inputs can reduce the biodiversity of plants and AM fungi, and influence the sustainability of ecosystems.  相似文献

Mycorrhizas are ubiquitous plant–fungus mutualists in terrestrial ecosystems and play important roles in plant resource capture and nutrient cycling. Sporadic evidence suggests that anthropogenic nitrogen (N) input may impact the development and the functioning of arbuscular mycorrhizal (AM) fungi, potentially altering host plant growth and soil carbon (C) dynamics. In this study, we examined how mineral N inputs affected mycorrhizal mediation of plant N acquisition and residue decomposition in a microcosm system. Each microcosm unit was separated into HOST and TEST compartments by a replaceable mesh screen that either prevented or allowed AM fungal hyphae but not plant roots to grow into the TEST compartments. Wild oat (Avena fatua L.) was planted in the HOST compartments that had been inoculated with either a single species of AM fungus, Glomus etunicatum, or a mixture of AM fungi including G. etunicatum. Mycorrhizal contributions to plant N acquisition and residue decomposition were directly assessed by introducing a mineral 15N tracer and 13C‐rich residues of a C4 plant to the TEST compartments. Results from 15N tracer measurements showed that AM fungal hyphae directly transported N from the TEST soil to the host plant. Compared with the control with no penetration of AM fungal hyphae, AM hyphal penetration led to a 125% increase in biomass 15N of host plants and a 20% reduction in extractable inorganic N in the TEST soil. Mineral N inputs to the HOST compartments (equivalent to 5.0 g N m?2 yr?1) increased oat biomass and total root length colonized by mycorrhizal fungi by 189% and 285%, respectively, as compared with the no‐N control. Mineral N inputs to the HOST plants also reduced extractable inorganic N and particulate residue C proportion by 58% and 12%, respectively, in the corresponding TEST soils as compared to the no‐N control, by stimulating AM fungal growth and activities. The species mixture of mycorrhizal fungi was more effective in facilitating N transport and residue decomposition than the single AM species. These findings indicate that low‐level mineral N inputs may significantly enhance nutrient cycling and plant resource capture in terrestrial ecosystems via stimulation of root growth, mycorrhizal functioning, and residue decomposition. The long‐term effects of these observed alterations on soil C dynamics remain to be investigated.  相似文献

Radiocaesium enters the food chain when plants absorb it from soil, in a process that is strongly dependent on soil properties and plant and microbial species. Among the microbial species, arbuscular mycorrhizal (AM) fungi are obligate symbionts that colonize the root cortex of many plants and develop an extraradical mycelial (ERM) network that ramifies in the soil. Despite the well-known involvement of this ERM network in mineral nutrition and uptake of some heavy metals, only limited data are available on its role in radiocaesium transport in plants. We used root-organ culture to demonstrate that the ERM of the AM fungus Glomus lamellosum can take up, possibly accumulate and unambiguously translocate radiocaesium from a 137Cs-labelled synthetic root-free compartment to a root compartment and within the roots. The accumulation of 137Cs by hyphae in the root-free compartment may be explained by sequestration in the hyphae or by a bottleneck effect resulting from a limited number of hyphae crossing the partition between the two compartments. Uptake and translocation resulted from the incorporation of 137Cs into the fungal hyphae, as no 137Cs was detected in mycorrhizal roots treated with formaldehyde. The importance of the translocation process was indicated by the correlation between 137Cs measured in the roots and the total hyphal length connecting the roots with the labelled compartment. 137Cs may be translocated via a tubular vacuolar system or by cytoplasmic streaming per se.  相似文献

The rate of global deposition of Cd, Pb, and Zn has decreased over the past few decades, but heavy metals already in the soil may be mobilized by local and global changes in soil conditions and exert toxic effects on soil microorganisms. We examined in vitro effects of Cd, Pb, and Zn on critical life stages in metal-sensitive ecotypes of arbuscular mycorrhizal (AM) fungi, including spore germination, presymbiotic hyphal extension, presymbiotic sporulation, symbiotic extraradical mycelium expansion, and symbiotic sporulation. Despite long-term culturing under the same low-metal conditions, two species, Glomus etunicatum and Glomus intraradices, had different levels of sensitivity to metal stress. G. etunicatum was more sensitive to all three metals than was G. intraradices. A unique response of increased presymbiotic hyphal extension occurred in G. intraradices exposed to Cd and Pb. Presymbiotic hyphae of G. intraradices formed presymbiotic spores, whose initiation was more affected by heavy metals than was presymbiotic hyphal extension. In G. intraradices grown in compartmentalized habitats with only a portion of the extraradical mycelium exposed to metal stress, inhibitory effects of elevated metal concentrations on symbiotic mycelial expansion and symbiotic sporulation were limited to the metal-enriched compartment. Symbiotic sporulation was more sensitive to metal exposure than symbiotic mycelium expansion. Patterns exhibited by G. intraradices spore germination, presymbiotic hyphal extension, symbiotic extraradical mycelium expansion, and sporulation under elevated metal concentrations suggest that AM fungi may be able to survive in heavy metal-contaminated environments by using a metal avoidance strategy.  相似文献

Mycorrhizal fungi can contribute to soil carbon sequestration by immobilizing carbon in living fungal tissues and by producing recalcitrant compounds that remain in the soil following fungal senescence. We hypothesized that nitrogen (N) fertilization would decrease these carbon stocks, because plants should reduce investment of carbon in mycorrhizal fungi when N availability is high. We measured the abundance of two major groups of mycorrhizal fungi, arbuscular mycorrhizal (AM) and ectomycorrhizal (ECM) fungi, in the top 10 cm of soil in control and N-fertilized plots within three Alaskan boreal ecosystems that represented different recovery stages following severe fire. Pools of mycorrhizal carbon included root-associated AM and ECM structures; soil-associated AM hyphae; and glomalin, a glycoprotein produced by AM fungi. Total mycorrhizal carbon pools decreased by approximately 50 g C m−2 in the youngest site under N fertilization, and this reduction was driven mostly by glomalin. Total mycorrhizal carbon did not change significantly in the other sites. Root-associated AM structures were more abundant under N fertilization across all sites, and root-associated ECM structures increased marginally significantly. We found no significant N effects on AM hyphae. Carbon sequestered within living mycorrhizal structures (0.051–0.21 g m−2) was modest compared with that of glomalin (33–203 g m−2). We conclude that our hypothesis was only supported in relation to glomalin stocks within one of the three study sites. As N effects on glomalin were inconsistent among sites, an understanding of the mechanisms underlying this variation would improve our ability to predict ecosystem feedbacks to global change.  相似文献

The hypothesis of this study was that cumulative P fertilization decreases the contribution of arbuscular mycorrhiza (AM) to crop growth and nutrient uptake in Northern European field conditions. The modes of action of P fertilization were evaluated through effects on mycorrhization, crop dependence on AM, and AM fungal (AMF) community. Field studies were carried out within long-term experiments on soils with low and intermediate initial content of extractable P, where no P fertilization and 45 kg ha–1 a–1 P were applied for 20 years. AM effectiveness in terms of growth and nutrient uptake of flax, red clover and barley, percentage root length colonized by AMF, P response of flax, and spore densities and species composition of the AMF communities, were assessed. In the soil with low initial P supply, cumulative P fertilization decreased AM contribution to crop growth and nutrient uptake. The higher AM effectiveness in soil with no added P compensated the cumulative P fertilization (soil PH2O 2.5 v. 9.5 mg kg–1) for flax, but not completely for clover. In contrast, barley obtained no benefit from AM at harvest and only a slight benefit from cumulated P. In the soil with intermediate initial P supply, AM reduced growth of flax and barley, especially with no added P, and no response to AM was obtained on clover due to retarded mycorrhization. Cumulative P fertilization reduced yield losses of flax by AM (PH2O 18.8 v. 5.4 mg kg–1), because fertilization inhibited mycorrhization. In both soils, root colonization and spore density were decreased by cumulative P fertilization, but no changes in AMF species composition were observed.  相似文献

Biochar may alleviate plant water stress in association with arbuscular mycorrhizal (AM) fungi but research has not been conclusive. Therefore, a glasshouse experiment was conducted to understand how interactions between AM fungi and plants respond to biochar application under water-stressed conditions. A twin chamber pot system was used to determine whether a woody biochar increased root colonisation by a natural AM fungal population in a pasture soil (‘field’ chamber) and whether this was associated with increased growth of extraradical AM fungal hyphae detected by plants growing in an adjacent (‘bait’) chamber containing irradiated soil. The two chambers were separated by a mesh that excluded roots. Subterranean clover was grown with and without water stress and harvested after 35, 49 and 63 days from each chamber. When biochar was applied to the field chamber under water-stressed conditions, shoot mass increased in parallel with mycorrhizal colonisation, extraradical hyphal length and shoot phosphorus concentration. AM fungal colonisation of roots in the bait chamber indicated an increase in extraradical mycorrhizal hyphae in the field chamber. Biochar had little effect on AM fungi or plant growth under well-watered conditions. The biochar-induced increase in mycorrhizal colonisation was associated with increased growth of extraradical AM fungal hyphae in the pasture soil under water-stressed conditions.  相似文献

温室盆栽条件下宿主植物高粱(SorghumvulgarePers.)的栽培密度对丛枝菌根(Arbuscularmycorrhizae,AM)真菌Glomusmosseae(Nicol.&Gerd.)Gerdemann&Trappe生长发育的影响试验结果表明:60株/盆密度处理的根外菌丝量及孢子数均高于其它处理。在一定栽培密度下(20~60株/盆),植株根系可溶性糖浓度与根外菌丝量呈显著负相关,与菌根侵染率呈显著正相关。植株根中磷浓度与根外菌丝量、根外菌丝量与孢子数均呈显著正相关。植株根中磷浓度与菌根侵染率呈显著负相关。结果说明:适当密植虽对植株生长有一定影响,但却促进了真菌的生长,此时菌根共生体有可能由互惠共生开始向偏利共生或弱寄生转化。密植作为一种调控手段,在菌剂生产中能获得较大数量的侵染根段、菌丝及孢子等繁殖体。  相似文献

采用四室隔网培养装置,以玉米为宿主植物,通过在植物生长室设0、50、250和500 mgPkg-1 4个施磷水平,研究了植株体内的磷营养状况对AM真菌Glomus sinuosum和Glomus intraradices生长及活性的影响。研究发现在不施磷条件下,接种AM真菌G. intraradices显著促进了植物生长和磷的吸收;低磷条件(50 mgPkg-1)下,接种菌根真菌显著促进了植物对磷的吸收,但对植物生长没有明显的影响;而在高磷条件(250 mgPkg-1 和500 mgPkg-1)下,接种菌根真菌不但没有促进植物的生长和磷的吸收,反而对其有抑制作用。随着施磷水平的提高, AM真菌根内菌丝的碱性磷酸酶活性显著下降;与不施磷相比,低量(50 mgPkg-1)供磷增加了AM真菌土壤中根外菌丝的密度,高磷(250 mgPkg-1 和500 mgPkg-1)降低了土壤中根外菌丝的密度。上述结果说明:⑴ 给宿主植物施用磷肥引起的植物磷营养状况的改变,对AM真菌生长和代谢活性具有一定的调控作用;⑵ G. sinuosum和G. intraradices两种AM真菌的生长和代谢活性对施磷水平的响应程度无显著性差异;⑶ 高磷抑制AM真菌生长和代谢活性,使真菌吸磷量减少,可能是造成菌根效应降低的原因之一  相似文献

Pot experiments were set up to determine the species-specific uptake of cesium (Cs) by mycorrhizal (AM) and non-mycorrhizal (non-AM) plants. Using stable Cs and K application, side-effects of mineral fertilization (K) on AM development and uptake of Cs and the other cations Na, Ca and Mg were investigated. AM colonization by the fungus Glomus mosseae led to a significant decrease in shoot Cs content of Agrostis tenuis from the first (4 weeks) to the third harvest (8 weeks). With regard to the root system, statistically significant differences were observed from the first (4 weeks) to the second harvest (6 weeks). Supply of additional K produced a significant decrease in Cs uptake by both AM and non-AM plants over a 10-week period. In the case of AM plant shoots, K fertilization did not very effectively reduce Cs uptake by A. tenuis. Cs contents of fertilized AM roots were similar to non-AM controls. Potassium application resulted in an increase in K content and a slight reduction in Na and Mg contents of shoots and roots. Without K fertilization, the Na content of non-AM controls was significantly enhanced over AM shoots. Shoot and root Ca contents were generally higher without than with K addition. Negative side-effects of K fertilization as a countermeasure to Cs uptake were not observed in relation to AM development. The intensity of colonization by G. mosseae was not significantly depressed by K treatment. AM development in plants appeared to decrease Cs uptake, at least at moderate nutrient levels. It is possible that Cs is sequestered by AM extraradical fungal hyphae and consequently not transferred to the plant to the extent found in non-AM roots. Accepted: 6 November 2000  相似文献

通过30μm尼龙网将根盒分成根室和菌丝室,菌丝室中的低磷土壤施加75 mg P/kg土壤的植酸钙,研究了菌丝室土壤中丛枝菌根(AM)真菌Glomus intraradices和解磷细菌Bacillus megaterium C4对有机磷的矿化和吸收.结果表明,在试验条件下,植酸钙的溶解性很低,对土壤溶液有机磷的贡献不大.接种解磷细菌C4提高了土壤中磷酸酶的活性,减少了土壤中有机磷的含量.但是,由于存在解磷细菌与AM真菌对磷的竞争,解磷细菌矿化出的磷大部分被自身利用,AM真菌的生长受到抑制,解磷细菌对植物磷营养的改善没有表现出显著的贡献.  相似文献

Various doses of glucose were added weekly to pots with maize growing in a mixture of soil and Perlite inoculated with the arbuscular mycorrhizal (AM) fungusGlomus fistulosum to define the effects of an additional carbon source on plant growth, mycorrhizae and other microbial features of the cultivation substratum. Higher doses of glucose (100 and 300 mg per pot) decreased plant growth and abundance of root hairs after 6 weeks of cultivation. Lower doses of glucose (10 and 30 mg per pot) had a positive effect on some characteristics of the development of the arbuscular fungus,e.g. root colonization, abundance of arbuscules, the length of extraradical mycelium associated with root surface, length of mycelium in the substratum as well as the length of both mycelia showing dehydrogenase activity as compared with variants not supplied with glucose solution. Glucose did not affect the number of spores of AM fungus. No effect of glucose was found on substratum respiration but glucose amendment increased microbial biomass and particularly the occurrence of saprophytic fungi. In a subsequent experiment focused on nonsymbiotic phase of fungal life cycle, the mycorrhizal root segments were incubated for 6 weeks in Petri dishes on membranes covered with a soil layer and supplied weekly with four glucose concentrations from 0.3 to 10 mg. Highest total length of hyphae associated with the root surface and the length of hyphae showing dehydrogenase activity was found when the lowest dose of 0.3 mg glucose was added to the soil weekly, whereas a 10 mg dose increased the length and activity of hyphae associated with a membrane. The possible mechanisms of the effects of additional labile carbon pool on the development of mycorrhizal fungus are discussed.  相似文献

The distribution of an arbuscular mycorrhizal (AM) fungus between soil and roots, and between mycelial and storage structures, was studied by use of the fatty acid signature 16:1(omega)5. Increasing the soil phosphorus level resulted in a decrease in the level of the fatty acid 16:1(omega)5 in the soil and roots. A similar decrease was detected by microscopic measurements of root colonization and of the length of AM fungal hyphae in the soil. The fatty acid 16:1(omega)5 was estimated from two types of lipids, phospholipids and neutral lipids, which mainly represent membrane lipids and storage lipids, respectively. The numbers of spores of the AM fungus formed in the soil correlated most closely with neutral lipid fatty acid 16:1(omega)5, whereas the hyphal length in the soil correlated most closely with phospholipid fatty acid 16:1(omega)5. The fungal neutral lipid/phospholipid ratio in the extraradical mycelium was positively correlated with the level of root infection and thus decreased with increasing applications of P. The neutral lipid/phospholipid ratio indicated that at high P levels, less carbon was allocated to storage structures. At all levels of P applied, the major part of the AM fungus was found to be present outside the roots, as estimated from phospholipid fatty acid 16:1(omega)5. The ratio of extraradical biomass/intraradical biomass was not affected by the application of P, except for a decrease at the highest level of P applied.  相似文献

在温室30%强度Hoagland营养液半水培条件下,对寄主植物玉米Zea mays接种丛枝菌根(Arbuscular myeorrhizae,AM)真菌Glomus mosseae后施加不同浓度的葡萄糖、蔗糖、脯氨酸(Pro)、吲哚-3-乙酸(IAA)、赤霉素(GA3)和6-苄基腺嘌呤(6-BA)。结果表明:除低浓度GA3处理外,其他处理尤其是高浓度处理抑制了玉米植株的生长。中浓度蔗糖、脯氨酸(Pro)和吲哚-3-乙酸(IAA)处理显著提高玉米菌根侵染率,其中以中浓度蔗糖的处理最高,达92.3%。葡萄糖、赤霉素(GA3)和6-苄基腺嘌呤(6-BA)处理则降低菌根侵染率。施加不同有机养分和植物生长物质并未提高AM真菌产孢数量,相反,却减少了孢子的形成。高浓度氨基乙酸(Gly)和低浓度6-BA处理的产孢量分别为50和48个/管,高于其他处理,但低于对照(78个/管);可溶性糖处理的产孢数量最少,平均仅18个/管。本试验条件下菌根侵染率与产孢数量不具有相关性,而产孢数与根外菌丝量具相关性。认为通过外界不同的化学调控,对植物生长、菌根侵染、根外菌丝量和孢子发育产生了一定的影响,具有一定的调节作用。  相似文献

We tested the prediction that the abundance and diversity of arbuscular mycorrhizal (AM) fungi are influenced by resource availability and plant community composition by examining the joint effects of carbon dioxide (CO(2) ) enrichment, nitrogen (N) fertilization and plant diversity on AM fungi. ? We quantified AM fungal spores and extramatrical hyphae in 176 plots after 7 yr of treatment with all combinations of ambient or elevated CO(2) (368 or 560 ppm), with or without N fertilization (0 or 4 g Nm(-2) ), and one (monoculture) or 16 host plant species (polyculture) in the BioCON field experiment at Cedar Creek Ecosystem Science Reserve, Minnesota, USA. ? Extramatrical hyphal lengths were increased by CO(2) enrichment, whereas AM spore abundance decreased with N fertilization. Spore abundance, morphotype richness and extramatrical hyphal lengths were all greater in monoculture plots. A structural equation model showed AM fungal biovolume was most influenced by CO(2) enrichment, plant community composition and plant richness, whereas spore richness was most influenced by fungal biovolume, plant community composition and plant richness. ? Arbuscular mycorrhizal fungi responded to differences in host community and resource availability, suggesting that mycorrhizal functions, such as carbon sequestration and soil stability, will be affected by global change.  相似文献
