In the Auerbach's plexus of guinea-pig ileum lower concentrations of oxotremorine (Oxo-T) produced an increase, whereas higher concentrations of Oxo-T caused inhibition of evoked acetylcholine (ACh) release, measured by bioassay using dorsal muscle of leech. Dopamine inhibited the increase in evoked release of ACh induced by Oxo-T as a function of its concentration and this inhibitory effect was nullified in presence of pimozide, the dopamine receptor antagonist. The results demonstrate existence of presynaptic dopamine receptors having inhibitory influence on excitatory presynaptic muscarinic receptors on regulation of ACh release. However, no physiological role of dopamine could be observed on ACh release in this preparation.  相似文献

The motility of the stomach and jejunum in 8 fed dogs with the intact vagus nerves was registered by the balloon method. Subcutaneous injection of benzohexonium (0.125--0.5 ml of 2.5% soltuion) and atropine (0.12--0.25 ml of 0.1% solution) or metacine (0.125--0.25 ml of 0.1% solution) to 6 dogs proved to induce a transition from digestive motility to the periodic form after a transient depression of the digestive motility. The same effect followed injection of 0.5--1.0 ml of 0.1% atropine only in 2 dogs and 1.0 ml of 0.1% metacine in 1 dog. Since retention of periodic motility following food consumption was inherent for vagotomized dogs, a conclusion was drawn that the experimental dog had "pharmacologic vagotomy". It was suggested that the muscarine receptors on the Auerbach's plexus cells exceeded the nicotine receptor in number.  相似文献

Summary The nature and distribution of cell contacts have been examined in the human enamel organ in bell stage. The lateral cell surfaces of secretory ameloblasts are linked at their distal (apical) and proximal (basal) parts by junctional complexes consisting of tight junctions, large intermediate junctions (zonulae adherentes), occasional gap junctions and one or more series of desmosomes. Scattered desmosomes and large gap junctions link epithelial cells of the external enamel epithelium, stellate reticulum, stratum intermedium and internal enamel epithelium including secretory ameloblasts. Furthermore the above-mentioned layers are also linked together by desmosomes and gap junctions.With increasing maturation of the enamel organ an increase in size and number of gap junctions is observed. Some possible implications of the role of the different junctions are considered. The gap junctions probably participate in cell differentiation in the normal morphogenesis of the teeth as well as in metabolic and ionic coupling of the cells of the enamel organ. By means of tight junctions, adjacent secretory ameloblasts cooperate to form a physical barrier which might prevent the diffusion of some types of molecules or substances (e.g. secretory material distally and acid mucopolysaccharides proximally) through the interspaces between the cells. Summary An electron-microscopic investigation, based on the suggestion that differences seen in progesterone levels under differing hormonal conditions might be reflected in the ultrastructural organisation of the lutein cells of the guinea-pig was undertaken. Comparisons were made between corpora lutea taken from animals during the normal oestrous cycle, pregnancy and lactation, and after hysterectomy or hypophysectomy.The lutein cells from the oestrous cycle corpus luteum appeared to be of two types, light and dark. The former were more numerous. The main difference between them lay in the arrangement of the endoplasmic reticulum. Lutein cells from corpora lutea (with the exception of the old degenerating corpora lutea) all contained well-developed agranular endoplasmic reticulum, little granular endoplasmic reticulum, several electron-dense lipid granules, lysosomal bodies which ranged from small spherical bodies to large autophagic vesicles and mitochondria. The mitochondria were numerous, and in the corpus luteum of pregnancy, they were closely associated with the parallel arrays of granular endoplasmic reticulum.With minor exceptions, the lutein cells of the guinea-pig present a strikingly uniform picture despite their hormonal condition.The manner in which this uniformity of ultrastructure may be related to observed differences in progesterone levels in the corpus luteum of the guinea-pig is discussed.Meat and Livestock Commission (MLC) Scholar.The authors wish to thank Dr. J. Myenteric plexus-longitudinal muscle strips prepared from tolerant/dependent guinea-pigs and continuously exposed to normorphine, display a contracture upon naloxone challenge. This phenomenon represents a sign of abstinence. Removal of the opiate by extensive washing resulted in the failure of naloxone to induce the abstinence sign, while the plexus still displayed considerable, although reduced, tolerance to morphine. Reexposure of withdrawn preparations to normorphine reinduced the ability to display the abstinence sign. Highly tolerant preparations exhibited a 30 fold increase in sensitivity to serotonin and prostaglandin E1 when tested a few minutes after naloxone-precipitated withdrawal. Supersensitivity rapidly declined when normorphine was washed off the preparation, while reincubation of withdrawn tissues with the opiate resulted in reinduction of supersensitivity. The data confirms a close relationship between a state of tolerance and dependence (including display of the abstinence sign) and supersensitivity to putative neurotransmitters or neuromodulators, becoming evident following administration of naloxone.  相似文献

The ultrastructural characteristics of oesophageal striated muscle from guinea-pig and marmoset have been examined using transmission electron microscopy and compared with ultrastructural features of skeletal muscle. The findings show that, although similar to skeletal muscle, oesophageal striated muscle exhibits important differences in the arrangement of its muscle fibres and their motor innervation. It was also found impossible to classify oesophageal striated muscle according to the usual ultrastructural criteria since its motor endplates bore a greater resemblance to those of intrafusal skeletal muscle fibres. The consideration of these differences in conjunction with characteristics revealed elsewhere by other techniques confirms the unique nature of oesophageal striated muscle.  相似文献

The distal portions of rat colon from 14-, 16-, 18-, and 21-day fetuses, newborns, and adults were histochemically examined for acetylcholinesterase (AChE) activity by light and electron microscopy. The specificity of AChE activity in Auerbach's plexus was confirmed by specific and/or nonspecific cholinesterase inhibition tests. Enzyme activity was first detectable after 18 days of gestation and became stronger with age. The reaction product was demonstrated by electron microscopy in and between the plasma membranes of the nerve fibers and their terminals. Ganglion cells also showed positive activity in the plasma membrane, nuclear envelope, and rough endoplasmic reticulum. The distribution pattern of the reaction product in fetal and newborn rat colons was basically the same as in adult rat colon. Therefore, the localization of AChE activity is considered to be a good marker for identifying premature ganglion cells in Auerbach's plexus, and the degree of AChE staining is a good indication of the degree of maturation of the plexus.  相似文献
