Transgenic lines of soybean, Glycine max (L.) Merrill, expressing a synthetic cry1A gene (tic107) from Bacillus thuringiensis (Bt), were evaluated in screenhouse and conventional field trials for efficacy against lepidopteran pests. In screenhouse trials, Bt soybean and negative checks (isogenic segregants and parental lines) were evaluated against Anticarsia gemmatalis Hübner and Pseudoplusia includens (Walker) in the United States and against A. gemmatalis, Epinotia aporema (Walsingham), Rachiplusia nu (Guenée), and Spilosoma virginica (F.) in Argentina. Bt soybean exhibited virtually complete efficacy against each of these pests, whereas negative checks suffered significant damage. Bt soybean and negative checks also were evaluated in conventional trials against native populations of A. gemmatalis and P. includens in the southeastern United States. Each of these insects caused significant damage to negative checks in one or more locations, whereas Bt soybean exhibited virtually complete efficacy against these pests. In the laboratory, lyophilized leaf tissues from Bt soybean incorporated in artificial diet at a concentration representing a 25-fold dilution of fresh tissue caused complete mortality of A. gemmatalis and near complete mortality of P. includens neonates after 11 d, whereas mortality on negative checks did not exceed 10% for either insect. Average TIC107 expression approached or exceeded 50 microg/g fresh weight at V3 stage of growth and 200 microg/g by R6 stage of growth. These results demonstrate that expression of TIC107 in soybean can not only achieve highly efficacious control of several lepidopterans under field conditions but also provide a high dose for effective insect resistance management.

Cry1Ac protoxin (the active insecticidal toxin in both Bollgard and Bollgard II cotton [Gossypium hirsutum L.]), and Cry2Ab2 toxin (the second insecticidal toxin in Bollgard II cotton) were bioassayed against five of the primary lepidopteran pests of cotton by using diet incorporation. Cry1Ac was the most toxic to Heliothis virescens (F.) and Pectinophora gossypiella (Saunders), demonstrated good activity against Helicoverpa zea (Boddie), and had negligible toxicity against Spodoptera exigua (Hübner) and Spodoptera frugiperda (J. E. Smith). Cry2Ab2 was the most toxic to P. gossypiella and least toxic to S. frugiperda. Cry2Ab2 was more toxic to S. exigua and S. frugiperda than Cry1Ac. Of the three insect species most sensitive to both Bacillus thuringiensis (Bt) proteins (including H. zea), P. gossypiella was only three-fold less sensitive to Cry2Ab2 than Cry1Ac, whereas H. virescens was 40-fold less sensitive to Cry2Ab2 compared with CrylAc. Cotton plants expressing Cry1Ac only and both Cry1Ac and Cry2Ab2 proteins were characterized for toxicity against H. zea and S.frugiperda larvae in the laboratory and H. zea larvae in an environmental chamber. In no-choice assays on excised squares from plants of different ages, second instar H. zea larvae were controlled by Cry1Ac/Cry2Ab2 cotton with mortality levels of 90% and greater at 5 d compared with 30-80% mortality for Cry1Ac-only cotton, depending on plant age. Similarly, feeding on leaf discs from Cry1Ac/Cry2Ab2 cotton resulted in mortality of second instars of S.frugiperda ranging from 69 to 93%, whereas exposure to Cry1Ac-only cotton yielded 20-69% mortality, depending on plant age. When cotton blooms were infested in situ in an environmental chamber with neonate H. zea larvae previously fed on synthetic diet for 0, 24, or 48 h, 7-d flower abortion levels for Cry1Ac-only cotton were 15, 41, and 63%, respectively, whereas for Cry1Ac/Cry2Ab2 cotton, flower abortion levels were 0, 0, and 5%, respectively. Cry1Ac and Cry2Ab2 concentrations were measured within various cotton tissues of Cry1Ac-only and Cry1Ac/Cry2Ab2 plants, respectively, by using enzyme-linked immunosorbent assay. Terminal leaves significantly expressed the highest, and large leaves, calyx, and bracts expressed significantly the lowest concentrations of Cry1Ac, respectively. Ovules expressed significantly the highest, and terminal leaves, large leaves, bracts, and calyx expressed significantly (P < 0.05) the lowest concentrations of Cry2Ab2. These results help explain the observed differences between Bollgard and Bollgard II mortality against the primary lepidopteran cotton pests, and they may lead to improved scouting and resistance management practices, and to more effective control of these pests with Bt transgenic crops in the future.

Transgenic crops that produce insecticidal toxins from the bacterium Bacillus thuringiensis (Bt) grew on >62 million ha worldwide from 1996 to 2002. Despite expectations that pests would rapidly evolve resistance to such Bt crops, increases in the frequency of resistance caused by exposure to Bt crops in the field have not yet been documented. In laboratory and greenhouse tests, however, at least seven resistant laboratory strains of three pests (Plutella xylostella [L.], Pectinophora gossypiella [Saunders], and Helicoverpa armigera [Hübner]) have completed development on Bt crops. In contrast, several other laboratory strains with 70- to 10,100-fold resistance to Bt toxins in diet did not survive on Bt crops. Monitoring of field populations in regions with high adoption of Bt crops has not yet detected increases in resistance frequency. Resistance monitoring examples include Ostrinia nubilalis (Hübner) in the United States (6 yr), P. gossypiella in Arizona (5 yr), H. armigera in northern China (3 yr), and Helicoverpa zea (Boddie) in North Carolina (2 yr). Key factors delaying resistance to Bt crops are probably refuges of non-Bt host plants that enable survival of susceptible pests, low initial resistance allele frequencies, recessive inheritance of resistance to Bt crops, costs associated with resistance that reduce fitness of resistant individuals relative to susceptible individuals on non-Bt hosts ("fitness costs"), and disadvantages suffered by resistant strains on Bt hosts relative to their performance on non-Bt hosts ("incomplete resistance"). The relative importance of these factors varies among pest-Bt crop systems, and violations of key assumptions of the refuge strategy (low resistance allele frequency and recessive inheritance) may occur in some cases. The success of Bt crops exceeds expectations of many, but does not preclude resistance problems in the future.

The efficacy of neem (1500 ppm azadirachtin (AI)), Delfin WG, a biological insecticide based on selected strain of Bacillus thuringiensis Berliner (Bt) subspecies kurstaki, and Cry1Ac protein, either individually or in combination, were examined against first to fourth instar Helicoverpa armigera (Hübner) larvae. Using an oral administration method, various growth inhibitory concentrations (EC) and lethal concentrations (LC) were determined for each bioagent. Combinations of sublethal concentrations of Bt spray formulation with azadirachtin at EC50 or EC95 levels not only enhanced the toxicity, but also reduced the duration of action when used in a mixture. The LC20 and LC50 values for Cry1Ac toxin were 0.06 and 0.22 microg ml-1, respectively. Bt-azadirachtin combinations of LC50+EC20 and LC50+EC50 result in 100% mortality. The mortality also was significant in LC20+EC20 and LC20+EC50 mixtures. These studies imply that the combined action is not synergistic but complimentary, with azadirachtin particularly facilitating the action of Bt. The Bt spray-azadirachtin combination is more economical than combinations that involve isolating the toxic protein, as the Bt spray formulations can be combined in a spray mixture with neem. These combinations may be useful for controlling bollworm populations that have acquired resistance to Bt as they may not survive the effect of mixture. Azadirachtin may be useful as a means of reducing the endotoxin concentrations in a mixture, to promote increased economic savings and further reduce the probability of resistance development to either insect control agent.

The effects of Cry toxins from Bacillus thuringiensis (Berliner) (Bt) on the anthocorid Orius albidipennis Reuter were studied under laboratory conditions. Tritrophic experiments were performed, in which Orius nymphs were fed Helicoverpa armigera (Hübner) larvae reared on a diet with Cry1Ac, Cry1Ab, or Cry2Ab toxins at different concentrations (0, 1, and 10 microg/ml), when supplemented with Ephestia kuehniella Zeller eggs. In complementary experiments, the Bt Cry1Ac toxin was directly fed to Orius nymphs at a very high concentration (1 mg/ml). No effects on prey consumption, developmental time, nymph survival, fecundity, and egg hatching of O. albidipennis were found in either experiment. It can be concluded that the toxins tested do not seem to pose a risk for the anthocorid O. albidipennis, especially when it is exposed through the prey.

Efficacy of thiamethoxam (Cruiser) and imidacloprid (Gaucho) were evaluated as seed treatments for controlling European corn borer, Ostrinia nubilalis (Hübner) and Indianmeal moth, Plodia interpunctella (Hübner) larvae in stored grain. At approximately 22-26 degrees C, all fifth instar European corn borers died after two or 4 d of exposure to corn treated with 250 and 500 ppm thiamethoxam, respectively, while mortality of larvae exposed for two and 4 d on corn treated with 6.3-937.5 ppm imidacloprid did not exceed 48% at any concentration. At 29 degrees C, all nondiapausing fifth instars were killed after 3, 4, and 6-d exposure to 400, 300 and 200-ppm thiamethoxam, respectively, while survival increased at successively lower concentrations of 100, 50, 25, and 12.5 ppm. At 29 degrees C, the LC50 decreased from 85.9 to 7.2 ppm as the duration of exposure on treated corn increased from 2 to 6 d. All second and third instar Indianmeal moth larvae died after a 5 d exposure period to corn grain treated with thiamethoxam at 50 ppm or higher, but as the larvae aged, higher concentrations and longer exposure periods were required to give 100% mortality of each larval instar. Similar results were obtained when larval Indianmeal moths were exposed on corn treated with imidacloprid, or on sorghum treated with thiamethoxam. Mature wandering phase fifth instars were the most tolerant larval stage of the Indianmeal moth.

Field studies were conducted from 1999 to 2001 to evaluate the efficacy of the transgenic cotton, Gossypium hirsutum (L.), genotype, Bollgard II (Monsanto 15985), which expresses two Bacillus thuringiensis Berliner (Bt) proteins (Cry1Ac + Cry2Ab) that are active against lepidopterous pests. Bollgard II was compared with Bollgard (DP50B), which expresses only one Bt protein (Cry1Ac), and, in all tests, the conventional variety, DP50, was used as a non-Bt control. Larval populations of the bollworm, Helicoverpa zea (Boddie), and the soybean looper, Pseudoplusia includens (Walker), were significantly lower in Bollgard II than in Bollgard and conventional cotton, and the proportion of fruit damaged by H. zea was also lower. Fall armyworm, Spodoptera frugiperda (J. E. Smith), populations were lower in Bollgard II than in Bollgard, although not significantly. Field tests were supplemented with laboratory bioassays in 2001 to compare mortality of S. frugiperda, and beet armyworms, Spodoptera exigua (Hübner), feeding on these genotypes. Mortality of both species was significantly greater on Bollgard II plant material than on either Bollgard or conventional cotton. This study demonstrated that the dual-toxin Bollgard II genotype is highly effective against lepidopterous pests that are not adequately controlled by the current single-toxin Bollgard varieties. If toxin expression in future Bollgard II varieties remains consistent with that of Monsanto 15985, supplemental insecticides will be reduced, and may be eliminated for lepidopterous pests in South Carolina.

We present a field-based approach to detect and monitor insects with resistance to insecticidal toxins produced by transgenic plants. Our objective is to estimate the phenotypic frequency of resistance in a population by relating the densities of insects on genetically transformed plants to densities on nontransformed plants. We focus on European corn borer, Ostrinia nubilalis (Hübner), in sweet corn, Zea mays L., expressing Cry1Ab from Bacillus thuringiensis subsp. kurstaki Berliner to illustrate principles underlying the method. The probability of detecting one or more rare, resistant larvae depends on sample size, the density of larvae on nontransformed plants, and an assumed frequency of resistant phenotypes in a given population. Probability of detection increases with increases in sample size, background density, or the frequency of resistant individuals. Following binomial probability theory, if a frequency of 10(-4) is expected, 10(3)-10(4) samples must be collected from a B. thuringiensis (Bt) crop to have at least a 95% probability of locating one or more resistant larvae. In-field screens using transgenic crops have several advantages over traditional laboratory-based methods, including exposure to a large number of feral insects, discrimination of resistant individuals based on Bt dosages expressed in the field, incorporation of natural and Bt-induced mortality factors, simultaneous monitoring for more than one insect species, and ease of use. The approach is amenable to field survey crews working in research, extension, and within the seed corn industry. Estimates of the phenotypic frequency of resistance from the in-field screen can be useful for estimating initial frequency of resistant alleles. Bayesian statistical methods are outlined to estimate phenotype frequencies, allele frequencies, and associated confidence intervals from field data. Results of the approach are discussed relative to existing complementary methods currently available for O. nubilalis and corn earworm, Helicoverpa zea (Boddie).

Abstract 1 Larval survival and development of Dipel‐susceptible and ‐resistant strains of European corn borer, Ostrinia nubilalis (Hübner), were assayed using diets incorporating low doses of a commercial formulation of Bacillus thuringiensis var. kurstaki Berliner (Dipel ES). 2 Larval mortality, growth and development, pupation rate and pupal weight were not significantly different between Dipel‐susceptible and ‐resistant strains when larvae were reared on a nontoxic control diet. 3 Larval mortality of Dipel‐resistant larvae did not significantly change as Dipel concentration increased at the tested concentrations, whereas mortality of Dipel‐susceptible larvae increased dramatically as Dipel concentration increased. 4 Larval development was significantly delayed when larvae were fed diets containing low doses of Dipel. 5 Pupation rate and pupal body weight declined as Dipel concentration increased but it decreased faster for the susceptible strain than for the resistant strain.

Laboratory experiments were conducted to evaluate the behavior of Helicoverpa armigera (Hübner) and Spodoptera litura (Fabricius) larvae on meridic diet with different concentrations of Bt spray formulation Delfin or isolated Cry1Ac protein or the foliage and bolls from transgenic cotton, Bollgard hybrid RCH-317 Bt. Both insect species selectively fed on nontreated diet compared with the diet treated with Delfin. While H. armigera exhibited concentration response with Cry1Ac, this protein did not affect S. litura larvae. In general Helicoverpa selected diet with low concentrations (EC20 and EC50 levels) of Cry1Ac compared with higher concentrations of Cry1Ac. In order to develop appropriate management strategies, a thorough understanding of the behavioral mechanisms leading to the responses of insects to the proteins in transgenic varieties is required. Thus, based on results of the insects fed individually on the leaf discs or bolls from transgenic cotton plants alone or under choice situation with meridic diet revealed that H. armigera larvae preferred meridic diet to transgenic leaves or bolls expressing Cry1Ac protein. H. armigera larvae preferred meridic diet to plant material; more than 70% larvae were seen on the meridic diet, and average larval weight gain was in the range of 121.7–130.5 mg. However, in case of S. litura the larvae showed no significant discrimination between meridic diet and the leaf discs. In fact more than 60% larvae preferred leaf discs for feeding, though Cry1Ac expression in leaf discs was in the range of 0.9–2.18 μg/g. Thus differences in behavioral response could potentially impact the level of efficacy of crop cultivars that have been genetically engineered to produce these proteins.

Susceptibility to Cry1Ab toxin from Bacillus thuringiensis (Bt) was determined for 12 field populations of neonate corn earworm, Helicoverpa zea (Boddie), from the United States. Earworm larvae were exposed to artificial diet treated with increasing Bt concentrations, and mortality and growth inhibition were evaluated after 7 d. The range of variation in Bt susceptibility indicated by growth inhibition was very similar to that indicated by mortality. Although interpopulation variation in susceptibility to both proteins was observed, the magnitude of the differences was small (less than or equal to fivefold). These results suggest that the observed susceptibility differences reflect natural variation in Bt susceptibility among corn earworm populations rather than variation caused by prior exposure to selection pressures. Therefore, corn earworms apparently are susceptible to Bt toxins across most of their geographic range.

A 2-yr field and laboratory study investigated insect resistance of the maize, Zea mays L., inbred Tex6, which has previously demonstrated resistance to Aspergillus ear rot and aflatoxin production, relative to susceptible inbred B73. Field studies indicated significantly greater resistance to insect feeding of V4-V8 growth stage Tex6 plants compared with B73 plants in both years, primarily to flea beetles (Chaetonema spp.). Field studies of natural (1999) and artificial (2000) infestations of corn earworms, Helicoverpa zea (Boddie), indicated much lower levels of kernel damage at milk stage (approximately three-fold) and smaller surviving larvae (approximately three-fold) in Tex6 compared with B73 ears. At harvest similar trends in reduction of numbers of damaged kernels per ear, as well as incidence and numbers of kernels per ear symptomatically infected by Fusarium spp. were noted. Laboratory studies indicated little difference in mortality or survivor weight of caterpillars or sap beetle adults caged with milk stage kernels of the two inbreds. However, assays with silks indicated significantly greater mortality of H. zea in both 1999 and 2000, and European corn borer, Ostrinia nubilalis (Hübner) in 1999 (only year tested) when fed Tex6 silks compared with B73 silks. Pollinated Tex6 silks were generally darker colored and more toxic than unpollinated silks. Thus, it is possible that commercially usable inbreds with resistance to insects, which also contribute to the mycotoxin problem through vectoring and damage, could be produced using Tex6 as a source.

One field strain each of the European corn borer, Ostrinia nubilalis (Hübner); southwestern corn borer, Diatraea grandiosella Dyar; and sugarcane borer, Diatraea saccharalis (F.); were collected from cornfields in northeastern Louisiana. Susceptibilities of the field strain and a corresponding laboratory strain of the three borer species to Cry1Ab protein in DK69-70 Bacillus thuringiensis (Bt) corn hybrid were determined by exposing neonates to intact leaf tissues from whorl stage plants or by feeding neonates or third instars on a meridic diet treated with different concentrations of Cry1lAb protein extracted from Bt corn leaves. Mortality and growth of larvae were evaluated after 2 and 4 d posttreatment in the bioassays by using intact leaf tissues or after 7 d in the bioassays by using diet incorporating Cry1Ab protein. D. saccharalis was the least susceptible species to Cry1Ab protein among the three species, followed by D. grandiosella, whereas O. nubilalis was most susceptible. The 2-d mortality of D. saccharalis neonates on intact Bt leaf tissues was lower than that of O. nubilalis and D. grandiosella. All neonates of O. nubilalis were killed on the diet treated with Cry1Ab protein at 0.5 and 1 mg/kg. The mortality of D. grandiosella was > 75% at 1 mg/kg, but it was < 6% for D. saccharalis at 1 mg/kg. The LC50 values of D. saccharalis were 3- and 11-fold higher than those of D. grandiosella and O. nubilalis, respectively. The LC90 values of D. saccharalis were 8- and 32-fold higher than those of D. grandiosella and O. nubilalis, respectively. Larval growth of the three species on Cry1Ab-treated diet was inhibited, but the inhibition was greater for O. nubilalis and D. grandiosella than for D. saccharalis. The lower susceptibility of D. saccharalis to Cry1Ab protein suggests that it is necessary to verify if a high-dose Bt corn for O. nubilalis and D. grandiosella is also a high dose for D. saccharalis.

Two candidate diagnostic concentrations of the Cry1Ab and Cry1Ac toxins from Bacillus thuringiensis corresponding to the LC99 and EC99 (effective concentration that causes 99% growth inhibition) for European corn borer, Ostrinia nubilalis (Hübner), were determined based on previously obtained baseline data. Validation experiments using field-collected European corn borer populations from across North America showed that for Cry1Ab, a concentration corresponding to the upper limit of the 95% confidence interval of the LC99, produced mortality > 99% for all populations tested. However, for Cry1Ac, adjustments and further validation are probably necessary. Development of B. thuringiensis resistance monitoring programs that rely on diagnostic techniques are discussed.

In 1995, ears of a experimental inbred (CG59-2) containing a synthetic Bacillus thuringiensis Cry IA (b) gene driven by PEPC, pith and pollen promoters and artificially infested with Ostrinia nubilalis (Hübner) larvae in small plot studies were free from insect damage, whereas 40-50% of the corresponding non-Bt inbred ears were damaged. Bt inbred ears that were inoculated with Aspergillus flavus Link and Fusarium proliferatum T. Matsushima (Nirenberg) or exposed to natural mold inoculum after infestation with O. nubilalis were free of visible signs of mold, as compared with approximately 30-40% of the non-Bt ears similarly treated. Results in 1996 using the same inbred with a single allele dose of the Bt gene showed similar trends. Mean total fumonisin levels for non-Bt versus Bt inbred ears were not significantly different (2.8 versus 0.8 ppm, respectively) in 1996. In paired hybrid studies run in 0.4-ha (1-acre) fields, an event 176 Bt hybrid had significantly lower amounts of damage and signs of Fusarium spp. mold, but not fumonisin, compared with a corresponding non-Bt hybrid from 1996 to 1998. However, two hybrid pairs that contained either MON810 or Bt11 constructs examined in similar fields at the same site had lower levels of fumonisin in both 1997 (30- to 40-fold) and 1998. High intrafield variability in insect infestation and presence of Helicoverpa zea (Boddie) in Bt hybrids was apparently responsible for fewer significant differences in fumonisin levels in 1998. Similar trends for all three hybrid pairs were noted in small plot trials at another site. Incidence of other ear pests or insect predators varied as much among non-Bt hybrids as they did for Bt/non-Bt hybrid pairs.

Larvae of the Old World corn earworm, Helicoverpa armigera (Hübner), were fed diets containing lyophilized silks from maize genotypes expressing varying levels of maysin, a flavone glycoside known to be toxic to the New World corn earworm, Helicoverpa zea Boddie. Three different H. armigera colonies were tested: a wild-type colony (96-S), a colony selected for resistance to deltamethrin (Del-R), and a colony selected for resistance to the Cry1Ac protoxin of Bacillus thuringiensis (Bt-R). A colony of H. zea was also tested as a control. High-maysin silk diets significantly slowed the growth and arrested the development of larvae from all H. armigera colonies compared with low-maysin silk diets, maysin-lacking silk diets, and no-silk control diets. The effects on the H. armigera and H. zea colonies were similar across maysin levels, although H. zea is a larger insect than H. armigera and this overall size difference was observed. Among the H. armigera colonies, maysin effects were generally similar, although 7-d-old Del-R larvae were significantly smaller than 7-d-old Bt-R and 96-S larvae for one no-silk control and two maysin-containing silk treatments. The toxic effect of maysin on the Bt-R and Del-R colonies suggests that physiological mechanisms of H. armigera resistance to Cry1Ac and deltamethrin do not confer cross-resistance to maysin.
