During late stages of neural development, synaptic circuitry is edited by neural activity. At neuromuscular synapses, the transition from multiple to single innervation is modulated by the relative pattern of activity among inputs competing for innervation of the same muscle fiber. While experimental perturbations of activity result in marked changes in the timing of neuromuscular synaptic competition, little is known about the patterns of activity present during normal development. Here, we report the temporal patterning of motor unit activity in the soleus muscle of awake, behaving neonatal mice, and that patterning is modulated by gap-junctional coupling. Our work suggests that neuromuscular synaptic competition is modulated by surprisingly low levels of activity and may be triggered by the disappearance of temporally correlated activity among inputs competing for innervation of the same muscle fiber.  相似文献

Glial cell-line derived neurotrophic factor (GDNF) is a potent survival factor for motor neurons. Previous studies have shown that some motor neurons depend upon GDNF during development but this GDNF-dependent motor neuron subpopulation has not been characterized. We examined GDNF expression patterns in muscle and the impact of altered GDNF expression on the development of subtypes of motor neurons. In GDNF hemizygous mice, motor neuron innervation to muscle spindle stretch receptors (fusimotor neuron innervation) was decreased, whereas in transgenic mice that overexpress GDNF in muscle, fusimotor innervation to muscle spindles was increased. Facial motor neurons, which do not contain fusimotor neurons, were not changed in number when GDNF was over expressed by facial muscles during their development. Taken together, these data indicate that fusimotor neurons depend upon GDNF for survival during development. Since the fraction of cervical and lumbar motor neurons lost in GDNF-deficient mice at birth closely approximates the size of the fusimotor neuron pool, these data suggest that motor neuron loss in GDNF-deficient mice may be primarily of fusimotor neuron origin.  相似文献

An analysis is given of the polyneuronal innervation of embryonic skeletal muscle and its subsequent elimination during development. The amount of polyneuronal innervation that has been observed is consistent with the notion that initially each motor neuron distributes its contacts at random among the available fibres of a particular muscle. The idea that the elimination of excess innervation proceeds through interactions between terminals is placed on quantitative basis. Each motor neuron is presumed to have a finite capacity for maintaining the structure and activity of its terminals, which is shared out among them; a survival strength can be assigned to each terminal. Survival strengths undergo a process of continual adjustment. A terminal with above average strength for its endplate is strengthened at the expense of the weaker terminals, subject to the total survival strength available to each motor neuron remaining constant. It is proved that this scheme will transform the initial pattern of innervation into one in which each muscle fibre has contact from a single axon. Interpretations of the following results are given: the decrease in the spread of motor unit size during the development of innervation of the rat soleus muscle; the time course of superinnervation; the effects of neonatal partial denervation. Various suggestions are made for future experimental approaches.  相似文献

Gap junctions (GJs) between neurons are present in both the newborn and the adult nervous system, and although important roles have been suggested or demonstrated in a number of instances, in many other cases a full understanding of their physiological role is still missing. GJs are expressed in the rodent lumbar cord at birth and mediate both dye and electrical coupling between motor neurons. This expression has been proposed to mediate: (i) fast synchronization of motoneuronal spike activity, in turn linked to the process of refinement of neuromuscular connections, and (ii) slow synchronization of locomotor-like oscillatory activity. Soon after birth this coupling disappears. Since in the adult rat regeneration of motor fibers after peripheral nerve injury leads to a recapitulation of synaptic refinement at the target muscles, we tested whether GJs between motor neurons are transiently re-expressed. We found that in conditions of maximal responsiveness of lumbar motor neurons (such as no depression by anesthetics, decerebrate release of activity of subsets of motor neurons, use of temporal and spatial summation by antidromic and orthodromic stimulations, testing of large ensembles of motor neurons) no firing is observed in ventral root axons in response to antidromic spike invasion of nearby counterparts. We conclude that junctional coupling between motor neurons is not required for the refinement of neuromuscular innervation in the adult.  相似文献

The evolutionary origin of novelties is a central problem in biology. At a cellular level this requires, for example, molecularly resolving how brainstem motor neurons change their innervation target from muscle fibers (branchial motor neurons) to neural crest-derived ganglia (visceral motor neurons) or ear-derived hair cells (inner ear and lateral line efferent neurons). Transplantation of various tissues into the path of motor neuron axons could determine the ability of any motor neuron to innervate a novel target. Several tissues that receive direct, indirect, or no motor innervation were transplanted into the path of different motor neuron populations in Xenopus laevis embryos. Ears, somites, hearts, and lungs were transplanted to the orbit, replacing the eye. Jaw and eye muscle were transplanted to the trunk, replacing a somite. Applications of lipophilic dyes and immunohistochemistry to reveal motor neuron axon terminals were used. The ear, but not somite-derived muscle, heart, or liver, received motor neuron axons via the oculomotor or trochlear nerves. Somite-derived muscle tissue was innervated, likely by the hypoglossal nerve, when replacing the ear. In contrast to our previous report on ear innervation by spinal motor neurons, none of the tissues (eye or jaw muscle) was innervated when transplanted to the trunk. Taken together, these results suggest that there is some plasticity inherent to motor innervation, but not every motor neuron can become an efferent to any target that normally receives motor input. The only tissue among our samples that can be innervated by all motor neurons tested is the ear. We suggest some possible, testable molecular suggestions for this apparent uniqueness.  相似文献

We have compared the development of fast and slow motor innervation in the neonatal rabbit soleus, a muscle which contains two distinct motor unit types during the early period of polyneuronal innervation. The innervation state of individual muscle fibers was ascertained using an intracellular electrode; a fluorescent dye was then injected into particular fibers to permit subsequent identification of histochemical type. We found no significant difference in the time course of synapse elimination for fast and slow motor units as judged by the percentage of fibers remaining polyneuronally innervated at two ages: 7-8 days, when most fibers are multiply innervated, and 10-11 days, when the level of polyinnervation is low. In a second experiment, we examined a phenomenon in which compound end-plate potentials were occasionally seen in muscle fibers at an age (17-23 days) well past the major episode of synapse elimination. We present evidence that this apparent polyinnervation in fact derives from an electrode-induced electrical coupling artifact and that genuinely polyinnervated fibers are very rare at this stage, if present at all.  相似文献

As lobsters grow from early juveniles to adults their body size increases more than 20-fold, raising the question of how function is maintained during these ongoing changes in size. To address this question we studied the pyloric 1 (p1) muscle of the stomach of the lobster, Homarus americanus. The p1 muscle receives multiterminal innervation from one motor neuron, the lateral pyloric neuron of the stomatogastric ganglion. Staining with antibodies raised against synaptotagmin showed that as the muscle fibers increased in length, the spacing between the terminal innervation increased proportionally, so the number of synaptic contact regions/muscle fiber did not change. Muscle fibers were electrically coupled in both juveniles and adults. The amplitude of single intracellularly recorded excitatory junctional potentials evoked by motor nerve stimulation was the same in both juveniles and adults. Nonetheless, the peak depolarizations reached in response to ongoing pyloric rhythm activity or in response to high-frequency trains of stimuli similar to those produced during the pyloric rhythm were approximately twofold larger in juveniles than in adults. This suggests that homeostatic regulation of synaptic connections may operate at the level of the amplitude of the single synaptic potential rather than on the summed depolarization evoked during strong rhythmic activity.  相似文献

To define alterations of neuronal connectivity that occur during motor neuron degeneration, we characterized the function and structure of spinal circuitry in spinal muscular atrophy (SMA) model mice. SMA motor neurons show reduced proprioceptive reflexes that correlate with decreased number and function of synapses on motor neuron somata and proximal dendrites. These abnormalities occur at an early stage of disease in motor neurons innervating proximal hindlimb muscles and medial motor neurons innervating axial muscles, but only at end-stage disease in motor neurons innervating distal hindlimb muscles. Motor neuron loss follows afferent synapse loss with the same temporal and topographical pattern. Trichostatin A, which improves motor behavior and survival of SMA mice, partially restores spinal reflexes, illustrating the reversibility of these synaptic defects. Deafferentation of motor neurons is an early event in SMA and may be a primary cause of motor dysfunction that is amenable to therapeutic intervention.  相似文献

1. A circadian pacemaker is located in the eyes of a variety of marine gastropods, including Aplysia and Bulla. It produces a circadian rhythm in the frequency of spontaneously occurring optic nerve (ON) compound action potentials (CAPs). The circadian pacemaker in Bulla includes a population of 100 retinal pacemaker neurons, that produce the rhythmic CAP output. Intracellular recording from the Bulla pacemaker neurons has yielded new insight into their time-keeping ability. 2. Intracellular injection of Lucifer yellow dye into a single pacemaker neuron results in the spread of dye to several neurons. This dye coupling is presumably mediated by the gap junctions among neurons that are responsible for the synchronous firing of the population of pacemaker neurons and the generation of ON CAPs. 3. The circadian pacemaker in each eye interacts with the paired pacemaker in the contralateral eye. The interaction results in the coordinating firing of CAPs from each eye and in the coordinated phasing of the circadian rhythms of CAP activity generated in each eye. This interaction occurs by reciprocal excitatory chemical synapses. These synaptic receptors occur in the ON as well as in the retinal neuropil and CAP synchrony occurs in the ON as well as in the basal retina. 4. Pacemaker neurons are depolarized by 5-HT and membrane permeable cAMP analogues. The membrane resistance increases during the depolarization suggesting a background potassium current is decreased. 5. The tetrapeptide FMRF-HN2 hyperpolarizes the pacemaker neurons. It reverses the effect of 5-HT and cAMP, suggesting 5-HT and FMRF-NH2 may be acting on the same membrane channel, the S channel.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Acetylcholine receptors (AChRs) mediate synaptic transmission at the neuromuscular junction, and structural and functional analysis has assigned distinct functions to the fetal (alpha2beta(gamma)delta) and adult types of AChR (alpha2beta(epsilon)delta). Mice lacking the epsilon-subunit gene die prematurely, showing that the adult type is essential for maintenance of neuromuscular synapses in adult muscle. It has been suggested that the fetally and neonatally expressed AChRs are crucial for muscle differentiation and for the formation of the neuromuscular synapses. Here, we show that substitution of the fetal-type AChR with an adult-type AChR preserves myoblast fusion, muscle and end-plate differentiation, whereas it substantially alters the innervation pattern of muscle by the motor nerve. Mutant mice form functional neuromuscular synapses outside the central, narrow end-plate band region in the diaphragm, with synapses scattered over a wider muscle territory. We suggest that one function of the fetal type of AChR is to ensure an orderly innervation pattern of skeletal muscle.  相似文献

Summary InLumbricus terrestris the wall of the pharynx is built up from obliquely striated longitudinal and circular muscle layers. The occurrence of perikarya and nerve bundles showing green fluorescence suggests the presence of aminergic innervation in the pharynx. A significant number of chemical synapses were detected in the neuropil among axon terminals. The junctional gap is generally 100–300 nm wide in type I junctions which resemble the cholinergic motor endplates of vertebrate skeletal muscle. A narrow junctional gap of about 25 nm is characteristic of the close contacts in the type II neuromuscular junction. Agranular spherical vesicles, together with small and large dense-cored granules, fill in these axon terminals.  相似文献

Gap junction channels (electrical synapses) are a major component of the central nervous system mediating both electrical and metabolic coupling between neurons and glia. Their roles are as diverse as the cell types in which they are expressed and only some of these are reviewed here. In the adult the plastic nature of the gap junction channel allows for changes in the writing of the retinal circuitry that optimize visual processing to suit ambient lighting conditions. Gap junctional communication has been proposed to play a key role in embryonic development in general and in particular during the development of the retina where its roles may include control of neurogenesis, cell specification, synaptogenesis, and the synchronization of the spontaneous electrical activity required for the sharpening of central visual projections. Here we review gap junctional coupling within the retina and present data correlating gap junction expression with development events in the chick retina.  相似文献

Using extracellular and intracellular stimulation, recording and dye-filling, we identified and studied the superficial extensor motor neurons of the crayfish, Cherax destructor. Functional associations of each neuron were characterised by recording its responses to sensory and abdominal cord inputs, its extensor muscle innervation pattern and its relationships with other neurons. Two clear associations were found among the six neurons of each segment. A medium-sized excitor (no. 3), that innervates a substantial percentage of extensor muscle fibres, and the largest excitor (no. 6), recruited during peak, excitation, were inhibited by input from unknown interneurons that excited the common inhibitor (no. 5). Likewise, these excitors received excitatory input when the inhibitor was silent. Another medium-sized neuron (no. 4) that innervates many muscle fibres was co-active with one of the small excitors (no. 2). The two medium-sized neurons were never active at the same time, and these two groupings may be determined by pre-motor interneurons. The implications of these findings for our understanding of motor control in this system are discussed. Accepted: 21 June 1998  相似文献

We have previously shown that knockout of fibroblast growth factor-2 (FGF-2) and potential compensatory effects of other growth factors result in amelioration of disease symptoms in a transgenic mouse model of amyotrophic lateral sclerosis (ALS). ALS is a rapidly progressive neurological disorder leading to degeneration of cortical, brain stem, and spinal motor neurons followed by subsequent denervation and muscle wasting. Mutations in the superoxide dismutase 1 (SOD1) gene are responsible for approximately 20% of familial ALS cases and SOD1 mutant mice still are among the models best mimicking clinical and neuropathological characteristics of ALS. The aim of the present study was a thorough characterization of FGF-2 and other growth factors and signaling effectors in vivo in the SOD1G93A mouse model. We observed tissue-specific opposing gene regulation of FGF-2 and overall dysregulation of other growth factors, which in the gastrocnemius muscle was associated with reduced downstream extracellular-signal-regulated kinases (ERK) and protein kinase B (AKT) activation. To further investigate whether the effects of FGF-2 on motor neuron death are mediated by glial cells, astrocytes lacking FGF-2 were cocultured together with mutant SOD1 G93A motor neurons. FGF-2 had an impact on motor neuron maturation indicating that astrocytic FGF-2 affects motor neurons at a developmental stage. Moreover, neuronal gene expression patterns showed FGF-2- and SOD1 G93A-dependent changes in ciliary neurotrophic factor, glial-cell-line-derived neurotrophic factor, and ERK2, implying a potential involvement in ALS pathogenesis before the onset of clinical symptoms.  相似文献

Ischemic preconditioning increases the heart's tolerance to a subsequent longer ischemic period. The purpose of this study was to investigate the role of gap junction communication in simulated preconditioning in cultured neonatal rat cardiac myofibroblasts. Gap junctional intercellular communication was assessed by Lucifer yellow dye transfer. Preconditioning preserved intercellular coupling after prolonged ischemia. An initial reduction in coupling in response to the preconditioning stimulus was also observed. This may protect neighboring cells from damaging substances produced during subsequent regional ischemia in vivo, and may preserve gap junctional communication required for enhanced functional recovery during subsequent reperfusion.  相似文献

Cobalt axonal iontophoresis and intracellular recordings were used to identify a cluster of several motor neurons innervating the penis-retractor muscle of Aplysia. Intracellularly recorded motor neuron action potentials elicited direct, one-for-one, constant latency excitatory junctional potentials (ejps) in individual muscle fibers. The axons of motor neurons could be recorded extracellularly in the penis-retractor nerve and stimulation of the nerve backfired the motor neurons. Perfusion of the ganglion, the muscle, or both with solutions of either increased Mg++/decreased Ca++ or increased Ca++ sea water indicated that the presumed motor neuron impaled was not a sensory cell and that interneurons were not intercalated in the pathway. Innervation of muscle fibers was found to be functionally polyneuronal and diffuse. The ejps were found to undergo marked facilitation with repetitive motor-neuron stimulation. The motor neurons were isolated in a distinct cluster in the right pedal ganglion. Their electrical activity was characterized by spontaneous irregular action potentials and a moderate input of postsynaptic potentials.  相似文献

Each adult mammalian skeletal muscle has a unique complement of fast and slow myofibers, reflecting patterns established during development and reinforced via their innervation by fast and slow motor neurons. Existing data support a model of postnatal "matching" whereby predetermined myofiber type identity promotes pruning of inappropriate motor axons, but no molecular mechanism has yet been identified. We present evidence that fiber type–specific repulsive interactions inhibit innervation of slow myofibers by fast motor axons during both postnatal maturation of the neuromuscular junction and myofiber reinnervation after injury. The repulsive guidance ligand ephrin-A3 is expressed only on slow myofibers, whereas its candidate receptor, EphA8, localizes exclusively to fast motor endplates. Adult mice lacking ephrin-A3 have dramatically fewer slow myofibers in fast and mixed muscles, and misexpression of ephrin-A3 on fast myofibers followed by denervation/reinnervation promotes their respecification to a slow phenotype. We therefore conclude that Eph/ephrin interactions guide the fiber type specificity of neuromuscular interactions during development and adult life.  相似文献

Competition among axon terminals is usually considered to contribute to the formation of patterned synaptic connections. During axonal regeneration of motor neurons in the cockroach, leg muscles initially become innervated by appropriate and inappropriate motor neurons. All axon terminals from inappropriate neurons eventually are eliminated, resulting in the reformation of the original innervation pattern. Destruction of an identified motor neuron by the intracellular injection of pronase did not prevent the elimination of inappropriate axon terminals in the muscle normally innervated by that motor neuron. Therefore, competition does not play a role in the reinnervation of the leg muscles. This indicates a major role for specific cell-cell recognition.  相似文献
