Carotenoids are nutritionally important tetraterpenoid pigments that upon oxidative cleavage give rise to apocarotenoid (norisoprene) aroma volatiles. beta-Carotene is the predominant pigment in orange-fleshed melon (Cucumis melo L.) varieties, reaching levels of up to 50 microg/gFW. Pale green and white cultivars have much lower levels (0-10 microg/gFW). In parallel, beta-ionone, the 9,10 cleavage product of beta-carotene, is present (12-33ng/gFW) in orange-fleshed melon varieties that accumulate beta-carotene, and in much lower levels (0-5 ng/gFW) in pale green and white fleshed varieties. A search for a gene putatively responsible for the cleavage of beta-carotene into beta-ionone was carried out in annotated melon fruit EST databases yielding a sequence (CmCCD1) highly similar (84%) to other plant carotenoid cleavage dioxygenase genes. To test its function, the clone was overexpressed in Escherichia coli strains previously engineered to produce different carotenoids. We show here that the CmCCD1 gene product cleaves carotenoids at positions 9,10 and 9',10', generating geranylacetone from phytoene; pseudoionone from lycopene; beta-ionone from beta-carotene, as well as alpha-ionone and pseudoionone from delta-carotene. CmCCD1 gene expression is upregulated upon fruit development both in orange, pale-green and white melon varieties, despite the lack of apocarotenoid volatiles in the later. Thus, the accumulation of beta-ionone in melon fruit is probably limited by the availability of carotenoid substrate.  相似文献

Japanese morning glory, Ipomoea nil, exhibits a variety of flower colours, except yellow, reflecting the accumulation of only trace amounts of carotenoids in the petals. In a previous study, we attributed this effect to the low expression levels of carotenogenic genes in the petals, but there may be other contributing factors. In the present study, we investigated the possible involvement of carotenoid cleavage dioxygenase (CCD), which cleaves specific double bonds of the polyene chains of carotenoids, in the regulation of carotenoid accumulation in the petals of I. nil. Using bioinformatics analysis, seven InCCD genes were identified in the I. nil genome. Sequencing and expression analyses indicated potential involvement of InCCD4 in carotenoid degradation in the petals. Successful knockout of InCCD4 using the CRISPR/Cas9 system in the white-flowered cultivar I. nil cv. AK77 caused the white petals to turn pale yellow. The total amount of carotenoids in the petals of ccd4 plants was increased 20-fold relative to non-transgenic plants. This result indicates that in the petals of I. nil, not only low carotenogenic gene expression but also carotenoid degradation leads to extremely low levels of carotenoids.  相似文献

采用RT-PCR和RACE技术从观赏向目葵‘闽葵3号’黄色花瓣中克隆到类胡萝卜素合成途径关键基因HaPDS的cDNA,该cDNA全长2017bp,具有一个1710bp的完整开放阅读框(ORF),编码一个570个氨基酸的蛋白质。序列分析表明,HaPDS编码的氨基酸序列与其他植物的PDs蛋白具有很高的同源性,在N-端有一个辅助因子结合结构域,C-端有一个类胡萝卜素结合域。系统进化树分析显示,观赏向日葵HaPDS与万寿菊、菊花蛋白亲缘关系较近。实时荧光定量RT-PCR技术分析表明,胁肋路因在花发育的盛花期表达量最高;不同组织中的表达量舌状花瓣〉苞片〉叶片〉绿色管状花〉黑色管状花:随着基因表达量的增加,花色由白色到黄色、金黄色转变。  相似文献

Sixteen xanthophylls were isolated from the petals of chrysanthemum (Dendranthema grandiflorum (Ramat.) Kitamura). Among them, (3S,5S,6R,3'R,6'R)-5,6-dihydro-5,6-dihydroxylutein (1) and five di-Z geometrical isomers of lutein-5,6-epoxide, i.e., 9Z,13'Z (2), 13Z,9'Z (3), 9'Z,13'Z (4), 9Z,13Z (5), and 9Z,9'Z (7), had never before been identified as natural products. All of the carotenoids isolated from chrysanthemum, except for (9Z)-violaxanthin, are beta,epsilon-carotene (alpha-carotene) derivatives. The analyses indicate that carotenoids from the petals of chrysanthemum have a very characteristic composition.  相似文献

月季(Rosa hybrida)花色丰富,是世界著名的观赏花卉。月季红双喜因其花瓣的变色特性而在市场上广受欢迎。该研究通过类黄酮和类胡萝卜素靶向代谢并结合转录组分析,发现红双喜的黄色花瓣呈色主要源于叶绿素、类胡萝卜素以及类黄酮的积累,红色花瓣呈色主要是花青素苷积累增加且糖苷化的结果。花青素苷合成关键基因CHI、ANS和UFGT,以及R2R3-MYB家族的AN2-like成员在红色花瓣中的强烈表达是花青素苷积累的分子基础;类胡萝卜素成分改变及相关基因的表达变化在红双喜花瓣变色过程中也起重要作用,并且miRNA156可能参与其调控过程。该研究揭示了月季红双喜花瓣变色的分子和化学基础,研究结果为观赏植物花色分子设计育种提供了重要理论依据。  相似文献

Orange petals of calendula (Calendula officinalis) accumulate red carotenoids with the cis-configuration at the C-5 or C-5' position (5-cis-carotenoids). We speculated that the orange-flowered calendula is a carotenoid isomerase (crtiso) loss-of-function mutant that impairs the cis-to-trans conversion of 5-cis-carotenoids. We compared the sequences and enzyme activities of CRTISO from orange- and yellow-flowered calendulas. Four types of CRTISO were expressed in calendula petals. The deduced amino acid sequence of one of these genes (CoCRTISO1) was different between orange- and yellow-flowered calendulas, whereas the sequences of the other three CRTISOs were identical between these plants. Analysis of the enzymatic activities of the CoCRTISO homologs showed that CoCRTISO1-Y, which was expressed in yellow petals, converted carotenoids from the cis-to-trans-configuration, whereas both CoCRTISO1-ORa and 1-ORb, which were expressed in orange petals, showed no activity with any of the cis-carotenoids we tested. Moreover, the CoCRTISO1 genotypes of the F2 progeny obtained by crossing orange and yellow lines linked closely to petal color. These data indicate that CoCRTISO1 is a key regulator of the accumulation of 5-cis-carotenoids in calendula petals. Site-directed mutagenesis showed that the deletion of Cys-His-His at positions 462-464 in CoCRTISO1-ORa and a Gly-to-Glu amino acid substitution at position 450 in CoCRTISO1-ORb abolished enzyme activity completely, indicating that these amino acid residues are important for the enzymatic activity of CRTISO.  相似文献

对开黄花菊花和开白花菊花材料分别和开红花菊花材料进行了正反交,结果表明,花色遗传比较复杂, 在以红花材料作为母本组合中表现为比较明显的偏母性遗传特征,而以黄花和白花材料为母本则不表现偏母性特征;除此之外,菊花花色遗传还表现出不完全显性和镶嵌显性的特点。在黄花材料3501和红花材料3509所得到杂种中发现了2个分枝出现花色嵌合体的现象,该嵌合体特征是花一边为红色,而另一边则出现镶嵌显性的现象。染色体分析表明,不同颜色嵌合体花瓣的染色体数目都是36条,因此,实验所得到的花色嵌合体不是由染色体数目变化造成的,而有可能是转座子插入影响色素合成基因造成的。  相似文献

对红色、黄色、粉紫色和白色菊花品种不同开放度的花序舌状花中CHS、CHI、DFR、F3H、F3′H和3GT基因的表达量进行了相对定量分析。结果表显示:6个基因的表达因不同花色、不同发育阶段而异。‘钟山红鹰’(红色)中各基因的表达量均较高,且均在Ⅱ(松蕾期)或Ⅲ(半开期)期达到峰值,其中DFR、3GT基因的表达量远高于其他花色品种。‘金陵娇黄’(黄色)中CHS、CHI基因表达量较高,且Ⅰ(紧蕾期)、Ⅱ期表达量高于Ⅲ、Ⅳ(盛开期)期;3GT、DFR基因表达量分别高或低于‘金陵笑靥’(粉紫色)品种中相应基因的表达量,但均比红色品种低;F3H在4个品种中表达量最低,F3′H表达量接近或略低于红色或粉紫色品种,且各阶段表达水平较稳定。‘金陵笑靥’中DFR表达量仅次于‘钟山红鹰’,3GT和CHS表达量低于红色与黄色品种。‘钟山雪桂’(白色)中各基因仅有微量表达,除F3H外各基因的表达量明显低于其他花色品种。研究表明,花色素结构基因DFR、3GT是菊花花色素合成的关键基因,DFR很可能是限速关键基因,一定表达水平的CHS、CHI也是菊花花色素合成所必须的,F3H基因与花色素合成不存在直接相关。  相似文献

以小苍兰(Freesia refracta)16个不同花色品种及后代为试验材料,对花瓣色素用特征显色反应和紫外-可见光谱扫描,分析其色素的成分和花色素苷的稳定性.结果表明,小苍兰花色的色素属于类黄酮化合物,含黄酮和花色素苷类物质,可能含有异黄酮,不含黄酮醇、二氢黄酮、二氢黄酮醇、查耳酮和橙酮,其中黄色系品种及后代还含有类胡萝卜素.避光下小苍兰花色素苷的稳定性要强于光照;温度对花瓣色素的稳定性有一定的影响.  相似文献

以红色、红心白边、粉红、玫红、黄色、黄心红边、浅粉和白色8种花色丽的格海棠花瓣为试验材料,采用目视测色法、RHSCC比色法和色差仪测定花瓣表型,通过组织切片法观察花瓣色素细胞的显微结构和分布特点,采用双光束紫外-可见光分光光度计和高效液相色谱-电喷雾离子化-质谱连用技术(HPLC-ESI-MS)测定分析花瓣中花青素苷的成分和含量,为探讨丽格海棠花色的呈色机理和花色育种提供参考。结果显示:(1)丽格海棠的明度L*随花瓣颜色变深而降低,红度a*则表现出相反趋势,红度(a*)和彩度(C*)值与明度(L*)呈显著负相关关系,且a*和C*是影响L*的主要因素。(2)红花品种花瓣色素主要分布于上表皮细胞和海绵组织中;红白花品种花瓣色素主要分布于上下表皮中,且下表皮积累量更多;粉色花和玫红花品种花瓣色素主要分布于上下表皮细胞;黄红花和粉白色花品种花瓣上表皮中含有少量色素,而黄花和白花品种花瓣几乎没有色素积累。各花色丽格海棠花瓣上表皮细胞均为圆锥形,且红花和红白花品种锥形化程度最高,它们花瓣下表皮细胞均呈扁平的长方形。(3)8个丽格海棠品种花瓣中共检测出15种花青素苷,其中10种为芍药素苷,3种为矢车菊素苷,1种为锦葵素苷,1种为飞燕草素苷,酰化花青素苷占多数;红花品种花瓣中总花青素苷含量最高,玫红花品种次之,黄花和白花品种中未检出;除粉红花品种外,其余含花青素苷的品种中芍药素苷含量最高,均占总花青素苷含量的50%以上,是花瓣的主要呈色物质。(4)丽格海棠花瓣中总花青素苷含量与其红度(a*)、彩度(C*)值呈正相关关系、与其L*值呈负相关关系。研究表明,花青素苷的积累有利于丽格海棠花瓣红色化,并影响其花瓣彩度(C*)及明度(L*);色素分布细胞数量和上表皮细胞锥形化明显影响花瓣呈色,且花瓣主要的呈色物质为芍药素苷,酰基化修饰可能影响其明度。  相似文献

以27个上海交通大学自育矮牵牛新种质为研究材料,对花色这一重要观赏性状及其花色素进行了系统研究。用RHSCC比色和色差仪测色方法描述了矮牵牛的花色表型,通过特征显色反应初步判断了矮牵牛的花色素类型,以标准曲线法和pH示差法等方法测定了矮牵牛3类花色素的含量。研究表明:这27个矮牵牛种质的花色可归于5个色系,以紫红色和红色为主;矮牵牛花色在CIELab表色系统中分布较广,而且不同色系花色参数的区分度较大。矮牵牛花瓣中含有类黄酮和花色苷,不含或含少量类胡萝卜素。13个被测种质的花瓣类黄酮含量在2.5~12.2 mg·/g–1 ·FW之间,花色苷含量在0.08~3.88 mg·g–1 FWmg/g·FW之间,而类胡萝卜素在矮牵牛花瓣中含量很低,远远低于类黄酮含量,在7个被测种质中,最高仅为0.216 mg·g–1 FWmg/g·FW,最低为0.004 mg·g–1 FWmg/g·FW。以上结果显示,5个色系矮牵牛所含花色素种类不尽相同,含量也有明显差异,其中紫红色系和红色系花瓣大多不含或含极少量类胡萝卜素,黄色系、白色系和紫色系花瓣的类黄酮含量较高,紫色系和紫红色系花瓣花色苷含量较高。  相似文献

We examined the role of carotenoid-binding protein (CBP) in yellow cocoon pigmentation. First, using yellow or white cocoon races, we investigated the linkage between the yellow pigmentation and CBP expression. CBP was expressed only in the silk gland of the yellow cocoon races, which utilize carotenoids for cocoon pigmentation. Furthermore, CBP expression in the silk glands of day 1-7 fifth instar larvae matched the period of carotenoid uptake into the silk gland. Finally, we gave double-stranded CBP RNA to Bombyx mori (B. mori) larvae to induce RNA interference. The significantly reduced expression of CBP in the silk gland of fifth instar larva was confirmed on day 4 and a decrease in yellow pigmentation was observed in the cocoon. We showed that CBP plays a key role in the yellow cocoon pigmentation caused by carotenoids.  相似文献

Ketocarotenoids and especially astaxanthin are high-valued pigments used as feed additives. Conventionally, they are provided by chemical synthesis. Their biological production is a promising alternative. For the development of a plant production system, Nicotiana glauca, a species with carotenoid-containing yellow pigmented flower petals, was transformed with a cyanobacterial ketolase gene. The resulting plants accumulated 4-ketozeaxantin (adinoxanthin), which is the first ketocarotenoid synthesized in flower petals by genetic modification. Due to the very late flowering in this tobacco species, N. tabacum was used to optimize the yield and ketocarotenoid product pattern by metabolic engineering of the ketolation steps of carotenogenesis. The highly carotenogenic nectary tissue in the flowers represents a model of a flower chromoplast system. By expression of a ketolase gene, it was possible to engineer the biosynthetic pathway towards the formation of 3'-hydroxyechinenone, 3-hydroxyechinenone, 4-ketozeaxanthin, 4-ketozeaxanthin esters, 4-ketolutein and 4-ketolutein esters. Some of these ketocarotenoids were also formed in the leaves of the trangenic plants. In particular, by co-expression of the ketolase gene in combination with a hydroxylase gene under an ubiquitous promoter, the formation of total carotenoids in nectaries increased by more than 2.5-fold. In the nectaries of this type of transformants, more than 50% of the accumulating carotenoids were keto derivatives. In addition, the levels of ketocarotenoid esters were much lower and a higher percentage of the free ketocarotenoids accumulated. These results open new promising perspectives for a successful metabolic engineering of keto-hydroxy carotenoid production in carotenogenic flowers.  相似文献

Xanthophylls, the pigments responsible for yellow to red coloration, are naturally occurring carotenoid compounds in many colored tissues of plants. These pigments are esterified within the chromoplast; however, little is known about the mechanisms underlying their accumulation in flower organs. In this study, we characterized two allelic tomato (Solanum lycopersicum L.) mutants, pale yellow petal (pyp) 1‐1 and pyp1‐2, that have reduced yellow color intensity in the petals and anthers due to loss‐of‐function mutations. Carotenoid analyses showed that the yellow flower organs of wild‐type tomato contained high levels of xanthophylls that largely consisted of neoxanthin and violaxanthin esterified with myristic and/or palmitic acids. Functional disruption of PYP1 resulted in loss of xanthophyll esters, which was associated with a reduction in the total carotenoid content and disruption of normal chromoplast development. These findings suggest that xanthophyll esterification promotes the sequestration of carotenoids in the chromoplast and that accumulation of these esters is important for normal chromoplast development. Next‐generation sequencing coupled with map‐based positional cloning identified the mutant alleles responsible for the pyp1 phenotype. PYP1 most likely encodes a carotenoid modifying protein that plays a vital role in the production of xanthophyll esters in tomato anthers and petals. Our results provide insight into the molecular mechanism underlying the production of xanthophyll esters in higher plants, thereby shedding light on a longstanding mystery.  相似文献
