SYNOPSIS. Three polycyclic, polyether, monocarboxylic acid antibiotics produced by and extracted from a strain of Streptomyces albus and designated A204 sodium, A28695A, or A204np, were specifically tested for activity against hepatic and incidentally against intestinal coccidia of rabbits. All were effective in preventing infections and acceptable to young rabbits when prepared in pelleted feed. Weight gain, however, was not as good in inoculated, medicated rabbits as in noninoculated, nonmedicated controls.
No gross pathologic changes were detected except in the livers of inoculated, nonmedicated controls which were severely infected and greatly enlarged. Livers of medicated rabbits were uninfected and normal in appearance and size.  相似文献

Abstract

Forty-eight male lambs were used to evaluate the effect of dietary supplementation of mannan-oligosaccharide (MOS) with or without parenteral Escherichia coli injection on their growth performance, feed conversion efficiency, blood metabolites, total serum immunoglobulin G (IgG) levels and antibody response. Lambs were randomly assigned to four groups of 12 animals each. In groups C (control) and CE (E. coli challenged), animals were fed commercial concentrate pellets and hay (50:50), and in groups M (MOS) and ME (MOS+E. coli challenged), animals were fed commercial concentrate pellets including MOS at 0.2% and hay (50:50). At day 15 and 30, animals in groups CE and ME were injected subcutaneously with 1 ml of phosphate buffered saline (PBS) suspension containing 106 cfu of heat inactivated non-toxigenic E. coli O157:H7, while animals in C and M groups were injected subcutaneously with 1 ml of PBS. The experimental period was 45 days. Data indicated that body weight of lambs at the end of the study were statistically non-significant among the groups. Blood metabolites, i.e. total protein, albumin, calcium and phosphorus concentrations were not affected significantly by MOS supplementation. However, administration E. coli lowered (p < 0.05) total protein, albumin and calcium concentrations in the serum on day 30. The IgG level was not different between groups. However, on day 45, the total IgG level was found to be higher (p < 0.05) in lambs that had received MOS and E. coli than in other groups. Application of MOS did not have any effect on the antibody response to E. coli as OD values.  相似文献

目的对普通级新西兰兔进行剖宫产,获得无菌兔,比较无菌兔、SPF兔和普通级兔的脏器系数。方法采取剖宫产手术获得无菌兔,在无菌隔离器内人工哺乳饲养。用电子天平和刻度尺测量无菌兔、SPF兔和普通兔心、肝、脾、肺、肾、肾上腺、胸腺、脑等脏器的脏器重量和长度,计算脏器系数。结果经检测无菌兔符合国家标准;无菌级、SPF级和普通级新西兰兔在左颌下腺、右颌下腺、心脏、肺、胆囊、脾脏、左肾上腺、右肾上腺、子宫+卵巢、左睾丸、胃长、蚓突长度、盲肠长度、盲肠总重、盲肠干重、胃总重、胃干重等17项指标差异显著(P〈0.05);脑、肝脏、左肾、右肾、胰脏、右睾丸、胃宽、小肠长度、直肠长度、大肠长度等10项指标差异不显著(P〉0.05)。结论不同等级微生物环境对新西兰兔脏器系数有显著影响。  相似文献

目的分析比较市售中国白兔与我院自繁培育的封闭群新西兰白兔血液流变学特性的差异。方法采集新西兰白兔和中国白兔的血液样品,分别检测比较其WBV(高、中、低切变率)、PV、HCT、ESR等14项血液流变学指标。结果 (1)新西兰白兔与中国白兔不同性别间血液流变学特性没有显著性差异(P〉0.05);(2)新西兰白兔和中国白兔血液流变学指标中,血浆粘度ηP,还原粘度低切Rls,还原粘度中切Rms,还原粘度高切Rhs及红细胞电泳指数RIE5个指标间有显著差异(P〈0.05)。其余9项指标差异不显著(P〉0.05)。结论实验兔的血液流变学特性与品种有一定的相关性。  相似文献

实验兔三个封闭群微卫星DNA多态性遗传分析   总被引:1,自引:0,他引:1  
目的 对日本大耳白兔、青紫蓝兔、新西兰兔三个封闭群体开展群体遗传学分析.方法 利用10个微卫星位点,进行Hardy-Weinberg平衡(HWE)检验,统计三个种群的基因频率、观测杂合度、期望杂合度、F值和遗传距离.结果 青紫蓝品种在12L1E11位点,新西兰品种在INRACCDDV0087位点与INRACCDDV0203位点,日本大耳白兔在Sat12位点与INRACCDDV0203,P<0.05,显著偏离HWE,多数表现为杂合子缺陷;三个群体在Sat13、So144、6L1F10、7L1F1、12L4A1、INRACCDDV0016点上均符合HWE;各位点平均等位基因数5.9,种群整体基因频率差别较大,其范围为0 -0.9060;三个种群的平均观测杂合度为0.6204,平均期望杂合度为0.6178;群体间分化系数(Fst)平均为0.0750,日本大耳白兔和青紫蓝兔遗传距离最近为0.1223,青紫蓝兔与新西兰兔遗传距离最远为0.1934.结论 三个种群的遗传结构均表现出遗传稳定性和均一性,在10个微卫星位点上呈现高度多态性,种群间遗传分化明显.  相似文献

Homologous skin-sensitizing antibodies were detected in the sera of rabbits infected with Taenia pisiformis. They were demonstrated in the sera of some rabbits as early as 8 days after first exposure and peaked at 30 days post-infection before declining over a period of 1 month. A second challenge dose of eggs boosted the titers markedly. The physico-chemical characteristics of the antibody in gel-filtration and anion-exchange chromatography, and in response to heat treatment and reduction and alkylation, conform to those previously described for reagin or IgE. PCA reactions first became positive 4 h after sensitization, and were maximal at 48 h ,although positive reactions occurred at sensitized sites when challenge was delayed for 8 weeks. The significance of this reaginic antibody response is discussed in relation to its potential role in the mechanism of acquired resistance to experimental cysticercosis.  相似文献

Using capillary high-performance liquid chromatography (HPLC)-fast atom bombardment (FAB) mass spectrometry (MS), both positive and negative FAB mass spectra of 24 cephem antibiotics with diethanolamine (DEA) and glycerol (GLY) as matrices are presented. In the positive mode, an internal quasi-molecular peak together with relatively abundant fragment peaks were obtained from all 24 drugs with both matrices, though DEA provided more information on molecular mass of a compound than did GLY for some drugs. In the negative mode, the background was generally lower than that in the positive, but neither the quasi-molecular nor molecular peak was detected in several drugs with either matrix. The drugs were isolated from serum samples using an octadecyl reversed-phase cartridge; recoveries were generally over 60%. With this isolation and the capillary HPLC-FAB-MS in the positive mode, ceftriaxone and cefazolin, two of the most popular cephem antibiotics, were successfully identified in 0.5 ml of sera obtained from a clinical or an autopsy case.  相似文献

In vitro gas production kinetics of six different substrates, pectin (PEC), xylan (XYL), starch (STA), cellulose (CEL), commercial compound feed (FEED; 201 g crude protein per kg, 155 g crude fibre per kg, 334 g neutral-detergent fibre (NDF) per kg and 190 g acid-detergent fibre (ADF) per kg) and an NDF prepared from commercial compound feed (NDFFEED) were determined using the caecum contents of weaned rabbits (36 days of age) and of rabbits at slaughter age (78 days of age) as inoculums. The cumulated gas production over 96 h of incubation was modelled with Gompertz model, and the kinetic parameters compared. The total potential gas production (parameter ‘B’ of the Gompertz model) was not affected (P>0.05) by the inoculum source, except with STA, where rabbits at slaughter weight had significantly higher total potential fermentability (314 ml/g dry matter (DM)) than those at weaning age (189 ml/g DM). Intensities of fermentation (maximum fermentation rate; MFR) of PEC (32.2 ml/h) and XYL (24.4 ml/h) were significantly greater in rabbits at weaning, while that of STA (45 ml/h) was significantly lower than at slaughter age (23.0, 14.3 and 14.0 ml/h for PEC, XYL and STA, respectively). The MFRs of CEL and NDFFEED were very similar between inoculum sources. In the first 10 h of fermentation which correspond to the normal retention time of the substrates in the caecum, the highest amount of gas was produced from PEC, followed by FEED and XYL. These substrates had a time of maximum fermentation rate (TMFR) at both rabbit ages short enough (8.0 and 9.5 h for PEC, 9.5 and 6.6 h for FEED, 13.7 and 14.2 h for XYL at weaning and at slaughter age, respectively) to be almost completely fermented in vivo.  相似文献

本文旨在研究电刺激家兔迷走神经诱导的黑-伯(Hering-Breuer,HB)反射中的学习和记忆现象。选择性电刺激家兔迷走神经中枢端(频率10~100Hz,强度20~60μA,波宽0.3ms,持续60s),观察对膈神经放电的影响。以不同频率电刺激家兔迷走神经可模拟HB反射的两种成分,即类似肺容积增大所致抑制吸气的肺扩张反射和类似肺容积缩小所致加强吸气的肺萎陷反射。(1)长时高频(≥40Hz,60s)电刺激迷走神经可模拟呼吸频率减慢,呼气时程延长的肺扩张反射。随着刺激时间的延长,膈神经放电抑制的程度逐渐衰减,表现为呼吸频率的减慢(主要由呼气时程延长所致)在刺激过程中逐渐减弱或消失,显示为适应性或“习惯化”的现象;刺激结束时呼吸运动呈现反跳性增强,表现为一过性的呼气时程缩短,呼吸频率加快,然后才逐渐恢复正常。长时低频(〈40Hz,60s)电刺激迷走神经可模拟呼吸频率加快、呼气时程缩短的肺萎陷反射。随着刺激时间的延长,膈神经放电增强的程度逐渐衰减,同样表现出“习惯化”现象;刺激结束后,膈神经放电不是突然降低,而是继续衰减,表现为呼气时程逐渐延长,呼吸频率逐渐减慢,直至恢复到前对照水平,表现了刺激后的短时增强效应。(2)HB反射的适应性或“习惯化”程度反向依赖于刺激强度和刺激频率,表现为随着刺激强度和频率的增加,膈神经放电越远离正常基线水平,即爿惯化程度减弱。结果表明,家兔HB反射具有“习惯化”这一非联合型学习现象,反映与其有关的呼吸神经元网络具有突触功能的可翅性,呼吸的中枢调控反射具有一定的适应性。  相似文献

Experiments were conducted to develop a simple and reliable technique to produce chimeric rabbits from morula stage embryos. In Experiments 1 and 2, an in-vitro test of viability was initially performed by culturing embryos to the blastocyst stage. Ninety-three percent of the “chimeric” embryos developed to the blastocyst stage compared to 94% for controls when embryos were manipulated soon after collection (Exp. 1). Eighty-one percent chimeric embryos and 78% control embryos developed to blastocyst stage when embryos were held at room temperature for 4 hr (Exp. 2). In Experiment 3, enough morula-stage embryos were available from true breeding Dutch-belted and albino rabbits to form potentially 67 diverse “color” pairs. These micromanipulated pairs of morulae were successfully combined to produce 64 chimeric embryos (96%, 64/67). They were transferred to the uteri of seven recipient does and three became pregnant producing 13 young. Four of the young exhibited substantial overt chimerism (31%) and one more was a possible chimera.  相似文献

The Introduction of antibiotics into the clinical use in the middle of the 20th century had a profound impact on modern medicine and human wellbeing. The contribution of these wonder molecules to public health and science is hard to overestimate. Much research has informed our understanding of antibiotic mechanisms of action and resistance at inhibitory concentrations in the lab and in the clinic. Antibiotics, however, are not a human invention as most of them are either natural products produced by soil microorganisms or semisynthetic derivatives of natural products. Because we use antibiotics to inhibit the bacterial growth, it is generally assumed that growth inhibition is also their primary ecological function in the environment. Nevertheless, multiple studies point to diverse nonlethal effects that are exhibited at lower levels of antibiotics. Here we review accumulating evidence of antibiosis and of alternative functions of antibiotics exhibited at subinhibitory concentrations. We also speculate on how these effects might alter phenotypes, fitness, and community composition of microbes in the context of the environment and suggest directions for future research.  相似文献

C1027 is a macromolecular antitumour antibiotic produced by Streptomyces globispourus C1027 and consists of an apoprotein and a non‐protein labile chromophore. Little is known about how the thermally unstable chromophore is stabilized by the apoprotein. The purified C1027 was monodisperse according to dynamic light‐scattering measurements and crystallized in two different crystal forms from two different starting conditions using the vapour‐diffusion method. Condition I yielded hexagonal prism crystals having space group P31/P32 and unit‐cell parameters a = b = 66.8, c = 55.4 Å. Diffraction data were collected to 2.1 Å resolution using an in‐house Rigaku rotating Cu anode X‐ray generator. Another condition produced rod‐like crystals with space group P3121/P3221 and unit‐cell parameters a = b = 55.15, c = 55.87 Å. A data set to 1.8 Å resolution was collected from a rod‐like crystal using a MAR CCD detector at the SRS synchrotron source.  相似文献

Hormonal effect on glycoproteins and glycosaminoglycans in rabbit uteri   总被引:1,自引:0,他引:1  
The effect of the female hormones on glycoproteins and glycosaminoglycans in uteri has been studied.The uteri taken from the ovariectomized rabbits treated with estrogen, estrogen plus progesterone, and sham administration (control) were incubated in vitro with [U-14C]glucose. Subsequently, the tissues were digested extensively with pronase, yielding crude glycan fractions. The amount and radioactivity of the crude glycan fraction increased by the treatment with estrogen, but reduced to certain level with progesterone.Separation of glycoproteins and glycosaminoglycans was achieved by stepwise elution from Dowex 1 (X2, chloride form) with increasing concentration of NaCl. The yield and radioactivity, together with the results of chemical, enzymatic, and electrophoretic studies on the resulting fractions indicated that the metabolism of a slightly acidic glycoprotein, hyaluronic acid, sulfated glycoproteins, low-sulf ated chondroitin sulfate, heparan sulfate, chondroitin 4-sulfate, and dermatan sulf ate were stimulated remarkably with estrogen, but the stimulation was restored to certain level with progesterone. The degree of the change with these hormones was, however, found to be different from each other. It was noticed that sulfated glycoproteins were the most sensitive to the hormones. On the other hand, the estrogenic stimulation of the metabolism of a neutral glycoprotein and oversulfated chondroitin sulfates was not restored with progesterone.  相似文献

目的:探究三七茎对家兔实验性高脂血症的影响。方法:将家兔随机分为4组,除对照组外,其余组加喂猪油和胆固醇,并对低剂量组和高剂量组喂以不同剂量的三七茎,利用酶学终点法于给药前和给药后2,4,6周测定其TC、TG、HDL-C、LDL-C值。结果:与对照组比较,高脂组家兔的TC、TG、LDL-C值升高,HDL-C值降低,差异有统计学意义(P<0.05)。与高脂组比较,低剂量组和高剂量组的TC、TG、LDL-C值降低,HDL-C值升高,差异有统计学意义(P<0.05)。结论:三七茎对实验性高脂血症家兔具有降血脂作用。  相似文献

Intravenous inoculation of young rabbits with 1.4 × 105 spores ml-1 of Absidia corymbifera produced gross and microscopic changes principally in the kidneys. Grossly, there was congestion, haemorrhages and greyish-white necrotic foci giving a mottled appearance to the kidneys. Microscopically, besides congestion and haemorrhages, there was a granulomatous reaction characterized by a central necrotic mass containing mycelium surrounded by a reactive zone of neutrophils, lymphocytes, a few macrophages and giant cells. Fibrous tissue proliferation was also present. Characteristic dichotomously branched non-septate fungal hyphae could be demonstrated in the sections of kidneys up to 14 days of infection. In one rabbit, which had died on the 8th day, small granulomatous lesions with fungal hyphae were noticed in the liver's parenchyma. Die vorliegenden Untersuchungen sind Bestandteil eines komplexen Forschungsprogramms zur Weiterentwicklung der energetischen Futterbewertung im Nettoenergie‐Fett‐System. Sie wurden mit dem Ziel durchgeführt, neue Ergebnisse zur Erfassung des Zusammenhanges zwischen Ort und Art der Nährstoffverdauung und der energetischen Verwertung von Rationen bei der Tierart Rind zu erarbeiten. Für 9 Rationsvarianten mit jeweils 3 Varianten der Stärkeherkunft (Gerste, Mais, Kartoffeln) und ihres Rationsanteiles (50, 25 und 10 %) wurde an weitgehend ausgewachsenen Ochsen mit Hilfe von duodenalen Brückenfisteln auf dem Ernährungsniveau 1.7 die ruminale Nährstoffverdaulichkeit gemessen. Bei Stärkeeinnahmen zwischen 484 und 2573 g je Tier und Tag wurden Mengen an ruminai und postruminal verdauter Stärke zwischen 444 und 2336 bzw. 10 und 284 g je Tier und Tag bestimmt. Für die organische Substanz, Stärke, wasserlöslichen Kohlenhydrate und N‐freien Reststoffe wurden hohe relative Anteile der ruminai verdauten an den scheinbar verdauten Nährstoffen mit Werten zwischen 78 und 88, 83 und 98, 93 und 97 bzw. 88 und 100% ermittelt.  相似文献

The presence of the fermentable sugar d-mannitol in the diet improves nitrogen (N) utilization in rabbits. To clarify the mechanism by which d-mannitol improves N utilization, we studied the effect of d-mannitol on the fate of blood urea N in growing rabbits. Growing rabbits received a control diet or a diet containing d-mannitol, which were formulated by adding 80 g/kg glucose or d-mannitol to timothy hay. After 9 days of feeding of the experimental diets, 15N-urea was administrated intravenously under anesthesia 1 h before slaughter. The blood urea level (concentration of both urea N (43.6% of the control group (CG), P < 0.05) and 15N (95% of the CG, P < 0.05) in blood serum) was reduced in the mannitol group. The concentration and amount of N, and 15N atom % excess in the contents of the cecum and colon were higher (P < 0.05) in the rabbits fed the mannitol diet than in rabbits fed the control diet, especially in the cecum. The consumption of mannitol caused bacterial proliferation in the cecum characterized by marked short-chain fatty acid production (165% of the CG, P < 0.05), decreased cecal ammonia N (73% of the CG, P < 0.05) and elevated cecal bacterial N (150% of the CG, P < 0.05). On the other hand, addition of d-mannitol to the diet decreased N (80% of the CG, P < 0.05) and 15N (77% of the CG, P < 0.05) excretion in the urine. These results indicate that d-mannitol increases the transfer of blood urea N to the large intestine, where it is used for bacterial N synthesis.  相似文献

抗生素耐药性及其在全球范围内的传播已成为国际关注的热点。本文结合最新文献,综述了抗生素抗性基因在环境中的来源、传播、分布以及新型抗性基因的发现等方面的研究进展。环境中抗生素抗性基因的来源主要是环境中细菌的内在抗性基因及随人或动物粪便排到体外的抗性细菌。功能宏基因组学技术的应用极大地丰富了人们对抗生素抗性组学的认知,并已从环境中筛选到多种新型抗性基因。近年来,由于抗生素在医疗以及养殖业中的大量使用,增加了抗性基因在环境中的丰度和多样性,加速了抗性基因在环境中的传播,在多种环境介质(如养殖水域、污水处理厂、河流、沉积物和土壤等)均检测到多种高丰度的抗生素抗性基因。我们建议今后在以下方面开展深入研究:(1)抗性基因传播和扩散的机制;(2)新型抗性基因筛选和抗性机制;(3)抗生素和抗性基因环境风险评估体系等。  相似文献
