The segregation of suri and huacaya fleece types in alpacas in the United States deviates from that expected from results of previously published studies from other regions. Suri × suri matings produced 278 huacaya and 1702 suri offspring. Suri × huacaya matings produced 89 huacaya and 56 suri offspring. Huacaya × huacaya matings produced 19,633 huacaya and 4 suri offspring. No linkage or other influence of sex was noted. These results are consistent with a single autosomal dominant gene controlling suri fleece production, with an additional relatively common genetic mechanism that can suppress the suri phenotype in some animals. These results are especially important in cases where the two fleece types are crossed with one another, as they result in a relative underproduction of suri fleeces.  相似文献

When exposed to high ambient temperatures, mammals lose heat evaporatively by either sweating from glands in the skin or by respiratory panting. Like other camelids, alpacas are thought to evaporate more water by sweating than panting, despite a thick fleece, unlike sheep which mostly pant in response to heat stress. Alpacas were brought to Australia to develop an alternative fibre industry to sheep wool. In Australia, alpacas can be exposed to ambient temperatures higher than in their native South America. As a young industry there is a great deal of variation in the quality and quantity of the fleece produced in the national flock. There is selection pressure towards animals with finer and denser fleeces. Because the fibre from secondary follicles is finer than that from primary follicles, selecting for finer fibres might alter the ratio of primary and secondary follicles. In turn the selection might alter sweat gland density because the sweat glands are associated with the primary follicle. Skin biopsy and fibre samples were obtained from the mid-section of 33 Huacaya alpacas and the skin sections were processed into horizontal sections at the sebaceous gland level. Total, primary, and secondary follicles and the number of sweat gland ducts were quantified. Fibre samples from each alpaca were further analysed for mean fibre diameter. The finer-fibred animals had a higher total follicle density (P<0.001) and more sweat glands (P<0.001) than the thicker-fibred animals. The fibre diameter and total follicle density were negatively correlated (R2=0.56, P<0.001). Given that the finer-fibred animals had higher follicle density and more sweat glands than animals with thicker fibres, we conclude that alpacas with high follicle density should not be limited for potential sweating ability.  相似文献

Hair follicle activity and fibre growth were studied using histological sections from the skin of five adult feral does sampled every four weeks for 18 months. The main period of guard hair growth in primary follicles was from November to April. Secondary follicles grew fine, long, nonmedullated fibres (cashmere) from December to June. Shedding of these fibres from secondary follicles had commenced by July and cashmere was absent from the fleece by November. From September to December a subsidiary hair cycle occurred in many secondary follicles which produced minute (vellus) fibres, less than 2.4 mm in length. Some secondary follicles probably shed their cashmere fibres and remain quiescent over spring. Annual pelage changes were therefore achieved with one main growth period, although many secondary follicles underwent another brief hair cycle in spring.  相似文献

Thyroidectomy surgery performed late in gestation results in perturbations in wool follicle development in foetal sheep, showing the importance of thyroid hormones for wool follicle development. The aim of this study was to determine the influence of transient manipulation of thyroid hormone status at a time corresponding with foetal primary wool follicle initiation. Pregnant Merino ewes (n = 12 per treatment) were treated daily between gestational days 55 and 64 with control (vehicle), exogenous thyroxine (T4) or propylthiouracil (PTU), an inhibitor of T4 synthesis, and conversion to the active form of the thyroid hormone (triiodothyronine). There were no significant differences in birth weight, gestational lengths and birth coat scores of the resultant lambs. The total primary and secondary follicle densities were significantly lower in lambs exposed to exogenous T4 compared with other treatments (P < 0.05). However, the T4 group displayed a higher proportion of mature secondary follicles (reflected by increased mature secondary follicle densities and mature secondary/primary follicle ratios) than the other treatment groups (P < 0.05). The skin morphology of the lambs differed 12 months later, with the T4 group having significantly higher total follicle densities compared with the PTU group, largely attributed to increased mature and total secondary follicle densities. However, this increase in wool follicle densities did not translate to differences in the fleece yields and weight, fibre diameter, staple lengths or any other fibre parameters. This study showed that transient manipulation of thyroid hormone status during foetal primary follicle initiation does have long-term consequences on the morphology of wool follicles, in particular the maturity of secondary wool follicles.  相似文献

Skin and fleece traits have been characterized in four lines of Merino sheep selected for high- and low-fibre diameter (D +/-) and staple length (L +/-) from a medium-woolled flock. Over a period of 20 years, each line responded in the desired direction, producing fleeces composed of thick or thin fibres and long or short wool staples. However, variations in the amounts of wool grown that might be expected from these procedures were compensated by changes in unselected characters. Thus a predicted difference in fleece weights between high and low staple length lines was reduced by an increase in fibre crimp frequency in L- sheep. Similarly, changes induced in fibre diameter in the D lines resulted in small effects on fleece weight in comparison to the large (and inverse) effects on follicle numbers. Towards the end of the selection regime, mean follicle density in D- sheep was twice that of D+ sheep. This intriguing response within the follicle population was examined further: an analysis of the relationship between follicle density and fibre diameter amongst the four lines revealed a highly significant, negative linear correlation. The implication of this statistical association is that the numbers of follicles initiated in skin during foetal life had a direct bearing on the sizes of wool fibres eventually produced. It was concluded that both features must be under the control of a single developmental mechanism. Since the expression of each of the characters is separated in time, the mechanism must be activated during the earlier event, i.e. at or before the phase of follicle initiation.  相似文献

A subpopulation of Yanshan cashmere goats with crimped fibre has emerged in a closed population in recent years, but little is known about differences in cashmere production performance, skin characteristics, and fibre-style-related genes between goats with different cashmere fibre styles. Therefore, the aim of this study was to investigate fibre characteristics, cashmere yield, hair follicle traits, and fibre-style-related genes in cashmere goats with the two cashmere fleece styles—non-crimped and crimped. Based on the cashmere fibre type, 80 six-month-old female Yanshan cashmere goats were used in this study: 40 goats with non-crimped fleece (NCF) and 40 with crimped fleece (CF). The growth performance and cashmere production of the goats were recorded. Skin samples were collected to determine hair follicle traits and gene sequencing. The results indicated that there were no differences in initial live weight, final live weight, average daily feed intake, and average daily gain between the two groups of goats (P > 0.05). The total yield of cashmere and the stretched length of fibre of the CF goats were higher (P < 0.01 and P < 0.05, respectively) and fibre diameter was lower (P < 0.05) than that of the NCF goats. There were no between-group differences in the density and activity of primary and secondary hair follicles, secondary-to-primary fibre ratio, depth of primary follicles, or epidermal thickness. However, the depth of secondary follicles and dermal thickness were higher (P < 0.05) in NCF goats than in CF goats. There were mutations in the KRT5, KAP8, KRT8, KRT74, KRT34, KRT1, KRT71, KRT6A, KAP6, KRT81, and KRT83 genes, four of which caused amino acid changes. The allele and genotype frequencies of base mutations in the KRT5, KAP8, KRT34, KRT1, KRT6A, KRT81, and KRT83 genes were different in the NCF and CF goats (P < 0.05). The distribution and content of the secondary structure elements and tertiary structures of proteins differed between the wide-type and mutated KRT1 and KRT6A proteins. KRT1, KRT6A, KRT71, and TGFα mRNA expression levels were significantly higher in CF goats than in NCF goats (P < 0.05). It is concluded that cashmere goats that have fleece with crimped fibres produce a greater yield of fleece with finer diameter fibres than those with conventional straight cashmere fibres. These differences in fibre properties may be associated with mutations in the genes coding for KRT1 and KRT6A.  相似文献

Crimp, a distinguishing feature of sheep fibres, significantly affects wool value, processing and final fabric attributes. Several explanations for fibre bending have been proposed. Most concentrate on relative differences in the physicochemical properties of the cortical cells, which comprise the bulk of the fibre. However, the associations between cortical properties and fibre crimp are not consistent and may not reflect the underlying causation of fibre curvature (FC). We have formulated a mechanistic model in which fibre shape is dictated primarily by the degree of asymmetry in cell supply from the follicle bulb, and the point at which keratinisation is completed within the follicle. If this hypothesis is correct, one would anticipate that most variations in fibre crimp would be accounted for by quantitative differences in both the degree of mitotic asymmetry in follicle bulbs and the distance from the bulb to the point at which keratinisation is completed. To test this hypothesis, we took skin biopsies from Merino sheep from sites producing wool differing widely in fibre crimp frequency and FC. Mitotic asymmetry in follicle bulbs was measured using a DNA-labelling technique and the site of final keratinisation was defined by picric acid staining of the fibre. The proportion of para- to ortho-cortical cell area was determined in the cross-sections of fibres within biopsy samples. Mitotic asymmetry in the follicle bulb accounted for 0.64 (P < 0.0001) of the total variance in objectively measured FC, while the point of final keratinisation of the fibre accounted for an additional 0.05 (P < 0.05) of the variance. There was no association between ortho- to para-cortical cell ratio and FC. FC was positively associated with a subjective follicle curvature score (P < 0.01). We conclude that fibre crimp is caused predominantly by asymmetric cell division in follicles that are highly curved. Differential pressures exerted by the subsequent asymmetric cell supply and cell hardening in the lower follicle cause fibre bending. The extent of bending is then modulated by the point at which keratinisation is completed; later hardening means the fibre remains pliable for longer, thereby reducing the pressure differential and reducing fibre bending. This means that even highly asymmetric follicles may produce a straight fibre if keratinisation is sufficiently delayed, as is the case in deficiencies of zinc and copper, or when keratinisation is perturbed by transgenesis. The model presented here can account for the many variations in fibre shape found in mammals.  相似文献

Intravenous infusion of 4.5-4.7 mg of mouse epidermal growth factor (mEGF) into nine castrated male Merino sheep for 26 h resulted in complete casting of the fleeces 6-8 days later. The morphological changes which occurred in the skin were studied in skin samples taken before infusion and at intervals between 1 h and 42 days after the infusion had begun. Wool fibres from the shed fleeces were examined with the scanning electron microscope. Increased cell proliferation occurred in the epidermis and sebaceous glands, whereas the wool follicles regressed. Transient dermal haemorrhages occurred during the first 3 h of infusion. The fibre and inner root sheath in the keratogenous zone of 30-40% of the follicles were partially disrupted within the first 6 h of mEGF infusion; catagen began in all follicle bulbs within 24 h. Fibre and inner root sheath production, although markedly reduced, continued in about 60% of follicles which had partially regressed, but production ceased in the remainder in which tapered ends formed on the fibres prior to shedding. Follicles began to regenerate asynchronously 4-8 days after the beginning of infusion and completed their development during the next 3 weeks. The follicle regression and fleece casting induced by mEGF infusion, and subsequent follicle regeneration were completed more rapidly than observed previously with other depilatory agents, and, except for prolonged epidermal thickening, there was no lasting cutaneous abnormality.  相似文献

This paper reviews genetics of fibre production and fleece characteristics in small ruminants, Angora rabbit and South American camelids with a special distinction between single-coated (SC) and double-coated (DC) species. Considering the biology of fibre production, there are variations in coat composition and structure, fibre growth pattern and fibre structure and quality between these two main kinds of fibre-producing animals. In SC species, all fibres are nearly similar in dimensions and are produced from individual follicles that have a very long period, essentially permanent, of active growth without a synchronous phase of rest between follicles. In contrast, in DC species the fleece comprises a coarse outer coat and a fine inner coat with variations of coat composition and structure, and fibre growth pattern according to the season with a well-defined duration of fibre growth. Genetic basis of hair growth pattern, coat composition and fibre structure are different between species. In small ruminants, these coat characters are additive and because of several genes whereas in rabbit, several autosomal recessive genes determine fibre growth, coat composition and structure. In alpaca, the fleece type (Suri or Huacaya) is determined by a single dominant gene. This paper also reviews genetic parameters of fibre production traits in Angora goat, Angora rabbit and alpaca in which many aspects of the genetic basis of fibre production are analogous. There are many traits controlling both fibre quality and fibre quantity, and most of these traits tend to be moderately to strongly inherited so that a rapid genetic progress in any traits is possible and indeed has been achieved. However, there are differences in breeding programmes. In DC Angora rabbit, selection for one single trait, the easy measurable total fleece weight has general beneficial effects on fleece quality. However, because of antagonistic relations between qualitative and quantitative traits in SC species, achieving this goal requires a multi-trait selection index approach. Gene mapping studies have recently identified several putative quantitative trait loci and major genes affecting fibre and fleece characteristics in sheep, goat and rabbit are reviewed. The whole genome sequence of sheep and rabbit will be available in the near future and the use of high-density single nucleotide polymorphism chip will allow fine mapping and dissection of the genetic basis of many production traits including fibre production and fleece characteristics. The application of these techniques will thus contribute to improving the efficiency, profitability and sustainability of small ruminant and rabbit fibre production.  相似文献

Mammalian hair follicles are complex multicellular structures in the skin, which produce hair fibre under the influence of locally produced and systemic signalling systems. Investigation to determine mechanisms of regulation, follicular responses and the importance of nutritional supply have utilised a number of in vivo and in vitro approaches. Included in these are studies on isolated intact anagen secondary follicles singly or in groups with incubation in culture medium. These utilise techniques developed for investigation of follicles from human skin. Results from selected studies reviewed here demonstrate differences in capacity for hair growth and protein synthesis between secondary follicles from Angora and cashmere-bearing goats. Mohair follicles were shown to exhibit faster hair shaft elongation both in vivo and in vitro, to have greater DNA content per follicle and to deposit significantly more protein per follicle and per unit of DNA. Incubation of anagen mohair and cashmere follicles in the presence of melatonin or prolactin showed positive responses in hair shaft growth and protein synthesis to both signalling molecules. This result indicated directly acting effects on the follicle in addition to any indirect effects arising at a whole animal level in response to, for example, variation in photoperiod. Similarly, epidermal growth factor was shown to alter elongation and protein synthesis in mohair follicles and to produce, at higher concentration, club hair structures similar to effects observed in other species. The vitamin biotin was shown to be important in maintaining viability of isolated sheep secondary hair follicles where supplementation increased the proportion continuing to grow. Effects on growth and apparent protein synthesis suggested comparatively lesser effects on follicles, which remained viable. Histology on follicles indicated effects of biotin deficiency in reducing proliferation of basal keratinocytes. The final study, included in this review, demonstrated that supply of the essential sulphur-containing amino acid l-methionine was necessary to maintain the viability and growth of mohair follicles. l-cysteine was not required in the presence of l-methionine, although there was evidence of an optimisation when both amino acids were present in adequate concentrations. Consideration is given to the importance of transport mechanisms and capacity to utilise absorbed nutrients when considering optimising nutritional supply to individual follicles. These may then provide targets for attainment in applied nutrition of animals in vivo.  相似文献

Studies with gene knockout mice have shown that Sonic hedgehog (Shh) is required for early development of hair follicles, but the role of this gene in the late stages of follicle development is not clear. By using an organ culture system of embryonic mouse skin, the role of Shh signaling in the early and late stages of follicle development was investigated. In the early stage of follicle development, the downward growth of the follicular epithelium was suppressed by cyclopamine, an inhibitor of Shh signaling, and accelerated by recombinant Shh. In addition, cyclopamine impaired dermal papilla formation, accompanied by the rearrangement of papilla cells, but not the elongation of the follicular epithelium at the later stage. These results suggest that Shh signaling is required for the proliferation of epithelial cells in the early development of hair follicles and for the morphogenetic movement of mesenchymal cells at the later stage of follicle development.  相似文献

The development of hemorrhagic anovulatory follicles (HAF) involves luteinization and hemorrhage of the follicle. This is observed on ultrasound as an increase in the echogenicity of the granulosa layer and formation of echoic particles in the antrum. The inhibition of prostaglandin synthesis with flunixin meglumine (FM) during the periovulatory period induces ovulatory failure with development of luteinized unruptured follicles (LUF). These two types of anovulatory follicles appear to share similar ultrasound features but they have not been compared critically. The following endpoints: follicle diameter, follicular contents score, interval from hCG administration to beginning of follicular hemorrhage, interval from hemorrhage to organization of follicular contents, and cycle length were studied and compared in mares with HAF (n = 11) and LUF (n = 13). The objective of this study was to elucidate whether these two unruptured follicles have a consistent clinical pattern of development and therefore can be considered as part of the same anovulatory syndrome. None of the endpoints analyzed differed significantly between HAF and LUF. However, there was a greater individual variation in HAF as compared with LUF in regards to interval from hCG to hemorrhage, follicular diameter at the administration of hCG, and beginning of hemorrhage. In conclusion, HAF share a similar cascade of ultrasound characteristics with the experimentally induced LUF. This finding may provide new insights in elucidating the pathogenesis of HAF.  相似文献

Ultrasonic evaluation of the preovulatory follicle in the mare   总被引:4,自引:0,他引:4  
Ultrasonically visible characteristics of preovulatory follicles in mares which single ovulated were studied daily for 79 preovulatory periods in 40 mares. The preovulatory follicle became the largest follicle in the ovary from which ovulation later occurred six or more days before ovulation in 65 of 79 (82%) preovulatory periods; the mean was day -7 (range, day -14 to day -4). The increase in mean diameter of the preovulatory follicle was linear (R(2)=99.5%) over day -7 (29.4 +/- 0.8 mm) to day -1 (45.2 +/- 0.5 mm; growth rate, 2.7 mm/day). Follicles which double-ovulated were smaller (P<0.05) on day -1 (36 +/- 1.6 mm; n=12 follicles). Preovulatory follicles exhibited a pronounced change in shape from a spherical to a conical or pear-shaped structure in 84% of the preovulatory periods. Remaining follicles retained a spherical shape. Scores representing thickness of the follicular wall increased (P<0.05) as the interval to ovulation decreased. There was no significant difference among days in mean gray-scale value of the follicular wall or in echogenicity of the follicular fluid. Although diameter and shape of the follicle and thickness of the follicular wall changed during the preovulatory period, no reliable ultrasonically visible predictor of impending ovulation was found.  相似文献

Ovarian follicular dynamics were determined in adult llamas by ultrasonography and palpation per rectum and hormone analysis (estradiol-17 beta and estrogen conjugates) of plasma and urine. The relationship of gonadotropin secretion to follicular development was determined by the analysis of plasma FSH and LH concentrations. Progesterone analysis of plasma was used to verify or deny the presence of CL. Final follicular development (from 3 mm) averaged 4.8 days, while the duration of the mature follicle (8-12 mm) averaged 5.0 days; regression of the follicle occurred over about 4 days. The development of a subsequent dominant follicle usually began within 2-3 days after onset of regression of the dominant follicle. While several follicles were present at the time of the demise of the dominant follicle, only one follicle continued to develop. The interval between ovarian follicle waves averaged 11.1 days. Dominant follicle activity alternated between ovaries in 81% of the cycles. The occurrence of dominant follicles was evenly distributed between ovaries. While plasma estradiol and estrogen conjugate concentrations were positively associated (p less than 0.05) with follicular activity, urinary estrogen conjugate concentrations best reflected ovarian follicular dynamics (p less than 0.001). Daily FSH concentrations in plasma were not correlated with follicular activity. LH concentrations in plasma were low in all animals throughout the study, indicating estrogen from developing ovarian follicles does not induce the release of LH. Progesterone values were low during the study, indicating that the llama does not spontaneously ovulate, at least under the conditions of this study. Atretic follicles regularly occur in the ovary of the house fly, Musca domestica. The frequency of ovarian follicular atresia and the proportion of atretic follicles per ovary are related to the stage of oögenesis and to the age of the females. Only vitellogenic follicles may become atretic. The atresia may occur at any stage of vitellogenesis, though most follicles become atretic in mid-vitellogenesis. Atretic follicles are completely resorbed within 24–36 hr. The follicle cells may play a synthesizing role during growth and disintegrating one during follicle resorption. The induction of glycogen synthesis by the cessation of RNA and protein synthesis and by vitellogenesis in normal follicles is discussed. The same processes occur prematurely in the atretic follicle which can be thus distinguished by a high content of glycogen.  相似文献

目的观察出生后小鼠不同部位皮肤毛囊早期发育生长差异及细胞色素C的表达分布。方法对新生1~9日龄的KM小鼠背部、尾部和触须部皮肤取材,进行HE染色,用二步法免疫组织化学对组织进行细胞色素C进行表达分布检测。结果新生小鼠不同部位皮肤毛囊发育差异很大,这种差异不仅体现在形态差异上,而发育时间的差异也十分明显。小鼠出生后背部皮肤和尾部皮肤的毛囊发育都经过了一个非线性的发育和生长期,过了非线性的发育和生长期才开始快速生长,相比较尾部发育略迟于背部。触须部毛囊发育特征和背部尾部差异很大,一出生便可看到较成熟的触毛,没有经过稳定期便开始发育。结论通过形态学比较,结合CytC表达分布水平,发现新生小鼠不同部位皮肤毛囊早期发育存在形态和时间上的差异。  相似文献

The follicle wall was previously shown to be involved in insulin induction of oocyte maturation in Rana pipiens ovarian follicles. Steroidogenic involvement in insulin induction of maturation was investigated following development of a radioimmunoassay (RIA) for progesterone to measure endogenous progesterone associated with in vitro incubates. Insulin and frog pituitary homogenate (FPH) were both found to elevate progesterone levels significantly in these incubates. FPH was more effective in elevating progesterone levels than insulin and caused progesterone increase of about 2 orders of magnitude greater than insulin. Removal of the follicle wall eliminated the steroidogenic effects of insulin. Considerable interanimal variation was observed in the ability of insulin to induce oocyte germinal vesicle breakdown (GVBD) in intact follicles. The hypothesis was proposed that differences in endogenous progesterone might explain this variation. To test this hypothesis, an experiment was carried out in which hormone production and follicular sensitivity to insulin were simultaneously determined in follicles obtained from the same animals. Results of the experiment show that the ability of insulin to induce GVBD, as indicated by the effective concentration needed for 50% response (ED50), was strongly correlated with the levels of endogenous progesterone as measured by RIA. The results provide direct evidence that insulin's action on the follicle wall involves steroid production. It was thus concluded that increased endogenous progesterone facilitates GVBD induction by insulin. It is unclear how the two hormones interact to produce an enhanced effect, but interactions at the receptor or postreceptor level may be involved. This follicle system may provide important insights into the mode of action and interaction of these two important hormones.  相似文献
