锌对镉胁迫下马蔺生长、镉积累及生理抗性的影响   总被引:10,自引:0,他引:10  
通过溶液培养研究了不同浓度锌(Zn)对镉(Cd)胁迫下马蔺(Iris lacteavar. chinen-sis)生长、Cd积累及抗氧化酶活性等的影响.结果表明:加入1~100mg.L-1Zn后,Cd胁迫下马蔺地上部Cd含量变化不大,而地下部Cd含量显著增加并呈先增后降的趋势;1mg.L-1Zn处理下马蔺地下部Cd含量最高,比对照显著增加51.4%;Zn浓度高于1mg.L-1后Cd含量均出现不同程度的下降,但仍高于对照.与单独Cd处理(10mg.L-1)相比,添加低浓度Zn(1~10mg.L-1)后,马蔺地上部生物量和叶绿素含量呈增加趋势,丙二醛(MDA)含量降低;在10mg.L-1Zn处理下,叶绿素a、b含量达到峰值,分别增加5.21%和22.27%,MDA含量降低25.46%,表明低浓度Zn缓解了Cd对马蔺的毒害.随Zn浓度的增加,Zn对Cd毒害的缓解作用逐渐降低,当溶液中Zn达到一定浓度(100mg.L-1)时,马蔺毒害加重,其生物量、叶绿素含量均下降,MDA含量显著增加.在试验胁迫浓度范围内超氧化物歧化酶(SOD)、过氧化物酶(POD)活性始终呈增加趋势,表明马蔺在受到Cd胁迫及Cd-Zn共存的条件下SOD和POD起着重要的抗氧化保护作用.  相似文献

通过室内水培试验,研究了不同浓度Pb2+(0、0.25、0.50、1.00和2.00mmol·L-1)胁迫对东方香蒲根和叶中Pb含量、叶绿素含量、丙二醛(MDA)含量、抗氧化酶(SOD、CAT和POD)活性以及亚细胞结构的影响。结果显示:(1)随着外源Pb2+浓度的增加,Pb在香蒲根和叶中的积累量均显著高于对照,且Pb在根中的含量明显高于叶中,并与外源Pb2+浓度呈显著正相关关系。(2)香蒲叶片中的叶绿素a和叶绿素b含量随着外源Pb2+浓度的增加呈先升后降趋势,均在处理浓度为0.50mmol·L-1时达到峰值。(3)胁迫处理叶片的MDA含量与对照相比变化不显著,但根中MDA含量呈显著下降趋势。(4)叶片中SOD活性在1.00mmol·L-1 Pb2+处理时达到峰值,然后下降,但始终高于对照,CAT和POD活性则均低于对照组;根中SOD活性除1.00mmol·L-1 Pb2+处理组外均显著低于对照组,CAT和POD活性分别在0.25和0.50mmol·L-1 Pb2+处理时达到峰值,然后随处理Pb2+浓度升高而下降。(5)电镜观察发现,Pb2+胁迫使香蒲叶细胞中叶绿体被膜破裂,类囊体膨胀、破损;根和叶细胞中的线粒体被膜均破裂、内腔空泡化,细胞核核膜破损、核仁消失、染色质凝集。研究表明,Pb2+胁迫致使东方香蒲根、叶生理代谢失衡,亚细胞结构出现不可逆的损伤,这为从分子水平研究Pb2+作用的具体机理以及香蒲在重金属污染修复中的应用提供了依据。  相似文献

对0、30、150、300、450和600μmol·L-1Pb胁迫条件下狭叶香蒲(TyphaangustifoliaLinn.)种子的萌发特性进行了研究,并分析了0、450、900、1800和2700μmol·L-1Pb胁迫对狭叶香蒲幼苗叶片及根系中部分生理生化指标的影响。结果表明:随Pb浓度提高,狭叶香蒲种子的发芽率、发芽势、发芽指数和活力指数以及下胚轴长度均逐渐下降且低于对照,而其下胚轴长度抑制指数则逐渐增大,但在30μmol·L-1Pb胁迫条件下各项萌发指标均与对照无显著差异。叶片叶绿素a、叶绿素b及总叶绿素含量随Pb浓度提高呈逐渐下降趋势,但在450和900μmol·L-1Pb胁迫条件下与对照无显著差异,而在1800和2700μmol·L-1Pb胁迫条件下显著低于对照。在Pb胁迫条件下叶片和根中SOD活性均显著高于对照但变化趋势不同;随Pb浓度提高,叶片SOD活性呈波动但整体上升的趋势,而根中SOD活性则呈逐渐降低的趋势。叶片和根中POD活性均随Pb浓度提高呈持续上升的趋势,其中,在450和900μmol·L-1Pb胁迫条件下叶片的POD活性低于对照、根的POD活性高于对照,但均与对照无显著差异;而在1800和2700μmol·L-1Pb胁迫条件下叶片和根的POD活性均显著高于对照。在Pb胁迫条件下叶片和根中AsA和MDA含量均高于对照。随Pb浓度提高,叶片的AsA含量总体上逐渐增加但在450和900μmol·L-1Pb胁迫条件下与对照无显著差异;而根的AsA含量则呈先增加后降低的趋势且均与对照差异显著。随Pb浓度提高,叶片的MDA含量先增后降但均与对照无显著差异;而根的MDA含量呈“高-低-高”的波动趋势且仅在450μmol·L-1Pb胁迫条件下与对照差异显著。综合分析结果显示:狭叶香蒲幼苗根系对Pb胁迫的敏感性可能强于叶片;狭叶香蒲种子可在轻度Pb污染水体中萌发和生长;其幼苗对Pb胁迫具有一定的耐性,可用于中度Pb污染水体的修复。  相似文献

采用水培法,研究不同浓度Cu~(2+)胁迫对狭叶香蒲生长及生理特性的影响。结果表明:狭叶香蒲叶宽、株高的增长量及整株干物质累积量在较低浓度Cu~(2+)处理时均未受影响,此后随Cu~(2+)浓度升高显著下降。Cu~(2+)浓度为o~30 mg·L~(-1)时,叶绿素含量显著上升,此后随Cu~(2+)浓度增加其含量显著下降。在Cu~(2+)浓度0~5 mg·L~(-1)范围内根系活力显著上升,此后随Cu~(2+)浓度升高则大幅下降。根系和叶片SOD、POD、CAT、活性随Cu~(2+)浓度增加均呈先显著升高后显著降低的趋势,根系SOD、POD活性在30 mg·L~(-1)时出现最大值,叶片SOD、POD活性在55 mg·L~(-1)时出现最大值,根系和叶片CAT活性均在80 mg·L~(-1)时出现最大值;同一Cu~(2+)浓度下,根系SOD、POD、CAT活性明显高于叶片,说明根系比叶片对Cu~(2+)胁迫反应更敏感。从根系和叶片SOD/POD、SOD/CAT的比值变化上看出,在Cu~(2+)为1 mg·L~(-1)和5 mg·L~(-1)时起主要保护作用的酶是SOD、POD,后来CAT起到主要作用。MDA含量自Cu~(2+)浓度为30 mg·L~(-1)时开始持续上升。说明在Cu~(2+)为30~55 mg·L~(-1)时狭叶香蒲表现为积极的生理响应。  相似文献

主要研究了重金属Cd污染对小麦生长和部分生理特性的影响。结果表明,叶绿素含量在0 2 5mmol·L-1Cd处理浓度时达到峰值,随Cd浓度的增加,含量下降。超氧化物歧化酶(SOD)、过氧化物酶(POD)活性随Cd浓度的增加而增加。丙二醛含量和细胞膜透性同样呈上升趋势。根系活力表现为先升高后下降的变化趋势。Cd胁迫下,小麦的生长受到一定程度的抑制,并随溶液中Cd浓度的增加,抑制加重  相似文献

采用半定量反转录聚合酶链式反应(RT-PCR)技术研究了Cd胁迫对拟南芥(Ara-bidopsis thaliana)幼苗错配修复和增殖细胞核抗原基因表达的影响,并结合幼苗的形态和生理指标,选取Cd胁迫敏感的生物标记物.结果表明:不同浓度(0.25、0.5、1.0 mg·L1)Cd处理对拟南芥幼苗叶片数、地上部鲜质量影响不大;Cd浓度为0.25 mg·L-1时,地上部分可溶性蛋白质含量明显升高(P<0.05),Cd浓度为0.5和1.0 mg·L-1时,可溶性蛋白质含量明显降低(P<0.05);叶绿素含量随着Cd浓度的增加而微弱增加(P>0.05).Cd浓度为0.25 mg·L-1时,以18S rRNA为内参照,PCNA1、PCNA2、MSH2、MSH3、MSH6、MSH7 6个基因均出现了诱导表达,当Cd浓度增加到1.0 mg·L-1时,除了MSH6持续表达诱导及MSH3基因与对照相比表达抑制外,其他基因的表达依然出现诱导,但都低于0.5 mg·L-1Cd处理下的基因表达水平.以上结果表明,基因表达的改变可作为检测Cd污染对植物遗传毒性效应潜在有用的生物标记物.  相似文献

研究了金鱼藻在不同浓度 Hg2 +、Cd2 +以及 Hg2 +、Cd2 +共同胁迫下叶绿素含量、叶绿素 a/b值、可溶性蛋白含量、SOD活性和游离脯氨酸含量等的变化。结果表明 :低浓度 Cd2 + (≤ 2 mg.L- 1)对上述指标有激应性反应 ,低浓度 Hg2 + (≤ 0 .5mg.L- 1)对叶绿素含量、叶绿素 a/b值、可溶性蛋白含量、SOD活性也有激应性反应 ;可以认为这是植物的一种保护性机制。但随着 H2 +、Cd2 +处理浓度增大 ,上述生理指标呈下降趋势 ,表明植物细胞遭受伤害。Hg2 + 、Cd2 + 共同胁迫下上述生理指标下降幅度明显增大 ,表现为协同作用。  相似文献

通过盆栽实验研究了Cd处理对油菜生长和抗氧化酶系统的影响.结果表明,油菜整株鲜重、叶鲜重随Cd胁迫呈显著降低趋势.逐步回归表明,叶鲜重下降导致油菜整株鲜重下降.油菜叶细胞膜渗透性和MDA随Cd胁迫增大而增加.Cd处理浓度为20 mg·kg-1时,叶细胞膜渗透性和MDA分别增加29.68%和15.19%,叶细胞膜渗透性和 MDA呈显著正相关,相关系数为0.823*.Cd处理浓度为5 mg·kg-1时,叶绿素a、b和a+b含量达到峰值,分别比对照高23.97%、33.63%和26.45%;Cd胁迫对类胡萝卜素含量无显著影响.几种色素对Cd敏感顺序为:叶绿素b>叶绿素a>叶绿素a+b>类胡萝卜素;3种抗氧化酶对Cd敏感顺序为:POD>CAT>SOD.各生理指标IC50表明,油菜只适宜种植在Cd含量小于5 mg·kg-1的土壤中.  相似文献

采用水培方法,研究不同浓度镉(Cd)(0、0.5、2.5、5、10和25 mg· L-1)胁迫下金银花的生长特征,分析低浓度Cd对金银花产生的毒物刺激效应.结果表明:低浓度Cd处理(≤5mg·L-1)对金银花最大根长、植株干质量以及叶绿素a、叶绿素b和类胡萝卜素含量均产生显著刺激效应,分别比对照增加13.6%、11.7%、14.0%、10.8%和54.5%;低浓度Cd(≤5mg·L-1)对叶片含水量表现出一定程度的促进作用,但影响不显著;当Cd浓度≥10mg·L-1,对金银花的生长表现出显著的抑制作用.当Cd浓度为25 mg·L-1时,金银花地上部Cd含量达到622.93 mg· kg-1,表明金银花对Cd具有较强的超富集能力.Cd对金银花产生毒物刺激效应的剂量范围为0.5 ~5 mg·L-1.  相似文献

采用砂基栽培,研究重金属镉(Cd)不同胁迫浓度(0、0.5、5、25、50、100和150mg·L-1)和胁迫时间(45和90 d)对红树植物白骨壤幼苗钾钠镁吸收与分配的影响.结果表明:白骨壤各器官对Cd的吸收和累积均表现为随Cd胁迫浓度上升而增加,随胁迫时间增加而增加的趋势;进入植物体内的Cd主要富集在根部,其次是凋落子叶,在150 mg·L-1浓度下分别占累积总量的66.9%和16.3%,这对减少Cd对植物全株的危害具有积极意义.受Cd胁迫影响90 d后,根和茎中Na含量呈上升趋势,叶和子叶中呈下降趋势;根和子叶中K含量呈下降趋势,茎和叶中变化不明显;Cd胁迫90 d后各器官(根、茎、叶和子叶)中Mg含量均低于对照,且与Cd含量呈显著负相关.  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

Fluid collected from hatching eggs of Haemonchus contortus contained a lipase which hydrolysed 2-naphthyl laurate (about 0·7 μmol naphthol freed /h/106 eggs). The fluid also hydrolysed l-leucinamide (about 2·3 μmol leucine freed/h/106 eggs). The fluid when added to normal or heated eggs caused ‘hatching’. ‘Hatching’ also occurred in exsheathing fluid from infective juveniles and in a preparation of pancreatic lipase containing leucine aminopeptidase. A purified mammalian leucine aminopeptidase in combination with several different lipases did not attack egg shells.The ‘spontaneous’ hatching of eggs of H. contortus was strongly inhibited by 1,10-phenanthroline, 10?3M, and this inhibition was reversed by Zn2+. However, the inhibition of ‘hatching’ of eggs in externally applied hatching fluid, or the hydrolysis of leucinamide in hatching fluid was generally less marked.  相似文献
