Synthetic chemical elicitors of plant defense have been touted as a powerful means for sustainable crop protection. Yet, they have never been successfully applied to control insect pests in the field. We developed a high-throughput chemical genetics screening system based on a herbivore-induced linalool synthase promoter fused to a β-glucuronidase (GUS) reporter construct to test synthetic compounds for their potential to induce rice defenses. We identified 2,4-dichlorophenoxyacetic acid (2,4-D), an auxin homolog and widely used herbicide in monocotyledonous crops, as a potent elicitor of rice defenses. Low doses of 2,4-D induced a strong defensive reaction upstream of the jasmonic acid and ethylene pathways, resulting in a marked increase in trypsin proteinase inhibitor activity and volatile production. Induced plants were more resistant to the striped stem borer Chilo suppressalis, but became highly attractive to the brown planthopper Nilaparvata lugens and its main egg parasitoid Anagrus nilaparvatae. In a field experiment, 2,4-D application turned rice plants into living traps for N. lugens by attracting parasitoids. Our findings demonstrate the potential of auxin homologs as defensive signals and show the potential of the herbicide to turn rice into a selective catch crop for an economically important pest.  相似文献

Nicotinic acetylcholine receptors (nAChRs) are major excitatory neurotransmitter receptors in both vertebrates and invertebrates. In insects, nAChRs are the target site for several naturally occurring and synthetic compounds that exhibit potent insecticidal activity. Several compounds isolated from plants are potent agonists or antagonists of nAChRs, suggesting that these may have evolved as a defence mechanism against insects and other herbivores. Nicotine, isolated from the tobacco plant, has insecticidal activity and has been used extensively as a commercial insecticide. Spinosad, a naturally occurring mixture of two macrocyclic lactones isolated from the microorganism Saccharopolyspora spinosa, acts upon nAChRs and has been developed as a commercial insecticide. Since the early 1990s, one of the most widely used and rapidly growing classes of insecticides has been the neonicotinoids. Neonicotinoid insecticides are potent selective agonists of insect nAChRs and are used extensively in both crop protection and animal health applications. As with other classes of insecticides, there is growing evidence for the evolution of resistance to insecticides that act on nAChRs.  相似文献

Herbivore microbial associates can affect diverse interactions between plants and insect herbivores. Some insect symbionts enable herbivores to expand host plant range or to facilitate host plant use by modifying plant physiology. However, little attention has been paid to the role of herbivore-associated microbes in manipulating plant defenses. We have recently shown that Colorado potato beetle secrete the symbiotic bacteria to suppress plant defenses. The bacteria in oral secretions from the beetle hijack defense signaling pathways of host plants and the suppression of induced plant defenses benefits the beetle’s performance. While the defense suppression by the beetle-associated bacteria has been investigated in local damaged leaves, little is known about the effects of the symbiotic bacteria on the manipulation of plant defenses in systemic undamaged leaves. Here, we demonstrate that the symbiotic bacteria suppress plant defenses in both local and systemic tissues when plants are attacked by antibiotic-untreated larvae.  相似文献

Landscape-scale intensification of individual crops and pesticide use that is associated with this intensification is an emerging, environmental problem that is expected to have unequal effects on pests with different lifecycles, host ranges, and dispersal abilities. We investigate if intensification of a single crop in an agroecosystem has a direct effect on insecticide resistance in a specialist insect herbivore. Using a major potato pest, Leptinotarsa decemlineata, we measured imidacloprid (neonicotinoid) resistance in populations across a spatiotemporal crop production gradient where potato production has increased in Michigan and Wisconsin, USA. We found that concurrent estimates of area and temporal frequency of potato production better described patterns of imidacloprid resistance among L. decemlineata populations than general measures of agricultural production (% cropland, landscape diversity). This study defines the effects individual crop rotation patterns can have on specialist herbivore insecticide resistance in an agroecosystem context, and how impacts of intensive production can be estimated with general estimates of insecticide use. Our results provide empirical evidence that variation in the intensity of neonicotinoid-treated potato in an agricultural landscape can have unequal impacts on L. decemlineata insecticide insensitivity, a process that can lead to resistance and locally intensive insecticide use. Our study provides a novel approach applicable in other agricultural systems to estimate impacts of crop rotation, increased pesticide dependence, insecticide resistance, and external costs of pest management practices on ecosystem health.  相似文献

Abstract Numerous studies indicate that target gene silencing by RNA interference (RNAi) could lead to insect death. This phenomenon has been considered as a potential strategy for insect pest control, and it is termed RNAi‐mediated crop protection. However, there are many limitations using RNAi‐based technology for pest control, with the effectiveness target gene selection and reliable double‐strand RNA (dsRNA) delivery being two of the major challenges. With respect to target gene selection, at present, the use of homologous genes and genome‐scale high‐throughput screening are the main strategies adopted by researchers. Once the target gene is identified, dsRNA can be delivered by micro‐injection or by feeding as a dietary component. However, micro‐injection, which is the most common method, can only be used in laboratory experiments. Expression of dsRNAs directed against insect genes in transgenic plants and spraying dsRNA reagents have been shown to induce RNAi effects on target insects. Hence, RNAi‐mediated crop protection has been considered as a potential new‐generation technology for pest control, or as a complementary method of existing pest control strategies; however, further development to improve the efficacy of protection and range of species affected is necessary. In this review, we have summarized current research on RNAi‐based technology for pest insect management. Current progress has proven that RNAi technology has the potential to be a tool for designing a new generation of insect control measures. To accelerate its practical application in crop protection, further study on dsRNA uptake mechanisms based on the knowledge of insect physiology and biochemistry is needed.  相似文献

Loss in seed yield and therefore decrease in plant fitness due to simultaneous attacks by multiple herbivores is not necessarily additive, as demonstrated in evolutionary studies on wild plants. However, it is not clear how this transfers to crop plants that grow in very different conditions compared to wild plants. Nevertheless, loss in crop seed yield caused by any single pest is most often studied in isolation although crop plants are attacked by many pests that can cause substantial yield losses. This is especially important for crops able to compensate and even overcompensate for the damage. We investigated the interactive impacts on crop yield of four insect pests attacking different plant parts at different times during the cropping season. In 15 oilseed rape fields in Sweden, we estimated the damage caused by seed and stem weevils, pollen beetles, and pod midges. Pest pressure varied drastically among fields with very low correlation among pests, allowing us to explore interactive impacts on yield from attacks by multiple species. The plant damage caused by each pest species individually had, as expected, either no, or a negative impact on seed yield and the strongest negative effect was caused by pollen beetles. However, seed yield increased when plant damage caused by both seed and stem weevils was high, presumably due to the joint plant compensatory reaction to insect attack leading to overcompensation. Hence, attacks by several pests can change the impact on yield of individual pest species. Economic thresholds based on single species, on which pest management decisions currently rely, may therefore result in economically suboptimal choices being made and unnecessary excessive use of insecticides.  相似文献

Hyperaccumulation of Ni by plants is hypothesized to function as an elemental defense against herbivores and pathogens. Laboratory experiments have documented toxic effects to herbivores consuming high-Ni plant tissues, but this paper reports the first experiment to examine the defensive effectiveness of Ni hyperaccumulation under field conditions. The experiment was conducted at an ultramafic soil site naturally inhabited by the Ni hyperaccumulator Streptanthus polygaloides (Brassicaceae). Experimental treatments examined the response of herbivores to hyperaccumulated Ni, using exclosure and insecticide treatments to divide herbivores into groups based primarily upon herbivore size. Three soils (Ni-amended greenhouse soil, unamended greenhouse soil, ultramafic soil), three exclosure treatments (exclosure, control exclosure, no exclosure), and a systemic insecticide treatment were combined in a fractional factorial experimental design. Streptanthus polygaloides plants were grown in a greenhouse for 2 mo, transplanted into the field by inserting potted plants into holes dug on the experimental site, and periodically examined for herbivore damage during a 41-d period. Initial surveys showed greater amounts of insect damage to plants with low tissue Ni levels, confirming the defensive effect of Ni against some insect herbivores, but large herbivores (probably vertebrates) later consumed entire plants regardless of plant Ni status. We concluded that Ni was not an effective defense against these large herbivores, probably because their diets mix high-Ni S. polygaloides foliage with that of associated non-hyperaccumulating species. We suggest that such dietary dilution is one mechanism whereby some herbivores can circumvent elemental plant defenses.  相似文献

This paper demonstrates the necessity to consider plants as an essential and interactive component of biological control practices. Plants not only possess direct chemical and morphological defenses against herbivores but also benefit from indirect defenses provided by parasitoids and predators, which use herbivores as hosts or prey. Plants play an active role in the interplay between entomophagous arthropods and herbivores and actually mediate many of the interactions, thereby influencing the intensity of protection received. Herein, we review how plant attributes influence natural enemy efficiency by providing shelter, mediating host/prey accessibility, providing host/prey finding cues, influencing host/prey suitability, mediating host/prey availability, and providing supplemental food sources for natural enemies. In light of this crucial role, we suggest ways of manipulating morphological and chemical attributes of crop plants for a more sustainable and balanced control of insect pests in agro-ecosystems.  相似文献

The ‘evolution of increased competitive ability’ (EICA) hypothesis is an extension of optimal defense theory and predicts that reduced pressure from insect herbivores in the introduced range results in evolution of reduced defenses in invading plant populations, allowing greater allocation of resources to competitive traits such as growth rate and reproduction. The EICA hypothesis considered levels of defensive chemistry to be fixed within a particular genotype. In this paper, we propose that if herbivory is reduced in the introduced range, but chemical defenses are inducible in response to herbivory, evolution of reduced defenses and any associated increase competitive ability should not occur. Rather, mean induced and constitutive levels of induced defenses should be similar in introduced and native ranges, but the variance about mean induced levels should be greater in the introduced range. This is predicted because induced levels will occur less frequently in the introduced range where herbivory is reduced, thereby insulating these levels from the stabilizing selection expected in the native range where induced levels occur more frequently. We conducted a preliminary study to examine this by comparing constitutive and induced concentrations of total pyrrolizidine alkaloids (PAs) from native (European) and introduced (western North America) populations of Cynoglossum officinale L. The mean constitutive and induced concentrations of PAs did not differ between continents, but the variability of the induced concentrations was significantly greater for plants from the introduced range. Although our study with C. officinale is provisional due to a small sample size, it supports our predictions for evolution of inducible defenses in introduced ranges where herbivore pressure is reduced. Most chemical defenses in plants have been found to be inducible, so similar patterns may occur widely. If so, this weakens the generality of EICA’s predictions concerning chemical defenses. The effects of inducible defenses should be considered in cross-continent comparisons of other invasive plant species.  相似文献

Increasingly, genetically modified crops are being developed to express multiple 'stacked' traits for different types of transgenes, for example, herbicide resistance, insect resistance, crop quality and tolerance to environmental stresses. The release of crops that express multiple traits could result in ecological changes in weedy environments if feral crop plants or hybrids formed with compatible weeds results in more competitive plants outside of agriculture. To examine the effects of combining transgenes, we developed a stacked line of canola (Brassica napus L.) from a segregating F(2) population that expresses both transgenic glyphosate resistance (CP4 EPSPS) and lepidopteran insect resistance (Cry1Ac). Fitness-associated traits were evaluated between this stacked genotype and five other Brassica genotypes in constructed mesocosm plant communities exposed to insect herbivores (Plutella xylostella L.) or glyphosate-drift. Vegetative biomass, seed production and relative fecundity were all reduced in stacked trait plants when compared with non-transgenic plants in control treatments, indicating potential costs of expressing multiple transgenes without selection pressure. Although costs of the transgenes were offset by selective treatment, the stacked genotype continued to produce fewer seeds than either single transgenic line. However, the increase in fitness of the stacked genotype under selective pressure contributed to an increased number of seeds within the mesocosm community carrying unselected, hitchhiking transgenes. These results demonstrate that the stacking of these transgenes in canola results in fitness costs and benefits that are dependent on the type and strength of selection pressure, and could also contribute to changes in plant communities through hitchhiking of unselected traits.  相似文献

Calcium oxalate is the most abundant insoluble mineral found in plants and its crystals have been reported in more than 200 plant families. In the barrel medic Medicago truncatula Gaertn., these crystals accumulate predominantly in a sheath surrounding secondary veins of leaves. Mutants of M. truncatula with decreased levels of calcium oxalate crystals were used to assess the defensive role of this mineral against insects. Caterpillar larvae of the beet armyworm Spodoptera exigua Hübner show a clear feeding preference for tissue from calcium oxalate-defective (cod) mutant lines cod5 and cod6 in choice test comparisons with wild-type M. truncatula. Compared to their performance on mutant lines, larvae feeding on wild-type plants with abundant calcium oxalate crystals suffer significantly reduced growth and increased mortality. Induction of wound-responsive genes appears to be normal in cod5 and cod6, indicating that these lines are not deficient in induced insect defenses. Electron micrographs of insect mouthparts indicate that the prismatic crystals in M. truncatula leaves act as physical abrasives during feeding. Food utilization measurements show that, after consumption, calcium oxalate also interferes with the conversion of plant material into insect biomass during digestion. In contrast to their detrimental effects on a chewing insect, calcium oxalate crystals do not negatively affect the performance of the pea aphid Acyrthosiphon pisum Harris, a sap-feeding insect with piercing-sucking mouthparts. The results confirm a long-held hypothesis for the defensive function of these crystals and point to the potential value of genes controlling crystal formation and localization in crop plants.  相似文献

1. When considering intercropping as a strategy to reduce pest oviposition, knowledge about the insect’s oviposition behaviour is very important. Physiological effects on the insect because of difficulties in finding a suitable oviposition site may also be important. 2. In the present study, the effects that delays in access to host plants have on lifetime fecundity on diamondback moth and leek moth were examined. The ability to postpone egg laying, fecundity and lifetime oviposition are discussed in relation to intercrop/cover crop as a strategy to reduce oviposition on crop plants. 3. When faced with host plant deprivation, the diamondback moth is relatively more dependent upon host plant stimuli for the onset of egg production. By contrast, leek moth is able to postpone egg production for a longer time. There even appeared to be a tendency for leek moth females to extend their lifetime when faced with host plant deprivation. 4. We conclude that leek moths have the ability to postpone production of eggs and lay them later in life when finally encountering host plants after a period of host plant deprivation. Therefore, the use of intercropping as a strategy to reduce oviposition is questionable. For such an insect, use of a trap crop might be a better option because the female will lay her eggs in the trap crop and not get the opportunity to lay them later in life when finally encountering crop plants.  相似文献

Winter squash is a vital agricultural commodity worldwide. In the Northeastern United States, the primary insect pest is the striped cucumber beetle, Acalymma vittatum F. Using a Blue Hubbard squash (Cucurbita maxima Duchesne) perimeter trap crop system can reduce insecticide use by >90% in butternut squash (C. moschata Poir), the primary winter squash grown in this region. Despite the savings in insecticide costs, growers may be reluctant to give up field space for a perimeter crop of Blue Hubbard squash, which comprises only 5% of the winter squash market in New England as compared with 19% for buttercup squash. Finding a more marketable trap crop would lower the barrier for adoption of this system. We tested eight varieties of three species of cucurbits for attractiveness to beetles relative to Blue Hubbard and butternut squash, and chose buttercup squash as the most promising replacement. We compared the effect of a buttercup border, Blue Hubbard border, or control (no border) on beetle numbers, herbivory, insecticide use, pollination, and pollen limitation in the main crop. We found that buttercup squash performed equally well as Blue Hubbard as a trap crop, with 97% reduction in total insecticide use compared with control fields. Honey bees (Apis mellifera L.) and squash bees (Peponapis pruinosa Say) were the predominant pollinators, and border treatments did not affect visitation. Hand pollination did not increase reproduction or yield, indicating that natural pollination was sufficient for full yield. This study confirms the effectiveness of perimeter trap crop systems and offers growers a more marketable trap crop for managing cucumber beetle damage.  相似文献

Hokkanen & Pimentel (1984) proposed a novel approach for the selection of biocontrol agents. They advocated the selection of agents from a relative of the weed plant rather than from the target weed species itself. The new relationship that such agents would have with the weed would be characterised by a relative lack of homeostasis compared with the old herbivore-plant associations traditionally used in weed biocontrol, and would consequently be more effective. There are few examples to support these contentions because of the traditional use of old herbivore-plant associations in weed biocontrol. In the present study, herbivore-plant associations in agricultural situations, which are analogous to agent-weed associations, are examined to assess the potential of new associations for weed biocontrol. The herbivores on 14 introduced crop plants which have salient similarities to the major weeds in the south-western Cape were surveyed: (a) 68% of the 188 insect and mite herbivores are indigenous species in new associations with these host plants, and (b) of the five most damaging pests on each of 13 of the crop plants, 53% are in new associations with the plants. Of the 40 most important agricultural pests in South Africa, 58% are in new associations, confirming these results. About 50% of the insect and mite herbivores in new associations with their host plants are oligophagous, indicating that new associations are not necessarily characterised by polyphagy and hence unsafe for use in weed biocontrol. We conclude that new associations between herbivore species and host plants have strong potential in weed biocontrol because (a) their frequency in agriculture indicates that they can easily be established in weed biocontrol situations, (b) they are as damaging as old associations and (c) they are not necessarily unsafe as regards host specificity. We therefore advocate the use of both the classical approach and that of Hokkanen & Pimentel (1984). Our support for Hokkanen & Pimentel (1984) is, however, based on evidence and rationale clearly different to theirs, and we provide novel guidelines, which can be routinely and practically applied in the selection of agents for weed biocontrol.  相似文献

Transgenic resistance to insects has been demonstrated in plants expressing insecticidal genes such as δ -endotoxins from Bacillus thuringiensis (Bt), protease inhibitors, enzymes, secondary plant metabolites, and plant lectins. While transgenic plants with introduced Bt genes have been deployed in several crops on a global scale, the alternative genes have received considerably less attention. The protease inhibitor and lectin genes largely affect insect growth and development and, in most instances, do not result in insect mortality. The effective concentrations of these proteins are much greater than the Bt toxin proteins. Therefore, the potential of some of the alternative genes can only be realized by deploying them in combination with conventional host plant resistance and Bt genes. Genes conferring resistance to insects can also be deployed as multilines or synthetic varieties. Initial indications from deployment of transgenics with insect resistance in diverse cropping systems in USA, Canada, Argentina, China, India, Australia, and South Africa suggest that single transgene products in standard cultivar backgrounds are not a recipe for sustainable pest management. Instead, a much more complex approach may be needed, one which may involve deployment of a combination of different transgenes in different backgrounds. Under diverse climatic conditions and cropping systems of tropics, the success in the utilization of transgenics for pest management may involve decentralized national breeding programs and several small-scale seed companies. While several transgenic crops with insecticidal genes have been introduced in the temperate regions, very little has been done to use this technology for improving crop productivity in the harsh environments of the tropics, where the need for increasing food production is most urgent. There is a need to develop appropriate strategies for deployment of transgenics for pest management, keeping in view the pest spectrum involved, and the effects on nontarget organisms in the ecosystem.  相似文献

Evolutionary interactions among insect herbivores and plant chemical defenses have generated systems where plant compounds have opposing fitness consequences for host plants, depending on attack by various insect herbivores. This interplay complicates understanding of fitness costs and benefits of plant chemical defenses. We are studying the role of the glucosinolate-myrosinase chemical defense system in protecting Arabidopsis thaliana from specialist and generalist insect herbivory. We used two Arabidopsis recombinant inbred populations in which we had previously mapped QTL controlling variation in the glucosinolate-myrosinase system. In this study we mapped QTL controlling resistance to specialist (Plutella xylostella) and generalist (Trichoplusia ni) herbivores. We identified a number of QTL that are specific to one herbivore or the other, as well as a single QTL that controls resistance to both insects. Comparison of QTL for herbivory, glucosinolates, and myrosinase showed that T. ni herbivory is strongly deterred by higher glucosinolate levels, faster breakdown rates, and specific chemical structures. In contrast, P. xylostella herbivory is uncorrelated with variation in the glucosinolate-myrosinase system. This agrees with evolutionary theory stating that specialist insects may overcome host plant chemical defenses, whereas generalists will be sensitive to these same defenses.  相似文献

硅不是植物必需营养元素,但硅在提高植物对一系列非生物和生物胁迫的抗性方面都具有重要作用。综述了硅对植物抗虫性的影响及其机制。在多数植物中,增施硅肥可增强其抗虫性;所增强的抗性与硅肥种类和施用方式之间存在关系。植物组织中沉积的硅可增加其硬度和耐磨度,降低植物可消化性,从而增强植物组成性防御,包括延缓昆虫生长发育、降低繁殖力、减轻植物受害程度;植物体内的硅含量以及硅沉积的位点和排列方式影响组成性防御作用的强度。此外,硅可以调节植物诱导性防御,包括直接防御和间接防御,直接防御涉及增加有毒物质含量、产生局部过敏反应或系统获得抗性、产生有毒化合物和防御蛋白,从而延缓昆虫发育;间接防御主要通过释放挥发性化合物吸引植食性昆虫的捕食性和寄生性天敌而导致植食性昆虫种群下降。  相似文献
