A strain of Escherichia coli with the fol gene deleted and a kan gene inserted in its place was created for use in cloning and isolation of mutant dihydrofolate reductase. Southern blot analysis and dihydrofolate reductase enzyme assays confirmed the delta fol::kan genotype. A thyA mutation accompanied the fol deletion and is required for survival of a dihydrofolate reductase-deficient strain.  相似文献

The dihydrofolate reductase (fol) gene in Escherichia coli has been deleted and replaced by a selectable marker. Verification of the delta fol::kan strain has been accomplished using genetic and biochemical criteria, including Southern analysis of the chromosomal DNA. The delta fol::kan mutation is stable in E. coli K549 [thyA polA12 (Ts)] and can be successfully transduced to other E. coli strains providing they have mutations in their thymidylate synthetase (thyA) genes. A preliminary investigation of the relationship between fol and thyA gene expression suggests that a Fol- cell (i.e., a dihydrofolate reductase deficiency phenotype) is not viable unless thymidylate synthetase activity is concurrently eliminated. This observation indicates that either the nonproductive accumulation of dihydrofolate or the depletion of tetrahydrofolate cofactor pools is lethal in a Fol- ThyA+ strain. Strains containing the thyA delta fol::kan lesions require the presence of Fol end products for growth, and these lesions typically increase the doubling time of the strain by a factor of 2.5 in rich medium.  相似文献

Saccharomyces cerevisiae is so far the only organism where a knock-out mutant in the gene encoding GTP cyclohydrolase I (FOL2) has been obtained. GTP cyclohydrolase I controls the de novo biosynthetic pathway of tetrahydrobiopterin and folic acid. Since deletion of yeast FOL2 leads to a recessive auxotrophy for folinic acid, we used a yeast fol2Delta mutant for an in vivo functional assay of heterologous GTP cyclohydrolases I. We show that the GTP cyclohydrolase I, encoded either by the E. coli folE gene or by the human cDNA, complements the yeast fol2Delta mutation by restoring folate prototrophy. Furthermore the folE-3x allele of the E. coli gene, carrying three base substitutions, failed to complement the yeast fol2Delta defect. This allele behaved as a negative semidominant to the wild type folE and, when overexpressed, completely abolished complementation of fol2Delta by folE. Thus, the yeast fol2 null mutant is a suitable system to characterize mutations in genes encoding GTP cyclohydrolase I.  相似文献

The gene order and orientation in the leu-pyrA region of the Salmonella typhimurium linkage map was established by phage P22-mediated transductions. The gene order, in counterclockwise orientation, is leuO-leuA-leuB-leuC-leuD-ara-fol-pyrA. The fol locus is co-transducible with either the ara and leu loci or the pyrA locus, whereas no co-transduction for the ara and pyrA loci can be found.  相似文献

An O-specific polysaccharide, containing 6-deoxy-L-talose (6dTal), N-acetyl-D-fucosamine (FucNAc), 3-amino-3,6-dideoxy-D-glucose with an unidentified N-acyl substituent (Qui3NR), and O-acetyl groups, was obtained on mild acid degradation of a Pseudomonas fluorescens strain 361 lipopolysaccharide. On the basis of O-deacetylation, acid hydrolysis, methylation, selective solvolysis with anhydrous hydrogen fluoride, and 13C NMR analysis, the polysaccharide is built up of trisaccharide repeating units of the following structure: (Formula: see text).  相似文献

A branch and bound algorithm is proposed for the two-dimensional protein folding problem in the HP lattice model. In this algorithm, the benefit of each possible location of hydrophobic monomers is evaluated and only promising nodes are kept for further branching at each level. The proposed algorithm is compared with other well-known methods for 10 benchmark sequences with lengths ranging from 20 to 100 monomers. The results indicate that our method is a very efficient and promising tool for the protein folding problem.  相似文献

The mitochondrial outer membrane is not just a barrier but a site of regulation of mitochondrial function. The VDAC family of proteins are the major pathways for metabolite flux through the outer membrane. These can be regulated in a variety of ways and the integration of these regulatory inputs allows mitochondrial metabolism to be adjusted to changing cellular conditions. This includes total blockage of the flux of anionic metabolites leading to permeabilization of the outer membrane to small proteins followed by apoptotic cell death.  相似文献

本文在“广义Fuzzy运算的综合评判”的基础上,并结合牧草产量预报实践,提出了一个经验“模型V”,简称“综合决策模型”。 该模型不仅具有计算简捷,容易为群众掌握的优点,而且还扩大了“广义模糊运算的综合评判”的应用范围,为了弥补其不足,还运用数学统计法建立如下的预报模型: (?)=-6.8223-0.4871R_1 0.3033R_2 0.0791R_3 2.8231R_4 0.6361R_5 (7)同时,还用周期方差分析来预报牧草产量。最后,综合运用了上述3种方法来预报牧草产量,提高了预报的精确度,效果更好。本文还举例说明了计算方法,并且讨论了同时运用几种方法进行产量预报的必要性和优越性。  相似文献

When platelet rich plasma is exposed to N-ethylmaleimide, a ten fold increase in measurable prostaglandin E synthesis occurs. This effect is almost completely abolished within 2 hours of ingestion of 600 mg of aspirin by human volunteers. Recovery of this platelet function is slow for the first two days, returning sharply to normal over the next six days and plateauing approximately 8 days following initial removal from aspirin. It is suggested from these studies that platelet prostaglandin E production following NEM may be a useful test of platelet function.  相似文献

The principles of enzyme chemistry, mechanism of action and inhibitor design are being applied to proteomics by the development of activity-based probes. This approach suggests a potentially broad method for interrogating enzyme family members, both known and unknown, in cells and proteomic fractions without the need for individual assay development and isolation. The serine hydrolases and cysteine proteases have provided the proofs of concept for activity-based proteomics, and other studies are rapidly following. The result will be a proteomics technology of great value to drug discovery and development.  相似文献

In this pathway, the Oxa1/Alb3/YidC proteins are believed to function as membrane insertases that play an important role in the membrane protein biogenesis of respiratory and energy transduction proteins. Total kininogen in plasma of Freund's adjuvant treated rats increased 20-fold 7 days following the injection. Analysis of the kininogens demonstrated that increases in T-kininogen was the major reason for the rise in kininogen. High molecular weight and low molecular weight kininogens showed little or no change. The increase in T-kininogen paralleled the inflammatory condition. Anti-inflammatory agents which reduced paw swelling also reduced plasma T-kininogen levels. Unidentified peaks on HPLC of kinin following plasma treatment by trypsin were shown to be oligopeptides containing T-kinin (Ile-serbradykinin). The relationship of T-kininogen to the inflammatory response is discussed.  相似文献

本文研究了渤海南部半滑鳎的食性及其摄食的季节性变化,胃食物分析结果表明,半滑舌鳎为底栖生物食性鱼类,以虾类,蟹类,双壳类及部分中下层小型鱼类为主要食物,兼食一些多毛类,头足类,腹足类,棘皮动物及海葵,半滑舌鳎终年摄食,其摄食强度的周年变化不大,半滑舌鳎常年捕食鲜明鼓虾,日本鼓虾,口虾蛄,隆线强蟹及泥古隆背蟹等。夏季加强捕食小刀蛏,凸壳肌蛤等双壳类,降春季外还捕食一些以虾虎鱼为主的中下层小型鱼类。  相似文献
