Summary The fine structure of ovarian steroid-producing cells of the domestic fowl as seen after administration of steroids is described. Diaethylstilboestrol, estradiol and hydroxyprogesterone were given as intramuscular injections for a 28-days period. The main cytoplasmic changes of the steroid-producing cells were an increase in the number and the size of lipid droplets due to a possible expansion of the endoplasmic reticulum and a confluence of the smooth endoplasmic reticulum and the lipid droplets. The mitochondria became paler, markedly reduced in size and number, and contained less cristae. The Golgi apparatus became smaller and tended to disappear. The most prominent nuclear changes were irregular outline of the nuclear membrane, shrinkage of the nucleus, formation of pseudo-inclusions, decreased number of nuclear bodies with a tendency to vacuolization and small nucleoli. These alterations have never been demonstrated in ovarian steroid-producing cells before, and they are consistent with atrophic changes observed in steroid-producing cells in other organs. The present study substantiates the view that the thecal gland must be concerned with endocrine function. The possible functions of the different cell types of the thecal gland are discussed. 相似文献

Summary The post-natal evolution of thecal gland in the domestic fowl has been explored, and a hypothesis for this development is proposed. It is suggested that the surface epithelium of the ovary forms crypts and submicroscopic clefts (ingrowing cords) which contribute with epithelial elements required for the regeneration of some of the ovarian elements, including the thecal glands. Complexes of these ingrowing epithelial cells are forming small islets which, after hormonal stimulation, are transformed into well defined structures, the thecal glands, with a specific function, viz. steroid bio-synthesis. 相似文献

Summary The fine structure of the interstitial gland of the rat ovary was studied at estrus and on Days 4, 6, 10, 14 and 18 of pregnancy. At estrus, ovarian interstitial cells have small nuclei with dense irregular clumps of heterochromatin. Mitochondria are small and rod-shaped and have predominantely lamellar cristae. Numerous osmiophilic lipid droplets are present. At Days 4 and 6, nuclear heterochromatin is reduced, and nucleoli are larger and complex. Mitochondria are enlarged and often bizarre-shaped and have tubular cristae. Golgi and smooth endoplasmic reticulum are more conspicuous. At Day 10, prominent ultrastructural features include nuclei with conspicuous heterochromatin, smaller mitochondria with both lamellar and tubular cristae, numerous ribosomes and lipid droplets with decreased osmiophilia. At Days 14 and 18, nuclei have increased heterochromatin, mitochondria are small and have lamellated cristae and an increase in the size and number of lipid droplets occurs. These observations suggest that steroidogenic activity of interstitial cells is highest during the first half of pregnancy and regresses during the last half. It is suggested that the interstitial gland is an important ovarian source of pregnancy hormone(s) during the first half of gestation and that LH may modulate steroidogenic activity in this ovarian component. 相似文献

Summary The fine structure of the interstitial gland of the ovary was studied in hypophysectomized rats and in hypophysectomized rats after denervation of the ovary or stimulation of the ovarian plexus. Hypophysectomized rats were used to eliminate gonadotropic influences on interstitial cells. In hypophysectomized rats, there was a large amount of intercellular space and cells had irregularly shaped nuclei and a large nuclear-cytoplasmic ratio. Prominent cytoplasmic features included small mitochondria with an electron-dense matrix, rough and smooth endoplasmic reticulum, polysomes and large osmiophilic lipid droplets. Interstitial cells from stimulated ovaries had reduced intercellular space and a reduced nuclear-cytoplasmic ratio. Mitochondria had tubular cristae; smooth endoplasmic reticulum-surrounded lipid droplets, and large polysomes were present. After section of the ovarian plexus, intercellular space was increased and filopodia were numerous. Cytoplasmic features included mitochondria with a dense matrix and indistinct cristae, large electronlucent lipid droplets, and variously sized multivesicular structures. These observations suggest that stimulation of the ovarian plexus in hypophysectomized rats causes regressed cells to assume the fine structural features of active steroidogenic cells. In contrast, interruption of the ovarian nerve supply causes a qualitative and quantitative increase in ultrastructural features characteristic of regressed steroidogenic cells. These responses of interstitial gland cells to denervation and stimulation provide morphological evidence for a functional role for the adrenergic nerves to this ovarian compartment. 相似文献

Summary The granulosa cells of the ovarian follicle of the rat and the domestic fowl have been studied with the light and electron microscope. The nuclei of the granulosa cells were irregular with indentations and large in proportion to the cytoplasm of the cell. The mitochondria had a dense, dark matrix with only few cristac. The Golgi apparatus was moderately developed, located towards the oocyte in a juxtanuclear position. The endoplasmic reticulum was rather sparse. Lipid droplets were only occasionally encountered. Microtubules were regularly observed. The functions of the granulosa cells are discussed. Compared with the steroid-producing cells of the theca interna of the same follicles, the granulosa cells primarily are the nursing cells for the growing oocyte and mainly have the characteristics of protein forming cells. 相似文献

Summary The ultrastructure of the lutein cells in the mink throughout pregnancy and the regression periodpost partum is described. To correlate the fine structure with the changes in the peripheral plasma progesterone levels, the concentrations of progesterone were measured by a rapid competitive protein-binding assay.Even during the delay period (e.g. as long the plasma progesterone levels remain at the basal level, <8ng/ml), the lutein cells in the mink exhibit structural criteria of functional activity. However, the increase in progesterone secretion is accompanied by some morphological transformations, characterized by the presence of more and more small dense homogenous bodies in the cytoplasm, which become irregular and scalloped during the stage with maximum release of progesterone. At this stage the agranular endoplasmic reticulum is often cisternal or vesicular.During the decline of the progesterone levels, typical and moderate electron-dense lipid droplets are found increasingly more within the lutein cells. The expanded agranular ER is now more sparse, while the granular ER becomes more pronounced, often forming parallel arrays. During this phase the mitochondria become elongated, dumb-bell, or cup shaped. After parturition the corpora lutea consist of cells in various stages of degeneration. At day 14post partum only a few lutein cells are still identifiable.Evidently the observed morphological changes take place in the lutein cells during the life span of corpora lutea. This feature lends further support to the concept that the mink lutein cells are steroid-producing cells and furthermore, that the corpora lutea may be the main sites of gestagen production during pregnancy. 相似文献

Summary The ultrastructure of the polymorphic vesicular component of the ultimobranchial gland of the domestic fowl (Gallus gallus domesticus) has been described in detail, together with the structure of the cell strands interconnecting the vesicles and the parathyroid nodules lying within the ultimobranchial stroma. The vesicles frequently appear to arise from the nodules by way of the cell strands. The strands show a structure of their component cells intermediate between that of the parathyroid and the vesicular cells, although the position at which the strand changes from an essentially parathyroid structure to an essentially vesicular structure is very variable. The degree and kind of secretory activity within different cell types has been described. A review of the structure of ultimobranchial glands throughout the vertebrates shows that similar tissue with a similar secretory potential has been observed in all vertebrate classes, suggesting a functional significance for this part of the gland. 相似文献

Résumé Chez les Silphes et en particulier chez Phosphuga atrata, la glande de la spermathèque présente une structure particulière liée à la présence d'une intima cuticulaire tapissant la lumière de la glande. Elle comporte trois types cellulaires: les cellules sécrétrices, les cellules de l'épithélium sous-cuticulaire et les cellules-manchons. Les cellules sécrétrices de grande taille contiennent une invagination de la membrane cytoplasmique formant une «vacuole» extracellulaire bordée de microvillosités. Dans cette vacuole plonge l'extrémité, différenciée en ampoule poreuse, d'un canalicule de nature cuticulaire, qui véhicule la sécrétion jusqu'à la lumière de la glande. Le canalicule est élaboré par une cellule-manchon qui l'accompagne sur toute sa longueur sauf à son extrémité intravacuolaire.Ce type de glande, qui se retrouve chez de nombreux Insectes, y assurant des fonctions diverses (sécrétion odorifique, sécrétion de défense, sécrétion spermale, etc.), est susceptible de nombreuses variations.
Ultrastructure of the spermathecal accessory gland in Phosphuga atrata L. (coleoptera: silphidae)
Summary The spermathecal accessory gland in the female of Phosphuga atrata (Silphidae), exhibits a special structure which is due to the presence of a cuticular intima lining the lumen. The wall of the gland shows three cellular types: the secretory cells, the epithelial cells and the ductule carrying cells. Each large secretory cell contains a cavity formed by an invagination of the cytoplasmic membrane and lined by many microvilli. The secretory cell is connected with a cuticular ductule ending in the cavity of the glandular cell, in a porous organelle. This ductule, which carries the secretory material to the lumen, is surrounded by the ductule carrying cell.This type of integumentary gland is very common in insects, where it assumes various functions (attraction, defense, conservation of sperm, etc.) and its morphology varies considerably.相似文献

Summary The structure of follicular layer of growing and atretic follicles in the ovary of the domestic goose, was studied by electron microscopy. In small follicles, the wall is lined with a narrow layer of tightly packed small, cuboidal cells separated from the thecal tissue by the basal lamina. During growth, they transform into tall, columnar cells arranged in a single row. The cells display several peculiar ultrastructural features. First, annulate lamellae are commonly observed. Second, cytoplasmic dense-cored granules accumulate in close association with fenestrated cisternae and networks of tubuli derived from the RER. They consist of spheres and strands of amorphous substance of unknown origin. Third, the cells contain many transosomes, a unique organelle of the avian follicle cell consisting of a dense plaque associated with ribosome-like particles. The mature forms of transosomes are located at the tips of lateral and apical cell projections, while bodies thought to be their precursors, are found in the apical cytoplasm. In follicles larger than 8 mm in diameter, most of the transosomes and their precursors have disappeared. Follicular atresia occurs in all of the size-classes of follicles investigated. A loss of transosomes (in follicles up to 8 mm in diameter) and an accumulation of lipid droplets are the first atretic events detectable by electron microscopy. Morphologic features, including deep nuclear indentations, accumulation of lipid droplets frequently encireled by membrane whorls, dilation and disintegration of RER cisterns, swelling of mitochondria and accumulation of dense irregular masses of unknown origin in the cytoplasm, are taken as evidence for advanced degradation. We conclude that necrosis is the dominant type of cell death of the follicular cells during atresia. However, a small fraction of cells, characterized by dark condensed cytoplasm, seems to die by apoptosis. 相似文献

Summary The position, structure and function of the valves within the lateral sinus of the medical leech, Hirudo medicinalis, are described on the basis of vital, light- and electron microscopy. In this species the valvular apparatus consists of multiple elongated fir cone-shaped fibrous villi surrounding the orifices of the latero-lateral and latero-dorsal vessel like a tentacular crest. Each villus is covered by a thin sheet of a continuous endothelium. The valves prevent the backflow of hemolymph during systolic contraction of the lateral sinus.The endothelium contains many small mitochondria and polyribosomes in the perinuclear cytoplasm and it develops deep projections into the underlying connective tissue. Each of these consists of a multilayered system of closely interwoven thin endothelial membranes. The endothelium is anchored to its basement membrane by means of a great number of poorly defined hemidesmosomes. The fibrous tissue of the villi consists mainly of a homogeneous vitreous matrix in which few cellular components and very fine filaments are dispersed. Close to the endothelium this matrix appears to be condensed to form a multilayered framework made out of a basement membrane-like material.Though the valves themselves are devoid of muscle cells, those situated at their base and thus belonging to the vascular wall proper, display some specific morphological features: in particular the nuclei of these cells show a distinct fibrous lamina. Moreover, these muscle cells seem to be innervated only by one type of axon, containing both small, lucent synaptic vesicles as well as some of the dense-core variety.These findings are compared with data from earlier works and are discussed in relation to the hemodynamic functions of this valvular apparatus. 相似文献

Summary The ultrastructural characteristics of the ovarian medulla of the newly hatched white leghorn chick were studied in control animals and compared with chicks that were treated with human chorionic gonadotropin during embryonic development. The ovarian medulla was mainly occupied by epithelial cells which formed cords or islets surrounded by a basal lamina. Within this epithelial compartment, steroidogenic cells, poorly differentiated cells and a lacunary system could be recognized. When chicks were treated with human chorionic gonadotropin, steroidogenic cells became discernible; there was an increment in the amount of cytoplasm and the area of mitochondria. Poorly differentiated cells exhibited signs of stimulation, and transitional images suggested the transformation of these cells into steroidogenic cells. The epithelial cells of the lacunar system also displayed stimulated cytoplasmic organelles. Evidence was supplied suggesting that relatively undifferentiated cells persist in the ovarian medulla until hatching and can develop into steroidogenic cells under gonadotropic stimulation. 相似文献

The role of the interstitial cells of Cajal (ICC) associated with the myenteric plexus (ICC-MP) as regulators of the motility
of the colonic external muscle remains unclear. Ultrastructural studies of myenteric interstitial cells are lacking in human
colon. We therefore characterized the distinctive ultrastructure of these cells in the myenteric region of the colon by transmission
electron microscopy of the region between the main muscle layers in all parts of the colon in unaffected areas of resected
specimens from nine adult human patients. ICC-MP were similar in various colonic regions and had myoid features such as scattered
caveolae, prominent intermediate filaments, and cytoplasmic dense bodies. We found characteristic dense membrane-associated
bands with a patchy basal lamina, invaginating cellular protrusions (peg and socket junctions) between ICC and between ICC
and muscle cells, and close contacts (<100 nm) between ICC and nerves. No gap junctions were observed. Fibroblast-like cells
(FLC) were abundant showing well-developed secretory organelles, including coated vesicles, but lacked prominent intermediate
filaments and caveolae. FLC had a patchy basal lamina, and peg and socket junctions were observed between them. Macrophage-like
cells frequently occurred in close apposition with FLC and, more seldomly, with ICC-MP. The ultrastructure of ICC and FLC
in the myenteric region of the human colon thus differs characteristically, but significant overlaps in the ultrastructure
between ICC and FLC might complicate any interpretation in pathological ultrastructural studies of the human colonic muscle
layer.

Summary The ultrastructure of the secretory cells of the adenohypophysis of juvenile sockeye salmon was investigated. Pituitary glands were collected from immature fish transferred experimentally to sea water and subsequently returned to fresh water. The rostral pars distalis contained three cell types: ACTH cells, prolactin cells, and non-secretory cells. The prolactin and non-secretory cells were joined together in the form of follicles by desmosomes and they both had cilia and microvilli projecting into the follicle lumen. Various follicular structures such as lumen, multivesicular structures, and peripheral basement membrane are discussed as possible sites of prolactin cell granule release. The columnar ACTH cells were found at the junction of the rostral pars distalis and the neurohypophysis. The cytoplasmic granules in these cells were characteristically separated from their limiting membrane by a clear space. Multivesicular structures were also found in association with this cell type. The caudal pars distalis also contained three cell types: one acidophil (putative somatotrop) and two basophils (putative thyrotrops and gonadotrops), all of which were similar to those described in adult fish. The pars intermedia contained only one cell type. They appeared to be active cells and were characterized by containing membrane-bounded granules similar to those found in the ACTH cells. Changes in ambient salinity had no apparent effect on any cell type described.The work was supported by a grant in aid of research from the National Research Council of Canada. The aim of this ultrastructural study was to examine the human detrusor for interstitial cells of Cajal (ICC)-like cells (ICC-L)
by conventional transmission electron microscopy (TEM) and immuno-transmission electron microscopy (I-TEM) with antibodies
directed towards CD117 and CD34. Two main types of interstitial cells were identified by TEM: ICC-L and fibroblast-like cells
(FLC). ICC-L were bipolar with slender (0.04 μm) flattened dendritic-like processes, frequently forming a branching labyrinth
network. Caveolae and short membrane-associated dense bands were present. Mitochondria, rough endoplasmic reticulum and Golgi
apparatus were observed in the cell somata and cytoplasmic processes. Intermediate filaments were abundant but no thick filaments
were found. ICC-L were interconnected by close appositions, gap junctions and peg-and-socket junctions (PSJ) but no specialised
contacts to smooth muscle or nerves were apparent. FLC were characterised by abundant rough endoplasmic reticulum but no caveolae
or membrane-associated dense bands were observed; gap junctions and PSJ were absent and intermediate filaments were rare.
By I-TEM, CD34 gold immunolabelling was present in long cytoplasmic processes corresponding to ICC-L between muscle fascicles
but CD117 gold immunolabelling was negative. Thus, ICC-like cells are present in the human detrusor. They are CD34-immunoreactive
and have a myoid ultrastructure clearly distinguishable from fibroblast-like cells. ICC-L may be analogous to interstitial

Summary
Hyrax possess luteal cells with many ultrastructural features similar to those described in other mammalian species. The process of luteinization is gradual. Cells with increasing amounts of smooth endoplasmic reticulum become predominant in the developing gland. The luteal cells of Procavia and Heterohyrax show many ultrastructural similarities. In both species the corpus luteum persists throughout pregnancy. 相似文献

Summary In intact male rats after TRH administration for 7 and 14 days, TSH cells showed similar morphological changes to those observed after thyroidectomy. These changes were paralleled by small numerical increases in TSH cell counts. After 34 days of TRH treatment, however, most of the TSH cells had a normal appearance and the number of TSH cells also had returned to normal. TRH treatment for 7, 14 and 34 days caused morphological changes in Prolactin cells similar to those obtained after a suckling stimulus. In the three groups these changes were also paralleled by small numerical increases in Prolactin cell counts. The cell replication after TRH for 7 and 14 days, as measured by incorporation of tritiated thymidine to obtain a labeling index, was slightly but significantly increased.This work was supported by grants MA-552 and MT-2701 from the Medical Research Council of Canada. Summary The interstitial cells associated with the myenteric plexus of the rabbit colon were studied by scanning and transmission electron microscopy. It was demonstrated that the interstitial cells were stellate or fusiform in shape and located over the ganglia, over nerve bundles and between muscle cells. They were characterized by many slender processes, and resemble fibroblasts. No basal lamina was observed between the interstitial cells and muscle cells. It was concluded that structural features of the interstitial cells are distinctly different from those of neurons, Schwann cells, or of smooth muscle cells, while they show clear similarities to those of fibroblasts. By scanning electron microscopy the shapes and the relations of these cells could be demonstrated in great detail. 相似文献
