Potted soybean (Glycine max (L.) Merr.) plants were grown in P-fertilized (+P) or low-P soil (-P), or colonized in -P soil by one of the arbuscular mycorrhizal (AM) fungi Glomus etunicatum (Ge), Glomus mosseae (Gm), or Gigaspora rosea (Gr). Treatment effects on plant development, on the soil microflora, and on the status of water-stable soil aggregates (WSA) were evaluated for all 5 treatments or for the 3 AM treatments alone. Dry weights of the AM plants, as a group, were half-way between the dry weights of the +P and -P plants, but within the AM group, Gm plants had the highest pod dry weights and pod/stem and root/stem ratios and the lowest specific root lengths, while Ge plants had high stem dry weights and were highly nodulated. High reproductive development and coarse roots in the Gm plants were associated with the most extensive growth of AM soil hyphae (km pot-1: Gm, 20; Gr, 12; Ge, 8), while nodulation was inversely related with AM-colonized root length. The soils colonized by AM fungi had significantly higher levels of WSA (size classes 1 to 2 and 2 to 4 mm), and within the larger size class, Gm soils had the highest percentage of WSA. Proliferation (plate counts) of Gram positive (G+) and Gram negative (G-) bacteria, Arthrobacter sp. (G+), and Pseudomonas sp. (G-) was greatest in the -P soils, but the bacterial populations of the +P and the AM soils were generally not significantly different. There were, however, differences among the AM treatments, where Gm soils had the lowest G- bacterial populations, while Ge soils had the highest populations of both G+ and G- bacteria. Correlations between plant and soil traits indicated that interactions within the plant-soil system were mediated by the AM fungi.  相似文献

Roots and mycorrhizal fungi may not associate with soil bacteria randomly, but rather in a hierarchical structure of mutual preferences. Elucidation of such structures would facilitate the management of the soil biota to enhance the stability of the plant-soil system. We conducted an experiment utilizing two isolates of soil bacteria to determine their persistence in distinct mycorrhizal regions of the root zone, and their effects on general rhizosphere populations of fluorescent pseudomonads (FP). Split-root sorghum (Sorghum bicolor L.) plants were grown in four-compartment containers, constructed so that the soils in individual compartments held either (1) roots colonized by the arbuscular-mycorrhizal (AM) fungus Glomus mosseae (M), (2) nonAM roots only (R), (3) hyphae of G. mosseae (H), or (4) no mycorrhizal structures (S). The soils were inoculated (107 cells g-1 dry soil) with antibiotic-resistant (rifampicin, rif; streptomycin, sm) strains of the soil bacteria, Alcaligenes eutrophus (rifr50) or Arthrobacter globiformis (smr250), or were left uninoculated as control. A. eutrophus had been isolated from a specific source (hyphosphere soil of G. mosseae), and A. globiformis from mycorrhizosphere soils of two AM fungi. After 10 wk of growth, the presence of A. eutrophus was barely detectable (<10 cfu g-1 dry soil) in nonAM (R and S) soils, but persisted well (104 cfu g-1 dry soil) in AM (H and M) soils. Numbers of A. globiformis were more evenly distributed between all soils, but were highest in the presence of AM roots (M soil). There were varied bacterial effects on root and AM-hyphal development: A. eutrophus decreased hyphal length in H soil, while A. globiformis stimulated root length in M soil. The two bacterial inoculants did not affect numbers of FP in H, R, and M soils, but the AM status of the soils did: the numbers of FP increased in the order M>R>H>S. There was a positive correlation of FP numbers with both bacterial inoculants in M and H soils. Numbers of FP changed with root or hyphal lengths, an effect that was related to changes in the numbers of the inoculated bacteria. The results indicate that the hyphosphere-specific A. eutrophus depended on the presence of G. mosseae, but that the nonspecific A. globiformis did not. The mycorrhizal status of soils may selectively influence persistence of bacterial inoculants as well as affecting the numbers of other native bacteria.  相似文献

The influence of arbuscular mycorrhizal (AM) fungi on aggregate stability of a semi-arid Indian vertisol was studied in a pot experiment in which Sorghum bicolor (L.) was grown as test plant for 10 weeks. Pasteurized soil inoculated with AM fungi was studied with pasteurized and unpasteurized soils as references. A part of the soil in each pot was placed in nylon mesh bags to separate effects of roots and hyphae. The sorghum plants were planted outside the mesh bags which permitted AM hyphae to enter while excluding roots. Aggregate stability of the soil was determined by wet-sieving and turbidimetric measurements. Development of the AM fungi was quantified as colonized root length and external hyphal length. Soil exposed to growth of roots and hyphae (outside mesh bags) showed aggregates with larger geometric mean diameter (GMD) in pasteurized soil inoculated with AM fungi than in pasteurized uninoculated soil. There was no significant difference in GMD of the inoculated, pasteurized soil and the unpasteurized soil. No significant effects of inoculation or plant growth were found in pasteurized soil exposed to hyphal growth only (inside the mesh bags). However, the unpasteurized soil had significantly higher GMD than the pasteurized soil, irrespective of plants and inoculum. Turbidimetric measurements of soil exposed to roots and hyphae (outside mesh bags) showed the highest aggregate stability for the inoculated pasteurized soil. These results demonstrate that AM fungi contribute to the stabilization of soil aggregates in a vertisol, and that the effect is significant after only one growing season. The effect was associated with both AM hyphae and the stimulation of root growth by AM fungi. The contribution from plant roots and AM hyphae to aggregate stability of different size fractions is discussed. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献

The effects of tomato and bean rhizospheres on hyphal spreading of the arbuscular mycorrhizal (AM) fungus Glomus mosseae were studied using a soil compartment system in combination with hydrophobic polytetrafluorethylene (PTFE) membranes. Both the nylon screen and the PTFE membrane were freely permeable to hyphae but not to roots. Furthermore, the hydrophobic PTFE membrane seemed to be a barrier to the flux of soil solutions containing root exudates. The results show that water soluble exudates of tomato and bean roots greatly stimulate hyphal growth in the soil compartment system used. Moreover, water soluble root exudates of bean exert a clear attractional effect on AM hyphal growth.  相似文献

How soil carbon balance will be affected by plant–mycorrhizal interactions under future climate scenarios remains a significant unknown in our ability to forecast ecosystem carbon storage and fluxes. We examined the effects of soil temperature (14, 20, 26 °C) on the structure and extent of a multispecies community of arbuscular mycorrhizal (AM) fungi associated with Plantago lanceolata. To isolate fungi from roots, we used a mesh‐divided pot system with separate hyphal compartments near and away from the plant. A 13C pulse label was then used to trace the flow of recently fixed photosynthate from plants into belowground pools and respiration. Temperature significantly altered the structure and allocation of the AM hyphal network, with a switch from more vesicles (storage) in cooled soils to more extensive extraradical hyphal networks (growth) in warmed soils. As soil temperature increased, we also observed an increase in the speed at which plant photosynthate was transferred to and respired by roots and AM fungi coupled with an increase in the amount of carbon respired per unit hyphal length. These differences were largely independent of plant size and rates of photosynthesis. In a warmer world, we would therefore expect more carbon losses to the atmosphere from AM fungal respiration, which are unlikely to be balanced by increased growth of AM fungal hyphae.  相似文献

The relative importance of roots and AM-fungi on soil physical processes was investigated by controlling the presence of roots and AM fungi in pot experiments using a mycorrhiza-defective tomato mutant and a wild-type tomato (Solanum lycopersicum L.). Root-Zone and Bulk Soil sections were established by splitting pots into two lengthwise halves using a nylon mesh that contained roots whilst allowing the free movement of fungal hyphae. Post-incubation microbial populations and fungal biomass were measured and related to soil stability, pore structure and water repellency. Unplanted controls consistently had the least fungal biomass, fatty acids, water-stable aggregates (WSA) and water repellency. Wild-type-planted treatments had significantly more WSA than mycorrhiza-defective treatments (P?<?0.01). Fluctuations in water content induced by transpiration caused significant changes in soil pore structure, measured using high-resolution X-Ray computer tomography. Porosity and mean pore size increased in soil aggregates from planted treatments, which had larger more heterogeneous pores than those in the unplanted soils. AM fungi accentuated soil stability. However, changes were not linked to repellency and fungal biomass. The presence of plants, regardless of AM fungi, appears to have the greatest impact on increasing soil stability.

The effect of 46 bacterial strains isolated from tilled and non-tilled soils collected at 3 localities on the growth of intraradical hyphae of the arbuscular mycorrhizal (AM) fungusGlomus claroideum was demonstrated. A larger number of stimulatory bacterial isolates was obtained from tilled soils, but the bacteria showing the strongest stimulation of hyphal growth were isolated from a soil that had not been cultivated. Isolates obtained from hyphae of AM fungi showed no substantial stimulatory effects, but produced more uniform effects on hyphal growth than the isolates of bacteria obtained from soil. Bacterial cenoses present in 3 different soils differ significantly in their effects on AM fungi.

Mycorrhizas are ubiquitous plant–fungus mutualists in terrestrial ecosystems and play important roles in plant resource capture and nutrient cycling. Sporadic evidence suggests that anthropogenic nitrogen (N) input may impact the development and the functioning of arbuscular mycorrhizal (AM) fungi, potentially altering host plant growth and soil carbon (C) dynamics. In this study, we examined how mineral N inputs affected mycorrhizal mediation of plant N acquisition and residue decomposition in a microcosm system. Each microcosm unit was separated into HOST and TEST compartments by a replaceable mesh screen that either prevented or allowed AM fungal hyphae but not plant roots to grow into the TEST compartments. Wild oat (Avena fatua L.) was planted in the HOST compartments that had been inoculated with either a single species of AM fungus, Glomus etunicatum, or a mixture of AM fungi including G. etunicatum. Mycorrhizal contributions to plant N acquisition and residue decomposition were directly assessed by introducing a mineral 15N tracer and 13C‐rich residues of a C4 plant to the TEST compartments. Results from 15N tracer measurements showed that AM fungal hyphae directly transported N from the TEST soil to the host plant. Compared with the control with no penetration of AM fungal hyphae, AM hyphal penetration led to a 125% increase in biomass 15N of host plants and a 20% reduction in extractable inorganic N in the TEST soil. Mineral N inputs to the HOST compartments (equivalent to 5.0 g N m?2 yr?1) increased oat biomass and total root length colonized by mycorrhizal fungi by 189% and 285%, respectively, as compared with the no‐N control. Mineral N inputs to the HOST plants also reduced extractable inorganic N and particulate residue C proportion by 58% and 12%, respectively, in the corresponding TEST soils as compared to the no‐N control, by stimulating AM fungal growth and activities. The species mixture of mycorrhizal fungi was more effective in facilitating N transport and residue decomposition than the single AM species. These findings indicate that low‐level mineral N inputs may significantly enhance nutrient cycling and plant resource capture in terrestrial ecosystems via stimulation of root growth, mycorrhizal functioning, and residue decomposition. The long‐term effects of these observed alterations on soil C dynamics remain to be investigated.  相似文献

We examined the effects of atmospheric carbon dioxide (CO2) enrichment on belowground carbon (C) pools and arbuscular mycorrhizal (AM) fungi in a chaparral community in southern California. Chambers enclosing intact mesocosms dominated by Adenostoma fasciculatum were exposed for 3.5 years to CO2 levels ranging from 250 to 750 ppm. Pools of total C in bulk soil and in water-stable aggregates (WSA) increased 1.5- and threefold, respectively, between the 250- and 650-ppm treatments. In addition, the abundance of live AM hyphae and spores rose markedly over the same range of CO2, and the community composition shifted toward dominance by the AM genera Scutellospora and Acaulospora. Net ecosystem exchange of C with the atmosphere declined with CO2 treatment. It appears that under CO2 enrichment, extra C was added to the soil via AM fungi. Moreover, AM fungi were predominant in WSA and may shunt C into these aggregates versus bulk soil. Alternatively, C may be retained longer within WSA than within bulk soil. We note that differences between the soil fractions may act as a potential feedback on C cycling between the soil and atmosphere.  相似文献

An investigation was carried out to test whether the mechanism of increased zinc (Zn) uptake by mycorrhizal plants is similar to that of increased phosphorus (P) acquisition. Maize (Zea mays L.) was grown in pots containing sterilised calcareous soil either inoculated with a mycorrhizal fungus Glomus mosseae (Nicol. and Gerd.) Gerdemann and Trappe or with a mixture of mycorrhizal fungi, or remaining non-inoculated as non-mycorrhizal control. The pots had three compartments, a central one for root growth and two outer ones for hyphal growth. The compartmentalization was done using a 30-m nylon net. The root compartment received low or high levels of P (50 or 100 mg kg–1 soil) in combination with low or high levels of P and micronutrients (2 or 10 mg kg–1 Fe, Zn and Cu) in the hyphal compartments.Mycorrhizal fungus inoculation did not influence shoot dry weight, but reduced root dry weight when low P levels were supplied to the root compartment. Irrespective of the P levels in the root compartment, shoots and roots of mycorrhizal plants had on average 95 and 115% higher P concentrations, and 164 and 22% higher Zn concentrations, respectively, compared to non-mycorrhizal plants. These higher concentrations could be attributed to a substantial translocation of P and Zn from hyphal compartments to the plant via the mycorrhizal hyphae. Mycorrhizal inoculation also enhanced copper concentration in roots (135%) but not in shoots. In contrast, manganese (Mn) concentrations in shoots and roots of mycorrhizal plants were distinctly lower, especially in plants inoculated with the mixture of mycorrhizal fungi. The results demonstrate that VA mycorrhizal hyphae uptake and translocation to the host is an important component of increased acquisition of P and Zn by mycorrhizal plants. The minimal hyphae contribution (delivery by the hyphae from the outer compartments) to the total plant acquisition ranged from 13 to 20% for P and from 16 to 25% for Zn.  相似文献

White clover (Trifolium repens L.) plants were grown in a calcareous soil in pots with three compartments, a central one for root growth and two outer ones for growth of vesicular-arbuscular (VA) mycorrhizal (Glomus mosseae [Nicol. & Gerd.] Gerdemann & Trappe) hyphae (hyphal compartments). Phosphorus (P) was applied at three levels (0, 20 and 50 mg kg−1 soil) in the outer compartments in mycorrhizal treatments. Root and shoot dry weight were increased in mycorrhizal plants with hyphal access to outer compartments. Growth of the mycorrhizal hyphae in the outer compartments was not significantly affected by variation in P level in these compartments. However, both concentration and amount of P in roots and shoots sharply increased with increasing P supply in the outer (hyphal) compartments. With increasing P levels the calculated delivery of P by the hyphae from the outer compartments increased from 34% to 90% of total P uptake. Hyphal access to the outer compartments also significantly increased both concentration and quantity of Cu in the plants. The calculated delivery of Cu by the hyphae from the outer compartments ranged from 53% to 62% of total Cu uptake, irrespective of the P levels and the amounts of P taken up and transported by the hyphae. However, the distribution of Cu over roots and shoots was largely dependent on P levels. With increase in P level in the outer compartments the calculated hyphal contribution to the total amount of Cu in the shoots increased from 12% to 58%, but decreased in the roots from 75% to 46%. In conclusion, uptake and transport by VA-mycorrhizal hyphae may contribute substantially not only to P nutrition, but also to Cu nutrition of the host.  相似文献

The origins and composition of soil organic matter (SOM) are still largely uncertain. Arbuscular mycorrhizal fungi (AMF) are recognized as indirect contributors through their influence on soil aggregation, plant physiology, and plant community composition. Here we present evidence that AMF can also make large, direct contributions to SOM. Glomalin, a recently discovered glycoprotein produced by AMF hyphae, was detected in tropical soils in concentrations of over 60 mg cm–3. Along a chronosequence of soils spanning ages from 300 to 4.1 Mio years, a pattern of glomalin concentrations is consistent with the hypothesis that this protein accumulates in soil. Carbon dating of glomalin indicated turnover at time scales of several years to decades, much longer than the turnover of AMF hyphae (which is assumed to be on the order of days to weeks). This suggests that contributions of mycorrhizae to soil carbon storage based on hyphal biomass in soil and roots may be an underestimate. The amount of C and N in glomalin represented a sizeable amount (ca. 4–5%) of total soil C and N in the oldest soils. Our results thus indicate that microbial (fungal) carbon that is not derived from above- or below-ground litter can make a significant contribution to soil carbon and nitrogen pools and can far exceed the contributions of soil microbial biomass (ranging from 0.08 to 0.2% of total C for the oldest soils).  相似文献

Radiocaesium enters the food chain when plants absorb it from soil, in a process that is strongly dependent on soil properties and plant and microbial species. Among the microbial species, arbuscular mycorrhizal (AM) fungi are obligate symbionts that colonize the root cortex of many plants and develop an extraradical mycelial (ERM) network that ramifies in the soil. Despite the well-known involvement of this ERM network in mineral nutrition and uptake of some heavy metals, only limited data are available on its role in radiocaesium transport in plants. We used root-organ culture to demonstrate that the ERM of the AM fungus Glomus lamellosum can take up, possibly accumulate and unambiguously translocate radiocaesium from a 137Cs-labelled synthetic root-free compartment to a root compartment and within the roots. The accumulation of 137Cs by hyphae in the root-free compartment may be explained by sequestration in the hyphae or by a bottleneck effect resulting from a limited number of hyphae crossing the partition between the two compartments. Uptake and translocation resulted from the incorporation of 137Cs into the fungal hyphae, as no 137Cs was detected in mycorrhizal roots treated with formaldehyde. The importance of the translocation process was indicated by the correlation between 137Cs measured in the roots and the total hyphal length connecting the roots with the labelled compartment. 137Cs may be translocated via a tubular vacuolar system or by cytoplasmic streaming per se.  相似文献

An experiment was set up to investigate the role of arbuscular mycorrhiza (AM) in utilization of P from organic matter during mineralization in soil. Cucumber (Cucumis sativus L.) inoculated with one of two AM fungi or left uninoculated were grown for 30 days in cross-shaped PVC pots. One of two horizontal compartments contained 100 g soil (quartz sand: clay loam, 1:1) with 0.5 g ground clover leaves labelled with32P. The labelled soil received microbial inoculum without AM fungi to ensure mineralization of the added organic matter. The labelling compartment was separated from a central root compartment by either 37 m or 700 m nylon mesh giving only hyphae or both roots and hyphae, respectively, access to the labelled soil. The recovery of32P from the hyphal compartment was 5.5 and 8.6% for plants colonized withGlomus sp. andG. caledonium, respectively, but only 0.6 % for the non-mycorrhizal controls. Interfungal differences were not related to root colonization or hyphal length densities, which were lowest forG. caledonium. Both fungi depleted the labelled soil of NaHCO3-extractable P and32P compared to controls. A 15–25% recovery of32P by roots was not enhanced in the presence of mycorrhizas, probably due to high root densities in the labelled soil. The experiment confirms that AM fungi differ in P uptake characteristics, and that mycorrhizal hyphae can intercept some P immobilization by other microorganisms and P-sorbing clay minerals.  相似文献

Penicillium pinophilum was isolated from the soil in a commercial strawberry field. The strain readily formed arbuscular mycorrhizae (AM) with the roots of strawberry 'Zoji' (Fragaria x ananassa Duch. CV.) when plants were inoculated with either fresh cultured hyphae or root/soil mixtures. Fresh hyphae, however, resulted in higher amounts of colonization than root/soil inoculum. Compared with uninoculated strawberries, inoculation increased plant dry weight by 31%, as well as nitrogen content (47%), phosphorus content (57%), and photosynthetic rate (71%). AM inoculation also shortened the blossom and ripening date by 3 and 4 days, respectively. This is the first report of a P. pinophilum strain resulting in mycorrhiza with strawberry roots. The significant advantages of this strain are that it is easy to culture and inoculation of plants results in significant growth benefits that may be useful in strawberry production.  相似文献

Experimental systems for measuring nutrient transport by arbuscular mycorrhizal (AM) fungi in soil are described. The systems generally include two soil compartments that are separated by fine nylon mesh. Both roots and root-external hyphae grow in one compartment, but only hyphae are fine enough to grow through the mesh into the other compartment. Application of tracer isotopes to the soil of this hyphal compartment can be used to measure nutrient uptake by plants via AM fungal hyphae. Use of compartmented systems is discussed with particular reference to phosphorus, which is the mineral nutrient transported in the largest quantity by AM fungi. Laboratory and field applications of the compartmentation methodology are presented with emphasis on the functioning of native AM fungal communities. Advantages and limitations of the method are considered and future important research directions are discussed in this context. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献
