The presence of muscarinic receptors in sheep and rat pineals was detected by binding of [3H]quinuclidinyl benzilate ([3H]QNB), a potent and specific muscarinic antagonist. [3H]QNB binding to sheep pineal membrane resuspensions was saturable and reversible, with a rate constant for association at 37°C of 6×108M?1min?1 and a rate constant for dissociation of 1×10?2min?1. Kinetic and saturation experiments yielded an equilibrium dissociation constant of 13–18 pM and a concentration of binding sites equivalent to 1.1 pmol/g of original wet weight. This is only about 5% of the level of β-adrenergic receptors. Competition by a variety of cholinergic drugs confirmed the muscarinic nature of the binding sites. Experiments in rats failed to detect a significant decrease in pineal [3H]QNB binding following bilateral superior cervical ganglionectomy, suggesting that the binding sites are not localized exclusively on sympathetic terminals.  相似文献

The interaction of alkylguanidines and decahydrohistrionicotoxin with the membrane-bound and solubilized muscarinic acetylcholine receptor (mAcChR) from porcine atria was described. Alkylguanidines with alkyl chain lengths from one to ten carbons displaced l-[3H]quinuclidinyl benzilate (l-[3H]QNB) competitively from a single class of sites for the membrane-bound mAcChR. From a plot of ?ln Ki versus alkyl carbon chain number, a value of ?(473 ± 30) cal/mol was estimated as the energetic contribution per methylene group to the total binding energy. The binding of alkylguanidines to the digitonin/cholate solubilized mAcChR was complex in nature resulting in titration curves that did not obey the law of mass action for simple competitive inhibition at higher alkyl carbon numbers and a sigmoidal plot of ?ln Ki versus carbon number. Decahydrohistrionicotoxin bound in a competitive manner versus l-[3H]QNB to both the membrane-bound (Ki = (6.9 ± 1.4) × 10?6 M) and the solubilized (Ki = (1.5 ± 0.3) × 10?5 M) preparations.  相似文献

Human erythrocyte ghosts contain a small population of muscarinic cholinergic receptors, as evidenced by their high affinity binding of radiolabeled quinuclinidinyl benzilate ([3H]QNB). The apparent KD is 1.3 × 10?9 M and the receptor sites are saturated at a QNB concentration of 5 nM. The number of sites is 23 fmoles/mg membrane protein. The pharmacological profile of the specific binding is similar to that of neural membranes. The binding is not stereoselective for the d and 1 isomers of QNB, a situation which prevails in the muscarinic receptors of another peripheral cholinergic system, the rat iris, but not in the central nervous system.  相似文献

Membranes of neuron-like NG108-15 hybrid cells bind [3H]quinuclidinyl benzilate (QNB) with high affinity and specificity. Greater than 90% of total [3H]QNB binding is to sites having the pharmacological specificity of muscarinic acetylcholine receptors. Three significant features characterize the interaction of ligands with these sites: (1) Specific binding of [3H]QNB at equilibrium follows a simple adsorption isotherm with an apparent KD of 1 × 10?10 M; (2) Rates of [3H]QNB association and dissociation are biphasic and, as the binding reaction proceeds, the fraction of readily dissociable [3H]QNB decreases; (3) Competition against [3H]QNB for specific binding sites by antagonists gives a slope of 1 when analyzed on Hill plots, but competition for binding sites by agonists gives a slope of less than 1. A simple two-step model for activation is proposed to account for these features.  相似文献

The true KDs of [3H] (?) QNB binding to muscarinic receptors were found to be 4.13 and 6.43 × 10?11 M in pancreas of 21 day fetal and adult rats. The competition curves of specific [3H] (?) QNB bound by two antagonists have shown that the affinity of these drugs did not change with age with Hill coefficients near unity. However, with the agonist carbamylcholine as the competitive drug, a more flat curve was obtained with a Hill coefficient below unity. At least two populations of carbamylcholine binding sites were found with different KDs: a KH around 7 × 10?7 M and a KL around 3 × 10?5 M. These two populations were present during all the developmental periods studied. The ED50 of bethanechol stimulated amylase secretion did not change within the age limit studied (from 11 to 365 days). The high affinity sites for carbamylcholine would seem to be the receptor implicated in the physiological response of the pancreas.  相似文献

Male rats were treated for 10 days with the organophosphorus insecticide, acetylcholinesterase inhibitor, O,O-diethyl S-[2-(ethylthio)ethyl]phosphorodithioate (disulfoton, 2 mg/kg/day by gavage). At the end of the treatment, binding of [3H]quinuclidinyl benzilate ([3H]QNB) to cholinergic muscarinic receptors and cholinesterase (ChE) activity were assayed in the pancreas. Functional activity of pancreatic muscarinic receptor was investigated by determining carbachol-stimulated secretion of α-amylase in vitro. ChE activity and [3H]QNB binding were significantly decreased in the pancreas from disulfoton-treated rats. The alteration of [3H]QNB binding was due to a decrease in muscarinic receptor density with no change in the affinity. Basal secretion of amylase from pancreas in vitro was not altered, but carbachol-stimulated secretion was decreased. The effect appeared to be specific since pancreozymin was able to induce the same amylase release from pancreases of control and treated rats. The results suggest that repeated exposures to sublethal doses of an organophosphorus insecticide lead to a biochemical and functional alteration of cholinergic muscarinic receptors in the pancreas.  相似文献

Specific thyroxine (T4) binding was identified in bovine mammary cytosol preparations. Binding specificity of 3,5,3′ triiodothyronine (T3) with respect to T4 was less than 1%. The halftime of T4 binding and displacement at 4°C was 1 and 20 minutes, respectively. Scatchard analyses demonstrated the presence of two T4 binding components with dissociation constants of 3.61 × 10?10 and 7.73 × 10?8M. The high affinity binding component had a molecular weight of ~ 100,000, a T4 binding capacity of 5.05 × 10?12moles/mg protein, and was destroyed by boiling or protease treatment. High-affinity, low capacity T4 binding was not found in bovine serum and was unique to mammary tissue.  相似文献

We have studied muscarinic acetylcholine (ACh) receptors in intact atrial and ventricular heart cells dissociated from 8-day chick embryos and maintained in sparse cell cultures. Two specific antagonists, [3H]quinuclidinyl benzilate (QNB) and [3H]N-methyl scopolamine (NMS), bind to surface sites with affinity (Kds ? 40 and 400 pM, respectively). The concentration of [3H]QNB sites in ventricular cell cultures (460 fmole/mg protein) was comparable to the concentration of sites in atrial cultures (420 fmole/mg protein). The same result was obtained with [3H]NMS. Autoradiography following incubation in saturating concentrations of [3H]QNB shows that nearly all of the atrial and ventricular myocytes were labeled and that the distribution of grains over individual cells was uniform. The mean binding site density was 109/μm2 for atrial cells 117/μm2 for ventricular cells. In contrast to the antagonist binding results, microelectrode recordings from individual myocytes or from small clusters of cells showed that many more atrial myocytes (89%) were sensitive to 10?4M carbachol than were ventricular myocytes (26%). Saline extract of embryonic brain tissue added to the culture medium did not alter the number or distribution of ligand binding sites but it produced a 2.6-fold increase in the number of carbachol-sensitive ventricular cells.  相似文献

Abstract

The pharmacological characteristics of muscarinic receptor (mAChR) subtypes in canine left ventricular membranes (LVM) were determined using [3H]quinuclidinyl benzilate ([3H]QNB) and [3H] N-methyl scopolamine ([3H]NMS) as ligands. Binding of [3H]QNB and [3H]NMS was saturable with respect to the radioligand concentrations. Analysis of binding isotherms by Scatchard plot showed that [3H]QNB and [3H] NMS bound to an apparently homogeneous population of mAChRs in LVM, with KD values of 390 ± 100 and 285 ± 34 pM and Bmax values of 240 ± 20 and 133 ± 9 fmol/mg protein, (n=6), respectively. The Hill coefficients for [3H]QNB and [3H]NMS binding were 0.95 ± 0.02 and 0.99 ± 0.01, respectively. Based on the competitive inhibition of [3H] ligand binding, atropine and NMS as well as the selective M1 antagonist PZ revealed no selectivity for these mAChRs. PZ competed with [3H]QNB or [3H]NMS for a single binding site with a Ki value of 0.23 ± 0.03 μM and 0.62 ± 0.10 μM, (n = 6), respectively, which is close to the values of M2 or M3 receptors. The data indicate that the M1 receptor subtype did not exist in canine LVM. Competition of [3H] ligand binding with selective M2 antagonists, AF-DX 116 and methoctramine and the selective M3 antagonists, 4-DAMP and hexahydrosiladifenidol, gave a best fit for a two-binding site model. The inhibition of carbachol-mediated phosphoinositide hydrolysis by PZ, AF-DX 116 and 4-DAMP, generated an affinity profile for this response also dissimilar to that described for the classical cardiac M2 response. Although no other muscarinic receptor mRNA has been detected in this tissue, these data suggest the presence of a second population of muscarinic sites, which may signify an M2 receptor diversity.  相似文献

[3H] quinuclidinyl benzilate (QNB), a specific muscarinic antagonist, was utilized to identify muscarinic cholinergic receptors on dispersed anterior pituitary cells. Scatchard analysis of [3H] QNB binding to receptors departs from linearity with upward concavity. A high affinity binding site having a dissociation constant (Kd) of 1.5 nM was observed when the [3H] QNB concentration was varied from 0.15 to 20 nM. A low affinity binding site (Kd 20 nM) was observed when [3H] QNB concentration was above 20 nM. Using 10 nM [3H] QNB for binding, the second order association rate constant (k1) of 0.064 nM?1 min?1 and first order dissociation rate constant (k2) of 0.078 min?1(T12 8 min) were observed. k2/k1 = Kd of 1.22 nM is in good agreement with Kd = 1.5 nM from equilibrium data. Muscarinic cholinergic receptor antagonists, atropine and scopolamine, and agonist oxtoremorine potently competed with [3H] QNB binding. A nicotinic cholinergic receptor agonist was 50 times less potent as a competitor of [3H] QNB binding than the muscarinic agonist.  相似文献

The effect of fluoride ion on the binding of the specific muscarinic agonist ligand [3H]c is methyldioxolane ([3H]CD) to the mouse cardiac muscarinic receptor was investigated. Utilizing equilibrium ligand binding experiments, sodium fluoride (10mM) was shown to decrease [3H]CD binding, measured at a concentration of 2 nM, by 52%. Studies with several different ions demonstrated that the reduction in [3H]CD binding was a specific effect of fluoride. This fluoride modulation was selective for agonist binding, as no effect of fluoride on the binding of the muscarinic antagonist [3H](?) quinuclidinyl benzilate (QNB) was observed.  相似文献

The saturable and specific high-affinity uptake of [3H]serotonin ([3H]5HT) (5 × 10?8 M) was studied in slices from the hippocampus, parietal cortex, septum-preoptic area, and hypothalamus of male 2, 6, 12 and 24–32 month old C57BL/6N mice. Hippocampal [3H]5-HT uptake showed a significant biphasic relationship to age, with lower values in the 2 and 24–32 month old mice compared to 6 month old mice. No significant age effects were seen in the other regions, or in [3H]norepinephrine high-affinity uptake in the hippocampus.Studies of the high-affinity uptake mechanism in synaptosomal preparations were made in a subgroup of 12 and 24 month old mice. A micro-assay using a tissue-harvester measured high-affinity uptake on 8–30 μl of the P2 suspension (crude-synaptosomal preparation). The high-affinity uptake was linear for 4 min at 37°C and inhibited in both the adult and aged tissue by 10?5 M cold 5-HT (83 and 78% respectively), 10?5 M fluoxetine (85 and 82% respectively) and 10?3 M NaCN (57 and 57% respectively). Kinetic analysis of the [3H]5HT high-affinity uptake in the hippocampus (3 min, 37°C) revealed the same apparent Km for serotonin at both ages (6.7 x 10?8 M), but a 44% decrease in Vmax in the aged hippocampal synaptosomal high-affinity uptake compared to adults (120 vs 215 pmol of 5-HT/g-tissue/3 min).These results are discussed in relationship to the reported age effects on the intrinsic neurons of the hippocampus.  相似文献

[3H]Nimodipine binding was studied in isolated myocytes from rat heart and in partially purified sarcolemma, sarcoplasmic reticulum and mitochondrial fractions from dog heart. In isolated myocytes, the density of [3H]nimodipine specific sites (106 per cell) was close to the density of [3H]QNB sites (0.8 × 106 per cell) and higher than that of [3H]DHA sites (0.2 × 106 per cell). During subcellular fractionation, [3H]nimodipine binding did not copurify with plasma membrane markers. The highest densities were found in fractions enriched in sarcolemma or in sarcoplasmic reticulum. No specific binding was found in mitochondria. These results indicate that the localization of [3H]nimodipine sites is not restricted to areas of the plasma membrane rich in β-adrenoceptors, muscarinic receptors and sodium pump sites.  相似文献

The effects of adrenergic and cholinergic receptor agonists and antagonists on single and clustered mouse cardiocytes in culture have been studied. Cardiocytes were obtained from mice, ranging in ages from 9 days in utero to 1 day postpartum, and were grown in culture for 2–14 days. Single isolated cells of every age tested possessed the ability to respond both via a muscarinic cholinergic receptor to the cholinergic agonist, carbamylcholine, and via α- and β-adrenergic receptors to norepinephrine and epinephrine. Thus, cholinergic and adrenergic receptors are simultaneously present on the same cell. Cardiocyte clusters had considerably higher sensitivity to both autonomic agents, but, because of the extensive functional specializations between cells, the localization of functional receptors to specific cells could not be made. [3H]Alprenolol, a potent β-adrenergic receptor antagonist, and [3H]quinuclidinyl benzilate ([3H]QNB), a potent muscarinic cholinergic receptor antagonist, were used to localize β-adrenergic and muscarinic cholinergic receptors by autoradiography. Quantitation of the muscarinic ACh receptor gave ~800 sites/μm2, a value comparable to that for the nicotinic ACh receptor on primary skeletal muscle in culture. Electrophysiological and fine-structural studies confirmed the myocardial nature of these cells.  相似文献

Abstract

The binding characteristics of [3H]quinuclidinyl benzilate ([3H]QNB) to isolated crude membranes of cultured bovine aortic endothelial cells were investigated. [3H]QNB bound to endothelial cell membranes with high affinity (kD = 0.056 nM) and limited capacity (132 fmol/mg DNA). The binding specificity, order of affinity and inhibition constants (Ki) were determined by displacement of bound [3H]QNB with unlabeled ligands. The order of affinity was QNB > atropine > 4-diphenylacetoxy-N-methyl-piperidine methiodide (4-DAMP) > p-fluoro-hexahydro-sila-difenidol (p-F-HHSiD) (M3 antagonist) > pirenzepine (M1 antagonist) > AFDX-116 (M2 antagonist) > (4-hydroxy-2-butynyl) trimethylammonium chloride m-chlorocarbanilate (McN-A-343, M1 agonist). These observations suggest that muscarinic receptors of endothelial cells in culture are likely to be of M3 and M1 subtype. Northern blot analysis of receptor subtypes using cDNA probes did not provide conclusive results due to the low level expression of these receptors in cultured cells. Solubilization of protein bound [3H]QNB with 1% digitonin and 0.02% cholate followed by analysis on sucrose density gradients demonstrated the presence of a specifically bound [3H]QNB-protein complex sedimenting at the 6.2S region of the gradient. These data demonstrate the presence of muscarinic acetylcholine receptor protein in cultured bovine aortic endothelial cells.  相似文献

Mouse neuroblastoma clone N1E-115 has muscarinic acetylcholine receptors that mediate cyclic GMP synthesis. This receptor-mediated response is not significantly higher than background until the cells have been maintained in the stationary phase for at least 1 week. The basis of the influence of time in culture on the cyclic GMP response was investigated. The relative amount of cyclic GMP synthesized by intact cells was measured by radioactively labeling the GTP pool with [3H]guanine, incubating cells with agonists, and then chromatographically isolating [3H]cyclic GMP. Carbamylcholine-, ionophore X-537A-, and sodium azide-induced cyclic GMP formation increased with time in culture to a maximum of 13-, 9-, and 2.5-fold above basal, respectively. There was no change in the number or the apparent affinity of the muscarinic receptors as measured by [3H]quinuclidinyl benzylate ([3H]QNB) binding. In addition, there was no change in the apparent affinity of the receptors for agonist as measured by the ability of carbamylcholine to displace the specific binding of [3H]QNB. Guanylate cyclase activity per milligram protein and per cell in-creased six- and sevenfold, respectively, from day 0 to day 22. However, this increase in guanylate cyclase appeared to precede the marked increase in sensitivity of the cells to agonists. These data suggest that, in addition to guanylate cyclase and muscarinic receptors, there is another factor which is responsible for the development of this muscarinic receptor-mediated response.  相似文献

α-[3H]Bungarotoxin was prepared by catalytic reduction of iodinated α-bungarotoxin with tritium gas. Crude mitochondrial fraction from rat cerebral cortex bound 40 · 10?15 ?60 · 10?15 moles of α-[3H]bungarotoxin per mg of protein. This binding was reduced by 50% in the presence of approx. 10?6 M d-tubocurarine or nicotine, 10?5 M acetylcholine, 10?4 M carbamylcholine or decamethonium or 10?3 M atropine. Hexamethonium and eserine were the least effective of the drugs tested. Crude mitochondrial fraction was separated into myelin, nerve endings, and mitochondria. The highest binding of toxin per mg of protein was found in nerve endings, as well as the greatest nhibition of toxin binding of d-tubocurarine. Binding of α-[3H]bungarotoxin to membranes obtained by osmotic shock of the crude mitochondrial fraction indicates that the receptor for the toxin is membrane bound. 125I-Labeled α-bungarotoxin, prepared with Na 125I and chloramine T, was highly specific for the acetylcholine receptor in diaphragm, however, it was less specific and less reliable than α-[3H]bungarotoxin in brain. We conclude that a nicotinic cholinergic receptor exists in brain, and that α-[3H]bungarotoxin is a suitable probe for this receptor.  相似文献

The administration of convulsant drugs has proven a powerful tool to study experimental epilepsy. We have already reported that the administration of convulsant 3-mercaptopropionic acid (mp) at 150 mg/kg enhances binding affinity of muscarinic antagonist [3H]quinuclidinyl benzilate ([3H]QNB) to certain rat CNS membranes during seizure and postseizure without affecting site number. Results obtained with a 100-mg/kg dose of mp have shown reversible increases in [3H]QNB binding to cerebellum and hippocampus, whereas a delayed response has been found in striatum. Neither a subconvulsant dose nor in vitro addition modifies binding. In order to evaluate preseizure, seizure as well as early (30 min) and late (24 h) postseizure stages, we employed a 50 mg/kg dose and tested [3H]QNB binding to CNS membranes. Changes in binding were as follows (in %): in cerebellum, +37, +86, and +40 at preseizure, seizure and early postseizure stages, respectively, but there was a decrease at late postseizure; in hippocampus, +27 at pre- and seizure stages, but a decrease at early and late postseizure. No changes were found in striatum or cerebral cortex membranes at any stage studied. Saturation curves analysed by Scatchard plots indicated that changes in [3H]QNB binding to cerebellar membranes are attributable to an increase in ligand affinity at seizure, followed by a decrease in binding site number at postseizure. A similar profile was observed for hippocampus except that the decrease in binding site number, though lower than at postseizure, was already evident at seizure stage. Results confirm a region-specific response to the convulsant and transient changes provide an example of neuronal plasticity.  相似文献
