The seagrass Posidonia oceanica plays a multifunctional role in the coastal area as an important and productive component of ecosystems in the Mediterranean Sea. We detected by RAPD analysis with two arbitrary primers genetic differences in P. oceanica collected from several sites in the Southern Mediterranean. By AMOVA analysis we observed a level of about 20% genetic difference among individuals within a population and 80% among populations. A common band of 200 bp was found in all the amplified samples. Cloning and sequencing analysis of this band revealed the presence of a simple tandem repeat sequence (minisatellite) that we called PoTR (Posidonia oceanica tandem repeat). Finally, the ability of PoTR to detect genetic variability in P. oceanica genome was demonstrated by the presence of amplification products of different lengths utilizing primers internal to this sequence.  相似文献

Mercury concentrations, non-protein thiol levels and the enzyme activities of glutathione-S-transferase (GST) were measured in the blades and sheaths of the marine phanerogam Posidonia oceanica. The seagrass was collected in January and June and at three sites: the Bay of Rosignano (Italy) known for its mercury contamination, the north of the Lérins islands (Bay of Cannes, France), the Bay of Tonnara (Corsica, France). The two latter sites are considered as free of any known industrial inputs. Mercury concentrations and GST activities in both tissues were always higher in samples from Rosignano, particularly in June. Non-protein thiol levels were significantly higher in the blades than in the sheaths of P. oceanica from Tonnara and Lérins. In contrast, at Rosignano, the sheaths presented a significantly higher non-protein thiol concentration than the blades, particularly in June. Levels in the sheaths appeared to increase with the degree of pollution. Western Blot performed on sheaths of P. oceanica collected in June at Rosignano and Lérins revealed a characteristic band of GSTs at 31 kDa, proving the presence of the GST enzyme in this tissue. Mercury seemed to exert an influence upon non-protein thiol metabolism, including GST induction, in P. oceanica collected from the NW Mediterranean.  相似文献

The location and dynamics of Posidonia oceanica beds were studied in the marine natural reserve of Cerbère-Banyuls-sur-Mer (eastern Pyrenees, France), through the mapping of bottom assemblages (transects and image processing) and the monitoring of fixed markers installed at the Posidonia oceanica lower limit. The surface area occupied by continuous beds is relatively limited and localized, whereas the presence of important areas of dead 'matter' would seem to indicate that bottom cover by Posidonia oceanica was more extensive in the past. Although the limited extension of the seagrass beds seems to be linked to both a high level of turbidity and severe hydrodynamic conditions, it is hypothesized that the regressions observed are due to lack of sediment.  相似文献

This study describes the changes in the structure of the fish assemblage associated with Posidonia oceanica meadows after the establishment of a marine protected area. The survey of the ichthyofauna in the Marine Reserve of Tabarca Island was carried out by visual censuses with SCUBA. Inside and outside the reserve was sampled six times at random from summer 1997 to summer 1999 and in three different depths (1, 5 and 15 m) by applying transects of 150 × 5 m over the P. oceanica meadows. The data were analyzed with multivariant methodologies and with analysis of variance. The most represented families in number of species were the sparids and labrids. Mean abundance, biomass and number of species were higher inside the reserve in all studied periods. Inside the marine protected area, these variables decreased as depth increased; outside the protected area the pattern was the opposite, reaching the maximum at 15 m.  相似文献

Densities of red mullet Mullus surmuletus have declined since 1996 at Cap Martin, Alpes‐Maritimes, France, following colonization by Caulerpa taxifolia . Foraging M. surmuletus were rarely observed over C. taxifolia or the seagrass Posidonia oceanica , but were observed over bare sand and endemic macroalgae. Within colonized sites (where the mean cover of C. taxifolia was 30–100%), fish were concentrated on areas with low cover of C. taxifolia (<21%). A difference in the frequency of observations of foraging groups (17%) was observed between sites with and without C. taxifolia . The observed patterns were probably due to a physical barrier to foraging that is presented by meadows of C. taxifolia . Changes to habitat structure that occur when the substratum is colonized by C. taxifolia influence the accessibility to benthic food resources, and consequently the foraging activities of fish that feed on benthic invertebrates. The decline in density of M. surmuletus at Cap Martin could be the result of fish emigration from colonized sites (unfavourable to the species) to uncolonized sites.  相似文献

The study of the dynamics of Posidonia oceanica beds, and more particularly, the position of their lower limit, are usually carried out by the installation and monitoring of markers set on the sea bottom. Acoustic telemetry could constitute an interesting alternative solution to the marking of the lower limit seagrass beds. In order to test the potentialities of this system, an experiment was conducted in the Natural Marine Reserve of Cerbère-Banyuls (eastern Pyrenees, France). The possibility of very precise mapping offers many potentialities with regards to the work of monitoring networks in the marine environment (species and assemblages of importance), but also in the monitoring of competition dynamics between the species.  相似文献

Posidonia oceanica supports mainly saprophytic marine flora, comprising predominantly lignicolous fungi. The frequency of occurrence of species recorded on this marine angiosperm, was high, indicating that they play a major role in the biological degradation of the sea grass Posidonia oceanica. In vitro experiments with Corollospora maritima (isolated from leaf material) were conducted in order to evaluate their role in the degradation of leaf material. Corollospora maritima actively degrade leaf material. Biophysical and biochemical changes (particle detritus formation, C and N variation), enzymatic activity involved and sterol production were studied during the transformation process of leaves to mycelial biomass.  相似文献

The Migjorn artificial reef (MAR) was deployed in the summer of 1990 and is made by 48 concrete blocks set 30 m deep on a sea bed of sand with a small proportion of Posidonia oceanica (L.) Delile meadow. Visual censuses of blocks on both substrata were carried out during the months of February, May and September in 1992 and 1993. The MAR fish assemblages were influenced by the surronding substrate. Thus, a group of ubiquitous species, frequent on both substrata and typical of natural rocky habitats (mostly Sparidae) and a group associated with the samples recorded on the Posidonia meadow (mostly Labridae) could be distinguished. Season had only a secondary and slight effect on fish assemblages. The MAR was colonized by 74% of the species in the first two years after deployment. Positive and lineal relationships between species richness, diversity, abundance of necto-benthic species and time since deployment on sandy substrata were found. On Posidonia oceanica deployment time has only increased very sedentary and cryptic species. Although some ways in which the MAR would have a productive function are not rejected, the most obvious MAR working pattern is that of fish aggregation. Interannual changes in leaf production and rhizome elongation rates of the seagrass Posidonia oceanica have been evaluated by means of reconstructive methods at the Travello meadow (Ligurian Sea, NW Mediterranean, Italy) to provide evidence of responses to the putative impact of a beach replenishment made with terrigenous materials in 1993. Two additional meadows (Genoa-Quinto and Noli) were sampled as controls. An asymmetrical sampling design ('beyond BACI': Before/After, Control/Impact) was thus used to detect the impact on the basis of a single sampling as dating methods obviated the lack of pre-impact data. At all three meadows investigated, leaf production and rhizome elongation rates were reconstructed for 12 previous years (from 1988 to 1999). A marked decrease in the leaf production rate (around 20%) was assessed only at Travello immediately after the putative disturbance. Control meadows, instead, did not display any significant variations in the pattern of change of this variable from before to after the putative impact. With regard to rhizome growth rates, no significant changes in space and time attributable to the putative impact have been detected. The present data also suggest that the impact studied may be considered as a pulse disturbance, since leaf production appeared to recover over a comparatively short-time scale (around 2-3 years) if compared to the low turn-over and high longevity of P. oceanica. The high potential of asymmetrical sampling designs in combination with dating methods is discussed in the light of the results presented here.  相似文献

Posidonia oceanica is an endemic Mediterranean seagrass species that has often been assumed to contain low levels of genetic diversity. Random amplified polymorfic DNA (RAPD) markers were used to assess genetic diversity among five populations from three geographical regions (north, central, and south) of the western Mediterranean Sea. Stranded germinating seeds from one of the central populations were also included in the analysis. Forty-one putative genets were identified among 76 ramets based on 28 RAPD markers. Genotypic diversity strongly depended on the spatial structure, age, and maturity of the meadows. The lowest clonal diversity was found in the less structured and youngest prairies. Conversely, a high genotypic diversity was found in the highly structured meadows. The genotypic diversity in these meadows was at the same level as in P. australis and higher than previously reported data for P. oceanica populations in the Tyrrhenian Sea near the coast of Italy.  相似文献

High-resolution multispectral imagery, aerial photography, and SCUBA diving surveys were used to determine if there had been any significant alteration in the dimensions of Posidonia oceanica beds in the Bay of Menton, French Riviera, since colonization of the green alga Caulerpa taxifolia began in the late 1980s. After 8 years of direct contact, there was no evidence of a decrease in the abundance of P. oceanica. On the contrary, the integrity of a seagrass bed had increased in the first area colonized by Caulerpa taxifolia on the eastern side of Cape Martin and, in a more recently colonized central part of the bay, established tufts of Posidonia were isolated from the alga by well-defined exclusion zones.  相似文献

The concentration of phenolic compound was measured in the seagrass Posidonia oceanica when interacting with two Bryopsidophyceae, Caulerpa taxifolia and Caulerpa racemosa, between May 1999 and May 2000. These measurements were performed on adult and intermediate leaves and in sheaths of the seagrass. Sampling was carried out at three stations subject to increasing levels of interaction with Caulerpa. The number of tannin cells was also analysed. Five phenolic compounds were identified in P. oceanica, with a predominance of caffeic acid in the adult and intermediate leaves. For a given level of interaction (and for both caulerpa sp.), a significant seasonal variation in phenolic compounds was shown in the adult leaves (higher in November and lower in September and March for example for the interaction with C. taxifolia). Only for two compounds (corresponding to a mixture containing ferulic acid and the ester methyl 12-acetoxyricinoleate) were significant differences observed as a function of the level of interaction with C. taxifblia, and only in the adult leaves: higher concentrations of phenols were observed with increasing level of interaction. Thus,adult leaves gave values of 55.5 +/- 14.1 microg g(-1) dm without interaction (OCt) and 94.9 +/- 23.4 microg g(-l) dm with high interaction (2Ct),corresponding to an increase of 70%. No significant difference was observed with intermediate leaves and sheaths, or for interaction with C. racemosa. The number of tannin cells (supposed to produce the phenolic compounds) largely increased in the adult and intermediate leaves when the degree of interaction with C. taxifolia increased: 90 mm above the base of the sheath (in adult leaves), 16.7 +/- 10.6 tannin cells cm(-2) were found without interaction (OCt), and 57.8 + 21.2 tannin cells cm(-2) with high interaction (2Ct). No significant difference was found for C. racemosa interaction. It thus appears that when the seagrass P. oceanica is in interaction with C. taxifolia, it accelerates its production of secondary metabolites so as to limit invasion of the beds.  相似文献

MFO enzyme activities were measured in microsomes from whole mussels (Mytilus galloprovincialis) comber livers (Serranus cabrilla), or Posidonia oceanica etiolated tissues, and PAH contents were determined in sediments collected in coastal locations of the French Riviera and Corsica during 3 oceanographic cruises in 1987-1988. 2. BaP activities in mussel and EROD activities measured in fish were strongly correlated to the log of PAH content in sediments. The first results for CA4H in Posidonia showed significant differences related to PAH pollution levels. The increase in MFO activities measured in Corsica in summer 1988 indicated a recent petroleum contamination.  相似文献

Temporal and spatial variation in density, biomass and body size of littoral fish species associated with nearshore Posidonia oceanica meadows was studied over an annual cycle in an area of the eastern Mediterranean Sea. A total of 109,350 littoral fishes were collected, belonging to 34 families and 88 species. Density of fishes peaked during the summer due to high numbers of juveniles. Season was a significant factor determining density, although number of species and biomass did not show any obvious seasonal pattern. Throughout the study, schooling planktivorous fish species such as the picarel Spicara smaris, the bogue Boops boops and the damselfish Chromis chromis were dominant, both in terms of density (80%) and biomass (70%). Temporal variation in density and body size of fishes was used to assess the seasonal and ontogenetic habitat use of each species, with their affinity to seagrass assessed by comparing their respective distribution on sand. Four functional guilds were created (juvenile migrants, seagrass residents, seasonal migrants and occasional visitors) to describe the habitat use of P. oceanica meadows by each species. Several species associated with P. oceanica meadows used this habitat mainly as juveniles during summer, although many others were present concurrently as adults and as juveniles. Among the species encountered, 11 were non-indigenous of Indo-Pacific origin, of which three used seagrasses mainly as juveniles and four as residents. The non-indigenous silverstripe blaasop Lagocephalus sceleratus ranked among the 10 most dominant species in terms of biomass (2%) and was classified as a seagrass resident.  相似文献

Late in summer 2003, extensive mass mortality of at least 25 rocky benthic macro-invertebrate species (mainly gorgonians and sponges) was observed in the entire Northwestern (NW) Mediterranean region, affecting several thousand kilometers of coastline. We were able to characterize the mortality event by studying six areas covering the main regions of the NW Mediterranean basin. The degree of impact on each study area was quantified at 49 sites by estimating the proportion of colonies affected in populations of several gorgonian species compared with reference data obtained in years without mortality signs. According to these data, the western areas (Catalan coast and Balearic Islands) were the least affected, while the central areas (Provence coast and Corsica-Sardinia) showed a moderate impact. The northernmost and eastern areas (Gulf of Genoa and Gulf of Naples) displayed the highest impact, with almost 80% of gorgonian colonies affected. The heat wave of 2003 in Europe caused an anomalous warming of seawater, which reached the highest temperatures ever recorded in the studied regions, between 1 and 3 °C above the climatic values (mean and maximum). Because this exceptional warming was observed in the depth ranges most affected by the mortality, it seems likely that the 2003 anomalous temperature played a key role in the observed mortality event. A correlation analysis between temperature conditions and degree of impact seems to support this hypothesis. Under the present climate warming trend, new mass mortality events may occur in the near future, possibly driving a major biodiversity crisis in the Mediterranean Sea.  相似文献

In the late summer of 1999, an extensive mortality of gorgonians and other epi-benthic organisms was observed in the Ligurian Sea (Mediterranean Sea) from the Tuscan Archipelago to Marseille. Quantitative data from Tino Island and Portofino Promontory indicated that the proportion of affected gorgonians ranged from 60% to 100% in populations having a density of 9–27.8 colonies m−2, suggesting that millions of sea fans died along the coast of Liguria. This mass mortality episode coincided with a sudden increase of sea water temperature down to more than 50 m depth. Laboratory analyses showed that the colonies stressed by high temperature also underwent extensive attack by microrganisms (protozoans and fungi), which are interpreted as opportunistic pathogens.  相似文献

The Platte River caddisfly (Ironoquia plattensis Alexander and Whiles 2000) was recently described from a warm-water slough along the Platte River in central Nebraska and was considered abundant at the type locality. Surveys of 48 sites in 1999 and 2004 found eight additional sites with this species on the Platte River. The caddisfly was not found at the type locality in 2004 and one additional site in 2007, presumably because of drought conditions. Because of its apparent rarity and decline, the Platte River caddisfly is a Tier I species in Nebraska. For this project, surveys for the caddisfly were conducted at 113 new and original sites primarily along the Platte, Loup, and Elkhorn Rivers between 2009 and 2011. These surveys identified 30 new sites with the caddisfly. Larval densities were quantified at a subset of inhabited sites, and there was a large variation of densities observed. Seven sites on other Nebraska drainages were found to support morphologically similar caddisflies, presumably the Platte River caddisfly. Because of the discovery of populations outside the Platte River drainage, amplified fragment length polymorphism (AFLP) was used to determine the amount of genetic variability and breeding among sites on the Platte, Loup, and Elkhorn Rivers. Analysis of molecular variance (AMOVA) suggested moderate gene flow among the three river systems and that there was more genetic variation within populations than between populations. Differentiation, but not total divergence, was exhibited by the northernmost population from the Elkhorn River. Because it may be considered an indicator species and is vulnerable to ongoing habitat loss and degradation, all Platte River caddisfly populations should be conserved.  相似文献

Mounting evidence indicates large-scale climatic phenomena such as El Niño Southern Oscillation (ENSO) can overwhelm endogenous factors that govern the population dynamics of wild species. We add to this evidence by documenting an ENSO-related decline of large mammals in the Kumbhalgarh Wildlife Sanctuary, in Rajasthan, India. This event coincided with the drought of 2000, following two consecutive monsoon failures. Time series of biennial counts (1991–2005) shared a common feature: all 13 species declined in abundance from 1999 to 2001, with 11 species experiencing an apparent decline exceeding 25%. An ENSO index explained much of the variability in population size, apparently reflecting mass mortality and/or recruitment failure caused by the major 1998–2000 La Niña event, followed by a rapid rebound. ENSO apparently overwhelmed endogenous factors and synchronized the dynamics of the mammalian community. Our findings may prove to be symptomatic of geographically broad impacts of large-scale climate on the dynamics of terrestrial vertebrate communities, even in protected areas. Our findings reinforce the growing recognition that we should not overlook global-scale causal agents of ecological change.  相似文献
