Genetic diversity of the house mouse Mus musculus from 12 local populations (n = 65) of the central and eastern parts of the former Soviet Union was examined using RAPD-PCR. About 400 loci were identified, encompassing approximately 500 kb of the mouse genome. Genetic diversity was assessed using NTSYS, POPGENE, TFPGA, and TREECON software programs. In general, the house mouse sample from the regions examined was characterized by moderate genetic variation: polymorphism P = 95.6%, P99 = 60.7%, P95 = 24.2%; heterozygosity H = 0.089; the mean observed number of alleles n(a) = 1.97; effective number of alleles n = 1.13; intrapopulation differentiation deltaS = 0.387; gene diversity h = 0.09. Individual local populations displayed different levels of genetic isolation: the genetic subdivision index G(st) varied from 0.086 to 0.324 at gene flow Nm varying from 5.3 to 1.05, while the interpopulation genetic distance D(N) ranged from 0.059 to 0.186. Most of the genetic diversity of the total sample resided within the local populations: H(S) = 0.6, total gene diversity H(T) = 0.09. The exact test for differentiation, however, did not confirm the affiliation of all the mice examined to one population: chi2 = 1446, d.f. = 724, P = 0.000. Molecular markers specific to four subspecies (musculus, castaneus, gansuensis, and wagneri) were identified. Moreover, in some cases the populations and individual animals exhibited traits of different subspecies, suggesting their introgressive hybridization. It was demonstrated that the house mouse fauna on the territories investigated was characterized by the prevalence of musculus-specific markers, while gansuensis-specific markers ranked second. The castaneus-specific markers were highly frequent in the Far East, but almost absent in Central Asia, where wagneri-specific markers were detected. It was suggested that house mice from Turkmenistan could belong to one of the southern subspecies, which had not deeply penetrated into the Asian fauna of the former Soviet Union. In phenogenetic (UPGMA) and phylogenetic (NJ) reconstructions this form with the high bootstrap support was placed at the tree base, while the isolation of other clusters was not statistically significant. It is thus likely that the house mice from Turkmenistan are closest to the ancestral form of the genus Mus on the territory of the former Soviet Union.  相似文献

The present study analyses the capacity of house mice (Mus musculus) to solve a problem, consisting in opening a door which must be rotated 50 times in the same direction to allow access to the food reward. This capacity emerged spontaneously in 4.8 % of 500 animals belonging to a random bred Swiss population, tested beforehand. The experiment then investigated the effects of genetic and experiential (social) factors on transmission of this capacity from one individual to another. Five groups of approximately 70 animals each were compared: a) controls; b) animals with no experience of the problem but offspring of parents able to solve the problem spontaneously; c) animals with experience of the problem being offspring of parents able to solve it spontaneously; d) naive offspring of parents incapable of solving the problem spontaneously; e) animals with experience of the problem and offspring of parents incapable of solving it. The results showed considerable effects both of genetic and experiential (social) factors with values ranging from only 3.9 % of successful naive mice among offspring of unsuccessful individuals, up to 32.3 % of successful offspring of successful parents, reared with a mother who solved the problem several times in their presence.  相似文献

Despite the increased number of anthrax outbreaks in Georgia and the other Caucasian republics of the former Soviet Union, no data are available on the diversity of the Bacillus anthracis strains involved. There is also little data available on strains from the former Soviet Union, including the strains previously used for vaccine preparation. In this study we used eight-locus variable-number tandem repeat analyses to genotype 18 strains isolated from infected animals and humans at different sites across Georgia, where anthrax outbreaks have occurred in the last 10 years, and 5 strains widely used for preparation of human and veterinary vaccines in the former Soviet Union. Three different genotypes affiliated with the A3.a cluster were detected for the Georgian isolates. Two genotypes were previously shown to include Turkish isolates, indicating that there is a regional strain pattern in the South Caucasian-Turkish region. Four of the vaccine strains were polymorphic, exhibiting three different patterns of the cluster A1.a genotype and the cluster A3.b genotype. The genotype of vaccine strain 71/12, which is considered an attenuated strain in spite of the presence of both of the virulence pXO plasmids, appeared to be a novel genotype in the A1.a cluster.  相似文献

Summary Of all the introduced mammalian pests, rodents are the most common invaders of islands and have triggered numerous extinctions around the globe. They have a short gestation period and large litter size, giving them the ability to colonise new areas rapidly. The same biological traits make them difficult to eradicate. Although the biodiversity benefits of removing exotic rodents from islands are increasingly being recognised, there are few published analyses of eradication attempts that critically evaluate eradication tools. This study examined the response of House Mouse (Mus musculus) populations to an eradication operation on Montague Island, Australia. While the specific impacts of mice have not been studied on this particular island, elsewhere they have been shown to have negative impacts on a range of species including plants, invertebrates, lizards and seabirds. On Montague Island, mouse abundance across different habitats was examined using mark–recapture data collected before and after the deployment of brodifacoum baits. Data obtained before baiting showed significantly lower numbers of mice in sites dominated by exotic grass compared with those dominated by native vegetation. Mouse numbers overall were declining during winter and the population was not breeding, making this an optimal time to undertake the eradication. Trapping immediately after the initial bait drop failed to capture any survivors, and no individuals have been detected during the 3 years since the deployment of baits. This study demonstrated that both small and large baits (0.6 and 2 g pellets) were effective in eradicating the House Mouse from Montague Island. While present, mice were probably slowing the re‐establishment of native vegetation by grazing seedlings and consuming seeds. With this pest now gone, the process of natural regeneration is expected to accelerate. The eradication of the House Mouse from Montague is likely to have other positive effects on the island’s biodiversity. The operation itself contributed to enhancing local capacity to eradicate exotic rodents from larger or more complex islands.  相似文献

The hybrid zone between the two subspecies of mice Mus musculus domesticus and Mus musculus musculus, which has been studied extensively in Denmark, crosses Europe to the Black Sea through the Alps and the Balkans. Two hundred and seventy-nine animals were captured in 22 localities along a transect across the Balkans. The animals were characterized for seven diagnostic nuclear loci by protein electrophoresis and by restriction pattern analysis of their mitochondrial DNA. The nuclear data show a sharp transition between the two subspecies, most of the variations in allele frequencies (from 0.9 to 0.1) occurring within a 36-km section of the transect. The introgression varies from one locus to the other and is more pronounced, in terms of distance, in M. m. musculus territory. Mitochondrial DNA introgression is important but occurs in one direction only, i.e. from M. m. musculus to M. m. domesticus, while a cytoplasmic transfer from M. m. domesticus to M. m. musculus has been reported. A previous study showed that no Y chromosome introgression occurs. The different behaviour of these three types of markers could be due to the interaction between selection against hybrid genomes and meiotic recombination. Objectively, it would appear that the genes that can introgress are neutral or nearly so and have been separated from deleterious genes they were linked to by recombination. This could explain the differential introgression between autosomal loci. The mitochondrial and Y chromosomes undergo no or very little recombination and each is transmitted as a whole. Their degree of introgression is thus indicative of the intensity of selection resulting from the amount of functional differentiation between the two taxa, which seems to be strong for the Y chromosome and weak for mitochondrial DNA. We propose that the asymmetry of nuclear introgression is due to different population structures. As M. m. musculus is relatively less structured, the rapid spreading of introgressed genes would be favoured. Such a scheme, however, can hardly account for the unidirectionality of the mitochondrial flow, which could be due to sex-dependent behaviour.  相似文献

Nineteen loci from 239 individuals of the house mouse Mus musculus domesticus (Rodentia, Muridae) were analyzed by means of thin layer electrophoresis. The mice were collected from 14 localities of Greece mainly confined to the area of NW Peloponnese, where a Robertsonian (Rb) system is observed. The individuals were chromosomally characterized by nine diploid numbers, the 2n = 24, 26, 27, 28, 29, 30, 31, 32, and 40. The statistic elaboration revealed that all 14 populations studied were not characterized by cohesive demic structure and high inbreed levels while the gene flow among them has resulted in low levels of genetic differentiation. The resulting values for Neis genetic distance corresponded to distances known for the level of geographical populations of, M. musculus. Wagners cladogram for the phylogenetic relations between the populations studied implied that it is the diploid number, rather than the geographical factor, that characterizes or dominates each population, which mainly influences the phylogenetic relationships.  相似文献

This study deals with the characterization of 10populations of M. musculus from differentgeographical locations in India. The genetics of Indianwild mice has been completely obscure and this is thefirst report on allozyme variations in the naturalpopulation. We have used a set of 24 biochemical geneticmarkers to measure levels of diversity within and amongpopulations. The allelic frequency data indicate extreme genetic variability, which is furtherenhanced by the presence of novel alleles. Overall thespecies shows a high level of heterogenity. The highlypolymorphic central populations of M. musculus cannot be assigned to any one particularsubspecies. The allelic profiles, however, indicate agradual differentiation toward the castaneus andbatcrianus subspecies lineages.  相似文献

Path integration is the animal's running computation of its position relative to a starting point based on recording all displacements. This process is known to be prone to cumulative errors and hence must somehow be corrected with the help of local spatial cues. In the reported experiments, the relative role of local spatial cues at two target locations was appraised. These two targets were the peripheral box of a female house mouse (Mus musculus) and a central box from where she retrieved her pups over a distance of 50 cm. The experimental conditions required the mouse to navigate between the two targets solely by means of path integration. To find out whether the mice continued integrating at the two target locations, directional misinformation was fed into their path integrating system. Hence, passive rotation of 90° proved to be sufficient to decide unambiguously whether the mice were misdirected. Such experimental interference consistently documented ongoing path integration at the target location outside the residential nest. In contrast, the same interference when the mouse was in the residential nest documented discontinuation of path integration in most cases. It is inferred that mice, when departing from residential nests, initially direct themselves by means of local guiding stimuli.  相似文献

The present study reports the biochemical genetic profile of commensal forms of Indian wild house mouse populations Mus musculus from ten localities as well as other species of genus Mus, M. platythrix, M. booduga and M. terricolor III. Extreme polymorphism seen by the presence of a large number of variants and novel alleles in these animals indicates that the Indian M. musculus holds considerable potential for development of useful models for biomedical research.  相似文献

General parameters of selection, such as the frequency and strength of positive selection in natural populations or the role of introgression, are still insufficiently understood. The house mouse (Mus musculus) is a particularly well-suited model system to approach such questions, since it has a defined history of splits into subspecies and populations and since extensive genome information is available. We have used high-density single-nucleotide polymorphism (SNP) typing arrays to assess genomic patterns of positive selection and introgression of alleles in two natural populations of each of the subspecies M. m. domesticus and M. m. musculus. Applying different statistical procedures, we find a large number of regions subject to apparent selective sweeps, indicating frequent positive selection on rare alleles or novel mutations. Genes in the regions include well-studied imprinted loci (e.g. Plagl1/Zac1), homologues of human genes involved in adaptations (e.g. alpha-amylase genes) or in genetic diseases (e.g. Huntingtin and Parkin). Haplotype matching between the two subspecies reveals a large number of haplotypes that show patterns of introgression from specific populations of the respective other subspecies, with at least 10% of the genome being affected by partial or full introgression. Using neutral simulations for comparison, we find that the size and the fraction of introgressed haplotypes are not compatible with a pure migration or incomplete lineage sorting model. Hence, it appears that introgressed haplotypes can rise in frequency due to positive selection and thus can contribute to the adaptive genomic landscape of natural populations. Our data support the notion that natural genomes are subject to complex adaptive processes, including the introgression of haplotypes from other differentiated populations or species at a larger scale than previously assumed for animals. This implies that some of the admixture found in inbred strains of mice may also have a natural origin.  相似文献

Genetic divergence between house mouse and gleaner mouse from different regions of Ukraine was estimated by electrophoresis at 26 loci. Four diagnostic loci were established among these species: IDH-1, Est-1,2,4. Genetic distance between species is 0.217, which is in accordance with genetic differences between other species of the genus Mus. In previous studies, 13 different recessive embryonic lethal genes have been associated with t haplotypes in the wild mice of the species Mus domesticus. In this communication we have analyzed five populations of Mus musculus for the presence and identity of t haplotypes. The populations occupy geographically distant regions in the Soviet Union: Altai Mountains, western and eastern Siberia, Azerbaijan and Turkmenistan. No t haplotypes were found in mice from eastern Siberia. In the remaining four populations, t haplotypes occurred with frequencies ranging from 0.07 to 0.21. All the t haplotypes extracted from these populations and analyzed by the genetic complementation test were shown to carry the same lethal gene tcl-w73. In one population (that of western Siberia), another lethal gene (tcl-w5) was found to be present on the same chromosome as tcl-w73. This situation is in striking contrast to that found in the populations of the western form of the house mouse, M. domesticus. In the latter species, tcl-w73 has not been found at all and the different populations are characterized by the presence of several different lethal genes. The low diversity of t haplotypes in M. musculus is consistent with lower genetic variability of other traits and indicates a different origin and speciation mode compared to M. domesticus. Serological typing for H-2 antigenic determinants suggests that most, if not all, of the newly described t haplotypes might have arisen by recombination of tw73 from M. musculus with t haplotypes from M. domesticus either in the hybrid zone between the two species or in regions where the two species mixed accidentally.  相似文献

Live and kill traps were used to remove House mice living in granaries, mixed-food stores, dairy units and ancillary stores on farms. Structural weaknesses were common to all buildings. Cavities in walls, in particular, provided favourable living and nesting quarters for mice and were rapidly exploited. The body-weight structure and reproductive traits of the populations in the different habitats were compared. The onset of maturity, in both sexes, was closely related to body-weight increase in the 10 to 14 g range. Breeding, unlike that in most out-door populations in temperate climates, continued throughout the year. Ancillary buildings holding limited food supported small reservoir populations. Mice inhabiting dairy units containing pelleted cattle feed tended to be heavier than those sampled elsewhere and females in the units produced the largest litters. Reproductive performance on farms was intermediate between that of mice occupying urban buildings and corn ricks. There was evidence that environmental and behavioural factors were operating to limit increase in numbers in the dairy units.  相似文献

The nucleic acid sequence relationship between mouse intracisternal type A-particles and type C and B viruses was examined by reciprocal complementary DNA-RNA hybridization; complementary DNAs prepared from the RNAs of intracisternal A-particles were hybridized with high-molecular-weight RNAs from a variety of murine tumor viruses, and complementary DNAs representing a variety of RNA tumor virus genomes were hybridized with the high-molecular-weight RNAs from A-particles. The criterion for homology between two types of virus was that the heterologous hybridization reaction occurs over the same RNA concentration range as the homologous reacton. The results of these hybridizations indicate that there are no major sequence homologies between the RNA of intracisternal A-particles and the RNA of representative members of type B and C viruses of Mus musculus.  相似文献

Divergence of dispersal regimens has been suggested to be the selective basis for the evolutionary differentiation of agonistic phenotypes found in natural populations of house mice. Dispersal propensity may, therefore, be expected to exhibit heritable variation in wild house mice, ultimately related to motivational differences causing observable differences in agonistic behaviour. To test for heritable components in dispersal propensity in wild house mice, father–offspring regressions of dispersal latencies from residential social groups were determined in standardized seminatural social settings. To evaluate potential motivational causes of phenotypic variation in dispersal behaviour, all test animals (fathers, sons, and daughters) were scored prior to the dispersal experiment in a standardized behavioural test, at 60 d of age. Activities were monitored in a 1 m2 square test arena during 10‐min observation periods. Test arenas exhibited four equidistant openings leading to cages containing fresh, own, sibling, or foreign bedding material. The apparatus allowed for scoring anxiety, exploratory activity, and kin preference. Subsequently, test animals were exposed to a resident population in a semi‐natural enclosure providing a dispersal opportunity. Father–son regressions of dispersal latencies were significantly positive, but no significant relationship was found for daughters. Dispersal latency decreased with increasing exploratory activity scores in males, but increased in females. Anxiety as well as kin preferences did not affect dispersal propensity. Hence sex‐linked, motivational components reflect heritable social behaviour variation in male house mice that may ultimately be caused by diverging dispersal regimens.  相似文献
