Combining measurements of electric potential and pH with such of chlorophyll fluorescence and leaf gas exchange showed heat stimulation to evoke an electrical signal (propagation speed: 3–5 mm s−1) that travelled through the leaf while reducing the net CO2 uptake rate and the photochemical quantum yield of both photosystems (PS). Two-dimensional imaging analysis of the chlorophyll fluorescence signal of PS II revealed that the yield reduction spread basipetally via the veins through the leaf at a speed of 1.6 ± 0.3 mm s−1 while the propagation speed in the intervein region was c. 50 times slower. Propagation of the signal through the veins was confirmed because PS I, which is present in the bundle sheath cells around the leaf vessels, was affected first. Hence, spreading of the signal along the veins represents a path with higher travelling speed than within the intervein region of the leaf lamina. Upon the electrical signal, cytoplasmic pH decreased transiently from 7.0 to 6.4, while apoplastic pH increased transiently from 4.5 to 5.2. Moreover, photochemical quantum yield of isolated chloroplasts was strongly affected by pH changes in the surrounding medium, indicating a putative direct influence of electrical signalling via changes of cytosolic pH on leaf photosynthesis.  相似文献

Observations were made of growth rate and variation in colour pattern of the leopard tortoise ( Geochelone pardalis babcocki ) in northern Tanzania between October 1993 and June 1996. Growth rate differed significantly between tortoises, with immature animals (6.9 ± 1.4 mm month−1) and females (5.7 ± 1.3 mm month−1) exhibiting higher rates of growth than adult animals (2.9 ± 0.8 mm month−1) and males (3.2 ± 1.1 mm month−1), respectively. The faster growth rate in immature animals suggested that growth rate declined with age. The difference in growth rates between the sexes was ascribed to sexual dimorphism, adult females being larger and therefore growing faster than males.
The carapacial colour pattern observed in the northern Tanzanian tortoises was generally similar to that described elsewhere in the species' range. The colour pattern varied between Arusha and Serengeti District tortoises. Animals at the Arusha sites tended to be sparsely coloured (55.6%) compared to the Serengeti tortoises, which had a buff colouration with radiating spots (89.3%).; The sparse colouration provides effective camouflage in the drier Arusha habitats, as does the buff colouration with black spots in the wetter, darker habitats in the Serengeti.  相似文献

Endurance swimming of diploid and triploid Atlantic salmon   总被引:1,自引:0,他引:1  
When groups of diploid (mean ±  s . e . fork length, L F) 33·0 ± 1·4 cm and triploid (35·3 ± 0·5 cm) Atlantic salmon Salmo salar were forced to swim at controlled speeds in a carefully monitored 10 m diameter 'annular' tank no significant difference was found between the maximum sustained swimming speeds ( U ms, maintainable for 200 min) where the fish swam at the limit of their aerobic capability. Diploids achieved 2·99 body lengths per second (bl s−1)(0·96 m s−1) and triploids sustained 2·91 bl s−1(1·02 m s−1). The selection of fish for the trials was based on their ability to swim with a moving pattern projected from a gantry rotating at the radius of the tank and the selection procedure did not prove to be significant by ploidy. A significant difference was found between the anaerobic capabilities of the fish measured as endurance times at their prolonged swimming speeds. During the course of the experimentation the voluntary swimming speed selected by the fish increased and the schooling behaviour improved. The effect of the curvature of the tank on the fish speeds was calculated (removing the curved effect of the tank increased the speed in either ploidy by 5·5%). Implications of the endurance times and speeds are discussed with reference to the aquaculture of triploid Atlantic salmon.  相似文献

Physiological impact of sea lice on swimming performance of Atlantic salmon   总被引:6,自引:0,他引:6  
Atlantic salmon Salmo salar were infected with two levels of sea lice Lepeophtheirus salmonis (0·13 ± 0·02 and 0·02 ± 0·00 sea lice g−1). Once sea lice became adults, the ventral aorta of each fish was fitted with a Doppler cuff to measure cardiac output ( ̇ ), heart rate ( f H) and stroke volume ( V S) during swimming. Critical swimming speeds ( U crit) of fish with higher sea lice numbers [2·1 ± 0·1 BL (body lengths) s−1] were significantly lower ( P  < 0·05) than fish with lower numbers (2·4 ± 0·1 BL s−1) and controls (sham infected, 2·6 ± 0·1 BL s−1). After swimming, chloride levels in fish with higher sea lice numbers (184·4 ± 11·3 mmol l−1) increased significantly (54%) from levels at rest and were significantly higher than fish with fewer lice (142·0 ± 3·7 mmol l−1) or control fish (159·5 ± 3·5 mmol l−1). The f H of fish with more lice was 9% slower than the other two groups at U crit. This decrease resulted in ̇ not increasing from resting levels. Sublethal infection by sea lice compromised the overall fitness of Atlantic salmon. The level of sea lice infection used in the present study was lower than has previously been reported to be detrimental to wild Atlantic salmon.  相似文献

Atlantic salmon Salmo salar were infected with sea lice Lepeophtheirus salmonis (0·08 ± 0·007 sea lice g−1) over a period of 4 h. Both infected and non‐infected fish were swim tested in sea water (SW) and fresh water (FW). The ventral aorta of each fish was fitted with a Doppler cuff in order to measure cardiac output, stroke volume and heart rate during swim testing. Blood samples were taken at rest and after exercise. Critical swimming speed of infected fish in SW (2·14 ± 0·08 body lengths, bl s−1) was significantly lower ( P  < 0·05) than infected fish switched to FW (2·81 ± 0·08 bl s−1) and non‐infected fish in SW (2·42 ± 0·04 bl s−1) and FW (2·61 ± 0·08 bl s−1). Cardiac and blood results indicated infected fish exposed to FW did experience stress, but relief from osmotic and ionic distress probably reduces energy expenditure, allowing the increase in performance. As the performance of sea lice‐infected fish improved upon transfer to FW, it is likely that heavily infected salmonids do return to FW to restore compromised osmotic and ionic balance, and remove sea lice in the process.  相似文献

Xylem maturation in elongating leaf blades of tall fescue ( Festuca arundinacea ) was studied using staining and microcasting. Three distinctive regions were identified in the blade: (1) a basal region, in which elongation was occurring and protoxylem (PX) vessels were functioning throughout; (2) a maturation region, in which elongation had stopped and narrow (NMX) and large (LMX) metaxylem vessels were beginning to function; (3) a distal, mature region in which most of the longitudinal water movements occurred in the LMX. The axial hydraulic conductivity ( K h) was measured in leaf sections from all these regions and compared with the theoretical axial hydraulic conductivity ( K t) computed from the diameter of individual inner vessels. K t was proportional to K h throughout the leaf, but K t was about three times K h. The changes in K h and K t along the leaf reflected the different stages of xylem maturation. In the basal 60 mm region, K h was about 0.30±0.07 mmol s−1 mm MPa−1. Beyond that region, K h rapidly increased with metaxylem element maturation to a maximum value of 5.0±0.3 mmol s−1 mm MPa−1, 105 mm from the leaf base. It then decreased to 3.5±0.2 mmol s−1 mm MPa−1 near the leaf tip. The basal expanding region was observed to restrict longitudinal water movement. There was a close relationship between the water deposition rate in the elongation zone and the sum of the perimeters of PX vessels. The implications of this longitudinal vasculature on the partitioning of water between growth and transpiration is discussed.  相似文献

Foliar respiration is a major component of ecosystem respiration, yet extrapolations are often uncertain in tropical forests because of indirect estimates of leaf area index (LAI). A portable tower was used to directly measure LAI and night-time foliar respiration from 52 vertical transects throughout an old-growth tropical rain forest in Costa Rica. In this study, we (1) explored the effects of structural, functional and environmental variables on foliar respiration; (2) extrapolated foliar respiration to the ecosystem; and (3) estimated ecosystem respiration. Foliar respiration temperature response was constant within plant functional group, and foliar morphology drove much of the within-canopy variability in respiration and foliar nutrients. Foliar respiration per unit ground area was 3.5 ± 0.2  µ mol CO2 m−2 s−1, and ecosystem respiration was 9.4 ± 0.5  µ mol CO2 m−2 s−1[soil = 41%; foliage = 37%; woody = 14%; coarse woody debris (CWD) = 7%]. When modelled with El Niño Southern Oscillation (ENSO) year temperatures, foliar respiration was 9% greater than when modelled with temperatures from a normal year, which is in the range of carbon sink versus source behaviour for this forest. Our ecosystem respiration estimate from component fluxes was 33% greater than night-time net ecosystem exchange for the same forest, suggesting that studies reporting a large carbon sink for tropical rain forests based solely on eddy flux measurements may be in error.  相似文献

When swimming at low speeds, steelhead trout and bluegill sunfish tilted the body at an angle to the mean swimming direction. Trout swam using continuous body/caudal fin undulation, with a positive (head-up) tilt angle ( 0 , degrees) that decreased with swimming speed ( u , cm s−1) according to: 0 =(164±96).u(−1.14±0.41) (regression coefficients; mean±2 s.e. ). Bluegill swimming gaits were more diverse and negative (head down) tilt angles were usual. Tilt angle was −3·0 ± 0.9° in pectoral fin swimming at speeds of approximately 0.2–1.7 body length s−1 (Ls−1; 3–24 cm s−1), −4.5 ±2.6° during pectoral fin plus body/caudal fin swimming at 1·2–1·7 L s−1 (17–24cm s−1), and −5.0± 1.0° during continuous body/caudal fin swimming at 1.6 and 2.5 L s−1 (22 and 35cm s−1). At higher speeds, bluegill used burst-and-coast swimming for which the tilt angle was 0.1±0.6°. These observations suggest that tilting is a general phenomenon of low speed swimming at which stabilizers lose their effectiveness. Tilting is interpreted as an active compensatory mechanism associated with increased drag and concomitant increased propulsor velocities to provide better stabilizing forces. Increased drag associated with trimming also explains the well-known observation that the relationship between tail-beat frequency and swimming speed does not pass through the origin. Energy dissipated because of the drag increases at low swimming speeds is presumably smaller than that which would occur with unstable swimming.  相似文献

Critical swimming speeds (mean ± s . e .) for juvenile shortnose sturgeon Acipenser brevirostrum were 34·4 cm s−1± 1·7 (2·18 ± 0·09 body lengths, BL s−1). Swimming challenges at 10, 20 and 30 cm s−1 revealed that juvenile A. brevirostrum are relatively poor swimmers, and that the fish did not significantly modify their swimming behaviour, although they spent more time substratum skimming ( i.e. contact with flume floor) at 30 cm s−1 relative to 10 cm s−1. When present, these behavioural responses are probably related to morphological features, such as flattened rostrum, large pectoral fins, flattened body shape and heterocercal tail, and may be important to reduce the costs of swimming.  相似文献

The growth rates of naturally sympatric juvenile pink Oncorhynchus gorbuscha and sockeye Oncorhynchus nerka salmon were compared in a common lacustrine environment in south‐west Alsaka, an unusual opportunity given the normal disparity in freshwater residence time of these two species. Fork length ( L F) frequency distributions of juvenile pink salmon caught in the lake during the summer in 1991 and 1999–2003 indicated a growth rate of 0·54 mm day−1, 54% greater than the estimated growth rate of juvenile sockeye salmon sampled from 1958 to 2003 (0·35 mm day−1). Examination of daily growth rings on otoliths indicated that pink salmon in Lake Aleknagik grew an average of 1·34 mm day−1 in 2003 but sockeye salmon grew only 0·63 mm day−1(average specific growth rates were 3·0 and 1·8% day−1, respectively). Pink salmon increased from c . 32 mm L F and 0·2 g at emergence to 78 mm L F and 3·0 g within 3–4 weeks. After experiencing these rapid growth rates, the pink salmon appeared to leave the lake by late July in most years. The diets of pink and sockeye salmon in the littoral zone of the lake were very similar; >80% of the stomach contents consisted of adult and pupal insects and the remainder was zooplankton. This high degree of diet overlap suggested that the observed differences in growth rate were not attributable to variation in prey composition.  相似文献

This study compared the specific activity of phosphoenolpyruvate carboxylase (PEPC) of Spartina densiflora Brongn., collected from four populations along its latitudinal distribution range. Spartina densiflora is a halophyte with C4 photosynthesis that has a very wide latitudinal distribution, from Patagonia to the southwest Iberian Peninsula. The basis of intraspecific differences in PEPC activity were analysed by recording the phosphorylation state and amount of the enzyme, comparing leaf anatomy and evaluating leaf gas exchange. S. densiflora individuals from Patagonia had 60% higher PEPC specific activity than plants from the other three populations due to higher levels of PEPC protein that coincided with lower activation mediated by phosphorylation, yielding similar net photosynthesis rate ( c . 29 μmol CO2·m−2·s−1). Patagonian plants had a higher area of photosynthetic mesophyll relative to total chlorophyll than plants from north Argentina and the southwest Iberian Peninsula. Ecotypic differentiation in PEPC activity and leaf anatomy were found, distinguishing a higher-latitude ecotype from lower-latitude populations. The higher PEPC protein levels of the Patagonian ecotype seemed to be a response to lower light activation level of the enzyme, as judged by the low PEPC phosphorylation state.  相似文献

Given the influence of photoperiod on reproductive development and whole-plant senescence in monocarpic plants, one would suspect that leaf senescence in these plants might be under photoperiodic control. In Arabidopsis thaliana , which is monocarpic and also a nonobligate long-day (LD) plant, LDs (16 h, 300 μmol m−2 s−1) caused leaves to die earlier than did short days (SDs, 10 h). Since leaf longevity was not paralleled by the reproductive development in the present study, the reproductive structures did not seem to be the primary controls of leaf senescence. The LD effect appeared to depend on the amount of light rather than on day length, for leaves given LDs at reduced light intensity (180 μmol m−2 s−1) lived longer than those in LDs with full light. In addition, the higher light intensity promoted chlorophyll loss and anthocyanin accumulation in LDs. Thus, senescence of these leaves seems to be governed by light dosage rather than photoperiod. Light may play a natural role in promoting the senescence of A. thaliana leaves.  相似文献

Nutrient acquisition in the mature root zone is under systemic control by the shoot and the root tip. In maize, exposure of the shoot to light induces short-term (within 1–2 min) effects on net K+ and H+ transport at the root surface. H+ efflux decreased (from −18 to −12 nmol m−2 s−1) and K+ uptake (∼2 nmol m−2 s−1) reverted to efflux (∼−3 nmol m−2 s−1). Xylem probing revealed that the trans-root (electrical) potential drop between xylem vessels and an external electrode responded within seconds to a stepwise increase in light intensity; xylem pressure started to decrease after a ∼3 min delay, favouring electrical as opposed to hydraulic signalling. Cutting of maize and barley roots at the base reduced H+ efflux and stopped K+ influx in low-salt medium; xylem pressure rapidly increased to atmospheric levels. With 100 m m NaCl added to the bath, the pressure jump upon cutting was more dramatic, but fluxes remained unaffected, providing further evidence against hydraulic regulation of ion uptake. Following excision of the apical part of barley roots, influx changed to large efflux (−50 nmol m−2 s−1). Kinetin (2–4  µ m ), a synthetic cytokinin, reversed this effect. Regulation of ion transport by root-tip-synthesized cytokinins is discussed.  相似文献

In tropical lowlands, peatlands are commonly reported from Southeast Asia, and especially Indonesian tropical peatlands are known as considerable C sinks and sources. In contrast, Amazonia has been clearly understudied in this context. In this study, based on field observations from 17 wetland sites in Peruvian lowland Amazonia, we report 0–5.9 m thick peat deposits from 16 sites. Only one of the studied sites did not contain any kind of peat deposit (considering pure peat and clayey peat). Historic yearly peat and C accumulation rates, based on radiocarbon dating of peat samples from five sites, varied from 0.94 ± 0.99 to 4.88 ± 1.65 mm, and from 26 ± 3 to 195 ± 70 g C m−2, respectively. The long-term apparent peat and C accumulation rates varied from 1.69 ± 0.03 to 2.56 ± 0.12 mm yr−1, and from 39 ± 10 to 85 ± 30 g C m−2 yr−1, respectively. These accumulation rates are comparable to those determined in the Indonesian tropical peatlands. Under altered conditions, Indonesian peatlands can release globally relevant amounts of C to the atmosphere. Considering the estimated total area of Amazonian peatlands (150 000 km2) close to that of the Indonesian ones (200 728 km2) as well as several factors threatening the Amazonian peatlands, we suggest that the total C stocks and fluxes associated with Amazonian peatlands may be of global significance.  相似文献

Radio‐tagged adult grayling Thymallus thymallus ( n  = 22), monitored from mid August to mid December 1999 in the River Kuusinkijoki, Finland, shifted by the end of September (water temperature 10·0–14·5° C) from riffle sites to deeper and slower pool sites 0·7–1·6 km up‐ or downstream. In early winter ( c . 0° C water temperature), eight of 13 fish still under study made a further shift into new pool sites, possibly triggered by ice formation. The summer range of grayling in the riffles was smaller (mean ±  s . d . length: 75 ± 146 m) than the autumn range (99 ± 46 m) in the pools, but gross daily movements were equally short in both the seasons (18 ± 34 m and 15 ± 7 m, respectively). In late summer, adult grayling preferred water depths 80–120 cm and mean velocities >40 cm s−1. In autumn, the preferred ranges were 100–240 cm and <30 cm s−1, respectively. Substratum was mainly boulders in the summer sites, and gravel or pebbles in the autumn sites.  相似文献

Saithe Pollachius virens , tracked diurnally with a split-beam echosounder, showed no relationship between size and swimming speed. The average and the median swimming speeds were 1·05 m s−1(±0·09 m s−1) and 0·93 m s−1, respectively. However, ping-to-ping speeds up to 3·34 m s−1 were measured for 25–29 cm fish, whose swimming speeds were significantly higher at night (1·08 m s−1) than during the day (0·72 m s−1). The high average swimming speed could be related to the foraging or streaming part of the population and not to potential weakness of the methodology. However, the uncertainty of target location increased with depth and resulted in calculated average swimming speeds of 0·15 m s−1 even for a stationary target. With increasing swimming speed the average error decreased to 0 m s−1 for speeds >0·34 m s−1. Species identity was verified by trawling in a pelagic layer and on the bottom.  相似文献

The impact of viruses and protists on bacterioplankton mortality was examined monthly during 2 years (May 2005–April 2007) in an oligotrophic coastal environment (NW Mediterranean Sea). We expected that in such type of system, (i) bacterial losses would be caused mainly by protists, and (ii) lysogeny would be an important type of virus–host interaction. During the study period, viruses and grazers together were responsible for 50.6 ± 40.1% day−1 of bacterial standing stock losses (BSS) and 59.7 ± 44.0% day−1 of bacterial production losses (BP). Over the first year (May 2005–April 2006), protists were the principal cause of bacterial mortality, removing 29.9 ± 20.4% day−1 of BSS and 33.9 ± 24.3% day−1 of BP, whereas viral lysis removed 13.5 ± 17.0% day−1 of BSS and 12.3 ± 12.3% day−1 of BP. During the second year (May 2006–April 2007), viruses caused comparable bacterial losses (29.2 ± 14.8% day−1 of BSS and 40.9 ± 20.7% day−1 of BP) to protists (28.6 ± 25.5% day−1 of BSS and 32.4 ± 20.0% day−1 of BP). In 37% of cases higher losses of BP due to viruses than due to protists were found. Lysogenic infection was detected in 11 of 24 samplings. Contrary to our expectations, lytic infections dominated over the two years, and viruses resulted to be a significant source of bacterial mortality in this oligotrophic site.  相似文献

Abstract: The reaction of muscimol as amino donor substrate for GABA transaminase (GABA-T) has been studied using enzyme purified from rabbit brain. Enzyme activity was assayed by measuring the glutamate produced using glutamate dehydrogenase. Kinetic parameters determined at 37°C were for GABA, K m (app) = 1.92 ± 0.24 m M , specific activity = 7.33 ± 0.27 μmol/min/mg ( k cat= 13.7s−1), and for muscimol, K m (app) = 1.27 ± 0.15 m M , specific activity = 0.101 ± 0.009 μmol/min/mg ( k cat= 0.19s−1). Addition of muscimol to the enzyme caused the spectral changes associated with conversion of the pyridoxaldimine form to the pyridoxamine form, and the first-order rate constant for the reaction showed a dependence on muscimol concentration that followed saturation kinetics, with a K = 1.1 ±0.18 m M and k max= 0.065 ± 0.004 s−1 (19°C). The rate of spectral change observed on addition of muscimol to ornithine transaminase was extremely slow—at least an order of magnitude slower than that seen with GABA-T.  相似文献

The prolonged swimming speed and metabolic rate of 0+ year Arctic grayling Thymallus articus were examined with respect to current velocity, water temperature and fish size, and compared to conditions fish occupy in the river. Oxygen consumption (mg O2 h−1) increased with fish mass and temperature (6–23° C), with a steep increase in metabolic rate between 12 and 16° C. Absolute prolonged swimming speed (cm s−1) increased rapidly with fish size (total length, L T, and mass), however, fish in the natural stream habitat occupied current velocities between 15 and 25 cm s−1 or 4  L T s−1, approximately half their potential prolonged swimming speed (10  L T s−1).  相似文献

Membrane-bound [NiFe]-hydrogenase from Hydrogenophaga sp. AH-24 was purified to homogeneity. The molecular weight was estimated as 100±10 kDa, consisting of two different subunits (62 and 37 kDa). The optimal pH values for H2 oxidation and evolution were 8.0 and 4.0, respectively, and the activity ratio (H2 oxidation/H2 evolution) was 1.61 × 102 at pH 7.0. The optimal temperature was 75 °C. The enzyme was quite stable under air atmosphere (the half-life of activity was c . 48 h at 4 °C), which should be important to function in the aerobic habitat of the strain. The enzyme showed high thermal stability under anaerobic conditions, which retained full activity for over 5 h at 50 °C. The activity increased up to 2.5-fold during incubation at 50 °C under H2. Using methylene blue as an electron acceptor, the kinetic constants of the purified membrane-bound homogenase (MBH) were V max=336 U mg−1, k cat=560 s−1, and k cat/ K m=2.24 × 107 M−1 s−1. The MBH exhibited prominent electron paramagnetic resonance signals originating from [3Fe–4S]+ and [4Fe–4S]+ clusters. On the other hand, signals originating from Ni of the active center were very weak, as observed in other oxygen-stable hydrogenases from aerobic H2-oxidizing bacteria. This is the first report of catalytic and biochemical characterization of the respiratory MBH from Hydrogenophaga .  相似文献
