The antimicrobial properties of acetone, methanol, and aqueous extracts of the lichens Lasallia pustulata, Parmelia sulcata, Umbilicaria crustulosa, and Umbilicaria cylindrica were studied comparatively in vitro. Antimicrobial activities of the extracts of different lichens were estimated by the disk diffusion test for Gram-positive bacteria, Gram-negative bacteria, and fungal organisms, as well as by determining the MIC (minimal inhibitory concentration). The obtained results showed that the acetone and methanol extracts of Lasallia pustulata, Parmelia sulcata, and Umbilicaria crustulosa manifest antibacterial activity against the majority of species of bacteria tested, in addition to selective antifungal activity. The MIC of lichen extracts was lowest (0.78 mg/ml) for the acetone extract of Lasallia pustulata against Bacillus mycoides. Aqueous extracts of all of the tested lichens were inactive. Extracts of the lichen Umbilicaria cylindrica manifested the weakest activity, inhibiting only three of the tested organisms.  相似文献

It has been suggested that the host specificity exhibited by some lichenicolous fungi depends on their ability to tolerate the secondary chemistry of potential host lichens. For example, the lichen parasite Marchandiomyces corallinus is able to degrade the tissues of the lichen Flavoparmelia baltimorensis irrespective of the presence or absence of endogenous phenolic compounds. In contrast, the degradation of tissues from the lichen Lasallia papulosa is suppressed when endogenous phenolics are not removed. We have investigated the physiological basis of this inhibition in order to understand more about how lichen chemistry influences host preference in lichenicolous fungi. Results showed that the secondary compounds from L. papulosa inhibit the overall growth of M. corallinus , but not the catalytic activity of its tissue-degrading polysaccharidases. This effect is different from that shown by another lichen parasite, Nectria parmeliae , where lichen compounds specifically inhibited polysaccharidase activity. Compared with the compounds of L. papulosa , the endogenous phenolics of F. baltimorensis inhibited the growth of M. corallinus substantially less and exhibited little or no inhibition of polysaccharidases. For M. corallinus , host preference appears to be associated with physiological adaptation to the chemistry of F. baltimorensis .  相似文献

Abstract:Four anthraquinones were isolated from the foliose lichen, Lasallia papulosa (Ach.) Llano. Two of the anthraquinones are known compounds, previously isolated from Lasallia papulosa, while the other two were reported previously as secondary metabolites from laboratory-cultured Nephroma laevigatum, and are isolated here for the first time from lichens in their natural habitat. All compounds were characterized by UV spectrophotometry, mass spectrometry, 1H NMR and 13C NMR. The products were identified as 7-chloroemodin, valsarin (7-chloro-5-hydroxyemodin), 5-chloro-1- O -methylemodin and 5-chloro-1- O -methyl-ω-hydroxyemodin.  相似文献

The lichenicolous basidiomycete Marchandiomyces corallinus is widely distributed in North America and Europe, where it commonly is found on a variety of lichens. Theoretically either of these characteristics, a wide geographic range or generalized host ecology, could provide opportunities for genetic differentiation within this species. To determine how genetic variation is partitioned in M. corallinus, 12 fungal isolates were obtained from locations in North America and Europe; at two locations, in Washington County, Maine, and on the Isle of Mull in Scotland, fungi also were isolated from different lichen hosts. Vegetative mycelial compatibility tests were used to determine compatibility groupings from among the isolates; in addition, several PCR amplification products (RAPD, nuITS rDNA) were obtained for each isolate. A number of distinct compatibility groups were recognizable based on geography, not host ecology. In addition compatible isolates always were restricted to either North America or Europe. However RAPD markers indicated that compatible isolates are not always genetically identical. The presence of sequence heterozygosity at specific positions indicated that the isolates are heterokaryotic and a number of distinct haplotypes could be identified based on ITS variation at three separate locations. This type of genetic variation in these fungi suggests that sexual recombination is possible and that genetic differentiation has taken place recently as a result of geographic isolation, not host switching.  相似文献

The large distributional areas and ecological niches of many lichenized fungi may in part be due to the plasticity in interactions between the fungus (mycobiont) and its algal or cyanobacterial partners (photobionts). On the one hand, broad‐scale phylogenetic analyses show that partner compatibility in lichens is rather constrained and shaped by reciprocal selection pressures and codiversification independent of ecological drivers. On the other hand, sub‐species‐level associations among lichen symbionts appear to be environmentally structured rather than phylogenetically constrained. In particular, switching between photobiont ecotypes with distinct environmental preferences has been hypothesized as an adaptive strategy for lichen‐forming fungi to broaden their ecological niche. The extent and direction of photobiont‐mediated range expansions in lichens, however, have not been examined comprehensively at a broad geographic scale. Here we investigate the population genetic structure of Lasallia pustulata symbionts at sub‐species‐level resolution across the mycobiont's Europe‐wide range, using fungal MCM7 and algal ITS rDNA sequence markers. We show that variance in occurrence probabilities in the geographic distribution of genetic diversity in mycobiont‐photobiont interactions is closely related to changes in climatic niches. Quantification of niche extent and overlap based on species distribution modeling and construction of Hutchinsonian climatic hypervolumes revealed that combinations of fungal–algal interactions change at the sub‐species level along latitudinal temperature gradients and in Mediterranean climate zones. Our study provides evidence for symbiont‐mediated niche expansion in lichens. We discuss our results in the light of symbiont polymorphism and partner switching as potential mechanisms of environmental adaptation and niche evolution in mutualisms.  相似文献

Aim:  The aim of this study was to investigate the nature and bioactivities of endolichenic fungi in three abundant lichens, Pseudocyphellaria sp., Usnea sp. and Parmotrema sp. in the lower elevation of Hakgala montane forest in Sri Lanka.
Methods and Results:  Endolichenic fungal strains, fungi that live asymptomatically in the lichen thallus, much the same way as endophytic fungi live within healthy plant tissues, were isolated from three abundant lichen species, Pseudocyphellaria sp., Usnea sp. and Parmotrema sp., at Hakgala montane forest in Sri Lanka, using the surface sterilization method. Nine endolichenic fungal strains were isolated from Parmotrema sp. and Usnea sp. separately, while 11 endolichenic fungi were recovered from the lichen Pseudocyphellaria sp. Isolation of endolichenic fungus Chrysosporium sp. 2 was common to all three lichen species. Substrate utilization patterns and antifungal activities of eight endolichenic fungal species were evaluated and the results revealed that all the test fungi were able to produce at least one enzyme to utilize the test substrates. Nigrospora sp., Chrysosporium sp. 1 and 2 and Cladosporium sp. showed antifungal activities on growth of some selected plant pathogenic fungi.
Conclusions:  Endolichenic fungal strains (29) were isolated from the lichens Parmotrema sp., Usnea sp. and Pseudocyphellaria sp. in Sri Lanka. Chrysosporium sp. 2 was common in all three lichens. Some of these endolichenic fungal strains showed antifungal activities against common plant pathogenic fungi and they are capable of utilizing the substrates by producing specific enzymes.
Significance and Impact of the Study:  The diversity and prevalence of the endolichenic fungi have not been studied extensively and this is the first report of isolation and identification of endolichenic fungi in lichens available in Sri Lanka.  相似文献

Lichens from the genus Umbilicaria were collected across a 5,000-km transect through Antarctica and investigated for DNA sequence polymorphism in a region of 480-660 bp of the nuclear internal transcribed spacer region of ribosomal DNA. Sequences from both fungal (16 ascomycetes) and photosynthetic partners (22 chlorophytes from the genus Trebouxia) were determined and compared with homologs from lichens inhabiting more temperate, continental climates. The phylogenetic analyses reveal that Antarctic lichens have colonized their current habitats both through multiple independent colonization events from temperate embarkation zones and through recent long-range dispersal in the Antarctic of successful preexisting colonizers. Furthermore, the results suggest that relichenization-de novo establishment of the fungus-photosynthesizer symbiosis from nonlichenized algal and fungal cells-has occurred during the process of Antarctic lichen dispersal. Independent dispersal of algal and fungal cultures therefore can lead to a successful establishment of the lichen symbiosis even under harsh Antarctic conditions.  相似文献

Lichenized and non-lichenized fungi produce a wide range of secondary metabolites. So far, type I polyketide synthases (PKSs) are the suggested catalysts for the biosynthesis of lichen compounds. We were interested whether lichen mycobionts also contain type III PKSs, representing a class that was only recently discovered in fungi. With an alignment of known type III CHS-like genes we applied the CODEHOP strategy to design degenerate PCR primers. We further screened available fungal genomes for type III PKS genes and aligned these sequences for a phylogenetic analysis. Type III-like genes from lichen mycobionts are closely related to those known from non-lichenized fungi, but not to those of bacteria and/or plants. We conclude that type III PKS genes are ubiquitous in fungi. They are present in diverse unrelated lichen mycobionts, but their function in lichens is so far unclear.  相似文献

Lichens are fungal and algal/cyanobacterial symbioses resulting in the production of specific metabolites. Some of these are forming an available biomass for phytochemical investigations, including the assessment of biological activities of the isolated compounds. The alpine or polar region are characterised by highly stressful environmental conditions for many organisms, but lichens are among the dominating organisms in these habitats. In the performant mutual protective system, lichen fungi often accumulate high amounts of metabolites with specific physicochemical properties (UV absorbents, hydrophobicity) which help the lichens to survive. Unique secondary metabolites and polysaccharides have been isolated and tested from these organisms. Even though this has been tested until now only with a low number of compounds so far, interesting activities have been recorded. We review here some of the antimicrobial, anti-inflammatory, antiproliferative and antioxidant activities properties described. Solutions with axenic biotechnological cultivation of each symbiotic partner and particularly the mycobiont to obtain the lichen secondary metabolites are challenging to overcome the limitations for the supply of these rare compounds. Additionally, these lichens appear to harbour a diversity of culturable microorganisms from which active compounds have also been isolated recently.  相似文献

Hobsonia christiansenii B. L. Brady & D. Hawksw. is a helicosporous hyphomycete reported to parasitize a number of lichen species. Quantitative study of a lichen community in Montgomery County, Maryland, revealed that H. christiansenii colonized one lichen species, Flavoparmelia baltimorensis, almost exclusively. This pattern of attack was unexpected because F. baltimorensis produces an array of phenolic secondary metabolites known to inhibit generalist herbivores and microorganisms. The chemical ecology of the pathogen was investigated further in a series of experiments designed to determine if acetone-soluble compounds from various lichens (including F. baltimorensis) inhibited growth of H. christiansenii under controlled laboratory conditions. Results demonstrated that acetone extracts of lichens other than F. baltimorensis were frequently quite inhibitory to the growth of H. christiansenii in culture, but that those of F. baltimorensis had no significant effect on the growth of the pathogen. Indeed, acetone extracts of F. baltimorensis stimulated the growth of Hobsonia on artificial media. These observations suggest H. christiansenii is a relatively specialized lichen pathogen that has been able to overcome the chemical defenses of F. baltimorensis.  相似文献

Individual variation of synthesis level is also observed among plants of a same line. Defence reactions are triggered by pre-inoculation of an avirulent strain of Fusarium oxysporum. Similar results are obtained by application of analogs of fungal elicitors like arachidonic acid. Quite the contrary, treatment of plants with a competitive inhibitor of PAL downs natural barriers and in a same way the effects of cross protection and arachidonic treatment. Seven phenolic compounds, mainly benzoic acid derivatives, inhibit in vitro the growth of pathogen like spores germination. The variability of the host reaction and its stimulation by elicitors could be used to improve the resistance.  相似文献

Endolichenic fungi live in close association with algal photobionts inside asymptomatic lichen thalli and resemble fungal endophytes of plants in terms of taxonomy, diversity, transmission mode, and evolutionary history. This similarity has led to uncertainty regarding the distinctiveness of endolichenic fungi compared with endophytes. Here, we evaluate whether these fungi represent distinct ecological guilds or a single guild of flexible symbiotrophs capable of colonizing plants or lichens indiscriminately. Culturable fungi were sampled exhaustively from replicate sets of phylogenetically diverse plants and lichens in three microsites in a montane forest in southeastern Arizona (USA). Intensive sampling combined with a small spatial scale permitted us to decouple spatial heterogeneity from host association and to sample communities from living leaves, dead leaves, and lichen thalli to statistical completion. Characterization using data from the nuclear ribosomal internal transcribed spacer and partial large subunit (ITS-LSU rDNA) provided a first estimation of host and substrate use for 960 isolates representing five classes and approximately 16 orders, 32 families, and 65 genera of Pezizomycotina. We found that fungal communities differ at a broad taxonomic level as a function of the phylogenetic placement of their plant or lichen hosts. Endolichenic fungal assemblages differed as a function of lichen taxonomy, rather than substrate, growth form, or photobiont. In plants, fungal communities were structured more by plant lineage than by the living vs. senescent status of the leaf. We found no evidence that endolichenic fungi are saprotrophic fungi that have been “entrapped” by lichen thalli. Instead, our study reveals the distinctiveness of endolichenic communities relative to those in living and dead plant tissues, with one notable exception: we identify, for the first time, an ecologically flexible group of symbionts that occurs both as endolichenic fungi and as endophytes of mosses.  相似文献

Aims The biochemical defense of lichens against herbivores and its relationship to lichen frequency are poorly understood. Therefore, we tested whether chemical compounds in lichens act as feeding defense or rather as stimulus for snail herbivory among lichens and whether experimental feeding by snails is related to lichen frequency in the field.Methods In a no-choice feeding experiment, we fed 24 lichen species to snails of two taxa from the Clausilidae and Enidae families and compared untreated lichens and lichens with compounds removed by acetone rinsing. Then, we related experimental lichen consumption with the frequency of lichen species among 158 forest plots in the field (Schw?bische Alb, Germany), where we had also sampled snail and lichen species.Important findings In five lichen species, snails preferred treated samples over untreated controls, indicating chemical feeding defense, and vice versa in two species, indicating chemical feeding stimulus. Interestingly, compared with less frequent lichen species, snails consumed more of untreated and less of treated samples of more frequent lichen species. Removing one outlier species resulted in the loss of a significant positive relationship when untreated samples were analyzed separately. However, the interaction between treatment and lichen frequency remained significant when excluding single species or including snail genus instead of taxa, indicating that our results were robust and that lumping the species to two taxa was justified. Our results imply lichen-feeding snails to prefer frequent lichens and avoid less frequent ones because of secondary compound recognition. This supports the idea that consumers adapt to the most abundant food source.  相似文献

As self‐supporting and long‐living symbiotic structures, lichens provide a habitat for many other organisms beside the traditionally considered lichen symbionts—the myco‐ and the photobionts. The lichen‐inhabiting fungi either develop diagnostic phenotypes or occur asymptomatically. Because the degree of specificity towards the lichen host is poorly known, we studied the diversity of these fungi among neighbouring lichens on rocks in an alpine habitat. Using a sequencing metabarcoding approach, we show that lichen mycobiomes clearly reflect the overlap of multiple ecological sets of taxa, which differ in their trophic association with lichen thalli. The lack of specificity to the lichen mycobiome is further supported by the lack of community structure observed using clustering and ordination methods. The communities encountered across samples largely result from the subsampling of a shared species pool, in which we identify three major ecological components: (i) a generalist environmental pool, (ii) a lichenicolous/endolichenic pool and (iii) a pool of transient species. These taxa majorly belong to the fungal classes Dothideomycetes, Eurotiomycetes and Tremellomycetes with close relatives in adjacent ecological niches. We found no significant evidence that the phenotypically recognized lichenicolous fungi influence the occurrence of the other asymptomatic fungi in the host thalli. We claim that lichens work as suboptimal habitats or as a complex spore and mycelium bank, which modulate and allow the regeneration of local fungal communities. By performing an approach that minimizes ambiguities in the taxonomic assignments of fungi, we present how lichen mycobiomes are also suitable targets for improving bioinformatic analyses of fungal metabarcoding.  相似文献

The ecology of many tropical rain forest organisms, not the least in Africa, remains poorly understood. Here, we present a detailed ecological study of epiphytic lichens in the equatorial montane rain forest of Bwindi National Park (331 km2), Uganda. We evaluated all major lichen growth forms, including selected groups of crustose lichens. In 14 transects at elevations of 1290 m to 2500 m, we sampled 276 trees belonging to 60 species. We recorded all lichen species on each tree trunk between ground level and 2 m above the ground, yielding 191 lichen species in 67 genera, with a mean of 4.7 species per tree. We used non‐metric multi‐dimensional scaling to separate epiphytic lichen assemblages according to tree species composition and elevation. Structural equation modeling indicated that elevation influenced tree species composition and that tree species composition largely determined lichen species composition. Thus, elevation acted indirectly on the lichen assemblages. Further studies examining factors such as bark properties and lichen colonization ecology may clarify what determines the association between tree species and lichen assemblages. The link between lichen assemblages and large‐scale elevation patterns, as well as disturbance and regrowth histories, warrants further study. An analysis of lichen species composition on individual tree species that occur over large elevation ranges would distinguish the effect of tree species on lichen assemblages from the effect of elevation and thus climate. Our study highlights the limited extent of our knowledge of tropical epiphytic lichens.  相似文献

Bacillus amyloliquefaciens NJN-6 produces volatile compounds (VOCs) that inhibit the growth and spore germination of Fusarium oxysporum f. sp. cubense. Among the total of 36 volatile compounds detected, 11 compounds completely inhibited fungal growth. The antifungal activity of these compounds suggested that VOCs can play important roles over short and long distances in the suppression of Fusarium oxysporum.  相似文献

Three long-term studies of lichen growth and colonization have been undertaken at Signy Island, South Orkney Islands, in the maritime Antarctic. Small individual thalli of several crustose species and uncolonized plots on 12 fresh rock surfaces were photographically monitored at intervals of 3–4 years over a period of up to 20 years. The development of Ochrolechia frigida colonies on a regenerating moss bank, recently uncovered by a receding glacier, was similarly monitored. The results indicate that many lichens growing in sites enriched by nitrogenous compounds derived from populations of sea birds, have relatively rapid colonization and growth rates. Mean percentage increase in thallus area can be as high as 15–32% per annum in some nitrophilous saxicolous species (e.g. Acarospora macrocyclos, Xanthoria elegans and species of Buellia and Caloplaca), but as low as 0·4–4% in nitrophobous species (Lecanora physciella, Lecidea sp., Rhizocarpon geographicum). Umbilicaria antarctica and Usnea antarctica also yielded data indicating high growth rates, with colonist plants reaching several centimetres after 20 years. Colonization by mixed assemblages of lichens of new rock surfaces can attain 40→90% cover after 20 years in nutrient-enriched sites, and even 20–25% in non-biotically influenced sites. Colonization by or increase in extant O. frigida on the regenerating moribund moss bank was also quite rapid. It is suggested that the ~exceptionally large thalli of several lichen species and the locally extensive dense lichen fellfield communities in the maritime Antarctic may be much younger than previously supposed.  相似文献

Some of the principal advances in our knowledge of lichens made since the time of E. M. Holmes are briefly reviewed. Floristic studies on British lichens have continued and distribution maps of particular species are now being produced. These studies have shown that many species have disappeared from areas affected by air pollution by sulphur dioxide. Lichen taxonomy has seen the acceptance of chemical characters as valid taxonomic criteria, and the realization that ascus types, ascocarp structure and ascocarp ontogeny are more important at the generic level than ascospore characters alone. Lichens are not now thought of as a single taxonomic group but as fungi related to various orders of non-lichenized Ascomycotina, united only in their common method of nutrition (symbiosis with algae); some lichenized and non-lichenized species with identical types of ascocarps are consequently being treated as members of the same genus. Considerable progress has been made in describing lichen communities characteristic of particular substrates and situations and in understanding the factors which may affect the occurrence of particular species. Lichens have proved to be particularly valuable indicators of mean air pollution levels and scales for the estimation of these levels based solely on the lichen vegetation have been devised. Physiological and ultrastructural studies on lichens have progressed rapidly in the last decade. Carbohydrates have been demonstrated to move from the algal to the fungal component and the mobile carbohydrates found to differ in lichens with different genera of algae. The algal and fungal partners have been isolated and studied in pure culture. Most attempts to synthesize lichens have met with little success but in 1970 a thallus was formed from its components in culture. It is now possible to grow some lichens under controlled conditions in the laboratory and these techniques, together with transplant experiments, will clearly assume considerable importance in the investigation of taxonomic and ecological problems in the future. Little progress has been made in the study of lichen cytology or the factors affecting the establishment of lichen thalli. Lichens constitute an important part of some ecosystems and their decline due to anthropogenic influences is consequently a matter of concern for all biologists.  相似文献

地衣是一种传统的民族药物,能产生多种具有活性的物质。该研究对地衣型真菌(Xanthoria elegans,Myelochroa indica,Ramalina peruviana,Cladonia macilenta,Nephromopsis pallescens,Cladonia coccifera)进行液体培养,2个月后,培养液用乙酸乙酯萃取后获得初提物。该研究采用抑菌圈法评价地衣型真菌初提物对7种致病细菌(Bacillus subtilis,Bacillus cereus,Vibrio parahaemolyticus,Straphylococcus haemolyticus,Pseudomonas aeruginosa,Staphylococcus aureus,Micrococcus luteus)的抗菌活性,并测定最低抑菌浓度(MIC)。结果表明:6种地衣型真菌的初提物均具有一定的抗菌活性,且不同培养基对地衣型真菌产生抗菌物质有显著影响。其中,R.peruviana在MY液体培养基中所产生的次级代谢产物对金黄色葡萄球菌、藤黄微球菌、溶血性葡萄球菌、铜尿假单胞菌具有抑制效果,但在YMG培养基中所得初提物对供试7种致病细菌不具有抑菌效果。X.elegans在YMG培养基中所得初提物对枯草芽孢杆菌具有明显抗菌活性,其抑菌圈直径可达17.77 mm。该研究证实不同地衣型真菌液体培养初提物具有抗菌活性,不同的培养基也直接影响地衣型真菌抗菌效果。该研究结果为地衣型真菌的进一步研究及民族药的开发利用奠定了基础。  相似文献
