花生幼胚在含2,4-D的诱导培养基上,形成近球状的致密的胚性愈伤组织、杆状两极结构及子叶期体胚。继代培养也有体胚发生。光照明显抑制体胚发生类似于自然栽培的情况。成熟培养基中诱导体胚根、芽两极发育完全。光下,具有根、芽的体胚于再生培养基中长成小植株后移载于盛沙土的盆中正常生长、结实。在较好的影响因素(光照、幼胚长度、激素、切分方式、接种密度)组合下,体胚发生频率达75%以上,每子叶形成体胚3个以上。  相似文献

分别以完整成熟胚、切去一个子叶的成熟胚和切下的子叶为外植体,以MS为基本诱导培养基、1/2MS为基本分化培养基,进行了花楸体细胞胚胎发生研究。结果表明:以完整合子胚作为外植体的体胚诱导率最高,为100%,最佳植物生长调节剂组合为5 mg.L-1NAA+2 mg.L-16-BA;NAA和6-BA浓度及二者的交互作用对愈伤组织和体胚诱导率的影响极显著;光照配合延长继代间隔时间有利于体胚发生。实体观察结果表明,花楸体胚发生方式有直接发生和间接发生两种;体胚发育经历了球形期、心形期、鱼雷形期和子叶期。组织学观察结果表明,体胚具有两极性,子叶期体胚结构完整。  相似文献

在含有2,4-D的培养基上所诱导和增殖的枳棋体细胞胚只能形成无再生能力的畸形胚。通过次生胚发生途径可以在不加植物生长调节物质的培养基诱导出发育正常的体细胞胚,子叶期胚的次生胚诱导能力高于早期体胚。相对高的温度(30℃)有利于次生胚的诱导,相对低的温度(20℃)更有利于次生胚的发育、植株再生和移栽成活。  相似文献

以橡胶树未成苗体细胞胚为试材,选取未成苗双子叶胚、连体胚、多子叶胚、单子叶胚和子叶愈伤化胚作为外植体,研究不同体细胞胚类型的外植体分化茎芽的情况,并利用其进行幼态微型芽条的培育研究。结果表明:在植株诱导培养基中培养80d后,未成苗双子叶胚、连体胚、多子叶胚、单子叶胚、子叶愈伤化胚均能分化出芽,双子叶胚分化率最高(达90%),单子叶胚最低(20%),多子叶胚和连体胚最多可分化出芽3个,双子叶胚2个。幼态微型芽条增殖的最优培养基为MS+6-BA2mg·L-1或MS+6-BA2mg·L-1+KT1mg·L-1,而添加NAA对芽的伸长有抑制作用;适宜幼态微型芽条伸长培养的基本培养基为MS。  相似文献

花生实生苗幼叶接种于MS TDZ 0.2mg/L NAA0.4mg/L诱导培养基上经诱导培养,继而转移到无激素培养基MS可获得不定芽和体细胞胚。组织学观察表明,花生不定芽和体细胞胚均起源于愈伤组织表层,不定芽为多细胞起源,而体细胞胚起源于单个胚性原始细胞。体细胞胚的发育经历多细胞原胚、球形胚、心形胚、鱼雷胚和子叶胚等时期发育成小植株。  相似文献

由6个苹果品种和1个梨品种的幼胚下胚轴及子叶基部直接诱导出不定芽。不定芽的发生是成从的。实验使用MS作基础培养基,其中附加细胞分裂素和生长素的浓度为BAP 1—3ppm,NAA 0.5ppm。但是,我们在进一步实验中表明,在上述培养基中除去了生长素亦得到同样结果。在含有0.5ppm IBA的土豆培养基上,离体诱导下胚轴及子叶基部处在发育中的不定芽生根,成长为完整植株。这一结果为克服果树及农作物杂交育种中常见的幼胚不育和扩大繁殖优良品种的无性系植株,提供一个可能的实用方法。  相似文献

以欧石楠茎段为外植体,研究其体细胞胚胎发生和植株再生。对影响茎段不定芽分化及胚性愈伤组织诱导的主导因子进行比较分析,并研究其体胚萌发、生根及移栽;同时,采用树脂切片法对茎段脱分化产生胚性愈伤组织及体胚发育过程进行组织细胞学观察。结果表明,接种在1/2WPM基本培养基上的茎段,胚性愈伤组织诱导率为88.7%,显著高于其他处理,不定芽诱导率可达90.6%,平均分化倍数为3.6个,平均分化苗高3.82cm;体细胞经过成熟培养后。在添加1.0mg·L-1 ZT和0.3mg·L-1 IBA的1/2WPM培养基上萌发,萌发的体胚在I/2WPM附加0.2mg·L-1 NAA和0.3mg·L-1 IBA的培养基上形成完整的体胚苗植株,体胚苗生根率达到87.4%,经炼苗后移栽到蛭石:珍珠岩=3:1(V/V)的栽培基质中,成活率可达63.7%。在显微镜下可观察到球形胚、心形胚、鱼雷形胚和子叶形胚;体细胞胚以间接方式发生,表现为愈伤组织外层细胞直接发生和愈伤组织组织内部细胞发生。  相似文献

研究了金花茶(Camellia chrysantha(Hu)Tuyama)子叶在离体培养中体细胞胚状体发生的条件。在MS基本培养基中附加苄基嘌呤(BA)或苄基嘌呤与萘乙酸(NAA)组合,诱导了胚状体发生。组织学观察表明,胚状体起源于子叶的表皮细胞。在增添细胞分裂素和生长素的MS或改良B_5液体培养基里振荡培养,明显地促进了胚状体根的生长和茎的发育。胚状体在继代培养中能保持旺盛的再生能力。已得到两个繁殖率较高的胚状体无性系。在合适的条件下,胚状体能长成正常的小植株。  相似文献

The study aimed at testing chronotype and gender differences in the time of day when humans feel the greatest need for sex and the time of day they actually undertake sexual activity. A Polish sample of 565 participants aged between 18 and 57 was tested. In females, regardless of chronotype, the greatest need for sex occurred between 18:00 and 24:00, but a secondary peak appeared only in morning types at 6:00–9:00. In males, the greatest need for sex occurred either in the morning or evening hours: in evening types at 9:00–12:00 and 18:00–3:00; in neither types at 6:00–9:00 and 18:00–24:00; in morning types at 6:00–12:00 and 18:00–24:00. Considering time of day when subjects were undertaking sexual activity most frequently, this appeared between 18:00 and 24:00 for all the participants, and prolonged until 3:00 at night in evening type males. Morningness preference was more strongly related to the timing of need for sex than to the timing of actual sexual activity (r?=??0.275 vs. r?=??0.174), while the timing of desire and the timing of sexual activity were positively, but moderately related (r?=?0.320).  相似文献

The debate over whether the medical profession should accommodate its members' conscientious objections (COs) has raged on in the bioethics literature and on legislative floors for decades. Unfortunately, participants on all sides of the debate fail to distinguish among different types of CO, a failure that obstructs the view of which cases warrant accommodation and why. In this paper, we identify one type of CO that warrants consideration for accommodation, called Nature of Medicine COs (NoMCOs). NoMCOs involve the refusal of physicians to perform actions they reasonably judge to be contrary to the nature of medicine and their professional obligations. We argue that accommodating NoMCOs can be justified based on the profession's need to preserve reformability. Importantly, this previously underdeveloped position evades some of the concerns commonly raised by opponents of CO accommodations.  相似文献

X-ray structural analysis provides no quantitative estimate of the relative contribution of specific and nonspecific or strong and weak interactions to the total affinity of enzymes for nucleic acids. We have shown that the interaction between enzymes and long nucleic acids at the molecular level can be successfully analyzed by the method of stepwise increase in ligand complexity (SILC). In the present review we summarize our studies of human uracil DNA glycosylase and apurinic/apyrimidinic endonuclease, E. coli 8-oxoguanine DNA glycosylase and RecA protein using the SILC approach. The relative contribution of structural (X-ray analysis data), thermodynamic, and catalytic factors to the discrimination of specific and nonspecific DNA by these enzymes at the stages of complex formation, the following changes in DNA and enzyme conformations and especially the catalysis of the reactions is discussed.  相似文献

Inosine is an important RNA modification, furthermore RNA oxidation has gained interest due, in part, to its potential role in the development/progression of disease as well as on its impact on RNA structure and function. In this report we established the base pairing abilities of purine nucleobases G, I, A, as well as their corresponding, 8-oxo-7,8-dihydropurine (common products of oxidation at the C8-position of purines), and 8-bromopurine (as probes to explore conformational changes), derivatives, namely 8-oxoG, 8-oxoI, 8-oxoA, 8-BrG, and 8-BrI. Dodecamers of RNA were obtained using standard phosphoramidite chemistry via solid-phase synthesis, and used as models to establish the impact that each of these nucleobases have on the thermal stability of duplexes, when base pairing to canonical and noncanonical nucleobases. Thermal stabilities were obtained from thermal denaturation transition (Tm) measurements, via circular dichroism (CD). The results were then rationalized using models of base pairs between two monomers, via density functional theory (DFT), that allowed us to better understand potential contributions from H-bonding patterns arising from distinct conformations. Overall, some of the important results indicate that: (a) an anti-I:syn-A base pair provides thermal stability, due to the absence of the exocyclic amine; (b) 8-oxoG base pairs like U, and does not induce destabilization within the duplex when compared to the pyrimidine ring; (c) a U:G wobble-pair is only stabilized by G; and (d) 8-oxoA displays an inherited base pairing promiscuity in this sequence context. Gaining a better understanding of how this oxidatively generated lesions potentially base pair with other nucleobases will be useful to predict various biological outcomes, as well as in the design of biomaterials and/or nucleotide derivatives with biological potential.  相似文献

Transglutaminases are a class of transferases known to form isopeptide bond between glutamine and lysine residues in a protein molecule. Increasing demand for transglutaminase in food and other industries and its low productivity have compelled researchers to isolate and screen micro-organisms with potential to produce it. In the present investigation around 200 isolates were screened for extracellular secretion of microbial transglutaminase (MTGase). Isolate B4 showed enzyme activity of 1.71?±?0.2?U/mL followed by isolate C2 which showed 1.61?±?0.17?U/mL activity, comparable with the activity of industrially used microbial strains. Biochemical analysis along with 16S r-RNA sequencing revealed these isolates (B4 and C2) to be Bacillus nakamurai and a variant of Bacillus subtilis, respectively. Amongst the various production media screened, a medium containing starch and peptone was found best for MTGase production. Correlation between growth, enzyme production, and sugar utilization was also studied and maximum enzyme production was obtained after 48 to 60?hr. Highest MTGase titer (3.95?±?0.03?U/mL for B4 and 2.65?±?0.17?U/mL for C2) was obtained by optimization of parameters. The enzyme was characterized for temperature and pH optima, pH and thermal stability, and effect of metal ions, suggesting its potential use in future applications.  相似文献

We designed, built or 3D printed, and screened tubular reactors that minimize axial dispersion to serve as incubation chambers for continuous virus inactivation of biological products. Empirical residence time distribution data were used to derive each tubular design's volume equivalent to a theoretical plate (VETP) values at a various process flow rates. One design, the Jig in a Box (JIB), yielded the lowest VETP, indicating optimal radial mixing and minimal axial dispersion. A minimum residence time (MRT) approach was employed, where the MRT is the minimum time the product spends in the tubular reactor. This incubation time is typically 60 minutes in a batch process. We provide recommendations for combinations of flow rates and device dimensions for operation of the JIB connected in series that will meet a 60‐min MRT. The results show that under a wide range of flow rates and corresponding volumes, it takes 75 ± 3 min for 99% of the product to exit the reactor while meeting the 60‐min MRT criterion and fulfilling the constraint of keeping a differential pressure drop under 5 psi. Under these conditions, the VETP increases slightly from 3 to 5 mL though the number of theoretical plates stays constant at about 1326 ± 88. We also demonstrated that the final design volume was only 6% ± 1% larger than the ideal plug flow volume. Using such a device would enable continuous viral inactivation in a truly continuous process or in the effluent of a batch chromatography column. The aftermath of Hurricane Katrina drew attention to commonplace landscape markers that create for a community a sense of place— that connection between people and places crucial to a sense of corporate and individual identity and heritage. There is a legal context for sense of place within extant federal preservation legislation. Nevertheless, many such markers with special meanings for residents have been overlooked in federal documentation, the cornerstone of which is the National Register of Historic Places. Grassroots efforts and national media coverage have helped forge a niche for sense of place within the recovery plans and policy emerging in the affected region. However, it is unclear whether this will carry over into practice. In terms of long-term policy shifts, remedying the shortcomings highlighted by Katrina may require changes to the National Historic Preservation Act and its associated guidelines and regulations, or it may entail a new approach altogether.  相似文献

Most of the endogenous free d-serine (about 90%) in the brain is produced by serine racemase (SR). d-Serine in the brain is involved in neurodegenerative disorders and epileptic states as an endogenous co-agonist of the NMDA-type glutamate receptor. Thus, SR inhibitors are expected to be novel therapeutic candidates for the treatment of these disorders. In this study, we solved the crystal structure of wild-type SR, and tried to identify a new inhibitor of SR by in silico screening using the structural information. As a result, we identified two hit compounds by their in vitro evaluations using wild-type SR.Based on the structure of the more potent hit compound 1, we synthesized 15 derivatives and evaluated their inhibitory activities against wild-type SR. Among them, the compound 9C showed relatively high inhibitory potency for wild-type SR. Compound 9C was a more potent inhibitor than compound 24, which was synthesized by our group based upon the structural information of the mutant-type SR.  相似文献

The quantity and quality of freshwater resources are now being seriously threatened, partly as a result of extensive worldwide changes in land use, and scientists are often called upon by policy makers and managers to predict the ecological consequences that these alterations will have for stream ecosystems. The effects of the urbanization of stream ecosystems in the United States over the next 20 years are of particular concern. To address this issue, we present a multidisciplinary research agenda designed to improve our forecasting of the effects of land-use change on stream ecosystems. Currently, there are gaps in both our knowledge and the data that make it difficult to link the disparate models used by economists, hydrologists, geomorphologists, and ecologists. We identify a number of points that practitioners in each discipline were not comfortable compromising on—for example, by assuming an average condition for a given variable. We provide five instructive examples of the limitations to our ability to forecast the fate of stream and riverine ecosystems one drawn from each modeling step: (a) Accurate economic methods to forecast land-use changes over long periods (such as 20 years) are not available, especially not at spatially explicit scales; (b) geographic data are not always available at the appropriate resolution and are not always organized in categories that are hydrologically, ecologically, or economically meaningful; (c) the relationship between low flows and land use is sometimes hard to establish in anthropogenically affected catchments; (d) bed mobility, suspended sediment load, and channel form—all of which are important for ecological communities in streams—are difficult to predict; and (e) species distributions in rivers are not well documented, and the data that do exist are not always publicly available or have not been sampled at accurate scales, making it difficult to model ecological responses to specified levels of environmental change. Meeting these challenges will require both interdisciplinary cooperation and a reviewed commitment to intradisciplinary research in the fields of economics, geography, quantitative spatial analysis, hydrology, geomorphology, and ecology.
