The intake of dietary fructose has undergone a marked increase around the world, especially the developed countries, in recent times. Stevioside, a glycoside contained in the leaves of Stevia rebaudiana Bertoni (Compositae), was used to screen the effect induced by a diet containing 60% fructose on insulin resistance in rats. Single oral administration of stevioside for 90 min decreased plasma glucose concentrations in a dose-dependent manner in rats receiving fructose-rich chow for four weeks. In addition, insulin action on glucose disposal rate was measured using the glucose-insulin index, the product of the areas under the curve of glucose, and insulin during the intraperitoneal glucose tolerance test. Oral administration of stevioside (5.0 mg/kg) in rats given four weeks of fructose-rich chow for 90 min reversed the value of glucose-insulin index, indicating that stevioside has the ability to improve insulin sensitivity in this insulin-resistant animal model. Time for the loss of plasma glucose lowering response to tolbutamide (10.0 mg/kg, i. p.) in fructose-rich chow fed rats was also markedly delayed by repeated stevioside treatment three times daily compared to the vehicle-treated group. The plasma glucose-lowering activity of tolbutamide was introduced to account for varying levels of endogenous insulin secretion, and is widely used as the indicator of insulin resistance development. Thus, it provided the supportive data that repeated oral administration of stevioside delayed the development of insulin resistance in rats on a high-fructose diet. Increased insulin sensitivity by stevioside administration was further identified using the plasma glucose-lowering action of exogenous insulin in streptozotocin-induced diabetic rats (STZ-diabetic rats). Oral administration of stevioside at 0.2 mg/kg three times daily into STZ-diabetic rats for ten days increased the response to exogenous insulin. Taken together, this demonstrated that oral administration of stevioside improves insulin sensitivity, and seems suitable as an adjuvant for diabetic patients and/or those that consume large amounts of fructose.  相似文献

This study was carried out to examine the seasonal difference in the magnitude of the suppression of melatonin secretion induced by exposure to light in the late evening. The study was carried out in Akita (39 degrees North, 140 degrees East), in the northern part of Japan, where the duration of sunshine in winter is the shortest. Ten healthy male university students (mean age: 21.9+/-1.2 yrs) volunteered to participate twice in the study in winter (from January to February) and summer (from June to July) 2004. According to Japanese meteorological data, the duration of sunshine in Akita in the winter (50.5 h/month) is approximately one-third of that in summer (159.7 h/month). Beginning one week prior to the start of the experiment, the level of daily ambient light to which each subject was exposed was recorded every minute using a small light sensor that was attached to the subject's wrist. In the first experiment, saliva samples were collected every hour over a period of 24 h in a dark experimental room (<15 lux) to determine peak salivary melatonin concentration. The second experiment was conducted after the first experiment to determine the percentage of melatonin suppression induced by exposure to light. The starting time of exposure to light was set 2 h before the time of peak salivary melatonin concentration detected in the first experiment. The subjects were exposed to light (1000 lux) for 2 h using white fluorescent lamps (4200 K). The percentage of suppression of melatonin by light was calculated on the basis of the melatonin concentration determined before the start of exposure to light. The percentage of suppression of melatonin 2 h after the start of exposure to light was significantly greater in winter (66.6+/-18.4%) than summer (37.2+/-33.2%), p<0.01). The integrated level of daily ambient light from rising time to bedtime in summer was approximately twice that in winter. The results suggest that the increase in suppression of melatonin by light in winter is caused by less exposure to daily ambient light.  相似文献

The objective of the study was to explore the changes in melatonin and zinc levels in rats with induced hypothyroidism. Thirty adult male rats used in the study were allocated to three groups with equal numbers. Group 1: General control group which was not subjected to any procedure. Group 2: Sham-hypothyroidism group to which was administered 10 mg kg(-1) intraperitoneal (i.p.) physiologic saline (0.09% NaCl) for 4 weeks. Group 3: Hypothyroidism group which was supplemented with intraperitoneal 10 mg kg(-1) propylthiouracil (PTU) for 4 weeks. Blood samples collected from all animals at the end of the study by decapitation were analysed for serum Total T4 (TT4), Total T3 (TT3), Free T4 (FT4), Free T3 (FT3) (ELISA) as well as for melatonin (RIA) hormones and zinc levels (atomic emission). Comparison of the study groups in terms of thyroid hormones, melatonin and zinc levels showed that TT4, TT3, FT4, FT3, melatonin and zinc levels in group 3 were lower than those in groups 1 and 2 (p < 0.01). These parameters were not different in groups 1 and 2. The results of the study demonstrate that PTU supplementation for 4 weeks results in a significant inhibition in both melatonin and zinc levels. Inhibited melatonin levels may result from the decrease in zinc levels.  相似文献

Daily afternoon injections of 25 micrograms melatonin for 12 weeks had no effect on testicular weights of male rats kept in long photoperiod (14L:10D); similarly, exposure of rats to short photoperiod (2L:22D) had no effect on gonadal weight. However, rats maintained in a long or short photoperiod and implanted every 2 weeks with a 15 mm Silastic pellet containing testosterone showed a significant reduction in testicular weight; this effect was more pronounced in rats exposed to a short photoperiod. Melatonin injections in testosterone-treated rats in a long photoperiod exacerbated the inhibitory effects of testosterone alone. Subcutaneous 2-weekly implants of a beeswax pellet containing 1 mg melatonin reversed the effects of the melatonin injections on relative testicular weights but not those due to short photoperiod exposure. Testosterone implants significantly reduced pituitary LH values in long and short photoperiod-exposed animals, more particularly in those exposed to short photoperiod. Melatonin injections alone or in combination with melatonin pellets did not further exaggerate the depression in pituitary LH due to testosterone alone in long photoperiod-exposed animals; similarly melatonin pellets did not reverse the depression in pituitary LH observed. No significant differences in plasma prolactin concentrations or in thyroxine concentrations or free thyroxine index were observed after any combination of treatments. We therefore suggest that the effects observed with short photoperiod may be due to melatonin.  相似文献

Surprisingly, in our modern 24/7 society, there is scant information on the impact of developmental chronodisruption like the one experienced by shift worker pregnant women on fetal and postnatal physiology. There are important differences between the maternal and fetal circadian systems; for instance, the suprachiasmatic nucleus is the master clock in the mother but not in the fetus. Despite this, several tissues/organs display circadian oscillations in the fetus. Our hypothesis is that the maternal plasma melatonin rhythm drives the fetal circadian system, which in turn relies this information to other fetal tissues through corticosterone rhythmic signaling. The present data show that suppression of the maternal plasma melatonin circadian rhythm, secondary to exposure of pregnant rats to constant light along the second half of gestation, had several effects on fetal development. First, it induced intrauterine growth retardation. Second, in the fetal adrenal in vivo it markedly affected the mRNA expression level of clock genes and clock-controlled genes as well as it lowered the content and precluded the rhythm of corticosterone. Third, an altered in vitro fetal adrenal response to ACTH of both, corticosterone production and relative expression of clock genes and steroidogenic genes was observed. All these changes were reversed when the mother received a daily dose of melatonin during the subjective night; supporting a role of melatonin on overall fetal development and pointing to it as a 'time giver' for the fetal adrenal gland. Thus, the present results collectively support that the maternal circadian rhythm of melatonin is a key signal for the generation and/or synchronization of the circadian rhythms in the fetal adrenal gland. In turn, low levels and lack of a circadian rhythm of fetal corticosterone may be responsible of fetal growth restriction; potentially inducing long term effects in the offspring, possibility that warrants further research.  相似文献

Adult male Syrian hamsters either placed in a short photoperiod alone or kept in a long photoperiod and given daily afternoon injections of the pineal indole melatonin (25 micrograms) exhibited splenic hypertrophy and extramedullary hematopoiesis in addition to a marked regression in testicular weight. The testicular regression as well as the changes in spleen weight and histology could be prevented if the animals in short photoperiod were either pinealectomized or implanted subcutaneously with a pellet containing 1 mg melatonin. Female Syrian hamsters given afternoon injections of melatonin for 7 or 12 weeks had ovaries devoid of corpora lutea; additionally, these animals had reduced relative spleen weights compared to the control animals. In conclusion, it is apparent that spleen weight varies with the functional status of the gonads. Splenic hypertrophy accompanied by pineal-induced testicular regression in males may be related to splenic extramedullary hematopoiesis.  相似文献

Adult female albino rats anovulatory as a result of exposure to constant light (CL rats) were shown to ovulate in response to limited coital stimulation without necessarily becoming either pseudopregnant or pregnant. A higher level of coital stimulation would induce pseudopregnancy provided that intromission was not prevented. The occurrence of an ejaculation was not essential for the induction of pseudopregnancy but the incidence of pseudopregnancy was higher when a series of intromissions included one or more intromissions associated with ejaculation than when it did not. Similar findings regarding the induction of pseudopregnancy were obtained in experiments on female rats maintained under normal light-dark conditions (LD rats). Increasing the interval between intromissions from less than 1 min to as much as 20 min did not reduce the incidence of pseudopregnancy in CL rats. Splitting a set number of intromissions into two groups separated by up to 180 min did not reduce the incidence of pseudopregnancy in CL or LD rats. Conditions for the induction of pregnancy were more critical than for the induction of pseudopregnancy. CL rats showed only a low incidence of pregnancy and, if pregnant, either had small litters or did not deliver living pups even when a high level of coital stimulation from several males, including multiple ejaculations, occurred over a limited period of time. A high success rate was only observed when CL rats were mated with a single male overnight. LD rats showed a higher incidence of pregnancy than CL rats in response to a restricted amount of coital stimulation over a short time period, especially when an ejaculation was the terminal event in the mating sequence. This dependence on the occurrence of an ejaculation as the terminal event was not observed in CL rats, probably because, unlike LD rats, their uteri were not distended with fluid at the time of mating.  相似文献

It has been shown in the Syrian hamster that a short photoperiod sensitizes the hypothalamo-hypophyseal axis of castrated animals to the negative feedback effect of testosterone. There is some evidence that even the reproductive system of the rat, which is generally considered not to be very sensitive to light, can respond to changes in illumination. Therefore, we found it of interest to examine whether alterations in lighting conditions produce changes of sensitivity in the negative feedback effect of testosterone in the rat. We kept intact, castrated, and castrated testosterone-treated animals either in periodic (L:D 12:12) or constant light for 7 days starting 4 weeks after castration. In all 3 testosterone-injected groups, serum luteinizing hormone (LH) was lower in constant than in periodic light. Exogenous testosterone did not decrease the castration-induced elevations of pituitary LH and follicle-stimulating hormone (FSH). On the contrary, testosterone increased the pituitary contents of LH and FSH, especially in constant light. We conclude that, in constant light, the hypothalamo-hypophyseal axis of the castrated rat becomes more sensitive to the negative feedback action of testosterone.  相似文献

There is some controversy whether phase response curves constructed from studies conducted after acute release into constant darkness (Type II) or after prolonged constant darkness are comparable. This study investigated the effects of brief low-intensity light pulses on the onset of 6-sulphatoxymelatonin excretion in rats 48 to 60 h after lights-off and after 14 days of continuous darkness. In the former condition, maximum phase delays occurred between 4 and 6 h after expected lights-off, but no phase advances were observed within 2 days of the presentation of the stimulus. When the times of the pulses were plotted in relation to the individual onsets, peak light-induced phase delays occurred 0 to 2 h after melatonin onset. After 14 days in continuous darkness, the peak phase delays also occurred 0 to 2 h after melatonin onset and were slightly but significantly smaller. No significant phase advances were observed. In a separate small series of experiments, the temperature rhythm of rats was shown to be delayed by a comparable degree to that of melatonin by light pulses 2 and 4 h after expected lights-off under the Type II conditions. It is concluded that phase response curves conducted under Type I and Type II conditions are comparable.  相似文献

Immature albino rats were exposed to continuous illumination for 5-93 days and the light induced ultrastructural and electroretinographic changes were studied. Another group was exposed to continuous light for 7-9 days and then kept in complete darkness, or in cyclic light-dark up to 90 days. By comparison with the results obtained in adult animals, lesions appeared faster in the immature group. Tubular transformation of rods, phagocytosis of altered outer and inner segments with resulting changes in retinal organization, synaptic degeneration in the outer plexiform layer, and cell lysis of some photoreceptor cell perikarya are described. ERG recovery, following the period of darkness or cyclic light-dark was only partial, the amplitude of the "b" wave reached only 50-60% of the control preillumination values. However, the fine structure of the recovered outer segments was similar to that found in normal retinae.  相似文献

Summary Immature albino rats were subjected to (a) continuous illumination for 5–9 days, or (b) continuous illumination followed by prolonged darkness. Their electroretinographic responses and the ultrastructural characteristics of the rod outer segments, as revealed by a mixture of zinc iodine-osmium tetroxide (ZIO) at different temperatures, were studied and compared with those of a control group maintained in a cyclic rhythm of light and darkness.Noteworthy differences in the distribution of ZIO reactive sites were observed in the rats exposed for 5–9 days to continuous illumination (no electroretinographic responses) as compared with normal controls. In rams, artificial long days followed by continuous light stimulate testosterone secretion during the non-breeding season. The objective of this study was to determine whether artificial long days followed by continuous light could stimulate testosterone secretion in Alpine bucks as well as in those exposed to long days followed by a melatonin treatment. All bucks were kept in shaded open pens. Control males were exposed to natural photoperiod conditions (n=5). Males of the two experimental groups were exposed to 2.5 months of long days from 1 December (n=5 each). On 16 February, one group of males was exposed to 24 h of light per day until 30 June; the other group was exposed to natural variations of photoperiod and received two s.c. melatonin implants. Testicular weight was determined every 2 weeks, and the plasma testosterone concentrations once a week. In the control and the two photoperiodic-treated groups, a treatment×time interaction was detected for testicular weight and plasma testosterone concentrations (P<0.001). In control bucks, testicular weight increased from January and peaked in June, whereas in both photoperiodic-treated groups, this variable increased from January, but peaked in April, when the values were higher than in controls (P<0.05). In the control group, plasma testosterone concentrations remained low from January to June, whereas in both photoperiodic-treated groups, this variable remained low from January to March; thereafter, these levels increased in both photoperiodic-treated groups, and were higher than controls in April and May (P<0.05). We conclude that continuous light after a long-day treatment stimulate testosterone secretion in Alpine male goats during the non-breeding season as well as the long days followed by a melatonin treatment. Therefore, continuous light could replace the implants of melatonin.  相似文献

Testis histometry and daily sperm production (DSP) were evaluated in adult (160-day-old) Wistar rats exposed to constant light for the first 25 days after birth, and compared with control animals which were exposed to a 12 h-light-12 h-dark light regimen. Significantly greater (P < 0.05) numbers of Sertoli cell nucleoli and round spermatids per cross-section of seminiferous tubule were found in animals exposed to constant light. In addition, epididymis weight, DSP per testis and per gram of testis, as well as Leydig cell compartment volume, were significantly increased in treated animals. Although there was a clear trend toward an increased Sertoli cell population per testis in animals exposed to constant light, this difference was not statistically significant (P < 0.05). The number of round spermatids as expressed per Sertoli cell was the same in both groups. Surprisingly, the diameter and volume of round spermatid nucleus at stages I and VII of the cycle of seminiferous epithelium were significantly lower (P < 0.05) in treated animals. In conclusion, constant illumination during neonatal testis development increased sperm production and Leydig cell compartment volume in adult rats probably through a mechanism involving elevated follicle stimulating hormone and luteinizing hormone during the prepubertal period. To our knowledge, this is the first study showing that altering the light regimen can affect sperm production in non-seasonal breeders.  相似文献

Light is the most important synchronizer of melatonin rhythms in fish. This paper studies the influence of the characteristics of light on plasma melatonin rhythms in sole. The results revealed that under long-term exposure to constant light conditions (LL or DD), the total 24 h melatonin production was significantly higher than under LD, but LL and DD conditions influenced the rhythms differently. Under LL, melatonin remained at around 224 pg/ml throughout the 24 h, while under DD a significant elevation (363.6 pg/ml) was observed around the subjective evening. Exposure to 1 h light pulses at MD (mid-dark) inhibited melatonin production depending on light intensity (3.3, 5.3, 10.3, and 51.9 microW/cm(2)). The light threshold required to reduce nocturnal plasma melatonin to ML (mid-light) values was 5.3 microW/cm(2). Melatonin inhibition by light also depended on the wavelength of the light pulses: while a deep red light (lambda>600 nm) failed to reduce plasma melatonin significantly, far violet light (lambda(max)=368 nm) decreased indoleamine's concentration to ML values. These results suggest that dim light at night (e.g., moonlight) may be perceived and hence affect melatonin rhythms, encouraging synchronization to the lunar cycle. On the other hand, deep red light does not seem to inhibit nocturnal melatonin production, and so it may be used safely during sampling at night.  相似文献

Male rats treated with estradiol dipropionate (250 mug) by subcutaneous injection on Day 5 of postnatal life failed to show maturation changes in the testis and epididymis at puberty. Histological examination at 60 days revealed inhibition of spermatogenesis at primary spermatocyte stage and atrophic Leydig cells. Epididymis presented infantile features and there was a marked reduction in the secretory activity as revealed by lowered concentration of sialic acid and glycerylphosphorylcholine. Testosterone therapy (1 mg/day) for a period 45 days prior to autopsy at 60 days failed to induce completion of spermatogenesis but restored the structural and functional integrity of the epididymis.  相似文献
