Studies have shown that frog skin secretes many types of peptides that are good for human skin. In this study, acid and enzymatic extracts of Rana skin peptides (acid/enzymatic Rana skin peptides, ARPs/ERPs) were obtained. The chemical and physical properties of the ARPs and ERPs were identified through UV scanning, HGLC, FRIT, and MS. MTS and flow cytometry were used to test the proproliferative and antiapoptotic effects of the ARPs and ERPs on human immortalized keratinocytes (HaCaT cells). To elucidate the antiapoptotic mechanisms, the mRNA and protein levels of EGF (epidermal growth factor, which enhances stimulation of cellular proliferation in both cells and epithelial tissues) and caspase-3 were evaluated using quantitative RT-PCR. The results indicated that the ARPs and ERPs were extracted from the Rana skin with yields of 0.65% and 0.52%, respectively. Treatment with ARPs (1.6 g/L) and ERPs (0.8 g/L) showed a 1.66-fold (p < 0.001) and 2.1-fold (p < 0.001) enhancement in the proliferation rates of HaCaT cells. The rate of apoptosis decreased by 2.6 fold (p < 0.01) and 3.4 fold (p < 0.01) under the UVB stimulation, respectively, at the same time, the up-regulation of EGF and down-regulation of caspase-3 were found. These results suggested that we can dig into the potential value of ARPs/ERPs in a new field.  相似文献

An experiment examined the effects of two field bean cultivar samples with different tannin contents, the effect of heat treatment (micronising) and the effect of dietary supplementation of a proprietary enzyme preparation containing tannase, pectinase, and xylanase activities on metabolisable energy (ME), total tract dry matter digestibility (DMD) and ether extract digestibility (EED), nitrogen retention (NR), tannin degradability, gastrointestinal tract (GIT) development, and endogenous mucin losses excretion in broiler chickens. The Control diet contained per kg 221 g crude protein and 12.83 MJ ME. Four additional diets contained 300 g/kg of each of the two untreated or micronised experimental field bean cultivar samples. Each diet was then split into two batches and one of them was supplemented with 3400 units tannase per kg diet resulting in 10 diets in total. Each diet was fed to seven pens with two randomly selected male broilers each. Birds fed the high tannin bean sample had a lower weight gain (p < 0.001), and a lower determined apparent ME (p < 0.05), and DMD (p < 0.001) but a higher tannin degradability (p < 0.001). Compared to the Control diet, feeding field beans increased (p < 0.001) the weights of the proventriculus and gizzard of the birds, and also increased endogenous mucin losses (p < 0.05). Supplementing diets with the tannase-containing enzyme preparation improved dietary ME (p < 0.001), DMD (p < 0.001), NR (p < 0.001) and DEE (p < 0.05), but did not change tannin degradability. Heat treatment of the beans reduced the degradability of condensed tannins and increased endogenous mucin losses (p < 0.05). The differences in the feeding value of the different field bean samples were not improved by heat treatment, but enzyme supplementation improved the feeding value of all diets regardless of the bean samples or heat treatment. Further research is warranted to study the effectiveness of tannase supplementation in poultry diet formulations by dose response trials with purified tannase preparations.  相似文献

Active individuals often perform exercises in the heat following heat stress exposure (HSE) regardless of the time-of-day and its variation in body temperature. However, there is no information concerning the diurnal effects of a rise in body temperature after HSE on subsequent exercise performance in a hot environnment. This study therefore investigated the diurnal effects of prior HSE on both sprint and endurance exercise capacity in the heat. Eight male volunteers completed four trials which included sprint and endurance cycling tests at 30 °C and 50% relative humidity. At first, volunteers completed a 30-min pre-exercise routine (30-PR): a seated rest in a temperate environment in AM (AmR) or PM (PmR) (Rest trials); and a warm water immersion at 40 °C to induce a 1 °C increase in core temperature in AM (AmW) or PM (PmW) (HSE trials). Volunteers subsequently commenced exercise at 0800 h in AmR/AmW and at 1700 h in PmR/PmW. The sprint test determined a 10-sec maximal sprint power at 5 kp. Then, the endurance test was conducted to measure time to exhaustion at 60% peak oxygen uptake. Maximal sprint power was similar between trials (= 0.787). Time to exhaustion in AmW (mean±SD; 15 ± 8 min) was less than AmR (38 ± 16 min; < 0.01) and PmR (43 ± 24 min; < 0.01) but similar with PmW (24 ± 9 min). Core temperature was higher from post 30-PR to 6 min into the endurance test in AmW and PmW than AmR and PmR (< 0.05) and at post 30-PR and the start of the endurance test in PmR than AmR (< 0.05). The rate of rise in core temperature during the endurance test was greater in AmR than AmW and PmW (< 0.05). Mean skin temperature was higher from post 30-PR to 6 min into the endurance test in HSE trials than Rest trials (< 0.05). Mean body temperature was higher from post 30-PR to 6 min into the endurance test in AmW and PmW than AmR and PmR (< 0.05) and the start to 6 min into the endurance test in PmR than AmR (< 0.05). Convective, radiant, dry and evaporative heat losses were greater on HSE trials than on Rest trials (< 0.001). Heart rate and cutaneous vascular conductance were higher at post 30-PR in HSE trials than Rest trials (< 0.05). Thermal sensation was higher from post 30-PR to the start of the endurance test in AmW and PmW than AmR and PmR (< 0.05). Perceived exertion from the start to 6 min into the endurance test was higher in HSE trials than Rest trials (< 0.05). This study demonstrates that an approximately 1 °C increase in core temperature by prior HSE has the diurnal effects on endurance exercise capacity but not on sprint exercise capacity in the heat. Moreover, prior HSE reduces endurance exercise capacity in AM, but not in PM. This reduction is associated with a large difference in pre-exercise core temperature between AM trials which is caused by a relatively lower body temperature in the morning due to the time-of-day variation and contributes to lengthening the attainment of high core temperature during exercise in AmR.  相似文献

Fermentation kinetics, digestibility, faecal characteristics and bacterial populations (aerobes, anaerobes, lactobacilli, lactic acid bacteria, enterococci, coliforms and clostridia) of dog food mixed with citrus pulp and apple pomace were evaluated. The in vitro gas production of a pre-digested dog food mixed with 0, 30, 50 and 70 g/kg dry matter (DM) of citrus pulp or apple pomace was measured, and also an experiment with dogs fed the same dog food with or without the addition of 70 g/kg of either fresh citrus pulp or apple pomace was conducted. Gas production increased linearly (p < 0.001) and quadratically (p < 0.001) as fibre levels augmented. The inclusion of fibre sources in the diets resulted in higher faecal output (p = 0.005) and defecation frequency (p < 0.001), and lower faecal pH (p < 0.001) and digestibility values (p < 0.01). Faecal consistencies and microbial populations did not differ among treatments. The addition of fresh citrus and apple was effective to stimulate the hindgut fermentation, but slightly depressed the digestion.  相似文献

Eating during the night may increase the risk for obesity and type 2 diabetes in shift workers. This study examined the impact of either eating or not eating a meal at night on glucose metabolism. Participants underwent four nights of simulated night work (SW1–4, 16:00–10:00 h, <50 lux) with a daytime sleep opportunity each day (10:00–16:00 h, <3 lux). Healthy males were assigned to an eating at night (NE; n = 4, meals; 07:00, 19:00 and 01:30 h) or not eating at night (NEN; n = 7, meals; 07:00 h, 09:30, 16:10 and 19:00 h) condition. Meal tolerance tests were conducted post breakfast on pre-night shift (PRE), SW4 and following return to day shift (RTDS), and glucose and insulin area under the curve (AUC) were calculated. Mixed-effects ANOVAs were used with fixed effects of condition and day, and their interactions, and a random effect of subject identifier on the intercept. Fasting glucose and insulin were not altered by day or condition. There were significant effects of day and condition × day (both < 0.001) for glucose AUC, with increased glucose AUC observed solely in the NE condition from PRE to SW4 (= 0.05) and PRE to RTDS (< 0.001). There was also a significant effect of day (= 0.007) but not condition × day (= 0.825) for insulin AUC, with increased insulin from PRE to RTDS in both eating at night (= 0.040) and not eating at night (= 0.006) conditions. Results in this small, healthy sample suggest that not eating at night may limit the metabolic consequences of simulated night work. Further study is needed to explore whether matching food intake to the biological clock could reduce the burden of type 2 diabetes in shift workers.  相似文献

ABSTRACT

Introduction: Two of the most ubiquitous fatigue countermeasures used by shift-working nurses are napping and caffeine. This mixed-methods case study investigated the ways nurses and midwives utilised napping and caffeine countermeasures to cope with shift work, and associated sleep, physical health and psychological health outcomes.

Materials and Methods: = 130 Australian shift-working nurses and midwives (mean age = 44 years, range = 21–67, 115F, 15M) completed the Standard Shiftwork Index. A sub-set of 22 nurses and midwives completed an in-depth interview.

Results: Nearly 70% of participants reported napping. Those who napped during night shifts had significantly less total sleep time before (F2,75 = 5.5, < 0.01) and between days off (F2,82 = 3.9, < 0.05). By the end of the night shift, average hours of time awake were significantly less for prophylactic and on-shift nappers compared to non-nappers (F2,85 = 97.2, p < 0.001). Since starting shift work, the percentage of high caffeine consumers (>400 mg/day) increased from 15% to 33% of the sample and an average of 4 (SD = 2) caffeinated beverages per day was reported. Increased caffeine consumption was associated with greater sleep disturbance (= 0.26, < 0.05), psychological distress (= 0.37, < 0.001), abdomen pain (= 0.27, < 0.05) and weight gain since starting shift work (= 0.25, < 0.05). Interviews confirmed these relationships and revealed that caffeine consumption on night shift was common, whereas napping on night shift was dependent on a number of factors including ability to sleep during the day.

Conclusion: This study identified reasons shift workers chose to engage in or abstain from napping and consuming caffeine, and how these strategies related to poor sleep and health outcomes. Further research is required to help develop recommendations for shift workers regarding napping and caffeine consumption as fatigue countermeasures, whilst taking into account the associated hazards of each strategy.  相似文献

Background

Complications of ostomy significantly affect the quality of life of ostomates. There is little evidence on the rate of long-term complications in ostomates, especially from the developing countries which include Sri Lanka. This study was aimed to describe the long-term complications of enteral ostomies and their contributory factors.

Methods

A retrospective analysis was carried out on 192 patients who underwent ostomy creation over a period of 5 years. Data on type of complications, age, sex, type of ostomy, type of surgery and perioperative care by enteric stoma therapist were gathered. Associations were established using Chi square test and multiple logistic regression.

Results

Out of 192 patients, only 146 patients presented regularly for follow up. The mean follow up duration was 28 months (range: 3–183). Around 34.2% developed surgical long-term complications related to the ostomy. Common complications were prolapse (n = 24, 16.4%), skin excoriation (n = 22, 15.1%) and parastomal hernia (n = 14, 9.6%). Overall complication rate was significantly less in loop ostomies (p < 0.05) and defunctioning ostomies (p < 0.05). Skin excoriation was significantly high in males (p < 0.05) and in ileostomies (p < 0.001). Parastomal hernia was commoner in end ostomies (p < 0.05). Perioperative care by enteric stoma therapist reduced the overall and specific complications (p < 0.001).

Conclusion

The overall complication rate in our cohort of patients was 34.2%. The perioperative care of a stoma therapist may be very effective in preventing complications particularly in a setting with limited resources.

The present study aimed to investigate the diurnal variation of postural stability, attentional capacities, and oral temperature in 5–6-year-old tennis players. A total of 24 young children were divided into two groups: Twelve tennis players with 2 years of training experience and twelve sedentary children without previous experience in any type of sport. They were asked to maintain an upright bipedal stance on a static force platform with eyes open (EO) and eyes closed (EC) at 07:00, 10:00, 14:00, and 18:00 h. Postural stability was evaluated by the center of pressure (CoP), surface area (CoPArea), CoP mean velocity (CoPVm), and Romberg’s index (RI). Oral temperature and simple reaction time (SRT) were also recorded at the beginning of each test session. Postural stability in 5–6-year-old tennis players was low at 07:00 h and improved at 10:00 h (p < 0.001); then, it worsened at 14:00 h (p < 0.001) and improved again at 18:00 h (p < 0.001) as found in sedentary children. This rhythm was also close to that of SRT in both groups. Body temperature increased significantly (p < 0.001) from the morning to the afternoon in both groups. However, the peak of postural performance, the peak of attentional capacities, and the greatest vision contribution to maintain balance observed at 18:00 h were only found in the trained group. Moreover, young tennis players were more stable (p < 0.001) and more attentive (p < 0.01) than sedentary ones at 18:00 h. The amplitude of the diurnal rhythm of CoP parameters was significantly larger (p < 0.01) in trained children compared to sedentary ones (43.1 ± 8.5 vs 31.7 ± 8.3 for the CoPArea; 27.5 ± 7.4 vs 17.7 ± 8.2 for the CoPVm). Therefore, time-of-day has a significant effect on static postural stability and attentional capacities in young tennis players with better performances in the late afternoon (habitual hour of training). Thus, we recommend planning some training sessions at 07:00 and/or 14:00 h to optimize postural stability at the time of its batyphase and to reduce the incidence of fall-related injury risks during this critical age in athletic environments.  相似文献

The accumulation and removal efficiency of Fe by Centella asiatica was carried out at various Fe concentrations in soil treatments (0, 50, 100, 150 and 200 mg Fe/kg soil). Iron accumulation in different parts of C. asiatica (leaf, stem and root) was analyzed by atomic absorption spectrophotometer (AAS). Factorial experiment with a completely randomized design and Duncan's test were used for data analyses. The results revealed that C. asiatica have the ability to uptake and accumulate Fe significantly (p < 0.05; r = 0.977) in the aerial parts. The different soil treatments had significant effect on the total Fe accumulations in C. asiatica (p < 0.05). The potential of C. asiatica as a metal hyperaccumulator plant, harvested for analysis, shows efficient accumulation of Fe at high concentration (p < 0.05; r = 0.977). The root showed the highest accumulation of Fe followed by the leaves (p < 0.05) and the stem (p < 0.05). The Pearson correlation coefficient between leaves and root have showed highly significant correlation (p < 0.01; r = 0.785) as compared to the leaves and stem (p < 0.01; r = 0.780). The efficiency of Fe removal by C. asiatica from the contaminated soil has been evaluated by bioconcentration factor and translocation factor, found to be >1 and <1, respectively, further supporting its metal hyperaccumulator properties.  相似文献

This study aimed to evaluate the effects of adding dried distillers’ grains with solubles (DDGS) to the nutritional regimens of ewes at different time periods on reproductive traits, serum hormones and serum metabolites. In Experiment 1, 100 ewes were divided into 4 groups (n = 25) according to diet. The four diets were grass hay (H) alone (group H), H with barley (group H + B), H with DDGS for the whole feeding period (27 d) (group H + DDGS) or H and DDGS for 5 d (d 8–12 of oestrus cycle) (group H + DDGS5). Serum progesterone concentrations were affected by oestrus cycle (p < 0.001), but not by dietary treatments. However, feeding H + DDGS caused significant increases in serum insulin, leptin and growth hormone concentrations (p < 0.05). In Experiment 2, 30 ewes were divided into 2 groups (n = 15), receiving DDGS or soybean meal (SBM) during the prepartum period. Diets had no significant effect on weights of dams or lambs at birth; however, the weaning weights of lambs born from ewes of group DDGS were significantly higher (p < 0.05). Moreover, serum glucose, blood urea nitrogen, triglyceride and beta hydroxybutyric acid (BHBA) concentrations during the prepartum period were affected by dietary treatments and by time (< 0.001). During the prepartum period, BHBA concentrations of ewes fed SBM were higher, while BHBA levels decreased during the last weeks of pregnancy regardless of diet. Lamb serum immunoglobulin G concentrations increased from 1 h to 24 h after birth (< 0.001). Colostrum of ewes fed SBM had higher fat-free dry matter (DM) and protein contents in comparison to colostrum of ewes fed DDGS (< 0.05). In conclusion, DDGS can be included as protein source in pregnancy rations up to 15% of DM to obtain reproductive performance outcomes equal to or exceeding those obtained with SBM.  相似文献

This study examined the sleep intra-individual variability (IIV) of rugby league athletes across senior and junior levels during one week of the competitive season. Forty-five rugby league athletes across elite senior, sub-elite senior and elite junior levels each wore actigraphy monitors for seven days during the competitive season, and completed a subjective sleep diary each morning upon waking. Linear mixed models were used to assess differences in sleep measures between playing levels. Intra-individual standard deviations for each sleep measure were calculated for each athlete to determine their respective IIV, allowing differences in IIV between groups to be assessed. Elite junior athletes went to bed later (ES = 0.94 ± 0.50, < 0.05) and woke later than elite senior athletes (ES = 0.94 ± 0.40, < 0.05), while also displaying greater IIV when considering time in bed (ES = 1.14 ± 0.60, < 0.05) and sleep duration (ES = 1.53 ± 0.64, < 0.05) compared with elite senior athletes. Similarly, IIV was more pronounced in elite junior players for time in bed (ES = 0.88 ± 0.60, < 0.05) and sleep duration (ES = 1.03 ± 0.64, < 0.05) compared with sub-elite senior athletes. Despite this, elite junior athletes still obtained sufficient sleep duration, efficiency and quality. The outcomes of this investigation show a distinct difference in the habitual sleep-wake patterns of senior and junior rugby league athletes, and show how sleep IIV differs between playing levels in rugby league.  相似文献

High solar radiation has been recognised as a contributing factor to exertional heat-related illness in individuals exercising outdoors in the heat. Although solar radiation intensity has been known to have similar time-of-day variation as body temperature, the relationship between fluctuations in solar radiation associated with diurnal change in the angle of sunlight and thermoregulatory responses in individuals exercising outdoors in a hot environment remains largely unknown. The present study therefore investigated the time-of-day effects of variations in solar radiation associated with changing solar elevation angle on thermoregulatory responses during moderate-intensity outdoor exercise in the heat of summer. Eight healthy, high school baseball players, heat-acclimatised male volunteers completed a 3-h outdoor baseball trainings under the clear sky in the heat. The trainings were commenced at 0900 h in AM trial and at 1600 h in PM trial each on a separate day. Solar radiation and solar elevation angle during exercise continued to increase in AM (672–1107 W/m2 and 44–69°) and decrease in PM (717–0 W/m2 and 34–0°) and were higher on AM than on PM (both < 0.001). Although ambient temperature (AM 32–36°C, PM 36–30°C) and wet-bulb globe temperature (AM 31–33°C, PM 34–27°C) also continued to increase in AM and decrease in PM, there were no differences between trials in these (both > 0.05). Tympanic temperature measured by an infrared tympanic thermometer and mean skin temperature were higher in AM than PM at 120 and 180 min (< 0.05). Skin temperature was higher in AM than PM at the upper arm and thigh at 120 min (< 0.05) and at the calf at 120 and 180 min (both < 0.05). Body heat gain from the sun was greater during exercise in AM than PM (< 0.0001), at 0–60 min in PM than AM (< 0.0001) and at 120–180 min in AM than PM (< 0.0001). Dry heat loss during exercise was greater at 0–60 min (< 0.0001), and lower at 60–120 min (< 0.05) and 120–180 min (< 0.0001) in AM than PM. Evaporative heat loss during exercise was greater in PM than AM at 120–180 min (< 0.0001). Total (dry + evaporation) heat loss at the skin was greater during exercise in PM than AM (< 0.0001), at 0–60 min in AM than PM (< 0.0001) and at 60–120 and 120–180 min in PM than AM (< 0.05 and 0.0001). Heart rate at 120–150 min was also higher in AM than PM (< 0.05). Neither perceived thermal sensation nor rating of perceived exertion was different between trials (both > 0.05). The current study demonstrates a greater thermoregulatory strain in the morning than in the afternoon resulting from a higher body temperature and heart rate in relation to an increase in environmental heat stress with rising solar radiation and solar elevation angle during moderate-intensity outdoor exercise in the heat. This response is associated with a lesser net heat loss at the skin and a greater body heat gain from the sun in the morning compared with the afternoon.  相似文献

The study evaluated the effects of different doses of 25-hydroxyvitamin D3 (25(OH)D3) on growth performance, immune function and antioxidative capacity in piglets. In a 21-d trial, 35 weaned pigs were divided into five groups and diets were supplemented with 5.5 (control), 43.0, 80.5, 118.0 and 155.5 μg 25(OH)D3/kg, respectively. No treatment effects were observed for average daily gain, average daily feed intake and feed to gain ratio. Increasing dietary 25(OH)D3 levels increased serum 25(OH)D3 concentrations linearly (< 0.01), decreased the frequency of CD3+CD4+ and CD3+CD8+ T cells (< 0.01), and the serum level of complement component 3 (< 0.05). Supplementation of 80.5 and 118.0 μg 25(OH)D3/kg enhanced the activity of serum glutathione peroxidase (< 0.05) and addition of 43.0 μg 25(OH)D3/kg increased the malondialdehyde concentration (< 0.05). Overall, feeding high-dose 25(OH)D3 to weaned pigs partly improved immune functions and the antioxidative capacity.  相似文献

Disruption of the rest–activity rhythm in patients with lung cancer can accelerate cancer progression and affect survival. Rest–activity rhythm changes with age. Therefore, we investigated the effects of rest–activity rhythm and age on patients’ survival. A total of 84 patients with lung cancer were recruited, then separated into two groups; younger patients aged under 65 years or elderly patients aged 65 and over. The dichotomy index (I < O) was used to estimate the rest–activity rhythm measured through the actigraphy motion detector. Cox proportional hazards models were adopted to investigate the effects of different variables on the patients’ survival. After adjusting for confounding, the risk of earlier mortality in the younger patients with disrupted I < O were 2.52 (95%CI = 1.09–5.82) times higher than that in the younger patients with robust I < O (= 0.03), the risk of earlier mortality in the elderly patients with disrupted I < O was 4.08 (95%CI = 1.91–8.68) times higher than that in the elderly patients with robust I < O (< 0.001). Therefore, age and I < O influence the survival period of patients with lung cancer. Moreover, disrupted I < O has a substantial influence on elderly patients. In conclusion, aging and disrupted rest–activity rhythm negatively and jointly influenced the survival period of the patients with lung cancer and significantly increased their death risk.  相似文献

The objective of this study was to evaluate the effect of a fresh sugarcane-based diet and different roughage-to-concentrate ratios (70:30, 60:40, 40:60 and 20:80) on the rumen microbiota associated with rumen fermentation parameters and the intake and apparent digestibility of nutrients in Nellore steers. Eight rumen-cannulated Nellore steers (331 ± 8 kg BW) were distributed in a double 4 × 4 Latin square design balanced for the control of the residual effect. The ruminal pH decreased (p < 0.01) and the concentrations of N–NH3, isovaleric and valeric acids increased linearly (p < 0.05) with an increase dietary concentrate level. Furthermore, an increased concentrate proportion reduced the population of Fibrobacter succinogenes and Ruminococus flavefaciens (p < 0.01) and increased the population of Selenomonas ruminantium and Megasphaera elsdenii (p < 0.01). The protozoa count revealed a predominance of the genus Entodinium. The synthesis of microbial N [g/d] and the efficiency of microbial synthesis [g of microbial N/kg of organic matter apparently digested in the rumen] increased as the proportion of concentrate was increased (p < 0.05). Therefore, it can be concluded that an increasing proportion of concentrate in sugarcane-containing diets enhances the synthesis of microbial protein and does not alter the fibre digestibility, although the population of fibre fermenting bacteria was reduced.  相似文献

This experiment examined if immune system stimulation of weaner pigs, initiated by inoculation an enterotoxigenic strain of Escherichia coli (ETEC), increased the requirement for dietary tryptophan (Trp), modulated the inflammatory response, altered plasma levels of Trp and its metabolite kynurenine (Kyn) and effected post-weaning diarrhoea. Individually housed pigs (n = 72) weaned at 21 d of age were allocated to one of six treatments (n = 12) according to a two by three factorial arrangement of (1) with or without ETEC infection and (2) three dietary ratios of standardised ileal digestible (SID) Trp to lysine (Lys) (SID Trp:Lys) of 0.16, 0.20 or 0.24, in a completely randomised block design. Pigs had ad libitum access to diets (per kg 14.13 MJ ME, 12.4 g SID Lys, 195 g crude protein) for 3 weeks after weaning. Pigs were infected with ETEC (O149:K98:K88) at 72, 96 and 120 h after weaning and then bled on day 3, 11 and 19. An increased dietary Trp:Lys ratio increased plasma Trp and Kyn (< 0.001) without effect of infection. On day 3, pigs fed 0.24 SID Trp:Lys had lower levels of plasma urea than at 0.20 Trp:Lys (= 0.047) and on day 11, plasma urea was lower at 0.20 than at 0.16 SID Trp:Lys (= 0.007). Infection increased (= 0.039) the diarrhoea index and deteriorated faecal consistency from day 4–10 (< 0.05). Treatments did not affect haptoglobin and acid soluble glycoprotein levels or daily gain and feed intake. However, 0.24 SID Trp:Lys improved (= 0.021) feed efficiency without an effect of infection. In conclusion, in the absence of dietary antibiotic growth promotants, increasing the dietary SID Trp:Lys ratio to 0.24 improved feed conversion ratio after weaning and increased plasma levels of Trp and Kyn regardless of infection with E. coli.  相似文献

Background

Irisin is a hormone released mainly from skeletal muscle after exercise which increases adipose tissue energy expenditure. Adipocytes can also release irisin after exercise, acting as a local adipokine to induce white adipose tissue to take on a brown adipose tissue-like phenotype, suggesting that irisin and its receptor may represent a novel molecular target for the treatment of obesity and obesity-related diabetes. Previous reports provide conflicting evidence regarding circulating irisin levels in patients with type 2 diabetes (T2DM).

Methods

This study investigated plasma irisin concentrations in 79 T2DM individuals, assessing potential associations with measures of segmental body composition, markers of endothelial dysfunction and peripheral blood mononuclear cell telomere length (TL).

Results

Resting, overnight-fasted plasma irisin levels were significantly higher in this group of T2DM patients compared with levels we previously reported in healthy volunteers (p < 0.001). Moreover, plasma irisin displayed a positive correlation with body mass index (p = 0.04), body fat percentage (p = 0.03), HbA1c (p = 0.03) and soluble E-selectin (p < 0.001). A significant negative association was observed between plasma irisin and visceral adiposity (p = 0.006) in T2DM patients. Multiple regression analysis revealed that circulating soluble E-selectin levels could be predicted by plasma irisin (p = 0.004). Additionally, cultured human umbilical vein endothelial cells (HUVEC) exposed to 200 ng/ml irisin for 4 h showed a significant fourfold increase in E-selectin and 2.5-fold increase in ICAM-1 gene expression (p = 0.001 and p = 0.015 respectively), and there was a 1.8-fold increase in soluble E-selectin in conditioned media (p < 0.05).

Conclusion

These data suggest that elevated plasma irisin in T2DM is associated with indices of adiposity, and that irisin may be involved in pro-atherogenic endothelial disturbances that accompany obesity and T2DM. Accordingly, irisin may constitute a potentially novel therapeutic opportunity in the field of obesity and cardiovascular diabetology.

In order to increase zinc (Zn) absorption and improve eggshell quality, diets for aged laying hens were supplemented with different Zn sources and the effects on egg production, eggshell quality and serum parameters were recorded. Seventy-five 64-week old brown layers were individually caged and randomly allotted to five treatment groups: an un-supplemented Control and four groups, where the following Zn sources were added to the diets, ZnO (group ZnO), Zn-methionine (group Zn-Met), nanosized ZnO (group nanoZnO) and γ-PGA-nano ZnO (group γ-PGA-nanoZnO). The Zn level was maintained at 80 mg/kg in the supplemented diet, while the Control group received Zn at 40 mg/kg diet. The results indicated that the average daily feed intake was highest in group nanoZnO (p < 0.05), whereas further performance parameters were not influenced by treatments. Eggshell thickness was increased in group γ-PGA-nanoZnO (p < 0.05). Zn content in eggshells and Zn concentration in serum were increased in groups Zn-Met, nanoZnO and γ-PGA-nanoZnO (p < 0.05). Serum ghrelin concentration was significantly elevated in all Zn-supplemented groups, but further increased in groups nanoZnO and γ-PGA-nanoZnO (p < 0.05). Carbonic anhydrase activity was highest in group γ-PGA-nanoZnO and lowest in group Zn-Met (< 0.05). Compared to the Control and group ZnO, supplementation of γ-PGA-nanoZnO and nanoZnO increased serum IgG levels (< 0.001). In conclusion, dietary supplementation of nanoZnO and γ-PGA-nanoZnO increased Zn content in eggshells, serum Zn concentration, ghrelin and IgG levels of aged layers when compared to regular ZnO. Compared to Zn-Met, the serum carbonic anhydrase activity and ghrelin levels were also increased. Thus, nanosized ZnO alone or mixed with γ-PGA has positive effects on the Zn status of aged layers.  相似文献

To examine the effects of maternal energy restriction along with selenium-methionine (Se-Met) and chromium-methionine (Cr-Met) supplementation on performance of pregnant sheep and their offspring, the following treatments were allotted randomly to 40 multiparous Baluchi ewes (53.9 ± 1.15 kg of body weight [BW]) from 5 weeks prior to 5 weeks after parturition: (1) Control diet (60% and 100% of NRC energy requirements in pre- and post-partum, respectively); (2) Control diet plus 5 mg Se-Met/kg dry matter (DM); (3) Control diet plus 3 mg Cr-Met/kg DM and (4) Control diet plus 5 mg Se-Met and 3 mg Cr-Met/kg DM (Se–Cr-Met) of concentrate diet. The results indicated that Cr-Met alone or in combination with Se-Met increased average DM intake of ewes. In addition, Group Cr-Met had higher average BW than the Control (< 0.05). Se-Met and/or Cr-Met supplementation led to decreased average serum cholesterol of the ewes (< 0.05). Groups Cr-Met and Se–Cr-Met displayed decreased average serum malondialdehyde compared to the Control (< 0.05). At 24 h post-partum, Group Se–Cr-Met had a greater serum Se content than the Control (= 0.006). Compared with the Control, the Se concentration in milk was significantly increased from 30 to 138 µg/l and 197 µg/l in Groups Se-Met and Se–Cr-Met, respectively (p < 0.01), which proved that Se-Met supplementation can increase the Se concentration of ewe milk. Furthermore, feeding Cr-Met may attenuate BW loss post-partum and Se-Met and/or Cr-Met supplements may ameliorate oxidative stress condition in ewes around parturition.  相似文献

The in vitro caecal fermentation of five substrates low in starch and protein content [d-(+)-glucose (GLU), d-cellobiose (CEL), sugar beet pectin (PEC), sugar beet pulp (SBP) and wheat straw (WS)] was investigated using soft faeces from rabbits receiving different levels of cellobiose and soluble fibre as inoculum. A total of 24 rabbits were supplemented 3 levels of cellobiose in the drinking water (0.0, 7.5, 15.0 g/l) and fed two experimental diets containing either low soluble fibre (LSF) or high soluble fibre (HSF) levels (84.0 and 130 g/kg dry matter). All substrates were subjected to a two-step pepsin/pancreatin in vitro pre-digestion, and the whole residue was used as substrate for the in vitro incubations. Gas production was measured until 144 h, and volatile fatty acid (VFA) production was determined at 24 h incubation. Experimental treatments did not affect SBP fermentation and had only a subtle influence on fermentation of WS and GLU. In contrast, cellobiose supplementation × donors’ diet interactions were detected for most gas production parameters for CEL. Both the fractional gas production (k) and maximal gas production rates were linearly increased (p ≤ 0.042) and the initial delay in the onset of gas production (Lag) linearly decreased (p < 0.001) by cellobiose supplementation with the HSF inoculum, with no differences between the 7.5 and 15.0 doses. In contrast, with the LSF inoculum cellobiose supplementation only affected k values, which were quadratically increased (p = 0.043) and had maximal values for the 7.5 dose. A quadratic effect (p ≤ 0.018) of cellobiose supplementation was observed for total VFA production at 24 h when CEL and PEC were fermented, obtaining the maximal VFA production for the 7.5 dose of cellobiose. Total VFA production for CEL was greater with LSF than with HSF inoculum (20.7 vs. 12.9 mmol/l; p = 0.014), but the opposite was found for WS (3.97 vs. 6.21 mmol/l; p = 0.005). The use of LSF inoculum for CEL fermentation sharply reduced acetate (p = 0.001) and increased butyrate proportions (p ≤ 0.001) compared with the HSF inoculum. A positive relationship between total VFA caecal concentrations in rabbits receiving the same experimental treatments and in vitro values was only observed when WS was used as substrate (r = 0.90; p = 0.015; n = 6). The results suggest that experimental factors influenced the fermentative activity of caecal digesta, but the observed response differed with the incubated substrate, being the CEL the most affected.  相似文献
