Summary P-protein and the changes it undergoes after wounding of sieve tubes of secondary phloem in one- to two-year old shoots ofHevea brasiliensis has been studied using electron microscopy. The P-protein in the form of tubules with a diameter of 8–9 nm and a lumen of 2–2.5 nm occurred in differentiating sieve elements and appeared as compact bodies which consisted of small aggregates of the tubules. As the sieve elements matured, these P-protein bodies dispersed with a disaggregation of the tubules before they turned into striated fibrils, 10–11 nm in diameter. In wounding experiments, as the mature sieve elements collapsed after cutting, their striated P-protein converted into tubules. These tubules were the same in ultrastructure as the tubules in differentiating sieve elements and they often were arranged in crystalline aggregates.  相似文献

Summary Haustoria ofCuscuta odorata R. & P. andC. grandiflora H.B.K. show continuous traces of sieve elements, connecting the phloem of the host with that of theCuscuta shoot. The continuity of this haustorial phloem is discernible by callose fluorescence after staining with aniline blue. The fine structural criteria for sieve tubes are analyzed electronmicroscopically, with special respect to sieve pores, P-protein, and a distinct wall-standing smooth surfaced ER. Within the central part of the haustorium sieve tubes are elongated, while the elements abutting the phloem of theCuscuta shoot are nearly isodiametric in shape. Both elements are associated with rather large companion cells, derived from an unequal division.

Summary After chemical fixation following two different preparation procedures, the ultrastructure of mature sieve elements (SEs) was systematically compared in the transport phloem ofVicia faba leaves andLycopersicon esculentum internodes. The SEs in samples obtained by gentle preparation were well preserved, while those in conventionally prepared samples were generally injured. (1) In well-preserved SEs, parietal P-proteins were associated with cisternae of the SE endoplasmic reticulum (ER). Additionally, theV. faba SEs had crystalline P-proteins, and a homogeneous network of filamentous P-proteins occurred in the lumen of theL. esculentum SEs. In injured SEs, all P-proteins were dispersed. (2) In well-preserved SEs, stacked ER cisternae associated with P-proteins lay also on the sieve-plate walls, but passages were kept free in front of the sieve pores. Injured SEs lacked these orderly arranged deposits. Instead, irregular filamentous and membranous materials occluded the sieve pores. (3) In well-preserved SEs, the sieve-pore lumen was free of obstructions, apart from small, lateral coatings of P-proteins. Sieve pores in injured SEs were always occluded. (4) The SE organelles and, in tomato SEs, also the parietal ER located at the longitudinal walls were firmly attached in the SE periphery and stayed in place after injury. The stable parietal attachment is likely exerted by minute, clamplike structures which link the outer membranes of the SE components with one another or to the SE plasma membrane. Single, straight clamps with a length of about 7 nm anchored the SE components directly to the SE plasma membrane. The connections between adjacent SE organelles and/or parietal ER cisternae were mostly twice as long (about 15 nm) and often were branched. Presumably, the long, branched clamps were constituted by the interaction of opposite short clamps. The ultrastructural results are discussed with respect to SE functioning.  相似文献

Pyrenoid ultrastructure has been investigated from cultures of all 26 species ofTrebouxia with the aim of establishing pyrenoids as a taxonomic character. Different arrangements and forms of thylakoid lamellae within the pyrenoid matrix allow eight pyrenoid types to be distinguished. Each type is characteristic of a group of species. Thegigantea- andimpressa-type are similar, differing only in the form of the tubules: short, branched tubules mark thegigantea-type; ± long and straight invaginations theimpressa-type. Thearboricola-type is characterized by meandering pyrenoid membranes developing from lamellae parallel with each other in young autospores. Pyrenoids of thegelatinosa-type are traversed by thin parallel-arranged tubules. Few thylakoids with a curved profile are typical of theirregularis-type. Thecorticola-type is different from all others in having a distinct starch sheath closely connected with the pyrenoid matrix and no pyrenoglobuli being associated with the pyrenoid membranes. No true pyrenoids have been found inT. magna andT. erici. Within the chloroplast, they have indistinct areas with pyrenoglobuli, but without differentiated thylakoids. Pyrenoid morphology is stable in culture on different media as well as in phycobionts within lichen thalli. Comparing the pyrenoid of a lichenizedTrebouxia with that from cultured species, the identification of the phycobiont within the lichen thallus is possible, without the need of culturing the algae. This has been shown in species ofParmelia andHypogymnia. Polysomes in sieve elements of rice (Oryza sativa L.) were studied with the electron microscope. The polysomes were found on the rough endoplasmic reticulum (ER) present in immature sieve elements and also on the cisternae of aggregated ER in the parietal layer of mature, enucleate sieve elements. In the immature sieve elements the ER cisternae existed as narrow profiles while in the mature sieve elements the ER cisternae were considerably dilated and contained a fibrillar material and, occasionally, electron-opaque inclusions. In addition to the aggregated ER, single profiles of ER were found applied to the lateral walls and also the sieve plates. These cisternae also bore ribosomes and were separated from the plasmalemma by a narrow, dense space. In the mature sieve elements much of the surface of the ER membranes was covered with polysomes. The dimensions of the polysomes are described and the possibility that they contribute to the formation of the fibrillar material in the intracisternal space is discussed.Abbreviations ER endoplasmic reticulum  相似文献

Summary Cell plate formation inChara zeylanica was compared with recent models of cytokinesis in higher plants in order to gain insight into the evolutionary origin of plant cytokinetic processes. Transmission electron microscopy (TEM) reveals that while cytokinesis inC. zeylanica bears many features in common with that in higher plants, there are significant differences. Unlike that in higher plants, cytokinesis inC. zeylanica begins with a congregation of smooth membrane tubules that are closely associated with endoplasmic reticulum (ER) and Golgi membranes. Mitochondria and other organelles excluded by the phragmoplast in higher plants are present as well. Unlike in higher plants, phragmoplast microtubules persist throughout cytokinesis inC. zeylanica, and the cell plate generally forms across the whole cell at once, though development is patchy, due to small regions developing at different rates; the ends of the plate form last. By identifying aspects of cytokinesis that are different inC. zeylanica and plants, our study indicates which cytokinetic features are more likely to be derived, and which are more likely to be ancestral. Summary Differences in the activity and structure of the vacuolar H+-ATPase (V-ATPase, EC 3.6.1.3) were investigated in the C3/CAM intermediate plantKalanchoë blossfeldiana Poellnitz cv. Tom Thumb, with lower or higher expression of CAM, andHordeum vulgare cv. Carina, grown with or without 150 mM NaCl. InK. blossfeldiana ATP-hydrolysis and H+-transport activity were higher with higher expression of CAM than in plants with very weak CAM. This was mainly due to a larger amount of V-ATPase. Statistical analysis of the diameter of intramembrane particles (IMPs) on freeze-fractures of tonoplast vesicles showed that IMPs were larger in tonoplast vesicle preparations ofK. blossfeldiana with strong CAM expression (9.1 nm) than in preparations ofK. blossfeldiana with low CAM expression (7.3 nm). As there is evidence that the majority of IMPs on freeze-fractures of tonoplast vesicles corresponds to the V0 domain of V-ATPase, the higher activity of V-ATPase inK. blossfeldiana with stronger CAM could be a result of additional structural changes in its membrane-integral domain. The higher activity of V-ATPase inK. blossfeldiana with stronger CAM is discussed in relation to the requirement for a higher proton pumping capacity for nocturnal malate accumulation in the vacuole. The ATP-dependent H+-pumping activity inH. vulgare was higher under salt stress than in control plants, while the rates of ATP-hydrolysis and the size of IMPs were not affected by the salt treatment. Summary The formation and development of linear terminal complexes (TCs), the putative cellulose synthesizing units of the red algaErythrocladia subintegra Rosenv., were investigated by a freeze etching technique using both rotary and unidirectional shadowing. The ribbon-like cellulose fibrils ofE. subintegra are 27.6 ± 0.8 nm wide and only 1–1.5 nm thick. They are synthesized by TCs which are composed of repeating transverse rows formed of four particles, the TC subunits. About 50.4 ± 1.7 subunits constitute a TC. They are apparently more strongly interconnected in transverse than in longitudinal directions. Some TC subunits can be resolved as doublets by Fourier analysis. Large globular particles (globules) seem to function as precursor units in the assembly and maturation of the TCs. They are composed of a central hole (the core) with small subunits forming a peripheral ridge and seem to represent zymogenic precursors. TC assembly is initiated after two or three gobules come into close contact with each other, swell and unfold to a nucleation unit resembling the first 2–3 transverse rows of a TC. Longitudinal elongation of the TC occurs by the unfolding of globules attached to both ends of the TC nucleation unit until the TC is completed. The typical intramembranous particles observed inErythrocladia (unidirectional shadowing) are 9.15 ± 0.13 nm in diameter, whereas those of a TC have an average diameter of 8.77 ± 0.11 nm. During cell wall synthesis membranes of vesicles originating from the Golgi apparatus and which seem to fuse with the plasma membrane contain large globules, 15–22 nm in diameter, as well as tetrads with a particle diameter of about 8 nm. The latter are assumed to be involved in the synthesis of the amorphous extracellular matrix cell wall polysaccharides. The following working model for cellulose fibril assembly inE. subintegra is suggested: (1) the ribbon-like cellulose fibril is synthesized by a single linear TC; (2) the number of glucan chains per microfibril correlates with the number of TC subunits; (3) a single subunit synthesizes 3 glucan chains which appear to stack along the 0.6 nm lattice plane; (4) lateral aggregation of the 3-mer stacks leads to the crystalline microfibril.Dedicated to Prof. Dr. Dr. h.c. The hypothesis ofHenriques andFernandes that several Iberian species ofNarcissus (Amaryllidaceae) are tristylous is reconsidered. Contrary to the opinion ofBateman and most subsequent authors, we believe that the available evidence indicates that some populations ofN. triandrus andN. fernandesii, at least, are tristylous; other populations ofN. triandrus are distylous.Hugonia cf.penicillanthemum (Linaceae) from new Caledonia is distylous, but it remains possible that other species ofHugonia are tristylous. The disputed occurrence of heterostyly in S. African species ofBauhinia (Leguminosae),Cleome (Capparaceae) andAneilema (Commelinaceae), and inAgelaea (Connaraceae) is discussed.  相似文献

Endoplasmic reticulum in the root protophloem of Nymphoides peltata (S.G. Gmel.) O. Kuntze changes form as sieve elements differentiate. In immature sieve elements the individual endoplasmic reticulum (ER) cisternae form large irregular aggregates in the cytoplasm. In older immature sieve elements the ER aggregates are more ordered and membranes in them are convoluted. Although convoluted ER predominates in immature sieve elements the ER of the mature sieve elements consists mainly of flattened stacks of ER cisternae. Some of these stacks of ER may be derived from the existing convoluted ER. Crystalline fibrils first appear in the cytoplasm of the sieve element when the ER starts to aggregate. The crystalline fibrils move to the parietal layer of the sieve element along with the aggregates of ER. The ultrastructure of the sieve elements ofAustrobaileya is compared with that of angiosperm sieve tubes and gymnosperm sieve cells (mostly fromCycadales). Except for the size of the sieve poresAustrobaileya shares all ultrastructural characters (e.g., chromatolytic nuclear degeneration, presence of p-protein, formation of sieve pores from unbranched plasmodesmata) and other features (e.g., companion cells) with angiosperm sieve tubes. Gymnosperm sieve cells on the contrary are characterized by pycnotic nuclear degeneration, absence of p-protein, formation of sieve areas from branched plasmodesmata with median cavities. — The exact ordinal assignment ofAustrobaileya within the subclassMagnoliidae is still disputed, a placement close to eitherMyristicaceae andWinteraceae orMonimiaceae being possible as judged from both S-type sieve-element plastids and p-protein bodies. — On the basis of the ultrastructural results fromAustrobaileya it is proposed to reconsider concepts and terminology of sieve elements. i.e., to include features from sieve pore development, nuclei degeneration and presence of specific proteins into the definitions and to restrict the term sieve cell to gymnospermous sieve elements which differ much from those of other vascular plants.  相似文献

The floral vasculature in three allied genera,Plagiorhegma, Jeffersoria andAchyls is investigated, and the results are compared with those ofEpimedium andVancouveria which are related closely toPlagiorhegma andJeffersonia. The vasculature in the receptacle ofPlagiorhegma andJeffersonia is similar, but that ofAchlys is much simpler. Slightly different trace patterns are observed in the sepals ofPlagiorhegma andJeffersonia. InJeffersonia, the 3-trace condition leaving 2 or 3 gaps is most frequently observed, but inPlagiorhegma traces of a double nature leaving a single gap are more frequent. The traces to the innermost sepals, petals and stamens are usually of a double nature leaving a single gap in both genera. Regular division and fusion are not observed in the receptacular stele. The vascular differentiation between sepals and petals is more advanced inPlagiorhegma andJeffersonia than inEpimedium andVancouveria. InAchlys, the traces are all staminal and single throughout their course. Two parts recognized in the pistils ofPlagiorhegma, Jeffersonia andAchlys are traversed by independent vasculature. The comparisons of pistil morphology including vasculature ofPlagiorhegma, Jeffersonia, Achlys, Epimedium andVancouveria lead to the interpretation that the pistils are based on the same morphological plan. The probable evolutionary trend in pistil is then suggested in these five genera.  相似文献

To assess the effect of water depth on the decomposition process, I measured the losses in dry mass of the above- and belowground materials ofCarex utriculata andNuphar luteum ssp.potysepalum as well as cellulose (Whatman filter paper) in the top 10 cm of sediment/soil in a subalpine marsh. Samples were examined by the litter bag technique at three flooding levels (0 to 5, 60, and 100 cm water depth). Over a 374-d period, the % mass losses of cellulose,Carex leaves and roots, andNuphar leaves and rhizomes ranged from 98.5 to 99.0, 74.8 to 81.8, 36.3 to 44.9, 95.8 to 97.7, and 78.4 to 91.5%, respectively. Rates for cellulose decay in this study were much higher than for samples from other wetlands; this difference resulted from the location of the litter bag (in the top 10 cm of soilvs in the water column). Water depth significantly affected the decomposition ofCarex roots andNuphar rhizomes. The rate of loss for K was highest in all tissues ofCarex andNuphar, followed by Na inCarex and P inNuphar. N and Ca loss rates generally were low. The C/N ratio tended to converge to a common value over the long term. This convergence has an important implication in the paleoecological interpretation of the C/N ratio change in sediment; i.e., this ratio shift in the sediment core results from a change in the environment, rather than the source material.  相似文献

An analysis of the morphology, anatomy and ontogeny of the flowers, particularly of the gynoecium ofLagoecieae is presented. 1. The gynoecial model of angiosperms can be applied to all three generaArctopus, Lagoecia andPetagnia. 2. In the case ofArctopus an additional Apikalseptum is developed. 3. In the synascidiate region of the gynoecium the adaxial carpel is reduced inArctopus andPetagnia, the abaxial inLagoecia. 4. The reduced carpel produces either one mature ovule inArctopus, a rudimentary ovule inPetagnia, or none inLagoecia. 5.Petagnia andLagoecia have a completely pseudomonomerous gynoecium. 6.Arctopus displays many flower characteristics which lack in theSaniculoideae but occur in theHydrocotyloideae. 7. ForPetagnia andLagoecia an independent phylogenetic development within theSaniculoideae is assumed.

Summary Mesophyll containing the minor veins from leaves ofTetragonia expansa Murr. was examined in preparation for a study of effects of beet yellows virus on the leaf tissues of this plant. The sieve elements throughout the minor veins exhibit the characteristics commonly found in this type of cell in dicotyledons. The cells are connected with one another by sieve plates and with contiguous parenchyma cells by branched plasmodesmata. Mature sieve elements are enucleate and lack ribosomes. No tonoplast was discerned in these cells. Mitochondria, plastids, and sparse endoplasmic reticulum are retained in mature cells. The plastids, which contain a massive fibrous ring of proteinaceous material, resemble the sieve element plastids ofBeta. The P-protein in the sieve elements is occasionally composed of tubules; more commonly it is represented by loose helices. The tracheary elements have scalariform secondary thickenings. In regions free of these thickenings, the primary wall largely disintegrates; only some loosely arranged fibrils remain. The mesophyll and vascular parenchyma cells contain the various organelles characteristic of living plant cells but vary in degree of vacuolation and in density of cytoplasm. Some vascular parenchyma cells have particularly dense protoplasts. They contain numerous ribosomes and their background matrix consists of a dense population of fine fibrils. Stem tissue of Lycopodium lucidulum Michx. was fixed in glutaraldehyde and postfixed in osmium tetroxide for electron microscopy. Although their protoplasts contain similar components, immature sieve elements can be distinguished from parenchymatous elements of the phloem at an early stage by their thick walls and correspondingly high population of dictyosomes and dictyosome vesicles. Late in maturation the sieve-element walls undergo a reduction in thickness, apparently due to an “erosion” or hydrolysis of wall material. At maturity, the plasmalemma-lined sieve elements contain plastids with a system of much convoluted inner membranes, mitochondria, and remnants of nuclei. Although the endoplasmic reticulum (ER) in most mature sieve elements was vesiculate, in the better preserved ones the ER formed a tubular network closely appressed to the plasmalemma. The sieve elements lack refractive spherules and P-protein. The protoplasts of contiguous sieve elements are connected with one another by pores of variable diameter, aggregated in sieve areas. As there is no consistent difference between pore size in end and lateral walls these elements are considered as sieve cells.  相似文献

The cytochemical localization of ATPase in differentiating and mature phloem cells of Pisum sativum L. has been studied using a lead precipitation technique. Phloem transfer cells at early stages of differentiation exhibit strong enzyme activity in the endoplasmic reticulum (ER) and some reaction product is deposited on the vacuolar and plasma membranes. As the phloem transfer cells mature and develop their characteristic wall structures, strong enzyme activity can be observed in association with the plasma membranes and nuclear envelopes. Mature phloem transfer cells with elaborate cell-wall ingrowths show ATPase activity evenly distributed on plasma-membrane surfaces. Differentiating sieve elements show little or no enzyme activity. When sieve elements are fully mature they have reaction product in the parietal and stacked cisternae of the ER. There is no ATPase activity associated with P-protein at any stage of sieve-element differentiation or with the sieve-element plasma membranes. It is suggested that the intensive ATPase activity on the plasma membranes of the transfer cells is evidence for a transport system involved in the active movement of photosynthetic products through these cells.Key to labeling in the figures ER endoplasmic reticulum - P parenchyma cell - PP P-protein - SE sieve element - SPP sieve-plate pore - TC transfer cell  相似文献

In a mature mixed subalpine stand ofTsuga mertensiana andAbies amabilis, significantly higher Al levels were found in foliage, branch and root tissues ofT. mertensiana.Tsuga mertensiana had significant increases in Al, Ca and Mn levels with increasing foliage age. In current foliage,T. mertensiana had lower levels of Ca, similar levels of Mg and P, and higher levels of Mn thanA. amabilis. Both tree species had Cu and Fe present at higher levels in branch than foliage tissues. Fine roots had the highest concentrations of Al, Fe and Cu but the lowest Ca and Mn concentrations of all tissues analyzed. In the roots of both species, phloem tissues always had significantly higher Al levels than xylem. Fine roots (< 1 and 1–2 mm) ofT. mertensiana had higher Al levels than were found inA. amabilis. Roots greater than 2 mm in diameter exhibited no significant differences in Al levels in phloem or xylem tissue betweenA. amabilis andT. mertensiana. The two species show a clear difference in their ability to accumulate specific elements from the soil.  相似文献

The occurrence ofSalmonella in endemic and subendemic species of lizard and frog of Gran Canaria,Gallotia stehlini andRana perezi, as well as captive reptiles from other regions of the world was investigated. The occurrence ofSalmonella was statistically higher in endemic and subendemic species than in captive animals (p<0.001). Seventy strains ofSalmonella were isolated.S. berta andS. grancanaria were the most frequently isolated serotypes. The study ofSalmonella in gall-bladder contents showed a high parasitation (85%), being higher inGallotia stehlini (100%) than inRana perezi (60%). None of the isolated salmonellae were resistant to tested antibiotics.  相似文献
