A probe to study the toxic interaction of tartrazine with bovine hemoglobin at the molecular level 下载免费PDF全文
Tartrazine is an artificial azo dye commonly used in food products, but tartrazine in the environment is potentially harmful. The toxic interaction between tartrazine and bovine hemoglobin (BHb) was investigated using fluorescence, synchronous fluorescence, UV–vis absorption, circular dichroism (CD) and molecular modeling techniques under simulated physiological conditions. The fluorescence data showed that tartrazine can bind with BHb to form a complex. The binding process was a spontaneous molecular interaction, in which van der Waals' forces and hydrogen bonds played major roles. Molecular docking results showed that the hydrogen bonds exist between the oxygen atoms at position 31 of tartrazine and the nitrogen atom NZ7 on Lys99, and also between the oxygen atoms at position 15 of tartrazine and the nitrogen atom NZ7 on Lys104, Lys105. The results of UV–vis and CD spectra revealed that tartrazine led to conformational changes in BHb, including loosening of the skeleton structure and decreasing α helix in the secondary structure. The synchronous fluorescence experiment revealed that tartrazine binds into the hemoglobin central cavity, and this was verified using a molecular modeling study. Maltol, a food additive, is extensively used in our daily life. To date, its biological safety is still debated. In this article, binding interaction of maltol with bovine hemoglobin (BHb), an important functional protein, was studied by molecular docking research and spectroscopic and calorimetric measurements. We found that maltol could cause structural changes of BHb. By interacting with Glu 101 (1.27 Å) and Lys 104 (2.49 Å) residues, maltol changed the cavity structure and induced a microenvironment change around tryptophan (Trp) residue. Thermodynamic parameters obtained from isothermal titration calorimetry (ITC) measurement showed that hydrophobic forces were the main forces existing in this system. The association constant of K (8.0 ± 3.4 × 104 M?1) shows the mild ligand–protein binding for maltol with BHb. The α‐helix amount in BHb increased (59.6–62.6%) with different concentrations of maltol and the intrinsic fluorescence intensity was quenched by maltol, indicating the conformation changes and denaturation of BHb. This work presents the interactions of maltol with BHb at the molecular level and obtains evidence that maltol induces adverse effects to proteins in vitro. 相似文献

The treatment of Rutaceae in the Chinese flora chose to follow Engler in recognizing Rutoideae and Toddalioideae as two separate subfamilies. Morphological and chemical comparisons, however, suggested group-ing those two subfamilies in one subfamily, Rutoideae. This move has received support from molecular phyloge-netic analyses, which also showed that the Chinese taxa in Euodia should be placed in Tetradium and Melicope following Hartley. Investigations into the chemistry and molecular phylogeny of Murraya also indicated that the species in the section Bergera without yuehchukene should be removed from Murraya. These findings clearly show the value of molecular cladistics to the taxonomy of Rutaceae in China and also directions for further inves-tigations. 相似文献

I investigate the extent to which interviewers and respondents in a 1995 national survey consistently classify race in Brazil, overall and in particular contexts. Overall, classification as white, brown or black is consistent 79 per cent of the time. However, persons at the light end of the colour continuum tend to be consistently classified, whereas ambiguity is greater for those at the darker end. Based on statistical estimation, the findings also reveal that consistency varies from 20 to 100 per cent depending on one's education, age, sex and local racial composition. Inconsistencies are in the direction of both ''whitening'' and ''darkening'', depending on whether the reference is interviewer or respondent. For example, interviewers ''whitened'' the classification of higher educated persons who self-identified as brown, especially in mostly non-white regions. Finally, I discuss the role of the Brazilian state in constructing race and the implications of these findings for survey research and comparative analysis. 相似文献

We report the results of a study which aim was the mycological identification of specimens coming from patients included in a clinical trial. A total of 445 specimens from patients with clinical diagnosis of dermatomicosis were processed during 8 months (138 pityriasis versicolor, 28 cutaneous candidosis and 279 dermatophytosis). A 48% of pityriasis versicolor cultures were positive for Malassezia furfur, 50% of candidosis cultures were positive for yeasts and 67% of dermatophytosis cultures were positive for dermatophytes. According to our results Candida albicans was the principal causative agent for cutaneous candidosis and Trichophyton mentagrophytes and Trichophyton rubrum were the most frequent isolated species causing dermatophytosis. 相似文献

Exploratory statistical analysis of aminobiphenyl hemoglobin adduct data obtained in a large-scale population study was performed to assess precision and sensitivity over the 7 years required to conduct the analyses. A time-dependent trend toward higher values was observed that may be attributable to aging of the internal standard used throughout the study. A several-fold improvement in sensitivity from the beginning to end of the study was also noted. Repeated analysis of duplicate blood specimens provided a worst-case estimate of the coefficient of variation to be 0.31, attributable almost entirely to sample preparation rather than instrumental analysis. Substantial variability in calibration curves for the deuterated internal standard (standard deviation was +/-15% of the mean) was observed. The results obtained here will be used in support of further analyses of the data with respect to factors of epidemiological interest. 相似文献
