The development of the stratified squamous epithelium of the tympanic membrane and external canal was studied in 167 embryonic, fetal, and postnatal human ears. It originates as a tube derived from the epithelium of the fundus of the primary external canal (zone 1). The tube is composed of a thin, flat epithelium on the medial side (zone 2), continuous with a thicker one (zone 3) on the lateral side; zone 3 thereafter merges with the external epithelium of the primary external canal (zone 4). Proliferative activity, as indicated by a thickened epithelium, with rete ridges in later fetal life, is present mainly in zones 1 and 3. Cornification at 18 weeks gestation is followed by clearing of keratinous debris to the exterior. Subsequently the canal widens, zone 1 now covering the pars flaccida region, a tongue-shaped area passing inferiorly from it and a part of the postero-superior deep canal adjacent to it; zone 2 covers the pars tensa and zone 3 most of the deep external canal. On the basis of the original embryonic growth, migratory epithelial movement throughout life is postulated to be generated in zone 1 by mitotic interposition and then to pass to zone 2. It then moves en masse through to zone 3, where unilateral progression by mitotic means takes the epithelium up to the cartilaginous canal. Such a pathway is approximately that seen in the marked, living eardrum and canal.  相似文献

The infective third-stage juvenile of Trichostrongylus colubriformis is surrounded by its own cuticle as well as the incompletely moulted cuticle of the second-stage juvenile, which is referred to as the sheath. The sheath comprises an outer epicuticle, an amorphous cortical zone, a fibrous basal zone and an inner electron-dense layer. The basal zone of the sheath consists of three layers of fibres; the fibres are parallel within each layer, but the fibre direction of the middle layer is at an angle to that of the inner and outer layers. The cuticle comprises a complex outer epicuticle, an amorphous cortical zone and a striated basal zone. The lateral alae of the cuticle and the sheath are aligned and overlie the lateral hypodermal cords. The lateral alae of the sheath consist of two wing-like expansions of the cortical zone with associated specializations of the inner electron-dense layer which form a groove. The cuticular lateral alae consist of two tube-like expansions of the cortical zone. The lateral alar complex of the cuticle and the sheath may maximise locomotory efficiency and prevent rotation of the juvenile within the sheath.  相似文献

Fenestration patterns in endothelial cells of rat liver sinusoids   总被引:2,自引:0,他引:2  
Endothelial fenestrae of both zone 1 and zone 3 acinar liver sinusoids have been studied in rats by an interactive analysis of scanning electron microscopical images. Two fenestration patterns have been recognized in the endothelial cells on the basis of local variation in size, distribution and clustering of pores in each acinar zone. Our data indicate that both the number of fenestrae per square micrometer of endothelial surface and the mean diameter of fenestrae are significantly larger in zone 3 than in zone 1. The number of sieve plates is about 1.74 times larger in zone 3 than in zone 1, and the number of fenestrae per plate in zone 3 is nearly twice that in zone 1. Two different classes of fenestrae have been considered: clustered pores, which prevail in zone 3 and have a mean diameter smaller than the other pores, and free pores, which prevail in zone 1 and are bigger.  相似文献

Bacteriophage DNAs annealed into linear oligomeric concatemers were used to examine the quantitative pulsed-field gel electrophoretic behavior of different-sized DNAs as a function of electrical field strength and pulse time. Three zones of resolution are observed for increasingly larger DNAs. In the first two zones, the electrophoretic mobility decreases linearly with increasing DNA size. The separation in zone 2 is roughly twice that in zone 1. The largest DNA molecules do not resolve at all and migrate in a compression zone. Mobility in zone 1 increases linearly with the electric field strength and decreases with the inverse of the pulse time. The behavior of DNA in zone 2 is qualitatively similar. However, the effect of field strength and pulse time on the separations in each zone is quite different. The results for zone 1 are generally consistent with the predictions of several existing physical models of pulsed-field gel electrophoresis, but no model accounts for all of the observed behavior in the three zones.  相似文献

确定了小叶锦鸡儿、中间锦鸡儿和柠条锦鸡儿3种植物的分布式样:小叶锦鸡儿为蒙古高原东部-F松辽平原西部-华北山地分布种,中间锦鸡儿为东戈壁-鄂尔多斯高原-黄土高原北部分布种,柠条锦鸡儿为南阿拉善-西鄂尔多斯分布种。小叶锦鸡儿适应分布于蒙古高原典型草原带和森林草原带以及华北山地落叶阔叶林带,在草原带高平原上可形成灌丛化草原的景观,在草原带的沙地上可形成以小叶锦鸡儿为建群种的沙地灌丛植被;中间锦鸡儿适应分布于蒙古高原的荒漠化草原及草原化荒漠带、鄂尔多斯高原的典型草原和荒漠化草原带的沙地及梁地上、黄土高原北部的黄土丘坡上,常形成以中间锦鸡儿为建群种的沙地灌丛植被;柠条锦鸡儿则适应分布于草原化荒漠和典型荒漠带的固定和半固定沙地上。近缘的3种锦鸡儿的地带性分布从东至西或从北向南形成明显的有规律的地理替代分布格局。  相似文献

Ghrelin is the endogenous ligand for the growth hormone secretagogue receptor. We investigated the distribution and morphological characteristics of ghrelin-immunopositive (ghrelin-ip) cells in the African ostrich adrenal gland. We found that the adrenal gland of the African ostrich consisted of three parts: capsule, inter-renal tissue and chromaffin cells. The inter-renal tissue and chromaffin cells interdigitated irregularly. The inter-renal tissue consisted of a peripheral zone and a central inner zone. The peripheral zone could be divided into an outer subcapsular zone and an inner zone. The subcapsular zone cells were arranged as a bow, while the inner area cells formed cords that were perpendicular to the capsule. The central inner zone exhibited irregular clumps and the cells were morphologically similar to chromaffin cells. Ghrelin-ip cells were located throughout the adrenal gland except the capsule. The majority of ghrelin-ip cells were found among the chromaffin cells. The number of ghrelin-ip cells in the inter-renal tissue decreased gradually from the central inner zone, to the inner zone to the subcapsular zone. The ghrelin-ip cells were oval or irregular in shape and exhibited cytoplasmic staining. Our findings suggest that ghrelin may play a role in regulating adrenal hormone secretion in the African ostrich.  相似文献

An otolith increment is composed of an incremental zone and a discontinous zone. Observation of the otoliths of Oreochromis niloticus with transmission and scanning electron microscopy reveals that organic fibers are concentrated in the discontinuous zone and are relatively scarce in the incremental zone. On the other hand, calcium carbonate crystals are chiefly packed in the incremental zone and are less dense in the discontinuous zone. Both fibers and crystals are oriented perpendicular to the growth increments. Otolith checks, or discontinuities, contain even denser fibers and fewer fine crystals than does the discontinuous zone. A higher proportion of individual fibers is prominently stained in the discontinuous zone and check than in the incremental zone. Other features of individual fibers appear to be the same among the three zones. The crystals on either side of a check or a discontinuous zone resemble each other both in size and orientation. Zonation of protein and mineral components of otoliths can be understood in terms of both incremental growth and the daily cycle of deposition.  相似文献

Gravity regulates peg formation because cucumber seedlings grown in a horizontal position develop a peg on the lower side of the transition zone (TR zone) but not on the upper side. Studies on peg formation have suggested the regulation of peg formation by gravity as follows. Cucumber seedlings potentially develop a peg on both the lower and upper sides of the TR zone. The development of the peg on upper side of the TR zone is suppressed in response to gravity. A phytohormone, auxin, induces peg formation. Upon gravistimulation the auxin concentration on the upper side of the TR zone is reduced to a level below the threshold value necessary for peg formation. The unequally distributed auxin across TR zone is caused by a change in accumulation of auxin influx carrier (CsAUX1) protein and auxin efflux carrier (CsPIN1) protein in response to gravity. In addition, TR zone before peg initiation expresses both CsARF2 (putative activator of auxin response factor) and CsIAA1 (putative repressor of auxin-inducible gene expression), by which TR zone could respond the auxin gradient regulated by gravity.  相似文献

Summary and Conclusions The shoot apices ofAraucaria columnaris andA. bidwillii exhibit a tunica-corpus organization. In the former the tunica is one-layered during spring and summer months and becomes more stratified during winter months. InA. bidwillii it is usually two-layered, with a tendency to further stratification.The corpus is divisible into three zones, the corpus initial zone, the flanking zone and the rib meristem zone. The corpus initial zone is the seat of formation of the rest two. The flanking zone is concerned with the production of leaf primordia, in which the covering tunica layer has no part.The rib meristem zone varies in depth according to the season of the year and the pith cells mature gradually in these species.InA. columnaris, where a seasonal study was undertaken, no period of complete dormancy was observed.  相似文献

The light-microscopical and ultrastructural differentiation of the disseminate prostate was studied in 12 boars and 8 barrows, ranging in age from 8 to 28 and 12 to 27 weeks, respectively. The barrows had been castrated at the age of 8 weeks. In young boars the prostate is organized in an inner zone and an outer zone. The glandular tubules of the latter are more differentiated than those in the inner zone. Sexual maturation in the prostate is characterized by almost complete reduction of the inner zone and concomitant enlargement of the outer zone. This differentiation of the inner zone occurs progressively and is manifested by a higher density of the glandular tubules, by a decrease in the number of basal cells and by an increase in the secretory tubular cells containing more microvilli, organelles and secretory vesicles. A few weeks after castration, the characteristic morphology of the undifferentiated inner zone is found throughout the entire glandular layer. The number of glandular tubules in barrows has decreased, and their cells show minimal secretory activity, nuclear indentations and a decrease in their organelles. A further castration effect is vacuolization of the perinuclear cytoplasm in numerous glandular cells.  相似文献

A new method for studying macromolecular interactions was devised. The principle is based on affinity chromatography with a mobile zone of affinity ligand instead of a column with immobilized ligand. In this method, the difference in migration velocities between the moving zone of affinity ligand and a sample in a conventional gel-permeation column is utilized. The fast migrating zone (A zone), which is later injected into the column, and the slow migrating zone (B zone), which is injected beforehand, interfere with each other at the passing point in the column if A and B interact such as A + B ? AB. The zone interference deforms each elution profile of A and B, because the complex AB has a migration velocity different from the others. The elution profiles in zone-interference chromatography are calculated by computer simulation in the framework of the plate theory. The binding constant is calculated from the peak shift of elution volume of B in the zone-interference chromatogram. The interaction between single-strand DNA (A zone) and ribonuclease A (B zone) was studied.  相似文献

The secondary X zone induced by castration in the adrenal cortex of adult male mice was examined by electron microscopy and radioautography with 3H-thymidine. 10-15 days after castration a thin layer of small eosinophilic cells is formed in the inner-most cortex. Such eosinophilic cells contain irregulary shaped nuclei and spherical or ellipsoidal mitochondria with tubulolamellar cristae, 20-25 days after castration a prominent zone of small eosinophilic cells was clearly identified as the secondary X zone. The typical secondary X zone cells were characterized by the formation of peculiar mitochondrial complexes and whorled sER. The X zone cells with their characteristic organelles incorporated 3H-thymidine. The ultrastructure and formation of the secondary X zone were very similar to those of the primary X zone which appears during normal postnatal development. We demonstrate here the capacity of reticularis cells of adult male mice to transform into typical X zone cells following castration.  相似文献

Rapid patterning has been observed in confined 2-D cultures of Dictyostelium discoideum Ax-2 cells as an outer dark zone and a inner light zone. The width of outer zone was usually approximately100 microm, irrespective of the size of cell masses under atmospheric conditions. The width of the outer zone, however, changed depending on external O2 concentrations and reached up to 250 microm at 100% O2. A clear regional difference in tetramethyl rhodamine methyl ester (TMRM) staining was noticed between the outer zone and the inner zone: the inner zone was more strongly stained with TMRM than the outer zone, which faced the air. Using inhibitors of oxidative phosphorylation (dinitrophenol (DNP) or NaN3) and a specific inhibitor of CN-resistant respiration (benzohydroxamic acid (BHAM)), it has been demonstrated that the outer zone is basically formed by the O2 threshold for oxidative phosphorylation, while the inner cells mainly perform cyanide-resistant respiration. When cells around the early mound stage (just before prestalk and prespore differentiation) were cultured as 2-D cell masses, ecmA-expressing cells (pstA cells), ecmB-expressing cells (pstB cells) and D19-expressing cells (prespore; psp cells), arose in a position-dependent manner in the outer zone. In the inner zone, cell motility seemed to be markedly impaired and neither prestalk nor prespore differentiation occurred. In addition, once-differentiated prespore cells were found to dedifferentiate rapidly in the inner zone. The reason for dedifferentiation as well as for failure of cells to differentiate in the inner zone is discussed with reference to O2 radicals.  相似文献

This study characterizes a hybrid zone that spans a migratory divide between subspecies of the Swainson's thrush (Catharus ustulatus), a long distance migratory songbird, in the Coast Mountains of British Columbia. To assess the potential for a barrier to gene flow between the subspecies, I: (1) analyzed the shape and width of genetic and morphological clines relative to estimates of dispersal distance, (2) assessed the ratio of parental to hybrid genotypes across the hybrid zone, (3) estimated population density across the hybrid zone, and (4) compared the spatial relationship between the hybrid zone and an existing environmental gradient. The results indicate that the hybrid zone is characterized by mostly concordant character clines that are narrow relative to dispersal, the absence of a hybrid swarm, and low population density at the center of the zone. This hybrid zone and additional regions of contact between these subspecies are found on the border between coastal and interior climatic regions throughout the Pacific Northwest. An identified shift in the location, but not the width, of the mtDNA cline relative to the nuclear clines is consistent with asymmetrical hybridization. Neutral diffusion of populations following secondary contact and hybrid superiority within an ecotone are insufficient explanations for the observed patterns. The hypothesis that best fits the data is that the Swainson's thrush hybrid zone is a tension zone maintained by dispersal and ecologically mediated barriers to gene flow.  相似文献

The cephalolichen Peltigera aphthosa (L.) Willd. is characterized by lateral heterogeneity, which manifests itself in the presence of three thallus zones, referred to as the apical, basal and medial zone. These zones differ in terms of interaction between lichen bionts and their physiological activity. The apical thallus zone is more efficient in establishing a contact with cyanobacteria, because of a higher lectin content and a larger overall thallus surface area due to the presence of numerous mycobiont hyphae. Cephalodia are formed in this zone. The interaction between the mycobiont and cyanobiont is more intense in the medial zone. However, the establishment of the contact with cyanobacteria in this zone less probable. The spatial distribution of lectins in the thallus was determined. To reveal the differences in photosynthetic activity in three thallus zones, transient analysis of chlorophyll a fluorescence and the assessment of non‐photochemical quenching of excited chlorophyll states were performed. Assimilation of absorbed light energy was more effective in the medial zone. The basal zone was characterized by decreased photosynthetic activity, lichen dissociation and thallus death.  相似文献

当环境中可利用的资源低于种群最佳生长需要时,种群内个体之间就会产生竞争。竞争常发生在邻近植物之间,植物地上部分和地下部分的竞争范围是不同的。目前,有很多度量竞争强度和确定竞争范围的方法,能否对这些方法进行改进,提出更具有说服力的方法,是本研究的出发点。黄山松是常见的针叶树种,分布范围广,生存能力强,是亚热带中部中山地区代表群系的建群种,也是较高海拔地区重要的造林树种,具有较强的代表性。对黄山松的邻体竞争研究发现:(1)改进的竞争指数有更强的说服力;(2)逐步扩大范围的方法能很好地确定植株间的竞争范围;(3)邻体竞争强度随影响范围的增加而增加,在一定的范围内增加的较快,而超出该范围后增加的幅度变小,可以此为依据来确定邻体范围;(4)不同径级的基株,邻体范围有一定的差异;(5)邻体竞争强度和影响范围的关系服从对数函数关系(CI=AlnC+B)。结果表明,改进的竞争指数能很好地度量竞争强度,采用逐步扩大范围的方法能有效地确定邻体范围。  相似文献

The shoot apex of Bougainvillea spectabilis consists of five zones: A two- or occasionally three-layered tunica, a central mother cell zone, a cambium-like zone, a rib meristem (central meristem), and a peripheral meristem. The presence of a cambium-like zone is somewhat unusual in the apex of vascular plants, having only been reported for a few taxa. In B. spectabilis the cambium-like zone is consistently present throughout the plastochron and all yearly seasonal periods.  相似文献

The transitional region (including the end of the secretion zone, the beginning of the maturation zone, and the transition zone between them) was studied in the lower incisor enamel organ of adult rats with the electron microscope. Towards the end of the secretion zone, the surface invaginations of the ameloblasts diminish, the enamel surface becomes smooth and a dense, granulated material appears in the extracellular spaces between ameloblasts. The presence of a 60 Å wide gap between enamel and ameloblasts, and a 100 Å thick enamel border are thought to indicate the end point of enamel secretion and the beginning of the transition zone. Ameloblasts begin to shorten very close to this point. Subsequently, the following events occur: (1) the ameloblast cell membrane lifts away from the enamel to form a 450 Å wide gap; (2) before completion of this gap, a granular material appears in vesicles within the ameloblast apex, possibly to be secreted into the gap; (3) half-desmosomes are formed at the apical cell membrane; (4) the extracellular dense material passes into the spaces between the papillary cells; finally, (5) a coarse-textured material, thought to be enamel constituents being resorbed, appears in the gap, indicating the end of the transition zone. Only following this is the 450 Å gap completed. The attachment sites of both apical and basal terminal bars remain intact throughout the transition zone. The length of the transition zone is about 170 μ. The morphologic features of the transition zone overlap each other and persist for various distances into the maturation zone.  相似文献

In contrast to the vitellolysis zone which is involved in the degradation of the yolk, the cytoplasmic zone of the yolk syncytial layer, composed of many cytoplasmic organelles, is implicated in a secretory process. The granular endoplasmic reticulum of the latter is extremely well developed and organized into trabeculae. The yolk nuclei show a RNA positive reaction. The Golgi apparatus is implicated in the elaboration of very low density lipoproteins (VLDL) and of acid phosphatase. The numerous mitochondria seem to suggest an important energy metabolism in the solubilisation area. The appearance of certain special structures (crystalline bodies, pseudovesicular structures, lipochondria) as well as their relation with the organelles of the cytoplasmic zone, re-inforces the impression of a layer with a secretory nature and suggests its participation in the remodelling of the yolk products produced in the vitellolysis zone. The results of the investigations concerning the acid phosphatase activity suggest that this enzyme plays a role on the one hand in the degradation of certain platelets which penetrate into the cytoplasmic zone, and on the other hand in the regulation of VLDL, in the lysis of secretory products elaborated by the cytoplasmic zone. The possible presence of microperoxisomes is discussed as well as the positive detection of glucose-6-phosphate dehydrogenase.  相似文献

Cell sorting within the prestalk zone of Dictyostelium discoideum   总被引:2,自引:0,他引:2  
Abstract. The prestalk zone of slugs of Dictyostelium discoideum has been shown to contain three subregions in which the extracellular matrix genes ecmA and ecmB are differentially expressed; it is generally thought that these regions are defined by extracellular signals. Using β-galactosidase as a cell marker, we have shown that cells can sort specifically to all three regions. Cells from the posterior-prestalk zone ("prestalk 0 zone") which are injected into the slug tip move within 60 min back to their position of origin. When cells from the anterior prestalk zone (presumably containing a mixture of ecmA and ecmB expressers) are transplanted to the posterior prestalk zone, they move to the tip ("prestalk A zone") within 1 h and about 30 min subsequently are often found in a cone-shaped region within the tip ("prestalk B zone"). Cells transplanted to their own positions do not move significantly within this period. Since the sub-regions of the prestalk zone can be defined by sorting, it is possible that they are normally formed in this way rather than by position-dependent signals. Cells transplanted without a change in anterior-posterior position and cells which have sorted back to their positions of origin eventually spread out throughout the prestalk zone. This suggests that sorting preferences of cells are respecified. When posterior prestalk cells are transplanted to the prespore zone, respecification of sorting preference is suspended until the cells return to the prestalk zone and anterior-prestalk cells acquire posterior-prestalk sorting preferences.  相似文献
