For growing roots of azuki bean (Phaseolus chrysanthos), an electric potential is measured minutely along the surface of the root, together with the surface pH. It was found that the root begins to display a band-type pattern of potential with a spatial period of about 2 cm in a mature region as soon as it grows to about 10 cm in root length, while the surface potential shows only one convex peak around a position 5-20 mm behind a root tip and a succeeding concave peak around 20-35 mm, providing the length of root is shorter than about 10 cm. Since the surface potential takes a relatively positive value on average at the side of the root base compared with that in an elongation zone near the tip, electric current is supposed to flow into the elongation zone, accompanied by some local current loops in the mature region. The present band-type pattern observed first in multi-cell systems seems to be a kind of dissipative structure appearing far from equilibrium, and hence its relationship to growth is discussed.  相似文献

The electric spatial pattern and invertase activity distribution in growing roots of azuki bean (Phaseolus chrysanthos) have been studied. The electric potential near the surface along the root showed a banding pattern with a spatial period of about 2 cm. It was found that the enzyme activity has a peak around 3-7 mm from the root tip, in good agreement with the position of the first peak of the electric potential, which is located a little behind the elongation zone. An inhomogeneous distribution of ATP content was also detected along the root. Experiments on the electric isolation of the elongation zone from the mature zone and acidification treatment showed that H+ is transported from the mature-side to elongation-side regions, causing tip elongation through an acid-growth mechanism. Both acidification and electric disturbance on growing roots affected growth significantly. Simultaneous measurements of electric potential and enzyme activity clearly showed a good correlation between these two quantities and growth speed. From an analogy with the Characean banding, the spatio-temporal organization via the cell membrane in electric potential and enzyme activity can be regarded as a dissipative structure arising far from equilibrium. These experimental results can be interpreted with a new mechanism that the dissipative structure is formed spontaneously along the whole root, accompanied by energy metabolism, to make H+ flow into the root tip.  相似文献

To identify the region in which a root perceives a decrease in the ambient water potential and changes its elongation rate, we applied two agar blocks (1 x 1 x 1 mm(3)) with low water potential bilaterally to primary roots of maize (Zea mays) at various positions along the root. When agar blocks with a water potential of -1.60 MPa (-1.60-MPa blocks) or lower were attached to a root tip, the rate of elongation decreased. This decrease did not result from any changes in the water status of elongating cells and was not reversed when the -1.60-MPa blocks were replaced by -0.03-MPa blocks. The rate decreased slightly and was unaffected, respectively, when -1.60-MPa blocks were applied to the so-called decelerating region of the elongating zone and the mature region. However, the rate decreased markedly and did not recover for several hours at least when such blocks were attached to the accelerating region. In this case, the turgor pressure of the elongating cells decreased immediately after the application of the blocks and recovered thereafter. The decrease in elongation rate caused by -1.60-MPa blocks applied to the root tip was unaffected by additional -0.03-MPa blocks applied to the accelerating region and vice versa. We concluded that a significant reduction in root growth could be induced by water stress at the root tip, as well as in the accelerating region of the elongating zone, and that transmission of some signal from these regions to the decelerating region might contribute to the suppression of cell elongation in the elongation region.  相似文献

High-resolution profiles of surface pH and growth along vertically growing maize (Zea mays) primary root tips were determined simultaneously by pH-sensitive microelectrodes and marking experiments. Methodological tests were carried out that proved the reliability of our kinematic growth analysis, while questioning the validity of an alternative technique employed previously. A distal acidic zone around the meristematic region and a proximal one around the elongation zone proper were detected. This pattern as such persisted irrespective of the bulk pH value. The proximal acidic region coincided with maximum relative elemental growth rates (REGR), and both characters reacted in a correlated manner to auxin and cyanide. The distal acidic band was unrelated to growth, but was abolished by cyanide treatment. We conclude that: (a) the pattern of surface pH as such is a regulated feature of growing root tips; (b) the correlation of extracellular pH and growth rate suggests a functional relationship only along proximal portions of the growing root tip; and (c) the distal acidic band is not caused by pH buffering by root cap mucilage, as suggested previously, but rather is controlled by cellular activity.  相似文献

Possible energy transmission along conjugating membranes was investigated. It was found by extracellular electrodes that under local illumination of blue-green algae Phormidium uncinatum the electrical current propagated along trichomes. The form and sign of the responses show that e. m. f. appears in the illumination region, which generates on the cellular membrane the potential difference with a negative charge inside the cell. The initiated transmembrane potential difference propagates along the filament of Ph, uncinatum like along a cable. The hypothesis concerning a cable structure of Ph, uncinatum was supported in the experiments with intersection of algae, and with intracellular injection of fluorescein showing the existence of direct diffusion exchange through cellular interfaces.  相似文献

Extracellular and intracellular electric potentials in bean roots are known to show electric oscillations along the longitudinal axis with a period of several minutes. The relationship between growth and the electric oscillations was studied using roots of adzuki (Phaseolus chrysanthos). We measured surface electric potentials with a multielectrode apparatus while simultaneously measuring elongation using a CCD camera and monitor. Roots having an electric oscillation grew faster than roots with no oscillation. Furthermore, elongation rate was higher in roots with higher oscillation frequency. Oscillation frequency had a strong dependence on temperature; i.e. Q10 was estimated at 1.7. These results suggest a correlation between electric oscillation and elongation.  相似文献

Growth of elongating primary roots of maize (Zea mays) seedlings was approximately 50% inhibited after 48 h in aerated nutrient solution under water deficit induced by polyethylene glycol 6000 at -0.5 MPa water potential. Proton flux along the root elongation zone was assayed by high resolution analyses of images of acid diffusion around roots contacted for 5 min with pH indicator gel. Profiles of root segmental elongation correlated qualitatively and quantitatively (r(2) = 0.74) with proton flux along the surface of the elongation zone from water-deficit and control treatments. Proton flux and segmental elongation in roots under water deficit were remarkably well maintained in the region 0 to 3 mm behind the root tip and were inhibited from 3 to 10 mm behind the tip. Associated changes in apoplastic pH inside epidermal cell walls were measured in three defined regions along the root elongation zone by confocal laser scanning microscopy using a ratiometric method. Finally, external acidification of roots was shown to specifically induce a partial reversal of growth inhibition by water deficit in the central region of the elongation zone. These new findings, plus evidence in the literature concerning increases induced by acid pH in wall-extensibility parameters, lead us to propose that the apparently adaptive maintenance of growth 0 to 3 mm behind the tip in maize primary roots under water deficit and the associated inhibition of growth further behind the tip are related to spatially variable changes in proton pumping into expanding cell walls.  相似文献

A model of electrical activity in the heart has been developed that treats the intracellular domain and the extracellular domain as electrical syncytia with anisotropic resistivities (bi-syncytial model). At the microscopic level, propagation is assumed to proceed primarily along the axes of individual cells. Considerations at the macroscopic level relate the transmembrane current to the intracellular and extracellular resistivity and the transmembrane potential. The result is a relationship between instantaneous extracellular potentials and cardiac action potentials.  相似文献

One to two cm long root tips of Triticum aestivum L., after three days' germination at 25 ℃, each were cut into three regions--the meristem region, elongation region and mature region. The cells in different regions were stained with Hochest 33258, Pymnin G and FITC respectively. Nuclear DNA was measured by micmfluorometry with VIDAS digital image analysis system. The relative contents of RNA and protein in the cells were measured with a MPV-Ⅲ microspectrofluorometer. It was shown that the nuclear DNA content increased during root tip cell differentiation, being maximum in the mature region. The relative content of RNA was maximum in the meristem region, but decreased continuously during the growth of tissue. The relative content of protein was maximum in the elongation region, but minimum in the meristem region. The relationships among DNA, RNA, protein and cell differentiation were discussed.  相似文献

Abstract. Electrical potential differences (PDs) were measured continuously at six points in line on the surface of primary roots of cress ( Lepidium sativum L.). The root-to-hypocotyl transition was used as the reference. A gradient of extracellular potentials existed along the root axis. The most negative potentials were observed at the root tip. Whenever the growth rate declined to 0·10 mm h−1 or less, the PDs were nearly constant in space and time. Otherwise, the PDs showed different types of changes with time. Roots with growth rates between 0·65 and 1·10 mm h−1 preferably showed PDs characterized by approximately sinoidal oscillations. Maximal amplitudes appeared in the middle of the elongation zone. Roots which were exposed to a N2-atmosphere exhibited a reversible disappearance of the electrical oscillations and a decrease in the PD-gradient. Similar effects were obtained by the application of the metabolic inhibitor azide (N3). These results proved that the electrical oscillations are dependent on the aerobic cell metabolism.  相似文献

Gravity-induced changes in intracellular potentials in primary roots of 2-day-old mung bean (Vigna mungo L. cv. black matpe) seedlings were investigated using glass microelectrodes held by 3-dimensional hydraulic micro-drives. The electrodes were inserted into outer cortical cells within the elongation zone. Intracellular potentials, angle of root orientation with respect to gravity, and position within the root of the impaled cortical cell were measured simultaneously. Gravistimulation caused intracellular potential changes in cortical cells of the elongation zone. When the roots were oriented vertically, the intracellular potentials of the outer cortical cells (2 mm behind the root apex) were approximately - 115 mV. When the roots were placed horizontally cortical cells on the upper side hyperpolarized to - 154 mV within 30 s while cortical cells on the lower side depolarized to about - 62 mV. This electrical asymmetry did not occur in cells of the maturation zone. Because attempts to insert the electrode into cells of the root cap were unsuccessful, these cells were not measured. The hyperpolarization of cortical cells on the upper side was greatly reduced upon application of N,N'-dicyclohexylcarbodiimide (DCCD), an inhibitor of respiratory energy coupling. When stimulated roots were returned to the vertical, the degree of hyperpolarization of cortical cells on the previous upper side decreased within 30 s and approached that of cortical cells in non-stimulated roots. This cycle of hyperpolarization/loss of hyperpolarization was repeatable at least ten times by alternately turning the root from the vertical to the horizontal and back again. The very short (<30 s) lag period of these electrical changes indicates that they may result from stimulus-perception and transduction within the elongation zone rather than from transmission of a signal from the root cap.  相似文献

小麦(Triticum aestivum L.)种子在25℃条件下萌发3d,根生长至1~2cm长时,于双筒解剖镜下严格切取根分生区、伸长区和成熟区。用专一性荧光染料Hochest33258、Pyronin G和FITC分别染细胞核DNA、RNA和蛋白质,并用自动图像分析技术和细胞荧光测定术分别测定三个区中各125个细胞核DNA的相对含量和各100个细胞中RNA和蛋白质的相对含量。核DNA相对含量随着根尖细胞分化的进程,DNA含量递增,成熟区细胞中含量最高。RNA的相对含量则与之相反,在分生区细胞中含量最高,成熟区细胞中含量最低。蛋白质的相对含量则在伸长区细胞中最高,分生区细胞中最低。讨论了根尖细胞分化过程中DNA、RNA和蛋白质三者之间变化的一些内在联系。  相似文献

Al localization was compared in three developmental regions of primary root of an Al-sensitive soybean (Glycine max) genotype using secondary ion mass spectrometry. In cryosections obtained after a 4-h exposure to 38 [mu]M [Al3+], Al had penetrated across the root and into the stele in all three regions. Although the greatest localized Al concentration was consistently at the root periphery, the majority of the Al in each region had accumulated in cortical cells. It was apparent that the secondary ion mass spectrometry 27Al+ mass signal was spread throughout the intracellular area and was not particularly intense in the cell wall. Inclusion of some cell wall in determinations of the Al levels across the root radius necessitated that these serve as minimal estimates for intracellular Al. Total accumulation of intracellular Al for each region was 60, 73, and 210 nmol g-1 fresh weight after 4 h, increasing with root development. Early metabolic responses to external Al, including those that have been reported deep inside the root and in mature regions, might result directly from intracellular Al. These responses might include ion transport events at the endodermis of mature roots or events associated with lateral root emergence, as well as events within the root tip.  相似文献

In the present study, we investigated the alteration of reactive oxygen species production along the longitudinal axis of barley root tips during Cd treatment. In unstressed barley root tips, H2O2 production decreased from the root apex towards the differentiation zone where again, a slight increase was observed towards the more mature region of root. An opposite pattern was observed for O 2 ?? and OH? generation. The amount of both O 2 ?? and OH? was highest in the elongation zone, decreased in the root apex and at the differentiation zone of root, then increased again towards the more mature region of root. An elevated Cd-induced O 2 ?? production started in the elongation zone and increased further along the differentiation zone of barley root tip. In contrast, Cd-induced H2O2 production was localised to the root elongation zone and to the beginning of the differentiation zone. In contrast to Cd-induced H2O2 and O 2 ?? production, Cd reduced OH? production along the whole barley root tip. Our results suggest that not only an increase but also the spatial distribution of reactive oxygen species production is involved in the Cd-induced stress response of barley root tip.  相似文献

Onions (Allium cepa L.) treated with external ascorbic acid or with the immediate precursor of its synthesis L-galactono-gamma-lactone show a stimulated elongation rate of the roots and an increase in the number of new radicles appearing at the bulb base. Treatment with both molecules resulted in an enhanced accumulation of ascorbate and dehydroascorbate along the root axis, but the distribution of these redox forms was not uniform along the root, as detected in intracellular (symplastic) and extracellular (apoplastic) compartments. Thus, those radicular zones metabolically more active, such as the meristem and the elongation zone, accumulated the highest amount of both redox forms of ascorbate. On the other hand, ascorbate and L-galactono-gamma-lactone also stimulated cytosolic glucose-6-phosphate dehydrogenase activity and inhibited peroxidase activity as deduced from in vivo and in vitro experiments. Differences were also found when comparing apoplastic and symplastic activities. These results are compatible with the idea of an ascorbate-mediated stimulation of root growth by inhibiting cell wall stiffening and increasing root metabolism.  相似文献

The extracellular proton activity along primary roots of Phleum pratense L. was measured using proton-selective microelectrodes. Removal of the root cap caused a reduction of the proton influx in the transitional region between the meristem and the apical elongation zone of the vertical root and inhibited the development of pH differences between the physically upper and lower flanks of the gravistimulated root. Disruption of the actin filament system of the root with 5 mmol m-3 cytochalasin D did not result in an altered proton flux and pH pattern compared with untreated vertical control roots, but inhibited the gravity-induced development of pH differences between the physically upper and lower root flanks as well as gravitropic curvature. These results provide evidence that pH changes following gravistimulation are induced by a signal transmitted from the root cap and that the actin filament system is involved in the gravity perception/transduction mechanism.  相似文献

In Arabidopsis (Arabidopsis thaliana; Columbia-0) roots, the so-called zone of cell elongation comprises two clearly different domains: the transition zone, a postmeristematic region (approximately 200-450 μm proximal of the root tip) with a low rate of elongation, and a fast elongation zone, the adjacent proximal region (450 μm away from the root tip up to the first root hair) with a high rate of elongation. In this study, the surface pH was measured in both zones using the microelectrode ion flux estimation technique. The surface pH is highest in the apical part of the transition zone and is lowest at the basal part of the fast elongation zone. Fast cell elongation is inhibited within minutes by the ethylene precursor 1-aminocyclopropane-1-carboxylic acid; concomitantly, apoplastic alkalinization occurs in the affected root zone. Fusicoccin, an activator of the plasma membrane H(+)-ATPase, can partially rescue this inhibition of cell elongation, whereas the inhibitor N,N'-dicyclohexylcarbodiimide does not further reduce the maximal cell length. Microelectrode ion flux estimation experiments with auxin mutants lead to the final conclusion that control of the activity state of plasma membrane H(+)-ATPases is one of the mechanisms by which ethylene, via auxin, affects the final cell length in the root.  相似文献

Roots of the agravitropic pea (Pisum sativum L.) mutant, ageotropum, responded to a gradient in water potential as small as 0.5 MPa by growing toward the higher water potential. This positive response occurred when a sorbitol-containing agar block was unilaterally applied to the root cap but not when applied to the elongation region. Unilateral application of higher concentrations of sorbitol to the elongation region caused root curvature toward the sorbitol source, presumably because of growth reduction on the water-stressed side. The control blocks of plain agar applied to either the root cap or the elongation region did not cause significant curvature of the roots. These results demonstrate that hydrotropism in roots occurs following perception of a gradient in water potential by the root cap.  相似文献

The curvature of roots in response to gravity is attributed to the development of a differential concentration gradient of IAA in the top and bottom of the elongation region of roots. The development of the IAA gradient has been attributed to the redistribution of IAA from the stele to cortical tissues in the elongation region. The gravistimulated redistribution of IAA was investigated by applying [3H]IAA to the cut surface of 5 mm apical primary root segments. The movement of label from the stele-associated [3H]IAA into the root, tip, root cap, and cortical tissues on the top and bottom of the elongation region was determined in vertically growing roots and gravistimulated roots. Label from the stele moved into the region of cell differentiation (root tip) prior to accumulating in the elongation region. Little label was observed in the root cap. Gravistimulation did not increase the amount of label moving from the stele; but gravistimulation did increase the amount of label accumulating in cortical tissues on the lower side of the elongation region, and decreased the amount of label accumulating in cortical tissues on the upper side of the elongation region. Removal of the cap prior to or immediately following gravity stimulation rendered the roots partially insensitive to gravity and also prevented gravity-induced asymmetric redistribution of label. However, removal of the root cap following 30 min of gravistimulation did not alter root curvature or the establishment of an IAA asymmetry across the region of root elongation. These results suggest that a signal originating in the root cap directs auxin redistribution in tissues behind the root cap, leading to the development of an asymmetry of IAA concentration in the elongation region that in turn causes the differential growth rate in the elongation region of a graviresponding root.  相似文献

The mechanistic basis for cation amelioration of Al rhizotoxicity in soybean was investigated through a series of studies comparing protective effects of Ca and Mg against Al inhibition of root elongation in a background 0.8 mM CaSO4 solution (pH 4.3). A modified Gouy-Chapman-Stern model was used to evaluate the effect of cations on electrical potential and Al3+ activity at root plasma membrane surfaces. Activities of Al3+ up to 4.6 microM in the background solution inhibited soybean tap root elongation by more than 80%. There was little or no response in root elongation when Ca and Mg were added to background solutions in the absence of AL: When added to Al-toxic solutions in the micromolar concentration range, Mg was 100-fold more effective than Ca in alleviating Al toxicity, whereas both cations were equally effective when added in the millimolar concentration range. The protective effect of micromolar additions of Mg on root elongation was specific for Al and it failed to alleviate La rhizotoxicity. In contrast to wheat, Mg amelioration of Al toxicity to soybean root elongation at low Mg concentration could not be explained by changes in potential and Al3+ activity at the root plasma membrane surfaces as predicted by a Gouy-Chapman-Stern model. These results suggest that Mg is not acting as an indifferent cation when present at low concentration and implies the involvement of a mechanism other than pure electrostatic effects at the root surface.  相似文献
