Proteomic tools allow large-scale, high-throughput analyses for the detection, identification, and functional investigation of proteome. For detection of antigens from Haemaphysalis longicornis, 1-dimensional electrophoresis (1-DE) quantitative immunoblotting technique combined with 2-dimensional electrophoresis (2-DE) immunoblotting was used for whole body proteins from unfed and partially fed female ticks. Reactivity bands and 2-DE immunoblotting were performed following 2-DE electrophoresis to identify protein spots. The proteome of the partially fed female had a larger number of lower molecular weight proteins than that of the unfed female tick. The total number of detected spots was 818 for unfed and 670 for partially fed female ticks. The 2-DE immunoblotting identified 10 antigenic spots from unfed females and 8 antigenic spots from partially fed females. Matrix Assisted Laser Desorption Ionization-Time of Flight Mass Spectrometry (MALDI-TOF) of relevant spots identified calreticulin, putative secreted WC salivary protein, and a conserved hypothetical protein from the National Center for Biotechnology Information and Swiss Prot protein sequence databases. These findings indicate that most of the whole body components of these ticks are non-immunogenic. The data reported here will provide guidance in the identification of antigenic proteins to prevent infestation and diseases transmitted by H. longicornis. 相似文献

The questing height of the tick Haemaphysalis longicornis Neumann was studied on the plant species Solidago altissima L., Pteridium aquilinum Kuhn, Miscanthus sinensis Anderss, and Pleioblastus chino Makino in an abandoned rice field and in an ume (Japanese Apricot) orchard in a forest. On all plant species, the vertical distribution of tick adults and nymphs was not significantly correlated with the distribution of plant surface area. This result suggests that the questing height of this tick is related to the body height of its main animal host, the sika deer. 相似文献

The Haemaphysalis longicornis longicin P4 peptide is an active part peptide produced by longicin which displays bactericidal activity against both Gram-negative and Gram-positive bacteria and other microorganisms. In the present study, the effect of the longicin P4 peptide on the infectivity of Toxoplasma gondii parasites was examined in vitro. Tachyzoites of T. gondii incubated with longicin P4 had induced aggregation and lost the trypan blue dye exclusion activity and the invasion ability into the mouse embryonal cell line (NIH/3T3). Longicin P4 bound to T. gondii tachyzoites, as demonstrated by fluoresce microscopic analysis. An electron microscopic analysis and a fluorescence propidium iodide exclusion assay of tachyzoites exposed to longicin P4 revealed pore formation in the cellular membrane, membrane disorganization, and hollowing as well as cytoplasmic vacuolization. The number of tachyzoites proliferated in mouse macrophage cell line (J774A.1) was significantly decreased by incubation with longicin P4. These findings suggested that longicin P4 conceivably impaired parasite membranes, leading to the destruction of Toxoplasma parasites in J774A.1 cells. Thus, longicin P4 is an interesting candidate for antitoxoplasmosis drug design that causes severe toxicity to T. gondii and plays an important role in reducing cellular infection. This is the first report showing that longicin P4 causes aggregation and membrane injury of parasites, leading to Toxoplasma tachyzoite destruction. 相似文献

The results of experiments on regeneration of the Haller's sensory organ in the metastriate ixodid tick Haemaphysalis longicornisNeumann in the presence of the juvenoid methoprene and retinoic acid and of studies of its structural changes by SEM confirmed the similarity of prostriate and metastriate ixodids as concerns the juvenalizing effect of the above morphogens on regenerative processes during nymphal-imaginal metamorphosis. Vaccination is considered a promising alternative for controlling tick infestations. Haemaphysalis longicornis midgut proteins separated by SDS-PAGE and transferred to polyvinylidene difluoride (PVDF) membrane were screened for protective value against bites. The western blot demonstrated the immunogenicity of 92 kDa protein (P92). The analysis of the P92 amino acid sequence by LC-MS/MS indicated that it was a H. longicornis paramyosin (Hl-Pmy). The full lenghth cDNA of Hl-Pmy was obtained by rapid amplification of cDNA ends (RACE) which consisted of 2,783 bp with a 161 bp 3'' untranslated region. Sequence alignment of tick paramyosin (Pmy) showed that Hl-Pmy shared a high level of conservation among ticks. Comparison with the protective epitope sequence of other invertebrate Pmy, it was calculated that the protective epitope of Hl-Pmy was a peptide (LEEAEGSSETVVEMNKKRDTE) named LEE, which was close to the N-terminal of Hl-Pmy protein. The secondary structure analysis suggested that LEE had non-helical segments within an α-helical structure. These results provide the basis for developing a vaccine against biting H. longicornis ticks. 相似文献

The acaricidal activity and residual effects of 63 commercialized pesticides against the Cheongju strain of Haemaphysalis longicornis in Korea were examined. Twenty‐two pesticides (4 carbamates, 5 organophosphates, 10 pyrethroids, 1 amitraz, 1 diamide, and 1 unclassified pesticide) caused greater than 80% mortality in H. longicornis adults under laboratory conditions using the spray method. These 22 pesticides were used to treat grassland under field conditions for investigation of the residual effects, and 100% mortality in H. longicornis adults was observed with all the carbamates (carbosulfan, benfuracarb, fenobucarb, and carbaryl) and γ‐cyhalothrin after 3 days of pesticide treatment. γ‐Cyhalothrin exhibited 56.7% mortality after 10 days of treatment, which was the longest residual effect of treated grass on H. longicornis adults. With regard to the residual effects on H. longicornis under field conditions, most of the 22 pesticides exhibited higher mortality in nymphs than in adults. In particular, benfuracarb exhibited 96.7% acaricidal activity until 10 days after treatment. These results indicate that carbamates are highly likely to be available under field conditions and, based on this preliminary data, could be used for the control of H. longicornis adults and nymphs. 相似文献

was detected from March to September with highest peak between late April and early May; adults were detected from April to
September with highest peak between late June and July, and an overwintering male population was found during late September
to March; infestation of larvae was observed from June to October and peaked between middle August and early September. Most
of this tick (91%) attached to head and ears of hosts. Additionally, we captured rodents from April to September 2008, but
only a negligible number of nymphs were detected. This result suggested that rodents are not the principal hosts for this

Life Cycle of the Tick Haemaphysalis Leporis-palustris (Acari: Ixodidae) Under Laboratory Conditions
The life cycles of two separate populations (colonies A and B) of the rabbit tick, Haemaphysalis leporis-palustris, were studied under laboratory conditions. Domestic New Zealand rabbits, Oryctolagus cuniculus, and wild rabbits, Sylvilagus brasiliensis, were used as hosts for ticks from colony B and only O. cuniculus rabbits were used as hosts for ticks from colony A. Developmental periods were observed in an incubator at 27±1°C and RH 90±5%. Larvae from colonies A and B fed for 8.0±3.7 days and 8.5±1.3 days, respectively, on O. cuniculus. On S. brasiliensis larvae from colony B fed for 7.2±1.3 days. Nymphs from colony A fed for 8.1±1.4 days on O. cuniculus and nymphs from colony B fed for 8.1±1.0 days on S. brasiliensis. Only one engorged nymph from colony B was recovered from O. cuniculus. Females from colony A fed for 20.9±5.9 days on O. cuniculus and females from colony B fed for 18.6±2.4 days on O. cuniculus and 18.7±3.7 days on S. brasiliensis. Engorged larvae from colony A required 13.7±3.7 days to molt while engorged larvae from colony B required 11.8±3.0 and 11.5±1.8 days to molt, after having fed on O. cuniculus and S. brasiliensis, respectively. Engorged nymphs from colonies A and B required 16.3±1.9 days and 14.7±1.4 days to molt, respectively. Engorged females from colonies A and B required 4–7 and 3–5 days, respectively, to start oviposition. Mean egg incubation periods lasted for 33–34 days. For ticks from colony B, host species accounted for significant differences (p<0.05) in larval and nymphal feeding periods, oviposition weights and CEIs. Significant differences (p<0.05) between the two colonies when ticks fed on O. cuniculus were observed for larval and nymphal feeding and premolt periods, engorged female and oviposition weights and conversion efficiency indexes (CEI). S. brasiliensis were always a more suitable host for H. leporis-palustris than O. cuniculus. Significantly more larvae and nymphs engorged and molted when fed on S. brasiliensis (p<0.001). Females fed S. brasiliensis were more successful to lay fertile eggs and showed the highest engorged and egg mass weights, and the highest CEIs. Data of H. leporis-palustris fed on wild rabbits (one of its natural host species) are reported for the first time. 相似文献

为阐明蜱类盾窝及其发育特点,用扫描电镜观察了长角血蜱Haemaphysalis longicornis不同发育期盾窝的结构,并分析了血餐对盾窝发育的影响.结果表明:幼蜱仅具1对盾窝原基,且每个盾窝原基有1个盾窝孔;若蜱盾窝有了一定的发育,面积(长径×短径)增大且盾窝孔数增多(2~6个);成蜱盾窝面积最大,且盾窝孔数达21~35个.盾窝的发育主要在幼蜱蜕皮阶段及若蜱的吸血和蜕皮阶段,雌蜱盾窝孔径显著大于雄蜱(P<0.01),成蜱、若蜱和幼蜱的盾窝孔孔径在吸血过程中(交配雌蜱除外)各虫期均无显著变化 (P>0.05).综合分析成蜱与未成熟蜱盾窝孔径,发现它们之间无显著差异 (P>0.05),这在一定程度上说明蜱类的盾窝孔径在未成熟期可能已经有了雌雄分化. 相似文献

用嗜菌异小杆线虫Heterorhabditis bacteriophora E-6-7(Hb E-6-7)感染长角血蜱Haemaphysalis longicornis雌蜱,测定感染后雌蜱血淋巴总蛋白含量、酯酶活性及酯酶同工酶的变化,以探讨昆虫病原线虫对蜱的致病机理。结果显示,雌蜱血淋巴总蛋白含量在Hb E-6-7感染后12 h显著增加,达到最大值92.21 μg/μL;在感染后24 h明显下降(49.06 μg/μL);至感染后48 h降至34.25 μg/μL。雌蜱血淋巴酯酶活性在线虫感染后0~12 h内无显著变化;在感染24 h后迅速增加(OD24h=0.1840,OD36h=0.1940,OD48h=0.2165),与对照组差异显著。PAGE结果表明,线虫感染后导致雌蜱血淋巴酯酶同工酶电泳图谱发生变化,主要为电泳图谱两端a带和b带的消失以及一条c带的增加。上述结果表明,长角血蜱雌蜱被Hb E-6-7感染后其血淋巴总蛋白含量和酯酶发生变化,这种变化可能与蜱的防御和对昆虫病原线虫的适应有关。 相似文献

The tick embryo basic protein (TEBP) is present in the nucleus as a counterpart of histones at early embryonic stages of the tick Hyalomma dromedarii. The sharp drop in the TEBP nuclear level and elimination of the N-terminal dipeptide (leucine–serine) between days 12 and 15 after oviposition suggested the transport of TEBP to the cytoplasm for protein turnover. The traffic of TEBP during tick embryogenesis was examined. The level of TEBP was detected in the cytoplasm from the different embryonic stages by the established enzyme-linked immunosorbent assay (ELISA) and confirmed by immunoblotting. At day 12, a 2-fold increase in the cytoplasmic TEBP level coincided with its decrease in the nucleus. This result indicates that TEBP starts to leave the nucleus for the cytoplasm at day 12. The changes in the cytoplasmic leucine aminopeptidase (LAP)-specific activity were followed during tick embryogenesis. The LAP activity started to increase at day 12 and reached its maximum level at day 21. The enzyme displayed an optimum pH between 7.5 and 8.8 and a Km value of 0.5 M for leucine-p-nitroanilide. The involvement of the exopeptidase activity in the TEBP turnover after its translocation to the cytoplasm is discussed. 相似文献

A large-scale field test using pheromone-acaricide-impregnated plastic tail-tag decoys demonstrated excellent efficacy of these devices for control of the bont tick,Amblyomma hebraeum, on cattle in Zimbabwe. The tail tags were impregnated with a mixture containingo-nitrophenol, methyl salicylate, 2,6-dichlorophenol and phenylacetaldehyde and one of three different acaricides (cyfluthrin, flumethrin or alphacypermethrin).o-Nitrophenol and methyl salicylate are components of theA. hebraeum attraction-aggregation-attachment pheromone, while 2,6-dichlorophenol and phenylacetaldehyde are proven attractants for this tick. Botho-nitrophenol and methyl salicylate were lost gradually from the tags over 12 and 14 week periods, respectively. In field trials, tick counts were compared between cattle that received tail tags either impregnated with pheromone mixture alone, cyfluthrin and pheromone mixture, flumethrin and pheromone mixture, alphacypermethrin and pheromone mixture or were left untreated. During the first 3 month trial period, control of adult bont ticks was 94.9% with cyfluthrin tail tags and 87.5% with flumethrin tail tags. In general, there was no significant difference in bont tick numbers on cattle without tags and those with tail tags containing pheromone only. When the trial was repeated for another 3 month period, control of bont ticks with tail tags containing cyfluthrin and flumethrin was 99.3 and 95.1%, respectively. However, control of bont ticks using alphacypermethrin was only 79.2%. Overall, retention of tail tags was excellent although some loss was encountered during the rainy season. In addition to controlling bont ticks, the tail tags provided moderate control of other tick species (Rhipicephalus evertsi evertsi, Rhipicephalus zambeziensis andHyalomma spp.) simultaneously infesting cattle in the trials.Deceased. 相似文献

Ticks are blood-feeding arthropods known for their long survivability off the host. Although ticks are terrestrial, they can survive extended periods of time submerged underwater. A plastron is an alternative respiration system that can absorb oxygen from water via a thin layer of air trapped by hydrophobic hairs or other cuticular projections. The complex spiracular plate of ticks has been postulated to serve as a plastron but that function has not been verified. This study provides evidence of plastron respiration in the American dog tick, Dermacentor variabilis, and for the first time confirmed the existence of plastron respiration in Ixodidae. Longer survival rates in oxygenated water indicate that underwater respiration requires oxygen. Wetting the spiracular plate with alcohol debilitates any potential plastron function and lowers the survival rate. Survival underwater may also be enhanced by metabolic depression and possibly anaerobic respiration. This study describes the first example of plastron respiration in the Ixodidae. 相似文献

Owing to the need to combat the spread of chemical acaricide resistance in ticks, we evaluated the efficacy of a mixture of the entomopathogenic fungus Beauveria bassiana AT17 and acaricides for the control of Hyalomma anatolicum anatolicum in China. A mixture of B. bassiana AT17 at the concentration of 108 conidia/mL and the synthetic pyrethroid deltamethrin at concentrations of 2500, 250, 25, 5, 2.5, 0.5 and 0.25 ppm was tested in vitro. The germination capability, vegetative growth, conidia production, and viability of B. bassiana AT17 were assessed and the efficacy of the mixture in killing engorged H. anatolicum anatolicum females was measured. High mortality rates were achieved when the entomopathogen was combined with different concentrations of deltamethrin. Neither B. bassiana AT17 nor deltamethrin alone at the same concentrations could cause the higher mortality rates seen with the combination. In addition the combination killed the ticks more rapidly than did either agent alone (3–5 days more rapidly). Our results indicate that the mixture of B. bassiana AT17 and deltamethrin has potential as a new type of reagent for integrated control of H. anatolicum anatolicum. 相似文献

The cutaneous hypersensitivity induced by Rhipicephalus sanguineus tick extract in dogs (natural host) and guinea-pigs (laboratory host) was evaluated. The left ear of infested and control (tick-bite naive) dogs and guinea-pigs was injected intradermally with an extract from unfed adult ticks and the right ear with phosphate buffered saline (PBS). Ear thickness variations were then measured after 10 min and 1, 2, 6, 18, 24, 48, 72 and 96 h post-injection. Results were expressed as percentual changes in the ear thickness in relation to pre-inoculation values. The final variation in ear thickness induced by the extract was given by subtracting, in each animal, the right ear percentual increase from that of the left ear. Guinea-pigs were tested at two different times following infestation and with two different doses of extract. Infested guinea-pigs from the three experiments developed an immediate (within the first 2 h post-inoculation) and a strong delayed reaction (24 h) to the extract. Dogs, unlike guinea-pigs, developed only a strong immediate reaction whereby an 80% increase in ear thickness was observed. Control animals, with the exception of one dog, did not develop any significant reaction to the extract. Only mild reactions were induced by PBS in the right ear of all animals. The correlation between the absence of a strong delayed type reaction to tick extract and the lack of resistance of the natural host to R. sanguineus tick is discussed. 相似文献

The Archaeoglobus fulgidis gene RS27_ARCFU encodes the 30S ribosomal protein S27e. Here, we present the high-quality NMR solution structure of this archaeal protein, which comprises a C4 zinc finger motif of the CX(2)CX(14-16)CX(2)C class. S27e was selected as a target of the Northeast Structural Genomics Consortium (target ID: GR2), and its three-dimensional structure is the first representative of a family of more than 116 homologous proteins occurring in eukaryotic and archaeal cells. As a salient feature of its molecular architecture, S27e exhibits a beta-sandwich consisting of two three-stranded sheets with topology B(decreasing), A(increasing), F(decreasing), and C(increasing), D(decreasing), E(increasing). Due to the uniqueness of the arrangement of the strands, the resulting fold was found to be novel. Residues that are highly conserved among the S27 proteins allowed identification of a structural motif of putative functional importance; a conserved hydrophobic patch may well play a pivotal role for functioning of S27 proteins, be it in archaeal or eukaryotic cells. The structure of human S27, which possesses a 26-residue amino-terminal extension when compared with the archaeal S27e, was modeled on the basis of two structural templates, S27e for the carboxy-terminal core and the amino-terminal segment of the archaeal ribosomal protein L37Ae for the extension. Remarkably, the electrostatic surface properties of archaeal and human proteins are predicted to be entirely different, pointing at either functional variations among archaeal and eukaryotic S27 proteins, or, assuming that the function remained invariant, to a concerted evolutionary change of the surface potential of proteins interacting with S27. 相似文献

The purpose of this study was to determine whether dogs develop acquired resistance to adult Ixodes scapularis infestation in an experimental model. Five dogs were each infested with ten mating pairs of ticks every week for 7 consecutive weeks, another five dogs were each infested with ten mating pairs once every 2 weeks for 10 weeks and four dogs served as controls not exposed to ticks. All ticks were allowed to feed to repletion and were collected only after dropping from the host. Several variables were measured to determine the extent of blood feeding success. Regression analysis indicated that the engorgement success, survival and mean tick engorgement weight declined with repeated infestation in both groups of dogs (p<0.05). Tick oviposition as well as the F1 viability declined with each successive infestation in both groups. These results suggest that repeated infestation with I. scapularis elicits a protective immune response against tick feeding and could serve as a limiting factor in the spread and transmission of Borrelia burgdorferi. 相似文献

The newly described tick Amblyomma vikirri has a narrow host range, being found mainly on the Australian lizard, Egernia stokesii and rarely on another lizard, Tiliqua rugosa. Both hosts are in the family Scincidae. Larvae of A. vikirri were as successful in locating E. stokesii as T. rugosa from a range of release distances between 20 and 120 mm from the host. Over this range the proportion of ticks which successfully located hosts declined and the time taken by successful ticks to locate hosts increased with increasing release distance. From 60 mm, larvae of A. vikirri located four other lizard species from the families Scincidae and Agamidae and two non-living targets as successfully as they did E. stokesii. The only evidence that there was any host specificity in the searching behaviour of larvae of A. vikirri was that A. vikirri larvae spent less time paused and questing when they were searching for E. stokesii than when they were searching for T. rugosa. Aponomma hydrosauri, a tick which commonly infests T. rugosa but not E. stokesii, spent less time paused and questing when it was searching for T. rugosa than when it was searching for E. stokesii. However, the results overall suggest that the narrow host range of A. vikirri cannot be explained by any ability of the larvae of that species to discriminate between their natural host and other reptile species. 相似文献
