Proceeding from three previously derived expressions for the intensity of nitrification in soil as a function of time (logΣN=K.logt+q), as a function of incubation moisture (logΣN=A.pF i+B), as a function of initial moisture (logΣN=C.pF v+D), it was shown that the nitrification intensity as a function of time and of moisture can be expressed by the bilinear function log ΣN=a.pF i.logT+b.pF i+c.logt+d; as a function of time and of initial moisture by the bilinear function logΣ=N=a.pF v.logt+b.pF v+c.logt+d; as a function of initial and incubation moisture by the bilinear function log ΣN=a.pF ipF v+b.pF i+c.pF v+d. The intensity of nitrification as a function of time, incubation moisture and initial moisture may be expressed by the multilinear function log ΣN=a.pF i.pF v.logt+b.pF i.pF v+c.pF i.logt+d.pF v.logt+e .pF i+f.pF v=g.logt+h. This function is valid for all the incubation moistures lying between pF i 3.0 and 4.0 and for all initial moistures between 3.5 and 5.9 provided that the incubation temperature remains constant.  相似文献

The objective of this study was to examine whether S-RNase plays a specific role in the pre-germinated Pyrus pollen. Effects of exogenous RNase and endogenous S-RNase on concentration of cytosolic-free calcium ([Ca2+]i) variation of pre-germinated Pyrus pollen were studied. [Ca2+]i variation caused by different RNases were complex. In 1 h after being cultured, exogenous RNase, RNase T1 and RNase A, and endogenous incompatible ‘Hohsui’ RNase promoted the [Ca2+]i of ‘Hohsui’ pollen. Acid proteins of ‘Hohsui’ had no remarkable influence on the [Ca2+]i of self-pollen. Endogenous compatible ‘Kohsui’ RNase reduced the [Ca2+]i of ‘Hohsui’ pollen, but compatible ‘Hohsui’ RNase can stimulate the [Ca2+]i of ‘Kohsui’ pollen. RNase T1, RNase A and incompatible ‘Kohsui’ S-RNase can also make ‘Kohsui’ pollen [Ca2+]i increase. Different from ‘Hohsui’ pollen, acid proteins of ‘Hohsui’ pull down the ‘Kohsui’ pollen [Ca2+]i remarkably. Conclusion can be made that during the prophase of pollen germination, endogenous S-RNase has no specific effect on pollen [Ca2+]i changes.  相似文献

The long-term, 26 years’ data set of observations on daily concentrations of airborne grass pollen in Leiden is analyzed to present the variations and trends in quantities, and season starting dates. Monitoring of airborne pollen has been done continuously at one location, with a volumetric pollen trap. Annual totals of daily average grass-pollen concentrations are within a normal range of an urban site between 3690 and 9277, averagely 5510. The annual totals are irregularly fluctuating from year to year, and show no increasing or decreasing trend. Each year’s seasonal fluctuation is different, probably under the influence of changing weather conditions. The typical grass-pollen month is June. Using the Σ 75 criterium, the average starting date is on 16 May, whereas with the 1% threshold criterium the start of the grass-pollen season averagely is on 3 June. The mean air temperature in the preceding period is taken as the main factor for the timing of the season start. Analyzing the relationships of the two different criteria for the season starts with a number of temperature observation periods, the best relations were found between the mean air temperature in the period 11 April to 20 May and the Σ 75 criterium season start on 16 May (r=−0.78); and between the mean air temperature in May and the 1% threshold criterium season start on 3 June (r=−0.76). Forecasts of the season start which are significantly better than the average starting date are only possible with the mean air temperature up to a few days before the actual start. This limits the practical usefulness of the forecasting system.  相似文献

We describe the characteristics of a sampling procedure called random median sampling that was proposed to enhance the precision of population estimates. In performing random median sampling, we first select a sampling item at random from the sampling area. We roughly compare the abundance of individuals in the selected item with that of the adjacent two items in order to identify the item that has median abundance, i.e., the item that has the second largest abundance among the three items. We count the number of individuals of the item having the median abundance. This procedure is repeated n times in the sampling area (i = 1, 2, ..., n). Let m i be the ith median abundance. The estimates of the mean abundance per sampling item and the variance of estimates are given by Σm i /n and Σ(m i –Σm i /n)2/n(n – 1), respectively. This method is a local application of the median ranked set sampling that was proposed by Muttlak (J Appl Stat Sci 6:245–255, 1997). Random median sampling is effective when the correlation coefficient between adjacent items is small. If the correlation coefficient is close to zero, random median sampling reduces the variance of estimates to 45 or 32% of that in simple random sampling when the distribution follows a normal distribution or a Laplace distribution, respectively. The sample size required to achieve a given precision of estimate decreases accordingly. The effectiveness of random median sampling, however, is small if the correlation coefficient is large. The condition in which random median sampling is superior to simple random sampling is also discussed.  相似文献

Accurate assessments of pollen counts are valuable to allergy sufferers, the medical industry, and health researchers; however, monitoring stations do not exist in most areas. In addition, the degree of spatial reliability provided by the limited number of monitoring stations is poorly understood. We developed and compared spatial models to estimate pollen concentrations in locations without monitoring stations. Daily Acer, Quercus, and overall tree, grass, and weed pollen counts, in grains/m3, were obtained from 14 aeroallergen monitoring stations located in the northeastern and mid-Atlantic region of the United States from 2003 to 2006. Pollen counts were spatially interpolated using ordinary kriging. Mixed effects and generalized estimating equations incorporating daily and seasonal weather characteristics, pollen season characteristics and land-cover information were also developed to estimate daily pollen concentrations. We then compared observed values from a monitoring station to model estimates for that location. Observed counts and kriging estimates for tree pollen differed (p = 0.04), but not when peak periods were removed (p = 0.29). No differences between observed and kriging estimates of Acer (p = 0.46), Quercus (p = 0.24), grass (p = 0.31) or weed pollen (p = 0.29) were found. Estimates from longitudinal models also demonstrated good agreement with observed counts, except for the extremes of pollen distributions. Our results demonstrate that spatial interpolation techniques as well as regression methods incorporating both weather and land-cover characteristics can provide reliable estimates of daily pollen concentrations in areas where monitors do not exist for all but periods of extremely high pollen.  相似文献

In order to study allergic people responding to daily changes in pollen concentrations, we compared personal diary data on allergic symptoms and the use of allergy medicines to daily pollen counts during the two unequal alder and birch pollen seasons of 2009 and 2010. Almost 90% of the 61 subjects with physician-diagnosed birch pollinosis developed conjunctival, nasal or other symptoms during the peak birch pollination. Most subjects (95%) also reported symptoms during the alder pollination. Despite a delay between the most severe symptoms and the pollen peaks and the increased risk of allergy symptoms between the alder and birch pollen peaks at much lower pollen concentrations, the number of subjects with allergy symptoms correlated with the daily pollen concentrations in both years (r 09 = 0.35, r 10 = 0.36, p < 0.01). The positive correlation was even stronger (r 09 = 0.69, r 10 = 0.74, p < 0.001) in relation to the cumulative sum of daily concentrations. The use of allergy medicines precisely followed the abundance of allergy symptoms in both years (r 09 = 0.96, r 10 = 0.70, p < 0.001). We conclude that there is a fair correlation between the daily allergy symptoms and the particular pollen concentrations, but the risk of developing symptoms at low, moderate and high concentrations is affected by the progression of the pollen season.  相似文献

Sequence alignment has been an invaluable tool for finding homologous sequences. The significance of the homology found is often quantified statistically by p-values. Theory for computing p-values exists for gapless alignments [Karlin, S., Altschul, S.F., 1990. Methods for assessing the statistical significance of molecular sequence features by using general scoring schemes. Proc. Natl. Acad. Sci. USA 87, 2264–2268; Karlin, S., Dembo A., 1992. Limit distributions of maximal segmental score among Markov-dependent partial sums. Adv. Appl. Probab. 24, 13–140], but a full generalization to alignments with gaps is not yet complete. We present a unified statistical analysis of two common sequence comparison algorithms: maximum-score (Smith-Waterman) alignments and their generalized probabilistic counterparts, including maximum-likelihood alignments and hidden Markov models. The most important statistical characteristic of these algorithms is the distribution function of the maximum score S max, resp. the maximum free energy F max, for mutually uncorrelated random sequences. This distribution is known empirically to be of the Gumbel form with an exponential tail P(S max > x) ∼ exp(−λx) for maximum-score alignment and P(F max > x) ∼ exp(−λx) for some classes of probabilistic alignment. We derive an exact expression for λ for particular probabilistic alignments. This result is then used to obtain accurate λ values for generic probabilistic and maximum-score alignments. Although the result demonstrated uses a simple match-mismatch scoring system, it is expected to be a good starting point for more general scoring functions.  相似文献

Pollen forecasts are a fundamental prerequisite to obtain prophylactic measures for allergic individuals. Mugwort belongs to the most relevant allergenic pollen types after grasses and birch. An approach to modeling of mugwort pollen concentrations has not been attempted previously in Germany. A process-oriented mathematical model for the relative local daily average mugwort airborne pollen concentration was developed on the basis of pollen and weather data measured during a 6-year period. The model depends on the daily minimum and maximum temperature, amount of precipitation and atmospheric pressure, which have to and can be supplied by measurement and prediction. The comparison of modeling results and pollen counting for an additional year confirms the fitness of the model. A computer program was written, which rests upon the model and supplies daily predictions of mugwort pollen flight during the period of the weather forecast. The latter should allow a pollen forecasting period of about 5 days, with an accuracy of about 32–63% explained variance, which in view of the low mugwort pollen counts (nine grains/m3 maximum in the validation year) represents a high relative measurement error. The mathematical model may serve to improve and rationalize of present pollen forecasts.  相似文献

Oryza rufipogon Griff. is the ancestor of Asian cultivated rice (O. sativa L.) and possesses valuable genes for rice breeding. Pollen abortion is one of the major causes of indicajaponica hybrid sterility in rice and it happens due to allelic interaction at the pollen sterility gene loci. A total of six loci (Sa, Sb, Sc, Sd, Se, and Sf) have been found to be associated with F1 pollen sterility between indica and japonica rice, and five of them (all except Sf) have been mapped. Neutral alleles (S n ) at each locus have the potential to overcome the pollen sterility associated with the respective locus. Therefore, exploitation and utilization of neutral alleles have significant importance in overcoming indicajaponica hybrid sterility. In this study, an accession (IRW28) of O. rufipogon, native to China, was selected as paternal to cross with typical japonica (Taichung 65) and indica (Guanglu’ai 4) tester lines, and two F2 populations were developed. The simple sequence repeat markers tightly linked to five pollen sterility loci were applied for genotyping the F2 populations. Chi-squared tests were applied to examine the normal segregation/distortion at each locus. The expected and observed pollen sterility for each locus were estimated. As a result, the genotypes at five pollen sterility gene loci for IRW28 were identified as: Sa i−1/Sa i−1, Sb n /Sb n , Sc i−2/Sc i−2, Sd n /Sd n and Se n /Se n . Our results suggest that IRW28 (O. rufipogon) has the neutral alleles for pollen fertility at the Sb, Sd and Se loci, and these alleles have a good affinity with indica and japonica rice. These neutral alleles provide valuable gene resources to overcome the inter-subspecific hybrid pollen sterility in rice.  相似文献

An extension to HN(CO-α/β-N,Cα-J)-TROSY (Permi and Annila in J Biomol NMR 16:221–227, 2000) is proposed that permits the simultaneous determination of the four coupling constants 1 J N′(i)Cα(i), 2 J HN(i)Cα(i), 2 J Cα(i−1)N′(i), and 3 J Cα(i−1)HN(i) in 15N,13C-labeled proteins. Contrasting the original scheme, in which two separate subspectra exhibit the 2 J CαN′ coupling as inphase and antiphase splitting (IPAP), we here record four subspectra that exhibit all combinations of inphase and antiphase splittings possible with respect to both 2 J CαN′ and 1 J N′Cα (DIPAP). Complementary sign patterns in the different spectrum constituents overdetermine the coupling constants which can thus be extracted at higher accuracy than is possible with the original experiment. Fully exploiting data redundance, simultaneous 2D lineshape fitting of the E.COSY multiplet tilts in all four subspectra provides all coupling constants at ultimate precision. Cross-correlation and differential-relaxation effects were taken into account in the evaluation procedure. By applying a four-point Fourier transform, the set of spectra is reversibly interconverted between DIPAP and spin-state representations. Following our established theoretical model to deal with the second-harmonic generation (SHG) excited by a linearly polarized focused beam in type I collagen, in this paper, we further quantitatively characterize the differences between SHG emissions in type I collagen excited by collimated and focused beams. The effects of the linear polarization angle (α) and the fibril polarity characterized by the hyperpolarizability ratio ρ on SHG emission has been compared under collimated and focused beam excitation, respectively. In particular, SHG emission components along the i axis ( I2w,i )\left( {I_{2\omega {,}i} } \right) (i = x,y,z), the induced SHG emission deviation angle γ ij , and the detected SHG signals (I 2ω,ij ) in the ij plane by rotating the applied polarizer angle φ ij have been investigated (i = x, x, y; j = y, z, z). Results show that under our simulation model, SHG emission in the xy plane, such as I 2ω,x ,I 2ω,y ,γ xy and I 2ω,xy varying as polarization angle (α) under collimated and focused light, presents no significant difference. The reverse of the fibril polarity has induced great impact on I 2ω,x ,γ xy and I 2ω,xy in both collimated and focused light. I 2ω,x and γ xy show similarity, but I 2ω,xy at α = 30° demonstrates a slight difference in focused light to that in collimated light. Under focused light, the reverse of fibril polarity causes obvious changes of the collected SHG intensity I 2ω,xz and I 2ω,yz at a special polarization angle α = 60° and γ xz , γ yz along α.  相似文献

Phosphorus magnetic resonance spectroscopy (31P-MRS) was used to investigate the influence of maximal aerobic power (˙VO 2max) on the recovery of human calf muscle from high-intensity exercise. The (˙VOO2max) of 21 males was measured during treadmill exercise and subjects were assigned to either a low-aerobic-power (LAP) group (n = 10) or a high-aerobic-power (HAP) group (n = 11). Mean (SE) ˙VO 2max of the groups were 46.6 (1.1) and 64.4 (1.4) ml · kg−1 · min−1, respectively. A calf ergometry work capacity test was used to assign the same relative exercise intensity to each subject for the MRS protocol. At least 48 h later, subjects performed the rest (4 min), exercise (2 min) and recovery (10 min) protocol in a 1.5 T MRS scanner. The relative concentration of phosphocreatine (PCr) was measured throughout the protocol and intracellular pH (pHi) was determined from the chemical shift between inorganic phospate (Pi) and PCr. End-exercise PCr levels were 27 (3.4) and 25 (3.5)% of resting levels for LAP and HAP respectively. Mean resting pHi was 7.07 for both groups, and following exercise it fell to 6.45 (0.04) for HAP and 6.38 (0.04) for LAP. Analysis of data using non-linear regression models showed no differences in the rate of either PCr or pHi recovery. The results suggest that ˙VO2max is a poor predictor of metabolic recovery rate from high-intensity exercise. Bacillus mycoides strain RIJ B-017, a growth-associated poly-3-hydroxybutyrate (PHB) producer was grown on sucrose-containing media. PHB accumulated in cells up to 72% of dry cell mass. The overall maximum value of PHB yield (Y p/s) and productivities (Q p andq p) 250 mgp/gs, 120 mgp L−1 h−1 and 30 mgp gx −1 h−1, respectively, were obtained at 15 g/L sucrose. Differential scanning calorimeter heating curve showed two peaks, one at 95.9 °C and another at 165.4°C with a shoulder around 154.6 °C. The viscosity-average molar mass in chloroform at 27°C was 505 kDa. The carbon content of PHB was 55.4% of the mass.  相似文献

Background  

Assembling genomic sequences from a set of overlapping reads is one of the most fundamental problems in computational biology. Algorithms addressing the assembly problem fall into two broad categories - based on the data structures which they employ. The first class uses an overlap/string graph and the second type uses a de Bruijn graph. However with the recent advances in short read sequencing technology, de Bruijn graph based algorithms seem to play a vital role in practice. Efficient algorithms for building these massive de Bruijn graphs are very essential in large sequencing projects based on short reads. In an earlier work, an O(n/p) time parallel algorithm has been given for this problem. Here n is the size of the input and p is the number of processors. This algorithm enumerates all possible bi-directed edges which can overlap with a node and ends up generating Θ(nΣ) messages (Σ being the size of the alphabet).  相似文献

In Melbourne, Australia, grass pollen is the predominant cause of hayfever in late spring and summer. The grass pollen season has been monitored in Melbourne, using a Burkard spore trap, for 13 years (1975–1981, 1985 and 1991–1997). Total counts for grass pollen were highly variable from one season to the next (approximately 1000 to >8000 grains/m3). The daily grass pollen counts also showed a high variability (0 to approximately 400 grains/m3). In this study, the grass pollen counts of the 13 years (12 grass pollen seasons, extending from October to January) have been compared with meteorological data in order to identify the conditions that can determine the daily amounts of grass pollen in the air. It was found that the seasonal total of grass pollen was directly correlated with the rainfall sum of the preceding 12 months (1 September–31 August): seasonal total of grass pollen (counts/m3)=18.161 × rainfall sum of the preceding 12 months (mm) −8541.5 (r s=0.74,P<0.005,n=12). The daily amounts of grass pollen in the air were positively correlated with the corresponding daily average ambient temperatures (P<0.001). The daily amount of grass pollen which was to be expected with a certain daily average temperature was linked to the seasonal total of grass pollen: in years with high total grass pollen counts, a lower daily average temperature was required for a high daily pollen count than in years with low total grass pollen counts. As the concentration of airborne grass pollen determines the severity of hayfever in sensitive patients, an estimation of daily grass pollen counts can provide an indication of potential pollinosis symptoms. We compared daily grass pollen counts with the reported symptomatic responses of hayfever sufferers in November 1985 and found that hayfever symptoms were significantly correlated to the grass pollen counts (P<0.001 for nasal,P<0.005 for eye symptoms). Thus, a combination of meteorological information (i.e. rainfall and temperature) allows for an estimation of the potential daily pollinosis symptoms during the grass pollen season. Here we propose a symptom estimation chart, allowing a quick prediction of eye and nasal symptoms that are likely to occur as a result of variations in meteorological conditions, thus enabling both physicians and patients to take appropriate avoidance measures or therapy.  相似文献

We propose a new alpha proton detection based approach for the sequential assignment of natively unfolded proteins. The proposed protocol superimposes on following features: HA-detection (1) enables assignment of natively unfolded proteins at any pH, i.e., it is not sensitive to rapid chemical exchange undergoing in natively unfolded proteins even at moderately high pH. (2) It allows straightforward assignment of proline-rich polypeptides without additional proline-customized experiments. (3) It offers more streamlined and less ambiguous assignment based on solely intraresidual 15N(i)-13C′(i)-Hα(i) (or 15N(i)-13Cα(i)-Hα(i)) and sequential 15N(i + 1)-13C′(i)-Hα(i) (or 15N(i + 1)-13Cα(i)-Hα(i)) correlation experiments together with efficient use of chemical shifts of 15N and 13C′ nuclei, which show smaller dependence on residue type. We have tested the proposed protocol on two proteins, small globular 56-residue GB1, and highly disordered, proline-rich 47-residue fifth repeat of EspFU. Using the proposed approach, we were able to assign 90% of 1Hα, 13Cα, 13C′, 15N chemical shifts in EspFU. We reckon that the HA-detection based strategy will be very useful in the assignment of natively unfolded proline-rich proteins or polypeptide chains.  相似文献

Summary.  Patch-clamp whole-cell and single-channel recording techniques were used to investigate the regulation of outward K+ channels by external and internal protons in Brassica chinensis pollen protoplasts. Outward K+ currents and conductance were insensitive to external pH (pHo) except at pH 4.5. Maximal conductance (G max) for the outward K+ currents was inhibited at acidic external pH. Half-activation voltage (E 1/2) for the outward K+ currents shifted to more positive voltages along with the decrease in pHo. E 1/2 can be described by a modified Henderson–Hasselbalch equation expected from a single titratable binding site. The activation kinetics of the outward K+ channels was largely insensitive to pHo. An internal pH (pHi) of 4.5 significantly increased outward K+ currents and conductance. G max for the outward K+ currents decreased with elevations in pHi. In contrast to the effect of pHo, E 1/2 was shifted to more positive voltages with elevations in pHi. The outward K+ currents, G max and E 1/2 can be described by the modified Henderson–Hasselbalch equation. Furthermore, acidifying pHi accelerated the activation of the outward K+ currents significantly. The differences in electro-physiological properties among previously reported and currently described plant outward K+ channels may reflect differences in the structure of these channels. The effect of high irradiance (HI, photosynthetically active photon flux density of 1 300 μmol m−2 s−1) on net photosynthetic rate (P N), chlorophyll fluorescence parameters, and xanthophyll cycle components were studied in fruit tree bayberry leaves. HI induced the photoinhibition and inactivation of photosystem 2 (PS2) reaction centres (RCs), which was characterized by decreased P N, maximum yield of fluorescence after dark adaptation (Fm), photochemical efficiency of PS2 (Fv/Fm) and quantum yield of PS2 (ΦPS2), and increased reduction state of QA (1-qP) and non-photochemical quenching (NPQ). Initial fluorescence (F0) showed a decrease after the first 2 h, and subsequently increased from the third hour exposure to HI. Furthermore, a greater increase in the ratio (Fi-F0)/(Fp-F0) which is an expression of the proportion of the QB non-reducing PS2 centres, whereas a remarked decrease in the slope of Fi to Fp which represents the rate of QA reduction was observed in leaves after HI exposure. Additionally, HI caused an increase in the pool size of the xanthophyll cycle pigments and sustained elevated contents of zeaxanthin (Z), antheraxanthin (A), and de-epoxidation state (DES) at the end of the irradiation period. During HI, decreased Fm, Fv/Fm, ΦPS2, NPQ, slope of Fi to Fp, V+A+Z, and DES, and increased F0, 1-qP, ratio (Fi-F0)/(Fp-F0), and V were observed in dithiothreitol (DTT)-fed leaves compared to control ones under the same conditions. Hence photoinhibition caused by HI in bayberry was probably attributed to inactivation of PS2 RCs, and photoprotection from photodamage were mainly related to the xanthophyll cycle-dependent heat dissipation in excess photons.  相似文献

In many cases the distribution of saccadic reaction times (SRT) deviates considerably from a unimodal distribution and may often exhibit several peaks. We present a statistical approach to determining the number and form of the individual peaks. The overall density of the reaction times f i (t), i=1…M obtained in M different experiments with the same subject is described as the sum of K basis functions x k (t),k=1…K with different weights and an error term. A change in the experimental conditions is assumed to cause a change in the weights, not in the basis functions. We minimize the square of the difference (measured data minus approximation), divided by the error of the data. Incrementing K step by step we determine the necessary number of basis functions. This method is applied to data of six subjects tested in different saccade tasks. We detect five different modes: two in the range 80–140 ms (express modes), two in the range 145–190 ms (fast-regular mode) and one at about 230 ms (slow-regular mode). These modes are located at about the same positions for different subjects. The method presented here not only proves statistically the existence of several modes in SRT distributions but also allows the distributions to be described by a few characteristic numbers that go beyond the mean values and standard deviations. We have characterized the promoter specificity of theArabidopsis thaliana α1-tubulin (α 1-tub) gene by studying expression patterns of gene fusions between the 2.2 kbp 5′ upstream region of theα 1-tub gene and each of three different reporters: chloramphenical acetyltransferase, β-glucuronidase or the diphtheria toxin chain A gene. Analysis of transgenic tobacco andArabidopsis plants carrying the transgene showed that the chloramphenicol acetyltransferase and β-glucuronidase activities were not detected in any vegetative or reproductive organs except mature pollen. Transgenic tobacco plants carrying the diphtheria toxin chain A gene under the control of theα 1-tub promoter were of normal phenotype but seed fertility was drastically reduced. Furthermore, the transgene could not be transmitted to the next generation through pollen, supporting the observation that theα 1-tub promoter is active only in pollen. It was observed that the promoter activity was most active in mature pollen and decreased significantly duringin vitro pollen germination, indicating that the promoter is inactive or subdued in germinating pollen. The promoter activity was not affected by various plant growth hormones during pollen maturation.  相似文献
