The products of C14O2 fixation by Chlamydomonas and Chlorella were studied under conditions most favorable for glycolate synthesis. The highest percentage of the C14 was incorporated into glycolate in the pH range of 8 to 9. After 1 to 2 minutes as much as 40% of the C14 was found in glycolate products and only a trace of C14 was present as phosphoglycerate. Below pH 8 the rate of photosynthesis was much faster, but only a small percent of the C14 was incorporated into glycolate in 1 or 2 minutes, while a high percent of the C14 accumulated in phosphoglycerate. C14 labeling of glycolate even at pH 8 or above did not occur at times shorter than 10 seconds. During the first seconds of photosynthesis, nearly all of the C14 was found in phosphoglycerate and sugar phosphates. Thus glycolate appears to be formed after the phosphate esters of the photosynthetic carbon cycle.

Washing Chlamydomonas with water 2 or 3 times resulted in the loss of most of their free phosphate. When a small aliquot of NaHC14O3 was added to washed algae in the absence of this buffering capacity, the pH of the algal medium became 8 or above and much of the fixed C14 accumulated in glycolate.

Tobacco (Nicotiana tabacum var Havana Seed) leaf discs were supplied tracer quantities of [2-14C]- and [3-14C]pyruvate for 60 minutes in steady state photosynthesis with 21% or 1% O2, and the glycolate oxidase inhibitor α-hydroxy-2-pyridinemethanesulfonic acid was then added for 5 or 10 minutes to cause glycolate to accumulate. The [3-14C]pyruvate was converted directly to glycolate as shown by a 50% greater than equallabeled 14C in C-2 of glycolate, and the fraction of 14C in C-2 increased in 1% O2 to 80% greater than equal-labeled. This suggests the pathway using pyruvate is less O2-dependent than the oxygenase reaction producing glycolate from the Calvin cycle. The formation of glycolate from pyruvate in the leaf discs was time-dependent and with [2-14C]- and [3-14C]pyruvate supplied leaf discs the C-2 of glyoxylate derived from C-2 of isocitrate was labeled asymmetrically in a manner similar to the asymmetrical labeling of C-2 of glycolate under a number of conditions. Thus glycolate was probably formed by the reduction of glyoxylate. Isocitric lyase activity of tobacco leaves was associated with leaf mitochondria, though most of the activity was in the supernatant fraction after differential centrifugation of leaf homogenates. The total enzyme activity was at least 35 micromoles per gram fresh weight per hour. The relative contribution of the pathway to the glycolate pool is unknown, but the results support the existence of a sequence of reactions leading to glycolate synthesis during photosynthesis with pyruvate, isocitrate, and glyoxylate as intermediates.  相似文献

Isolated soybean (Glycine max [L.] Merr.) leaf cells were treated with three inhibitors of the glycolate pathway in order to evaluate the potential of such inhibitors for increasing photosynthetic efficiency. Preincubation of cells under acid conditions in α-hydroxypyridinemethanesulfonic acid increased 14CO2 incorporation into glycolate, but severely inhibited photosynthesis. Isonicotinic acid hydrazide (INH) increased the incorporation of 14CO2 into glycine and reduced label in serine, glycerate, and starch. Butyl 2-hydroxy-3-butynoate (BHB) completely and irreversibly inhibited glycolate oxidase and increased the accumulation of 14C into glycolate. Concomitant with glycolate accumulation was the reduction of label in serine, glycerate, and starch, and the elimination of label in glycine. The inhibitors INH and BHB did not eliminate serine synthesis, suggesting that some serine is synthesized by an alternate pathway. The per cent incorporation of 14CO2 into glycolate by BHB-treated cells or glycine by INH-treated cells was determined by the O2/CO2 ratio present during assay. Photosynthesis rate was not affected by INH or BHB in the absence of O2, but these compounds increased the O2 inhibition of photosynthesis. This finding suggests that the function of the photorespiratory pathway is to recycle glycolate carbon back into the Calvin cycle, so if glycolate metabolism is inhibited, Calvin cycle intermediates become depleted and photosynthesis is decreased. Thus, chemicals which inhibit glycolate metabolism do not reduce photorespiration and increase photosynthetic efficiency, but rather exacerbate the problem of photorespiration.  相似文献

The influence of pH upon the O2 inhibition of 14CO2 photoassimilation (Warburg effect) was examined in intact spinach (Spinacia oleracea) chloroplasts. With conditions which favored the Warburg effect, i.e. rate-limiting CO2 and 100% O2, O2 inhibition was greater at pH 8.4 to 8.5 than at pH 7.5 to 7.8. At pH 8.5, as compared with 7.8, there was an enhanced 14C-labeling of glycolate, and a decrease of isotope in some phosphorylated Calvin cycle intermediates, particularly triose-phosphate. The 14C-labeling of starch was also more inhibited by O2 at higher pH. The enhanced synthesis of glycolate during 14CO2 assimilation at higher pH resulted in a diminution in the level of phosphorylated intermediates of the Calvin cycle, and this was apparently a causal factor of the increased severity of the Warburg effect.  相似文献

Photosynthetic 14CO2 fixation, [14C]glycolate formation, and the decarboxylation of [1-14C]glycolate and [1-14C]glycine by leaf mesophyll protoplasts isolated from isogenic diploid and tetraploid cultivars of ryegrass (Lolium perenne L.) were examined. The per cent O2 inhibition of photosynthesis in protoplasts from the tetraploid cultivar was less than that of the diploid line at both 21 and 49% O2. Kinetic studies revealed that the Km (CO2) for photosynthesis by the diploid protoplasts was about twice that of the tetraploid line. In contrast, the Ki (O2) for protoplast photosynthesis was similar in both cultivars, as was the potential for oxidizing glycolate and glycine to CO2 via the photorespiratory carbon oxidation cycle. Although the maximal rates of glycolate accumulation by the isolated protoplasts in the presence of 21% O2 and a glycolate oxidase inhibitor were similar in the two cultivars, the percentage of total fixed 14C entering the [14C]glycolate pool and the ratio of the rate of [14C]glycolate formation to 14CO2 fixation at 21% O2 and low pCO2 were about two times greater in protoplasts and intact chloroplasts isolated from the diploid line compared to the tetraploid. These results fully support the recent observation that a doubling of ploidy in various ryegrass cultivars reduced the Km (CO2) of purified ribulose bisphosphate carboxylase-oxygenase by about one-half without affecting the Ki (O2) (Garrett 1978 Nature 274: 913-915).  相似文献

To study the effect of O2 on the photosynthetic and glycolate pathways, maize leaves were exposed to 14CO2 during steady-state photosynthesis in 21 or 1% O2. At the two O2 concentrations after a 14CO2 pulse (4 seconds) followed by a 12CO2 chase, there was a slight difference in CO2 uptake and in the total amount of 14C fixed, but there were marked changes in 14C distribution especially in phosphoglycerate, ribulose bisphosphate, glycine, and serine. The kinetics of 14C incorporation into glycine and serine indicated that the glycolate pathway is inhibited at low O2 concentrations. In 1% O2, labeling of glycine was reduced by 90% and that of serine was reduced by 70%, relative to the control in 21% O2. A similar effect has been observed in C3 plants, except that, in maize leaves, only 5 to 6% of the total 14C fixed under 21% O2 was found in glycolate pathway intermediates after 60 seconds chase. This figure is 20% in C3 plants. Isonicotinyl hydrazide did not completely block the conversion of glycine to serine in 21% O2, and the first carbon atom of serine was preferentially labeled during the first seconds of the chase. These results supported the hypothesis that the labeled serine not only derives from glycine but also could be formed from phosphoglycerate, labeled in the first carbon atom during the first seconds of photosynthesis.  相似文献

Experiments in which [1-14C]glycolate uptake is carried out in conjunction with measurements of stromal pH indicate that only glycolic acid and not the glycolate anion is crossing the pea (Pisum sativum var. Progress No. 9, Agway) chloroplast envelope. This mechanism of glycolate transport appears to be too slow to account for observed photorespiratory carbon fluxes in C3 plants.  相似文献

Fixation of 14CO2 by synchronized cultures of Ankistrodesmus braunii was highest for young growing cells, low for mature cells, and lowest for dividing cells. The amount of 14C excreted during photosynthesis followed the same trend. Cells at the end of the growing phase, after 10 hours of a 16-hour light phase, excreted nearly 35% of the total 14C fixed as one product, glycolate. Dividing cells from the dark phase, when tested in the light, excreted only 4% as much glycolate-14C as the young growing cells. Dividing cells also excreted as much mesotartrate as glycolate and also some isocitrate lactone and an unidentified acid. None of these excreted acids were found inside the cells in significant amounts. Methods for isolation and identification of the excreted acids are present. With 14C-labeled algae, it was shown that the excretion of glycolate was light-dependent and inhibited by 1,1-dimethyl-3-(p-chlorophenyl) urea. The excretion of labeled mesotartrate, isocitrate lactone, and an unknown acid, but not glycolate, also occurred in the dark. The excreted mesotartrate was predominantly carboxyl-labeled even after long periods of 14CO2 fixation. Since glycolate is known to be uniformly labeled, glycolate could not be the precursor of the carboxyl-labeled mesotartrate. The reason for the specific excretion of glycolate, mesotartrate, and isocitrate lactone is not known, but the metabolism of all three acids by the algae may be limited and each can form dilactides or lactones by dehydration. In this context isocitrate lactone was excreted rather than the free acid.  相似文献

When glycolate was metabolized in peroxisomes isolated from leaves of spinach beet (Beta vulgaris L., var. vulgaris) formate was produced. Although the reaction mixture contained glutamate to facilitate conversion of glycolate to glycine, the rate at which H2O2 became “available” during the oxidation of [1-14C]glycolate was sufficient to account for the breakdown of the intermediate [1-14C]glyoxylate to formate (C1 unit) and 14CO2. Under aerobic conditions formate production closely paralleled 14CO2 release from [1-14C]glycolate which was optimal between pH 8.0 and pH 9.0 and was increased 3-fold when the temperature was raised from 25 to 35 C, or when the rate of H2O2 production was increased artificially by addition of an active preparation of fungal glucose oxidase.  相似文献

Summary The amount of 14C-glycolate excreted by Oscillatoria sp. and Anabaena flos-aquae is less than 1% of the 14C fixed by the algae during photosynthesis. Transfer of cells grown on 5% CO2 in air to a medium of low bicarbonate concentration or treatment of the cells with isonicotinyl hydrazide (INH) during photosynthesis, caused little increase in glycolate excretion. -Hydroxysulfonates failed to stimulate massive excretion of glycolate. Although these blue-green algae excreted little glycolate, a significant proportion of the photosynthetically fixed carbon was excreted in the form of basic, neutral and acidic compounds, and such excretion was greater in 5% CO2-grown cells than in air-grown cells.  相似文献

Growth and shorter term incorporation measurements with both wild type Chlamydomonas reinhardtii and a mutant (F-60, lacking phosphori-bulokinase activity) indicate that the rate of glycolate utilization is always relatively low. Growth support with external glycolate is restricted to cells with full photosynthetic capacity. A high concentration of glycolate is required for optimal growth support and incorporation of [14C]glycolate. Glycolate incorporation is low at pH 3.8 even with the relatively free permeability. The F-60 mutant can take up only small quantities of glycolate in spite of photosynthetic electron transport and photophosphorylation competencies. This requirement for photosynthetic carbon metabolism indicates a significant difference in the glycolate pathway of this alga. No growth condition significantly increases glycolate incorporation rates. There is no evidence that one of the primary enzymes, glycolate dehydrogenase, is limiting utilization; measurements of glycolate uptake and excretion do not always correlate with its activity. Since the maximal utilization rate of glycolate is low, control of glycolate formation is important in preventing the loss of this fixed carbon from the algal cell.  相似文献

The effect of 1% and 21% O2 upon 14CO2 assimilation by desert plants exposed for 10 to 90 seconds has been studied. The plants studied can be divided into three groups with respect to O2. The C3 plants display the usual Warburg effect. No changes could be observed in the intensity of photosynthesis as a function of O2 content in another group of plants (showing signs of Crassulacean acid metabolism). In still another group of plants (C4 plants) the stimulating effect of O2 on photosynthesis could be detected. In C3 plants, O2 inhibits the processing of carbon through the Calvin cycle intermediates. The involvement of carbon in the glycolate pathway fails to explain completely the inhibiting effect of O2 on photosynthesis. It is assumed that O2 inhibits the enzymes of the Calvin cycle. In C4 plants O2 stimulates the incorporation of 14C into malate and aspartate. The incorporation of 14C into the intermediates of the Calvin cycle in C4 plants is inhibited much like that in typical C3 plants.  相似文献

Rates and products of photosynthetic 14CO2 fixation by division synchronized cultures of Euglena gracilis strain Z were determined over the cycle. Rate of 14CO2 fixation doubled in a continuous manner throughout the light phase followed by a slight reduction of photosynthetic capacity in the dark phase. Greater 14C incorporation into the nucleic acid-polysaccharide fraction occurred with mature cells. Products of 14CO2 fixation varied markedly over the cycle: although with mature cells 14C-labeled sucrose was not detected, with dividing cells this was the main sugar labeled; in young cells 14C maltose was formed. Cells removed at end of dark phase accumulated 14C in glycolate, whereas at other stages over the cycle less 14C was present in glycolate, and this was accompanied by a rapid incorporation of 14C into glycine and serine. Glycerate was an early and major product of photosynthesis with cells at the mature stage of the cycle.  相似文献

14CO2 assimilation was studied with diploid, tetraploid, hexaploidspecies of the genera Triticum and their wild relatives Aegilops.Attached mature leaves of 3–4 weekold plants were allowedto undergo photosynthesis under air at ambient temperature.The pattern of distribution of 14C was notably similar in Triticumand Aegilops species whatever the level of ploidy. Sucrose wasthe sink for photosynthetic carbon. 14C for sucrose synthesis was supplied either through the glycolatepathway by glycolate, the product of the photorespiration orby the Calvin cycle intermediates exported into the cytoplasm.Depending on the species, the glycolate pathway provided 40to 75%of the sucrose 14C. The higher labeling of sucrose was associated with the greaterparticipation of the glycolate pathway in the wild diploid (DD)A. squarrosa and in the cultivated hexaploid (AABBDD) T. aestivum.The results suggest that the expression of the male D genomeis dominant over the female AB genome in T. aestivum. In T. aestivum under ambient conditions lowering (low temperature)or hindering (1% O2 ) photorespiration, sucrose labeling decreased,but serine and glycine labeling was favoured. Long-term carbon dioxide enrichment, 14CO2 feeding, and partial defoliation were employed as probes to investigate source/sink limitations of photosynthesis during the development of symbiotically grown alfalfa. In the mature crop, long-term CO2 enrichment does not affect the rates of net photosynthesis, relative growth, 14C export to nonphotosynthetic organs, or the rates of 14C label incorporation into leaf sucrose, starch, or malate. The rate of glycolate labeling is, however, substantially reduced under these conditions. When the mature crop was partially defoliated, a considerable increase in net photosynthesis occurred in the remaining leaves. In the seedling crop, long-term CO2 enrichment increased dry matter accumulation, primarily as a result of increases in leaf starch content. Although the higher rates of starch synthesis are not maintained, the growth enhancement of the enriched plants persisted throughout the experimental period. These results imply a source limitation of seedling photosynthesis and a sink limitation of photosynthesis in more mature plants. Consequently, both the supply and the utilization of photosynthate may limit seasonal photosynthesis in alfalfa.  相似文献

Aminooxyacetate (1 millimolar) did not inhibit photosynthetic 14CO2 fixation by Chlamydomonas reinhardtii Dangeard, (−) strain (N.90) but greatly stimulated the biosynthesis and excretion of glycolate. Similar results were obtained from cells grown with 5% CO2 or low CO2 (air). After 2 minutes with air-grown cells, [14C]glycolate increased from 0.3% of the total 14C fixed by the control to 11.7% in the presence of aminooxyacetate and after 10 minutes from 3.8% to 41.1%. Ammonium nitrate (0.2 millimolar) in the media blocked the aminooxyacetate stimulation of glycolate excretion. Chromatographic analyses of the labeled products in the cells and supernatant media indicated that aminooxyacetate also completely inhibited the labeling of alanine while some pyruvate accumulated and was excreted. A high percentage (35%) of initial 14CO2 fixation was into C4 acids. Initial products of 14CO2 fixation included phosphate esters as well as malate, aspartate, and glutamate in treated or untreated cells. Lactate was also a major early product of photosynthesis, and its labeling was reduced by aminooxyacetate. Inasmuch as lactate was not excreted, glycolate excretion seemed to be specific. When photosynthesis was inhibited by 3-(3,4-dichlorophenyl)-1,1-dimethylurea, labeled organic and amino acids but not phosphate esters were lost from the cells. Aminooxyacetate did not inhibit the enzymes associated with glycolate synthesis from ribulose bisphosphate.  相似文献

When photosynthesis of the blue-green alga Anacystis nidulans was measured as 14CO2-fixation, the inhibitory effect of DCMU at low concentrations was greatest when mainly Photosystem 1 (PS 1) (excitation at 446 or 687 nm) was operative. At concentrations above 10-6M the inhibition on 14CO2-fixation was greatest when mainly Photosystem 2 (PS 2) was operative (excitation at 619). During excitation of PS 1, the excretion of glycolate was stimulated at low concentrations of DCMU (5 × 10-8M and lower), while higher concentrations inhibited excretion. All concentrations of DCMU inhibited glycolate excretion when mainly PS 2 was excited. The curves showing the relative effect of DCMU on the two photosystems, measured as PS 1/PS 2, had opposite shapes for 14CO2-fixation and glycolate excretion. An increase in 14CO2-fixation coincided with a decrease in glycolate excretion and vice versa. It appears that the increased rate of photosynthesis when mainly PS 1 was operative relative to that when mainly PS 2 was excited, increases the consumption of glycolate in an oxidation process associated with the excitation of PS 1, resulting in less excretion of glycolate to the medium. The influence of DCMU inhibition on labelled amino acid pools connected to the glycolate pathway (glycine-serine) is quite similar to that for 14CO2-fixation. At concentrations below 10-6M DCMU, inhibition of 14CO2- incorporation into the amino acids was greatest when PS 1 was excited, while at the higher concentrations tested, inhibition was greater when PS 2 was excited. We conclude that the metabolism of glycine and serine is closely connected to the rate of photosynthesis.  相似文献

Glycolate synthesis was inhibited 40–50% in illuminated tobacco leaf disks, which have rapid rates of photorespiration, when floated on 20 mm potassium glycidate (2,3-epoxypropionate), an epoxide similar in structure to glycolate. The inhibitor also decreased the release of photorespiratory CO2 about 40%, and the specificity of glycidate was demonstrated by the 40–50% increase in rate of photosynthetic CO2 uptake observed in its presence. The importance of glycolate synthesis and metabolism in the production of photorespiratory CO2 and the role of glycolate in diminishing net photosynthesis in species with rapid rates of photorespiration was thus further confirmed. L-(or 2S)-Glycidate was slightly more active than DL-glycidate, but glycidate was more effective as a specific inhibitor in leaf tissue than several other epoxide analogs of glycolate examined. The products of photosynthetic 14O2 fixation after 3 or 4 min of uptake were proportionately altered in the presence of glycidate, and the specific radioactivity of the [14C]glycolate produced was closer to that of the 14CO2 supplied. Glycidate inhibited glycolate synthesis in tobacco leaf disks irreversibly, since the degree of inhibition was the same for at least 2 hr after the inhibitor solution was removed. Glycidate also blocked glycolate synthesis in maize leaf disks, tissue with low rates of photorespiration, but large increases in net photosynthesis were not observed in maize with glycidate, because glycolate synthesis is normally only about 10% as rapid in maize as in tobacco. The demonstration of increases in net photosynthesis of 40–50% when glycolate synthesis (and photorespiration) is blocked with glycidate indicates in an independent manner that the biochemical or genetic control of photorespiration should permit large increases in plant productivity in plant species possessing rapid rates of photorespiration.  相似文献

The δ13C values of several seagrasses were considerably less negative than those of terrestrial C3 plants and tended toward those of terrestrial C4 plants. However, for Thalassia hemprichii (Ehrenb.) Aschers and Halophila spinulosa (R. Br.) Aschers, phosphoglycerate and other C3 cycle intermediates predominated among the early labeled products of photosynthesis in 14C-labeled seawater (more than 90% at the earliest times) and the labeling pattern at longer times was brought about by the operation of the C3 pathway. Malate and aspartate together accounted for only a minor fraction of the total fixed label at all times and the kinetic data of this labeling were not at all consistent with these compounds being early intermediates in seagrass photosynthesis. Pulse-chase 14C-labeling studies further substantiated these conclusions. Significant labeling of photorespiratory intermediates was observed in all experiments. The kinetics of total fixation of label during some steady-state and pulse-chase experiments suggested that there may be an intermediate pool of inorganic carbon of variable size closely associated with the leaves, either externally or internally. Such a pool may be one cause for the C4-like carbon isotope ratios of seagrasses.  相似文献

A method was devised to quantify short-term photorespiratory rates in terrestrial plants using 18O-intermediates of the glycolate pathway, specifically glycolate, glycine, and serine. The pathway intermediates were isolated and analyzed on a GC/MS to determine molecular percent 18O-enrichment. Rates of glycolate synthesis were determined from 18O-labeling kinetics of the intermediates, derived rate equations, and nonlinear regression techniques. Glycolate synthesis in wheat (Triticum aestivum L.), a C3 plant, and maize (Zea mays L.), a C4 plant, was stimulated by high O2 concentrations and inhibited by high CO2 concentrations. The synthesis rates were 7.3, 2.1, and 0.7 micromoles per square decimeter per minute under a 21% O2 and 0.035% CO2 atmosphere for leaf tissue of wheat, maize seedlings, and 3-month-old maize, respectively. Photorespiratory CO2 evolution rates were estimated to be 27, 6, and 2%, respectively, of net photosynthesis for the three groups of plants under the above atmosphere. The results from maize tissue support the hypothesis that C4 plants photorespire, albeit at a reduced rate in comparison to C3 plants, and that the CO2/O2 ratio in the bundle sheath of maize is higher in mature tissue than in seedling tissue. The pool size of the three photorespiratory intermediates remained constant and were unaffected by changes in either CO2 or O2 concentrations throughout the 10-minute labeling period. This suggests that photorespiratory metabolism is regulated by other mechanism besides phosphoglycolate synthesis by ribulose-1,5-bisphosphate carboxylase/oxygenase, at least under short-term conditions. Other mechanisms could be alternate modes of synthesis of the intermediates, regulation of some of the enzymes of the photorespiratory pathway, or regulation of carbon flow between organelles involved in photorespiration. The glycolate pool became nearly 100% 18O-labeled under an atmosphere of 40% O2. This pool failed to become 100% 18O-enriched under lower O2 concentrations.  相似文献
