Summary The viability of the chrysophycean flagellate,Poterioochromonas malhamensis, was examined after treatment of the cells with high temperatures. The fine structure of the cells was studied after heat-shock (42 °C, 16 minutes). Heat injury effects are visible at the nucleus, the chloroplast (distortion of the thylakoids), the mitochondria (increased occurrence of matrical crystalline inclusions) and, especially, at the dictyosome which is completely destroyed into some single vesicles and remnants of cisternae. Most damages are repaired within one hour; the reconstitution of the dictyosome takes 3–6 hours. It is inhibited severely by actinomycin D.The effect on the dictyosome reflects its labile position in the endomembrane system. The dependence of its reconstitution on protein synthesis indicates that membrane components are destroyed by the heat-shock.  相似文献

Summary Cytochemical analysis of the endomembrane system of the chlamydomonad flagellate,Gloeomonas kupfferi (Chlorophyta), reveals distinct compartmentalization. Phosphatase localization shows that: IDPase is located throughout all cisternae of the dictyosome and vesicles associated with the contractile vacuole. Other alkaline phosphatases like TPPase, ATPase and ITPase were localized within the trans-face cisternae and vesicles of the contractile vacuole. IMPase was localized at the plasmamembrane and not within the endomembrane system. Acid phosphatases, incl. CMPase, NADPase and -glycerophosphatase, were localized in vesicles emerging from the central terminus of the trans-face of the dictyosome and in the peripheral vacuolar network. Silver proteinate labeling was noted in the dictyosome, contractile vacuole and on the anterior plasmamembrane. Summary The connections and structural inter-relations of dictyosomes and endoplasmic reticulum (ER) in cotyledons of germinating mung beans were studied using thick (0.3 m) sections of aldehyde fixed, zinc iodide-osmium tetroxide (ZIO) impregnated tissue. The sections were examined by conventional (100 kV), rather than high voltage, transmission electron microscopy.Continuity of cisternal ER with tubular ER was confirmed and a direct connection of tubular ER totrans dictyosome cisternae was observed as were GERL networks associated withtrans dictyosome cisternae.Dictyosomes also gave rise to an extensive system of very fine tubules (10–20 nm diam) which have not been described previously in plant tissue. These tubules, which originated at thetrans dictyosome face, extended throughout the cytoplasm and were found connected to cisternal ER and tubular ER.The implications of these observations are discussed with regard to present ideas concerning endomembrane flow and protein sorting by the Golgi apparatus.  相似文献

Summary The toxic effect of pentachlorophenol (PCP) on the growth and ultrastructure of tobacco pollen tubes was tested using a semivivo technique of tube culture. In this technique the pollen tubes were allowed to grow in the pistilin situ for 24 hr before they protruded from the cut end of the style and came into contact with the medium containing PCP. The inhibitory effect of different PCP concentrations was determined by measuring the length of tube bundles. The intracellular action of PCP was analysed by electron microscopy. This biocide caused four obvious alterations in the pollen tube ultrastructure: (1) swelling of the mitochondrial saccules; (2) enlargement of the dictyosomes by the increase of the cisternal diameter and the number of cisternae per stack; (3) formation of cup-shaped Golgi apparatus-endoplasmic reticulum hybrid structures (GER hybrids) showing continuities of ER and Golgi cisternae; (4) formation of stacked and/or concentric arrangements of rough ER cisternae. It is suggested that swelling of saccules was directly due to the uncoupling of oxidative phosphorylation whereas the changes of the endomembrane system were caused by energy depletion due to the inhibition of ATP synthesis. These changes are consistant with dynamic concepts of dictyosome and ER function when membrane formation exceeds membrane use in the production of secretory and transition vesicles. Thus, the enlargement of the dictyosomes and the formation of GER hybrids are thought to result from inhibition of budding of vesicles from the Golgi apparatus or from both the ER and the Golgi apparatus, respectively.  相似文献

Abstract. The highly complex and symmetric endomembrane network of the chlamydomonad alga, Gloeomonas kupferri , can be disrupted by the herbicide, isopropyl N-phenyl carbamate (IPC). Upon incubation with IPC for between 30 min and 1 h, curling of the dictyosomes, proliferation of small vesicles from the trans face and swelling of the peripheral vacuolar complex takes place. Longer treatments result in degeneration of dictyosome cisternae into vesicular masses and the formation of supervacuolar complexes. Enzyme cytochemistry revealed that endomembrane 'marker' enzymes are altered/inhibited by IPC. A second but striking I PC-induced effect is the production of branched, fibrillar inclusions in the cell nucleus. A discussion of I PC-induced affects on ion regulation and microtubule/MTOC distribution in the cell is presented.  相似文献

Summary The ultrastructural changes which occur during the foliar abscission ofImpatiens sultani Hook. andColeus blumei Benth. have been described. In both cases fracture of the separation zone results from a modification of the wall and cleavage along the line of the middle lamella. This process starts at the corners of the cells and in regions rich in plasmodesmata.During the period of wall breakdown, cellular integrity is maintained and the membrane degradation described by others was not observed. Plasmolysis studies confirmed that the separation zone cells retained their selective permeability characteristics until well after wall fracture. Quantitative data are presented to show that there is an increase in the frequency of rough endoplasmic reticulum, dictyosomes and dictyosome vesicles during the period when wall-hydrolyzing enzyme secretion is occurring. These changes are interpreted as reflecting an increase in demand for the secretory machinery of the endomembrane system. The possible involvement of plasmodesmata in this process is also discussed.  相似文献

Summary The peripheral reticulum (PR) inCyphomandra betacea chloroplasts originates as vesicles budding from the inner membrane of the chloroplast envelope which elongate to form tubules then aggregate or branch to form discrete PR units. Individual PR units of many coiled tubules may be connected with other units by narrow tubules. Serial sectioning revealed the discrete units to be approximately 650–1,000 nm wide, 400–500 nm high and 500–600 nm deep and to possess a compact morphology. TheCyphomandra PR structure is compared to the morphologies of chloroplast reticula reported for other plant species. A scheme to group PR from different species into 3 distinct morphological categories is outlined and discussed.  相似文献

Summary Uniseriate filaments of the freshwater red algaCompsopogon coeruleus were examined by transmission electron microscopy for details of vegetative organization and cell division with the goal of providing useful taxonomic characters. Each cell's single, complex chloroplast contains a peripheral encircling thylakoid, and unlike the vast majority of red algae, the cis-regions of dictyosomes are not consistently juxtaposed with mitochondria. These subcellular features, which are present in all examined genera in theCompsopogonales, Erythropeltidales, andRhodochaetales, along with certain unique reproductive characteristics, unify these three orders. During mitosis in uncorticated axial cells, a small, ring-shaped nucleus associated organelle (NAO) is located at each division pole, an intranuclear spindle comes to a moderately acute focus at the flattened, fenestrated metaphase-anaphase division poles and perinuclear ER partially encloses dividing nuclei, including a well-developed interzonal midpiece. The cleavage furrow penetrates the large, central vacuolar region to separate daughter nuclei. These cell division features most closely resemble the pattern described for the orderCeramiales. Summary This study follows the maturation of the pollen grain of cotton (Gossypium hirsutum L.), particularly the development of the vegetative cytoplasm and the various storage products formed. CTEM, HVEM, stereoscopy, and cyto-histochemistry were used to examine the events occurring during the 9 days before anthesis. Starch began to accumulate in plastids at anthesis minus 9 days and reached a peak concentration shortly before anthesis; lipid deposition followed a similar pattern, but started at 6 days before anthesis. Lipid bodies were always seen closely oppressed to the endoplasmic reticulum (ER). Dictyosomes appear active during the entire 9 days; first producing vesicles involved in the formation of the intine and, later, producing vesicles stored in the pollen grain. The dictyosome vesicles appear to contain polysaccharides and concentrate in layers around the lipid bodies. Ribosomes increase in number from 6 days before anthesis and are particularly numerous in the mature pollen. From anthesis minus 6 days until anthesis, the ER cisternae become increasingly inflated and, in the hours immediately before pollen release, form pockets filled with lipid bodies and dictysosome vesicles. Young sieve elements from petioles and rachises of Davalliafijiensis Hook were examined with an electron microscope. Evidencewas obtained that implicated both the endoplasmic reticulum(ER) and the Golgi apparatus in the formation of refractivespherules. Numerous connections were observed between smooth,tubular ER and peripheral tubules of the dictyosomes, indicatingthat these two cytoplasmic components are parts of a singleendomembrane system. Summary The ultrastructure and composition of the pollen tube of cotton (Gossypium hirsutum) growing in the tissues of the stigma and style of the flower were examined. The distal portion of the tube is densely cytoplasmic and contains the vegetative nucleus and the two sperms. The vegetative nucleus is highly convoluted and the membrane contains many pores and connections with the ER. No organized nucleolus is present but 4–6 membrane-bound, protein containing bodies are found in the nucleus. Mitochondria containing numerous cristae are abundant in the cytoplasm. Dictyosomes are also plentiful and are engaged in the production of many large vesicles. Rough ER is conspicuous and polysomes are found in the cytoplasm. Plastids are few in number, poorly developed, and contain little starch. Many uniform, small vesicles are found throughout the cytoplasm. Lipid bodies frequently with small vesicles associated with them are found in the tube. In the proximal region vacuoles form and the cytoplasm becomes pressed against the wall. In the transition zone the ER frequently becomes distended and filled with protein. The wall has two distinct layers: one strongly PAS positive, the other faintly PAS positive. The inner wall is apparently formed by the deposition of large dictyosome vesicles. Plug structure and development were studied.  相似文献

Summary Placental cells line the ovarian transmitting tract inLilium regale and produce exudates for secretion. Sections through the highly lobed nuclei of these cells reveal the presence of membrane profiles which form vesicles with varying dimensions in cross section. Computer reconstruction of the nucleus reveals that the vesicle profiles form a complex reticulum of tubular cisternae, which spans the whole nucleus, enclosing a maze of continuous lumen space. Connections between the vesicles and the inner nuclear envelope are visible at various points along the nuclear envelope. This complex network of tubules which constitutes the reticulum arises from the inner nuclear membrane. The nuclear reticulum dramatically increases the inner-envelope surface area, comprising 82% of the total membrane perimeter of inner nuclear envelope and nuclear reticulum. The inner nuclear envelope invaginates into the nucleus forming the nuclear reticulum and the outer nuclear envelope evaginates into the endoplasmic reticulum (ER), indicating that there is a continuity between the lumens of the nuclear reticulum and the ER. The nuclear reticulum is labelled with zinc iodide-osmium tetroxide, a staining pattern identical to that seen in the ER. Positive reaction to the zinc iodide-osmium tetroxide indicates that the nuclear reticulum is a site for Ca2+ deposition. The nuclear reticulum forms an extension of the endomembrane system which reaches deep into the nucleoplasm. Summary Protophloem sieve elements (PSEs) in roots of wheat (Triticum aestivum L.) are arranged in single vertical files. The number of PSEs within the files increases by symmetrical divisions, which take place after the completion of asymmetrical (formative) divisions and before the initiation of differentiation. The divisions are preceded by well defined pre-prophase bands (PPB) of microtubules, which surround the nucleus in an equatorial position. In the cytoplasmic region between the nuclear surface and the PPB, perinuclear and endoplasmic microtubules were observed. The perinuclear microtubules are considered as part of the developing spindle, while the endoplasmic ones interlink the perinuclear microtubules with the PPB. Dividing cells do not show any signs of incipient differentiation. The first and most reliable indication of a commencing differentiation is provided by the sieve-element plastids that begin to accumulate dense crystalloid inclusions in the very young PSEs. In mature PSEs plastids contain two kinds of crystalloid inclusions, dense and thin, in a translucent stroma. Depending on the plastid-inclusions criterion it was shown that: (a) the PSEs of a given root do not initiate differentiation at exactly the same stage, (b) the developmental sequence extends to a span of 7–9 actively differentiating PSEs arranged in a single vertical file, and (c) each PSE needs about 16–21 h to pass through the whole developmental sequence. In the last two differentiating PSEs of a file, mitochondria were found to be enveloped by single cisternae of ER. The association is temporary as it is lost in the first PSEs with an autolysed lumen. During differentiation, Golgi bodies were abundant and active in producing vesicles involved in cell wall development. Golgi vesicles were also found among the microtubules of the PPB, but no local thickening was observed. The peripheral secretion tissue of the root cap of Lepidium sativum L. was investigated by electronmicroscopy and freeze-fracturing in order to study structural changes of membranes involved in the secretion process of polysaccharide slime. Exocytosis of slime-transporting vesicles occurs chiefly in the distal region of the anticlinal cell walls. The protoplasmic fracture face (PF) of the plasmalemma of this region is characterized by a high number of homogenously distributed intramembranous particles (IMPs) interrupted by areas nearly free of IMPs. Near such areas slime-transporting vesicles are found to be underlying the plasma membrane. It can be concluded that areas poor in particles are prospective sites for membrane fusion. During the formation of slime-transporting vesicles, the number of IMPs undergoes a striking change in the PF of dictyosome membranes and their derivatives. It is high in dictyosome cisternae and remarkably lower in the budding region at the periphery of the cisternae. Slime-transporting vesicles are as poor in IMPs as the areas of the plasmalemma. Microvesicles rich in IMPs are observed in the surroundings of dictyosomes. The results indicate that in the plasmalemma and in membranes of the Golgi apparatus special classes of proteins — recognizable as IMPs — are displaced laterally into adjacent membrane regions. Since the exoplasmic fracture face (EF) of these membranes is principally poor in particles, it can be concluded that membrane fusion occurs in areas characterized by a high quantity of lipid molecules. It is obvious that the Golgi apparatus regulates the molecular composition of the plasma membrane by selection of specific membrane components. The drastic membrane transformation during the formation of slime-transporting vesicles in the Golgi apparatus causes the enrichment of dictyosome membranes by IMPs, whereas the plasma membrane probably is enriched by lipids. Summary Cytokinesis in Impatiens sultani microspore mother cells is simultaneous. It starts with the formation of small ingrowths of the surrounding callosic wall. Next, an incomplete cell plate is formed by fusion of small dictyosome vesicles. The cell plate consists of a network of anastomosing tubules and sacs. Aggregates of fusing vesicles are associated with bundles of microtubules, which are oriented perpendicular to the plane of the future cell walls. In the sacculate parts of the cell plate, some callose is deposited, while the associated microtubules disappear. The cell walls ultimately develop by enlargement of the previously formed wall ingrowths, which successively incorporate the elements of the cell plate. The enlargement and thickening of the walls is not accompanied by a further fusion and incorporation of dictyosome vesicles.  相似文献

Summary Using cryo-fixation and freeze-substitution electron microscopy, the effects of brefeldin A (BFA) on the structure of the trans-Golgi network (TGN), the endoplasmic reticulum (ER), and Golgi bodies in the unicellular green algaBotryococcus braunii were examined at various stages of the cell cycle. In the presence of BFA, all the TGNs of interphase and dividing cells aggregated to form a single tubular mass. In contrast, the TGNs decomposed just after cell division and disappeared during cell wall formation. Throughout the cell cycle, the TGN produced at least six kinds of vesicles, of which two were not formed in the presence of BFA: vesicles with a diameter of 200 nm and fibrillar substances, which formed in interphase cells; and vesicles with a diameter of 180–240 nm, which may participate in septum formation. In addition, the number of clathrin-coated vesicles attaching to the TGN decreased. In interphase cells, BFA induced the disassembly of Golgi bodies and an increase in the smooth-ER cisternae at the cis-side of Golgi bodies. This result may suggest the existence of retrograde transport from the Golgi bodies to the ER in the presence of BFA. Summary The metabolism of P-particles (polysaccharide particles) was investigated in mature pollen grains ofEryngium campestre L. Numerous P-particles, originating from dictyosome activity, are found to be accumulated near the apertures, followed by mitochondria. A single layer of ER profiles seems to prevent the fusion of the P-particles with the intine. Instead of this, they fuse with each other forming nonmembrane-bounded polysaccharide-aggregates, which subsequently change their granulated structure to an amorphous. Mitochondria together with small vesicles are involved in the conversion-process. The so formed wall precursors pass through the ER and fuse into the intine.  相似文献

Giardia lamblia,a primitive eukaryotic cell, lacks organelles such as mitochondria, peroxisomes, and a typical Golgi complex and presents a system of vesicles located below the plasma membrane. We used fluorescence and electron microscopy to better characterize the peripheral vesicles. Incubation of living cells with acridine orange showed that the peripheral vesicles correspond to an acidic compartment. Incubation with lucifer yellow, and with horseradish peroxidase, showed labeling of the peripheral vesicles even after several hours. Acid phosphatase was localized in the endoplasmic reticulum and in most of the peripheral vesicles. On the other hand, glucose 6-phosphatase, an endoplasmic reticulum marker, was observed in the endoplasmic reticulum cisternae and in some peripheral vesicles. A similar labeling pattern was observed using the zinc iodide technique, which reveals SH-containing proteins. Three-dimensional reconstruction and electron microscopy tomography of cells stained for acid phosphatase and glucose-6-phosphatase revealed the connection between some vesicles and profiles of the endoplasmic reticulum. Taken together, our observations suggest that trophozoites ofG. lambliapresent an endosomal–lysosomal system concentrated in a single system, the peripheral vesicles, which may represent an ancient organellar system that later on subdivided into compartments such as early and late endosomes and lysosomes.  相似文献

Electronmicroscopic study of Coleps, Colpidium, Stylonychia, and especially of Paramecium confirmed the presence of the Golgi complex in these fresh-water ciliates. The complex consisted of numerous dictyosomes scattered throughout the cytoplasm. Each dictyosome included a few flat, partly reticulated saccules lying parallel to a cistern of rough endoplasmic reticulum which was free of ribosomes on the side exposed to the dictyosome. A unique layer of vesicles, characterized by constant size and a thick wall, separated the endoplasmic reticulum from the dictyosomes. The vesicles could be regarded as transition vesicles. Coated vesicles were seen in continuity with some of the flattened saccules. The possible role of the Golgi complex in the physiology of ciliates is discussed.  相似文献

Summary Secretory trichomes develop from epidermal cells on the leaf primordia and stem ofPharbitis nil. Following an initial growth phase, trichomes begin active secretion of a protein-carbohydrate mucilage. This mucilage covers the shoot apex and developing leaves ofPharbitis.The secretory cells possess cellular organelles in forms usually associated with actively secreting cells: many mitochondria, an elaborate network of rough endoplasmic reticulum (RER), many free ribosomes, and numerous dictyosomes. The role of the dictyosomes is twofold: 1. dictyosome vesicles bud coated vesicles which transport materials from the cell and, 2. dictyosome vesicles coalesce, forming large storage vesicles. The storage vesicles are surrounded by, and often in contact with, poculiform RER. The RER forms an interconnected network throughout the cytoplasm, extending from the nuclear envelope to the plasmalemma. Distended profiles of RER are frequently in direct contact with the plasmalemma. Thus, inPharbitis secretory trichomes, it is the coated vesicles and RER which are active in secretion export. These findings imply a secretory pathway which deviates from the usual pattern in glandular cells.Predoctoral fellow of National Science Foundation during part of the investigation.  相似文献
