The preference of guinea pigs for bedding materials, wood shavings (WS) or paper cutting sheets (PS), was studied. Animals aged 8 weeks and 20 weeks showed a similar behaviour pattern during 30 min in the light, preferring WS to PS regardless of ages. Over both light and dark periods for 24 h, guinea pigs apparently preferred WS in the light, spending much more time resting in them than in PS. In the dark, the border-crossing was significantly more frequent than in the light, and the staying time was rather longer in PS than WS. The results suggest that guinea pigs prefer different bedding materials under light and dark conditions.  相似文献

Differences in morningness‐eveningness among humans are commonly ascribed to circadian parameters, such as circadian period and responsivity to environmental time cues, as well as homeostatic sleep drive. Light is the primary synchronizer of the human biological clock, and if circadian differences exist between morning and evening types, they should have different phase angles of entrainment to the light/dark cycle; that is, morning and evening types should have different patterns of light exposure relative to endogenous circadian phase (ECP). When phase angle of entrainment is strictly defined as the relationship between a marker of ECP and the timing of light exposure, such differences have been demonstrated in the laboratory under controlled light/dark cycles and have recently been shown under conditions of spring and summer light exposure outside the laboratory, taking into account the variable intensity of light. Here, we report similar results from a large (n=66), diverse cohort of morning and evening types across the age span studied at all different times of the year. Differences between morning and evening types in light exposure relative to ECP, indicative of a difference in the phase angle of entrainment to the external light/dark cycle, were found. Specifically, evening types, compared to morning types, had a higher ratio of phase advancing to phase delaying by light. We interpret this as indicating a longer circadian period (τ) in evening types.  相似文献

The changes in the distribution of dark, light and intermediate pyriform neurocytes (Purkinje's cells) in the cerebellum of mice which developed under the conditions of protein-caloric deficiency were assayed from the 10th to the 40th day of mouse life. In the control animals, the number of dark cells was 7 +/- 3%, of intermediate 44 +/- 7%, and that of light ones 49 +/- 7%. Under malnutrition the number of dark cells rose to 26 +/- 5% (P less than 0.01), that of intermediate cells fell to 33 +/- 6% (P less than 0.01), and the number of light cells changed insignificantly (41 +/- 6%, P less than 0.1). Electron microscopy of the ultrastructure of dark cells has disclosed dystrophic and destructive changes in the nucleus and in the main organelles of the cytoplasm. Pronounced astroglial reaction was not infrequently observed around dark cells.  相似文献

Ozone resistance of spores of 2 strains of Bacillus isolated from laboratory animals was examined. Each of 0.02 ml of phosphate-buffered saline at pH 7.0 containing 10(6) Bacillus spores was dropped onto sterilized filter strips, wood chip bedding, pellets of diet, cloth pieces and stainless steel plates. After drying at room temperature, the test materials were exposed to ozone gas of different concentrations at 90% RH. Exposure to 200ppm ozone for 6 hours was sufficient to kill spores in filter strips, but a little higher concentration or a little longer period of ozone fumigation was necessary for sterilization of wood chips, cotton cloth pieces and steel plates. The present results indicated that 600ppm ozone fumigation for 6 hours might be effective for routine sterilization of cages, wood bedding, working clothes and other materials used in laboratory animal facilities. However, exposure to ozone gas of 500 or 1,000 ppm for 6 hours or 200 ppm for 24 hours could not kill spores in pellets of diet, suggesting that dietary protein inhibited the bactericidal activity of ozone.  相似文献

To investigate the mechanism involved in the reduction of body core temperature (T(core)) during fasting in rats, which is selective in the light phase, we measured T(core), surface temperature, and oxygen consumption rate in fed control animals and in fasted animals on day 3 of fasting and day 4 of recovery at an ambient temperature (T(a)) of 23 degrees C by biotelemetry, infrared thermography, and indirect calorimetry, respectively. On the fasting day, 1) T(core) in the light phase decreased (P < 0.05) from the control; however, T(core) in the dark phase was unchanged, 2) tail temperature fell from the control (P < 0.05, from 30.7 +/- 0.1 to 23.9 +/- 0.1 degrees C in the dark phase and from 29.4 +/- 0.1 to 25.2 +/- 0.2 degrees C in the light phase), 3) oxygen consumption rate decreased from the control (P < 0.05, from 24.37 +/- 1.06 to 16.24 +/- 0.69 ml. min(-1). kg body wt(-0.75) in the dark phase and from 18.91 +/- 0.64 to 14.00 +/- 0.41 ml. min(-1). kg body wt(-0.75) in the light phase). All these values returned to the control levels on the recovery day. The results suggest that, in the fasting condition, T(core) in the dark phase was maintained by suppression of the heat loss mechanism, despite the reduction of metabolic heat production. In contrast, the response was weakened in the light phase, decreasing T(core) greatly. Moreover, the change in the regulation of tail blood flow was a likely mechanism to suppress heat loss.  相似文献

为探明不同波段LED光对毛健夜蛾Brithys crini (Fabricius)行为的影响, 在实验条件下观察了4种不同波段(红光: 620~625 nm; 绿光: 520~523 nm; 蓝色: 465~467 nm; 白光: 460~465 nm)LED光源下毛健夜蛾幼虫的假死、 爬行、 静止、 取食、 排便、 瞭望及其他行为和成虫交配及产卵行为。结果表明: (1)毛健夜蛾幼虫在红光、 绿光、 蓝光和白光4种LED光源下, 爬行、 静止和取食行为时间分配差异极显著(爬行、 静止和取食: P=0.000), 红光组爬行的时间占98.06%, 绿光组取食时间占83.65%, 蓝光组爬行和静止时间占91.56%, 白光组爬行时间占61.16%; 爬行、 静止、 取食、 瞭望及其他行为发生率差异显著, 红光组和白光组爬行行为发生率最高, 分别为47.37%和32.00%, 绿光组取食行为发生率最高为40.00%, 蓝光组爬行行为发生率最高为32.35%; (2)毛健夜蛾跌落幼虫在4种LED光源下, 假死发生率差异不显著, 但假死持续时间有显著性差异, 红光组假死持续时间显著长于蓝光组和白光组; (3)4种LED光源处理后, 交配和未交配的单雌产卵量无显著差异; (4)第1-2天的暗期分别使用4种光源, 则交配和产卵高峰均出现在第3天的暗期, 而第1-2天的暗期不使用光源, 则交配和产卵高峰出现在第1天的暗期。这些研究结果初步揭示了不同波段光源对毛健夜蛾行为影响不同。  相似文献

Gastric dilatation (GD) has been observed in Tac:(SW)fBR surveillance mice, with mean age of 10 months, that are exposed to high levels of environmental antigens during routine exposure to dirty bedding. The aim of the study reported here was to determine whether GD was associated with other systemic conditions affecting mice. Three groups of nine animals including-surveillance mice not exposed to dirty bedding (control), surveillance mice with out GD (NGD), and surveillance mice with GD (group GD)-had mean stomach weight with ingesta of 0.5 +/- 0.02 g, 1.09 +/- 0.07 g (P < 0.0001), and 2.54 +/- 0.4 g (P < 0.0001), respectively. Mean serum creatinine concentration was significantly higher in GD (1.6 +/- 0.25 mg/dl), compared with NGD (0.17 +/- 0.22 mg/dl, P < 0.0001) and control (0.2 +/- 0.16 mg/ dl, P < 0.0001) mice. In addition, lesions consistent with severe chronic nephropathy and mild gastritis were common in GD, compared with NGD and control mice. Finally, serum amidated gastrin concentration was significantly high in GD (179.37 +/- 53.86 pM, P < 0.03) and NGD (264.89 +/- 115.89 pM, P < 0.009), compared with control (60.77 +/- 8.39 pM) mice. Gastric dilatation syndrome is associated with chronic nephropathy, hypergastrinemia, and gastritis in surveillance mice exposed to high levels of environmental antigens.  相似文献

This study aimed to determine whether Syrian (golden) hamsters, Mesocricetus auratus, prefer certain bedding materials and whether bedding material can affect paw condition, body weight gain and wheel-running activity. In a first experiment, 26 male hamsters had access to two connected cages, each cage containing a different bedding material (either pine shavings, aspen shavings, corn cob or wood pellets). In a second experiment, 14 male hamsters had access to four connected cages that contained the different bedding materials and also a piece of paper towel to serve as nest material. In a third experiment, 30 male hamsters were each placed in a single cage, 10 of them with pine shavings, 10 with aspen shavings and 10 with corn cob, and they were monitored for 50 days. Significant preferences in the first experiment were: pine shavings over aspen shavings, corn cob over wood pellets, pine shavings over corn cob and aspen shavings over wood pellets (aspen shavings versus corn cob was not tested). However, there was no significant preference expressed in the second experiment, suggesting that the general preference for shavings in the first experiment was based on bedding material suitability as a nesting material. No significant effect of bedding material on paw condition, body weight gain and wheel-running activity was detected. None of the four bedding materials tested in this study can be judged to be inappropriate in the short term if nesting material is added to the cage and if the litter is changed regularly.  相似文献

Unlike mice, adult laboratory rats do not spontaneously build nests when nesting material is offered. As a result, nesting material is often regarded as unsuitable environmental enrichment for laboratory rats. Wild rats and pet rats, however, have been observed to build complex nests from nesting material at hand. It was hypothesized that nest building in rats is an acquired behaviour, rather than genetically predisposed. To test this hypothesis, the progeny of three Wistar rats provided with nesting material (Kleenex tissues) during pregnancy and three standard-housed rats were divided in 34 same-sex couples with access to nesting material: (1) from the age of 8 weeks (n=7); (2) from weaning (n=8); (3) from birth (n=17). The latter were subdivided into two groups after weaning, one provided with Kleenex tissues (n=9), the other with Enviro-dri (n=8). At the age of 12 weeks, all couples were provided with both types of nesting material for one week. Amount, shape, and soiling of the nesting material were scored on weekdays, and behaviour of the rats was scored twice weekly. Results indicated that the older the rats were when first provided with nesting material, the greater the amount of nesting material eaten and soiled, and the nests built were less elaborate. Overall, Enviro-dri was eaten less than Kleenex tissues. It is concluded that rats need to learn the proper use of nesting material. When provided from birth, nesting material is a suitable type of environmental enrichment for laboratory rats.  相似文献

The renin-angiotensin system (RAS) has been implicated in the cardiovascular complications of diabetes. We showed that a high-fructose diet increases blood pressure and plasma angiotensin and impairs glucose tolerance. We investigated the role of angiotensin AT(1a) receptors in the development of fructose-induced cardiovascular and metabolic dysfunction. Male angiotensin AT(1a) knockout (AT1aKO) and wild-type (AT1aWT) mice with arterial telemetric catheters were fed a standard diet or one containing 60% fructose. Fructose increased mean arterial pressure (MAP) in AT1aWT but only during the dark phase (8% increase). In AT1aKO mice, fructose unexpectedly decreased MAP, during both light and dark periods (24 and 13% decrease, respectively). Analytical methods were used to measure systolic arterial pressure (SAP) and pulse interval (PI) variability in time and frequency domains. In fructose-fed AT1aWT mice, there was an increase in SAP variance and its low-frequency (LF) domain (11 +/- 3 vs. 23 +/- 4 mmHg(2), variance, and 7 +/- 2 vs. 17 +/- 3 mmHg(2), LF, control vs. fructose, P < 0.004). There were no changes in SAP variance in AT1aKO mice. Depressor responses to alpha(1)-adrenergic blockade were augmented in fructose-fed AT1a WT compared with AT1aKO mice. Fructose inhibited glucose tolerance with a greater effect in AT1aWT mice. Fructose increased plasma cholesterol in both groups (P < 0.01) and reduced ANG II in AT1aKO mice. Results document prominent interactions between genetics and diet with data showing that in the absence of angiotensin AT(1a) receptors, a fructose diet decreased blood pressure.  相似文献

In order to identify indicators of the preferences for bedding materials, the paper bedding material preferences of laboratory mice were investigated in the present study. Four cages, each containing a different structure of paper bedding material were connected to allow free access to each cage. The preferences for paper bedding materials of laboratory mice were judged by the differences in the length of stay and sleep in each cage. The mice preferred the bedding material that allowed them to easily hide and build nests and was soft. We conclude that the comfort and well-being of laboratory mice can be increased through the appropriate selection of bedding material.  相似文献

Several confounding factors may influence the outcome of an experiment and the extent of inter-individual variation. The aim of this study was to investigate if cage enrichment induces an effect on experimental mean values and on inter-individual variation in the light/dark paradigm using diazepam as the anxiolytic drug. The behaviour of 216 naive adult male mice of two different strains (BALB/c and C57BL/6) was studied. The animals were housed in groups of four in 'non-enriched', 'enriched' (nesting material) or 'super-enriched' (nest-box, nesting material, wooden gnawing stick and PVC tube) cages. After 5 weeks the animals were assigned to one of three treatments: control (no injection), sham (saline injection i.p.) or diazepam (1 mg/kg bw i.p.) and tested in the light/dark test for 5 min. Variation data were analysed using three different methods (mean absolute deviation, coefficient of variation and power analysis). The C57BL/6 mice scored higher than BALB/c mice in activity related measurements and showed a less 'emotional' behaviour profile in the pharmacological control situation of the light/dark test. In this study the anxiolytic effect of diazepam was clear in BALB/c mice but absent in C57BL/6 mice. Mice housed in enriched and super-enriched cages gained more weight than mice in non-enriched cages, although food intake was not affected. Generally, the strain of mouse had the greatest impact on both mean values and variation. However, there was no consistent increase for one particular strain. The choice of statistical method for analysing variation may influence the interpretation of within-group variability, but none of the methods showed any significant differences between standard and enriched conditions on variability in any of the parameters measured.  相似文献

Aspen wood-wool, provided as nesting material, was evaluated as a possible improvement of cage environment for 10-14-week-old inbred male mice maintained in groups of six (BALB/c n = 72 and C57BL/6J n = 36). The daily behaviour of mice was video recorded and their body weight, food consumption, weights of some organs and serum corticosterone concentrations were measured. Aggressive interactions between cage mates and against a strange intruder as well as the number of wounds on the back of the animals was monitored in order to evaluate the effect of nesting material on intermale aggression. Nesting material did not affect the daily active/passive behaviour patterns of mice, although animals clearly preferred it as a resting place. BALB/c mice given nesting material showed less weight gain and smaller brown adipose tissue weights than animals without nesting material. The other characteristics measured were not affected by the presence of nesting material in either strain. The presence of nesting material had no effect on fighting in cages. C57BL/6J mice were more aggressive than BALB/c mice according to the number of wounded animals in a cage. Wounded BALB/c mice had enlarged spleens and decreased epididymal adipose tissue weights. In conclusion, the nesting material used in this study did not adversely affect the animals. On the other hand, the material was clearly preferred to conventional bedding as a resting place. These findings suggest that nesting material may improve the cage environment of laboratory mice. Furthermore, there was an indication of strain differences in aggressive behaviour. It could be suggested that C57BL/6J mice are less tolerant towards intruders and housing six mice per cage is not suitable for this strain.  相似文献

Bedding influences various parameters in the housing of laboratory mice, such as health, physiology and behaviour (often considered as being integral parts of welfare). Notwithstanding existent studies about bedding preferences of individually tested mice, data about group-housed mice are still lacking. The aim of this study was to find out the structure preference for softwood bedding of group-housed mice. One hundred and eight 8-week-old female mice (C57BL6/JOlaHsd and BALB/cOlaHsd) were housed in groups of three and were given one-week free access to two different bedding structures at a time. In three test combinations, softwood shaving bedding was tested versus softwood chip bedding products of three different particle sizes (fine/medium/coarse-grained). The preference test was performed in a DoubleCage system composed of two Makrolon type IIL cages, connected by a perspex tunnel. This validated system was able to detect the crossings of each individual animal with correct crossing time and direction. On the basis of these data, dwelling times on the particular bedding structures were statistically analysed as a parameter for bedding preferences. In all three test combinations, a highly significant shaving preference was detected. On average, mice spent 70% of their dwelling time on the shavings. This preference was more explicit during the light period and in C57BL/6J mice. The relative ranking of the bedding structures was: shavings > coarse-grained chips > medium chips = fine chips. By means of these results, a shaving structure as bedding can be recommended for laboratory mice, whereas fine chip structures should be avoided.  相似文献

Melanin-concentrating hormone (MCH) plays an important role in energy balance. The current studies were carried out on a new line of mice lacking the rodent MCH receptor (MCHR1(-/-) mice). These mice confirmed the previously reported lean phenotype characterized by increased energy expenditure and modestly increased caloric intake. Because MCH is expressed in the lateral hypothalamic area, which also has an important role in the regulation of the autonomic nervous system, heart rate and blood pressure were measured by a telemetric method to investigate whether the increased energy expenditure in these mice might be due to altered autonomic nervous system activity. Male MCHR1(-/-) mice demonstrated a significantly increased heart rate [24-h period: wild type 495 +/- 4 vs. MCHR1(-/-) 561 +/- 8 beats/min (P < 0.001); dark phase: wild type 506 +/- 8 vs. MCHR1(-/-) 582 +/- 9 beats/min (P < 0.001); light phase: wild type 484 +/- 13 vs. MCHR1(-/-) 539 +/- 9 beats/min (P < 0.005)] with no significant difference in mean arterial pressure [wild type 110 +/- 0.3 vs. MCHR1(-/-) 113 +/- 0.4 mmHg (P > 0.05)]. Locomotor activity and core body temperature were higher in the MCHR1(-/-) mice during the dark phase only and thus temporally dissociated from heart rate differences. On fasting, wild-type animals rapidly downregulated body temperature and heart rate. MCHR1(-/-) mice displayed a distinct delay in the onset of this downregulation. To investigate the mechanism underlying these differences, autonomic blockade experiments were carried out. Administration of the adrenergic antagonist metoprolol completely reversed the tachycardia seen in MCHR1(-/-) mice, suggesting an increased sympathetic tone.  相似文献

Ninety adult males divided in six equal groups were exposed to different photoperiods for 21 days. Exposures included natural light (ca 11 hr), long photoperiod (16L:8D) and short photoperiod (8L:16D). The first three groups received these exposures at room temperature (13-20 degrees C) while the remaining three at raised temperature (36-38 degrees C). Soiled bedding of the above males was introduced in the cages of unisexually housed noncyclic females and their potentiality to induce oestrus was assessed. It was noticed that the bedding of all the males proved to be a stimulus inducing oestrus in the majority of the females during the 7 day exposure. There was no significant difference in the number of females returning to oestrus following exposure to soiled bedding of different males. These results elucidate that environmental factors, especially light and temperature do not influence the production/release of the oestrus-inducing pheromone in wild mice.  相似文献

Genetic variations in the wild-derived inbred mouse strains are more diverse than that of classical laboratory inbred mouse strains, including C57BL/6J (B6). The sleep/wake and monoamine properties of six wild-derived inbred mouse strains (PGN2, NJL, BLG2, KJR, MSM, HMI) were characterized and compared with those of B6 mice. All examined mice were nocturnal and had a polyphasic sleep pattern with a “main sleep period” identified during the light period. However, there were three sleep/wake phenotypic differences between the wild-derived mouse strains and B6 strain. First, the amount of sleep during the dark phase was comparable with that of B6 mice. However, the amount of sleep during the light phase was more varied among strains, in particular, NJL and HMI had significantly less sleep compared with that of B6 mice. Second, PGN2, NJL, BLG2, and KJR mice showed a “highly awake period” (in which the hourly total sleep time was <10%) immediately after the onset of the dark period, which was not seen in B6 mice. Third, relative to that of B6 mice, PGN2 and KJR mice showed longer duration of wakefulness episodes during the 12-h dark phase. Differences in whole brain noradrenaline, dopamine, and 5-hydroxy-tryptamine contents between the wild-derived mouse strains and B6 strain were also found. These identified phenotypes might be potentially under strong genetic control. Hence, wild-derived inbred mice could be useful for identifying the genetic factors underlying the regulation of sleep and wakefulness.  相似文献

Sister chromatid exchange (SCE) values were determined in bone marrow cells isolated from mouse (Mus musculus) femurs after injections of 5-bromo-2'-deoxyuridine (BrdU) and 5-fluorodeoxyuridine (FrdU). Male mice of C3H/J, C57BL/6J, and DBA/2 strains maintained in the laboratory gave mean SCE values of 3.42 +/- 0.07, 3.62 +/- 0.08, and 3.97 +/- 0.13, respectively. Males obtained from natural populations of southwestern Ontario had a higher mean SCE value (6.02 +/- 0.16), as did inbred males maintained in outdoor enclosures for at least 3 weeks (5.07 +/- 0.22). Wild mice housed in the laboratory for 9 months or longer had SCE values similar to laboratory bred mice (3.46 +/- 0.05). The SCE values in wild-caught mice were inversely proportional (r = -0.49) to the distance between the sites where these animals were collected and the nearest major industrial center. Based on these results, SCE analysis in mice is proposed as a possible first-line monitoring procedure for the detection of general changes in environmental genotoxicity.  相似文献

The light-dark box (LDB) and the open-field (OF) tests are widespread experimental models for studying locomotion and anxiety in laboratory rats and mice. The fact that rodents are nocturnal animals and more active at night raises a critical question of whether behavioral experiments carried out in the light phase are methodologically correct. Parameters of behavior of four mouse strains (C57BL/6J, DBA2/J, AKR/J and CBA/LacJ) in the light-dark box and open-field tests in the light and dark phases were compared. No significant influence of the phase of testing on anxiety in LDB and OF tests was revealed. In the OF test CBA mice showed increased locomotor activity, whereas AKR and C57BL/6 mice showed increased defecation in the dark phase. It was concluded that: 1) the phase of testing is not crucial for the expression of anxiety in LDB and OF; 2) the sensitivity to the phase of testing depends on the genotype; 3) the indices of behavior in the genotypes sensitive to the phase of testing (locomotion in the CBA and defecation in the AKR and C57BL/6 mouse strains) are increased in the dark phase.  相似文献
