The aim of the present work was to determine phylogenetic relationships among Astraeus species and to support macroscopic and microscopic characters of Astraeus with analysis of the ITS rDNA region. Collections of Astraeus basidiomes were made from different geographical areas in Thailand and compared with existing collections made worldwide. The marriage of observations on morphological features, including basidiospore ornamentation and molecular data demonstrated the presence of several Astraeus species. Sequences for 41 Astraeus collections were compared and the phylogenetic analyses grouped Thai Astraeus collections into two distinct groups. One contained A. odoratus and an Asian species described herein as A. asiaticus. There are at least two additional species: A. pteridis, and one so far un-named from North America. Our results show that molecular data can be used in combination with traditional morphological characteristics to resolve taxonomic uncertainties in the genus Astraeus.  相似文献

Torymus koreanus (Hymenoptera: Torymidae) was reared from galls of Dryocosmus kuriphilus (Hymenoptera: Cynipidae) for the first time in Japan. We here report morphological features and partial mtDNA sequencing data of T. koreanus. Torymus koreanus has several common characteristics with species of the cyaneus group defined by Zavada (2003 ). According to the key to species groups defined by Graham and Gijswijt (1998 ), it does not belong to any species group because of the entire posterior margin of metasomal tergum five.  相似文献

Seedlings of three Fagaceae species planted on acidic, infertile colluvial soil showed accelerated growth when inoculated with ectomycorrhizal fungi.

Abstract

We conducted a study with seedlings of Fagaceae species inoculated with ectomycorrhizal fungi to estimate their utility for growth in acidic soil conditions. We selected Quercus glauca, Quercus salicina and Castanopsis cuspidata as typical evergreen, broad-leaved, woody species of southwestern Japan. Seedlings were inoculated with Astraeus hygrometricus or Scleroderma citrinum, and planted in acidic, infertile colluvial soil collected from an abandoned site. Six months after planting, seedlings of the three species inoculated with A. hygrometricus were growing well, especially, Q. salicina. The growth of seedlings inoculated with S. citrinum was inferior to seedlings inoculated with A. hygrometricus. In contrast, seedlings without ectomycorrhizal fungi did not grow well. Differences in growth among the three types of seedlings were related to differences in the levels of nutrient acquisition. We concluded that Fagaceae seedlings inoculated with A. hygrometricus were best suited for acidic, infertile environments.  相似文献

The East Asian giant water bug species Appasus japonicus Vuillefroy and Appasus major Esaki are aquatic hemipteran insects whose ranges overlap, particularly in the Japanese Archipelago and on the Korean Peninsula. In rare cases, the two species co‐occur. Furthermore, they are very similar ecologically and also morphologically, making their identification extremely difficult, and the possibility of hybridization has also been suggested. In the present study, we re‐examined their taxonomic validity, and the characteristics useful for identifying them. To re‐examine the morphological traits useful for distinguishing these two species, 222 specimens of A. japonicus collected from Japan, Korea, and China, and 132 specimens of A. major from Japan and Korea, were examined. We also performed molecular phylogenetic analyses based on the mitochondrial DNA 16S rRNA and cytochrome oxidase subunit I (COI) regions and the nuclear DNA Histone 3 region. Although the two species are very similar ecologically and also morphologically, they showed significant genetic differentiation. Thus, there is likely some form of reproductive isolation acting between them. Major morphological characteristics overlap extensively between A. japonicus and A. major, and no particular trait was identified as being effective for differentiating these species. All the morphological characteristics examined overlapped between A. japonicus and A. major. However, a principal component analysis based on all of the morphological characteristics revealed that, despite the overlap between these species, it was possible to morphologically distinguish them. Therefore, a more accurate identification becomes possible using multiple characteristics rather than a single characteristic. The male genital paralobes, evaluated as the most useful morphological characteristic, was effective with 100% probability for the Japanese Appasus species. However, for the Asian (i.e. Korean) specimens, this characteristic was not useful. On the other hand, the results of molecular phylogenetic analyses based on the mitochondrial DNA 16S rRNA and COI regions and the nuclear DNA Histone 3 region clearly showed significant genetic differentiation between the two species. Notably, the results for the mitochondrial COI region strongly supported the independence of each monophyletic group (i.e. validity of each species). Relationships among three Japanese Laetiporus taxa (“L. sulphureus var. sulphureus” auct. jap., L. sulphureus var. miniatus, and L. versisporus) were assessed with phylogenetic analysis and incompatibility tests. Gene phylogenies inferred from the internal transcribed spacer region of nuclear ribosomal DNA, elongation factor 1α, and β-tubulin gene regions suggested that Japanese Laetiporus was divided into four groups: the yellow pore form of L. sulphureus var. miniatus, the white pore form of L. sulphureus var. miniatus, and two “L. sulphureus var. sulphureus”/ L. versisporus groups. A morphologically distinct species, Laetiporus versisporus, sharing a clade with “L. sulphureus var. sulphureus” auct. jap., was proved to be an anamorphic form of “L. sulphureus var. sulphureus” auct. jap. The “sulphureus/versisporus” isolates showed two divergent sequence types in each region. Some isolates had intraindividual polymorphism assigned to both sequence types. This finding suggests that speciation via hybridization is ongoing in the “sulphureus/versisporus” group. Single spore isolates from the “sulphureus/versisporus” group, white pore group, and yellow pore group were incompatible with each other. Our results provided strong support for the new recognition of three Laetiporus taxa in Japan.  相似文献

Interstitial nemerteans of the genus Ototyphlonemertes are difficult to organize into traditional morphospecies. They occur in a multitude of slightly different local varieties that form a seemingly continuous morphological cline. In this paper, we summarize most published morphological data on the group, plus 73 new records of geographic varieties from the Mediterranean Sea in the East to the Sea of Japan in the West. We summarize morphological variation, partition traits into character and character states, propose a standardized protocol for examination of live specimens and discuss the phylogenetic structure of the group. The phylogenetic discussion leads to a hypothesis that partitions all varieties (including the established species) into six groups. These are the smallest morphologically homogenous sets of varieties (corresponding to traditional morphospecies) we can diagnose on phylogenetically reliable traits. Variation within the groups appears to be unreliable phylogenetic markers that may distinguish ecological forms rather than relatedness. However, we distinguish four of the groups by combinations of two traits, one apomorphy for a more inclusive group and one plesiomorphy, and the remaining two by one trait each that may be either a unique plesiomorphy or an apomorphy depending on the rooting, and most of them may thus join paraphyletic groups of cryptic monophyletic units. We call this kind of group phylomorph and name them the Duplex-, Pallida-, Cirrula-, Fila-, Lactea- and Macintoshi-morph (referring to the first established species within each group, i.e. Ototyphlonemertes duplex, O. pallida, O. cirrula, O. fila, O. lactea and O. macintoshi respectively). The phylogenetic scheme provides a simple tool to allocate geographical varieties to a group of possible `species' and a phylogenetic null-hypothesis for further tests with genetic data.  相似文献

Polygonum cuspidatum in Japan, we analyzed the chloroplast DNA sequences of a region from the rbcL to the accD gene (ca. 1,420 bp), and found nucleotide variations at 22 sites in 68 samples. The phylogenetic relationship deduced from the sequence variations revealed the existence of at least five groups. The first group consisted of P. cuspidatum var. cuspidatum in the central part of Honshu; in Nagano, Yamanashi, and Shizuoka. The second, a sister of the first, consisted of those plants in Shizuoka-Itoigawa Line. The third group consisted of plants in the northern part of Japan including P. sachalinense in Hokkaido, P. cuspidatum var. cuspidatum in Aomori and var. uzensis in Akita. The fourth consisted of var. uzensis in the Tohoku District. The fifth consisted of var. terminalis in the Izu Islands. P. cuspidatum are differentiated according to their distribution, and two varieties, var. terminalis and var. uzensis, are differentiated genetically. Polygonum sachalinensis, a distinct species morphologically, fell into the accessions of P. cuspidatum on the phylogenetic tree obtained in the present study. The name Aconitum japonicum Thunb. subsp. japonicum (syn. A. japonicum var. montanum Nakai; Japanese name, Yama‐torikabuto) is well known to botanists and horticulturists as a representative of Japanese monkshoods and is occasionally found even in the suburbs of Tokyo. However, this aconite is restricted to the mountains on the Pacific Ocean side of Honshu Island, central Japan. Although the geographical range of A. japonicum is rather narrow, it shows a wide range of morphological variation as a member of a tetraploid species group (2n = 4x = 32).  相似文献

Laboulbenia coneglanensis isolated from Japanese harpaline carabids is described and photographed. It is compared morphologically withL. flagellata from Japan and Europe, which is similar in appearance. The most important difference between the two species is in antheridial character and host range. Other characters such as coloration, perithecial shape, especially the shape of the perithecial apex, and size of ascospores are also useful for distinction.Laboulbenia ophoni var.dilatata described by Maire (1920) is regarded as a synonym ofL. coneglanensis.  相似文献

A setiform dematiaceous hyphomycete,Kionochaeta spissa was newly collected and isolated from evergreen broad-leaved forests in the southern parts of Japan. Except for its dematiaceous nature, the species is morphologically similar to a nectriaceous hyphomycete,Chaetopsina fulva. The morphology and cultural properties of the Japanese isolates ofK. spissa were described, and the phylogenetic relationships betweenChaetopsina (C. fulva (type species)) andKionochaeta (K. ramifera (type species),K. spissa andK. ivoriensis) were inferred based on nuclear encoded small subunit (18S) rDNA sequences using the neighbor-joining method.Chaetopsina andKionochaeta were found to be phylogenetically related to the Hypocreales and Sordariales, respectively. Both should be maintained as separate genus for phylogenetic classification. The morphological resemblance especially betweenC. fulva andK. spissa is an example of the convergent evolution.  相似文献

The effects of temperature, ionic composition, and conductivity on growth rates of ten strains of Chaetoceros muelleri Lemmerman (mostly var. subsalsum Johan. & Rushf.) were studied. Lipid content of stressed and unstressed cells and fatty acid composition were also determined. Considerable physiological variability was observed in the ten strains, although principal components analysis of physiological data indicated that all strains fell into one of two major groups: C. muelleri (var. muelleri and var. subsalsum) and an undescribed Chaetoceros species morphologically close to C. muelleri var. subsalsum. A high degree of agreement was found among morphological, physiological, and biochemical data sets, indicating that physiological and biochemical data may be helpful in making taxonomic decisions in diatoms, particularly in taxa with few morphological characters. We also conclude that nonmorphological characters such as those employed in the present study can be used to test phylogenetic hypotheses formulated from traditional morphological data.  相似文献

The phylogenetic relationships among the Japanese members of the genus Eubrianax (Coleoptera: Psephenidae) were examined using the mitochondrial cytochrome oxidase subunit I (COI) gene and nuclear 28S rRNA gene sequences. Based on the molecular phylogeny as well as morphological features, the species status of Eubrianax brunneicornis Nakane, 1952 was proposed. The phylogenetic analyses recovered monophyly of the previously proposed pellucidus species group with four Japanese species, whereas a single Japanese species of the granicollis group was included in the lineage of the ramicornis group with five Japanese species. The divergence times of the species were estimated by dating the phylogenetic tree against the fossil record and a molecular clock based on the COI gene. The divergence of the Japanese species was inferred to have occurred during the Pliocene epoch.  相似文献

We investigated nine strains of the Micrasterias crux‐melitensis (Ehrenb.) Hassall ex Ralfs and M. radians W. B. Turner species complex. A combination of molecular, morphological, and geometric morphometric data was used to reveal the patterns of their phenotypic and phylogenetic differentiation. The molecular data based on internal transcribed spacer (ITS) rDNA, glycine transfer RNA (trnGuuc) intron, and SSU rDNA sequences revealed three phylogenetic lineages. One of them comprised the six European and North American strains that were morphologically identified as M. crux‐melitensis. Phenotypic data illustrated high morphological variability of strains within this genetically homogenous lineage that spanned several traditional infraspecific taxa, including strains corresponding to M. crux‐melitensis var. janeira (Racib.) Grönblad and M. crux‐melitensis var. superflua W. B. Turner, whose morphometric characteristics profoundly differed. Three strains of M. radians formed two separate phylogenetic lineages corresponding to traditional varieties M. radians var. evoluta (W. B. Turner) Willi Krieg. and M. radians var. bogoriensis (C. J. Bernard) G. S. West. The morphological types corresponding to the former variety have, so far, only been reported from Africa. Therefore, we cannot preclude that geographic isolation may play a role in species differentiation of relatively large freshwater protists, such as Micrasterias.  相似文献

Molecular variations of Spiranthes sinensis Ames var. australis (R.Br.) H. Hara et Kitam. ex Kitam. in Japan were examined to evaluate the validity of the seasonally differentiated groups and a dwarf form of the species, which is endemic to Yakushima Island, Japan. Sequence differences in the plastid trnL-F locus clearly distinguished Japanese S. sinensis var. australis from S. sinensis var. sinensis collected from Ryukyu. In contrast, the trnL-F sequence of S. sinensis var. australis from Sabah, Malaysia, clearly differed from that of Japanese S. sinensis var. australis, suggesting genetic heterogeneity of Spiranthes sinensis var. australis in Asia. Moreover, a molecular analysis based on the sequences of nuclear ITS1 regions indicated that there are two major groups of S. sinensis var. australis in Japan, with a geographic distribution boundary on Kyushu Island. However, the trnL-F and ITS1 sequences did not support the genetic differentiation of the seasonally differentiated groups or the dwarf form from the other Japanese individuals. Based on these molecular data, the systematic treatment of physiological and morphological variations in the Japanese population of S. sinensis. var. australis is discussed.  相似文献

Phylogenetic analyses were conducted using complete nucleotide sequences of the D-loop in the mitochondrial genome of Mola specimens, collected mainly in Japanese waters, to clarify the genetic features and distribution patterns of Mola sunfishes. Two significantly distinct groups (designated A and B) were present in the genus, with a considerable net nucleotide sequence divergence between the two (8.4%). The two groups occurred sympatrically around the Japanese coast, as previously suggested by Sagara et al. (2005). Group A occurred mostly on the Pacific coast of eastern Japan, while group B was widely distributed along the Kuroshio Current, strongly suggesting different migration routes for each group. The morphological characteristics of the two group specimens were differentiated via the head bump, body proportions and shape of the clavus. Through the addition of Mola sequence data taken from outside Japan to our phylogenetic analyses, three independent groups, including groups A and B, were found, each with a wide geographical distribution, which suggests the presence of at least three independent species within the genus Mola.  相似文献

The morphological and molecular differentiation of the Micrasterias truncata (Corda) ex Bréb species complex was investigated. In total, 17 strains traditionally assigned to M. truncata were isolated from different European localities (Czech Republic, southwest France, Ireland), and obtained from public culture collections. In addition, strains of the morphologically similar species, M. decemdentata (Nägeli) W. Archer and M. zeylanica F. E. Fritsch, were also included. Molecular phylogenetic analysis based on trnGucc intron sequences revealed five well supported clades. Two Australian strains assigned to M. truncata var. pusilla G. S. West formed a lineage sister to M. zeylanica. This was evident from a concatenated phylogeny based on small subunit rDNA and trnGucc intron sequences. The isolated position of these strains was also illustrated by parallel landmark‐based geometric morphometric analysis of cell shapes. The strains NIES 783 and NIES 784 probably represent a separate species. Particular analysis, including additional strains, is needed to resolve the relationship inside this lineage. The second phylogenetic lineage, containing two strains of M. truncata var. semiradiata (Kützing) Wolle, was also different from other strains on the basis of morphometric data. We suggest recognizing this variety as a separate species, Micrasterias semiradiata L.A. Brébisson ex F. T. Kützing. The remaining three clades formed a firmly supported group of the ‘core’M. truncata recognized by both molecular markers. However, neither any morphological, morphometric, nor geographical pattern was detected among members of these three clades. This pattern could be caused by a relatively recent origin of these lineages that may represent a sympatric, truly cryptic species. Strains attributable to traditional morphologically defined variety M. truncata var. neodamensis were nested within the ‘core’M. truncata.  相似文献

Varanid lizards, which vary considerably in body mass both interspecifically and intraspecifically, are generally considered to be morphologically similar. However, significant and non-isometric variation in the relative appendage dimensions for 17 species of Western Australian goannas suggest that these lizards are not morphologically conservative. The first and second canonical variates clearly distinguish the two subgeneral Odatria and Varanus, and species are generally sexually dimorphic. The morphological variation observed among these 17 species of goanna is associated with foraging mode and ecology. However, no single or small group of morphological dimensions discriminates phylogenetic groups, sexes, or ecological groups, and body size is an important component in these analyses. J. A putative new limpet species (Patellogastropoda) from the Sea of Japan is revealed by molecular genetic analyses using the mitochondrial markers 16S rRNA and cytochrome c oxidase subunit I (CO1), as well as the DNA marker 18SrRNA. Our data indicate that the limpet, collected in the Peter the Great Bay (Russian Federation), is not, as its morphology suggests, the Japanese species Lottia kogamogaiSasaki and Okutani, 1994, and might also hint towards another putative species complex in the Sea of Japan. The different currents between the Far East Asian mainland (cold, subpolar jet running southwards) and the Japanese archipelago (warm, subtropical jet running northwards) are likely to act as a barrier that has a substantial influence on species distribution in these waters. Accordingly, our results indicate that it is about time for a revision of patellogastropod species with a reported distribution in Japanese and Far Eastern Russian waters by an integrative approach using molecular genetic and morphological characters. The species investigated herein is referred to as Lottia cf. kogamogai until it is morphologically re‐examined and compared with primary type specimens of known species.  相似文献
