Biomachining has become a promising alternative to micromachining metal pieces, as it is considered more environmentally friendly than their physical and chemical machining counterparts. In this research work, two strategies that contribute to the development of this innovative technology and could promote its industrial implementation were investigated: preservation of biomachining microorganisms (Acidithiobacillus ferrooxidans) for their further use, and making valuable use of the liquid residue obtained following the biomachining process. Regarding the preservation method, freeze‐drying, freezing, and drying were tested to preserve biomachining bacteria, and the effect of different cryoprotectants, storage times, and temperatures was studied. Freezing at –80°C in Eppendorf cryovials using betaine as a cryoprotective agent reported the highest bacteria survival rate (40% of cell recovery) among the studied processes. The treatment of the liquid residue in two successive stages led to the precipitation of most of the total dissolved iron and divalent copper (99.9%). The by‐products obtained (iron and copper hydroxide) could be reused in several industrial applications, thereby enhancing the environmentally friendly nature of the biomachining process.  相似文献

土壤重金属污染问题日益严重,具有普遍性、隐蔽性、表聚性、不可逆性等特点,已经成为环境污染治理中的热点、难点问题。解磷微生物能够依靠自身的代谢产物或通过与其他生物的协同作用,将土壤中的难溶性磷转化为可供植物吸收利用的磷,具有多重植物促生长功能和重金属解毒能力,可在重金属毒害水平下,促进植物生长、提高植物抗病能力、克服重金属对植物生长的不利影响,从而增强重金属修复植物的生存竞争力。从解磷微生物的研究现状入手,介绍了解磷微生物对土壤重金属污染的修复能力,综述了解磷微生物对土壤重金属污染修复的作用机制,分析了目前解磷微生物在重金属修复过程中存在的问题,并提出了今后研究的方向,为重金属污染土壤的修复提供了新思路。  相似文献

通过对重金属胁迫下土壤微生物和微生物过程研究的进程和研究进展的归纳综述,分析了该研究尚存在的问题,并阐述了其可能原因.认为土壤微生物和微生物学过程的重金属胁迫研究存在如下问题:一是从实验室、田间试验和实地监测得到的结果间无法进行比较,从而使实验室和田间试验的研究丧失了其科学指导意义,并且在实地监测研究中缺乏相应的“精确”对照;二是在重金属的胁迫下土壤微生物不但数量有消长,而且区系结构上也发生了变化,但是用于校园微生物区系结构变化的手段(PLFA、BI-OLOG和DNA等方法)尚处在探索阶段并需要昂贵的设备,难以普及,需发展一些可广泛普及的新方法来代替传统的平板分离法分析土壤微生物结构;三是重金属对土壤微生物和微生物过程产生胁迫的形态、离子效应和根际效应尚未得到有效的研究和探讨;四是土壤微生物和微生物过程重金属胁迫的表征体系尚未建立.  相似文献

微生物在地下水和土壤环境中的迁移与地下水资源保护、地下水污染修复及土壤污染防治等息息相关。自然界中多孔介质具有结构复杂性和空间异质性。这导致微生物在其中的迁移易受多重环境因素的影响。本文总结了几种典型多孔介质中微生物迁移模型、理论与研究方法,并对多孔介质中影响微生物迁移行为的3种因素——物理、化学和生物因素进行了梳理。其中物理因素的影响主要包括多孔介质的粒径、表面粗糙度、饱和度、环境温度、水体流速等相关;化学因素主要包含流体pH、离子种类与强度、可溶性有机物含量、多孔介质自身化学性质等;生物因素不但涉及微生物种类、细胞大小和细胞表面特性,还与胞外聚合物的分泌、鞭毛运动及趋化性等相关。本综述旨在总结近年来有关微生物在多孔介质中迁移的相关研究,深入理解微生物在多孔介质中的迁移行为,为其在地下水和土壤污染修复中的实际应用提供理论依据。  相似文献

生物脱氮是污水处理厂的核心,掌握生物脱氮过程相关微生物代谢特性,对于探索微生物资源和提高污水处理厂脱氮性能具有重要意义。近年来,分子生物学方法不断发展和改进,已被广泛应用于揭示脱氮微生物群落多样性、组成结构和潜在功能等方面,大幅提升了研究者们对污水生物脱氮系统中微生物,尤其是不可培养微生物的代谢机理、抑制调控原理及新型生物脱氮工艺途径的认识。本文对流行的分子生物学方法(16S rRNA基因测序、实时荧光定量PCR技术、宏基因组学、宏转录组学、宏蛋白质组学和代谢组学)进行了介绍,综述了其在硝化细菌、反硝化细菌、完全氨氧化细菌、厌氧氨氧化细菌、厌氧铁氨氧化细菌、硫酸盐型厌氧氨氧化细菌及亚硝酸盐/硝酸盐型厌氧甲烷氧化微生物等方面的研究进展,阐明了这些氮素转化微生物在氮循环过程的代谢途径和酶促反应,并从标准测定方法构建、不同方法的联用及跨学科结合和检测方法的简易化这3个方面展望了分子生物学方法的技术突破及其在污水生物处理系统中的应用前景。本综述从系统角度全面认识脱氮微生物群落及其结构,为未来污水处理生物脱氮微生物的研究提供了新方向。  相似文献

With the depletion of global petroleum and its increasing price, biodiesel has been becoming one of the most promising biofuels for global fuels market. Researchers exploit oleaginous microorganisms for biodiesel production due to their short life cycle, less labor required, less affection by venue, and easier to scale up. Many oleaginous microorganisms can accumulate lipids, especially triacylglycerols (TAGs), which are the main materials for biodiesel production. This review is covering the related researches on different oleaginous microorganisms, such as yeast, mold, bacteria and microalgae, which might become the potential oil feedstocks for biodiesel production in the future, showing that biodiesel from oleaginous microorganisms has a great prospect in the development of biomass energy. Microbial oils biosynthesis process includes fatty acid synthesis approach and TAG synthesis approach. In addition, the strategies to increase lipids accumulation via metabolic engineering technology, involving the enhancement of fatty acid synthesis approach, the enhancement of TAG synthesis approach, the regulation of related TAG biosynthesis bypass approaches, the blocking of competing pathways and the multi-gene approach, are discussed in detail. It is suggested that DGAT and ME are the most promising targets for gene transformation, and reducing PEPC activity is observed to be beneficial for lipid production.  相似文献

微生物的代谢活动易受到环境变化的影响,当环境中存在重金属污染时微生物会通过调节代谢降低自身所受的重金属的毒害。本文通过微生物代谢组学研究探讨重金属胁迫下微生物代谢活动的响应情况,介绍了微生物代谢组学的相关技术和方法,对其应用进行说明;基于重金属对微生物细胞的毒害作用,对重金属胁迫下微生物代谢组学的相关内容进行综述,发现在重金属胁迫下,微生物可以通过增加代谢活动进而产生更多的代谢物质来响应重金属的胁迫,其中微生物产生的胞外聚合物、草酸和柠檬酸等代谢物在微生物响应重金属胁迫中具有重要作用。微生物通过产生相应代谢物不仅使自身可以在重金属胁迫下生存,这些代谢物还可以使环境中重金属有所减少,这对于利用微生物资源修复重金属污染具有重要意义。  相似文献

In Northern Ireland, phosphorus enrichment of lakes due to agriculture is a significant problem. Heavy metal exports from landfill sites, often located on water-logged land, are also of concern. Locally available laterite, a low grade bauxite which is rich in iron and aluminium, is used in acid solution with subsequent precipitation to remove phosphorus and heavy metals at several sewage treatment works. Constructed wetlands offer an attractive alternative to conventional waste water treatment in certain circumstances but removal of phosphorus is strongly dependent on the bed medium. Calcium-, iron- and aluminium-rich solid media are recommended. A brief introduction to the use and cost-effectiveness of constructed wetlands (CWs) in treating a range of effluents is given. This study, using both laboratory tests and pilot-scale constructed wetlands, reports the effectiveness of granular laterite in removing phosphorus and heavy metals from landfill leachate. Initial laboratory studies have shown that laterite is capable of 99% removal of phosphorus from solution. A pilot-scale experimental CW containing laterite achieved 96% removal of phosphorus. This removal is much greater than that reported in other systems. Initial removals of aluminium and iron by pilot-scale CWs have been up to 85% and 98% respectively. Percolating columns of laterite reduced Cd, Cr and Pb to undetectable concentrations. Possible application of this low cost, low technology, visually unobtrusive yet efficient system to rural areas with dispersed point sources of pollution is discussed.  相似文献

Metal-binding receptors are one of the extracellular components of ATP-binding cassette transporters that are essential for regulation of metal homeostasis in bacteria. Laminin-binding adhesin (Lmb) of Streptococcus agalactiae falls under this class of solute binding proteins. It binds to zinc with a high affinity. Crystal structure of Lmb solved previously by our group reveals that the zinc is tetrahedrally coordinated by three histidines and a glutamate at the interdomain cleft. Lmb contains a long disordered loop close to the metal-binding site whose precise function is unknown. Several experimental attempts to produce apo-Lmb failed and this prompted us to carry out in silico studies to analyse the structural importance of the metal in Lmb. Here, we present the results of the molecular dynamics (MD) simulation studies of native, apo-(metal removed) and the long loop truncated Lmb models along with a homologous protein, TroA from Treponema pallidum that was taken up for validating the MD results of Lmb. Absence of a metal results in significant structural changes in Lmb, particularly at the metal-binding pocket and with the long loop, although the overall fold is retained. This study thus revealed that the Lmb can exist in different conformational states with subtle differences in the overall fold based on the presence or absence of the metal. This could be functionally important for a putative metal uptake and release and also for the adhesive function of Lmb in recognizing laminin, which contains a high number of zinc finger motifs.  相似文献

This presentation comprises a review on the use of immobilized algae for wastewater nitrogen, phosphorus and metal removal purposes. Details of the use of immobilized algae, the techniques of immobilization and the effects of immobilization on cell function are included. Particularly relevant in their use for heavy metal removal from wastewaters; upon enriching the biomass in metal, can be recoverd, thereby providing economic advantages. The use of immobilized microalgae in these processes is very adequate and offers significant advantages in bioreactors. The future of this area of algal cell biotechnology is considered.  相似文献

The database BiolumBase was designed for the collection and systematization of available information on microorganisms containing bioluminescent systems; it includes two sections: natural and transgenic luminous microorganisms. By now, logic schemes of these sections have been developed, classification of the objects has been performed, ways of presentation of characteristics and structure of fields for input of information have been elaborated, and the necessary program modules have been developed. The database is filled on the basis of published data and our own experimental results; subsequent linkage of the database to the Internet is envisaged. Users will be able to obtain not only catalogues of strains but also information concerning the properties and functions of the known species of luminous bacteria, the structure, regulatory mechanisms, and application of bioluminescent systems and genetically engineered constructions with lux genes, as well as to find references and to search strains by using any set of attributes. Biodiversity and potentials of marine-derived microorganisms   总被引:14,自引:0,他引:14  
The marine environment is a prolific resource for the isolation of less exploited microorganisms. As a matter of fact, in the sea, untapped habitats exist with unique characteristics. In addition, the potential contribution of marine sources to the discovery of new bioactive molecules was recently recognized. Biosearch Italia possesses a collection of about 40 000 microorganisms, isolated from different ecological niches. In the search of new bioactive entities, investigations were expanded to marine habitats including marine sediments and organisms. More then 800 microorganisms have been isolated. About half belong to fungal genera, the others being actinomycetes. The frequency of antibiotic activities produced by these marine strains has been determined. Initial data are encouraging: marine isolates produce antibiotic activities with frequencies comparable to terrestrial ones. These activities probably represent a mixture of novel metabolites and known products previously discovered from terrestrial isolates. Further investigations are ongoing to assess the novelty of these observed microbiological activities.  相似文献

红树林土壤生境的独特性决定了其中微生物的多样性及其资源的珍稀性,对于红树林土壤微生物的研究正在成为热点。然而由于传统研究方法等因素的限制,至今人们对红树林土壤微生物的系统了解仍较为有限。近年来,基于16S rRNA,18S rRNA基因的各种分子微生物学技术的迅速发展,红树林土壤微生物的研究亦面临着崭新的局面。文中主要从红树林土壤微生物物种的多样性、生理生化类型的多样性及其在治理污染环境、生物修复作用中的可能性、有效性等方面阐述了红树林土壤微生物的研究进展,并以更合理、有效地开发利用红树林土壤微生物资源为目标,展望了21世纪,以新理念、新技术、新方法进行红树林土壤微生物研究及资源开发的巨大前景。  相似文献

白藜芦醇是植物源的多酚化合物,具有清除自由基、抗氧化、延长寿命等生理活性,在医药、保健品、化妆品等方面有着广阔的应用前景。目前,白藜芦醇主要采用植物提取的方法,对自然植物资源依赖严重,而且受植物成分含量和提取效率低的限制。近年来,合成生物学的迅速发展已使人们将注意力转向利用微生物合成白藜芦醇。通过在微生物中转入外源基因,成功构建出了白藜芦醇工程菌株,并从基因序列、组合方式及发酵工艺等方面进行了优化改造。本文综述了微生物合成白藜芦醇的研究进展。  相似文献

城市绿地微生物的稳定性是城市绿地发挥其重要生态系统功能的重要因素。人类世时代的快速城市化会改变城市绿地土壤理化性质、输入新兴污染物、加剧微生物生态系统潜在风险、改变微生物群落多样性及生态系统功能多样性,深远地影响了城市绿地的生态系统服务功能。本文综述了城市绿地微生物的特征,以及城市化进程对城市绿地微生物组(包括土壤、植物叶际和空气)、抗生素抗性基因、病原微生物和稀有物种群落的影响。与自然微生物相比,城市绿地微生物普遍具有较高的异质性,受人类活动影响大。同时,抗生素抗性基因水平以及人类致病菌数量则显著增加,体现了城市化对城市绿地生态系统健康和功能的扰动。今后研究中应更加关注城市化对于城市绿地微生物的影响,为其对人群健康影响风险评价提供可靠理论支持。  相似文献

作为环境中广泛存在的污染物,微塑料(microplastics)的相关研究备受关注。基于已有研究,本文综合分析了微塑料与土壤微生物(soil microorganisms)的互作关系,微塑料会通过直接或间接的方式影响微生物群落结构与多样性,影响的程度取决于微塑料的类型、剂量和形状。土壤微生物会通过形成表面生物膜和群落选择效应来适应微塑料这一外来物所引起的变化。本文还特别关注了微塑料的生物降解机理,同时探究了影响这一过程的因素,微生物首先会定殖在微塑料表面,分泌多种胞外酶在特定位点发挥作用,将聚合物转化成低聚物或单体,解聚的小分子进入胞内进一步分解代谢,而影响这一降解过程的因素除了分子量、密度、结晶度等微塑料自身理化性质,还包括一些生物因素和非生物因素对相关微生物生长代谢和酶活性的作用。未来研究应注重与实际环境的联系,在深入探究微塑料生物降解研究的同时,开发解决微塑料污染问题的新技术。  相似文献
