以文冠果油为原料,采用皂化法提取文冠果植物甾醇.通过均匀试验设计研究了碱液用量、提取温度、提取时间等因素对文冠果油中植物甾醇纯度和提取率影响,初步确定了皂化法提取工艺条件,并采用抑菌圈法分析了甾醇粗提物对大肠杆菌、枯草芽孢杆菌、毛霉和青霉的抑菌特性.结果表明,文冠果植物甾醇提取的较佳工艺参数为:浓度1 moL/L的氢氧化钠碱液与油用量比为14:1(mL/g),提取温度75℃,提取时间40 min.文冠果甾醇粗提物对毛霉和青霉无抑制作用,对大肠杆菌和枯草芽孢杆菌有明显抑制作用,且对大肠杆菌的抑制作用尤为突出,文冠果植物甾醇是一种较好的抗菌剂.  相似文献

本文以花椒、高良姜等11种食药同源植物提取物为供试材料,分别研究了其对扩展青霉、灰葡萄孢、链格孢和大肠杆菌、金黄色葡萄球菌等5种常见果蔬腐败菌的抑制作用。结果表明,花椒对三种霉菌抑制作用最强,高良姜和莱菔子对扩展青霉和链格孢有一定抑制作用。花椒对三种霉菌的最小抑菌浓度分别为25. 00、25. 00、50. 00 mg/m L,最小杀菌浓度分别为50. 00、100. 00、100. 00 mg/m L。花椒对三种霉菌的孢子萌发有显著抑制作用。花椒提取液处理樱桃番茄可显著降低果实发病率,延长果实保鲜期。11种食药同源植物提取液对大肠杆菌抑菌效果较差。高良姜和天麻对金黄色葡萄球菌抑菌效果突出,可显著抑制金黄色葡萄球菌生长,二者对金黄色葡萄球菌的MIC分别为7. 50和30. 00 mg/m L,MBC分别为15. 00和60. 00 mg/m L。  相似文献

利用硅胶、Sephadex LH-20、HPLC等各种色谱分离技术,从华泽兰根的乙醇提取物中分离得到9个化合物,通过理化性质和NMR波谱数据对它们进行了结构鉴定,分别为:达玛二烯醇乙酸酯(1)、(2R,3S)-5-乙酰基-6-羟基-2-异丙烯基-3-乙氧基苯并二氢呋喃(2)、豆甾醇(3)、邻苯二甲酸二丁酯(4)、12,13-dihydroxyeuparin(5)、5-乙酰-6-羟基-2-异丙烯基苯并呋喃(6)、2,5-二乙酰基-6-羟基苯并呋喃(7)、ruscodibenzofuran(8)、2-乙酰-5-(1-炔丙基)-噻吩-3-O-β-D-吡喃葡萄糖苷(9)。化合物1、2、4和8为首次从该种植物中分离得到,其中2、4和8为首次从该属植物中分离得到。采用牛津杯法评价部分化合物对5种病原菌的抑菌活性,并用肉汤稀释法测定各自的MIC值,结果显示,测试化合物对5种菌具有不同程度的抑菌活性,其中,1、6和8对肺炎克雷伯菌有较强抑菌活性,MIC值分别为0.98、0.98μg/m L和0.49μg/m L,化合物7对大肠埃希菌显示较强抑菌活性(MIC≤3.91μg/m L),化合物1、2、6和7对铜绿假单胞菌有较强抑菌活性,MIC值均为7.81μg/m L,可见化合物1及苯并呋喃类化合物2、6、7、8为华泽兰根主要抑菌活性成分。  相似文献

对实验室现有3种真菌产纤维素酶能力的分析及培养条件优化。比较了3种菌在刚果红培养基上的透明圈大小、并分析产纤维素酶酶活;通过单因素与响应面分析的方法优化毛酶产纤维素酶的培养条件。通过试验得出3种真菌均能产纤维素酶,毛霉能产较多的纤维素酶。毛霉产纤维素酶的最佳条件为:pH 5.0,转速220 r/min,发酵时间47 h,发酵温度35℃,纤维素酶活为6.99U/mL。毛霉、青霉、曲霉均产纤维素酶,毛霉能降解玉米芯纤维素。  相似文献

木榄内生真菌菌株ZD6及其代谢产物的抑菌活性   总被引:1,自引:0,他引:1  
根据形态学特征和ITS序列分析结果,将一株从木榄茎中分离到的内生真菌菌株ZD6鉴定为桔青霉Penicillium citrinum Thom.,该菌株在优化后的发酵培养基(1%麦芽糖,2%甘露醇,1%谷氨酸钠,0.5%蛋白胨,0.15%酵母膏,200g/L土豆汁,pH6.5)中28℃、160r/min振荡培养7d后的发酵液具有明显的抑菌活性。利用柱层析及重结晶等方法从该发酵液的乙酸乙酯和正丁醇提取物中分离得到4个化合物:大黄素、环(丙氨酸-甘氨酸)、赤藓醇和甘露醇,其中的大黄素和赤藓醇为首次在桔青霉中发现,它们对枯草芽孢杆菌Bacillus subtilis的生长具有明显的抑制作用,最低抑菌浓度(MIC)分别为25μg/mL和50μg/mL;大黄素对绿脓杆菌Pseudomonas aeruginosa的生长也具有一定的抑制作用,MIC为100μg/mL。  相似文献

为研究蓼科植物水蓼、火炭母体外抑菌效果,制备水煎煮液,测定其对枯草芽胞杆菌、大肠埃希菌、金黄色葡萄球菌、黑曲霉、青霉、酿酒酵母的抑菌圈直径、最小抑菌浓度及灭菌前后水煎煮液抑菌效果的变化。结果表明:当水蓼液浓度大于0.6 g/mL,火炭母液浓度大于0.2 g/mL时,2者对细菌的抑菌效果均依次为:金黄色葡萄球菌〉大肠埃希菌〉枯草芽胞杆菌。2种水煎煮液对3种真菌无抑制作用。火炭母液对3种细菌的最小抑菌浓度分别为1/8、1/16和1/64 g/mL,都比水蓼液的结果1/4、1/8和1/16 g/mL低。药液经121℃高压蒸汽灭菌处理后,水蓼液对3种细菌的抑制作用有所增强,火炭母液则对金黄色葡萄球菌的抑制作用有所增强。2种水煎煮液对3种细菌均有不同程度的抑制作用,可考虑以其为原料进行植物源抗菌剂的开发,拓宽其应用领域。  相似文献

以稀土(Re~(3+))和落叶松单宁(LT)为原料,采用液相合成法合成了5种廉价的稀土-落叶松单宁(Re~(3+)-LT)配合物,并通过红外光谱、X射线光电子能谱、紫外光谱以及配位数测定确定了配合物的结构.采用牛津杯法、琼脂稀释法测定配合物对黑曲霉、红曲霉、白腐菌、毛霉4种真菌的抑制作用.在抑菌方面,5种配合物对上述4种真菌均具有较强的抑制作用,其抑菌活性大小顺序为Ce~(3+)-LTGd~(3+)-LTLa~(3+)-LTNd~(3+)-LTYb~(3+)-LT,其中Ce~(3+)-LT对4种真菌的最小抑菌浓度分别为:1.6、1.6、0.8和1.6 g·L~(-1);Yb~(3+)-LT对4种真菌的最小抑菌浓度分别为:3.2、1.6、3.2和3.2 g·L~(-1).在杀菌方面,Yb~(3+)-LT的杀菌活性最强,其对4种真菌的最小杀菌浓度分别为:6.4、3.2、3.2和6.4 g·L~(-1).此外,尽管Nd~(3+)-LT和Gd~(3+)-LT具有较强的抑菌活性,但对黑曲霉和毛霉的杀菌作用较弱.  相似文献

本文研究了迷迭香中天然防腐剂的提取方法和抑菌作用,结果表明,用食用乙醇提取迷迭香中防腐物质的最佳提取工艺参数为:固液比1:15、提取温度80℃、提取时间为15 h。迷迭香醇提取物对实验用常见食品污染菌有较强的抑制作用,对金黄色葡萄球菌和枯草芽孢杆菌的最低抑菌质量浓度(MIC)为6.25 g.L-1,对大肠杆菌和汉逊氏酵母菌为12.5 g.L-1,对青霉和黑曲霉为25 g.L-1,对金黄色葡萄球菌、大肠杆菌和枯草芽孢杆菌的抑菌活性pH范围均为4~7,对汉逊氏酵母菌、青霉和黑曲霉为4~6。  相似文献

目的研究泊沙康唑对临床来源的接合菌体外抗菌活性。方法对临床来源的43株接合菌采用ITS区序列分析进行准确鉴定,采用E-test纸片法研究泊沙康唑对43株菌的体外药物敏感性,观察其MIC值。结果经ITS序列分析鉴定,43株临床来源接合菌中最为常见的是小孢根霉和不规则毛霉(各12株),其次为米根霉(10株),ramosa(3株)。其他菌种分别为ornata、总状共头霉、微小根毛霉、卷曲毛霉、印度毛霉和冻土毛霉各1株。E-test纸片法测得泊沙康唑对毛霉属、根霉属和Lichtheimia spp.的MIC值范围分别为0.19~>32μg/mL、0.19~3μg/mL、0.002~0.38μg/mL,对总状共头霉和微小根毛霉的MIC值分别为1μg/mL和0.19μg/mL。结论泊沙康唑对大部分接合菌有效,不同种属MIC值范围不同,毛霉属真菌的MIC值差异较大。  相似文献

The chitinolytic activity of nine species of filamentous fungi, classified with seven genera (specifically, Aspergillus, Penicillium, Trichoderma, Paecilomyces, Sporotrichum, Beaueria, and Mucor), was studied. When cultured in liquid medium containing 1% crystalline chitin, all fungi produced extracellular chitosans with activity varying from 0.2 U/mg protein (Sporotrichum olivaceum, Mucor sp., etc.) to 4.0-4.2 U/mg protein (Trichoderma lignorum, Aspergillus niger).  相似文献

The aim of this study was to determine the in vitro activity of propolis extract against 67 yeasts isolated from onychomycosis in patients attending at the Teaching and Research Laboratory of Clinical Analysis of the State University of Maringá. The method used was an adaptation made from the protocol approved by the National Committee for Clinical Laboratory Standards. The yeasts tested were: Candida parapsilosis 35%, C. tropicalis 23%, C. albicans 13%, and other species 29%. The propolis extract showed excellent performance regarding its antifungal activity: the concentration capable of inhibiting the all of the yeasts was 5 x 10(-2) mg/ml of flavonoids and 2 x 10(-2) mg/ml of flavonoids stimulated their cellular death. Trichosporon sp. were the most sensitive species, showing MIC50 and MIC90 of 1.25 x 10(-2) mg/ml of flavonoids, and C. tropicalis was the most resistant, with CFM50 of 5 x 10(-2) mg/ml of flavonoids and MFC90 of 10 x 10(-2) mg/ml. In view of the fact that propolis is a natural, low cost, nontoxic product with proven antifungal activity, it should be considered as another option in the onychomycosis treatment.  相似文献

Heavy metal analysis of agricultural field soil receiving long-term (>20 years) application of municipal and industrial wastewater showed two- to five-fold accumulation of certain heavy metals as compared to untreated soil. Metal-resistant fungi isolated from wastewater-treated soil belonged to genera Aspergillus, Penicillium, Alternaria, Geotrichum, Fusarium, Rhizopus, Monilia and Trichoderma. Minimum inhibitory concentrations (MIC) for Cd, Ni, Cr, Cu, and Co were determined. The MIC ranged from 0.2 to 5 mg ml(-1) for Cd, followed by Ni (0.1-4 mg ml(-1)), Cr (0.3-7 mg ml(-1)), Cu (0.6-9 mg ml(-1)) and for Co (0.1-5 mg ml(-1)) depending on the isolate. Aspergillus and Rhizopus isolates were tested for their metal biosorption potential for Cr and Cd in vitro. Biosorption experiments were conducted with initial metal concentrations of 2, 4, 6 and 8 mM with a contact time of 4 h and wet fungal biomass (1-5 g) at 25 degrees C. Maximum biosorption of Cr and Cd ions was found at 6 mM initial metal concentration. Aspergillus sp.1 accumulated 1.20 mg of Cr and 2.72 mg of Cd per gram of biomass. Accumulation of these two metals by very tolerant Aspergillus sp.2 isolate was at par with relatively less tolerant Aspergillus sp.1 isolate. Rhizopus sp. accumulated 4.33 mg of Cr and 2.72 mg of Cd per g of biomass. The findings indicated promising biosorption of cadmium and chromium by the Rhizopus and Aspergillus spp. from aqueous solution. There is little, if any, correlation between metal tolerance and biosorption properties of the test fungi.  相似文献

The essential oil (EO) of Ocimum gratissimum inhibited Staphylococcus aureus at a concentration of 0.75 mg/ml. The minimal inhibitory concentrations (MICs) for Shigella flexineri, Salmonella enteritidis, Escherichia coli, Klebsiella sp., and Proteus mirabilis were at concentrations ranging from 3 to 12 microg/ml. The endpoint was not reached for Pseudomonas aeruginosa (>=24 mg/ml). The MICs of the reference drugs used in this study were similar to those presented in other reports. The minimum bactericidal concentration of EO was within a twofold dilution of the MIC for this organism. The compound that showed antibacterial activity in the EO of O. gratissimum was identified as eugenol and structural findings were further supported by gas chromatography/mass spectra retention time data. The structure was supported by spectroscopic methods.  相似文献

The minimal inhibitory concentration (MIC) values of sulfadiazine, penicillin, and rifampin for meningococcal strains isolated from civilians during 1970 were compared. The strains were isolated from various sources and geographical areas and represented several serogroups. The ranges of MIC values were as follows: 0.05 to 20 mg/100 ml for sulfadiazine, 0.01 to 0.4 mug/ml for penicillin, and 0.01 to 0.8 mug/ml for rifampin. There was no significant relationship between MIC values of sensitive or resistant sulfadiazine strains and the MIC values to the other two antimicrobial agents. Comparisons of sulfadiazine MIC values with inhibition zones around sulfathiazole discs showed excellent correlation, provided the strains were separated into sensitive and resistant groups on the basis of growth at 1 mg/100 ml. Regression curves for penicillin and rifampin sensitivity showed homologous sensitive populations with the strains studied.  相似文献

目的 观察天然药物对牙龈卟啉单胞菌(Porphyromonas gingivalis,P.gingivalis)生长的影响,筛选抗菌作用最强的天然药物。方法 选用7种天然药物,采用液体稀释法,结合吸光度测定以确定7种天然药物对P.gingivalis的最小抑菌浓度(MIC)与最小杀菌浓度(MBC)。结果 黄芩、三七、白芷、大黄、五倍子、血藤和川芎对P.gingivalis的MIC值分别为1、1、0.5、0.0156、1、0.5、0.125 mg/ml;MBC值分别为1、1、0.5、0.0625、1、1、0.25mg/ml。结论 7种天然药物对P.gingivalis的生长均表现出明显的抑制作用,大黄的抑菌作用最强,川芎其次。  相似文献

Previous studies showed that methylxanthines increased the antimicrobial activity of gentamicin against Staphylococcus aureus and Pseudomonas aeruginosa. In this study, the effect of non-selective phosphodiesterase (PDE) inhibitors (methylxanthines: aminophylline and caffeine) and partially selective PDE inhibitors, dipyridamole and sildenafil, was evaluated on the antimicrobial activity of gentamicin using checkerboard method. Aminophylline at concentrations of 0.5 and 1 mg/ml reduced the minimal inhibitory concentration (MIC) of gentamicin (2 μg/ml) 2 and 4 times against S. aureus, and at concentrations of 0.5 and 2 mg/ml reduced the MIC of gentamicin (4 μg/ml) 2 and 4 times, respectively, against P. aeruginosa. Caffeine at concentrations of 1 and 2 mg/ml reduced the MIC of gentamicin (2 μg/ml) 4 and 32 times against S. aureus, and at concentrations of 0.12 and 2 mg/ml reduced the MIC of gentamicin (4 μg/ml) 2 and 4 times, respectively, against P. aeruginosa. However, dipyridamole and sildenafil (32 μg/ml) did not show any effect on MIC of gentamicin against S. aureus and P. aeruginosa. These results suggest that methylxanthines could increase gentamicin effects against S. aureus and P. aeruginosa but this effect is not mediated by inhibition of PDE 5, 6, 8, 10 and 11.  相似文献

为了研究金银花中抗菌消炎成分总黄酮、绿原酸的提取方法及其抑菌效果,本实验中用金银花的提取物对11种菌种进行筛选,并对其中有抑制作用的菌种进行了最小抑菌浓度的实验。结果表明金银花的提取物对金黄色葡萄球菌和大肠杆菌有较强的抑制作用,对金黄色葡萄球菌的MIC为6.25mg/ml,对大肠杆菌的MIC为12.5mg/ml。  相似文献

Resistant pathogens are the cause of clinical infections which threatening the patients lives and challenging the health systems through their economic importance. Therefore, new antibacterial agents with a broader spectrum of activity that protect against development of resistance are required. Tigecycline (Tygacil, Wyeth) is a relatively new FDA and EMEA approved glycylcycline antimicrobial with an expanded broad-spectrum activity against pathogens involved in complicated skin and skin structure infections. In this study we evaluated the in vitro activity of tigecycline in comparison to 14 other antibiotics against 182 clinical pathogens by use of the micro dilution method. In overall, tigecycline exhibited the lowest Minimum Inhibitory Concentration (MIC) values in almost all bacteria with a mean of 0.52 ± 1.25 mg/L, followed by meropenem and levofloxacin (mean MIC values 1.29 ± 2.52 and 1.45 ± 3.078 mg/L, respectively). MIC50 and MIC90 values of tigecycline were: 0.06 and 0.15 mg/L for E. coli, 0.12 and 1.00 mg/L for Klebsiella sp., 0.12 and 0.85 mg/L for various Enterobacter sp., 1.00 and 8.00 mg/L for Pseudomonas sp., 0.25 and 1.00 mg/L for Acinetobacter sp., 0.06 and 0.12 mg/L for Serratia sp., 0.12 and 0.25 mg/L for Staphylococcus aureus, 0.5 and 5.00 mg/L for Streptococcus sp. The MIC values recorded were among the lowest in recent literature for Acinetobacter sp. (included A. baumannii), and comparable to those obtained for Klebsiella, Serratia and Enterobacter indicating that tigecycline has a promising in vitro activity.  相似文献
