56个中国地方猪种微卫星基因座的遗传多样性   总被引:33,自引:0,他引:33  
采用FA0—ISAG联合推荐的27个微卫星DNA标记对56个中国地方猪品种和3个引进猪种(杜洛克猪、长白猪、大白猪)进行遗传多样性分析,通过计算等位基因频率、有效等位基因数、平均遗传杂合度、基因分化系数、多态信息含量和遗传距离并进行系统聚类分析,评估其种内遗传变异和种间遗传关系。以聚类结果为基础,将56个中国地方猪种分为12类:Ⅰ、Ⅱ和Ⅲ类都是《中国猪品种志》中的华北型猪种;Ⅳ类相当于其中的江海型猪种;Ⅴ、Ⅵ、Ⅶ、Ⅷ和Ⅸ类的品种大部分属于华中型;Ⅹ和Ⅺ类相当于华南型品种;Ⅻ类相当于西南型品种。提出保种场结合保护区是一种比较符合我国地方猪种实际状况的保种模式。研究结果可为我国地方猪种种质特性研究提供基础数据,为我国地方猪品种资源的合理保护和利用提供科学依据。  相似文献

三峡库区5 个鲢群体遗传变异的微卫星分析   总被引:1,自引:0,他引:1

研究利用10 个高度多态的微卫星标记对三峡水库秭归、巫山、云阳、忠县、木洞等5 个库区鲢(Hypophthalmichthys molitrix)的野生群体进行了遗传多样性分析。检测到161 个等位基因, 群体共有等位基因84 个, 每个微卫星位点的等位基因数729 不等。平均观测杂合度Ho 为0.7840.846, 平均期望杂合度He 为0.8280.847, 平均多态信息含量PIC 为0.7970.817。Fst 值为-0.0010.009, 表明5 个鲢群体间没有遗传分化。Hardy-Weinberg 平衡检验表明巫山、云阳、木洞群体在一些位点上偏离遗传平衡。Bottleneck 分析显示长江三峡库区江段的鲢群体可能在历史上经历了遗传瓶颈。5 个群体间的遗传相似系数为0.8910.950, 遗传距离为 0.0500.115, 根据 Nei's 遗传距离所绘制的聚类图, 表明鲢群体间的遗传距离与其地理距离基本一致。贝叶斯分析结果也证实三峡库区5 个鲢群体可视为一个类群。尽管没有检测到遗传分化, 数据清晰地表明三峡库区的鲢群体仍有很高的遗传多样性, 研究结果为三峡地区和长江上游的鲢种质资源保护和种群评估提供了参考。    相似文献

三个虹鳟养殖群体遗传结构的微卫星分析   总被引:2,自引:0,他引:2  
利用20个微卫星标记对3个虹鳟(Oncorhynchus mykiss)养殖群体进行遗传结构分析。结果表明,(1)3个群体检测的平均等位基因数为3·6~4·1,平均观测杂合度为0·5224~0·6328,平均期望杂合度为0·4736~0·5522,平均多态信息含量为0·4354~0·5084,说明这几个群体多态性属于偏高水平,遗传多样性高。通过d值,确定了Hard-Weinberg平衡的偏离情况,发现AY039638、AY039646在3个群体中都表现为不平衡。对3个群体的遗传距离进行了估算,并进行聚类分析,本溪的两个群体先聚为一支,再与渤海群体相聚,显示出明显的地理特征。(2)本溪虹鳟群体在位点AF352770出现部分等位基因消失的现象;AF352754在本溪群体中表现为位点消失,可作为区分本溪群体和渤海群体的分子标记。(3)综合评价3个群体,渤海站虹鳟群体的遗传多样性最高,与前人研究结果一致。  相似文献

青鱼野生与养殖群体遗传变异的微卫星分析   总被引:3,自引:0,他引:3  
研究利用自主开发的12个微卫星标记, 对来自于长江水系4个野生群体(湖北石首、湖南湘江、江苏邗江、浙江嘉兴)和1个养殖群体(江苏吴江)青鱼(Mylopharyngodon piceus)进行遗传多样性和遗传结构分析。遗传多样性分析结果显示这12个位点多态信息含量(PIC)介于0.660—0.923, 表明这12个位点均具有高度多态性(PIC>0.5)。5个群体的平均等位基因数(Na)介于7.917—11.667, 平均有效等位基因数(Ne)介于4.837—6.035; 平均观测杂合度介于0.713—0.861; 平均期望杂合度介于0.749—0.819; 平均多态信息含量介于0.711—0.788, 表明这5个群体均具有较高的遗传多样性。遗传距离分析结果表明4个野生群体之间遗传距离较近, 养殖群体与这4个野生群体遗传距离均较远。UPGMA系统进化树显示, 湘江群体和石首群体首先聚为一支, 然后与邗江群体和嘉兴群体聚为一支, 最后与吴江养殖群体聚为一支。这12个微卫星位点具有较为丰富的遗传多样性特征, 可以用于青鱼不同群体的种质资源的评估和遗传多样性的分析。  相似文献

文章旨在比较广东地方猪、华南野猪(S.s.chirodontus)和引入品种白细胞抗原复合体(Swine leukocyteantigen complex,SLA)的遗传多样性,为猪的抗病选育提供理论依据。在SLA区域内选取18个多态性丰富的微卫星(SLA-MS),分别对皮特兰、杜洛克、大花白、蓝塘猪以及华南野猪进行遗传分型。结果表明,SLA不同区域平均遗传多样性高低依次为SLAⅡ(He=0.628,PIC=0.581)>SLAⅠ(He=0.530,PIC=0.474)>SLAⅢ(He=0.526,PIC=0.458);5个群体间分子多样性指数(MDI)依次为华南野猪(0.716)>蓝塘猪(0.614)>大花白(0.559)>皮特兰(0.550)>杜洛克(0.507)。总体看来,SLA-MS的遗传多样性高低依次为华南野猪>广东地方猪>引入品种。基于Garza-Williamson指数(GWI)分析,杜洛克和大花白相较于华南野猪瓶颈效应严重;遗传距离分析表明蓝塘猪和华南野猪首先聚为一类,然后与大花白聚为一大类,而皮特兰和杜洛克则单独聚为一支。文章为广东地方猪种质资源的保护、抗病选育和配套系的生产提供了理论基础。  相似文献

为从分子水平上对我国双峰驼(Camelus bactrianus)群体的遗传多样性、群体间遗传关系、群体遗传分化及近交情况进行全面、系统地研究,为双峰驼种质资源保护和新品种培育提供基础数据,本文利用18对微卫星引物,分析了我国9个双峰驼群体和1个蒙古双峰驼群体的遗传多样性和遗传关系。结果显示:10个群体均具有较高的遗传多样性,共检测到242个等位基因,平均等位基因数为13.44,平均有效等位基因数为4.18,平均观察杂合度(Ho)为0.5528。10个群体间存在显著的遗传分化,有9.6%的遗传变异来自群体间,90.4%的遗传变异来自群体内部的个体间。聚类分析、主成分分析和群体遗传结构分析结果都表明10个群体被分成2个明显的分支,新疆4个群体单独聚为一类,剩下的6个群体聚为一类。这一结果可能与它们的地理分布和群体间的地理屏障有关。  相似文献

利用微卫星标记分析山东地方鸡品种的遗传多样性   总被引:53,自引:1,他引:53  
微卫星是近几年来应用较多的一种分子标记,可有效地进行基因鉴定与系谱分析,并可估算群体间的遗传距离。通过选用5个微卫星标记,检测了山东省5个地方鸡种:日照麻鸡、寿光鸡、莱芜黑鸡、济宁百日鸡、鲁西斗鸡以及一个外来鸡种——安卡黄鸡和一个外省地方鸡种——广西黄鸡共7个鸡种的遗传多样性。根据测试结果计算了每个等位基因的频率,以基因频率为基础分析了品种内的遗传变异和品种间的DA遗传距离,并讨论了微卫星多态性在应用于群体遗传变异及亲缘关系等方面的意义。结果表明:共检测到40个等位基因,其中等位基因数最多的位点为。ADL0136(10个);等位基因数最低的位点为ADL0146(5个);而且每个位点的等位基因分布并不均匀,都有一种或几种优势基因存在。在7个品种中,杂合度最低的为寿光鸡,杂合度值为0.3327,因为此鸡种多年来一直由寿光市慈伦种鸡场进行纯繁保种,未与其他鸡种杂交,因此杂合度最小;其他鸡种杂合度也都低于0.4,据分析可能是由于日照麻鸡、济宁百日鸡群体较小;莱芜黑鸡是正在选育的一个品种,个体间遗传关系也不远;安卡黄鸡和广西黄鸡自从引人嘉明公司后,群体近交现象普遍,因此各鸡种杂合度都偏低。由此可见,通过对杂合度的计算,微卫星可以较好地反应群体内的变异。各品种PIC值的变动范围从0.6196(寿光鸡)到0.7027(莱芜黑鸡),PIC值的大小与杂合度的高低体现了较高的一致性。对DA遗传距离的计算表明:日照麻鸡与济宁百日鸡的距离最近,而鲁西斗鸡与其他鸡种距离均较远。用UPGMA法进行聚类分析,结果7个鸡种被聚为3类:山东的4个地方鸡种寿光鸡、日照麻鸡、莱芜黑鸡与济宁百日鸡聚为一类;安卡黄鸡和广西黄鸡聚在一起;鲁西斗鸡独自为一类。这与几个鸡种的分化与选育历史是一致的,因此聚类图能够比较正确地反映7个品种之间的亲缘关系。  相似文献

应用微卫星标记技术以6个品种鹅(东北白鹅、籽鹅、皖西白鹅、豁眼鹅、莱茵鹅、朗德鹅)为实验材料分析不同品种鹅的遗传多样性。利用等位基因频率计算出各群体的平均遗传杂合度(H)多态信息含量(PIC)和群体间的遗传距离DS, 结果表明: 7个微卫星位在6种鹅群体中均表现为高度多态性, 可作为有效的遗传标记来分析各鹅群体的遗传多样性和系统发生关系。实验各群体的杂合度均较高, 平均杂合度在0.6617(莱茵鹅)~0.8814(籽鹅)之间。各品种的PIC值变动大小在0.6145(莱茵鹅)~0.7846(籽鹅)这与杂合度的高低一致。依据DS遗传聚类进行UPGMA聚类分析结果6个品种被分为2类: 国内地方品种东北白鹅、籽鹅、豁眼鹅及皖西白鹅为一类; 外来鹅种莱茵鹅与朗德鹅为一类。表明微卫星标记可准确地反映6个品种的亲缘关系及其所在地域分布上的差异, 适宜于群体遗传结构及遗传关系的研究, 是畜禽遗传多样性研究与保护的有效分析手段。  相似文献

通过选用30个多态性较好的微卫星标记,检测了四川省8个地方鸡品种:峨嵋黑鸡、泸宁鸡、旧院黑鸡、米易鸡、石棉草科鸡、凉山崖鹰鸡、兴文乌骨鸡、沐川乌骨鸡的遗传多样性。利用等位基因频率计算出各群体的平均遗传杂合度(H)、多态信息含量(PIC)和群体间的DA遗传距离。结果表明:30个微卫星位点中有24个微卫星位点在8个鸡群体中的多态信息含量均为高度多态,可作为有效的遗传标记用于各鸡品种的遗传多样性和系统发生关系的分析。各鸡种的杂合度都较高,平均杂合度范围是0.629~0.681,最高的是泸宁鸡(0.681),最低的是旧院黑鸡(0.629)。据分析可能是由于交通闭塞,形成了家禽品种的多种多型,而且杂合度的高低与PIC值的大小体现了较高的一致性。对DA遗传距离的计算表明:用UPGMA法进行聚类分析,结果8个鸡品种被聚为3类:Ⅰ类:峨嵋黑鸡、米易鸡、泸宁鸡、旧院黑鸡聚为一类。米易鸡、泸宁鸡先聚在一起,然后又与峨嵋黑鸡在较近的距离聚在一起,然后再与旧院黑鸡聚在一起;Ⅱ类:石棉草科鸡、兴文乌骨鸡、沐川乌骨鸡聚为一类。兴文乌骨鸡、沐川乌骨鸡在较近的距离聚在一起,然后又与石棉草科鸡在较近的距离聚在一起;Ⅲ类:凉山崖鹰鸡独自聚为一类。这与几个鸡品种的来源、分化、选育历史及地理分布是一致的。  相似文献

选用联合国粮农组织(FAO)和国际动物遗传学会(ISAG)推荐的12对微卫星引物, 采用荧光标记–多重PCR技术, 检测了9个中国地方黄牛品种和3个外来牛品种的遗传多样性。利用等位基因频率计算出各群体的平均遗传杂合度(H)、多态信息含量(PIC)和群体间的DA及DS遗传距离。基于DA遗传距离, 用UPGMA法进行聚类分析, 结果12个中外牛品种被聚为4类: Ⅰ类属于南方黄牛品种, 包括恩施牛、黎平牛、昭通牛和川南山地牛; Ⅱ类属于中原黄牛品种, 包括郏县红牛、早胜牛和平陆山地牛; Ⅲ类属于北方黄牛, 包括延边牛和长白地方牛; Ⅳ类属于外来牛品种, 包括西门塔尔牛、夏洛来牛和德国黄牛。研究结果为中国地方牛品种的保护和利用提供了理论依据。  相似文献

Chinese indigenous pig breeds are recognized as an invaluable component of the world''s pig genetic resources and are divided traditionally into six types. Twenty-six microsatellite markers recommended by the FAO (Food and Agriculture Organization) and ISAG (International Society of Animal Genetics) were employed to analyze the genetic diversity of 18 Chinese indigenous pig breeds with 1001 individuals representing five types, and three commercial breeds with 184 individuals. The observed heterozygosity, unbiased expected heterozygosity and the observed and effective number of alleles were used to estimate the genetic variation of each indigenous breed. The unbiased expected heterozygosity ranged between 0.700 (Mashen) and 0.876 (Guanling), which implies that there is an abundant genetic variation stored in Chinese indigenous pig breeds. Breed differentiation was shown by fixation indices (FIT, FIS, and FST). The FST per locus varied from 0.019 (S0090) to 0.170 (SW951), and the average FST of all loci was 0.077, which means that most of the genetic variation was kept within breeds and only a little of the genetic variation exists between populations. The Neighbor-Joining tree was constructed based on the Nei DA (1978) distances and one large cluster with all local breeds but the Mashen breed, was obtained. Four smaller sub-clusters were also found, which included two to four breeds each. These results, however, did not completely agree with the traditional type of classification. A Neighbor-Joining dendrogram of individuals was established from the distance of – ln(proportions of shared alleles); 92.14% of the individuals were clustered with their own breeds, which implies that this method is useful for breed demarcation. This extensive research on pig genetic diversity in China indicates that these 18 Chinese indigenous breeds may have one common ancestor, helps us to better understand the relative distinctiveness of pig genetic resources, and will assist in developing a national plan for the conservation and utilization of Chinese indigenous pig breeds.  相似文献

There are three native Portuguese horse breeds: Lusitano, Sorraia and Garrano. This study compares diversity patterns of 17 protein and 12 microsatellite markers in these three as well as 30 other breeds to infer relationships among the breeds and to compare levels of polymorphism of these breeds for use in conservation efforts. The Garrano and the Lusitano showed a high level of genetic diversity, similar to that observed for most of the other analysed breeds, while the Sorraia and Friesian breeds showed low levels of variation for both genetic marker types. The combined protein and microsatellite data produced a tree that fit historical records well and with greater confidence levels than those for either data set alone. The combined genetic diversity and relationship information provides important baseline data for future breed conservation efforts, especially for a critically endangered breed such as the Sorraia.  相似文献

The genetic diversities and relationships of four Chinese indigenous pig breeds and one Australian commercial pig breed have been evaluated using 27 microsatellites recommended by the International Society of Animal Genetics (ISAG) and the Food and Agriculture Organization (FAO). The allele frequencies, effective numbers of alleles and the polymorphic information content have been calculated. Nei's standard genetic distances have been used to construct a UPGMA dendrogram, which has been evaluated by the Bootstrap test. The utility of microsatellites for evaluating genetic diversity of pigs is discussed.  相似文献

Six local chicken breeds are registered in Hungary and are regarded as Hungarian national treasures: Hungarian White, Yellow and Speckled, and Transylvanian Naked Neck White, Black and Speckled. Three Hungarian academic institutes have maintained these genetic resources for more than 30 years. The Hungarian Yellow, the Hungarian Speckled and the Transylvanian Naked Neck Speckled breeds were kept as duplicates in two separate subpopulations since time of formation of conservation flocks at different institutes. In this study, we investigated genetic diversity of these nine Hungarian chicken populations using 29 microsatellite markers. We assessed degree of polymorphism and relationships within and between Hungarian breeds on the basis of molecular markers, and compared the Hungarian chicken populations with commercial lines and European local breeds. In total, 168 alleles were observed in the nine Hungarian populations. The F ST estimate indicated that about 22% of the total variation originated from variation between the Hungarian breeds. Clustering using structure software showed clear separation between the Hungarian populations. The most frequent solutions were found at K  = 5 and K  = 6, respectively, classifying the Transylvanian Naked Neck breeds as a separate group of populations. To identify genetic resources unique to Hungary, marker estimated kinships were estimated and a safe set analysis was performed. We show that the contribution of all Hungarian breeds together to the total diversity of a given set of populations was lower when added to the commercial lines than when added to the European set of breeds.  相似文献

A set of eleven pig breeds originating from six European countries, and including a small sample of wild pigs, was chosen for this study of genetic diversity. Diversity was evaluated on the basis of 18 microsatellite markers typed over a total of 483 DNA samples collected. Average breed heterozygosity varied from 0.35 to 0.60. Genotypic frequencies generally agreed with Hardy-Weinberg expectations, apart from the German Landrace and Schwäbisch-Hällisches breeds, which showed significantly reduced heterozygosity. Breed differentiation was significant as shown by the high among-breed fixation index (overall FST = 0.27), and confirmed by the clustering based on the genetic distances between individuals, which grouped essentially all individuals in 11 clusters corresponding to the 11 breeds. The genetic distances between breeds were first used to construct phylogenetic trees. The trees indicated that a genetic drift model might explain the divergence of the two German breeds, but no reliable phylogeny could be inferred among the remaining breeds. The same distances were also used to measure the global diversity of the set of breeds considered, and to evaluate the marginal loss of diversity attached to each breed. In that respect, the French Basque breed appeared to be the most "unique" in the set considered. This study, which remains to be extended to a larger set of European breeds, indicates that using genetic distances between breeds of farm animals in a classical taxonomic approach may not give clear resolution, but points to their usefulness in a prospective evaluation of diversity.  相似文献

We determined the genetic diversity and evolutionary relationships among 26 Chinese indigenous horse breeds and two introduced horse breeds by genotyping these animals for 27 microsatellite loci. The 26 Chinese horse breeds come from 12 different provinces. Two introduced horse breeds were the Mongolia B Horse from Mongolia and the Thoroughbred Horse from the UK. A total of 330 alleles were detected, and the expected heterozygosity ranged from 0.719 (Elenchuns) to 0.780 (Dali). The mean number of alleles among the horse breeds ranged from 6.74 (Hequ) to 8.81 (Debao). Although there were abundant genetic variations found, the genetic differentiation was low between the Chinese horses, which displayed only 2.4% of the total genetic variance among the different breeds. However, genetic differentiation (pairwise FST) among Chinese horses, although moderate, was still apparent and varied from 0.001 for the Guizou–Luoping pair to 0.064 for the Jingjiang–Elenchuns pair. The genetic differentiation patterns and genetic relationships among Chinese horse breeds were also consistent with their geographical distribution. The Thoroughbred and Mongolia B breeds could be discerned as two distinct breeds, but the Mongolia B horse in particular suffered genetic admixture with Chinese horses. The Chinese breeds could be divided into five major groups, i.e. the south or along the Yangtze river group (Bose, Debao, Wenshan, Lichuan, Jianchang, Guizhou, Luoping, Jinjiang and Dali), the Qinghai‐Tibet Plateau group (Chaidamu, Hequ, Datong, Yushu, Tibet Grassland and Tibet Valley), the Northeast of China group (Elenchuns, Jilin and Heihe), the Northwest of China group (Kazakh, Yili and Yanqi) and the Inner Mongolia group (Mongolia A, Sanhe, Xinihe,Wuzhumuqin and Sengeng). This grouping pattern was further supported by principal component analysis and structure analysis.  相似文献

Analysis of the genetic variation of an endangered population is an important component for the success of conservation. Animals from two local Romanian pig breeds, the Mangalitsa and Bazna, were analysed for variation at a number of genetic loci using PCR-based DNA tests. Polymorphism was assessed at loci which 1) are known to cause phenotypic variation, 2) are potentially involved in trait differences or 3) are putative candidate genes. The traits considered are disease resistance, growth, coat colour, meat quality and prolificacy. Even though the populations are small and the markers are limited to specific genes, we found significant differences in five of the ten characterised loci. In some cases the observed allele frequencies were interesting in relation to gene function and the phenotype of the breed. These breeds are part of a conservation programme in Romania and marker information may be useful in preserving a representative gene pool in the populations. The use of polymorphisms in type 1 (gene) markers may be a useful complement to analysis based on anonymous markers.  相似文献

Genetic variation in four indigenous chicken breeds from the Veneto region of Italy was assessed using amplified fragment length polymorphism (AFLP) markers. A total of 99 individuals were analysed using three AFLP primer combinations that produced 70 polymorphisms. Four indigenous Veneto chicken breeds (Ermellinata, Padovana, Pépoi and Robusta) and a reference broiler line were included in the analysis. Breed-specific markers were identified in each breed. The expected heterozygosity did not differ significantly among the indigenous Veneto chicken breeds and the broiler line. The coefficient of gene variation (Gst) value across loci indicated that almost half of the total variability was observed among breeds. Nei's standard genetic distance between pairs of breeds showed that the distance between the broiler line and the Pépoi breed was greater than the distances between the broiler line and the other three chicken breeds. Cluster analysis based on standard genetic distances between breeds indicated that the Padovana and Pépoi breeds were closely related. Factorial analysis based on a binary matrix of the AFLP data showed a clear distinction of all breeds.  相似文献
