Relative nuclear DNA contents in cortex parenchyma cells in root segments of 3- and 7-d-old soybean seedlings grown at 25 °C and in plants grown for 3 d at 25 °C, and then for 4 d at 10 °C, were determined with cytophotometry. Measurements revealed that in each variant the cortex cell nuclei with DNA content between 2C and 8C were in all the examined segments and nuclei with 8C – 16C DNA appeared in higher parts of roots. However, in chilled plant cells the number of 8C – 16C DNA nuclei was very low. Therefore, chilling inhibited endoreplication in comparison with plants grown at 25 °C for 7 d, and even reduced endopolyploidy level as compared to the initial seedlings, i.e. 3-d-old plants. DNA contents in root hairs grown at 25 °C (control) and in root hairs emerged at 10 °C were also determined. In controls 4C – 8C DNA nuclei predominated while in chilled plants an additional population of 2C – 4C DNA appeared. Thus a reduction of DNA synthesis was brought about by low temperature. The occurrence of an intermediate DNA contents besides those with full endoreplication cycles suggests the possibility of differential DNA replication. This suggestion seems to be supported by the lack of 3H-thymidine incorporation into root hair nuclei at the examined developmental stage both in control and chilled root hairs. The same number, but larger, chromocentric lumps in polyploid cortex cell nuclei of higher root zones, in comparison to meristematic nuclei, suggests that endoreduplication process occurred. Soybean (Glycine max L.) plants grown in nutrient solution were exposed to 1 mM Cd(NO3)2 for 24 h. Dynamics of distribution of cadmium among its different forms (water soluble, Ca-exchangeable and complexed) in the intercellular space and the ratio of the intercellular and intracellular cadmium in roots, stems and leaves were studied. In roots, in the beginning of treatment the largest portion of Cd was found in the intercellular space and 1 h later Cd content started to decrease, so that between 13- and 24-h treatment an equilibrium was reached in which about 70 % of Cd was found inside cells. In stems, already after 1-h treatment, the Cd concentrations in the cells and intercellular space were similar, the equilibrium being disturbed after 13 h, so that after 24-h treatment 80 % of Cd was found inside cells. In leaves, up to the 13 h Cd distribution showed fluctuation, after that equilibrium was reached, with 70 % of intracellular Cd. The highest contents of all Cd forms in the intercellular space was observed in roots.  相似文献

Aluminium effect on the mobility of apoplast protein in root tips was studied. Two-day seedlings of soybean (Glycine max. (L.) Merr. cv. Tsurunoko) were treated with 50 μM AlCl3 for 2 h. Using infiltration method, the apoplast protein in root tips was extracted with 20 or 100 mM MgCl2. When 20 mM MgCl2 was used to collect weakly bound protein to apoplast, the amount of protein extracted was reduced to be about 20 % compared with that of control and the band of 97 kDa disappeared in SDS-PAGE gel. However, the 97 kDa protein could be extracted by 100 mM MgCl2, which were used for extraction of more tightly bound protein to apoplast, and the amount was estimated to be the same as that of control. When the protein was further developed in two-dimensional electrophoresis, three spots were found between pI 6.4 and 6.5. This is the first report of an Al effect on the mobility of apoplast protein. The dynamics of seed storage protein biosynthesis was studied under field conditions during two vegetative seasons. Two soybean (Glycine max L. Merr.) genotypes were examined: BOSA (drought tolerant) and L 121 (drought susceptible). Seed samples were taken from plants at three stages of seed maturation (50 and 70 d after flowering, and at full maturity). The earlier synthesis of the -subunit of the 7S protein occurred in the drought susceptible cultivar. We have not found such differences in the synthesis of the - and -subunits of the 7S protein. Our results did not confirm significant genotypic differences in protein composition of the mature seeds between the cultivars studied, but have pointed out to the differences in the dynamics of protein biosynthesis during seed maturation and desiccation.  相似文献

The influence of media components on the initiation of somatic embryogenesis in three genotypes of soybean was investigated. The following genotypes were used: Iroquois, Macon, and Savoy. Media modifications included sucrose concentration, type and concentration of auxin at two pH levels, and pH level independently. Immature cotyledons were used as the source of explant. Cotyledons were placed on a medium containing MS salts, B5 vitamins, sucrose, and auxin. Gelrite (0.2%) was used as the solidifying agent. Sucrose concentrations of 1, 2, 3, 4.5, or 6% were used. The auxins used included 2,4-dichlorophenoxyacetic acid (2,4-D) and naphthaleneacetic acid (NAA) with each at concentrations of 45.2, 90.4, 135.7, 180.9, and 226.2 M. The pH of each the media was adjusted to either 5.7 or 7.0 with 1 N NaOH. In an additional experiment, the effect of the two pH levels, 5.7 and 7.0, was investigated independently. Overall, the frequency of somatic embryogenesis significantly varied among the different genotypes used in this study, with Iroquois showing the highest response. Frequency of somatic embryogenesis also varied in response to the different treatments used, including sucrose and auxin. The highest initiation (91.7%) and mean number of somatic embryos per responding explant (14.9) of Iroquois was observed in a medium containing 2% sucrose. The highest initiation (97.1%) and mean number of somatic embryos per responding explant (19.5) was observed in Iroquois on 135.7 M 2,4-D and Savoy on 135.7 M 2,4-D, respectively, for the auxin by pH level experiment. No significant differences were observed among the two pH treatments used.  相似文献

The stability of a plastid transgene has been evaluated in soybean transformants over six generations. These transformants had integrated the aadA selection cassette in the intergenic region between the rps12/7 and trnV genes. Three independent homoplasmic T0 transformation events were selected and ten plants from each event propagated to generation T5 in the absence of selection pressure. No transgene rearrangement nor wild-type plastome were detected in generation T5 by Southern blot analysis. All tested progenies were uniformly resistant to spectinomycin. Therefore, soybean transformants of generations T0 and T5 appear to be genetically and phenotypically identical.  相似文献

In 10-d-old soybean seedlings, the growth of roots and shoots was significantly inhibited at 50 and 100 M and more Cd2+, respectively, and by 50 M or more Ni2+. Although total protein content of roots exposed to 200 M Cd2+ or Ni2+ was similarly decreased compared to the control, the activity of nitrate reductase was much more inhibited by Cd2+. Ni2+-treatment (200 M) induced an accumulation of all free amino acids in roots associated with a decrease in alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities reflecting the accumulation of both alanine and aspartic acid, respectively. Cd2+-treatment (200 M) decreased the amount of all free amino acids. In addition, cysteine which is the main amino acid consisting the phytochelatin complexes constituted about 17.5 % of total free amino acids. The activities of both ALT and AST in Cd2+-treated roots were higher than in Ni2+-treated roots suggesting higher conversion of alanine and aspartate to pyruvate and oxaloacetate. Primary leaves excised from either Cd2+ or Ni2+-treated seedlings showed similar pattern of enzyme activities as roots.  相似文献

The protection compound benzo-(1,2,3)-thiadiazole-7-carbothioic acid S-methyl ester (BTH) was applied to soybean roots or leaves in a dose of 20 cm3 of a solution containing 25 μg g−1 of the active ingredient. Electrophoretic profiles of chitinase and superoxide dismutase were not altered by the product. Increased activity of two root anionic peroxidases and three differential isoforms of these enzymes were observed in plants with roots treated by BTH, which can be used as biochemical markers of the BTH effect. Interspecific competition between four soybean cultivars (PI471938, Stressland, Essex and Forrest) and the weed, common waterhemp was investigated under increasing weed densities (i.e. 0, 1, 4 and 16 plants per pot). Soybean height and leaflet number were measured over a 45-d period and used to calculate relative growth rates (RGR). Trigonelline (TRG) concentration was determined within the V1 leaf of 45-d-old soybean plants. Soybean leaflet number (P[lt ]0.05), soybean height (P[lt ]0.05) and soybean RGRh (expressed in terms of height) differed significantly (P[lt ]0.05) according to waterhemp density. At each waterhemp density Stressland matured at a significantly faster rate whereas the maturation rate of Essex decreased in the presence of waterhemp. Final TRG concentrations were affected by the interaction between soybean cultivar and waterhemp density. Under no competition, TRG concentration was significantly lower in Forrest relative to PI471938, Stressland and Essex. TRG concentrations in Essex declined in higher waterhemp densities.  相似文献

Trigonelline Concentration in Field-Grown Soybean in Response to Irrigation   总被引:2,自引:0,他引:2  
Trigonelline (TRG) is a conjugate of nicotinic acid, and is postulated to function as a compatible solute in response to salinity- and water deficit-stresses. TRG concentrations and several agronomic characteristics were measured under irrigated field and non-irrigated field conditions within 18 soybean (Glycine max) genotypes using leaves taken from different growth stages (vegetative, flowering and pod development). Under irrigation, relative water content (RWC) ranged from 90.0 to 99.6 %. Under non-irrigation, RWC ranged from 86.3 to 97.5 %. TRG concentration ranged from 364 to 555 μg g−1(d.m.) under irrigation, and from 404 to 570 μg g−1(d.m.) under non-irrigation. TRG concentrations increased in the majority of genotypes (15 of 18) under non-irrigation even though RWC did not significantly differ in many genotypes between treatments. TRG decreased as plants progressed to pod development and seed filling. Mean seed yield under non-irrigated conditions declined 55 % relative to the irrigated controls. The effects of temperature and initial inoculum density of Meloidogyne incognita on soybean growth and nematode reproduction were investigated in greenhouse temperature tanks and in controlled-growth chambers. The interactions of initial inoculum density (Pi) and soil temperature in effects on shoot growth were adequately described by multiple-regression models. At the highest temperatures (30 or 32/28 C), moderate to high inoculum killed many plants. A Pi of 27,000 eggs/15-cm-diam pot retarded shoot growth at 26 C. Only the greatest Pi (81,000 eggs/15-cm pot) suppressed shoot growth at 18, 22, or 20/16 C. Inoculation with 3,000 or 9,000 eggs/plant resulted in heavier root systems at all temperatures except 30 C. At that temperature, 9,000 eggs suppressed root growth. At 18 and 26 C, a Pi of 81,000 eggs was required to retard root growth. Nematode reproduction was related directly to temperature and Pi except at a density of 81,000 eggs/15-cm pot.  相似文献

The effect of aluminium (0.5 –1.0 mM) on contents of phosphorus, pigments, and pigment-protein complexes was studied in soybean (Glycine max Merril.) grown in different nutrient medium with and without P. Increased Al concentrations led to the decrease in the contents of chlorophylls (Chl) a and b, and carotenoids (Car) in soybean leaves, but Chl a/b ratio did not vary significantly. High irradiance (HI) effects on xanthophyll cycle pigments (XCP) and activity of violaxanthin de-epoxidase (VDE) in terms of de-epoxidation index (DEI) were studied in soybean calli. The calli from the hypocotyl segments of 5-d seedlings were induced on a solid (1.1 % agar) MS medium (pH 5.8) supplemented with 4.52 M 2,4-dichloro-phenoxyacetic acid, 2.32 M kinetin, and 3 % sucrose. After a 30 d cultivation, the green calli were irradiated for 24 h with white light (HI, 1 300 mol m–2 s–1) and VDE was isolated from the photosystem 2 (PS2) particles. In the control (0 h irradiation) callus, the reaction of PS2 particles with VDE in the presence or absence of Tween 20 resulted in the decrease of VIO content and the increase of ZEA content. In the 24 h HI-callus, the reaction of PS2 particles in the absence of VDE led to the decrease of VIO and ANT contents and increase of ZEA content. In the control, DEIs in the presence of VDE with or without 0.1 %Tween 20 (1.04 and 1.06, respectively) were significantly higher than the DEI (0.76) in the absence of VDE. In the HI-callus, DEIs in the presence of VDE with or without 0.1 %Tween 20 (0.98 and 0.96, respectively) were similar to that (1.03) in the absence of VDE.  相似文献

该研究基于大豆基因组数据库,根据拟南芥ABI4蛋白的氨基酸序列,经比对分析,获得了大豆中的2个GmABI4基因,分别命名为GmABI4-1(GenBank登录号为XM_014766551.1)和GmABI4-2(GenBank登录号为NM_001249003)。TMHMM软件和系统进化转录分析表明,这2个基因编码的蛋白均不具有信号肽,二级结构主要以无规则卷曲和延伸链为主;进化树分析表明,大豆GmABI4和野生大豆亲缘关系较近。荧光定量PCR分析表明,GmABI4-1与GmABI4-2基因在大豆种子与豆荚中的表达量均高于根、茎、叶、花等其他组织,推测可能与调控大豆种子生命活动相关。  相似文献

Bragg and Cobb soybean [Glycine max (L.) Merr.] seeds were germinated in sand at temperatures ranging from 25 to 40°C. Emergence decreased with increasing temperature above 37°C, with virtually no emergence at 40°C. Emergence of 12 other cultivars at 38°C ranged from 25 to 95%. Photosynthesis in iron-deficient soybean and maize leaves decreased drastically. The quantum yield of photosystem 2 (PS2) electron transport (ΦPS2), the efficiency of excitation energy capture by open PS2 reaction centres (Fv′/Fm′), and photochemical quenching coefficient (qP) under high irradiance were lowered significantly by iron deficiency, but non-photochemical quenching (NPQ) increased markedly. The analysis of the polyphasic rise of fluorescence transient showed that iron depletion induced a pronounced K step both in soybean and maize leaves. The maximal quantum yield of PS2 photochemistry (Φpo) decreased only slightly, however, the efficiency with which a trapped exciton can move an electron into the electron transport chain further than QA0) and the quantum yield of electron transport beyond QAEo) in iron deficient leaves decreased more significantly compared with that in control. Thus not only the donor side but also the acceptor of PS2 was probably damaged in iron deficient soybean and maize leaves. Summary Silicon during the early vegetative stage did not affect the oven dry weight of any of the various tissues of the soybean plant. Silicon did, however, decrease the Mn concentration in the youngest fully mature leaf at intermediate levels of Mn. This effect did not occur at the lowest or highest Mn levels. Deficiency and toxicity symptoms were moderated to a slight degree by Si except at the highest level of Mn.  相似文献

Embryogenic suspension cultures of soybean (Glycine max L. cv. Iroquois) were subjected to mutagenesis using varying concentrations (1, 3, 10, and 30 mM) of ethyl methanesulfonate (EMS). Depending on the concentration of EMS used, the mean survival rate of embryogenic cultures decreased from 74 % (1 mM EMS) to 43 % after 30 mM EMS treatment. Random amplified polymorphic DNA (RAPD) analysis was used to determine whether induction of genetic variability in embryogenic cultures in response to the different EMS treatments may result in identification of polymorphic markers. Two of 35 ‘core’ primers tested revealed polymorphisms. One of the primers, OPO-01/1150, revealed polymorphism in tissue treated with 10 mM EMS, while the other primer, OPO-05/1200, revealed polymorphism in tissue treated with either 1 or 30 mM EMS. These results suggest that RAPD markers are useful in detecting mutations in embryogenic cultures of soybean. Leaf photosynthetic rates were measured on field-grown soybeans during the 1980 season. Comparisons were made between different cultivars and isolines representative of maturity groups I–IV. Mature, fully expanded leaves at different nodes on the plant were measured in high light to determine which had the highest potential photosynthetic rates at any one time. Successive leaves during the growing season had maximum rates which increased from about 22 mol CO2 m-2 s-1 on 25 June to a peak of 30–44 mol CO2 m-2 s-1 in early August.The persistency and eventual decline in the maximum rate was associated with the maturity group and related dates of flowering, pod fill and onset of senescence. Early maturing cultivars (groups I and II) had higher peak rates (38–44 mol CO2 m-2 s-1) than later maturing cultivars (30–35 mol CO2 m-2 s-1, groups III and IV). However, the photosynthetic rates of early maturing cultivars declined rapidly after attaining their peak, whereas the leaves of later maturing cultivars maintained their photosynthetic activity for much longer.  相似文献
