Cell wall structure of wheat coleoptiles grown under continuous hypergravity (300 g) conditions was investigated. Length of coleoptiles exposed to hypergravity for 2-4 days from germination stage was 60-70% of that of 1 g control. The amounts of cell wall polysaccharides substantially increased during the incubation period both in 1 g control and hypergravity-treated coleoptiles. As a results, the levels of cell wall polysaccharides per unit length of coleoptile, which mean the thickness of cell walls, largely increased under hypergravity conditions. The major sugar components of the hemicellulose fraction, a polymer fraction extracted from cell walls with strong alkali, were arabinose (Ara), xylose (Xyl) and glucose (Glc). The molar ratios of Ara and Xyl to Glc in hypergravity-treated coleoptiles were higher than those in control coleoptiles. Furthermore, the fractionation of hemicellulosic polymers into the neutral and acidic polymers by the anion-exchange column showed that the levels of acidic polymers in cell walls of hypergravity-treated coleoptiles were higher than those of control coleoptiles. These results suggest that hypergravity stimuli bias the synthesis of hemicellulosic polysaccharides and increase the proportion of acidic polymers, such as arabinoxylans, in cell walls of wheat coleoptiles. These structural changes in cell walls may contribute to plant resistance to hypergravity stimuli.  相似文献

Elongation growth of dark grown maize (Zea mays L cv. Cross Bantam T51) coleoptiles and mesocotyls was suppressed by hypergravity at 30 g and above. Acceleration at 300 g significantly decreased the mechanical extensibility of cell walls of both organs. Hypergravity increased the amounts of hemicellulose and cellulose per unit length in mesocotyl walls, but not in coleoptile walls. The weight average molecular masses of hemicellulosic polysaccharides were also increased by hypergravity in both organs. On the other hand, the activities of beta-glucanases extracted from coleoptile and mesocotyl cell walls were decreased by hypergravity. These results suggest that the decreased activities of beta-glucanases by hypergravity cause an increase in the molecular mass of hemicellulosic polysaccharides of both organs. The upshift of molecular mass of hemicellulosic polysaccharides as well as the thickening of cell walls under hypergravity conditions seems to be involved in making the cell wall mechanically rigid, thereby inhibiting elongation growth of maize coleoptiles and mesocotyls.  相似文献

The present study was conducted to investigate the mechanism inducing the difference in the cell wall extensibility of rice ( Oryza sativa L. cv. Koshihikari) coleoptiles grown under various temperature (10–50°C) conditions. The growth rate and the cell wall extensibility of rice coleoptiles exhibited the maximum value at 30–40°C, and became smaller as the growth temperature rose or dropped from this temperature range. The amounts of cell wall polysaccharides per unit length of coleoptile increased in coleoptiles grown at 40°C, but not at other temperature conditions. On the other hand, the molecular size of hemicellulosic polysaccharides was small at temperatures where the cell wall extensibility was high (30–40°C). The autolytic activities of cell walls obtained from coleoptiles grown at 30 and 40°C were substantially higher than those grown at 10, 20 and 50°C. Furthermore, the activities of (1→3),(1→4)- β -glucanases extracted from coleoptile cell walls showed a similar tendency. When oat (1→3),(1→4)- β -glucans with high molecular mass were incubated with the cell wall enzyme preparations from coleoptiles grown at various temperature conditions, the extensive molecular mass downshifts were brought about only by the cell wall enzymes obtained from coleoptiles grown at 30–40°C. There were close correlations between the cell wall extensibility and the molecular mass of hemicellulosic polysaccharides or the activity of β -glucanases. These results suggest that the environmental temperature regulates the cell wall extensibility of rice coleoptiles by modifying mainly the molecular mass of hemicellulosic polysaccharides. Modulation of the activity of β -glucanases under various temperature conditions may be involved in the alteration of the molecular size of hemicellulosic polysaccharides.  相似文献

Effects of polyethylene glycol (PEG)-induced osmotic stress on the mechanical properties of cell walls and the levels of their components were investigated along intact wheat (Triticum aestivum L.) coleoptiles. Stress-relaxation analysis showed that the cell walls of stressed coleoptiles were loosened as compared with those of unstressed ones not only in the apical but in the basal regions. The amounts of wall-bound ferulic acid (FA) and diferulic acid (DFA) of stressed coleoptiles were substantially lower than those of unstressed ones in all regions. The cellulose and hemicellulose contents increased toward the coleoptile base. Osmotic stress reduced the cellulose content in the basal region but it slightly affected the hemicellulose content. The molecular weight of hemicellulose in the apical region of stressed coleoptiles was higher than that of unstressed ones, while that in the basal region was almost the same in both coleoptiles. FA, DFA and cellulose contents correlated with the cell wall mechanical property. The amount and molecular weight of hemicellulose, however, did not correlate. These results suggest that the reduced levels of FA and DFA in all regions and cellulose in the basal region of wheat coleoptiles are involved in maintaining the cell wall extensibility under osmotic stress.  相似文献

Elongation growth of dark-grown azuki bean (Vigna angularis Ohwi et Ohashi cv. Takara) epicotyls was suppressed by hypergravity at 30 x g and above. Acceleration at 300 x g significantly decreased the mechanical extensibility of cell walls. The amounts of cell wall polysaccharides (pectin, hemicellulose-II and cellulose) per unit length of epicotyls increased under the hypergravity condition. Hypergravity also increased the amounts and the weight-average molecular mass of xyloglucans in the hemicellulose-II fraction, while decreasing the activity of xyloglucan-degrading enzymes extracted from epicotyl cell walls. These results suggest that hypergravity increases the amounts and the molecular mass of xyloglucans by decreasing xyloglucan-degrading activity. Modification of xyloglucan metabolism as well as the thickening of cell walls under hypergravity conditions seems to be involved in making the cell wall mechanically rigid, thereby inhibiting elongation growth of azuki bean epicotyls.  相似文献

Application of abscisic acid (ABA) to dark-grown wheat (Triticumaestivum L.) roots interfered the cell wall hardening of coleoptilesduring several days of the treatment. Although the amounts ofwall-bound diferulic (DFA) and ferulic (FA) acids in coleoptilesincreased as the coleoptiles grew, ABA substantially reducedtheir increases. When ABA was removed, however, these contentsincreased and reached levels near those of control coleoptiles.A close correlation was observed between the levels of DFA andFA and the mechanical properties of cell walls. The ratio ofthe amount of DFA to FA was almost constant irrespective ofgrowth conditions. The activities of phenylalanine- (PAL) andtyrosine-ammonia-Iyase (TAL) increased rapidly in the controlcoleoptiles. ABA greatly reduced the increases in these enzymeactivities. In response to ABA removal, the enzyme activitiesincreased rapidly. There was a close correlation between theincrease in FA level and the changes in enzyme activities. Theseresults suggest that ABA suppresses the increases in PAL andTAL activities in wheat coleoptiles, resulting in the reducedlevel of wall-bound FA, which, in turn, may cause the reducedDFA level and thereby maintain cell wall extensibility. Sections excised from maize coleoptiles incorporated radioactivity from proline, tyrosine, and phenylalanine into structural components of the cell wall. Only about 2% of radioactivity from proline taken up by sections was incorporated into cell wall; about 24% of that incorporated was in hydroxyproline and the rest remained in proline. In contrast, as much as 40% of the radioactivity from phenylalanine and 30% from tyrosine was incorporated into cell wall material. Most of this radioactivity was in saponifiable ferulic acid. Small amounts of p-coumaric and diferulic acid were found, but only a small fraction of the hemicellulose can possibly be immobilized directly through cross-linking of diferulic esters. Substantial amounts of radioactivity from aromatic amino acids remained insoluble after strong alkali extractions of wall material, and a large fraction of polysaccharide was solubilized by dilute alkali following oxidation of phenolics by acidic NaClO2. Hence, hemicellulosic material in the cell walls of maize coleoptiles may be organized and cross-linked primarily through alkali-resistant etherified aromatics.  相似文献

Elongation growth of etiolated hypocotyls of cress (Lepidium sativum L.) was suppressed when they were exposed to basipetal hypergravity at 35 x g and above. Acceleration at 135 x g caused a decrease in the mechanical extensibility and an increase in the minimum stress-relaxation time of the cell wall. Such changes in the mechanical properties of the cell wall were prominent in the lower regions of hypocotyls. The amounts of cell wall polysaccharides per unit length of hypocotyls increased under the hypergravity condition and, in particular, the increase in the amount of cellulose in the lower regions was conspicuous. Hypergravity did not influence the neutral sugar composition of either the pectin or the hemicellulose fraction. The amount of lignin was also increased by hypergravity treatment, although the level was low. The data suggest that hypergravity modifies the metabolism of cell wall components and thus makes the cell wall thick and rigid, thereby inhibiting elongation growth of cress hypocotyls. These changes may contribute to the plants' ability to sustain their structures against hypergravity.  相似文献

Cotton (Gossypium herbaceum L.) fiber development consists of a fiber elongation stage (up to 20 d post-anthesis) and a subsequent cell wall thickening stage. Cell wall analysis revealed that the extractable matrix (pectic and hemicellulosic) polysaccharides accounted for 30-50% of total sugar content in the fiber elongation stage but less than 3% in the cell wall thickening stage. By contrast, cellulose increased dramatically after the fiber elongation ceased. The amounts of extractable xyloglucans and arabinose- and galactose-containing polymers per seed increased in the early fiber elongation stage and decreased thereafter. The amounts of extractable acidic polymers and non-cellulosic beta-glucans (mainly composed of beta-1,3-glucans) increased in parallel with fiber elongation and then decreased. The molecular masses of extractable non-cellulosic beta-glucans, and arabinose- and galactose-containing polymers decreased during both fiber elongation and cell wall thickening stages. The molecular mass of extractable xyloglucans also decreased during the fiber elongation stage, but this decrease ceased during the cell wall thickening stage. Conversely, the molecular size of acidic polymers in the extractable pectic fraction increased during both stages. Thus, not only the amounts but also the molecular size of the extractable matrix polysaccharides showed substantial changes during cotton fiber development.  相似文献

The relationship between the mechanical properties of cell walls and the levels of wall-bound ferulic (FA) and diferulic (DFA) acids was investigated in wheat (Triticum aestivum L.) coleoptiles grown under osmotic stress (60 mM polyethylene glycol [PEG] 4000) conditions. The cell walls of stressed coleoptiles remained extensible compared with those of the unstressed ones. The contents of wall-bound FA and DFA increased under unstressed conditions, but the increase was substantially reduced by osmotic stress. In response to PEG removal, these contents increased and reached almost the same levels as those of the unstressed coleoptiles. A close correlation was observed between the contents of FA and DFA and the mechanical properties of cell walls. The activities of phenylalanine ammonia-lyase and tyrosine ammonia-lyase increased rapidly under unstressed conditions. Osmotic stress substantially reduced the increases in enzyme activities. When PEG was removed, however, the enzyme activities increased rapidly. There was a close correlation between the FA levels and enzyme activities. These results suggest that in osmotically stressed wheat coleoptiles, reduced rates of increase in phenylalanine ammonia-lyase and tyrosine ammonia-lyase activities suppress phenylpropanoid biosynthesis, resulting in the reduced level of wall-bound FA that, in turn, probably causes the reduced level of DFA and thereby maintains cell wall extensibility.  相似文献

The correlation between hypocotyl elongation, cell wall loosening and changes in cell wall polysaccharides was studied using intact lettuce seedlings grown in the dark or in light together with gibberellic acid (GA) and/or 5-fluorodeoxyuridine (FUDR). The following results were obtained:
1) The production of pectic, hemicellulosic and cellulosic polysaccharides look place in parallel with hypocotyl elongation, which was substantially affected by different growth conditions.
2) The mole percentage sugar composition of pectic and hemicellulosic polysaccharides changed in response to dark, light, GA, or FUDR treatments.
3) The amounts of xylose and glucose in hemicellulosic polysaccharides and those of galactosc, rhumnose and uronic acid in pectic polysaccharides increased in parallel with hypocotyl elongation.
4) Statistical analysis of the quantitative relationship between sugars composing polysaccharides revealed that the uronic acid content changed in parallel with those of rhamnose and galactose in pectic polysaccharides, and the content of xylose varied in parallel with those of fucose and glucose.
5) The content of hemicellulosic polysaccharides was correlated with cell wall loosening represented by a decrease in the minimum stress-relaxation time. Changes in the stress-relaxation time value were correlated with those in the content of araltinose and galactose in hemicellulosic polysaccharides.
Based on these results, the relationship between hypocotyl elongation, changes in cell wall polysaccharides, and cell wall loosening is discussed with respect to the effect of GA and FUDR on hypocotyl elongation.  相似文献

The relationship between the formation of cell wall-bound ferulic acid (FA) and diferulic acid (DFA) and the change in activities of phenylalanine ammonia-lyase (PAL) and cell wall-bound peroxidase (CW-PRX) was studied in rice shoots. The length and the fresh mass of shoots increased during the growth period from day 4 to 6, while coleoptiles ceased elongation growth on day 5. The amounts of FA and DFA isomers as well as cell wall polysaccharides continued to increase during the whole period. The activities of PAL and CW-PRX greatly increased in the same manner during the period. There were close correlations between the PAL activity and ferulate content or between the CW-PRX activity and DFA content. The expression levels of investigated genes for PAL and putative CW-PRX showed good accordance with the activities of these enzymes. These results suggest that increases in PAL and CW-PRX activities are cooperatively involved in the formation of ferulate network in cell walls of rice shoots and that investigated genes may be, at least in part, associated with the enzyme activities. The substantial increase in such network probably causes the maturation of cell walls and thus the cessation of elongation growth of coleoptiles.  相似文献

According to the acid-growth hypothesis, auxin-induced secretion of hydrogen ions activate “wall loosening” enzymes that change the rheological properties of the cell wall. The wall loosening process may yield monosaccharides by the enzymic cleavage of load-bearing polysaccharides. Alkaline hydrolysis liberated ferulic and diferulic acid from polysaccharides of the Avena coleoptile ( Avena sativa L. cv. Victory I) cell walls. The amount of the two phenolic acids bound to cell walls increased substantially at day 4–5 after sowing, when the growth rate of the coleoptile started to decrease. The level of these acids was almost constant from the tip to base in 3-day-old coleoptiles, but increased toward the basal zone in 4- and 5-day-old ones. The ratio of diferulic acid to ferulic acid was almost constant irrespective of coleoptile age and zone. An increase in the amount of ferulic and diferulic acids bound to cell wall polysaccharides correlated with a decrease in extensibility and with an increase in minimum stress-relaxation time and relaxation rate of the cell wall. The level of lignin in the cellulose fraction increased as coleoptiles aged, but this increase did not correlate with changes in mechanical properties of the cell walls. These results suggest that ferulic acid, ester-linked to cell wall polysaccharides, is oxidized to give diferulic acid, which makes the cell wall mechanically rigid by cross-linking matrix polysaccharides and results in limited cell extension growth. In addition, it is probable that the step of feruloylation of cell wall polysaccharides is rate-limiting in the formation of in-termolecular bridges by diferulic acid in Avena coleoptile cell walls.  相似文献

To elucidate the mechanism by which white fluorescent light (5 W m-2) stimulates the formation of diferulic acid (DFA) in cell walls, the effect of light on phenylalanine-and tyrosine-ammonia-lyase (PAL, EC 4.3.1.5 and TAL, EC 4.3.1.5) and peroxidase activities was studied using coleoptiles of maize ( Zea mays L. cv. Cross Bantam T51). Growth rate of dark-grown coleoptiles was highest at the basal zone and decreased towards the tip, while continuous irradiation caused an inhibition of growth, especially at the basal zone. Light decreased the cell wall extensibility in all zones of the coleoptile. The amounts of DFA, ferulic acid (FA) and p -coumaric acid ( p -CA) increased by severalfold in cell walls of light-grown maize coleoptiles as compared with those grown in the dark. Strong correlations were observed between the increase in the contents of either DFA, FA or p -CA and the decrease in cell wall extensibility. Light decreased the wall-bound peroxidase activity. No correlation was found between DFA content and peroxidase activity. The activities of PAL and TAL were enhanced upon white light irradiation. The increment in either DFA, FA or p -CA content was correlated with an increase in PAL activity, but not with that in TAL activity. White light may promote DFA formation in the cell walls of maize coleoptiles by enhancing PAL activity.  相似文献

The growth kinetics and variations in cell wall matrix polysaccharides and glycoside hydrolases during seedling development of the drought-tolerant wheat cultivar (cv. Hong Mang Mai) were compared with the drought-sensitive cultivar (cv. Shirasagikomugi). After 15d of culture in water at 22 degrees C under constant irradiance of 98mumolm(-2)s(-1), the length of the coleoptile and leaf sheath of Hong Mang Mai seedlings was 1.7 times longer than those of Shirasagikomugi seedlings. In the cell walls isolated from coleoptiles and leaf sheaths of the seedling of the two cultivars, the contents of arabinose, xylose, and glucose changed during development. The cell walls were fractionated progressively with 50mM CDTA, 50mM Na(2)CO(3), 1M KOH and 4M KOH, and sugar composition was determined. The amount of CDTA-soluble fraction from the Hong Mang Mai cell walls was 2.4-fold higher than that from the Shirasagikomugi cell walls at 6d of culture, and a considerable decrease was observed during development. The ratio of arabinose to xylose in 1M KOH-soluble fraction from the two cultivars decreased. The amount of 4M KOH-soluble fraction from the Shirasagikomugi cell walls was affected much more than those of the Hong Mang Mai cell walls. Many glycoside hydrolase activities were detected in the protein fractions from coleoptiles and leaf sheaths of the two cultivars, and the activities of licheninase, 1,3-1,4-beta-glucanase, and 1,3-beta-glucanase in the LiCl-soluble protein fraction increased drastically during development of the Shirasagikomugi seedlings. These findings suggest that the metabolism of the cell wall matrix polysaccharides of the drought-tolerant wheat cultivar is far different from that of the drought-sensitive wheat cultivar during seedling development.  相似文献

Resurrection plants have the unique capacity to revive from an air-dried state. In order to tolerate desiccation they have to overcome a number of stresses, mechanical stress being one. In leaves of the Craterostigma species, an extensive shrinkage occurs during drying as well as a considerable cell wall folding. Our previous microscopically analysis using immunocytochemistry on the resurrection plant Craterostigma wilmsii , has shown an increase in labelling of xyloglucan and unesterified pectins in the cell wall during drying. In this study, we have undertaken a biochemical approach to separate, quantify and characterize major cell wall polysaccharides in fully hydrated and dry leaves of C. wilmsii . Our results show that the overall cell wall composition of C. wilmsii leaves was similar to that of other dicotyledonous plants with respect to the pectin content. However, the structure of the hemicellulosic polysaccharide xyloglucan was characterized to be XXGG-type. The data also demonstrate marked changes in the hemicellulosic wall fraction from dry plants compared to hydrated ones. The most conspicuous change was a decrease in glucose content in the hemicellulosic fraction of dry plants. In addition, xyloglucan from the cell wall of dry leaves was relatively more substituted with galactose than in hydrated walls. Together these findings show that dehydration induces significant alteration of polysaccharide content and structure in the cell wall of C. wilmsii , which in turn might be involved in the modulation of the mechanical properties of the wall during dehydration.  相似文献

The effects of aluminum (Al) on root elongation, the mechanical extensibility of the cell wall, and the amount of cell-wall polysaccharides in the roots of Al-resistant (Atlas 66) and Al-sensitive (Scout 66) cultivars of wheat ( Triticum aestivum L.) were examined. Exposure to 10 μ M AlCl3 for 6 h inhibited root elongation in Scout 66 but not in Atlas 66. It also decreased the mechanical extensibility of the cell wall in the roots of both cultivars, but prominently only in the roots of Scout 66. The amount of hemicellulose in the 10-mm region of root apex of Scout 66 was increased by the exposure to Al, especially in the apical regions. Al did not influence the neutral sugar composition of either pectin or hemicellulose in Scout 66 roots. However, Al increased the weight-average molecular mass of hemicellulosic polysaccharides and the amounts of wall-bound ferulic and diferulic acids in Scout 66 roots. These findings suggest that Al modifies the metabolism of cell-wall components and thus makes the cell wall thick and rigid, thereby inhibiting the growth of wheat roots.  相似文献
