Cucumis sativus L.) seedlings were irradiated or not irradiated with UV-B for several days in environment-controlled growth chambers. The first leaves irradiated with UV-B were retarded in growth but simultaneously acquired a remarkably high tolerance to oxidative stress, as induced by paraquat treatment, compared with the non-irradiated leaves. This enhanced tolerance was observed within 1d after the start of UV-B irradiation and was maintained during the 12 d period of UV-B treatment. The effects of UV-B on several antioxidative enzymes were examined, and activities of superoxide dismutase, ascorbate peroxidase and guaiacol peroxidase, but not of glutathione reductase, were found to be enhanced. However, activation of these enzymes occurred only from 6 d after the start of irradiation. In contrast, accumulation of phenolic compounds was observed within 1d after the start of UV-B irradiation. HPLC analysis of phenolic compounds showed the distinct enhancement of a substance, which may have antioxidative properties in cucumber seedlings irradiated with UV-B. On the basis of these results, we conclude that not only antioxidative enzymes but also other factors in cucumber seedlings irradiated with UV-B, such as phenolic compounds, may participate in the enhanced tolerance to oxidative stress. Using two barley (Hordeum vulgare) cultivars (cvs. Tokak and Hamidiye) nutrient solution experiments were conducted in order to study the genotypic variation in tolerance to Cd toxicity based on (i) development of leaf symptoms, (ii) decreases in dry matter production, (iii) Cd concentration and (iv) changes in antioxidative defense system in leaves (i.e., superoxide dismutase, ascorbate peroxidase, glutathione reductase, catalase, ascorbic acid and non-protein SH-groups). Plants were grown in nutrient solution under controlled environmental conditions, and subjected to increasing concentrations of Cd (0, 15, 30, 60 and 120 micromol/L Cd) for different time periods. Of the barley cultivars Hamidiye was particularly sensitive to Cd as judged by the severity and earlier development of Cd toxicity symptoms on leaves. Within 48 h of Cd application Hamidiye rapidly developed severe leaf Cd toxicity symptoms whereas in Tokak the leaf symptoms appeared only slightly. Hamidiye also tended to show more decrease in growth caused by Cd supply when compared to Tokak. The differences in sensitivity to Cd between Tokak and Hamidiye were not related to Cd concentrations in roots and shoots or Cd accumulation per plant. With the exception of catalase, activities of the enzymes involved in detoxification of reactive oxygen species (ROS) were markedly enhanced in Hamidiye by increasing Cd supply. By contrast, in Tokak there was either only a slight increase or no change in the activities of the antioxidative enzymes. Similarly, levels of ascorbic acid and especially non-protein SH-groups were increased in Hamidiye by Cd supply, but not affected in Tokak. The results indicate the existence of a large genotypic variation between barley cultivars for Cd tolerance. The differential Cd tolerance found in the barley cultivars was not related to uptake or accumulation of Cd in plants, indicating importance of internal mechanisms in expression of differential Cd tolerance in barley. As a response to increasing Cd supply particular increases in antioxidative mechanisms in the Cd-sensitive barley cultivar Hamidiye suggest that the high Cd sensitivity of Hamidiye is related to enhanced production and oxidative damage of ROS.  相似文献

The impacts of heavy metals on oxidative stress and growth of spring barley   总被引:1,自引:0,他引:1  
Oxidative stress is accepted to play a significant role in stress symptoms, caused by different stressors in a variety of organisms. In this study seedlings of spring barley (Hordeum vulgare L.) were exposed to a wide range of copper, zinc, chromium, nickel, lead and cadmium concentrations in order to determine the relationships between heavy metals-induced oxidative stress and plant growth inhibition. All investigated heavy metals induced an essential increase in lipid peroxidation and a reduction of dry biomass along with an increase in metal concentration in the nutrient solution. A very close and statistically significant exponential relationship between lipid peroxidation and growth inhibition was detected in this study. According to the results of analysis of variance (ANOVA), the intensity of nonspecific oxidative stress is identified as the main factor of barley growth inhibition, explaining 75% of total variance. Almost 10% of growth inhibition is attributed to the specific impact of heavy metals. The most pronounced increase of malondialdehyde content and growth inhibition was observed in Cu and Cd treatments, whereas the lowest changes in observed indicators were detected after exposure to Zn and Pb.  相似文献

The present study examined the effect of salicylic acid (SA) pre-treatment on soybean seedlings exposed to cadmium and/or UV-B stress. Dry mass, pigment content, net photosynthetic rate (PN), stomatal conductance (gs), and transpiration rate (E) were decreased by the Cd and/or UV-B stress. SA alleviated the adverse effects of Cd and/or UV-B on growth, pigment content, PN, and gs, but did not mitigate the inhibitory effect of Cd/UV-B on E, or that of Cd on chlorophyll fluorescence parameters. Cd and/or UV-B induced oxidative stress and increased lipid peroxidation that was significantly decreased by SA pre-treatment. The Cd and/or UV-B increased superoxide dismutase (SOD) activity, decreased peroxidase (POD) activity, and catalase (CAT) activity was mostly unaltered. SA might act as one of the potential antioxidants as well as a stabilizer of membrane integrity to improve plant resistance to the Cd and/or UV-B stress.  相似文献

The hypothesis that mitogen‐activated protein kinase (MAPK) signalling is important in plant defences against metal stress has become accepted in recent years. To test the role of oxidative signal‐inducible kinase (OXI1) in metal‐induced oxidative signalling, the responses of oxi1 knockout lines to environmentally realistic cadmium (Cd) and copper (Cu) concentrations were compared with those of wild‐type plants. A relationship between OXI1 and the activation of lipoxygenases and other initiators of oxylipin production was observed under these stress conditions, suggesting that lipoxygenase‐1 may be a downstream component of OXI1 signalling. Metal‐specific differences in OXI1 action were observed. For example, OXI1 was required for the up‐regulation of antioxidative defences such as catalase in leaves and Fe‐superoxide dismutase in roots, following exposure to Cu, processes that may involve the MEKK1‐MKK2‐WRKY25 cascade. Moreover, the induction of Cu/Zn superoxide dismutases in Cu‐exposed leaves was regulated by OXI1 in a manner that involves fluctuations in the expression of miRNA398. These observations contrast markedly with the responses to Cd exposure, which also involves OXI1‐independent pathways but rather involves changes in components mediating intracellular communication.  相似文献

Plant species capable of hyperaccumulating heavy metals are of considerable interest for phytoremediation and phytomining. This work aims to identify the role of antioxidative metabolism in heavy metal tolerance in the Cd hyperaccumulator, Thlaspi caerulescens. Hairy roots of T. caerulescens and the non-hyperaccumulator, Nicotiana tabacum (tobacco), were used to test the effects of high Cd environments. In the absence of Cd, endogenous activities of catalase were two to three orders of magnitude higher in T. caerulescens than in N. tabacum. T. caerulescens roots also contained significantly higher endogenous superoxide dismutase activity and glutathione concentrations. Exposure to 20 ppm (178 microM) Cd prevented growth of N. tabacum roots and increased hydrogen peroxide (H(2)O(2)) levels by a factor of five relative to cultures without Cd. In contrast, growth was maintained in T. caerulescens, and H(2)O(2) concentrations were controlled to low, nontoxic levels in association with a strong catalase induction response. Treatment of roots with the glutathione synthesis inhibitor, buthionine sulfoximine (BSO), exacerbated H(2)O(2) accumulation in Cd-treated N. tabacum, but had a relatively minor effect on H(2)O(2) levels and did not reduce Cd tolerance in T. caerulescens. Lipid peroxidation was increased by Cd treatment in both the hyperaccumulator and non-hyperaccumulator roots. This work demonstrates that metal-induced oxidative stress occurs in hyperaccumulator tissues even though growth is unaffected by the presence of heavy metals. It also suggests that superior antioxidative defenses, particularly catalase activity, may play an important role in the hyperaccumulator phenotype of T. caerulescens.  相似文献

Exposure of roots to low Cd concentration induced morphogenic responses including the inhibition of root growth and the radial swelling of root tip. High Cd concentrations within a few minutes caused a robust induction of superoxide generation leading to the cell death and root growth arrest. This toxic superoxide generation blocked the development of low Cd concentration-activated morphogenic responses. While the morphogenic responses of roots to low Cd concentration are induced very rapidly and probably due to the interaction of Cd with the apoplast of root tissue, high Cd concentration-induced superoxide production required the entry of Cd into the symplast. Auxin signaling is involved in the activation of Cd-induced morphogenic defence responses but not in the Cd-induced toxic superoxide generation. These results suggest that oxidative stress is not a primary cause for the Cd-induced morphogenic responses such as growth reduction and radial cell expansion in barley root tips.  相似文献

The response of barley seedlings, subjected to 150 mmol/L NaCl for 4 days at different light regimes (4 d in the light, 4 d in darkness and a 12 h light/dark cycle) before UV-B radiation was investigated. NaCl treatment resulted in a decrease of total chlorophyll content and an increase in H2O2, free proline and lipid peroxidation, as quantified by measurement of malondialdehyde. Significantly more proline was accumulated in the light than in darkness. The combination of UV-B and NaCl treatment produced an additive effect on most of the parameters studied. UV-B radiation reduced the chlorophyll/carotenoids ratio and photochemical efficiency of PSII as estimated by chlorophyll fluorescence. NaCl pre-exposure decreased H2O2 generation and lipid peroxidation and alleviated the inhibitory effect of UV-B on PSII activity. Proline accumulated under salt stress conditions might be one of the reasons for the observed tolerance of barley seedlings to UV-B radiation.  相似文献

Physiological and biochemical responses of wheat seedlings to drought, UV-B radiation, and combined stress were investigated. Drought, UV-B, and combined stresses retarded seedling growth by 26.5, 29.1, and 55.9%, respectively. One reason for growth retardation may be the oxidative damage indicated by an increase in the H2O2 content and lipid peroxidation degree. Furthermore, there was negative correlation between shoot fresh weight and H2O2 content, fresh weight and the content of thiobarbituric acid-reacting substances (TBARS), and the positive correlation between H2O2 content and TBARS (R 2 = 0.9251, 0.9005, and 0.9007, respectively). The activities of superoxide dismutase, guaiacol peroxidase, and ascorbate peroxidase increased under drought, UV-B, and the combination of stresses, while catalase activity decreased under the combined stress as compared to the control. The combination of drought and UV-B caused more severe damage to wheat seedlings than stress factors applied separately. Thus, the combined application of drought and UV-B had more strong adverse effects on wheat seedlings. The addition of 0.2 mM sodium nitroprusside (SNP) enhanced wheat seedling growth under drought, UV-B, and combined stress, likely, due to decreasing the accumulation of H2O2 and lipid peroxidation as well as activating the antioxidant enzymes. When rice seedlings grown for 10 and 20 days were subjected to in vitro drought stress of −0.5 and −2.0 MPa for 24 h, an increase in the concentration of superoxide anion (O2.−), increased level of lipid peroxidation and a decrease in the concentration of total soluble protein and thiols was observed in stressed seedlings compared to controls. The concentration of H2O2 as well as ascorbic acid declined with imposition of drought stress, however glutathione (GSH) concentration declined only under severe drought stress. The activities of total superoxide dismutases (SODs) as well as ascorbate peroxidase (APX) showed consistent increases with increasing levels of drought stress, however catalase activity declined. Mild drought stressed plants had higher guaiacol peroxidase (GPX) and chloroplastic ascorbate peroxidase (c-APX) activity than control grown plants but the activity declined at the higher level of drought stress. The activities of enzymes involved in regeneration of ascorbate i.e. monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) and glutathione reductase (GR) were higher in drought stressed plants compared to controls. Results suggest that drought stress induces oxidative stress in rice plants and that besides SOD, the enzymes of ascorbate-glutathione cycle, which have not been studied in detail earlier under stressful conditions, appear to function as important component of antioxidative defense system under drought stress.  相似文献

There is limited information on the impacts of present-day solar ultraviolet-B radiation (UV-B) on biomass and grain yield of field crops and on the mechanisms that confer tolerance to UV-B radiation under field conditions. We investigated the effects of solar UV-B on aspects of the biochemistry, growth and yield of barley crops using replicated field plots and two barley strains, a catalase (CAT)-deficient mutant (RPr 79/4) and its wild-type mother line (Maris Mink). Solar UV-B reduced biomass accumulation and grain yield in both strains. The effects on crop biomass accumulation tended to be more severe in RPr 79/4 (≈ 32% reduction) than in the mother line (≈ 20% reduction). Solar UV-B caused measurable DNA damage in leaf tissue, in spite of inducing a significant increase in UV-absorbing sunscreens in the two lines. Maris Mink responded to solar UV-B with increased CAT and ascorbate peroxidase (APx) activity. No effects of UV-B on total superoxide dismutase (SOD) activity were detected. Compared with the wild type, RPr 79/4 had lower CAT activity, as expected, but higher APx activity. Neither of these activities increased in response to UV-B in RPr 79/4. These results suggest that growth inhibition by solar UV-B involves DNA damage and oxidative stress, and that constitutive and UV-B-induced antioxidant capacity may play an important role in UV-B tolerance.  相似文献

Changes in MAO and antioxidative enzymes copper-zinc superoxide dismutase (CuZnSOD), manganese superoxide dismutase (MnSOD) and catalase (CAT) activities were examined in the hypothalamus and the hippocampus of Wistar rats exposed to cold stress (6 °C) for 180 min and heat stress (38 °C) for 60 min.

2. Extreme environmental temperatures caused stressor-specific changes in the hypothalamic and hippocampal MAO and antioxidative enzyme activities, being dependent on the stressor applied (cold or heat) but not on the brain region studied (the hypothalamus or hippocampus).

Water stress is one of the main abiotic factors that reduces plant growth, mainly due to high evaporative demand and low water availability. In order to evaluate the effects of drought stress on certain morphological and physiological characteristics of two canola cultivars, we conducted a factorial experiment based on a completely randomized design. The findings show that drought stress exacerbations result in the plant's response to stress due to increased canola resistance caused by changes in plant pigments, proline, catalase, ascorbate peroxidase, peroxidase, superoxide dismutase and malondialdehyde, glucose, galactose, rhamnose and xylose. These in turn ultimately influence the morphological characteristics of canola. Drought stress reduces the concentration of carotenoids, chlorophyll a, chlorophyll b, total chlorophylls; however, glucose, galactose, rhamnose, xylose, proline, catalase, ascorbate peroxidase, peroxidase, superoxide dismutase, malondialdehyde (in leaves and roots) and the chlorophyll a and b ratios were increased. Reduction of plant height, stem height, root length, fresh and dry weight of canola treated with 300 g/l PEG compared to non‐treatment were 0.264, 0.236, 0.394, 0.183 and 0.395, respectively. From the two canola cultivars, the morphological characteristics of the NIMA increased compared to the Ks7 cultivar. Interaction effects of cultivar and drought stress showed that NIMA cultivar without treatment had the highest number of morphological characteristics such as carotenoid concentration, chlorophyll a, chlorophyll b, total chlorophylls a and b, whereas the cultivar with 300 g/l PEG (drought stress) had the highest amount of proline, malondialdehyde, soluble sugars and enzymes in leaves and roots. Increasing activity of oxidative enzymes and soluble sugars in canola under drought stress could be a sign of their relative tolerance to drought stress.  相似文献

Barley seedlings grown on filter paper moistened with 1mM Cd showed 50% root growth inhibition within 24h of exposure. The amount of cadmium after 24h Cd treatment was highest in the first 2mm-long apical root segment, while it was slightly higher in the fourth segment, 6-8mm behind the root tip, after 48h. In recovery experiments, when Cd-treated plants were transferred onto filter paper moistened with distilled water, a large amount of Cd was localised in the apoplast and considerable cell death was detected even though root growth was renewed. This indicates that cell death is likely an active physiological process that contributes to the removal of Cd from the root during root growth recovery. Elevated lipid peroxidation and thiol contents were detected in all individual segments of Cd-treated barley root. On the other hand, proline accumulation was disturbed during Cd stress, showing a significant decrease in all of the studied segments except the first. Cd-induced alteration in the expression of genes involved in metal signalling and detoxification and in drought and oxidative stress responses indicates that Cd-induced water and oxidative stress is responsible for the root growth inhibition, probably through an accelerated differentiation of root tissues.  相似文献

F26对H2O2胁迫的响应程度最高, 过氧化氢酶的快速合成对细胞抵抗H2O2胁迫至关重要, 当细胞及时分解进入胞内的H2O2, 胁迫对细胞的氧化损伤程度并不高, 相反会刺激细胞的生长和底物消耗, 当胁迫超过过氧化氢酶的分解能力时, H2O2会迅速抑制细胞生长和过氧化氢酶合成; 由于 ·与细胞作用的方式和效果与H2O2不同, 超氧化物歧化酶和过氧化氢酶的快速合成并不能保证细胞及时有效地清除胞内的活性氧, 因此, 细胞对 ·胁迫的响应程度要低于H2O2胁迫; 在所考察的3种活性氧中, OH·胁迫(Fenton反应体系)对细胞的氧化损伤程度最大, 胁迫强烈地抑制了细胞生长和抗氧化物酶的合成。由此表明, 由于不同活性氧的化学性质有所不同, 细胞对不同种类、程度和形式的活性氧胁迫会表现出不同的生物学效应, 为了提高自身对氧胁迫的抵抗能力, 微生物会通过自身的代谢调节适应新的环境, 包括调整抗氧化物酶合成水平、改变生长速度以及底物消耗速率等。  相似文献

Magnesium-deficiency conditions applied to spinach cultures caused an oxidative stress status in spinach chloroplast monitored by an increase in reactive oxygen species (ROS) accumulation. The enhancement of lipids peroxide of spinach chloroplast grown in magnesium-deficiency media suggested an oxidative attack that was activated by a reduction of antioxidative defense mechanism measured by analysing the activities of superoxide dismutase, catalase, ascorbate peroxidase, guaiacol peroxidase, and glutathione reductase, as well as antioxidants such as carotenoids and glutathione content. As the antioxidative response of chloroplast was reduced in spinach grown in magnesium-deficiency media, it caused a significant reduction of spinach plant weight, old leaves turning chlorosis. However, cerium treatment grown in magnesium-deficiency conditions decreased the malondialdehyde and ROS, and increased activities of the antioxidative defense system, and improved spinach growth. Together, the experimental study implied that cerium could partly substitute for magnesium and increase the oxidative stress-resistance of spinach chloroplast grown in magnesium-deficiency conditions, but the mechanisms need further study.  相似文献

UV-B (0.4 W m–2) irradiation and cadmium (2 and 8 M) treatments separately inhibited the survival, growth, pigment content, and photosynthetic electron transport in Plectonema boryanum. Phycocyanin was the main target to UV-B and Cd and it was followed by chlorophyll a and carotenoids. UV-B and Cd caused strong inhibition on activities of photosystem 2 (PS2) and the whole electron transport chain, whereas photosystem 1 (PS1) was the least affected. UV-B and Cd treatment accelerated respiration, lipid peroxidation, and the activities of superoxide dismutase and catalase. However, enhancement in catalase activity was considerably less (5 – 50 %) as compared to SOD activity. As compared to individual treatment, the effect of their combination (UV-B + Cd) was more detrimental to the above parameters. A synergistic interaction of UV-B and Cd is probably due to increased cadmium uptake as a result of increased membrane permeability caused by lipid peroxidation in P. boryanum after UV-B exposure.  相似文献

Drought is a severe environmental constraint, causing a significant reduction in crop productivity across the world. Salicylic acid (SA) is an important plant growth regulator that helps plants cope with the adverse effects induced by various abiotic stresses. The current study investigated the potential effects of SA on drought tolerance efficacy in two barley (Hordeum vulgare) genotypes, namely BARI barley 5 and BARI barley 7. Ten-day-old barley seedlings were exposed to drought stress by maintaining 7.5% soil moisture content in the absence or presence of 0.5, 1.0 and 1.5 mM SA. Drought exposure led to severe damage to both genotypes, as indicated by phenotypic aberrations and reduction of dry biomass. On the other hand, the application of SA to drought-stressed plants protected both barley genotypes from the adverse effects of drought, which was reflected in the improvement of phenotypes and biomass production. SA supplementation improved relative water content and proline levels in drought-stressed barley genotypes, indicating the osmotic adjustment functions of SA under water-deficit conditions. Drought stress induced the accumulation of reactive oxygen species (ROS), such as hydrogen peroxide (H2O2) and superoxide (O2 •− ), and the lipid peroxidation product malondialdehyde (MDA) in the leaves of barley plants. Exogenous supply of SA reduced oxidative damage by restricting the accumulation of ROS through the stimulation of the activities of key antioxidant enzymes, including superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX) and glutathione peroxidase (GPX). Among the three-applied concentrations of SA, 0.5 mM SA exhibited better mitigating effects against drought stress considering the phenotypic performance and biochemical data. Furthermore, BARI barley 5 showed better performance under drought stress than BARI barley 7 in the presence of SA application. Collectively, our results suggest that SA played a crucial role in improving water status and antioxidant defense strategy to protect barley plants from the deleterious effects of water deficiency.  相似文献
