The nickel(II) complexes with the quinolone antibacterial agents oxolinic acid, flumequine, enrofloxacin and sparfloxacin in the presence of the N,N′-donor heterocyclic ligand 2,2′-bipyridylamine have been synthesized and characterized. The quinolones act as bidentate ligands coordinated to Ni(II) ion through the pyridone oxygen and a carboxylato oxygen. The crystal structure of [(2,2′-bipyridylamine)bis(sparfloxacinato)nickel(II)] has been determined by X-ray crystallography. UV study of the interaction of the complexes with calf-thymus DNA (CT DNA) has shown that they bind to CT DNA with [(2,2′-bipyridylamine)bis(flumequinato)nickel(II)] exhibiting the highest binding constant to CT DNA. The cyclic voltammograms of the complexes have shown that in the presence of CT DNA the complexes can bind to CT DNA by the intercalative binding mode which has also been verified by DNA solution viscosity measurements. Competitive study with ethidium bromide (EB) has shown that the complexes can displace the DNA-bound EB indicating that they bind to DNA in strong competition with EB. The complexes exhibit good binding propensity to human or bovine serum albumin protein having relatively high binding constant values. The biological properties of the [Ni(quinolonato)2(2,2′-bipyridylamine)] complexes have been evaluated in comparison to the previously reported Ni(II) quinolone complexes [Ni(quinolonato)2(H2O)2], [Ni(quinolonato)2(2,2′-bipyridine)] and [Ni(quinolonato)2(1,10-phenanthroline)]. The quinolones and their Ni(II) complexes have been tested for their antioxidant and free radical scavenging activity. This article interrogates the relationship between the Italian island of Lampedusa and trans-Mediterranean migration. It explores how the construction of Lampedusa as a border zone has been implicated in the rise and fall in numbers of migrants reaching the island's shores over the last two decades. It proceeds to consider the appropriateness of interpreting death and detention on Lampedusa in terms of ‘bare life’. While acknowledging how Giorgio Agamben's formulation of bare life has been problematized in relation to irregular migration and taking into account the frequent acts of migrants' political agency on the island itself, it is argued that the transformation of Lampedusa by the media and political establishment into a spectacle of bare life is not only instrumental to the functioning of migration management at Europe's southern border but is also constitutive of the subordinate position of migrants in Italian society and its labour market.  相似文献

This article provides a theoretical framework for analysing discrimination against second-generation immigrant girls in education and the labour market by proposing an intersectional approach. Drawing upon selected elements of the findings of our Neskak Gora Project – a qualitative research conducted between 2009 and 2011 in Denmark, France, Italy, the Netherlands, Spain and the UK and funded by the EC Daphne III Programme – we show that the intersection of class, gender and ‘race’/ethnicity, which is at play at the structural, institutional and discursive levels of systems of intersectional discrimination, works in variable ways. While gendered educational structures seem partly to benefit female immigrant youth at school, gendered disadvantages are experienced particularly in the transition to the labour market. This highlights the necessary acknowledgement of the ‘discontinuity’ of axes of inequality that are manifested in different ways, according to specific contexts, institutional settings and moments of the individual's life cycle.  相似文献

Research on the new second generation has paid much attention to testing one of the hypotheses posed by segmented assimilation theory – downward assimilation into America's underclass – and has neglected to examine other possible outcomes. In this paper, I address a much understudied pathway – assimilation by way of the ethnic community – based on a case study of Chinese immigrant children in the USA. I show that the children of Chinese immigrants have made inroads into mainstream America through educational achievement, not only because of the strong value their parents put on education but also because resources generated in the ethnic community help actualize that value. The Chinese American experience suggests that, in order to advance to the rank of middle-class Americans, immigrant parents have chosen the ethnic way to facilitate children's social mobility and achieved success. Paradoxically, ‘assimilated’ children have also relied on ethnicity for empowerment to fight negative stereotyping of the racialized other.  相似文献

Insecurity theory states that religiosity is predominantly affected by insecurities experienced during childhood, instead of present insecurities. The empirical research of these aspects, however, has been hampered by the difficulty to disentangle past and present contextual effects. In this respect, first-generation migrants offer an interesting case study that allows us to discern: (1) contextual effects experienced during childhood (i.e. associated with the origin country); (2) contextual effects experienced during later life (i.e. associated with the destination country); and (3) individual effects experienced during later life in the destination country. We test hypotheses using the European Social Survey (ESS) in cross-classified multi-level analyses on 5,900 individuals within on the one hand twenty-five destination countries and on the other 146 origin countries. While insecurity theory offers interesting prospects of explaining origin country variance, the applicability of insecurity theory to migrants at the individual and destination level is questioned by the results.  相似文献

Summary Twelve alfalfa cultivars inoculated with an indigenous strain (RM9) ofRhizobium meliloti, were compared for their seedling morphological characters, and growth characters, including net assimilation rate (NAR), relative growth rate (RGR), leaf area ratio (LAR) and relative nitrogen assimilation rate (RN). Highly significant differences were obtained between cultivars for most characters.Simple correlation showed that NAR influenced RGR (r=0.91) more than leaf area ratio (LAR) (r=–0.44), and that most characters measured were highly correlated with seedling dry weight. Factor analysis showed that NAR, RGR and RN contributed 25% of the total variation in the dependence structure. The grouping indicated that the higher the NAR and RN the greater was the RGR. Path-coefficient analysis showed that NAR had more important direct and indirect effects than RN in dry matter accumulation. The relationship implied that selection for plants with high NAR, or high efficiency in converting light energy to dry matter production could contribute greater N2 fixation in alfalfa.  相似文献

This article focuses on ageing in transnationalism. Drawing on the experiences of Italians in the UK as a paradigmatic example of settled European migrants, it explores the lived experiences of this group of older migrants. Using Levitt and Glick Schiller's framework, it concentrates first on migrants' ways of being and then on their ways of belonging. The article argues that a transnational lens is necessary to understand the experiences of older migrants and that a focus on older people needs to be incorporated into studies of transnationalism. Through a discussion of their narratives and experiences, the article offers a long view on the migration process and brings attention to the significance of gender, time and the life course to understand both migrants' transnationalism and their integration.  相似文献

  • Stomata modulate the exchange of water and CO2 between plant and atmosphere. Although stomatal density is known to affect CO2 diffusion into the leaf and thus photosynthetic rate, the effect of stomatal density and patterning on CO2 assimilation is not fully understood.
  • We used wild types Col‐0 and C24 and stomatal mutants sdd1‐1 and tmm1 of Arabidopsis thaliana, differing in stomatal density and pattern, to study the effects of these variations on both stomatal and mesophyll conductance and CO2 assimilation rate. Anatomical parameters of stomata, leaf temperature and carbon isotope discrimination were also assessed.
  • Our results indicate that increased stomatal density enhanced stomatal conductance in sdd1‐1 plants, with no effect on photosynthesis, due to both unchanged photosynthetic capacity and decreased mesophyll conductance. Clustering (abnormal patterning formed by clusters of two or more stomata) and a highly unequal distribution of stomata between the adaxial and abaxial leaf sides in tmm1 mutants also had no effect on photosynthesis.
  • Except at very high stomatal densities, stomatal conductance and water loss were proportional to stomatal density. Stomatal formation in clusters reduced stomatal dynamics and their operational range as well as the efficiency of CO2 transport.

Abstract Among sulfur compounds, thiosulfate and polythionates are present at least transiently in many environments. These compounds have a similar chemical structure and their metabolism appears closely related. They are commonly used as energy sources for photoautotrophic or chemolithotrophic microorganisms, but their assimilation has been seldom studied and their importance in bacterial physiology is not well understood. Almost all bacterial strains are able to cleave these compounds since they possess thiosulfate sulfur transferase, thiosulfate reductace or S -sulfocysteine synthase activities. However, the role of these enzymes in the assimilation of thiosulfate or polythionates has not always been clearly established.
Elemental sulfur is, on the contrary, very common in the environmental. It is an energy source for sulfur-reducing eubacteria and archaebacteria and many sulfur-oxidizing archaebacteria. A phenomenon still not well understood is the 'excessive assimilatory sulfur metabolism' as observed in methanogens which perform a sulfur reduction which exceeds their anabolic needs without any apparent benefit. In heterotrophs, assimilation of elemental sulfur is seldom described and it is uncertain whether this process actually has a physiological significance.
Thus, reduction of thiosulfate and elemental sulfur is a common by incompletely understood feature among bacteria. These activities could give bacteria a selective advantage, but futher investigations are needed to clarify this possibility. Presence of thiosulfate, polythionates and sulfur reductase activities does not imply obligatorily that these activities play a role in thiosulfate, polythionates or sulfur assimilation as these compounds could be merely intermediates in bacterial metabolism. The possibility also exists that the assimilation of these sulfur compounds is just a side effect of an enzymatic activity with a completely different function.  相似文献

胆固醇氧化酶是胆固醇代谢过程中的关键酶,临床上用胆固醇氧化酶作为检测血清胆固醇含量的应用潜力巨大。将来源于红球菌Rhodococcus ruber的胆固醇氧化酶Ch OG,分别转化到大肠杆菌宿主BL21(DE3)和Rosetta(DE3)中,在不同条件下进行诱导表达,结果表明:BL21(DE3)菌株在诱导温度为16℃、IPTG浓度为0.1 mmol/L时,Ch OG可溶性表达量最高(0.49 mg/ml)。Ch OG的最适反应温度为30℃,最适反应p H为7.5。最适反应条件下,酶活性达到8.0 U/mg。利用TLC、HPLC对Ch OG催化产物胆甾-4-烯-3-酮进行了鉴定分析。三维结构及定点突变分析表明Glu406及Arg408、Glu261在进行胆固醇C3羟基的脱氢、质子传递,以及底物异构化方面发挥着重要作用。  相似文献

The net uptake, distribution and assimilation of NO 3 were studied in pea plants subjected to either long-term continuous Cd treatment for 10 d (10 or 50 M Cd) or short-term treatment (72 h) with 50 M Cd. In the latter treatment, the effects of transferring the plants to a Cd-free nutrient solution for a 'recovery period' of 96 h were also studied. All these treatments were compared with 'controls', plants which received no Cd. In both experiments, the reduction in fresh weight was associated with a decrease in the content (%) of shoot and root water and in transpiration rate as Cd concentration increased. The concentration of 3 in the shoots and sap decreased dramatically and net 3 uptake was severely inhibited, effects associated with a loss of shoot nitrate reductase (NR) activity. In the short-term Cd treatment, net 3 uptake was almost completely inhibited after 24 h, but recovered after the transfer of plants to a Cd-free nutrient solution. Similarly, a dramatic decrease in the shoot NR activity was observed. The uptake, distribution and tissue partitioning of K was also studied, which is considered to be the major counterion of 3 . Potassium uptake was similarly affected by Cd, as inferred from the ratio 3 /K uptake, which was ca. 10. The ratio K/ 3 tissue content increased in the shoot concomitantly to Cd in both long-term and short-term metal supply. These parameters showed a tendency of K similar to that observed for 3 , although its relative tissue distribution was not affected by Cd.  相似文献

ABSTRACT

Can the negative aspects of incorporation – discrimination, marginalization, and frustration with life in a country of settlement – help us to understand next generation transnationalism? This question is applied to a small, non-representative sample of next generation individuals involved in Mexican and Salvadoran transnational political and philanthropic organizations operating in California and Washington, DC. The findings suggest that negative incorporation had some explanatory potential for the transnational mobilization of some respondents. However, the study also suggests the multiple trajectories and contexts that give rise to next generation transnationalism. Involvement in cross-border organizations appears not only to be a refuge for those who perceive the country of settlement negatively or a means through which individuals can respond to negative experiences; it can also be pursued by individuals that positively identify with the country of settlement and perceive its values favourably, indicating the need for a more synthetic understanding of this phenomenon.  相似文献

While white ethnics and immigrants of colour have been studied in terms of their attempts to assimilate into the American mainstream, sociologists assume that ongoing racial discrimination obviates the need for an extensive examination of the actual assimilation trajectories of middle-class blacks. Many middle-class blacks travel from the black to the white world rather than existing exclusively in one racially distinct environment. Yet, we do not fully understand how middle-class blacks conceptualize their own integration into American society. Drawing on data collected through in-depth interviews with middle-class blacks and ethnographic research in a white and a black suburb, I establish the link between an affinity for black spaces and the alternative assimilation trajectories of middle-class blacks. I find that middle-class blacks engage in a variant of segmented assimilation, privileging the black world as a site for socializing even if they live in a white suburb. This selective pattern of assimilation, what I term strategic assimilation, suggests that this population of middle-class blacks does not perceive itself as permanently constrained to the bottom rung of a racial hierarchy.  相似文献

Using longitudinal data, this paper analyses the effect of different forms of social capital on the likelihood of employment and the occupational status of first generation immigrant men in Germany. This allows me to examine to what extent social capital of the bonding and the bridging types yield different returns. The study considers how contacts with natives, co-ethnic ties and family-based social capital are beneficial to the economic position of immigrant men. Random effects and fixed effects models show that strong inter-ethnic ties are beneficial both for employment and occupational status. There is no effect of co-ethnic ties and family-based social capital. It is concluded that, when using panel data, bridging social capital contributes to a better economic position and bonding social capital does not.  相似文献

Previous results in our laboratory indicated that a reduced Mn concentration in the leaves of barley was highly correlated with the reduced relative growth and net assimilation rates of salt-stressed plants. If Mn deficiency limits the growth of salt-stressed barley, then increasing leaf Mn concentrations should increase growth. In the present study, the effect of supplemental Mn on the growth of salt-stressed barley ( Hordeum vulgare L. cv. CM 72) was tested to determine if a salinity-induced Mn deficiency was limiting growth. Plants were salinized with 125 mol m−3 NaCl and 9.6 mol m−3 CaCl2. Supplemental Mn was applied in 2 ways: 1) by increasing the Mn concentration in the solution culture and 2) by spraying Mn solutions directly onto the leaves. Growth was markedly inhibited at this salinity level. Dry matter production was increased 100% in salt-stressed plants treated with supplemental Mn to about 32% of the level of nonsalinized controls. The optimum solution culture concentration was 2.0 mmol m−3, and the optimum concentration applied to the leaves was 5.0 mol m−3. Supplemental Mn did not affect the growth of control plants. Further experiments showed that supplemental Mn increased Mn concentrations and uptake to the shoot. Supplemental Mn increased the relative growth rate of salt-stressed plants and this increase was attributed to an increase in the net assimilation rate; there were no significant effects on the leaf area ratio. Supplemental Mn also increased the net photosynthetic rate of salt-stressed plants. The data support the hypothesis that salinity induced a Mn deficiency in the shoot, which partially reduced photosynthetic rates and growth.  相似文献

Glutathione (gamma-glu-cys-gly; GSH) is usually present at high concentrations in most living cells, being the major reservoir of non-protein reduced sulfur. Because of its unique redox and nucleophilic properties, GSH serves in bio-reductive reactions as an important line of defense against reactive oxygen species, xenobiotics and heavy metals. GSH is synthesized from its constituent amino acids by two ATP-dependent reactions catalyzed by gamma-glutamylcysteine synthetase and glutathione synthetase. In yeast, these enzymes are found in the cytosol, whereas in plants they are located in the cytosol and chloroplast. In protists, their location is not well established. In turn, the sulfur assimilation pathway, which leads to cysteine biosynthesis, involves high and low affinity sulfate transporters, and the enzymes ATP sulfurylase, APS kinase, PAPS reductase or APS reductase, sulfite reductase, serine acetyl transferase, O-acetylserine/O-acetylhomoserine sulfhydrylase and, in some organisms, also cystathionine beta-synthase and cystathionine gamma-lyase. The biochemical and genetic regulation of these pathways is affected by oxidative stress, sulfur deficiency and heavy metal exposure. Cells cope with heavy metal stress using different mechanisms, such as complexation and compartmentation. One of these mechanisms in some yeast, plants and protists is the enhanced synthesis of the heavy metal-chelating molecules GSH and phytochelatins, which are formed from GSH by phytochelatin synthase (PCS) in a heavy metal-dependent reaction; Cd(2+) is the most potent activator of PCS. In this work, we review the biochemical and genetic mechanisms involved in the regulation of sulfate assimilation-reduction and GSH metabolism when yeast, plants and protists are challenged by Cd(2+).  相似文献
