Intracellular potentials were measured in beetroot tissue during the steady-state uptake of K+ from various solutions. In solutions containing bicarbonate, the membrane potential becomes up to 70 mv more negative than the estimated equilibrium potential for K+. The uptake of K+ from such solutions is correlated with variations in the potential, both when the bicarbonate concentration is changed and also when the metabolic activity of the tissue is changed by washing in water for various periods. However, the estimated permeability to K+ varies from 0.4 x 10-7 to 1.5 x 10-7 cm·sec-1. It is postulated that the change of potential arises from the metabolic transport of HCO3- into the cell or H+ outwards, and that the associated uptake of K+ is partly or entirely by passive diffusion across the cell membrane. In contrast, K+ uptake from KCl solutions is not accompanied by any significant change in the membrane potential, which remains relatively close to the K+ equilibrium potential. In solutions containing both KHCO3 and KCl, it appears that an amount of K+ equal to the influx of Cl- is taken up independently of the potential, while the component of K+ uptake which is not balanced by Cl- uptake is related to the potential in the manner described. These results suggest that K+ uptake is linked to Cl- uptake in an electrically neutral active transport process.  相似文献

The influence of NO3 uptake and reduction on ionic balance in barley seedlings (Hordeum vulgare, cv. Compana) was studied. KNO3 and KCl treatment solutions were used for comparison of cation and anion uptake. The rate of Cl uptake was more rapid than the rate of NO3 uptake during the first 2 to 4 hours of treatment. There was an acceleration in rate of NO3 uptake after 4 hours resulting in a sustained rate of NO3 uptake which exceeded the rate of Cl uptake. The initial (2 to 4 hours) rate of K+ uptake appeared to be independent of the rate of anion uptake. After 4 hours the rate of K+ uptake was greater with the KNO3 treatment than with the KCl treatment, and the solution pH, cell sap pH, and organic acid levels with KNO3 increased, relative to those with the KCl treatment. When absorption experiments were conducted in darkness, K+ uptake from KNO3 did not exceed K+ uptake from KCl. We suggest that the greater uptake and accumulation of K+ in NO3-treated plants resulted from (a) a more rapid, sustained uptake and transport of NO3 providing a mobile counteranion for K+ transport, and (b) the synthesis of organic acids in response to NO3 reduction increasing the capacity for K+ accumulation by providing a source of nondiffusible organic anions.  相似文献

The uptake of Na+ and K+ by barley seedlings grown on aerated or non-aerated solutions was studied. Plants growing in culture solution took up K+ with high selectivity whether the solution was aerated or not. Roots of plants grown on aerated CaSO4 and transferred to a solution of KCl and NaCl had a lower preference for K+ than roots of plants grown on non-aerated CaSO4. Both kinds of low-salt roots were much less able to discriminate between K+ and Na+ than high-salt roots grown on a culture solution. The different levels of K+ selectivity are suggested to be related to H+ release from the tissue.  相似文献

1. When yeast oxidizes propan-2-ol in the presence of KCl no uptake of K+ occurs. 2. When propionate is added to suspensions containing propan-2-ol, or if the suspensions are bubbled with CO2, a considerable uptake of K+ occurs. 3. Maximum K+ uptake occurs at a propionate concentration of 2mm. 4. The addition of 20mm-propionate to the suspension lowers the intracellular pH of the yeast from a resting value in the region of 6.2 to approx. 5.6. 5. When K+ uptake is measured in the presence of 20mm-propionate, progressive changes in the rate of K+ uptake and intracellular pH occur. The optimum rate of K+ uptake occurs at an intracellular pH of 5.70. 6. The effect of both intra- and extra-cellular pH on K+–K+ exchange was studied and an optimum rate was found at an extracellular pH of 5.35, the corresponding intracellular pH being 6.44. 7. When a Na+-loaded yeast oxidizes propan-2-ol in the presence of KCl, a steady efflux of Na+ and influx of K+ occurs. The addition of 10mm-propionate to the suspension markedly inhibited the Na+ efflux but only slightly decreased the K+ influx. 8. The effect of both extra- and intra-cellular pH on Na+ efflux was studied with propan-2-ol and with glucose. The results can be best interpreted in terms of intracellular pH changes, and an optimum was obtained at approx. pH6.40.  相似文献

The effect of various ions on stomatal opening was studied in isolated epidermal strips of Vicia faba L. Stomata in strips floating on 10 mm KCl and in CO2-free air opened in light, closed in subsequent darkness, then opened fully again when illuminated. A light-activated highly specific effect of K- (and Rb+) on opening was found. When strips were floated on high concentrations (50 or 100 meq/liter) of Li+, Na+ or Cs+, stomata opened but light had very little effect on the concentrations required for opening. With K+, the opening produced in the dark was the same as with the other alkali ions. Light, however, lowered more than 100-fold the concentration of K+ required for maximal opening. Thus only the effect of K+ (and Rb+) was greatly accentuated by light. NH4+ and Mg2+ did not produce opening.  相似文献

Earlier studies have demonstrated close inverse relationships between Rb+ concentrations in plants and pH or base (including K+) saturation of soils. This study aims at elucidating conditions in soils influencing plant uptake of Rb+. Growth experiments with Carex pilulifera L. were performed, modifying the acidity and K+ supply of acid soils and solutions. We were unable to assess any reduction in Rb+ uptake by adding precipitated CaCO3 to acid soil unless pH was raised to near neutrality. Though not fully compensating the loss of soil solution K+and exchangeable K+ from uptake by the growing plants, soil treated with 0.5 mM K+ (as KCl) reduced the Rb+ concentration in the shoots by 40% without measurably changing soil pH. Experiments varying the pH and K+ concentration of a nutrient solution (20% Hoagland), spiked with 6 uM Rb+, clearly demonstrated that plant uptake of Rb+ and K+ was unaffected by acidity in the pH range 3.6–5.0 tested, whereas Rb+ uptake was reduced by ca. 50%, when K+ concentration was increased from 1.2 to 3.6 mM. The sensitivity of this reaction indicates that shortage or low availability of K+ controls Rb+ uptake from acid soils, being probably more important than soil acidity per se. Secondary effects of high soil acidity, such as leaching losses of K+, might also be of importance in accounting for the high uptake of Rb+ from such soils. It is suggested that leaf analysis of Rb+ may be used as a method to assess early stages of K+ deficiency in plants on acid soils.  相似文献

The influx of K+ from 86Rb-labeled solutions in the concentration range 0.008 to 0.2 mm into roots of intact plants and excised roots of barley plants (Hordeum vulgare [L.]) previously grown in 5 mm CaSO4 (low K+ roots) or 0.5 mm CaSO4 plus 5 mm KCl (high K+ roots) was measured. A consistent observation of these experiments was a substantial reduction of influx (usually by about 50%) following excision. The possible leakage of K+ into the medium and subsequent dilution of specific activity of labeled solutions was eliminated as an explanation for influx reduction in excised low K+ roots. Reduction of transpirational rates was also without effect upon influx into low K+ roots. Excision followed by 2 hours aging in 0.5 mm CaSO4 solution revealed that influx values recovered within the 2 hours to the values obtained in intact roots. It is concluded that much of the literature which describes the enhancement of ion uptake following excision actually describes excision damage followed by recovery.  相似文献

Mitochondrial volume regulation depends on K+ movement across the inner membrane and a mitochondrial Ca2+-dependent K+ channel (mitoKCa) reportedly contributes to mitochondrial K+ uniporter activity. Here we utilize a novel KCa channel activator, NS11021, to examine the role of mitoKCa in regulating mitochondrial function by measuring K+ flux, membrane potential (ΔΨm), light scattering, and respiration in guinea pig heart mitochondria. K+ uptake and the influence of anions were assessed in mitochondria loaded with the K+ sensor PBFI by adding either the chloride (KCl), acetate (KAc), or phosphate (KH2PO4) salts of K+ to energized mitochondria in a sucrose-based medium. K+ fluxes saturated at ∼ 10 mM for each salt, attaining maximal rates of 172 ± 17, 54 ± 2.4, and 33 ± 3.8 nmol K+/min/mg in KCl, KAc, or KH2PO4, respectively. NS11021 (50 nM) increased the maximal K+ uptake rate by 2.5-fold in the presence of KH2PO4 or KAc and increased mitochondrial volume, with little effect on ΔΨm. In KCl, NS11021 increased K+ uptake by only 30% and did not increase volume. The effects of NS11021 on K+ uptake were inhibited by the KCa toxins charybdotoxin (200 nM) or paxilline (1 μM). Fifty nanomolar of NS11021 increased the mitochondrial respiratory control ratio (RCR) in KH2PO4, but not in KCl; however, above 1 μM, NS11021 decreased RCR and depolarized ΔΨm. A control compound lacking KCa activator properties did not increase K+ uptake or volume but had similar nonspecific (toxin-insensitive) effects at high concentrations. The results indicate that activating K+ flux through mitoKCa mediates a beneficial effect on energetics that depends on mitochondrial swelling with maintained ΔΨm.  相似文献

Photosynthesis, stroma-pH, and internal K+ and Cl concentrations of isolated intact chloroplasts from Spinacia oleracea, as well as ion (K+, H+, Cl) movements across the envelope, were measured over a wide range of external KCl concentrations (1-100 millimolar).

Isolated intact chloroplasts are a Donnan system which accumulates cations (K+ or added Tetraphenylphosphonium+) and excludes anions (Cl) at low ionic strength of the medium. The internally negative dark potential becomes still more negative in the light as estimated by Tetraphenylphosphonium+ distribution. At 100 millimolar external KCl, potentials both in the light and in the dark and also the light-induced uptake of K+ or Na+ and the release of protons all become very small. Light-induced K+ uptake is not abolished by valinomycin suggesting that the K+ uptake is not primarily active. Intact chloroplasts contain higher K+ concentrations (112-157 millimolar) than chloroplasts isolated in standard media. Photosynthetic activity of intact chloroplasts is higher at 100 millimolar external KCl than at 5 to 25 millimolar. The pH optimum of CO2 fixation at high K+ concentrations is broadened towards low pH values. This can be correlated with the observation that high external KCl concentrations at a constant pH of the suspending medium produce an increase of stroma-pH both in the light and in the dark. These results demonstrate a requirement of high external concentrations of monovalent cations for CO2 fixation in intact chloroplasts.

The role of cation and anion uptake in salt-stimulated growth of light-grown, GA3-treated lettuce (Lactuca sativa L.) hypocotyl sections was investigated. Potassium chloride (10 mm) causes a 2-fold increase in the growth rate of GA3-treated hypocotyl sections without affecting the growth rate of sections incubated in the absence of GA3. Salt uptake is the same in both treatments, and furthermore the uptake of cation and anion is stoichiometric during the first 24 hours under all incubation conditions. The importance of the anion for cation uptake is demonstrated in experiments with benzenesulfonate and iminodiacetate2−. When K+ and Na+ are supplied only as the benzenesulfonate and iminodiacetate salts, growth and cation uptake are markedly reduced compared to KCl and NaCl. Calculation of the osmotic potential of salt-treated sections based on measurement of K+ and Cl uptake suggests that the observed increase in tissue osmolality is a result of salt uptake. Similarly, uptake of ions can account for the shift in water potential when sections are incubated in 10 mm KCl. We conclude that the change in growth rate of light-grown, GA3-treated sections caused by the addition of KCl or NaCl to the incubation medium results solely from decreased water potential of the tissue due to ion uptake.  相似文献

The effect of external inorganic nitrogen and K+ content on K+ uptake from low-K+ solutions and plasma membrane (PM) H+-ATPase activity of sorghum roots was studied. Plants were grown for 15 days in full-nutrient solutions containing 0.2 or 1.4 mM K+ and inorganic nitrogen as NO3-, NO3-/NH4+ or NH4+ and then starved of K+ for 24, 48 and 72 h. NH4+ in full nutrient solution significantly affected the uptake efficiency and accumulation of K+, and this effect was less pronounced at the high K+ concentration. In contrast, the translocation rate of K+ to the shoot was not altered. Depletion assays showed that plants grown with NH4+ more efficiently depleted the external K+ and reached higher initial rates of low-K+ uptake than plants grown with NO3-. One possible influence of K+ content of shoot, but not of roots, on K+ uptake was evidenced. Enhanced K+-uptake capacity was correlated with the induction of H+ extrusion by PM H+-ATPase. In plants grown in high K+ solutions, the increase in the active H+ gradient was associated with an increase of the PM H+-ATPase protein concentration. In contrast, in plants grown in solutions containing 0.2 mM K+, only the initial rate of H+-pumping and ATP hydrolysis were affected. Under these conditions, two specific isoforms of PM H+-ATPase were detected, independent of the nitrogen source and deficiency period. No change in enzyme activity was observed in NO3--grown plants. The results suggest that K+ homeostasis in NH4+-grown sorghum plants may be regulated by a high capacity for K+ uptake, which is dependent upon the H+-pumping activity of PM H+-ATPase.  相似文献

Potassium ions at low concentrations stimulate cytokinin-dependent betacyanin synthesis in Amaranthus tricolor seedlings more than other alkali metal ions when tested as the chloride salts. The sequence of relative stimulation is K+ > Rb+ > (Na+ = Li+). Calcium and Mg2+ ions are inhibitory at concentrations > 1 millimolar when tested as chlorides. Anions also have an effect on the degree of alkali metal stimulation in the order PO43− > NO3 > Cl. The high activity of phosphate may be partly due to its chelating effect on inhibitory Ca2+ ions, or to effects on K+ uptake. A mixture of Na+ and K+ in the presence of phosphate is more effective than either cation alone. This result may be due either to effects on tyrosine transport or on the potassium uptake system. Phytochrome-dependent betacyanin synthesis shows the same stimulation by Na+ plus K+. The effect of a number of inhibitors of transport systems on betacyanin accumulation is reported. The possible role of the ionic environment of cells in their metabolic regulation is discussed, particularly in relation to cytokinin action.  相似文献

The commercial tomato, lycopersicon esculentum Mill. cv Walter, and its wild relative, Lycopersicon cheesmanii ssp. minor (Hook.) C.H. Mull., were grown for 30 days under controlled conditions and in solution culture varying in its content of Na+ and K+. Subsequently, 86Rb-labeled K+ uptake and distribution were studied. From all solutions, `Walter' consistently absorbed 86Rb-labeled K+ at a higher rate in micromoles per gram fresh weight per 30 minutes than L. cheesmanii. L. cheesmanii distributed a greater proportion of the absorbed K+ from its root to its shoot. When 0.6 millimolar NaNO3 replaced 0.6 millimolar KNO3 in the pretreatment solution, intact plants of both genotypes followed a similar pattern as when they were pretreated with K+ only, but a greater percentage of the absorbed K+ remained in the roots. Leaf slices of L. cheesmanii plants deprived of K+ for 6 days showed a greater rate of K+ uptake than did slices from `Walter' plants pretreated the same way. Stem slices of L. cheesmanii, however, had a lower uptake rate than did those of `Walter'. Both leaf and stem slices of `Walter' plants, pretreated 6 days with 0.6 millimolar NaNO3 substituting for 0.6 millimolar KNO3 in their growth medium, had greater rates of 86Rb-labeled K+ uptake from 0.5 and 20 millimolar KCl solutions than did slices of L. cheesmanii. These marked differences in patterns of ion uptake and translocation indicate that these genotypes of tomato have evolved different mechanisms to deal with K+ and Na+ in their environments.  相似文献

The effect of addition of Ca salts on accumulation of K from 5 mN KCl or K2SO4 solutions was found to depend on whether Ca was added as Cl or SO4 salt. Chloride as well as K uptake was increased when Ca and Cl concentrations in culture solutions were increased. Pre-treatment of roots with CaCl2 stimulated subsequent K uptake from K2SO4 solutions as compared to pre-treatment with distilled water but pre-treatment with CaSO4 did not. The results indicate that addition of Ca salts to KCl or K2SO4 solutions increased anion uptake and the effect of the addition of the Ca salts on K uptake was in part the result of increased anion uptake and not entirely a direct effect of Ca.  相似文献

The effect of uncouplers and diffusible acids on K+ transport was studied in yeast.Although the K+ transport system seems to depend on ATP to function, the effects of uncouplers are not due primarily to its action on the energy conserving systems of the cell.Other uncouplers with different structures to that of DNP showed also an inhibitory effect on K+ transport, which agrees with their reported ability to conduct protons through membranes.Uncouplers, besides inhibiting K+ uptake, produce an efflux of this cation; however, the rate of efflux produced is quantitatively important only when the cells have previously taken up the cation; there seems to exist a mechanism which prevents the loss of cations by yeast.In the absence of substrate, at pH 8.5, with 0.5 m KCl, TCS produces the efflux of H+, and when 86Rb+ was used as a substitute for K+, an increase of the entrance of the cation could be detected in the presence of the uncoupler. It seems that the effect of the uncoupler depends on the direction of the combined H+ and K+ gradients, or the electrochemical potential of the cell.As reported by other authors, weak diffusible acids increase the uptake of K+ by yeast, and this effect is not due to changes in the metabolism, but to the magnitude of the entrance of the molecules to the yeast cell.It was found that the efflux of the acids (H2CO3), on the other hand, can produce an efflux of K+, which means that anions are important not only for the entrance of the cations, but for its permanence within the cell as well.The data seem to be in agreement with the hypothesis of the existence of a proton pump, responsible for the creation of an electrochemical potential, involved in K+ transport. At low pH, this pump seems to be activated by the transport of K+ into the cell.  相似文献

Summary Detopped root systems of Ricinus communis plants were used for the study of the effects of temperature and DNP on the uptake of K and Na ions supplied as KNO3 and NaNO3.When K and Na ions were offered together in equivalent concentrations, the steady state uptake rates for K+ and Na+ at 23 to 25° gave a K+/Na+ ratio of 3. Increasing the Na+ concentration relative to K+ 3-fold did not alter the preferential uptake of K+. The uptake of K+ was more sensitive to temperature in the range 10 to 40° and to the application of DNP at 1.5x10-4 M than was the uptake of Na+. When NaNO3 was the only salt supplied Na+ uptake became more sensitive to DNP than when both K+ and Na+ nitrates were supplied. Prolonged application of DNP led to net K+ efflux from the roots, even when no K+ was being supplied to the roots. Net Na+ efflux under the influence of DNP occurred only in roots previously grown on Na-containing nutrient medium.The different responses of the K+ and Na+ uptake processes to temperature and DNP suggest the operation of different uptake mechanisms for K+ and Na+ These results have been considered in relation to the recent concept of dual mechanisms for the absorption of alkali cations by plant tissues.  相似文献

The effects of K fertiliser (160 kg ha-1) applied with Cl- or SO4 2- as the accompanying anion on the K nutrition of kiwifruit (Actinidia deliciosa var. deliciosa) were assessed in a field experiment, using vines with varying degrees of K deficiency. Leaf K concentrations in spring were significantly higher for vines receiving KCl, compared to those receiving K2SO4. This effect did not interact significantly with the degree of K deficiency, and persisted for about 6 weeks. Subsequently there was no significant difference between the leaf K concentrations for the vines receiving KCl or K2SO4. Applying K as KCl increased the leaf Cl concentration, especially in spring, while applying K as K2SO4 had no significant effect on the leaf S concentration at that time. These results implied a greater requirement for organic acid anions for K+ uptake from K2SO4 than from KCl, and the importance of organic acid anions for K+ uptake from different sources of K fertiliser is discussed. This transient effect of the accompanying anion on leaf K status was associated with large effects on flowering, and fruit yields were about 28% higher for plants receiving KCl rather than K2SO4.The effects on growth and tissue nutrient composition of varying the concentrations of Cl-, NO3 -, SO4 2- and H2PO4 - around the roots of kiwifruit vines were examined in a solution culture experiment. For H2PO4 -, plant growth was very similar over a wide range of rates of addition. For the other anions, the range between deficiency and toxicity was clearly delineated. For Cl- and NO3 -, toxicity was associated with high tissue concentrations of Cl and N, respectively, and was consistent with competition for uptake between Cl- and NO3 -. However, for SO4 2-, toxicity was associated with only a small increase in the tissue S concentration relative to that associated with maximum growth, and appeared to result more from effects on uptake of other anions and cations rather than from direct effects of high tissue S concentrations.It is concluded that the sensitivity of kiwifruit to the anion accompanying K+ in fertiliser may be related to the unusually high requirement for Cl previously reported for this species.  相似文献

The effect of potassium ions on succinic dehydrogenase activity of mitochondria was studied. The results showed that in these organelles K+ induces inhibition of the respiratory control; moreover, in submitochondrial particles potassium inhibits the rate of oxidation of succinate. The results showed also that K+ does not changes theK m for succinate but diminishes theV max. In addition, the data provide evidence that mitochondria oxidizing glutamatemalate in a sucrose medium show a higher activity of succinate dehydrogenase than mitrochondria incubated in KCl.  相似文献

The uptake of glycine,l-aspartic acid andl-lysine at pH 5 was not systematically affected by the presence of up to 0.1m NaCl or KCl, without or with 0.3mm actidione. The observed effects, either positively or negatively altering the rate of amino acid uptake, depended on the concentration of the ion but were always qualitatively the same for Na+ and K+. The inhibition prevailing with 0.1m ions was significantly more pronounced with Na+ than with K+. The presence of other anions (phosphate, nitrate, sulphate) had a slight but significant effect on uptake as compared with the variant using chloride. It is concluded that inSaccharomyces cerevisiae the transport of amino acids is not essentially coupled with the transport of different ions.  相似文献

Summary In order to investigate the question whether ammonium reabsorption in the thick ascending limb of Henle's loop (TALH) proceeds via the Na+,K+,Cl-cotransporter, plasma membrane vesicles were prepared from TALH cells isolated from rabbit kidney outer medulla and the effect of NH 4 + on their transport properties was investigated. It was found that, in the presence of a 78-mmol/liter NaCl gradient, 5 mmol/liter NH 4 + inhibited bumetanide-sensitive rubidium flux by 86%; a similar decrease was observed for 5 mmol/liter, K+. Inhibition of bumetanide-sensitive rubidium uptake by NH 4 + was competitive and an apparentK i of 1.9 mmol/liter was found Bumetanide-sensitive sodium uptake measured in the presence of a 83 mmol/liter KCl gradient was not inhibited by 5 mmol/liter NH 4 + . A 100-mmol/liter NH4Cl gradient was, however, capable of stimulating bumetanide-sensitive sodium uptake to the same extent as a KCl gradient. These data suggest that NH 4 + is accepted by the K+ site of the Na+,K+,Cl-cotransport system and that the transporter can function in a Na+, NH 4 + ,2Cl mode. Since the affinity of the transporter for NH 4 + lies in the concentration range found in the TALH lumen in vivo, it is concluded that Na+, NH 4 + 2Cl-cotransport can contribute to the NH 4 + reabsorption in this tubular segment.  相似文献
