Bacteria are known to be associated endophytically with plants. Research on endophytic bacteria has identified their importance in food safety, agricultural production and phytoremediation. However, the diversity of endophytic bacterial communities and the forces that shape their compositions in non-cultivated plants are largely uncharacterized. In this study, we explored the diversity, community structure, and dynamics of endophytic bacteria in different plant species in the Tallgrass Prairie Preserve of northern Oklahoma, USA. High throughput sequencing of amplified segments of bacterial rDNA from 81 samples collected at four sampling times from five plant species at four locations identified 335 distinct OTUs at 97% sequence similarity, representing 16 phyla. Proteobacteria was the dominant phylum in the communities, followed by the phyla Bacteriodetes and Actinobacteria. Bacteria from four classes of Proteobacteria were detected with Alphaproteobacteria as the dominant class. Analysis of molecular variance revealed that host plant species and collecting date had significant influences on the compositions of the leaf endophytic bacterial communities. The proportion of Alphaproteobacteria was much higher in the communities from Asclepias viridis than from other plant species and differed from month to month. The most dominant bacterial groups identified in LDA Effect Size analysis showed host-specific patterns, indicating mutual selection between host plants and endophytic bacteria and that leaf endophytic bacterial compositions were dynamic, varying with the host plant’s growing season in three distinct patterns. In summary, next generation sequencing has revealed variations in the taxonomic compositions of leaf endophytic bacterial communities dependent primarily on the nature of the plant host species.  相似文献

The leaves of fescue grasses are protected from herbivores by the production of loline alkaloids by the mutualist fungal endophytes Neotyphodium sp. or Epichloë sp. Most bacteria that reside on the leaf surface of such grasses can consume these defensive chemicals. Loline-consuming bacteria are rare on the leaves of other plant species. Several bacterial species including Burkholderia ambifaria recovered from tall fescue could use N-formyl loline as a sole carbon and nitrogen source in culture and achieved population sizes that were about eightfold higher when inoculated onto plants harboring loline-producing fungal endophytes than on plants lacking such endophytes or which were colonized by fungal variants incapable of loline production. In contrast, mutants of B. ambifaria and other bacterial species incapable of loline catabolism achieved similarly low population sizes on tall fescue colonized by loline-producing Neotyphodium sp. and on plants lacking this endophytic fungus. Lolines that are released onto the surface of plants benefiting from a fungal mutualism thus appear to be a major resource that can be exploited by epiphytic bacteria, thereby driving the establishment of a characteristic bacterial community on such plants.  相似文献

Fast-growing, aerobic, heterotrophic bacteria from the root surface of young sugar beet plants were inventoried. Isolation of the most abundant bacteria from the root surface of each of 1,100 plants between the second and tenth leaf stage yielded 5,600 isolates. These plants originated from different fields in Belgium and Spain. All isolates were characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis of total cellular proteins. Comparison of protein fingerprints allowed us to inventory the bacteria of individual plants of different fields or leaf stages and to analyze the composition and variability of the rhizobacterial population of young sugar beet plants. Each field harbored a specific population of bacteria which showed a highly hierarchic structure. A small number of bacteria occurring frequently at high densities dominated in each field. The major bacteria were identified as Pseudomonas fluorescens, Xanthomonas maltophilia, Pseudomonas paucimobilis, and Phyllobacterium sp. The former three species showed a high genetic variability as they were represented by different protein fingerprint types on the same or different fields or leaf stages. Twinspan analysis and relative abundance plots showed that the structure and composition of the bacterial populations varied strongly over time. The phyllosphere is one of the largest habitats for terrestrial microorganisms. To gain a better insight into the factors underlying the composition of bacterial communities inhabiting leaf surfaces we performed culture-dependent and independent (Denaturing Gradient Gel Electrophoresis) analyses on the bacteria associated with the leaves of three plant species: Amygdalus communis, Citrus paradisi, and Nicotiana glauca. We found that the culturable classes Bacilli and Actinobacteria were the predominant classes on the phyllosphere of all three plant species. In contrast to this consistency on the bacterial class level, we found a significant variation on the bacterial species-level based on the culturable methods. Although some variation was detected among individual plants within one plant species, the inter-specific variability exceeded the intra-specific variability. C. paradisi leaf surface had the highest predicted total species richness (Chao 2 and ICE) and the highest species diversity (βw) among the three plant species. Our findings demonstrate that environmental conditions, mainly the plant species within a site, govern the bacterial community composition on leaf surfaces.  相似文献

Five native species of the flora of Egypt: Adonis dentata Delile, Ranunculaceae; Papaver dubium L., Papaveraceae; Matthiola longipetala (Vent.) DC., Brassicaceae; Malva parviflora L., Malvaceae; and Erodium laciniatum (Cav.) Willd., Geraniaceae have been investigated morphologically, palynologically and mycologically. The total counts of phyllosphere fungi fluctuated between 223 and 388 colonies/100 leaf segments giving maximum on Malva parviflora followed by Matthiola longipetala, Erodium laciniatum, Papaver dubium and Adonis dentata. The widest spectrum of genera (20) and species (39 and l variety) was recorded on Malva parviflora and the narrowest on Adonis dentata (9, 14). Sixty species and one variety belonging to 27 genera were collected from leaves (24 genera and 56 species and 1 variety) and anthers/pollen (16 genera and 27 species) of the five studied plants with the most frequently species were some members of Alternaria, Cladosporium, Fusarium and Mucor. Members of Aspergillus and Penicillium were prevalent only on leaves of Matthiola longipetala and Malva parviflora, while these members were infrequently recorded on leaf surfaces of Adonis dentata and Papaver dubium. Results revealed that density of hairs on the leaf surface and size of leaf blade, almost seem to be the most reliable factors of the biodiversity of the fungal spores on the studied taxa. On the other hand, reticulate or striate pollen surface had a wider spectrum of fungal biodiversity and higher densities of spores compared with echinate and scabrate surface.  相似文献

Of the 20 plant species of hydrophytes screened for Arbuscular Mycorrhizal (AM) fungal root colonization, 10 aquatic plants (out of 14 species) and five marshy plants (out of six species) were found to be mycorrhizal, while the remaining species were non mycorrhizal. Vesicular colonization occurred in 12 plant species while arbuscular colonization was restricted to only three plant species. A rooted submerged pteridophyte viz., Isoetes coromandelina L. was found to be mycorrhizal exhibiting vesicular colonization. In all, two genera viz., Glomus and Scutellospora, the former being dominant, were recorded. The most common AM fungal species Glomus claroideum was recovered from 14 plant species.  相似文献

We have isolated 576 endophytic bacteria from the leaves, stems, and roots of 10 rice cultivars and identified 12 of them as diazotrophic bacteria using a specific primer set of nif gene. Through 16S rDNA sequence analysis, nifH genes were confirmed in the two species of Penibacillus, three species of Microbacterium, three Bacillus species, and four species of Klebsiella. Rice seeds treated with these plant growth-promoting bacteria (PGPB) showed improved plant growth, increased height and dry weight and antagonistic effects against fungal pathogens. In addition, auxin and siderophore producing ability, and phosphate solubilizing activity were studied for the possible mechanisms of plant growth promotion. Among 12 isolates tested, 10 strains have shown higher auxin producing activity, 6 isolates were confirmed as strains with high siderophore producing activity while 4 isolates turned out to have high phosphate-solubilizing activity. These results strongly suggest that the endophytic diazotrophic bacteria characterized in this study could be successfully used to promote plant growth and inducing fungal resistance in plants.  相似文献

A total of 121 seed samples of cabbage, cauliflower, cress, radish and turnip collected from five localities in Upper Egypt were assayed for their fungal flora. The highest count of fungi was recorded on cabbage seeds (75%), whereas the lowest count was observed on turnip seeds (33%). Thirty-five fungal species and two varieties belonging to 16 genera were identified. The broadest number of species (22 species + 1 variety) were isolated from cress seeds. However, only five species,viz. Aspergillus flavus, A. niger, Penicillium chrysogenum, P. funiculosum andRhizopus stolonifer, were found to be associated with seeds of the five plants. The relative efficacy of aqueous seed extracts against these five fungal species was tested. The antifungal drug Trosyd (tioconazole) was taken as a standard inhibitor. The aqueous extract of cabbage seeds inhibited three fungal species, cress extract two and cauliflower and turnip one species; radish seed extract was ineffective.  相似文献

Endophytes play an important role in protection of host plants from infection by phytopathogens. Endophytic bacteria were isolated from five different parts (root, stem, petiole, leaf and seed) of Panax notoginseng and evaluated for antagonistic activity against Fusarium oxysporum, Ralstonia sp. and Meloidogyne hapla, three major pathogens associated with root-rot disease complex of P. notoginseng. From 1000 endophytic bacterial strains evaluated in vitro, 104 strains exhibited antagonistic properties against at least one of these three pathogens. Phylogenetic analyses of their 16S rRNA gene sequences showed that these 104 antagonistic bacteria belong to four clusters: Firmicutes, Proteobacteria, Actinobacteria and Bacteroidetes/Chlorobi. Members of the Firmicutes, in particular the Bacillus spp., were predominant in all analyzed tissues. The root was the main reservoir for antagonistic bacteria. Of the 104 antagonists, 51 strains showed antagonistic activities to one pathogen only, while 43 and 10 displayed the activities towards two and all three pathogens, respectively. The most dominant species in all tissues were Bacillus amyloliquefaciens subsp. plantarum and Bacillus methylotrophicus, which were represented by eight strains with broad antagonistic spectrum to the all three test pathogens of root-rot disease complex of P. notoginseng.  相似文献

The flora and fauna associated with the serrated wrack, Fucus serratus L., in Strangford Lough have been studied. Fucus plants were larger where they were most abundant and their distribution was determined largely by the availability of suitable substratum; however, plant size was reduced under turbulent conditions. The fauna contained 79 taxa, of which eleven were common: these comprised four polyzoans, two tunicates, two sponges, two hydroids and a serpulid. All except three were more abundant on plants with low silt loads in turbulent, weedy areas. All species except Electra pilosa (L.) tended to occur on the concave surfaces of the plant. Evidence of zonation in each species' distribution along the plants was obtained. Analysis of association coefficients for the commonest species suggested that there are two distinct species groupings within the community.  相似文献

Pink-pigmented facultative methylotrophic bacteria (PPFMs) have been found on the surfaces of leaves of most plants tested. We found PPFMs on the leaf surfaces of all 40 plants (38 species) tested and on soybean pods by pressing onto AMS medium with methanol as the sole carbon source. The abundance ranged from 0.5 colony forming unit (cfu) /cm2 to 69.4 cfu/cm2 on the leaf surfaces. PPFMs were found in homogenized leaf tissues of only 4 of the species after surface disinfestation with 1.05% sodium hypochlorite and were rarely found in cultures initiated from surface disinfested Datura innoxia leaves or inside surface disinfested soybean pods. Of 20 antibiotics tested for PPFM growth inhibition, rifampicin was the most effective and of seven others which also inhibited PPFM growth, cefotaxime should be the most useful due to the expected low plant cell toxicity. These antibiotics could be used in concert with common surface sterilization procedures to prevent the introduction or to eliminate PPFM bacteria in tissue cultures. Thus, while PPFMs are present on the surfaces of most plant tissues, surface disinfestation alone can effectively remove them so that uncontaminated tissue cultures can be initiated in most cases.  相似文献

A total of 1,897 isolates of endophytic fungi from Madhuca indica Gmel., representing 40 morphologically distinct fungal taxa were obtained from 2,700 segments of stem, bark and leaf from three different locations (Loc 1, Loc 2 and Loc 3) in Uttar Pradesh, India. Out of 40 taxa, 28 were identified microscopically and the remaining 12 by molecular methods. Coelomycetes (62.41 %) were the most prevalent fungal group followed by hyphomycetes (28.89 %) and ascomycetes (8.70 %). Colonisation frequency (CF) was greater in stem (82.55 %) than in leaf (65.00 %) and bark (63.22 %). Due to the dominance of a few taxa, species richness and Shannon and Simpson diversity indices were lower in stem than in leaf and bark at each location. Interestingly, less rainfall and lower temperatures disfavoured the overall colonisation of fungal endophytes at Loc 2. The stem samples from all locations were very similar in their endophytic composition, whereas bark and leaf samples showed differences. The dominant endophytic fungi isolated were Phomopsis sp. 1 (9.185 %), and Colletotrichum gloeosporioides (7.00 %). Principal component analysis showed 55 % tissue specificity with 51.08 % maximum variance. Antibacterial activity revealed that 58.33 % endophytic fungi were active against at least one or more bacterial pathogens, whereas the crude extract of five endophytic fungi inhibited the growth of five or more than five (50 %) of the pathogens tested. This report illustrates the value of having an adequate sample size from different tissues and different locations for species and chemical diversity in search of novel natural products.  相似文献

Infections by yeast strains of the genus Candida are among the most prevalent fungal infections of humans. These yeasts are common residents of the oral mucosa and other body surfaces. Since most yeast infections are due to endogenous strains and that species of Candida differ in virulence properties and in intrinsic susceptibilities to antifungal drugs, understanding the human commensal yeast flora can help designing effective treatment and prevention strategies against yeast infections. Here, we report the patterns of yeast species distributions in the oral cavities of 1,799 people from Hainan Island in southern China. Based on sequence information at the fungal barcode locus ITS regions, 368 of the 415 obtained oral yeast strains were identified as belonging to 26 yeast species, while the remaining 47 strains all showed significant sequence divergence to the currently described species. The four most common yeast species were C. albicans (42 %), C. tropicalis (20 %), C. glabrata (5.5 %), and C. parapsilosis (4.1 %) and 10 of the 26 yeast species were represented by only one strain each. Our analyses identified that the gender of hosts and ethnical background showed no contribution to oral yeast species distributions. However, the health status, place of birth, current residency, and the age of hosts all showed significant contributions to the distributions of the four dominant yeast species. We compared our results with those reported previously and discussed the potential mechanisms for the observed differences in oral yeast species distributions.  相似文献

【背景】植物内生菌(endophyte)对寄主植物的益生作用有利于植物的生存与扩散,而菌群互作网络为内生菌生态功能的实现提供了基础保障。【目的】了解影响西藏畜牧业发展的主要疯草冰川棘豆(Oxytropis glacialis)内生菌核心微生物组及其菌群互作网络,为青藏高原疯草类有毒植物的科学治理和利用提供基础参考依据。【方法】利用高通量测序技术分析冰川棘豆中内生菌核心微生物组,构建内生菌相关性网络,并以与苦马豆素代谢相关内生菌为例分析冰川棘豆内生菌的互作模式。【结果】内生细菌测序共得到175791条有效序列,注释到428个细菌OTU,分属于19个门和267个属;内生真菌测序共得到757 113条有效序列,注释到391个真菌OTU,分属于7个门和149个属。Venn图分析表明,根、叶组织的核心内生细菌菌属数目(54、62)大于核心内生真菌(22、13),根组织中的核心内生菌种类与叶组织相当(76、75)。系统发育树分析表明,冰川棘豆中存在产生苦马豆素内生真菌链格孢属(Alternaria)和降解苦马豆素的内生细菌短波单胞杆菌属(Brevundimonas)。相关性网络分析表明,内生菌菌群间以正向反馈的互作网络关系为主,核心菌群可能主要通过间接性的互作方式将影响传递到微生物群落,其中链格孢属与短波单胞杆菌属作为核心菌属通过间接性的显著相关关系(|ρ|0.6,P0.01)参与菌群间互作网络。【结论】冰川棘豆核心菌群以间接的方式参与内生菌菌群间的互作网络,高度的菌属连接性为内生菌生态功能的实现提供可能。  相似文献

The composition and diversity of fungal communities associated with three endangered orchid species, Hadrolaelia jongheana, Hoffmannseggella caulescens, and Hoffmannseggella cinnabarina, found in different vegetation formations of the Atlantic Forest were determined by constructing clone libraries and by applying diversity and richness indices. Our results demonstrated the presence of Basidiomycetes. Sebacinales (81.61 %) and Cantharellales (12.10 %) were the dominant orders and are potential candidates for orchid mycorrhizal fungi. The Ascomycetes identified included the Helotiales (29.31 %), Capnodiales (18.10 %), and Sordariales (10.34 %), among others. These orders may represent potentially endophytic fungi. A Shannon–Wiener diversity index (H′) analysis showed a relatively high fungal community diversity associated with these tropical orchids. This diversity may offer greater flexibility in terms of the adaptation of the plants to changing environmental conditions and the potential facilitation of reintroduction programs. The Simpson diversity index values showed that all of the libraries included dominant species, and a LIBSHUFF analysis showed that the fungal communities were structurally different from each other, suggesting an influence of local factors on this diversity. This study offers important information for the development of conservation strategies for threatened and endemic species of Brazilian flora in an important and threatened hotspot.  相似文献

The fungal colonization of the angiosperm root parasiteCynomorium coccineum and the halophytic hostsLimonium delicatulum andArthrocnemum glaucum were investigated in a Mediterranean salt marsh in March 1992. The main fungal inhabitants on the leaves or shoot surface of the test plants wereAspergillus niger, Penicillium chrysogenum andCladosporium herbarum. The qualitative analysis of the fungal species associating the parasite, the hosts and the non-infected plants showed similar pattern. However, the total population exhibited quantitative differences coupled with the amount and the chemical composition of the exudates on plant surface and the quantity of transpired water. The fungal catch from the aerial shoot (inflorescence) of the parasite was higher than that collected from either the leaves or aerial shoots of non-infected or host plants. The fungal density on the leaves ofL. delicatulum was higher than those isolated from the aerial shoots ofA. glaucum. Infection byC. coccineum caused a marked drop in the total fungal population on leaves or shoot surfaces of the hosts as compared to the corresponding non-infected individuals. The stimulative effect of washings on spore germination of some isolated fungal species was matched with the density of fungi on the target plants.  相似文献

Little is known about the amount of fungal biomass in the phyllosphere of bryophytes compared to higher plants. In this study, fungal biomass associated with the phyllosphere of three bryophytes (Hylocomium splendens, Pleurozium schreberi, Polytrichum commune) and three vascular plants (Avenella flexuosa, Gymnocarpium dryopteris, Vaccinium myrtillus) was investigated using ergosterol content as a proxy for fungal biomass. Phyllosphere fungi accounted for 0.2-4.0 % of the dry mass of moss gametophytes, representing the first estimation of fungal biomass associated with bryophytes. Significantly more fungal biomass was associated with the phyllosphere of bryophytes than co-occurring vascular plants. The ergosterol present in moss gametophytic tissues differed significantly between species, while the ergosterol present in vascular plant leaf tissues did not. The photosynthetic tissues of mosses had less associated fungal biomass than their senescent tissues, and the magnitude of this difference varied in a species-specific manner. The fungal biomass associated with the vascular plants studied varied significantly between localities, while that of mosses did not. The observed differences in phyllosphere community biomass suggest their size could be affected by host anatomical and physiological attributes, including micro-niche availability and chemical host defenses, in addition to abiotic factors like moisture and nutrient availability.  相似文献

以感染内生真菌的天然禾草羊草为实验材料,通过体外纯培养条件下的内生真菌、感染内生真菌的离体叶片和在体叶片对3种病原菌的抑菌实验,以探讨内生真菌对宿主植物羊草在抗病性方面的贡献。结果表明:体外纯培养条件下,分离自羊草的内生真菌Epichlobromicola对新月弯孢(Curvularia lunata)、根腐离蠕孢(Bipolaris sorokiniana)和枝孢霉(Cladosporium sp.)这3种病原菌都具有抑制作用,抑菌率分别达56.22%,46.93%和45.15%,且内生真菌培养滤液可以有效抑制这3种病原菌的孢子萌发,平均萌发率分别为30.4%,15.7%和16.4%;宿主植物叶片在离体条件下,内生真菌感染可以有效降低羊草叶片受C.lunata和C.sp.侵染后的病斑数或病斑长度,但对B.sorokiniana不起作用,甚至提高了叶片的病斑数及病斑长度,而离体叶片提取液对不同病原菌均有不同程度的抑制作用;在体条件下,内生真菌均可以通过降低叶片病斑数来增强羊草植株对这3种病原菌的抗性。由此看来,内生真菌E.bromicola对宿主植物羊草在抗病原菌侵染方面有一定的增益作用。  相似文献

Air pollution causes serious problems in spring in northern China; therefore, studying the ability of different plants to accumulate particulate matter (PM) at the beginning of the growing season may benefit urban planners in their attempts to control air pollution. This study evaluated deposits of PM on the leaves and in the wax layer of 35 species (11 shrubs, 24 trees) in Beijing, China. Differences in the accumulation of PM were observed between species. Cephalotaxus sinensis, Euonymus japonicus, Broussonetia papyriferar, Koelreuteria paniculata and Quercus variabilis were all efficient in capturing small particles. The plants exhibiting high amounts of total PM accumulation (on leaf surfaces and/or in the wax layer), also showed comparatively high levels of PM accumulation across all particle sizes. A comparison of shrubs and trees did not reveal obvious differences in their ability to accumulate particles based on growth form; a combination of plantings with different growth forms can efficiently reduce airborne PM concentrations near the ground. To test the relationships between leaf traits and PM accumulation, leaf samples of selected species were observed using a scanning electron microscope. Growth forms with greater amounts of pubescence and increased roughness supported PM accumulation; the adaxial leaf surfaces collected more particles than the abaxial surfaces. The results of this study may inform the selection of species for urban green areas where the goal is to capture air pollutants and mitigate the adverse effects of air pollution on human health.  相似文献
