白Qian体细胞胚胎发生及其植株再生条件的优化研究   总被引:1,自引:0,他引:1  
为建立白Qian体细胞胚胎发生及其植株再生的高频率实验系统,对聚乙二醇及干化处理的影响进行了系统研究。结果表明,在分化培养基中附加50g/L聚乙二醇可显著提高愈伤组织的分化频率和每块愈伤组织产生的体细胞胚个数,而干化处理又能使体细胞胚的萌发率大幅度上升。  相似文献

早熟棉体细胞胚胎发生和植株再生体系的建立   总被引:4,自引:0,他引:4  
以8个早熟或特早熟棉花为材料,通过不同激素组合(1.0 mg/L IBA+0.5 mg/L KT;0.1 mg/L 2,4-D+0.1 mg/L KT)诱导其愈伤组织,为早熟棉花的遗传转化以及基因功能研究奠定基础.结果表明,2种激素组合均能诱导产生4种主要类型的愈伤组织:淡黄色颗粒状愈伤组织,褐化的愈伤组织,翠绿致密的愈伤组织,疯长型愈伤组织.4种愈伤组织转入增殖培养基中前2种愈伤组织能够分化出胚性愈伤组织,胚性愈伤组织再转到分化培养基上培养能够产生胚状体,即体细胞胚;体细胞胚进一步发育成为再生小苗.用该组织培养再生系统,成功地使晋棉5号、中棉27号和辽棉10号等3个早熟棉品种在5~7个月内通过体细胞胚胎分化获得再生小苗.  相似文献

香雪兰的体细胞胚胎发生可通过两种途径进行,即直接发生与间接发生。在直接发生方式中,体细胞胚直接来源于尚未完全分化的外植体表皮细胞;体细胞胚与母体组织以一种类似胚柄的结构相联系。间接发生方式中,体细胞胚的形成要经过一个愈伤组织阶段。以是否能形成体细胞胚分类,可将愈伤组织分为胚性和非胚性愈伤组织。以间接方式形成的体细胞胚是由胚性愈伤组织中的一种决定细胞发育来的。这种体细胞胚不具有类似胚柄的结构,而与母体组织共同形成一个复合体。体细胞胚具有自己独立的维管束系统,在脱离母体组织后能够独立发育成株。  相似文献

在建立水稻体细胞悬浮培养体系的实验中,我们观察到当将胚性愈伤组织从固体培养基转入液体分化培养基进行悬浮培养并去掉2.4-D 时,随着培养时间的延长,愈伤组织往往出现褐化现象。与这种情况类似的是,当将悬浮培养的愈伤组织和成熟胚状体转入固体培养基时,也出现严重的褐化现象。这种褐化现象一般认为是不可逆的,伴随着胚性愈伤组织和成熟胚状体的褐化,胚性愈伤组织逐渐丧失分化能力,成熟胚状体则逐渐死亡。  相似文献

脱分化的枸杞叶片外植体愈伤组织转入含有2,4-D的MS培养基上分化培养后有大量胚性细胞的分化和体细胞胚发生;加入一定量的外源Ca2 或45Ca2 ,明显地提高了胚性愈伤组织中体细胞胚发生的频率;加入Ca2 的鳌合剂EGTA则显著降低了体细胞胚发生频率;胚性愈伤组织中CaM的水平在多细胞原胚期和球形胚期显著升高,加入外源Ca2 后CaM含量几乎成倍增加;胚性愈伤组织中蛋白质组分与活性都远远多于或高于非胚性愈伤组织,加Ca2 后蛋白质组分种类也增加.  相似文献

粗枝云杉愈伤组织在增殖后期体细胞胚的分化能力显著降低,转变愈伤组织增殖方式和体细胞胚分化培养方式有利于体细胞胚发生能力的提高。采用液体悬浮增殖取代半固体增殖更有利于胚性的保持。在增殖后期,首选的体细胞胚发生方式为"液体增殖-滤纸分化",其次为"块状增殖-块状分化",最后是"块状增殖-滤纸分化"。  相似文献

石刁柏胚性愈伤组织继代过程中,添加浓度为10 mg·L^-1的外源腐胺能有效地保持愈伤组织的胚胎发生能力,并减少愈伤组织的褐化,但不能提高愈伤组织的体细胞胚的诱导率.腐胺处理过的胚性愈伤组织的内源腐胺含量明显提高.这可能是外源腐胺保持细胞胚性、降低褐化程度的原因.  相似文献

大蕉未成熟雄花接种到胚性愈伤组织诱导培养基中,4~5个月后可诱导出胚性愈伤组织,并可在继代培养基上增殖.胚性愈伤组织转移到体细胞胚诱导培养基中可诱导出体细胞胚.体细胞胚在成熟培养基上培养2个月后转移到含有0.2mg·L-1 6-BA的分化培养基上可以萌发,进而形成再生植株.组织学切片证明所诱导的愈伤组织是胚性组织,其所产生的体胚具有典型的单子叶植物体细胞胚的组织结构.  相似文献

对 1 5种柑橘胚性愈伤组织进行体细胞胚诱导 ,发现逆境处理有利于体细胞胚发生 ,并可以恢复部分品种的体细胞胚发生能力。对具有和失去体细胞胚发生能力的两种纽荷尔脐橙 (CitrussinensisOsb .)愈伤组织进行随机扩增多态性DNA (RAPD)分析没有检测到带型的差异 ,而对它们的甲基化敏感扩增多态性 (MSAP)进行分析则发现两种愈伤组织间具有明显的DNA甲基化差异 ,具体细胞胚发生能力的愈伤组织的甲基化水平较失去体细胞胚发生能力的低  相似文献

研究了影响大豆幼胚培养体细胞胚胎发生频率的9个因素。诱导体细胞胚胎发生的适宜幼胚长度为4mm;随着供体植株发育阶段的提高诱导频率下降;最适基本培养基为MS培养基; 蔗糖浓度从1.5%提高到9%,诱导频率逐渐下降;过高的维生素B1浓度对胚胎发生不利;2,4 — D的诱导效果优于NAA,适宜的2,4—D浓度为20ppm; 光、暗处理与生长素种类和浓度之间存在交互作用,接种方式对诱导频率影响很大,体细胞胚只在下表皮与培养基接触的幼子叶的上表皮上产生,当上表皮与培养基接触时,两个表皮都不能产生体细胞胚;被试的所有基因型都能被诱导胚胎发生,不同基因型的诱导频率存在差异。  相似文献

用异常苗的茎段和叶片进行培养,可快速高效诱导获得棉花体细胞胚胎发生,激素组膈及其浓度配比影响异常苗的直接胚胎发生,在附加有0.1mg/L IAA和0.1mg/LZT的改良MS培养基上,异常苗不仅体细胞胚胎发生率高,而且形成的体细胞胚数目多,用异常苗作外植体获得胚性愈伤组织仅需要时间10d,获得成熟胚需要20-30d,获得再生植株需要60d,大大短于常规方法获得胚性愈伤组织、体细胞胚和再生植株的时间  相似文献

A simple, rapid and effective system to regenerate Arabidopsis plants via direct somatic embryogenesis has been established. Somatic embryogenesis was induced directly during culture of immature zygotic embryos. The frequency of somatic embryogenesis was strongly influenced by the stage of development of the explants. Explants in different developmental stages were cultured on B5 agar medium containing 5 M 2,4-dichlorophenoxyacetic acid and the highest frequency (up to 90%) of somatic embryogenesis was observed in zygotic embryos with fully-developed cotyledons. The first somatic embryos developing directly from explant tissue were noticed after 8 days of culture. Somatic embryogenesis of a high frequency (87–96%) was observed in cultures of the all six genotypes tested (Columbia, C-24, RLD, Wassilewskaja, Landsberg erecta and Wilna). Subculture of somatic embryos onto auxin-free medium resulted in their conversion into plants with an average frequency of 79.5%. The regenerates showed normal morphological characteristics and were fertile. All 56 analysed plants displayed a diploid number of chromosomes and two out of 96 (2.1%) tested plants carried a chlorophyll or embryo-lethal mutation.  相似文献

以作者自己的工作为背景,结合国内外近几年的有关报道,综述了几种外源和内源激素对植物体细胞胚胎发生的诱导与调节作用。外源生长素和细胞分裂素是诱导离体培养细胞分化与增殖所必需的,2,4-D是诱导胚性愈伤组织的重要激素。在体细胞胚胎发生中内源激素含量和代谢的平衡起着关键的作用,而且外源和内源激素对诱导体细胞胚胎发生起相互调节作用。ABA在提高体细胞胚胎发生频率和质量上具有重要作用,同时,外源与内源ABA对体细胞胚胎发生起相互促进作用。本文还较为深入地讨论了这些激素诱导体细胞胚胎发生的可能作用机制。 Abstract:The paper summarizes the induced and regulatory effects of a few exogenous and endogenous hormones in plant somatic embryogenesis by our studies and related international reports.The exogenous auxin and cytokinin are necessary to induced differentiation and proliferation of cells of culture in vitro.2,4-D is an important hormone of induced embryogenic calluses.The contents and the metabolic balances of endogenous hormones have key effects for somatic embryogenesis.In addition,the exogenous and endogenous hormones have mutual regulatory effects for somatic embryogenesis.ABA has an important effect to improving the frequency and quality of somatic embryogenesis.Meanwhile,the exogenous and endogenous ABA have mutual promoted effects for somatic embryogenesis.The paper discusses possible mechanism of hormones-induced somatic embryogenesis in a deep-going way.  相似文献

The system of high synchronous frequency of somatic embryogenesis and somatic embryo seedling formation was established by means of embryonic cell hne 1 ( CL1 ) of Camellia sinensis var. assamica Kitamura. Modified MS was used as the basic medium. Cultures of CL1 was transferred to the aqueous induced medium (0.05 mg/L 2,4-D + 0.50 mg/L 6-BA) from the maintenance medium (0.1 mg/L 2,4-D + 0.5 mg/L 6-BA) for somatic embryos induction under dark condition. 28 days later, they were cultured in the liquid differentiation medium. Various kinds of somatic embryos were obtained after another 28 days. The frequency of somatic embryos was 81.5 %. Various mesh sizes of sieves were applied to collect the somatic embryos in different developmental stages which could develop to mature stage in the aqueous growth medium ( 1/2 MS + 1.0 mg/L GA3 + 0.5 mg/L 6-BA). ABA was effective to promote the formation of highly qualified somatic embryo. The mature somatic embryos sized 20 to 70 mesh had the conversion frequency 75 %. The development of somatic embryogenesis studied under a cell suspension culture system was similar to the zygotic embryogenesis.  相似文献

利用云南大叶茶(Camellia sinensis var.assamica Kitamura)胚性细胞系(CL_1)中悬浮培养物,建立了高频率同步化体细胞胚发生及体胚苗形成体系。以改良的MS为基本培养基,将CL_1中培养物由液体保持培养基(0.1mg/L 2,4-D 0.5mg/L 6-BA)继代转入液体诱导培养基(0.05mg/L 2,4-D 0.50mg/L6-BA),暗培养诱导28d,转入不含任何激素的液体分化培养基中再培养28d,获得了不同发育时期的体细胞胚,其发生频率为81.5%。不同发育时期的体细胞胚用不同目的细胞筛收集,在液体生长培养基(1/2 MS 1.0mg/L GA_3 0.5mg/L 6-BA)中培养发育成熟。ABA有利于高质量体细胞胚的形成。20~70月大小的体细胞胚在固体生长培养基中成苗转换率为75%。在液体悬浮培养条件下观察记录了体细胞胚发育过程,证实其过程与合子胚的形态发生过程相似。  相似文献

Immature and mature zygotic embryos of Paspalum scrobiculatum L. cv. PSC 1 cultured on MS or N6 nutrient medium supplemented with different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D), formed embryogenic callus. Induction of embryogenic callus and subsequent somatic embryogenesis was possible at a lower concentration of 2,4-D on N6 than MS medium. Immature embryos were highly totipotent, forming somatic embryos at a higher frequency than mature embryos. Addition of amino acids (L-proline or L-tryptophan) to 2,4-D medium resulted in significant enhancement of embryogenesis on culture of mature embryos. Silver nitrate also supported an increased frequency of embryogenesis. Thus it is possible to have high frequency of somatic embryogenesis on culture of mature embryos, which are available in abundance and with ease than immature embryos. The somatic embryos readily germinated and formed plantlets on hormone-free regeneration medium. The regenerated plantlets were successful on transfer to soil and set seed.  相似文献

Significant differences in somatic embryogenesis from melon seeds were observed among 18 cultivars; especially, cultivars Earl's Favorite and Barnett which produced a large number of somatic embryos. F1 seeds were obtained by reciprocal crosses between cultivars. Some lines produced a large number of somatic embryos whereas others showed no or poor embryogenic response. Most of the F1 seeds formed somatic embryos. The frequency of somatic embryogenesis decreased as compared to the parents with the highest potential. Transfer of the frequency of somatic embryogenesis from superior responding cultivars to inferior cultivars was proved. Somatic embryogenesis in the carrot was used to model zygotic embryogenesis because the spatial and temporal changes in somatic and zygotic embryogenesis are quite similar. To establish an enhancer-trapping system for somatic embryogenesis in the carrot, we constructed 2 enhancer-trap vectors (pETVs) using the GUS reporter gene with a minimal promoter. We also constructed several positive control vectors (pPCVs) using the CaMV 35S promoter. These are models in which pETVs are inserted near a native enhancer region in correct or reverse orientation. First, we tested whether these vectors could be used as enhancer-trap vectors using transgenic hairy root of tobacco. Histochemical GUS assays revealed that pETVs could be used as enhancer-trap vectors, even when the reporter gene in the pETVs was inserted near the native enhancer. Subsequently, we examined the availability of pETVs in somatic embryogenesis in the carrot. The constructed vector was activated in transgenic carrot embryogenic cells at high frequency (64%). This suggests that the enhancer-trapping vector is suitable as a carrot somatic embryogenesis system.  相似文献

Summary Genetic factors controlling the differential expression of somatic embryogenesis and plant regeneration of maize from tissue culture were studied in two crosses. Inbred, hybrid, F2 and backcross generations developed from crossing maize inbred A188 with two commercially important inbred maize lines (B73 and Mo17) demonstrated genetic and environmental effects on somatic embryogenesis and plant regeneration when immature zygotic embryos were cultured on MS medium. Additive gene effects were more important in both crosses than dominant gene effects for precent somatic embryogenesis and percent or number of plants regenerated per embryo when generation means were analyzed. In backcross generations of each cross, cytoplasmic, maternal and/or paternal effects were significant for frequency of somatic embryos three weeks after culture as well as frequency, or number of plants regenerated per embryo, nine weeks after culture. Analysis of genetic variances suggests at least one gene (or block of genes) controls the expression of the frequency of somatic embryogenesis in these crosses.
