Rhodotorucine A which induces mating tube formation of a cells in Rhodosporidiumtoruloides is metabolized rapidly by a cells. By use of labeled rhodotorucine A, the degradation was found to be proteolytic. Two peptide fragments Tyr-Pro-Glu-Ile-Ser-Trp-Thr-Arg and Asn-Gly-Cys(S-farnesyl) were identified as the metabolites. Proteolysis of the pheromone mainly occurred on the cell surface. Culture filtrate of a cells at log phase did not metabolize rhodotorucine A.  相似文献

The transducing phage λdarg14, carrying a portion of the E. coli chromosome including argECBH, is derived from the heat-inducible, lysis-defective strain λy199, which has the b519 and b515 deletions. Cleavage of λy199 DNA by EcoRI endonuclease, followed by agarose slab gel electrophoresis, results in bands corresponding to the known C, D, E, and F segments of λ, and a segment A′ (A plus B minus b519 minus b515, the cleavage site between A and B being eliminated). Cleavage of λdarg14 DNA by EcoRI yields the expected D, E, and F segments of λ and four other segments, termed 14-1 through 14-4, whose length is 17.5, 6.2, 3.0, and 2.0 kilobases, respectively, as determined by electron microscopy and corroborated by electrophoretic mobility. Heteroduplex analysis shows that the E. coli argECBH cluster is on the 14-1 segment.  相似文献

An inhibitor of Streptococcus,mutans endodextranase was detected in proteins prepared from batch cultures of S.,mutans strains representing serotypes a through g. Affinity chromatography of strain 6715-49 proteins, which apparently were free of endodextranase activity, yielded an active endodextranase and, in a separate peak, the endodextranase inhibitor. The presence of the inhibitor in culture fluids accounts for the absence of endodextranase activity in batch-grown cultures of S.,mutans known to produce this enzyme.  相似文献

Purified cytochrome P450SCC from bovine adrenocortical mitochondria was incorporated into liposomes by the cholate-dilution method utilizing either dialysis or Sephadex gel filtration. Among synthetic phospholipids tested, dioleoylglycerophosphocholine showed the best stability during the incorporation of P450SCC into liposomes. A maximum amount of heme was incorporated into liposomes at a molar ratio of phospholipid to the cytochrome of approx. 200. When P450SCC was incorporated into the dioleoylglycerophosphocholine liposomes by the cholate-filtration method, the P450SCC-containing liposomes showed two major populations on the elution pattern of the Sepharose 4B gel filtration, and were seen at a diameter of 200–600 Å and its aggregated forms. When the cytochrome was incorporated into dioleoylglycerophosphocholine liposomes or cholesterol-free adrenocortical mitochondrial liposomes, P450SCC was less stable than P450SCC in aqueous solution. Cholesterol or adrenodoxin markedly stabilized the liposomal P450SCC. Liposomal P450SCC required cholesterol for its optimum reduction with adrenodoxin, adrenodoxin reductase, and NADPH in the presence of CO. About 70% of the total heme in the dioleoylglycerophosphocholine liposomes was reduced by the enzymatic reduction in the presence of cholesterol, indicating that 70% of the total molecules are exposed to the surface of the outer monolayer. In order to see the location of the heme in membrane, the dioleoylglycerophosphocholine-liposomal P450SCC was subjected to p-chloromercuriphenyl sulfonic acid treatment. This reagent destroyed the liposomal P450SCC. These results suggest that the heme is located in the proximity of the p-chloromercuriphenyl sulfonic acid reacting sites which are exposed to the surface, or located on the vincinity of polar heads of the membrane.  相似文献

The partial purification of (Na+ + K+)-ATPase from pig lens has been achieved by treatment with deoxycholate followed by density gradient centrifugation. The specific activity of the final preparation, ranging from 300 to 500 nmol/h per mg protein, is increased approx. 100-fold compared to the homogenate. A parallel increase in p-nitrophenylphosphatase activity is also observed. Sodium dodecyl sulfate (SDS) gel electrophoresis reveals six major protein bands, one of which is the 93 kDa α subunit of (Na+ + K+)-ATPase which can be phosphorylated by reaction with [γ-32P]ATP. A second band contains a glycoprotein which displays an apparent molecular weight of 51 000 and thus appears to be the β subunit of the enzyme. The enzyme is sensitive to ouabain with the I50 for (Na+ + K+)-ATPase and p-nitrophenylphosphatase inhibition being 1.2 and 1.3 μM, respectively. Several agents which inhibit Na+ + K+)-ATPase from other tissues such as oligomycin, Ca2+, vanadate, N-ethylmaleimide, p-chloromercuribenzenesulfonic acid (PCMBS) and 5,5′-dithiobis-(2-nitrobenzoic acid) (DTNB) also inhibit the lens enzyme. Monovalent cations other than K+ are partially effective in activating the (Na+ + K+)-ATPase and p-nitrophenylphosphatase activities. The K+ congeners were relatively more effective in supporting (Na+ + K+)-ATPase compared to p-nitrophenylphosphatase activity. Other kinetic properties of the lens enzyme are also comparable to those of the enzyme from other tissues. Utilizing the partially purified membrane bound enzyme, discontinuities in Arrhenius plots of (Na+ + K+)-ATPase activity, p-nitrophenylphosphatase activity and fluoresence polarization of the fluidity probe, 1,6-diphenyl-1,3,5-hexatriene (DPH), are observed near the physiological temperature of lens. The possible significance of these observations for the mechanism of cataract formation are discussed.  相似文献

Phospho-N-acetylmuramyl-pentapeptide translocase, the initial membrane enzyme in the biosynthesis of peptidoglycan, requires a lipid microenvironment for function. n-Butanol was reversibly intercalated into membranes to perturb the hydrophobic interactions in this microenvironment in order to define further the role of lipid. In the concentration range for maximal stimulation of enzymic activity (0.12–0.18 M), n-butanol causes a 40% decrease in the fluorescence emission of the dansylated product, undecaprenyl diphosphate-(N?-dansyl)pentapeptide. Since no change in emission maximum occurs below 22°C in the presence of 0.12 M n-butanol, it is concluded that intercalation of this alkanol causes an increase in fluidity. Above 22°C this concentration of n-butanol causes both a decrease in the fluorescence emission and a red shift in the emission maximum. It is concluded that a polarity change as well as fluidity change occurs above 22°C. n-Butanol also causes a significant change in the phase transition experienced by the dansylated lipid product. Thus, it is possible with n-alkanols, e.g. n-butanol, to perturb lipid-translocase interactions resulting in an increase in fluidity in the microenvironment of the enzyme. This change in fluidity correlates with a stimulation of enzymic activity.  相似文献

Evidence is presented for the presence of a new lectin activity in soybean seeds [Glycinemax (L.) Merrill] that has specificity towards the 4-O-methyl-D-glucurono-L-rhamnan exopolysaccharide produced by certain strains of Rhizobiumjaponicum. Bacterial agglutination and precipitin reactions revealed the lectin activity in phosphate-buffered saline extracts of seeds of all cultivars tested, including the “lectinless” varieties. Reaction of such extracts with carbohydrate haptens demonstrated that the specificity of the binding was towards 4-O-methyl-D-glucuronic acid, D-glucuronic acid and their methyl glycosides.  相似文献

Uterine stage embryos collected from the hamster (8-cell) and cow (morula, early blastocyst) were monitored for development invitro (embryo culture) and invivo (embryo transfer) following premature removal of the zona pellucida.Removal of the zona pellucida did not significantly affect invitro development to the blastocyst stage of (1) 8-cell hamster embryos (zonae removed by a combined enzymic-mechanical procedure), (2) bovine morulae (zonae removed by mechanical means only) (3) early bovine blastocysts (zonae removed by the enzymic-mechanical technique).Zona-free hamster embryos formed significantly fewer viable fetuses than did zona-intact embryos. The lower incidence of fetal development observed following transfer of zona-free 8-cell hamster embryos may have resulted in part from the formation of chimeras by fusion of these embryos inutero. Such fusion was observed to occur invitro between zona-free embryos placed in close proximity. The proportion of pregnancies resulting from transfer of bovine blastocysts cultured from zona-free morulae was similar to that of zona-intact embryos.In this study we have demonstrated that (1) enzymic and mechanical procedures used to remove zonae pellucidae from uterine-stage hamster and bovine embryos do not adversely affect subsequent development of these embryos invitro and invivo and (2) zonae pellucidae are not required for normal development of these embryos. These findings have implications for microsurgery of mammalian embryos and for embryo transfer.  相似文献

In previous studies on the modification of polar head groups of membrane phospholipids with the unnatural base analog, N-isopropylethanolamine, we reported an unidentified phospholipid in addition to phosphatidyl-N-isopropylethanolamine in the various membrane fractions of rat liver. The structure of this phospholipid has now been identified as phosphatidyl-N-methyl-N-isopropylethanolamine by nuclear magnetic resonance spectroscopy, and by chromatographic and enzymic analysis. In addition, we found that when rats were injected intraperitoneally with the N-methyl-N-isopropylethanolamine, 19% of teh liver microsomal phospholipid was phosphatidyl-N-methyl-N-isopropylethanolamine.  相似文献

The dependence of the replication of several plasmids on the chromosome-determined initiation products, dna A and dna C, has been studied. The initiation of the replication of Col E1 DNA requires the chromosomal dna A product. In contrast two de-repressed transfer factors (R 1 drd 16 and Hly152) seem to determine a corresponding plasmid-specific factor. The dna C-product is necessary for the ordered initation of all plasmids studied. The addition of low concentrations of chloramphenicol leads to a relaxed replication of Col E1 DNA at the restrictive temperature in dna A-mutants, but not in dna C-mutants.  相似文献

OKY-1581 is an effective inhibitor of thromboxane synthesis invivo and invitro. The generation of thromboxane B2 (TxB2), prostaglandin E (PGE) and prostaglandin F (PGF) was measured following clotting and during platelet aggregation induced by collagen. The presence of OKY 1581 either invivo or invitro caused a reduction in TxB2 generation during clotting and platelet aggregation with a concomitant increase in PGE and PGF. The effect could be observed two hours after oral or subcutaneous administration of 5 to 100 mg per rabbit and lasted for 24 to 48 hours. The reduction in TxB2 was not accompanied by an inhibition of clotting or platelet aggregation. OKY-1581 appears to be a suitable agent for studying the role of TxB2 in atherosclerosis.  相似文献

The specific synthesis of argF mRNA directed by the argF gene carried on the specialized transducing bacteriophage λh80C1857dargF, performed in vitro, is described with the use of an S180 extract from a strain carrying argR?. Synthesis of argF mRNA is biphasic at approximately 7 minutes. Stereosecific synthesis of trans-hydrindanone 2a, a bicyclic analog of prostaglandin E1, via the trans-hydrindane β-keto ester 8, is described. When tested in the guinea pig, 2a exhibited no effects on blood pressure and no broncho-constriction or dilation activity. Additionally, 2a failed to inhibit both ADP and collagen induced blood platelet aggregation.  相似文献

The thermally-induced changes of a cytochrome P-450 dependent activity (ethoxycoumarin-O-deethylase) and of the fluorescence polarization of diphenylhexatriene were compared in microsomes from lean, obob and dbdb mice. In lean mice, biphasic plots were obtained with break points in the same range of temperature by both methods, whereas, in obob and dbdb mice, no discontinuities were observed. These results may be related to a modified fatty acid composition of microsomal membranes in mutant mice. They exemplify the influence of the lipid environment on the monooxygenase system as also shown by the modified binding constants of cytochrome P-450 towards type II substrates in dbdb mice.  相似文献

The pesticide o,p'-DDT stimulates the production of a specific uterine protein, the so-called induced protein or IP, normally associated with an estrogenic response of the uterus. Invivo stimulation of IP production is observed 1 hour after the administration of 250 mg/kg of o,p'-DDT to immature rats. Invitro stimulation of IP production is observed after a 1 hour incubation of uteri with 100 μM o,p'-DDT. This invitro response is blocked by Actinomycin D. In contrast to o,p'-DDT, which binds to the cytoplasmic estrogen receptor and stimulates IP production, p,p'-DDT which does not bind well to the estrogen receptor does not stimulate IP production invitro. These findings represent the first report of an estrogenic effect of o,p'-DDT in a completely invitro system.  相似文献

(1) A membrane fraction enriched in (Na+ + K+)-ATPase (EC 3.6.1.3) was obtained from optic ganglia of the squid (Loligo pealei) by density gradient fractionation of membranes followed by treatment with either SDS or Brij-58. The resulting membrane had an (Na+ + K+)-ATPase specific activity of approx. 2 units/mg and was >95% ouabain-sensitive. (2) The (Na+ + K+)-ATPase had a Km for ATP of 0.42 ± 0.04 mM and a pH optimum of 7.0. It was inhibited by ouabain with a Ki of 0.32 ± 0.04 μM. (3) Optimum monovalent cation concentrations were: 240 mM NaCl, 60 mM KCl, tested with NaCl + KCl = 300 mM. (4) The Mg2+ dependence of hydrolysis varied with the absolute ATP concentration. At 3 mM ATP, theKm for Mg2+ was 0.86 ± 0.10 mM, and at 6 mM ATP, the Km was 1.86 ± 0.44 mM. High levels of Mg2+ caused inhibition of hydrolysis. (5) The interactions of Na+ and K+ were examined over a range of conditions. K+ levels caused modulations in the Na+ dependence in the range of 1–150 mM. (6) The (Na+ + K+)-ATPase prepared from squid optic ganglion displays properties similar to those of the sodium pump in injected nerves.  相似文献

The paromomycin producing organism Streptomycesrimosusforma paromomycinus is resistant to this antibiotic and contains a phosphotransferase which inactivates paromomycin. The gene encoding this enzyme has been inserted in the Streptomyces vector pIJ702 and then cloned in Streptomyceslividans, selecting for paromomycin-resistance. Three plasmids have been isolated and one of them, pMJ1, contains a 2.2 kb insert with a single HindIII restriction site. Insertion of foreign DNA in this site blocks the expression of the phosphotransferase enzyme indicating that it is within the cloned gene. These findings provide a new dominant selective marker for Streptomyces cloning vectors with the versatility of insertional inactivation.  相似文献

Short, mild treatments of sarcoplasmic reticulum vesicles with aqueous n-alcohols from methanol to n-heptanol caused an inhibition of calcium uptake and an enhancement of ATPase activity. The n-alcohol treatments increased both calcium-dependent (extra) ATPase activity and calcium-independent (basic) ATPase activity of vesicles. The apparent initial reaction rate of ATPase of n-alcohol-treated vesicles was about twice that of control vesicles. With increasing number (n) of carbon atoms of the n-alcohols, the maximum increment of ATPase activity increased, and both the alcohol concentration (NCa) required to inhibit calcium uptake by 50% and the alcohol concentration (NATPase) required to enhance ATPase activity by 50% of the maximum increment of ATPase activity decreased as follows.

NATPase=35.5·10?0.593nM
The ratio, NATPase to NCa, was constant for all n values. The apparent free energy of binding of the methylene groups of n-alcohols to sarcoplasmic reticulum vesicles was evaluated (?796 cal/mole) and compared with data from the partition of n-alcohols in octanol and water (?670 cal/mole). The effects of n-alcohols on membrane vesicles are discussed on the basis of these data.  相似文献

Both pairs of dl-ll-desoxy- and dl-13-cis-erythro-15, 16-dihydroxyprostaglandins have been synthesized via 1,4-conjugate additions of an appropriately functionalized cis-vinyl cuprate to the requisite cyclopentenone.
