C‐type lectins (CTLs) play a variety of roles in plants and animals. They are involved in animal development, pathogen recognition, and the activation of immune responses. CTLs carry one or more non‐catalytic carbohydrate‐recognition domains (CRDs) to bind specific carbohydrates reversibly. Here, we report the molecular cloning and functional analysis of a single‐CRD CTL, named C‐type lectin‐S2 (BmCTL‐S2) from the domesticated silkmoth Bombyx mori (Lepidoptera: Bombycidae). The ORF of CTL‐S2 is 666 bp, which encodes a putative protein of 221 amino acids. BmCTL‐S2 is expressed in a variety of immune‐related tissues, including hemocytes and fat body among others. BmCTL‐S2 mRNA level in the midgut and the fat body was significantly increased by bacterial challenges. The recombinant protein (rBmCTL‐S2) bound different bacterial cell wall components and bacterial cells. rBmCTL‐S2 also inhibited the growth of Bacillus subtilis and Staphylococcus aureus. Taken together, we infer that BmCTL‐S2 is a pattern‐recognition receptor with antibacterial activities.  相似文献

Beta-N-acetylglucosaminidase is a major glycosidase involved in several physiological processes, such as fertilization, metamorphosis, glycoconjugate degradation, and glycoprotein biosynthesis in insects. A search using the Bombyx mori cDNA database revealed the existence of two putative beta-N-acetylglucosaminidase genes. Their full-length cDNAs were cloned by rapid amplification of cDNA ends and polymerase chain reaction using specific primers, and named BmGlcNAcase1 and BmGlcNAcase2. A BLAST search revealed that BmGlcNAcase1 and BmGlcNAcase2 are homologous to a beta-subunit homolog encoded by Drosophila melanogaster HEXO2 and the Spodoptera frugiperda beta-N-acetylglucosaminidase gene respectively. The recombinant proteins of BmGlcNAcase1 and BmGlcNAcase2 without putative transmembrane domains were expressed in the yeast Pichia pastoris. Both enzymes showed broad substrate specificity, and cleaved terminal N-acetylglucosamine residues from the alpha-3 and alpha-6 branches of a biantennary N-glycan substrate, and also hydrolyzed chitotriose to chitobiose.  相似文献

To identify genes involved in the innate immunity of the silkworm Bombyx mori, we constructed a cDNA library from the fat body of Escherichia coli-challenged B. mori larvae. Based on the expressed sequence tag (EST) data and whole genome shotgun sequence analysis, we found four Gloverin-like genes, BmGlov1-4, in the Bombyx genome. Northern blot and RT-PCR analysis showed that BmGlov1-4 were induced in the larval fat body after an immune challenge by the injection of E. coli; however, less induction was observed after the injection of a yeast Candida albicans. In silico sequence analysis revealed the presence of a motif homologous to NF-kappaB binding site in the upstream region of each BmGlov gene. Moreover, we expressed recombinant BmGlov1-4 proteins using the baculovirus expression system, and found that all the recombinant BmGlov1-4 significantly inhibited the growth of E. coli.  相似文献

气味结合蛋白(odorant binding proteins, OBPs)在昆虫与外界环境化学信息交流过程中起着重要作用, 对昆虫的觅食、 求偶、 繁殖具有重要意义。触角结合蛋白(antennal binding protein, ABP)是OBP家族中的重要成员之一。为进一步探明家蚕Bombyx mori ABP与ABPX基因的结构、 表达及功能, 本研究利用染色体定位、 基因分析及半定量表达分析方法对其进行了研究。染色体定位分析表明, BmABP和BmABPX分别位于家蚕第5和第26染色体上, 基因结构差异较大, 可能功能上有较大差异。对家蚕胚胎、 幼虫和成虫不同发育阶段的雌、 雄虫多种组织进行基因表达谱分析发现, BmABP在家蚕发育的各个虫态、 多种组织器官中都有较高表达, 无时间特异性和组织特异性; BmABPX在不同发育时期和不同组织间差异显著(P<0.05), 相对表达量以触角中最高, 其他非嗅觉组织中也多有表达, 性别间差异不大。结果提示, BmABP和BmABPX除了具有嗅觉相关功能外, 很可能还具有其他未知的生理功能。  相似文献

Hox基因(homeobox genes)在昆虫躯体模式(body plan)的发育调控机制中扮演着重要角色,其表达具有严格的组织特异性和胚胎发育的程序性。家蚕Bombyx mori作为鳞翅目昆虫的代表,其Hox基因也陆续得到鉴定。在家蚕中存在一个拟复等位基因群--E群基因,其突变表型均与过剩斑纹和过剩附肢有关,这可能与Hox基因有着密切联系。家蚕全基因组测序完成后,发现其Hox基因簇中存在12个特有的homeobox基因(Bmshx1~Bmshx12), 说明家蚕Hox基因可能具有独特的生物学意义。我们还利用家蚕基因芯片数据分析了Bmlab与Bmpb基因的组织表达特征。通过对家蚕Hox基因的研究,探索家蚕躯体模式建立机制,可望为解析其他鳞翅目昆虫的躯体模式的建立机制提供理论依据。本文就家蚕Hox基因的表达、功能及其与E群突变的关系等方面进行了综述。  相似文献

LIM protein cDNA, from Bombyx mori that contains an open reading frame of 622 bp encoding 94 amino acids, was identified and characterized. The B. mori LIM protein homologue is classified into group 2 LIM proteins that contain glycine-rich LIM domain. Mutations in the human Crumbs homologue 1 (CRB1) gene cause severe retinal dystrophies. CRB1 is homologous to Drosophila Crumbs, a protein essential for establishing and maintaining epithelial polarity. We have isolated the mouse orthologue, Crb1, and analyzed its expression pattern in embryonic and post-natal stages. Crb1 is expressed exclusively in the eye, and the central nervous system. In the developing eye, expression of Crb1 is detected in the retinal progenitors, and later on becomes restricted to the differentiated photoreceptor cells where it remains active up to the adult stage. In the developing neural tube, expression of Crb1 is restricted to its most ventral structures, coinciding with the expression domain of Nkx2.2. In the adult brain, Crb1 expression is defined to areas where the production and migration of neurons occurs in adulthood.  相似文献

Based on the advances in the silkworm genome project, a new genome-wide analysis of cytochrome P450 genes was performed. A total of 84 CYP-related sequences were identified and could be classified into 26 families and 47 subfamilies according to standard nomenclature. Seventy eight of the eighty four genes appear to be functional and six are probable pseudogenes. The distribution of Bombyx mori P450s in the genome shows that most of them are tandem arranged on chromosomes, only 34 genes are present as singletons, with 8 clusters including 3 or more than 3 genes. Sequence alignments were used to reconstruct phylogenetic trees and to analyze the intron-exon organizations of the functional genes. The conserved intron positioning agrees perfectly with their common grouping on the tree. The presence of three extremely ancient introns which are conserved across different clans indicates that a few introns are still highly conserved after they have undergone extensive evolutionary changes of B. mori P450 duplication and divergence. Comparison of the P450s from B. mori to the P450s from Drosophila melanogaster shows that the expansion is not uniform across the gene families. Remarkably, two mitochondrial families, the B. mori CYP333 and D. melanogaster Cyp12, formed two orthologous groups in the phylogenetic tree. All CYP333s can be proposed to be related to xenobiotic metabolism in accordance with the D. melanogaster Cyp12s. The characterization and evolutionary analysis of P450s from B. mori in the current study provide useful information for understanding the characteristics and diversity of P450s from B. mori and the baseline for functional analyses of individual P450s in this model Lepidopteran insect.  相似文献

TheBombyx mori (B. mori) serine protease-like protein (BmSp) coding region (946 bp, GenBank accession number of mRNA, DQ118520; protein, AAZ40503) was generated from two separate and overlapping cDNA fragments using sequence homology withTrichoplusia ni azurocidin in aBombyx EST database (Silkbase; http://www.ab.a.u-tokyo.ac.jp/silkbase/). The deduced amino acid sequence of BmSp, which encodes 303 amino acids, shows 44% amino acid identity toA. gambiae serine protease (CAA89967), 43% amino acid identity toSarcophagi peregrina 26-kDa protease, an antibacterial protein and 31% identity toB. mori serine protease-2 (BmSP-2), a potential antiviral protein. Typical features of the BmSp included the serine protease active site triad His / Asp / Ser, three pairs of cysteine residues for disulfide bridges, and three residues, Asp / Gly / Gly, that help to confer trypsin-like specificity to the enzymes. Based on the result of sequence comparison and characterization, our results suggest that the BmSp probably the new subfamily of trypsin-like serine protease. Using RT-PCR and enzyme digestion, the full encoding sequence for BmSp was cloned into theE. coli expression vector pGEX-5X-1. The fusion protein GST-BmSp was effectively expressed inE. coli BL21(DE3) pLysS as inclusion bodies, and a denaturation and refolding procedure were performed to obtain soluble GST-BmSp. The purified protein was tested for antibacterial activity against Gram-positive and Gram-negative bacteria, but it did not show antibacterial activity in the agar well diffusion assay and liquid growth inhibition assay.  相似文献

In many organisms, dosage compensation is needed to equalize sex-chromosome gene expression in males and females. Several genes on silkworm Z chromosome were previously detected to show a higher expression level in males and lacked dosage compensation. Whether silkworm lacks global dosage compensation still remains poorly known. Here, we analyzed male:female (M:F) ratios of expression of chromosome-wide Z-linked genes in the silkworm using microarray data. The expression levels of genes on Z chromosome in each tissue were significantly higher in males compared to females, which indicates no global dosage compensation in silkworm. Interestingly, we also found some genes with no bias (M:F ratio: 0.8–1.2) on the Z chromosome. Comparison of male-biased (M:F ratio more than 1.5) and unbiased genes indicated that the two sets of the genes have functional differences. Analysis of gene expression by sex showed that M:F ratios were, to some extent, associated with their expression levels. These results provide useful clues to further understanding roles of dosage of Z chromosome and some Z-linked sexual differences in silkworms.  相似文献

昆虫能够特异性识别同类异性。雄蚕蛾对雌蚕蛾感知定位过程中, 性信息素结合蛋白PBP1、气味受体OR1和OR3起重要作用。为研究家蚕Bombyx mori和野桑蚕Bombyx mandarina杂交困难的分子机制, 了解性识别相关基因的进化, 本研究克隆得到了野桑蚕的性信息素结合蛋白基因pbp1(GenBank注册号:GQ246497)和气味受体基因or1(Genank注册号:GQ246496)和or3(GenBank注册号:GQ246498)。序列分析发现, 家蚕与野桑蚕相比, pbp1基因存在4个SNP位点, 分别为C10A, A40T, T270C和A333G, 其中2个SNP位点引起氨基酸的改变, 分别为Q→K和N→Y; or1基因存在5个SNP位点, 分别为T910C, A1147C, A1192T, T1276C和G1282A, 其中1个SNP位点引起氨基酸F→L的改变; or3基因存在4个SNP位点, 分别为A507G, A513G, T605C和G672A, 其中1个SNP位点引起氨基酸I→T的改变。3个基因的遗传距离很近, 进化速率也很慢。氨基酸的分子量和等电点有细微差异或无差异。PHD预测的二级结构表明, 变异位点对附近区域的结构没有任何影响, 功能位点也没有变化。推测家蚕与野桑蚕之间, 这些基因功能可能没有差异, 即二者的雌雄性个体间可以相互感知、识别, 这与实验观察结果一致。  相似文献

家蚕Bombyx mori是一种重要的经济昆虫, 在中国约有5 000年的驯化历史。家蚕分子免疫学方面的最新研究已经初步勾勒出其先天免疫的轮廓。本研究基于更新的家蚕基因组数据, 通过与黑腹果蝇Drosophila melanogaster、冈比亚按蚊Anopheles gambiae、意大利蜜蜂Apis mellifera和赤拟谷盗Tribolium castaneum基因组的比较分析, 鉴定了家蚕21个免疫相关基因家族的218个基因, 其编码产物包括模式识别受体、信号传导因子、效应分子和氧化防御相关的酶类。尽管信号传导因子的序列分化较大, 但系统进化分析显示它们在不同昆虫间呈明显的直系同源关系。相反, 与识别、调制和效应因子相关的基因的序列保守性更高, 但是这些基因家族明显缺乏直系同源基因, 由此推测这些基因是由物种特异的基因复制机制产生的。结果提示家蚕拥有与其他昆虫相同的免疫应答调控的分子机制, 而且家蚕同样可以通过基因复制及其序列分化等方式调节防御策略。  相似文献

Proteome analysis of silk gland proteins from the silkworm, Bombyx mori   总被引:10,自引:0,他引:10  
The silk gland of Bombyx mori is an organ specialized for the synthesis and secretion of silk proteins. We report here the resolution of silk gland proteins by 2-DE and the identification of many of those proteins. This was accomplished by dissecting the glands into several sections, with each exhibiting more than 400 protein spots by 2-DE, of which 100 spots were excised and characterized by in-gel digestion followed by PMF. Ninety-three proteins were tentatively identified. These were then categorized into groups involved in silk protein secretion, transport, lipid metabolism, defense, etc. Western blotting of a 2-DE gel using an antibody of the carotenoid binding protein confirmed the presence of this protein in the silk gland. Proteins including fibroin L-chain and P25 were found as multiple isoforms, some of which contained differential amounts of phosphate residues as analyzed by on-probe dephosphorylation. The current analysis contributes to our understanding of proteins expressed by the silk gland not only of the model lepidopteran B. mori, but also to proteins from other silk-producing insects such as Philosamia cynthia ricini.  相似文献

Cocoonase (CCN) which facilitates the degradation of a cocoon is recognized as a trypsin-like serine protease. In this study, CCN from the silkworm Bombyx mori was purified and comprehensively characterized. Its activity was maximal at about pH 9.8. It was stable above pH 3.4 at 4?°C and below 50?°C at pH 7.5. CuSO4, FeSO4, and ZnSO4 showed inhibitory effects on CCN, but other salts improved activity. Typical trypsin inhibitors inhibited CCN, but the relative inhibitory activities were much lower than those against bovine trypsin. An extract of cocoon shells inhibited trypsin, but it was only slightly inhibitory against CCN. There were significant differences in catalytic efficiencies and substrate specificities as between CCN and bovine trypsin.  相似文献

Sulfoconjugation plays a vital role in the detoxification of xenobiotics and in the metabolism of endogenous compounds. In this study, we aimed to identify new members of the sulfotransferase (SULT) superfamily in the silkworm Bombyx mori. Based on amino acid sequence and phylogenetic analyses, two new enzymes, swSULT ST1 and swSULT ST2, were identified that appear to belong to a distinct group of SULTs including several other insect SULTs. We expressed, purified, and characterized recombinant SULTs. While swSULT ST1 sulfated xanthurenic acid and pentachlorophenol, swSULT ST2 exclusively utilized xanthurenic acid as a substrate. Based on these results, and those concerning the tissue distribution and substrate specificity toward pentachlorophenol analyses, we hypothesize that swSULT ST1 plays a role in the detoxification of xenobiotics, including insecticides, in the silkworm midgut and in the induction of gametogenesis in silkworm ovary and testis. Collectively, the data obtained herein contribute to a better understanding of SULT enzymatic functions in insects.  相似文献
