The expression of the genes for two types of myrosinase (EC 3.2.3.1), designated MA and MB, during embryo and seedling development was investigated in Sinapis alba L. by in-situ and RNA slot-blot analyses. The expression of MA and MB genes followed similar temporal profiles during embryogenesis, but MB mRNA was present in considerably higher amounts than MA mRNA. In the embryo, both MA and MB genes are activated in cotyledons and axis. The MB genes are preferentially expressed in the cotyledons whereas MA genes are preferentially expressed in the axis. In the developing seedling, MA mRNA was not present in the organs investigated. By contrast, MB mRNA was found in appreciable amounts in hypocotyls, cotyledons and developing leaves. Ca-polygalacturonate is a demethoxylated component of pectins which are constitutive of plant root mucigel. In order to define the role of root mucigel in myrosinase immobilization and activity at root level, a myrosinase enzyme which had been isolated from Sinapis alba seeds was immobilized into Ca-polygalacturonate. The activity profile for the immobilized and free enzyme was evaluated using the pH-Stat method as a function of time, temperature, and pH. The Michaelis-Menten kinetic parameters change between the immobilized (V max ?=?127?±?13 U mg?1 protein; K M ?=?6.28?±?0.09?mM) and free (V max ?=?17?±?1 U mg?1 protein; K M ?=?0.96?±?0.01?mM) forms of myrosinase, probably due to conformational changes involving the active site as a consequence of enzyme immobilization. Immobilized enzyme activity evaluated as a function of different substrates gave the highest value with nasturtin, the glucosinolate that is typical of several brassicaceae plant roots containing the glucosinolate-myrosinase defensive system. No feedback regulation mechanism was found in the presence of an excess of enzymatic reaction products (i.e. allyl isothiocyanate or sulphate). The high enzyme immobilization yield into Ca-polygalacturonate and its activity preservation under different conditions suggest that the enzyme released by plants at root level could be entrapped in root mucigel in order to preserve its activity.  相似文献

Abstract The effect of light on the rate of formation of leaf primordia was investigated at the apex of seedlings of Sinapis alba and Xanthium strumarium. It was found that light accelerates this rate. On the other hand, no significant light effect was found on the angles of divergence of successive leaves during the transition from the almost decussate leaf position of the cotyledons to the helical phyllotaxis of the stem leaves. In fact, light and dark grown plants use the same leaves for the transition from decussate to helical phyllotaxis. Thus, if time is plotted in ‘biological units’ (number of primordia) there is no difference between light and dark grown plants. Using scanning electron microscope techniques it was found that the ‘primordia free apical area’ enlarges during development. The rate of enlargement is accelerated by light. However, if time is expressed in biological units (number of primordia) no difference between light and dark grown plants exists. It is concluded that light accelerates the realization of the apical pattern without interfering with the specification of the pattern. In other words, light accelerates the development of an apex without affecting the temporal and spatial coordination of the events.  相似文献

Atomic absorption spectrophotometry was used to determine thetotal amount of Ca in the leaves and apical bud of the long-dayplant Sinapis alba during the floral transition induced by asingle long day. The level of Ca increased in the apical budbut did not change significantly in the leaves. Ca was furtherlocalized cytochemically and measured in the meristem cellsby X-ray microprobe analysis. In the meristem, the level ofCa was found to increase in all cellular compartments, but mainlyin the nucleolus. Plants of Sinapis alba L. were induced to flower by either a single long day or a single displaced short day. The levels of three cations. Ca2+, Mg2+ and K+, were measured by atomic absorption spectrophotometry in exudates from roots, leaves and apical stem tips. The export of all three cations out of the root system (root exudate) was increased in induced as compared to non-induced plants. No changes were observed in cation export out of the mature leaves (leaf exudate). The supply of cations to the apical bud (apical exudate) did not originate from the phloem and, so, should mainly be of apoplastic origin. Only the supply of Ca2+ to the apical bud was increased, not the supply of Mg2+ or K+. The increase in Ca2+ supply was transient and occurred at about the same time as a conspicuous stimulation of cell division, previously detected in the apical bud.  相似文献

Summary The cells of the embryo of Sinapis alba L. include either aleurone or myrosin grains and all cells contain oil bodies. Aleurone grains and oil bodies are degraded during germination. The myrosin grains of each myrosin cell, on the other hand, gradually turn into one big vacuole containing the myrosin. Probably very little, if any, new myrosin is formed in the cotyledons and hypocotyl of the seedling after germination. No difference was found between aleurone and myrosin cells in the development of organelles. The cells of provascular bundles of the mature embryo contain different amounts of aleurone grains in different stages of development, and their organelles are more developed than those of all other cells.  相似文献

Gibberellins and the floral transition in Sinapis alba   总被引:3,自引:0,他引:3  
The putative role of gibberellins in the transition to flowering was investigated in Sinapis alba , a caulescent long-day (LD) plant. It was observed that: (1) physiological doses of exogenous gibberellins (GA1, GA3, GA9) do not cause the floral shift of the meristem when applied to plants grown in short days but have some positive effect on the flowering response to a suboptimal LD; no inhibition was observed in any case; (2) GA-biosynthesis inhibitors (prohexadione-Ca and paclobutrazol) considerably inhibit stem growth but have some negative effect on flowering only when a suboptimal LD is given; and (3) the floral transition induced by one 22-h LD does not correlate with any detectable change in GA content of the apical bud, of the leaves, and of the phloem exudate reaching the apex. Taken together, these results suggest that GAs do not act as a major signal for photoperiodic flower induction in Sinapis .  相似文献

In Arabidopsis thaliana, vernalization promotes flowering by repressing the floral inhibitor FLOWERING LOCUS C (AtFLC). This repression is mediated through epigenetic modifications at the AtFLC locus, leading to gene silencing. Whether the well-known quantitative effect of vernalization is due to the degree of AtFLC repression and/or its stability after return to normal temperature conditions has not been clarified. The aim of the work was to report morphological changes whichoccur in the shoot apex during the morphogenetic switch to floweringin the model long day (LD) plant, Sinapis alba. During the floraltransition induced by 1 LD the growth rate of all componentsof the shoot apex is modified profoundly. The earliest changes,detected at 24 h after start of LD, include a decrease in plastochronduration and an increase of growth of leaf primordia. One daylater, the meristem dome starts to increase in volume, apicalinternodes have an increased height and there is a precociousoutgrowth of axillary meristems. All these changes precede initiationof flower primordia, which starts at about 60 h after the startof LD. Later changes include meristem doming, a decrease inthe plastochron ratio and a shift to a more complex phyllotaxis.All the changes, except the decreased plastochron ratio, arecharacteristics of an apex with an increased tempo of growth.The stimulation of longitudinal growth (height of apical intemodes)is more marked and occurs earlier than the reduction of radialgrowth (plastochron ratio). Noncyclic electron transport to ferricyanide and photophosphorylation as well as the methylviologen mediated aerobic and anaerobic photophosphorylation with dichlorophenolindophenol-ascorbate as the electron donor of photosystem I were measured during the development of high-light and low-light adapted leaves of Sinapis alba. Anaerobic methylviologen-catalyzed phosphorylation is more than twice as high as aerobic phosphorylation. The difference between the rates of aerobic and anaerobic phosphorylation is sensitive to dibromothymoquinone. Thus, under anaerobic conditions, methylviologen mediates a cyclic phosphorylation including plastoquinone. All photochemical activities of high-light chloroplasts are about twice as high as that of low-light chloroplasts and show a permanent decline with increasing plant age. The lower activities of low-light chloroplasts correlate with a decrease of electron transport components, such as cytochrome f. Phytochrome in Sinapis alba L. (white mustard) seedlings undergoes both decay and reversion after an exposure to red light. This is typical of other crucifers and of dicotyledons in general. In the presence of sodium azide, decay is inhibited, and reversion continues at about the same rate as in buffer alone. The reversion has been demonstrated both in cotyledon plus hypocotyl hook and in hypocotyl hook samples alone and is of the same order of magnitude in both. Contrary conclusions in the literature that there is no reversion in Sinapis are based on indirect measurements and are unjustified.  相似文献

Sinapis alba is a long-day (LD) plant that can be induced to flower by a single LD. A number of changes normally occurring in the meristem of plants subjected to the LD can be produced in short day by a single application of a cytokinin to the apical bud. However, flower buds are not produced indicating that evocation by the cytokinin is only partial. In this work, the cytokinin content of root exudate, obtained under vacuum, and of leaf exudate, obtained by the EDTA-method, has been analyzed comparatively in vegetative and induced plants, using reversed-phase HPLC coupled to the Amaranthus bioassay. The results show that, as early as 16 hours after the start of the LD, there is an increase of cytokinin activity in both the root and leaf exudates of induced plants. These observations fit nicely with previous results obtained on Sinapis, and they indicate that cytokinins are part of the floral stimulus in this species.  相似文献

An electron microscopic investigation of fine structural changes in post-meristematic cotyledon mesophyll cells during the period of storage protein accumulation (16–32 d after pollination) showed that the rough ER, the Golgi apparatus and the developing vacuome are intimately involved in the formation of storage protein bodies (aleurone bodies). At the onset of storage protein accumulation (16–18 d after pollination) storage protein-like material appears within Golgi vesicles and preformed vacuoles. At a later stage (24 d after pollination) similar material can also be detected within vesicles formed directly by the rough endoplasmic reticulum (ER). It is concluded that there are two routes for storage protein transport from its site of synthesis at the ER to its site of accumulation in the vacuome. The first route involves the participation of dictyosomes while the second route bypasses the Golgi apparatus. It appears that the normal pathways of membrane flow in the development of central vacuoles in post-meristematic cells are used to deposit the storage protein within the protein bodies. Thus, the protein body can be regarded as a transient stage in the process of vacuome development of these storage cells.Abbreviation ER endoplasmic reticulum  相似文献

Shoot apical meristcms of vegetative and induced plants of Sinapis alba L. were labelled with [35S] methioninc for 2 h and the proteins were then separated by isoelectric focussing and polyacrylamide gel electrophoresis. Quantitative and possibly qualitative changes in the complement of proteins being synthesised during evocation were detected in the meristem, distal to the primordia, 50 to 52 h after the beginning of the inductive long day. This was before morphological changes in the meristem, and before the initiation of flower bud primordia.  相似文献

对不同来源的8份白芥材料,采用人工自交法分析其自交亲和性。结果显示:白芥的自交亲和性存在较大幅度的变异,自交亲和指数在0.01~4.10之间,8份参试材料中,自交亲和指数小于1的材料有3个,自交亲和指数大于1的材料有5个。表明白芥中存在自交亲和材料,白芥自交亲和性变异不仅存在于材料间,而且也存在于同一材料内不同个体间。按自交亲和指数的高低,可将参试材料分为3种类型:高自交亲和类型(自交亲和指数大于3.00,如民乐洪水芥麻、04(X)等)、自交亲和类型(自交亲和指数为1.00~2.99)、自交不亲和类型(自交亲和指数0.00~1.00)。  相似文献

Phototropic responses were studied in light-grown mustard hypocotyls.Phototropism was induced by adding 0.27 µmol m–2s–1 unilateral blue light to a background of low pressuresodium (SOX) lamp light. Curvatures of some 6° from thevertical were reached by 60 min, the curvature rate between20 min and 60 min being 0.14° min–1. From the axialgrowth rate and tissue geometry the local growth rates of illuminatedand shaded sides of the hypocotyl were calculated to be 1.5and 4.5 µmin–1 respectively. Turgor pressures ofexpanding cells in control plants and in the shaded and illuminatedsides of the blue light illuminated hypocotyls were measuredto be 0.40-0.55 MPa with a pressure probe. No changes in turgorpressure were observed on initiation of curvature. The decayof pressure in the cells of non-transpiring plants followingexcision indicated that the yield stress threshold of the tissuemay be as low as 0.1 MPa.
