以香菇菌株“沪香F2”及其自交优良F2代菌株“申香1504”为实验材料,收集孢子单核体,对其交配型进行鉴定,然后通过单孢自交的方法,构建F2和F3代群体,并对孢子单核体、F2、F3代群体各阶段培养、出菇情况以及重要农艺性状进行详细统计分析,研究各性状表型分化的情况及遗传规律。结果表明:2个亲本所获得的孢子单核体中A2B1交配型比例均为最高,根据孢子单核体交配型数量分别设计了1 028和972个F2和F3代自交配对组合。在2个群体中,配对阶段,分别有15.47%和23.56%的配对组合由于菌丝生长缓慢无法获得后代双核体菌株,且F3代显著高于F2代;生产种培养阶段,出现不良性状的菌株数量分别为7.78%和9.57%;菌棒培养阶段,出现不良性状的菌株数量分别为41.05%和49.28%,且F3代退化菌株比例显著高于F2代,不转色菌株比例显著低于F2代;出菇阶段,分别有3.11%和4.32%的菌株不现蕾,分别有13.04%和4.32%的菌株出畸形菇,分别有19.55%和8.95%的菌株能出正常菇,且F2代出正常菇的菌株比例显著高于F3代。“沪香F2”和“申香1504”分别有26个和8个孢子单核体,多次配对获得的杂交子,出正常菇的概率达50%以上。2个群体的平均单棒产量、平均单棒菇数、平均单菇重表现出明显的分化现象,且两个群体之间均存在极显著性差异。与F2代相比,F3代的产量分布、菇数分布表现出偏分离现象,平均单棒产量低于F2代43.84%,平均单棒菇数低于F2代56.77%。香菇“沪香F2”菌株在F3代中的培养、出菇情况以及农艺性状整体表现劣于F2代,且在F2代中获得表现优于亲本的高产品种,在F3代中获得大朵型品种,对香菇优良菌株选育具有重要的指导作用。  相似文献

 This study examines sporophytic and gametophytic vigor in wild and cultivated varieties of Cucurbita pepo L. and their hybrids in order to determine whether hybrid vigor extends to the microgametophyte generation. It also examines the variation in sporophytic and gametophytic vigor to discern the non-genetic influences of pollen provisioning by the sporophyte on pollen performance from the genetic influences of the microgametophyte’s own genotype on pollen performance. A cultivated and a wild C. pepo and their F1 and the F2 generations were grown under field conditions and flower and fruit production were monitored over one summer. We found that the four types of plants differed significantly in the number of male and female flowers and the number of fruits they produced. The F1 plants produced significantly more male flowers and marginally more female flowers and fruits than the parental lines. To estimate gametophytic vigor pollen was germinated in vitro and pollen tube length measured after 30 min. We found that pollen tubes from the F1 plants had significantly greater growth than tubes from the parental lines or the F2 generation, indicating that hybrid vigor extends to the microgametophytic generation. By partitioning the variance of pollen tube growth into ’within’ and ’among’ plant components of variation, we were able to show that the genotype of the microgametophyte influences pollen performance in vitro, but that expression of hybrid vigor in the microgametophyte is likely to be due to an environmental effect related to provisioning of the pollen grains during development. Meiosis in hybrids obtained in direct and return crossings between Haynatricum and wheat was studied. In F1 hybrids the possibility of stimulation of homoeologous pairing between the chromosomes of T. aestivum and T. dicoccum and, probably, H. villosum, if Haynatricum was used as pollinator, has been shown. This process is considerably intensified when the genes regulating chromosome pairing, in particular ph1b mutation, are used. In reciprocal crosses it was shown that wheat genotypes can differently influence on homoeologous chromosome pairing. In BC1 and F2 generations the chromosome compositions are determining which arise as a result of stochastic processes in premeiotic mitoses and in meiosis in F1 hybrids.  相似文献

Long-distance transplantation of seed material as done in restoration programs has raised concerns about the risks associated with the introduction of maladapted genotypes that may hybridize with neighboring native conspecifics and decrease local population fitness (outbreeding depression). We studied the consequences of gene flow from foreign provenances into local populations in the common grassland species Plantago lanceolata (Plantaginaceae). Three generations of intraspecific hybrids (F(1), F(2), and backcross to the local plants) were produced by controlled crossings between local plants and plants from geographically or environmentally distant populations. Their performance was compared to that of within-population crosses in a field experiment. Early growth in some interpopulation hybrids was significantly reduced, and this decrease in performance was higher in progeny of crosses with the local population from a different habitat than with geographically distant populations. At the end of the growing season, most fitness-related traits of the interpopulation hybrids were close to the average of their parents. Crosses with low-performing foreign parents therefore resulted in reduced fitness of the hybrids compared to the local plants and dilution of local adaptation. We conclude that the introduction of maladapted populations from distant or ecologically distinct environments might, at least temporarily, decrease the fitness of neighboring local plants.  相似文献

The genetic architecture of floral traits involved in the evolution of self-pollination provides a window into past processes of mating system divergence. In this study, we use two generations of crosses between highly selfing and predominantly outcrossing populations of Arenaria uniflora (Caryophyllaceae) to determine the minimum number, average dominance relationships, and pleiotropic effects of genetic factors involved in floral divergence. Comparison of the F1 and F2 phenotypic means with the expectations of a completely additive model of gene action revealed a primarily additive genetic basis for floral characters associated with mating system variation. The exception was flower life span, which showed partial dominance of the outcrosser phenology. In contrast to similarly divergent species, the substantial differences in flower size between these A. uniflora populations appear to involve relatively few genes of large effect (minimum number of effective factors = 2.2 +/- 2.8 SE). In addition, correlations among traits in the F2 generation indicate that pleiotropy may be an important feature of the genetic architecture of floral evolution in A. uniflora. The evolution of selfing via major modifiers of floral morphology is consistent with other evidence for ecological selection for preemptive self-pollination in A. uniflora. Analyses of the genetic basis of autonomous selfing were complicated by hybrid breakdown in both F1 and F2 generations. Only F1 hybrids showed reductions in female fertility, but about 30% of F1 and F2 hybrids exhibited partial or complete male sterility. Male sterile flowers were characterized by short stamens, reduced petals, and a lack of protandry, as well as indehiscent anthers. This morphological breakdown mimics environmental disruptions of floral development and may result from novel genic interactions in hybrids.  相似文献

Natural hybridization is increasingly recognized as an important process for the ecology and evolution of natural plant populations and species. There is a great need to initiate more studies based on natural populations in order to elucidate the possible role of hybrids in nature. The reproductive success of early generation hybrids can make or break hybrid lineages and may determine the genetic structure of hybrid swarms or the potential for gene flow through future generations, but studies of hybrid reproductive success are lacking. Here we measured components of male and female reproductive success in Senecio jacobaea and S. aquaticus (Asteraceae) species and F(1) hybrids between these species under laboratory conditions, and we measured reproductive output from crosses producing F(1), F(2), and backcross (BC) generation hybrids. F(1) hybrids were readily produced, and on average, the success of crosses producing subsequent generations (F(2), BC) decreased (though remained substantial), but the success of crosses was highly dependent on the genotypes involved. Also, F(1) hybrids were bigger, produced more flowers, and therefore produced more pollen than parental plants. Finally, crosses between parents were asymmetrical, such that S. aquaticus produced more and larger F(1) seeds than did S. jacobaea.  相似文献

Reciprocal crossing of the Mt Alford (A) strain of the cattle tick B. microplus with a susceptible (S) strain and phenotype analysis of F1, testcross and F2 progeny showed that high chlorpyrifos resistance in strain A was due to two genes that were complementary and jointly exhibited incomplete dominance. Diazinon resistance in the Gracemere (G) strain appeared to be similarly inherited. The 'average' degree of dominance ('average dominance', Dav) of high chlorpyrifos resistance over susceptibility, exhibited by F1 hybrids from A X S reciprocal crossings, was +0.54 on a -1 to +1 scale and was not significantly different from the parametric value of +0.5 (semi-dominance). The corresponding Dav values revealed by G X S crossings were +0.42 for diazinon resistance (significantly less than +0.5) and -0.031 for chlorpyrifos resistance (not significantly different from zero and therefore exhibiting zero dominance/recessivity). Resistance factors for chlorpyrifos in strains A and G for homozygotes were 74 and 35, respectively, and for F1 hybrids were 25-29 and 5-7, respectively. The resistance factors for diazinon in strain G for homozygotes and F1 hybrids were 174 and 37-41, respectively.  相似文献

Summary Combining ability studies for grain yield and its primary component traits in diallel crosses involving seven diverse wheat cultivars of bread wheat (Triticum aestivum L.) over generations F1-F5 are reported. The general and specific combining ability variances were significant in all generations for all the traits except specific combining ability variance for number of spikes per plant in the F5. The ratio of general to specific combining ability variances was significant for all the traits except grain yield in all the generations. This indicated an equal role of additive and non-additive gene effects in the inheritance of grain yield, and the predominance of the former for its component traits. The presence of significant specific combining ability variances in even the advanced generations may be the result of an additive x additive type of epistasis or evolutionary divergence among progenies in the same parental array. The relative breeding values of the parental varieties, as indicated by their general combining ability effects, did not vary much over the generations. The cheap and reliable procedure observed for making the choice of parents, selecting hybrids and predicting advanced generation (F5) bulk hybrid performance was the determination of breeding values of the parents on the relative performance of their F2 progeny bulks.  相似文献

Concerns exist that transgenic crop x weed hybrid populations will be more vigorous and competitive with crops compared with the parental weed species. Hydroponic, glasshouse, and field experiments were performed to evaluate the effects of introgression of Bacillus thuringiensis (Bt) cry1Ac and green fluorescent protein (GFP) transgenes on hybrid productivity and competitiveness in four experimental Brassica rapa x transgenic Brassica napus hybrid generations (F1, BC1F1, BC2F1 and BC2F2). The average vegetative growth and nitrogen (N) use efficiency of transgenic hybrid generations grown under high N hydroponic conditions were lower than that of the weed parent (Brassica rapa, AA, 2n = 20), but similar to the transgenic crop parent, oilseed rape (Brassica napus, AACC, 2n = 38). No generational differences were detected under low N conditions. In two noncompetitive glasshouse experiments, both transgenic and nontransgenic BC2F2 hybrids had on average less vegetative growth and seed production than B. rapa. In two high intraspecific competition field experiments with varied herbivore pressure, BC2F2 hybrids produced less vegetative dry weight than B. rapa. The competitive ability of transgenic and nontransgenic BC2F2 hybrids against a neighbouring crop species were quantified in competition experiments that assayed wheat (Triticum aestivum) yield reductions under agronomic field conditions. The hybrids were the least competitive with wheat compared with parental Brassica competitors, although differences between transgenic and nontransgenic hybrids varied with location. Hybridization, with or without transgene introgression, resulted in less productive and competitive populations.  相似文献

Fertility in first‐generation hybrids of roach, Rutilus rutilus, and silver bream, Blicca bjoerkna, was investigated. Sperm and egg production of hybrids at first sexual maturity were examined. Eggs from female hybrids were artificially fertilized with the sperm of a corresponding hybrid male; a hybrid male from the reciprocal crossbreeding; a parental species male R. rutilus; and a parental species male B. bjoerkna. The results revealed that gametogenesis was normal in female hybrids. However, in male hybrids, a low efficiency of gametogenesis was observed. The semen of male hybrids was extremely dilute, with spermatozoa concentration lower than that in parental species. Nevertheless, these F1 hybrids (males and females) from reciprocal crossbreeding were fertile. F2 and backcross generations were produced, but F2 crosses from the female hybrid and corresponding hybrid male displayed a drastically slower hatching rate. Also higher proportions of deformed embryos were hatched than in other post‐F1‐generation crosses.  相似文献

A possible effect of interpopulation hybridization is either outbreeding depression, as a consequence of breakdown of coadapted gene complexes which can increase developmental instability (DI) of the traits, or increased heterozygosity, which can reduce DI. One of the principal methods commonly used to estimate DI is the variability of fluctuating asymmetry (FA). We analysed the effect of interpopulation hybridization in Drosophila subobscura through the variability in the wing size and the FA of wing length and width for both sexes in parental, F1 and F2 generations. The results of the wing size per se in intra- and interpopulation hybrids of D. subobscura do not explicitly reveal the significance of either of the two hypotheses. However, the results of the FA of the wing traits give a different insight. The FA of wing length and width generally increases in interpopulation crosses in F1 with respect to the FA in the parental generation, which suggests the possibility that outbreeding depression occurred in the first generation after the hybridization event. We generally observed that the FA values for the wing length and width of interpopulation hybrids were higher in F1 and F2 generations, compared to intrapopulation hybrids in same generations. These results suggest that the association between coadaptive genes with the same evolutionary history are the most probable mechanism that maintains the developmental homeostasis in Drosophila subobscura populations.  相似文献

White crappie, Pomoxis annularis, and black crappie, P. nigromaculatus, were hybridized to produce Fi and F2 generations. There was considerable tissue specific esterase gene function within each parental species. Many of the esterases were homologous but possessed different electrophoretic mobilities between the two species which permitted their use as gene markers. The pattern of inheritance and expression of a number of esterase genes was determined in the Fi and F2 interspecific hybrids. Although certain key morphological phenotypes characteristic of the black crappie predominate in the Fi hybrid, the enzyme phenotypes reflect the contributions of both co-dominant parental genomes. There is a higher percentage of heterozygotes than expected in the F2 generation which may be indicative of heterosis. The genetic analysis also provided a better understanding of the polypeptide composition of these esterases.  相似文献

Heterosis is of utmost economic importance in plant breeding. However, its underlying molecular causes are still unknown. Given the numerous advantages of Arabidopsis thaliana as a model species in plant genetics and genomics, we assessed the extent of heterosis in this species using five hybrids derived from five ecotypes. Parents, F(1) and F(2), generations in both reciprocal forms were grown in a greenhouse experiment with four replications. Mid-parent heterosis (MPH) and best-parent heterosis (BPH) averaged across hybrids were surprisingly high for biomass yield (MPH: 60.3%; BPH: 32.9%) and rosette diameter (MPH: 49.4%; BPH: 34.8%), but smaller for flowering date (MPH: 27.5%; BPH: 18.5%), seed yield (MPH: 18.9%; BPH: 1.7%), and yield components. Individual hybrids varied considerably in their MPH and BPH values for all traits, one cross displaying 140.1% MPH for biomass yield. MPH was not associated with parental genetic distance determined from molecular markers. Reciprocal effects were significant only in a few cases. With a proper choice of hybrids, our results encourage the use of Arabidopsis as a model species for investigating the molecular causes of heterosis.  相似文献

Hereditary links between different behavioral and physiological traits were studied in rat strains SHR, WKY and their hybrids F1 and F2 by analysis of correlations in segregating and nonsegregating gene-rations. Genetic correlation of arterial pressure with body mass, intake of KC1 solution and the ability to avoidance learning (directed positively) and with characteristics of exploratory activity (directed negatively) was shown. These traits seemed to be possible genetic markers of arterial hypertension. Genetical correlates of the ability to elaborate active avoidance conditional reaction (arterial pressure and pain sensitivity) testify to relief of active avoidance learning in animals with inherited active-defensive type of behaviour, manifested by predisposition to pressor reactions and by suppressed emotional reactions to pain stimulation. Frequency in intertrial crossings during avoidance training displays positive correlation with characteristics of exploratory behaviour. Positive correlation between different characteristics of exploratory activity was established both in segregating and nonsegregating generations.  相似文献

Recombination of selected genotypes plays a key role in plant breeding for generating new base populations. We investigated the influence of recombination in two parent populations on the means and combining ability variances of their hybrid population by (1) quantitative genetic theory and (2) experiments with maize. The two parent populations were founded by four early flint and four early dent inbred lines, respectively. Each population was studied in three generations: Syn-0, the four inbred lines themselves; Syn*-1, the six intrapool single crosses (SC); and Syn*-2, the three intrapool double crosses (DC). Four interpool hybrid populations were created: (1) all 16 SC and (2) all 36 DC were produced from generations Syn-0 and Syn*-1, respectively, (3) 168 biparental progenies (BIP) of type flint x dent (female x male), and (4) 168 BIP of type dent x flint were produced according to NC-design I with randomly sampled plants of generation Syn*-2. The half-sib and full-sib families obtained in this manner were evaluated for grain yield, dry matter concentration and plant height. According to theoretical results, differences in the population means of these hybrid populations indicate the presence of various types of epistasis. Changes in combining ability variances from SC to DC reflect different levels of parental inbreeding (F = 1 vs F = 0), whereas changes from DC to BIP only reflect the effects of recombination and are attributable to covariances between additive and dominance effects caused by linkage disequilibrium in the Syn-0 generations. The experimental results showed a significant decline in yield from DC to BIP due to a loss of gene combinations with favourable epistatic effects. Estimates of sigma(2)(GCA) attributable to flint or dent lines decreased or remained unchanged from SC to DC, but generally increased in the BIP populations. The consequences of these trends for developing improved interpool hybrids are discussed.  相似文献

The effect of rainbow trout growth hormone complementary DNA on body shape, dress-out yield, and body composition were assessed in the F1 and F2 generations of transgenic common carp (Cyprinus carpio). All measurements were compared with those for nontransgenic full-sibling common carp in their respective families, and the fish were communally evaluated in earthen ponds. The body weight and length were highly correlated (P <0.01) in both genotypes in all the families. Head morphometrics were negatively correlated (P <0.05) to weight and length of the fish. Various head, body, and caudal traits grew disproportionately faster in transgenic fish in both generations. The altered body shape of transgenic fish resulted in improved dressing percentage in the F2 generation. The carcass composition of transgenic muscle had a lower percentage of (P <0.01) moisture and lipids and higher (P <0.01) percentage of protein in both generations. Six of the 18 amino acids analyzed in F1 transgenic common carp muscle were higher F1 (P <0.05) than the control genotype; however, amino acid ratios were minimally changed. Also, the fatty acid profiles of both genotypes were minimally altered. Higher histidine and lysine ratios in the diet are recommended for maximum growth and health of transgenic common carp in intensive culture systems on the basis of essential amino acid ratios.  相似文献

We estimated the genetic distances among 10 spring wheat genotypes based on pedigree data, morphological traits and AFLP markers, used individually and combined with morphological traits, to find the best predictors of general- and specific-combining abilities among parental genotypes. Ten wheat parents were crossed in a diallel form, disregarding reciprocal hybrids, totaling 45 combinations. The F(1) hybrids, F(2) populations and parents were evaluated in the field in 2007. The experimental plots consisted of 20 plants for F(1) hybrids and 40 plants for parental and F(2) populations. All methods (pedigree data, AFLP markers and morphological traits, used individually and combined) were found to be useful for the assessment of genetic diversity. The significant coefficient correlations ranged from low (0.45) to moderate (0.67) between the distance measures and hybrid performance. There was significant agreement between the distance measures based on AFLP markers vs morphological traits + AFLP markers (r = 0.47) and between pedigree data vs morphological traits + AFLP markers (r = 0.43). The pedigree distance was positively associated with traits 100-kernel weight and grain yield per plant in F(1) (correlations of 0.67 and 0.62, respectively) and F(2) (correlations of 0.62 and 0.59, respectively) generations. These correlation values indicate that the genetic distance, based on pedigree data, could replace diallel crosses for the selection of parents with higher combining ability and with moderate reliability.  相似文献

Crosses between populations of the copepod Tigriopus californicus typically result in outbreeding depression. In this study, replicate hybrid populations were initiated with first generation backcross hybrids between two genetically distinct populations from California: Royal Palms (RP) and San Diego (SD). Reciprocal F(1) were backcrossed to SD, resulting in expected starting frequencies of 25% RP/75% SD nuclear genes on either a pure RP cytoplasmic or a pure SD cytoplasmic background. After 1 year of hybridization (up to 15 generations), seven microsatellite loci were scored in two replicates on each cytoplasmic background. Frequencies of the rarer RP alleles increased significantly in all four replicates, regardless of cytoplasmic source, producing a mean hybridity of 0.97 (maximum = 1), instead of the expected 0.50. Explicit tests for heterozygote excess across loci and replicates showed significant deviations. Only the two physically linked markers showed linkage disequilibrium in all replicates. Subsequent fitness assays in parental populations and early generation hybrids revealed lower fitness in RP than SD, and significant F(2) breakdown. Computer simulations showed that selection must be invoked to explain the shift in allele frequencies. Together, these results suggest that hybrid inferiority in early generations gave way to hybrid superiority in later generations.  相似文献

A closed colony of Lutzomyia longipalpis was established with specimens collected in the Raposa - Serra do Sol indian reservoir, one of the main foci of visceral leishmaniasis in the State of Roraima, Brazil. Biological observations were made on four generations of a L. longipalpis colony with emphasis on productivity. Aspects studied were the number of laid and retained eggs, and the number of adults (male and female) per generation. During the four generations the percentage of engorged females that laid eggs varied from 64.2% (third generation-F3) to 90.3% (second generation-F2). The mean number of eggs laid per female varied from 23.6 (F3) to 39. 9 (first generation-F1). The maximum number of eggs laid per female varied from 84 (F3) to 124 (F1). The mean number of retained eggs per female was 12.7 (parental generation-P and F1) to 22.1 (F2). The number of females exceeded the number of males in all generations. However, significant difference for male/female ratio was found only for F3. Fecundity rates were between 42.1 (F3) and 58.3 (F2). From a total of 439 blood-fed females, 355 females laid 12,257 eggs that yield 5,354 adults (2,525 males and 2,829 females) in four generations. F2 presented maximum productivity and fecundity rates.  相似文献

The consequences of combining divergent genomes among populations of a diploid species often involve F1 hybrid vigor followed by hybrid breakdown in later recombinant generations. As many as 70% of plant species are thought to have polyploid origins; yet little is known about the genetic architecture of divergence in polyploids and how it may differ from diploid species. We investigated the genetic architecture of population divergence using controlled crosses among five populations of the autotetraploid herb, Campanulastrum americanum. Plants were reciprocally hybridized to produce F1, F2, and F1-backcross generations that were grown with parental types in a greenhouse and measured for performance. In contrast to diploid expectations, most F1 hybrids lacked heterosis and instead showed strong outbreeding depression for early life traits. Recombinant hybrid generations often showed a recovery of performance to levels approximating, or at times even exceeding, the parental values. This pattern was also evident for an index of cumulative fitness. Analyses of line means indicated nonadditive gene action, especially forms of digenic epistasis, often influenced hybrid performance. However, standard diploid genetic models were not adequate for describing the underlying genetic architecture in a number of cases. Differences between reciprocal hybrids indicated that cytoplasmic and/or cytonuclear interactions also contributed to divergence. An enhanced role of epistasis in population differentiation may be the norm in polyploids, which have more gene copies. This study, the first of its kind on a natural autotetraploid, suggests that gene duplication may cause polyploid populations to diverge in a fundamentally different way than diploids.  相似文献
