Extracts of whole nerve (chelipeds of Cancer magister) cause inhibition of impulse generation of the crayfish stretch receptor preparation, similar to that produced by gamma-aminobutyric acid (GABA). This is not found with extracts containing only sensory or sensory and motor fibers. Extracts of inhibitory fibers inhibit the stretch receptor discharge—indicating an inhibitory action equivalent to that of up to 30,000 micrograms of GABA per gm. wet weight of inhibitor fiber. This high value is taken as an indication that the inhibitory substance in crab inhibitory fibers is not identical with gamma-aminobutyric acid. Whole nerves were found to contain 1.7 to 6.7 µg. acetylcholine per gm. nerve tissue (clam ventricle and frog rectus abdominis muscle). No acetylcholine could be detected in extracts of motor and inhibitory fibers. The acetylcholine content of sensory fibers can account for the acetylcholine activity of whole nerve extract. It is concluded that the factor I of crustacean nerve is an exclusive property of the inhibitory fibers. The results support the assumption that factor I is the transmitter substance of inhibitory neurons in these animals. The absence of acetylcholine in motor fibers indicates that this substance does not function as a transmitter of motor impulses in Crustacea, and explains the previously observed failure of the substance to elicit motor responses in these animals. The function of acetylcholine in sensory fibers is not yet clarified.  相似文献

Response variability of the single neurons of the inferior colliculus of mouse (Mus musculus) to series of noise bands and of notch noises with regular 1/12 octave steps of the band/notch center frequency and width of noise band/notch 1/3 octave, was studied. Neurons with strong inhibitory influence in excitatory response area (inhibitory-dominated) show low impulse activity when noise band exceeded excitatory response area. Spectral contrasts crossing the center of excitatory response area (at CF or nearly CF) were found to be the most efficient stimuli for such neurons. Neuron responses to spectral contrasts derived both from noise band and noise notch were identical. Approaching of inhibitory and excitatory inputs is expected to sharpen the auditory neurons frequency tuning to position of spectral contrasts, similar to neuronal processing in visual system. Neuron selectivity to the direction of spectral contrasts movement was determined in neuron response differences when the noise band or notch shifted from excitatory area to inhibitory areas as compared with shift in the opposite direction. Functional role of contrast mechanism for sound localization on the base spectral cues related to external ear transfer characteristics is discussed.  相似文献

Summary By modulating sinusoidally the impulse frequency of the inhibitory input to the slowly adapting stretch receptor of Crustacea, the impulse frequency of the sensory neuron becomes sinusoidally modulated. The systems involved are linear for amplitudes of modulation up to 70% of the steady state frequency. The transfer function is given and some of its implications are discussed.  相似文献

Experiments were carried out on cultured hippocampal neurons using a patch-clamp technique in the whole-cell configuration. We studied the characteristics of regular series of action potentials (APs), which were generated with a low frequency by inhibitory and excitatory interneurons after their direct stimulation with long-lasting (500 msec) current pulses. Nearly all parameters of the evoked impulse activity (except the frequency of generation and duration of APs) in excitatory and inhibitory neurons were significantly different. According to immunocytochemical analysis, Kv1.2- and Kv4.2-type potassium channels were expressed in the membrane of excitatory neurons (granular cells), and somatostatin was present in all these cells. As to inhibitory interneurons, only a part of such cells (large units) demonstrated immunopositivity with respect to somatostatin. In inhibitory neurons, only Kv1.2-type potassium channels were expressed. Therefore, mechanisms responsible for the ability of hippocampal interneurons to generate impulse activity under conditions of direct stimulation (in our experiments, regular low-frequency series of APs) in inhibitory and excitatory neurons are rather dissimilar. Neurons that synthesize melanin-concentrating hormone (MCH) may modulate arousal and energy homeostasis. The scattered MCH neurons have been difficult to study, as they have no defining morphological characteristics. We have developed a viral approach with AAV for selective long-term reporter gene (GFP) expression in MCH neurons, allowing the study of their cellular physiology in hypothalamic slices. MCH neurons showed distinct membrane properties compared to other neurons infected with the same virus with a cytomegalovirus promoter. Transmitters of extrahypothalamic arousal systems, including norepinephrine, serotonin, and the acetylcholine agonist muscarine, evoked direct inhibitory actions. Orexigenic neuropeptide Y was inhibitory by pre- and postsynaptic mechanisms; an anorexigenic melanocortin agonist had no effect. In contrast, the hypothalamic arousal peptide hypocretin/orexin evoked a direct inward current and increased excitatory synaptic activity and spike frequency in the normally silent MCH neurons. Together, these data support the view that MCH neurons may integrate information within the arousal system in favor of energy conservation.  相似文献

Sensitivity of sensorimotor cortical neurons to microiontophoretically applied morphine and acetylcholine has been studied in the experiments on unanesthetized rabbits. The predominant reaction to morphine and acetylcholine was decrease and increase in the rate of neuronal impulse activity, respectively. There was no correlation in the responses to morphine and acetylcholine. Atropine failed to influence the morphine effect. When both drugs are simultaneously applied to neurons, morphine decreases both excitatory and inhibitory responses to acetylcholine. This effect of morphine may occur in the case when the drug is applied in doses which do not change spontaneous neuronal activity. On the contrary, excitatory effect of glutamic acid decreased only when morphine was applied in doses causing local anesthetic effect and decreasing background neuronal activity. It is suggested that morphine can exercise a modulating influence on choline receptors of cortical neurons.  相似文献

Changes in central neural processing are thought to contribute to the development of chronic osteoarthritis pain. This may be reflected as the presence of inflammatory mediators in the cerebral spinal fluid (CSF). We therefore exposed organotypically cultured slices of rat spinal cord to CSF from human subjects with osteoarthritis (OACSF) at a ratio of 1 part CSF in 9 parts culture medium for 5-6 days, and measured changes in neuronal electrophysiological properties by means of whole-cell recording. Although OACSF had no effect on the membrane properties and excitability of neurons in the substantia gelatinosa, synaptic transmission was clearly altered. The frequency of spontaneous excitatory postsynaptic currents (sEPSC) in delay-firing putative excitatory neurons was increased, as was sEPSC amplitude and frequency in tonic-firing inhibitory neurons. These changes could affect sensory processing in the dorsal horn, and may affect the transfer of nociceptive information. Although OACSF also affected inhibitory synaptic transmission (frequency of spontaneous inhibitory synaptic currents; sIPSC), this may have little bearing on sensory processing by substantia gelatinosa neurons, as sEPSC frequency is >3× greater than sIPSC frequency in this predominantly excitatory network. These results support the clinical notion that changes in nociceptive processing at the spinal level contribute to the generation of chronic osteoarthritis pain.  相似文献

In acute experiments on cats, we studied the impulse activity of 262 neurons of the parietal associative zone (PAZ, field 5). Among them, 129 cells [100 silent units and 29 units generating background activity (BA)] were identified as output neurons, while 133 cells with the BA were interneurons of the intrinsic cortical neuronal circuits. Electrical stimulation of the primary visual, auditory, or somatosensory cortices evoked no impulse responses in silent output PAZ neurons, while output neurons with the BA and interneurons (more than 65 and 80% of the cell units, respectively) generated clear responses (more frequently, phasic). Stimulation of the auditory and visual cortices exerted mostly inhibitory effects, while stimulation of the somatosensory cortex provided mostly excitatory influences. The ratios of neurons generating primary excitatory and inhibitory responses to stimulation of the visual, auditory, and somatic cortices were 0.3:1, 0.6:1, and 3.2:1, respectively. More than 95% of the field-5 neurons were influenced from the primary sensory zones via di- and/or polysynaptic pathways. Monosynaptic excitatory inputs from the visual cortex were identified for 3.8% of interneurons and 6.9% of output PAZ neurons; for the auditory cortical inputs, the respective figures were 1.7 and 3.5%. Monosynaptic connections with the somatic cortex were found only for 4% of the interneurons under study. It has been concluded that interaction of heteromodal signals coming to the PAZ via the corticopetal and associative inputs occurs on neurons of all the cortical layers.  相似文献

Summary The interactions of excitatory and inhibitory neural input controlling feeding behaviours of fliesPhormia regina Meigen, at three different levels of deprivation were assessed by studying the proboscis extension reflex, volume of sucrose imbibed and the responsiveness of the contact chemosensory neurons. The responsiveness of the sensory cells tested did not change with respect to length of deprivation. The two known inhibitory systems consist of stretch receptors associated with a) the foregut, and b) the abdomen. These two systems were demonstrated to act at different times. The effect of the foregut stretch receptors was shown to be delayed, cumulative and slowly decaying. The abdominal stretch receptors were demonstrated to act immediately and to be important in ending a drink.  相似文献

During development, alterations in sensory experience modify the structure of cortical neurons, particularly at the level of the dendritic spine. Are similar adaptations involved in plasticity of the adult cortex? Here we show that a 24 hr period of single whisker stimulation in freely moving adult mice increases, by 36%, the total synaptic density in the corresponding cortical barrel. This is due to an increase in both excitatory and inhibitory synapses found on spines. Four days after stimulation, the inhibitory inputs to the spines remain despite total synaptic density returning to pre-stimulation levels. Functional analysis of layer IV cells demonstrated altered response properties, immediately after stimulation, as well as four days later. These results indicate activity-dependent alterations in synaptic circuitry in adulthood, modifying the flow of sensory information into the cerebral cortex.  相似文献

The number of neurons in mammalian cortex varies by multiple orders of magnitude across different species. In contrast, the ratio of excitatory to inhibitory neurons (E:I ratio) varies in a much smaller range, from 3:1 to 9:1 and remains roughly constant for different sensory areas within a species. Despite this structure being important for understanding the function of neural circuits, the reason for this consistency is not yet understood. While recent models of vision based on the efficient coding hypothesis show that increasing the number of both excitatory and inhibitory cells improves stimulus representation, the two cannot increase simultaneously due to constraints on brain volume. In this work, we implement an efficient coding model of vision under a constraint on the volume (using number of neurons as a surrogate) while varying the E:I ratio. We show that the performance of the model is optimal at biologically observed E:I ratios under several metrics. We argue that this happens due to trade-offs between the computational accuracy and the representation capacity for natural stimuli. Further, we make experimentally testable predictions that 1) the optimal E:I ratio should be higher for species with a higher sparsity in the neural activity and 2) the character of inhibitory synaptic distributions and firing rates should change depending on E:I ratio. Our findings, which are supported by our new preliminary analyses of publicly available data, provide the first quantitative and testable hypothesis based on optimal coding models for the distribution of excitatory and inhibitory neural types in the mammalian sensory cortices.  相似文献

The release of vasopressin and oxytocin from the supraoptic nucleus (SON) neurons is tonically regulated by excitatory glutamatergic and inhibitory GABAergic synaptic inputs. Acetylcholine is known to excite SON neurons and to elicit vasopressin release. Cholinergic receptors are located pre- and postsynaptically in the SON, but their functional significance in the regulation of SON neurons is not fully understood. In this study, we determined the role of presynaptic cholinergic receptors in regulation of the excitatory glutamatergic inputs to the SON neurons. The magnocellular neurons in the rat hypothalamic slices were identified microscopically, and the spontaneous miniature excitatory postsynaptic currents (mEPSCs) were recorded using the whole cell voltage-clamp technique. The mEPSCs were abolished by the non-NMDA receptor antagonist 6-cyano-7-nitroquinoxaline-2,3-dione (20 microM). Acetylcholine (100 microM) significantly increased the frequency of mEPSCs of 38 SON neurons from 1.87 +/- 0.36 to 3.42 +/- 0.54 Hz but did not alter the amplitude (from 19.61 +/- 0.90 to 19.34 +/- 0.84 pA) and the decay time constant of mEPSCs. Furthermore, the nicotinic receptor antagonist mecamylamine (10 microM, n = 16), but not the muscarinic receptor antagonist atropine (100 microM, n = 12), abolished the excitatory effect of acetylcholine on the frequency of mEPSCs. These data provide new information that the excitatory effect of acetylcholine on the SON neurons is mediated, at least in part, by its effect on presynaptic glutamate release. Activation of presynaptic nicotinic, but not muscarinic, receptors located in the glutamatergic terminals increases the excitatory synaptic input to the SON neurons of the hypothalamus.  相似文献

Spatiotemporal pattern formation in neuronal networks depends on the interplay between cellular and network synchronization properties. The neuronal phase response curve (PRC) is an experimentally obtainable measure that characterizes the cellular response to small perturbations, and can serve as an indicator of cellular propensity for synchronization. Two broad classes of PRCs have been identified for neurons: Type I, in which small excitatory perturbations induce only advances in firing, and Type II, in which small excitatory perturbations can induce both advances and delays in firing. Interestingly, neuronal PRCs are usually attenuated with increased spiking frequency, and Type II PRCs typically exhibit a greater attenuation of the phase delay region than of the phase advance region. We found that this phenomenon arises from an interplay between the time constants of active ionic currents and the interspike interval. As a result, excitatory networks consisting of neurons with Type I PRCs responded very differently to frequency modulation compared to excitatory networks composed of neurons with Type II PRCs. Specifically, increased frequency induced a sharp decrease in synchrony of networks of Type II neurons, while frequency increases only minimally affected synchrony in networks of Type I neurons. These results are demonstrated in networks in which both types of neurons were modeled generically with the Morris-Lecar model, as well as in networks consisting of Hodgkin-Huxley-based model cortical pyramidal cells in which simulated effects of acetylcholine changed PRC type. These results are robust to different network structures, synaptic strengths and modes of driving neuronal activity, and they indicate that Type I and Type II excitatory networks may display two distinct modes of processing information.  相似文献

The Shannon's information theory in multiway channels (Shannon, 1961) is applied to multi-input-output relations of the stochastic automaton models for interaction of excitatory and inhibitory impulse sequences proposed in the previous papers (Tsukada et al., 1977). In these models, the output spike train depends upon several statistical characteristics (mean frequency, standard deviation, form, order-dependence or order-independence, etc.) of the excitatory and inhibitory input spike trains. By the use of the multiple-access channel in information theory, some stochastic properties of temporal pattern discrimination in neurons are analyzed and discussed with biological systems.  相似文献

The characteristics of extra- and intracellular responses of neurons in the AI region were studied in experiments with unanesthetized cats. It was established that auditory cortex neurons with similar best frequencies showed different forms of responses to tones of the corresponding frequency. About 40% of the auditory cortex neurons generated on responses to tone presentation. On — off and off responses were found in 27% of the neurons. Cortical neurons (27%) in which stimulation or inhibition of impulse discharge persisted throughout tone action were assigned to the tonic type group of cells. Approximately 6% of neurons in the AI region did not respond to a tone. During intracellular recording about 85% of the neurons responded to the turning on and/or off of a tone by generating an action potential followed by an IPSI. In 96% of the cortical neurons studied the IPSPs were a constant component of the intracellular responses to a tone. It is concluded that the inhibition of the impulse activity of the given neurons is of primarily a postsynaptic origin. Neurons showing one or another form of response differ from one another in the relative intensity and time characteristics of excitatory and inhibitory processes interacting on their postsynaptic membranes. In neurons of the phasic type inhibitory processes are dominant over excitatory, while excitatory processes are predominant in neurons of the tonic type.A. A. The pectines of scorpions are ventral bilateral appendages supporting 104–105 chemosensory sensilla called pegs. Each peg contains 10–18 sensory neurons, some of which show ultrastructural evidence of axo-axonic synapses with other sensory neurons in the same sensillum. In extracellular recordings from single-peg sensilla, individual sensory units can be distinguished by impulse waveform and firing frequency. Cross-correlation analysis of impulse activity showed that at least two of these units, types `A1' and `A2', are inhibited during the 100-ms period immediately following activity of a third unit, type `B'. This interaction between sensory units in a single sensillum also occurs in surgically isolated pectines, indicating that it does not involve efferent feedback from the central nervous system. Other sensillar neurons appear to have excitatory interactions. Thus, in scorpion pectine, chemosensory information undergoes some form of processing within individual sensilla prior to its relay to the CNS, making this an unusually accessible preparation for study of first-order chemosensory processing events. Acetylcholine is a ubiquitous cortical neuromodulator implicated in cognition. In order to understand the potential for acetylcholine to play a role in visual attention, we studied nicotinic acetylcholine receptor (nAChR) localization and function in area V1 of the macaque. We found nAChRs presynaptically at thalamic synapses onto excitatory, but not inhibitory, neurons in the primary thalamorecipient layer 4c. Furthermore, consistent with the release enhancement suggested by this localization, we discovered that nicotine increases responsiveness and lowers contrast threshold in layer 4c neurons. We also found that nAChRs are expressed by GABAergic interneurons in V1 but rarely by pyramidal neurons, and that nicotine suppresses visual responses outside layer 4c. All sensory systems incorporate gain control mechanisms, or processes which dynamically alter input/output relationships. We demonstrate that at the site of thalamic input to visual cortex, the effect of this nAChR-mediated gain is an enhancement of the detection of visual stimuli.  相似文献

A fundamental property of neuronal circuits is the ability to adapt to altered sensory inputs. It is well established that the functional synaptic changes underlying this adaptation are reflected by structural modifications in excitatory neurons. In contrast, the degree to which structural plasticity in inhibitory neurons accompanies functional changes is less clear. Here, we use two-photon imaging to monitor the fine structure of inhibitory neurons in mouse visual cortex after deprivation induced by retinal lesions. We find that a subset of inhibitory neurons carry dendritic spines, which form glutamatergic synapses. Removal of visual input correlates with a rapid and lasting reduction in the number of inhibitory cell spines. Similar to the effects seen for dendritic spines, the number of inhibitory neuron boutons dropped sharply after retinal lesions. Together, these data suggest that structural changes in inhibitory neurons may precede structural changes in excitatory circuitry, which ultimately result in functional adaptation following sensory deprivation.  相似文献

A study was made of excitatory and inhibitory responses elicited by acetylcholine (ACh) in neurons of the snail Eobania vermiculata. At resting potential, ACh evoked a depolarizing inward current in some neurons (D-cells) and a hyperpolarizing current in others (H-cells). The currents elicited by ACh were nonlinearly dependent on membrane potential. After either D- or H-cells were equilibrated in chloride-free isotonic calcium, ACh evoked a depolarizing inward current which reversed sign at about -55 mV. These results suggest that ACh causes an influx of Ca2+ in both types of neurons.  相似文献

i) In the awake animal, neither a late response nor a silent period could be evoked from the tibial nerve. Somatic afferentation with impulse trains failed to inhibit efferent sympathetic activity. On the other hand, vagal afferentation had an inhibitory action also in the awake animal. In the awake animal, the excitatory processes are dominant. ii) Urethan anaesthesia did not influence the sympathetic nervous processes; the reflex response were practically the same as in the awake animal. iii) Chloralose anaesthesia altered the sympathetic reflex observable in the awake animal. Somatic afferentation of low threshold voltage already elicited a late response and a silent period; in addition, a high degree of summation ability of silent periods was apparent. Thus, chloralose anaesthesia seems to raise the excitatory level of the sympathetic centres in the direction of inhibition. iv) Combined chloralose+urethan anaesthesia, under which investigations are usually performed, was seem to affect the reactivity of the sympathetic centres in the same way as did chloralose anaesthesia.  相似文献
