Lipids,Proteins, and Structure of Seed Oil Bodies from Diverse Species   总被引:31,自引:3,他引:28       下载免费PDF全文
Oil bodies isolated from the mature seeds of rape (Brassica napus L.), mustard (Brassica juncea L.), cotton (Gossypium hirsutum L.), flax (Linus usitatis simum), maize (Zea mays L.), peanut (Arachis hypogaea L.), and sesame (Sesamum indicum L.) had average diameters that were different but within a narrow range (0.6-2.0 [mu]m), as measured from electron micrographs of serial sections. Their contents of triacylglycerols (TAG), phospholipids, and proteins (oleosins) were correlated with their sizes. The correlation fits a formula that describes a spherical particle surrounded by a shell of a monolayer of phospholipids embedded with oleosins. Oil bodies from the various species contained substantial amounts of the uncommon negatively charged phosphatidylserine and phosphatidylinositol, as well as small amounts of free fatty acids. These acidic lipids are assumed to interact with the basic amino acid residues of the oleosins on the surface of the phospholipid layer. Isoelectrofocusing revealed that the oil bodies from the various species had an isoelectric point of 5.7 to 6.6 and thus possessed a negatively charged surface at neutral pH. We conclude that seed oil bodies from diverse species are very similar in structure. In rapeseed during maturation, TAG and oleosins accumulated concomitantly. TAG-synthesizing acyltransferase activities appeared at an earlier stage and peaked during the active period of TAG accumulation. The concomitant accumulation of TAG and oleosins is similar to that reported earlier for maize and soybean, and the finding has an implication for the mode of oil body synthesis during seed maturation.  相似文献

The opportunistic Candida species existing as part of commensal microbiota in humans are usually the etiological agents causing infections. We investigated whether isolates collected from different age groups, hospital units, and sources have distinct characteristics. A total of 913 isolates comprising 395 Candida albicans, 230 Candida tropicalis, 202 Candida glabrata, 62 Candida parapsilosis, 13 Candida krusei, and 11 of other six species were analyzed. Urine was the most common source (41.2%), followed by sputum (16.3%), blood (15.2%), and others (27.3%). Candida albicans and C. parapsilosis were more prevalent in the working group [from 19 to 65 years], whereas C. tropicalis and C. glabrata were more prevalent in the elder one (≥ 66 years). We found that the age of patients and the source of isolates affect the distribution of species. On the other hand, the drug susceptibility of isolates was associated with fungal species and whether patients were hospitalized.  相似文献

为了完善有关裂叶苔科植物孢子、弹丝和油体的研究,为该科植物的分类提供详细的资料,利用光学显微镜和扫描电镜观察裂叶苔科(Lophoziaceae)11种植物的孢子、弹丝和油体的形态特征,其中有6种具备了孢子和弹丝的形态特征,9种具备了油体的形态特征。结果表明:(1)裂叶苔科的6种植物的孢子为球形、近球形和椭圆形;颜色为褐色、红褐色和黄褐色;直径在10~16μm,属于小型孢子(MI);近极面向内凹,纹饰为疣状、棍棒状、刺状、颗粒状和短棒状。(2)弹丝均为紧密双螺旋状,两端圆钝,表面平滑或具稀的颗粒2种。(3)裂叶苔科的9种植物的油体形状为球形、椭圆形、球形或卵形,直径在3~10μm。本研究对这11种植物的孢子、弹丝和油体进行了详细的描述并附图示,为裂叶苔科植物的系统分类、演化研究提供基础资料。  相似文献

Abstract: With a combination of enzymatic treatment and micromanipulation, the contents of oil bodies of Radula complanata were obtained. A GC‐MS analysis revealed that 3‐methoxybibenzyl is the main constituent of these compartments. Its structure was elucidated by NMR spectroscopy.  相似文献

Studies on the Mediterranean Undercurrent in the Gulf of Cádiz showed that bacterial abundance and biomass as well as heterotrophic activity were higher in the water of Mediterranean origin in 500–800 m depth than in the adjacent Atlantic water. Upwelling processes off Mauretania and Portugal were accompanied by high bacterial numbers (bacterial plate counts) in the mixed surface layer. Changes in the qualitative composition of the bacterial flora in the waters off West Africa and in the Arabian Gulf were explained by the introduction of dust from desert regions into the sea by aeolian transport. In the Western Baltic migration of fish was detected by the presence of special bacteria, which normally live on or in these animals. Regions with complex hydrographic structures such as the Western and Central Baltic Sea revealed interesting relationships between bacteriological abundance and activity on the one hand and characteristic physical and chemical properties, such as origin, salinity and O2/H2S-content, on the other. The importance of bacteriological variables for the characterization of different water bodies is discussed.  相似文献

ABSTRACT. We have used ultrastructural techniques in different malarial species to demonstrate a lysosomal system. First, we have tried to localize acid phosphatase, a typical lysosomal label. Its activity was localized in the endoplasmic reticulum and in endocytic vesicles, and in dense-cored vesicles near the digestive vacuoles, especially in Plasmodium falciparum (FCR3 strain). Then, we have studied the different cellular compartments of the malarial parasite by the zinc iodide-osmium tetroxide technique that heavily contrasted the cellular compartments of the parasite. This experiment led to the observation of a profound rearrangement of the endoplasmic reticulum, especially in P. berghei. A very atypical but functional Golgi apparatus was demonstrated in all the growing stages of the parasite and lysosome-like vesicles were observed, showing a structure very similar to those of the coated vesicles of a true Golgi complex. The presence of these organelles are in favor of the existence of a lysosomal system and of the endogenicity of some enzymes involved hemoglobin degradation.  相似文献

Specific acyltransferase activities were detected in microsomesand oil bodies from palm mesocarp in the presence of exogenouslysophosphatidic acid, diacylglycerol and glycerol-3-phosphate.These activities were 2–3 times higher per mg proteinin oil bodies than in microsomes. Diacylglycerol acyltransferasein both preparations required Mg2+, dithiothreitol, gelatineand fat free bovine serum albumin for maximum activity at pH8.5. The enzyme had an apparent Km of 0.18 mM for dipalmitin.In both preparations, the enzyme was active with myristoyl-CoA,palmitoyl-CoA, stearoyl-CoA and oleoyl-CoA. When presented witha mixture of two acyl- CoAs, microsomal enzyme showed no selectivitybut oil body enzyme had a preference for oleoyl-CoA over palmitoyl-CoA. From ontogeny of tapetum in Anemarrhena asphodeloides, the ultrastructnral features of tapetal cells are as follows: 1. The profuse rough endoplasmic reticula are often closely associated with lipid bodies and vesicles, and linking each other into compound organelles. This is one of the striking features in Anemarrhena tapetal cell. 2. After meiosis of the micro- spore mother cell, the tapetal cytoplasm contains a large number of vesicles, in which the electron opaque substances are accumulated. Then they fuse to form a large zone of storage material similar to lipid bodies. Before accumulation of opaque material, these vesicles in the tapetal cytoplasm are larger than those in elaioplast (see Plate II, Fig. 2). 3. During stage of pollen maturation the tapetal cytoplasm becomes disorganized and the cells are almost occupied by the elaioplasts at various degree of development. On the basis of the report of Dickinson (1973), the formation of a pollen coatings of Lilium is different from that of Raphanus. The osmiophilic bodies in the former have originated from membrane lamellae or membranous system of plastid, and those in the latter are formed from the plastid vescles. It is intereting to note that the mode of origin of the plastid osmiophilic bodies in Anemarrhena is rather similar to that of Raphanus than to Lilium. About the origin of the pro-Ubisch bodies in tapetal cytoplasm of Anemarrhena studies revealed that a large number of the medium electron dense bodies appear in the tapetal cytoplasm. This is the first sign of the formation of the pro-Ubisch bodies and its character is very similar to spherosome in many respects. From many ultrasections, it can be seen that the ER profile is closely associated with the pro-Ubisch bodies. Thus we can conclude that the proubisch bodies of Anemarrhena are derived from rough endoplasmic reticulum. Although Heslop-Harrison et al. (1969) has considered that the compound Ubisch bodies do not occur in Lilium, there are prominent aggregation of Ubisch bodies in Anemarrhena, same as reported in Oxalis (Cariel, 1967), Silene (Heslop-Harrison, 1963a) and Helleborus (Echlin et al., 1968). After investigation on certain angiosperm in 1972, Gupta and Nanda have reported that the peritapetal membrane belonging to tapetum of secretory type lies against the inner tang- ential wall; in the plasmodial type of tapetum, it is formed on the outer tangential wall. But in some species of Poaceae and Solanaceae, the peritapetal membrane is formed on both sides of the tapetal cells (Banerjee, 1967; Reznickov & Willemse, 1980). In the secretory tapetum of Anemarrhena, the peritapetal membrane, which do not comply with the conclusion of Gupta & Nanta (1972), is formed on outer tangential wall.  相似文献

Mercury pollution is a major environmental problem that arises as a result of natural processes as well as from anthropogenic sources. In response to toxic mercury compounds, microbes have developed astonishing array of resistance systems to detoxify them. To address this challenge, this study was aimed in screening bacterial isolates for their tolerance against varied concentrations of phenylmercuric acetate. Mercury transformation by bacteria being sensitive to factors such as available carbon source, etc. that affect mer-mediated transformation, screened mercury tolerant bacteria were also studied for their tolerance to different antimicrobials and carbon sources, followed by identification using biochemical as well as 16S rRNA approach. Following identification, gene encoding organomercurial lyase catalyzing protonolytic cleavage of C-Hg bond of organic mercury was amplified using gene specific primers, cloned in pGEMT(?) easy vector and sequenced. Microbe-based approach using organomercurial lyase encoded by merB gene being potentially economic, provides foundation to facilitate genetic manipulation of this environmentally important enzyme to remove high concentrations of obstinate mercury using holistic, multifaceted approach for use in bioremediation through generation of transgenics or as catalyst for use in bioreactors.  相似文献

Abstract: Three Coffea species (C. arabica cv. Icatu, C. canephora cv. Apoatã and C. dewevrei) were tested in order to identify and study the mechanisms of tolerance to low, non‐freezing temperatures. Several photosynthesis‐related parameters were monitored during a 20‐day period of gradual temperature decrease, from 25/20 °C (day/night) down to 15/10 °C, during chilling treatments (15/4 °C), and upon rewarming (25/20 °C). Differences were found among species, both during low temperature exposure and during rewarming. In general, Coffea species showed cold‐induced photoinhibition of photosynthesis, which was attributable to biochemical (in vivo ribulose‐1,5‐bisphosphate carboxylase/oxygenase activity and carbohydrate synthesis) and biophysical (antennae functioning, photosystem II efficiency and linear electron transport) inactivation, rather than to stomatal constraints. The moderately low temperature of 15/10 °C was enough to cause a negative impact on net photosynthesis (A), mostly due to low (initial) rubisco activity in all species. However, C. arabica cv. Icatu showed a higher tolerance to chilling and recovered quickly and completely upon rewarming, as assessed from the impacts on the photosynthetic machinery (e.g. Amax, Fo, Fv/Fm, Fv′/Fm′, qP, ?e, rubisco activity) and on carbohydrate metabolism. Such lesser effects are likely to be related to the strong increases and higher contents of zeaxanthin, lutein and β‐carotene that presumably increased the ability to dissipate excitation energy and contributed to protect the photosynthetic apparatus. During cold exposure, a significant reduction of the α/β carotene ratio, which is considered an acclimation feature, was observed solely in C. arabica cv. Icatu. However, C. canephora cv. Apoatã and, especially, C. dewevrei showed to be highly cold‐sensitive. In these latter species, the photoinhibitory impairments to photosynthesis were stronger, probably due to the lower contents of protecting pigments during chilling conditions that lead to a higher vulnerability to excess excitation energy. Moreover, the mesophyll impairments (e.g. Amax, Fv/Fm, ?e) became significant even at moderately low temperatures of 15/10 °C, and a lower ability to recover after chilling exposure was observed. The limitation of in vivo rubisco activity and Amax may have been due to substrate limitation, but disturbances in sugar metabolism could also play an important role in the expression of chilling sensitivity in C. canephora cv. Apoatã and C. dewevrei.  相似文献

Using Nycodenz, a novel density gradient medium, we isolated intact protein bodies from developing seeds of Lupinus angustifolius L. (cultivar Unicrop) and achieved excellent separation from the endoplasmic reticulum, mitochondria, and other organelles. The distribution of the storage protein conglutin-β was taken as evidence that up to 96% of the protein bodies remained intact on the gradients and banded at 1.25 grams per milliliter. The protein bodies also contained the three other abundant proteins present in L. angustifolius seeds: conglutins-α, -γ, and -δ. Pulse labeling experiments were carried out to determine the site of proteolytic processing of conglutin-α, a legumin-like 11Svedberg unit storage protein. Cotyledons aged either 33 or 40 days after flowering were pulsed with [3H]leucine. Protein bodies obtained from the cotyledons aged 33 days after flowering contained only the labeled precursors of conglutin-α with molecular weights 85,000, 72,000, and 64,000, even after a 4 hour chase of the radioactivity. Protein bodies obtained from the cotyledons aged 40 days after flowering contained the same radioactive precursors if the tissue had been pulsed for 2 hours, and the processing products of these precursors when the tissue had been chased for 4 hours. These studies confirm that the subcellular location of proteolytic cleavage of this legumin-like protein is the protein body, that this activity is detected only in protein bodies from lupin seeds aged between 33 and 40 days of seed development after flowering and that protein bodies from seeds younger than this contain only unprocessed conglutin-α.  相似文献

为了解不同产地薰衣草(Lavandula angustifolia Mill.×Lavandula latifolia Medik.)鲜花精油的化学成分,采用水蒸气蒸馏法提取了产自濮院、厦门、昆明的薰衣草花精油,并用GC-MS方法对其化学成分及相对含量进行了分析。结果表明,三地薰衣草花精油的得油率分别为6.60%、3.07%、3.17%。精油中分别检出50、59、44个化合物,其中4个化合物(桉叶油醇、α-红没药醇、樟脑和芳樟醇)的相对含量占精油总量的75%以上。三地薰衣草花精油中的主要化学成分相同,但含量不同,且精油的组成成分也存在差异。此外,薰衣草精油中的桉叶油醇、樟脑及α-红没药醇的相对含量与海拔及年均降雨量具有一定相关性。  相似文献

The anion conductance of the plasma membrane of Coffea arabica protoplasts was isolated and characterized using the whole-cell patch clamp technique. Voltage pulse protocols revealed two components: a voltage-gated conductance (G s ) and a voltage-independent one (G l ). G s is activated upon depolarization (e-fold activation every +36 mV) with time constants of 1 sec and 5 sec at all potentials. G l and G s also differ by their kinetic and biophysical properties. In bi-ionic conditions the current associated with G s shows strong outward rectification and its permeability sequence is F > NO3 > Cl. In the same conditions the current associated with G l does not rectify and its permeability sequence is F≫ NO3 = Cl. Furthermore, at potentials over +50 mV G s , but not G l , increases with a time constant of several minutes. Finally the gating of G s is affected by stretch of the membrane, which leads to an increased activation and a reduced voltage sensitivity. Anion conductances similar to the ones described here have been found in many plant preparations but G l -type components have been generally interpreted as the background activation of the slow voltage-gated channels (corresponding to G s ). We show that in coffee protoplasts G l and G s are kinetically and biophysically distinct, suggesting that they correspond to two different molecular entities. An oil body preservesa matrix of triacylglycerols surrounded by a monolayer of phospholipidsembedded with abundant structural proteins termed oleosins andprobably some uninvestigated minor proteins of higher molecularmass. Three polypeptides of 27, 37, and 39 kDa (temporarilydenominated as Sopl, Sop2, and Sop3) were regularly co-purifiedwith seed oil bodies of sesame. Comparison of amino acid compositionindicated that they were substantially less hydrophobic thanthe known oleosins, and thus should not be aggregated multimersof oleosins. The results of immuno-recognition to sesame proteinsextracted from subcellular fractions of mature seeds, varioustissues, and oil bodies purified from different stages of seedformation revealed that these three polypeptides were uniqueproteins gathered in oil bodies, accompanying oleosins and triacylglycerols,during the active assembly of the organelles in maturing seeds.Both in vivo and in intro, immunofluorescence labeling usingsecondary antibodies conjugated with FITC (fluorescein isothiocyanate)confirmed the localization of these three polypeptides in oilbodies. 1To whom correspondence should be addressed  相似文献
