Cycloheximide given to insulin-treated alloxan diabetic rats results in the inhibition of insulin-induced liver glycogen synthase bintoa conversion without affecting the level of synthase b. The effect of cycloheximide, believed to elevate cAMP in liver of normal rats, is independent of cAMP levels of the insulin-treated diabetic rat. The inhibition of insulin-mediated synthase b to a conversion by cycloheximide does not appear to be the result of a cycloheximide-induced cAMP-dependent phosphorylation of synthase a to b and suggests that insulin control of synthase b and a interconversions is dependent upon cycloheximide-sensitive protein synthesis.  相似文献

Secretion of lysosomal enzymes by human monocytes in response to various stimuli and the effect of conditioned media from lymphocytes and neutrophils was studied. Monocytes were found to release β-glucosaminidase in response to NH4Cl and to particles (zymosan, opsonised zymosan, asbestos and latex), but do not respond to some soluble stimuli like formyl-methionyl-leucyl-phenylalanine, phorbol myristate acetate, cytochalasin B, concanavalin A and N-acetylmuramyl-l-alanyl-d-isoglutamine. Neutrophil conditioned medium or neutrophil components did not have any effect on secretion. When treated with lymphokines the cells are more responsive, especially to zymosan. Even through there are similarities in the secretory activities of mouse macrophages and human monocytes, there are several differences both in the quantity of the response and in the mechanisms involved.  相似文献

Barley embryo 5S rRNA hybridizes efficiently with barley embryo 18S rRNA but not with 26S rRNA. Mouse sarcoma 5S rRNA also selectively hybridizes, to a smaller extent, with mouse sarcoma 18S rRNA. The barley embryo 5S–18S rRNA complex has a sharp melting profile and a “Tm” of ca. 59° in 0.1 M NaCl. The mouse sarcoma 5S–18S rRNA complex has a broader transition breadth and a “Tm” of ca 52°. The conditions used for hybridization lead to very specific reconstitution of the “natural” complex between 5.8S and 26S–28S rRNA since both the invivoandinvitro complexes between 5.8S and 26S–28S rRNA from barley embryos and mouse sarcoma have equally sharp melting profiles and a “Tm” of ca. 52° in 0.1 M NaCl.  相似文献

The electron spin relaxation of iron-sulphur centres and ubisemiquinones of plant mitochondria was studied by microwave power saturation of the respective EPR signals. In the microwave power saturation technique, the experimental saturation data were fitted by a least-squares procedure to a saturation function which is characterized by the power for half-saturation (P12) and the inhomogeneity parameter (b). Since the theoretical saturation curves were based on a one-electron spin system, it became possible to differentiate between EPR signals of iron-sulphur centres which have similar g values but different P12 values. If the difference in the P12 values of the overlapped components was small, no significant deviation from these theoretical saturation curves was observed, as shown for the overlapped signals of centre S-3 and the Ruzicka centre of mung bean mitochondria. By contrast, the microwave power saturation data for the g = 1.93 signal (17–26 K) of Arum maculatum submitochondrial particles reduced by succinate could not be fitted using one-electron saturation curves. Reduction by NADH resulted in a stronger deviation. Since the iron-sulphur centres of Complex I were present only in an unusually low concentration in A. maculatum mitochondria, it was proposed that an iron-sulphur centre of the external NADH dehydrogenase contributes to the spectrum of centre S-1. For mung bean mitochondria, the g = 1.93 signal below 20 K could be attributed mainly to centre N-2. The microwave power saturation technique was also suitable for detecting magnetic interactions between paramagnetic centres. From the saturation data of the complex spectrum attributable to centre S-3 and an interacting ubisemiquinone pair in mung bean mitochondria (oxidized state) followed that centre S-3 has a faster electron spin relaxation than the ubisemiquinone molecules. It is noteworthy that the differences in the relaxation rates were maintained despite the interaction between centre S-3 and the ubisemiquinones. Furthermore, a relaxation enhancement was observed for centre S-1 of A. maculatum submitochondrial particles upon reduction of centre S-2 by dithionite. This indicated a magnetic interaction between centres S-1 and S-2.  相似文献

The 2′(3′)-O-l-phenylalanyl-N2,5′-anhydroformycin (1c) and 2′(3′)-O-l-phenylalanyl-N4,5′-anhydroformycin (2c), obtained by chemical synthesis, are substrates for ribosomal peptidyltransferase from Escherichia coli. Nucleoside 1c, which mimics an anti conformation of antibiotic formycin, has 80% of the acceptor activity of puromycin at 5 · 10?4 M determined by the release of N-Ac-Phe residue from the 70 S ribosome-poly(U)-N-Ac-[14C]Phe-tRNA complex. The reaction product, 2′(3′)-O-(N-acetyl)-l-phenylalanyl-l-phenylalanyl-N2,5′-anhydroformycin (1d), was characterized by paper electrophoresis before and after alkaline hydrolysis. By contrast, nucleoside 2c, which resembles a syn conformation of formycin, exhibited only 20% of the acceptor activity of puromycin at 5 · 10∮4 M and essentially none in the concentration region between 1 · 10?6 and 1 · 10?4 M. The results which are in accord with previous models have shown that a substrate with its base in an anti conformation is preferable for the acceptor site of peptidyltransferase than the corresponding syn counterpart, Nevertheless, it is possible that an intermediate conformation, for example, high anti (amphi-minus), is an optimal arrangement for acceptor site substrates.  相似文献

An ATP-dependent mechanism for Ca2+ uptake in human platelet membrane fractions has been identified and characterized. Ca2+ uptake into a membrane fraction is shown to be stimulated at low concentrations of ATP and Ca2+ and to require magnesium ions. Initial rate kinetics, using Eadie-Scatchard analysis, indicated a single class of calcium uptake sites in the presence of ATP, with a Kd for free [Ca2+] of 0.145 μM. Ca2+ uptake in the presence of several ATP concentrations demonstrates that ATP binds to at least two sites, representing high and low affinities of 3.21 and 80.1 μM, respectively. The neuroleptic drug fluphenazine inhibited ATP-stimulated calcium uptake (IC50 = 55 μM), suggesting this ATP-dependent Ca2+ uptake system may provide a useful ion-transport model with which to study neuroleptic therapy in humans.  相似文献

An epithelial cell line, designated CHK-ACE, was established from the kidney of a spontaneously diabetic Chinese hamster from the highly inbred AC line. CHK-ACE was separated into two sublines, CHK-ACE-100 and CHK-ACE-400, by successive passages in 100 and 400 mg/dl glucose respectively. Extra- and intracellular activities of N-acetyl-β-D-glucosaminidase and β-D-galactosidase were measured in these cultures after exposure to varying concentrations of glucose (100, 200, 300 and 400 mg/dl) for one passage and 10% heated fetal calf serum for 6.5 h before enzyme measurements were taken; no apparent dependence on medium-glucose concentration was found. In serum-free medium, the time-dependent release of both N-acetyl-β-D-glucosaminidase and β-D-galactosidase was sustained for up to 24 h; no significant difference in their activities was found between CHK-ACE-100 cultures grown in 100 and 400 mg/dl glucose for one passage.  相似文献

Insulin, glucagon, and somatostatin release in vitro from perfused pancreases of normal and alloxan-diabetic rats were compared. Insulin and glucagon responses to arginine were decreased in the diabetic group whereas both basal and arginine-stimulated somatostatin release was increased. These results suggest that alterations in pancreatic D cell function as well as in D cell mass may contribute to the abnormal insulin and glucagon secretion found in alloxan diabetes.  相似文献

A new invivo test of insulin sensitivity is described. It utilizes closed-loop insulin delivery device (GCIIS, Biostator®) capable of infusing glucose and insulin according to preselected algorithms. In euglycemic patients, insulin was infused by GCIIS to maintain euglycemia in the face of challenges with gradually increasing doses of intravenously administered glucose. Under the described experimental conditions, the endogenous insulin release was minimized as evidenced by serum C-peptide levels of less than 2 ng/ml, and thus the peripheral disposal of glucose should have depended entirely on the exogenous insulin. The amount of the insulin infused was considered to be a measure of peripheral insulin sensitivity. The test was applied to normal and non diabetic obese individuals, and to diabetics, both insulin dependent and independent. Significant insulin resistance was demonstrated in the obese and diabetic patients. In two obese females, the test was repeated after a prolonged period of starvation, and showed marked increase in insulin sensitivity. In two poorly controlled insulin dependent diabetics, marked increase in insulin sensitivity was also observed, here following a prolonged period of euglycemia (48 hours). It is concluded that the GCIIS controlled insulin sensitivity test is a simple, reliable test of peripheral insulin sensitivity, most convenient for clinical and experimental studies invivo  相似文献

Alloxan diabetes in rats was found to decrease the level of phospholipids in the heart. Measurement of specific phosphatides showed that the decrease was restricted only to phosphatidylethanolamine and lysophosphatidylcholine. Study of invivo incorporation of 32Pi indicated an impairment of phosphatidylethanolamine synthesis and conversion of phosphatidylcholine into lysophosphatidyl choline in the heart of diabetic rats. Treatment of diabetic rats with insulin restored the levels of phosphatidylethanolamine and lysophosphatidylcholine and incorporation of 32Pi into these phosphatides to almost normal.  相似文献

The relative effectiveness of oxidizing (.OH, H2O2), ambivalent (O2?) and reducing free radicals (e? and CO2?) in causing damage to membranes and membrane-bound glyceraldehyde-3-phosphate dehydrogenase of resealed erythrocyte ghosts has been determined. The rates of damage to membranebound glyceraldehyde-3-phosphate dehydrogenase (R(enz)) were measured and the rates of damage to membranes (R(mb)) were assessed by measuring changes in permeability of the resealed ghosts to the relatively low molecular weight substrates of glyceraldehyde-3-phosphate dehydrogenase. Each radical was selectively isolated from the mixture produced during gamma-irradiation, using appropriate mixtures of scavengers such as catalase, superoxide dismutase and formate. .OH, O2? and H2 O2 were approximately equally effective in inactivating membrane-bound glyceraldehyde-3-phosphate dehydrogenase, while e? and CO2? were the least effective. R(enz) values of O2? and H2O2 were 10-times and of .OH 15-times that of e?. R(mb) values were quite similar for e? and H2O2 (about twice that of O2?), while that of .OH was 3-times that of O2?. Hence, with respect to R(mb): .OH >e? = H2O2 >O2? , and with respect to R(enz): .OH >O2? = H2O2 >e?. The difference between the effectiveness of the most damaging and the least damaging free radicals was more than 10-fold greater in damage to the enzyme than to the membranes. Comparison between H2O2 added as a chemical reagent and H2O2 formed by irradiation showed that membranes and membrane-bound glyceraldehyde-3-phosphate dehydrogenase were relatively inert to reagent H2O2 but markedly susceptible to the latter.  相似文献

Cultured mouse peritoneal macrophages containing previously endocytosed zymosan or small-fibre asbestos (but not latex or sucrose) were shown to release selectively into the medium the lysosomal hydrolase β-N-acetylglucosaminidase. Thus macrophage lysosomal enzyme secretion was experimentally dissociated from endocytosis (as the residual external particles were washed away from the cells). The cells remained viable, and total activities of both N-acetyl-β-D-glucosaminidase and of lactate dehydrogenase (a cytosol enzyme) rose with time. The relevance of such secretion by macrophages containing stored materials to chronic inflammatory processes is discussed.  相似文献

Livers from normal, adrenalectomized, and diabetic rats were perfused invitro in order to investigate the mode of action of insulin in the control of glycogenesis by glucose. Control of glycogen synthase and phosphorylase by glucose is completely lost in livers from 2 and 6 day alloxan diabetic rats. Three hour treatment of normal rats with anti-insulin serum results in a decrease in the effect of glucose on hepatic glycogenesis. Glucose infusion into isolated perfused livers from fed normal and adrenalectomized rats promotes an increase in glycogen synthase activation and phosphorylase inactivation. These data clearly demonstrate that the presence of insulin rather than glucocorticoids is an absolute requirement in the control of hepatic glycogen synthesis by glucose.  相似文献

(1) Treatment of (Na+ + K+)-ATPase from rabbit kidney outer medulla with the γ-35S labeled thio-analogue of ATP in the presence of Na+ + Mg2+ and the absence of K+ leads to thiophosphorylation of the enzyme. The Km value for [γ-S]ATP is 2.2 μM and for Na+ 4.2 mM at 22°C. Thiophosphorylation is a sigmoidal function of the Na+ concentration, yielding a Hill coefficient nH = 2.6. (2) The thio-analogue (Km = 35 μM) can also support overall (Na+ + K+)-ATPase activity, but Vmax at 37°C is only 1.3 γmol · (mg protein)? · h?1 or 0.09% of the specific activity for ATP (Km = 0.43 mM). (3) The thiophosphoenzyme intermediate, like the natural phosphoenzyme, is sensitive to hydroxylamine, indicating that it also is an acylphosphate. However, the thiophosphoenzyme, unlike the phosphoenzyme, is acid labile at temperatures as low as 0°C. The acid-denatured thiophosphoenzyme has optimal stability at pH 5–6. (4) The thiophosphorylation capacity of the enzyme is equal to its phosphorylation capacity, indicating the same number of sites. Phosphorylation by ATP excludes thiophosphorylation, suggesting that the two substrates compete for the same phosphorylation site. (5) The (apparent) rate constants of thiophosphorylation (0.4 s?1 vs. 180 s?1), spontaneous dethiophosphorylation (0.04 s?1 vs. 0.5 s?1) and K+-stimulated dethiophosphorylation (0.54 s?1 vs. 230 s?1) are much lower than those for the corresponding reactions based on ATP. (6) In contrast to the phosphoenzyme, the thiophosphoenzyme is ADP-sensitive (with an apparent rate constant in ADP-induced dethiophosphorylation of 0.35 s?1, KmADP = 48 μM at 0.1 mM ATP) and is relatively K+-insensitve. The Km for K+ in dethiophosphorylation is 0.9 mM and in dephosphorylation 0.09 mM. The thiophosphoenzyme appears to be for 75–90% in the ADP-sensitive E1-conformation.  相似文献

1. In the presence of ATP, the Ca2+ pump of human red cell membranes catalyzes the hydrolysis of p-nitrophenyl phosphate. The requirement for ATP of the Ca2+-p-nitrophenylphosphatase activity was studied in relation to the two classes of site for ATP that are apparent during Ca2+ -ATPase activity. 2. (a) The K0.5 for ATP as activator of the Ca2+ -p-nitrophenylphosphatase extrapolated at 0 mM PNPP is equal to the Km of the Ca2+ -ATPase. (b) PNPP competes with ATP and its effectiveness is the same regardless the nucleotide acts as the substrate of the Ca2+ -ATPase or as activator of the Ca2+ -p-nitrophenylphosphatase. 3. PNPP at the high-affinity site does not substitute for ATP as activator of the Ca2+ -p-nitrophenylphosphatase. 4. At ATP concentrations that almost saturate the high-affinity site, Ca2+ -p-nitrophenylphosphatase activity increases as a function of PNPP along an S-shaped curve, while Ca2+ -ATPase activity is partially inhibited along a curve of the same shape and apparent affinity. The fraction of Ca2+ -ATPase activity which is inhibited by PNPP is that which results from occupation of the low-affinity site by ATP. 5. Activation of the Ca2+ -ATPase by ATP at the low-affinity site is associated with inhibition of the Ca2+ -p-nitrophenylphosphatase activity. Both phenomena take place with the same apparent affinity and along curves of the same shape. 6. Experimental results suggest that: (a) the Ca2+ -p-nitrophenylphosphatase activity depends on ATP at the high-affinity site; (b) PNPP is hydrolyzed at the low-affinity site; (c) Ca2+ -ATPase activity at the high-affinity size persists during Ca2+ -p-nitrophenylphosphatase activity.  相似文献

The kinetic mechanism of chlorpromazine inhibition of erythrocyte hexose transport was investigated using the non-metabolizable glucose analog 3-O-methylglucose. It was found that chlorpromazine added to the external medium is a non-competitive inhibitor of both equilibrium exchange and net 3-O-methylglucose transport at pH 7.8, 15°C. The Ki for equilibrium exchange is 76 ± 21 μM. When net efflux and equilibrium exchange were measured on the same population of cells the equilibrium exchange was 2.5-times the maximum net efflux. The percent reduction of 3-O-methylglucose flux by chlorpromazine is dependent upon chlorpromazine concentration and not 3-O-methylglucose concentration as expected for a non-competitive inhibitor. Equilibrium exchange and net efflux show the same extent of inhibition at each concentration of chlorpromazine evaluated. These results suggest that exchange and net efflux of 3-O-methylglucose in the human erythrocyte may share a common transport system.  相似文献

Two valine-sensitive acetohydroxy acid synthase activities were separable from Escherichiacoli K-12 cells by virtue of their different affinities for DEAE-cellulose eluted with a KC1 gradient. These activities appeared to be independent from a valine-resistant cryptic component expressed only in ilvO regulatory mutants. The properties of the first and second activity were coincident to those of extracts of ilvB and ilvHI mutants, respectively. These data prove that the ilvB and ilvHI gene products exist in the cell as physically distinct acetohydroxy acid synthase isoenzymes.  相似文献

The effects of experimentally induced diabetes on the conversion of glucose to lipid in the isolated perfused rat lung were examined. Alloxan diabetes and streptozotocin diabetes reduced the incorporation of glucose into the neutral lipid and phospholipid fractions of the lung to a rate less than 40% of that observed in normal animals. This phenomenon appears to be related to insulin deficiency as lungs from diabetic rats treated for one week with insulin were capable of incorporating glucose at a rate comparable to that observed in normal animals. While insulin invivo altered lipid metabolism in perfused lung, invitro insulin had no demonstrable effect on lipid metabolism in the perfused lung, an indication that the effects of the hormone may be long term rather than short term. These data indicate that pulmonary lipid metabolism may be regulated by the action of insulin.  相似文献
