The neuropeptide FF2 (NPFF2) receptor belongs to the rhodopsin family of G protein-coupled receptors and mediates the effects of several related RFamide neuropeptides. One of the main pharmacological interests of this system resides in its ability to regulate endogenous opioid systems, making it a potential target to reduce the negative effects of chronic opioid use. Phosphorylation of intracellular residues is the most extensively studied post-translational modification regulating G protein-coupled receptor activity. However, until now, no information concerning NPFF2 receptor phosphorylation is available. In this study, we combined mass spectrometric analysis and site-directed mutagenesis to analyze for the first time the phosphorylation pattern of the NPFF2 receptor and the role of the various phosphorylation sites in receptor signaling, desensitization, and trafficking in a SH-SY5Y model cell line. We identified the major, likely GRK-dependent, phosphorylation cluster responsible for acute desensitization, 412TNST415 at the end of the C terminus of the receptor, and additional sites involved in desensitization (372TS373) and internalization (Ser395). We thus demonstrate the key role played by phosphorylation in the regulation of NPFF2 receptor activity and trafficking. Our data also provide additional evidence supporting the concept that desensitization and internalization are partially independent processes relying on distinct phosphorylation patterns.  相似文献

Extracellular-signal-regulated kinase 5 (ERK5) is a member of the mitogen-activated protein kinase (MAPK) family and, similar to ERK1/2, has the Thr-Glu-Tyr (TEY) activation motif. Both ERK5 and ERK1/2 are activated by growth factors and have an important role in the regulation of cell proliferation and cell differentiation. Moreover, both the ERK5 and the ERK1/2 pathways are sensitive to PD98059 and U0126, which are two well-known inhibitors of the ERK pathway. Despite these similarities, recent studies have revealed distinctive features of the ERK5 pathway: ERK5 has a key role in cardiovascular development and neural differentiation; ERK5 nuclear translocation is controlled by its own nuclear localizing and nuclear export activities; and the carboxy-terminal half of ERK5, which follows its kinase catalytic domain, has a unique function.  相似文献

Drosophila melanogaster Held Out Wings (HOW) is a conserved RNA-binding protein (RBP) belonging to the STAR family, whose closest mammalian ortholog Quaking (QKI) has been implicated in embryonic development and nervous system myelination. The HOW RBP modulates a variety of developmental processes by controlling mRNA levels and the splicing profile of multiple key regulatory genes; however, mechanisms regulating its activity in tissues have yet to be elucidated. Here, we link receptor tyrosine kinase (RTK) signaling to the regulation of QKI subfamily of STAR proteins, by showing that HOW undergoes phosphorylation by MAPK/ERK. Importantly, we show that this modification facilitates HOW dimerization and potentiates its ability to bind RNA and regulate its levels. Employing an antibody that specifically recognizes phosphorylated HOW, we show that HOW is phosphorylated in embryonic muscles and heart cardioblasts in vivo, thus documenting for the first time Serine/Threonine (Ser/Thr) phosphorylation of a STAR protein in the context of an intact organism. We also identify the sallimus/D-titin (sls) gene as a novel muscle target of HOW-mediated negative regulation and further show that this regulation is phosphorylation-dependent, underscoring the physiological relevance of this modification. Importantly, we demonstrate that HOW Thr phosphorylation is reduced following muscle-specific knock down of Drosophila MAPK rolled and that, correspondingly, Sls is elevated in these muscles, similarly to the HOW RNAi effect. Taken together, our results provide a coherent mechanism of differential HOW activation; MAPK/ERK-dependent phosphorylation of HOW promotes the formation of HOW dimers and thus enhances its activity in controlling mRNA levels of key muscle-specific genes. Hence, our findings bridge between MAPK/ERK signaling and RNA regulation in developing muscles.  相似文献

FUS3转录因子是调控植物种子油脂合成的关键因子。为探讨亚麻荠CsFUS 3基因在脂质合成和积累过程中的作用,该研究对CsFUS 3基因家族进行全基因组鉴定,分析CsFUS 3基因的时空表达模式,并解析CsFUS 3-1和CsFUS 3-2基因在植物油脂合成中的功能,为深入解析CsFUS 3基因在亚麻荠油脂合成中的功能及亚麻荠高油品种选育提供理论基础。结果表明:(1)利用AtFUS3蛋白序列,在亚麻荠基因组数据库中鉴定出2条完整的CsFUS3蛋白序列,分别命名为CsFUS3-1和CsFUS3-2,亚细胞定位发现2个CsFUS3蛋白均位于细胞核。(2)亚麻荠CsFUS3-1和CsFUS3-2蛋白与拟南芥AtFUS3蛋白的亲缘关系最近,具有与拟南芥AtFUS3蛋白相似的理化性质、高级结构以及完整的B3功能域。(3)qRT-PCR结果显示,CsFUS 3-1和CsFUS 3-2基因仅在种子中表达,且随着种子的发育成熟,2个CsFUS 3基因的表达量均呈先增高后降低的变化趋势,并在花后30 d时表达量达到最高。(4)CsFUS3和CsWRI1蛋白互作以及CsFUS 3对OLE和ABI 3基因的转录调控可能是亚麻荠高油性状的关键调控途径。(5)烟草瞬时表达分析表明,与野生型相比,转CsFUS 3-1和CsFUS 3-2基因的烟草叶片总油脂含量分别提高了0.95%和1.12%,表明亚麻荠CsFUS 3基因能够提高烟叶总油脂的合成积累。  相似文献

目的:构建人E2F1基因原核表达质粒p GEX-KG-E2F1,并在大肠杆菌中诱导表达。随后验证纯化得到的E2F1蛋白可作为底物被甲基化转移酶修饰。方法:构建原核表达质粒p GEX-KG-E2F1,在大肠杆菌BL-21中经异丙基硫代半乳糖苷(IPTG)诱导表达,利用GST亲和层析法纯化表达的E2F1蛋白。随后将纯化的E2F1蛋白作为底物,组蛋白甲基化转移酶SET7/9作为酶进行体外同位素标记放射自显影实验,检测纯化的E2F1蛋白能否被甲基化。结果:酶切鉴定和测序结果证明成功构建了原核表达载体p GEX-KG-E2F1,SDS-PAGE检测结果证明实现了人E2F1基因在大肠杆菌中的可溶性表达,放射自显影证明纯化得到的E2F1蛋白可作为底物被甲基化转移酶SET7/9甲基化。结论:成功构建了转录因子E2F1体外甲基化体系,为筛选新的能甲基化E2F1的酶奠定基础。  相似文献
