Selenoprotein W (SelW) is an existing form of selenium (Se). Se influences the levels of SelW in mammals. However, little is known about the pattern of SelW expression in the gastrointestinal tract tissue of bird. The present paper describes the effects of different dietary levels of Se on the SelW mRNA expression in the gastrointestinal tract tissue of chicken. The expression levels of SelW mRNA and the Se contents in the gastrointestinal tract tissues (glandular stomach, gizzard, duodenum, small intestine, and rectum) were determined on days 15, 25, 35, 45, and 55, respectively. The results showed that the Se contents and the SelW mRNA expression were significantly higher (p < 0.05) in the high-Se group, and the Se contents and SelW mRNA expression in the low-Se group were significantly lower (p < 0.05) than in the controls. The Se contents were the highest in the duodenum and the lowest in the rectum, while the SelW mRNA expression was the highest in the gizzard and the lowest in the rectum. In addition, the SelW mRNA levels in the gastrointestinal tract tissue were found to increase in a time-dependent manner with increasing feeding time. Furthermore, the expression of the SelW mRNA in the gastrointestinal tract tissues of chickens was found to correlate with the dietary Se concentrations, but not with the tissue Se contents.  相似文献

Selenium (Se), selenoprotein N (SelN) and selenoprotein W (SelW) play a crucial role in muscle disorders. Se status highly regulates selenoprotein mRNA levels. However, few attempts have been performed on the effect of dietary Se supplementation on muscle SelN and SelW mRNA levels in birds. To investigate the effects of Se on the regulation of SelN and SelW mRNA levels in muscle tissues, one-day-old male chickens were fed either a commercial diet or a Se-supplemented diet containing 1.0, 2.0, 3.0 or 5.0 mg/kg sodium selenite for 90 days. Muscle tissues (breast, flight, thigh, shank and cardiac muscles) were collected and examined for Se content and mRNA levels of SelN and SelW. Moreover, Selenophosphate synthetase-1 (SPS-1) and selenocysteine-synthase (SecS) mRNA levels were analyzed. Significant increases in SelN mRNA levels were obtained in breast, thigh and shank muscles treated with Se, with maximal effects at 3.0 mg Se/kg diet, but 2.0 mg Se/kg diet resulted in peak levels of Sel N mRNA in flight muscles. Changes in SelW mRNA abundance in thigh and shank muscles increased in response to Se supply. After reaching a maximal level, higher Se supplementation led to a reduction in both SelN and SelW mRNAs. However, SelN and SelW mRNA levels displayed a different expression pattern in different skeletal and cardiac muscles. Thus, it suggested that skeletal and cardiac muscles SelN and SelW mRNA levels were highly regulated by Se supplementation and different muscle tissues showed differential sensitivity. Moreover, Se supplementation also regulated the levels of SPS1 and SecS mRNAs. The mRNA levels of SPS1 and SecS were enhanced in the Se supplemented groups. These data indicate that Se regulates the expression of SelN and SelW gene and affect the mRNA levels of SecS and SPS1.  相似文献

Selenoprotein W (SelW) is expressed in the immune systems of mammals. However, its pattern of expression in the immune organs of birds is still unclear. To investigate the distribution of SelW and effects of dietary Se levels on the SelW mRNA expression in the immune organs of birds, 1-day-old male chickens were fed either a commercial diet or an Se-supplemented diet containing 0.601, 1.058, 1.514, or 2.427?mg Se per kilogram, and 1.0, 2.0, 3.0 or 5.0?mg sodium selenite per kilogram for 90?days. The immune organs (spleen, thymus, and bursa of Fabricius) were collected and examined for Se content and SelW mRNA levels. The mRNA expression of SelW was detected in all the tissues. Although Se content was the highest in the spleen, the remarkable stability of the SelW mRNA level was observed in this organ during different times of dietary Se supplementation. Se-supplemented diet can make the SelW expression levels higher within a certain range in thymus and bursa of Fabricius. The present study demonstrates that SelW is widely expressed in immune organs of birds and that Se-supplementation of the feed increases SelW expression in the thymus and the bursa of Fabricius.  相似文献

Selenoprotein W (SelW) is abundantly expressed in skeletal muscles of mammals and necessary for the metabolism of skeletal muscles. However, its expression pattern in skeletal muscle system of birds is still uncovered. Herein, to investigate the distribution of SelW mRNA in chicken skeletal muscle system and its response to different selenium (Se) status, 1-day-old chickens were exposed to various concentrations of Se as sodium selenite in the feed for 35 days. In addition, myoblasts were treated with different concentrations of Se in the medium for 72 h. Then the levels of SelW mRNA in skeletal muscles (wing muscle, pectoral muscle, thigh muscle) and myoblasts were determined on days 1, 15, 25, and 35 and at 0, 24, 48, and 72 h, respectively. The results showed that SelW was detected in all these muscle components and it increased both along with the growth of organism and the differentiation process of myoblasts. The thigh muscle is more responsive to Se intake than the other two skeletal muscle tissues while the optimal Se supplementation for SelW mRNA expression in chicken myoblasts was 10−7 M. In summary, Se plays important roles in the development of chicken skeletal muscles. To effect optimal SelW gene expression, Se must be provided in the diet and the media in adequate amounts and neither at excessive nor deficient levels.  相似文献

应用ABC免疫组织化学方法,对红腹锦鸡(Chrysolophus pictus)胃肠道5-羟色胺(5-hydroxtryptamine,5-HT)、胃泌素(gastfin,GAS)、生长抑素(somatostatin,ss)3种内分泌细胞的分布密度和形态进行了观察.结果显示,5-HT细胞在空肠和直肠分布密度最高,回肠和盲肠次之,十二指肠较少,腺胃和肌胃最少;GAS细胞在十二指肠和直肠分布密度最高,其次是空肠和盲肠,腺胃部最低,肌胃则呈免疫阴性;SS细胞数量较少,在直肠、盲肠处分布密度相对高,其次是十二指肠和空肠,腺胃部最低,肌胃则呈免疫阴性.3种内分泌细胞的形态多样,有圆形、椭圆形、锥体形、杆状和不规则形,其中以圆形、椭圆形为主.细胞分布于固有膜、黏膜上皮细胞基部、黏膜上皮细胞之间、腺泡上皮细胞基部或腺泡上皮细胞之间.红腹锦鸡胃肠道内分泌细胞的形态与其内、外分泌功能是相适应的.  相似文献

The biological function of selenium (Se) is mainly elicited through Se-containing proteins. Selenoprotein W (SelW), one member of the selenoprotein family, is essential for the normal function of the skeletal muscle system. To investigate the possible relationship of Se in the process of differentiation in chicken myoblasts and the expression of SelW, the cultured chicken embryonic myoblasts were incubated with sodium selenite at different concentrations for 72?h, and then the mRNA levels of SelW and myogenic regulatory factors (MRFs) in myoblasts were determined at 12, 24, 48, and 72?h, respectively. Furthermore, the correlation between SelW mRNA expression and MRF mRNA expression was assessed. The results showed that the sodium selenite medium enhanced the mRNA expression of SelW, Myf-5, MRF4, and myogenin in chicken myoblasts. The mRNA expression levels of MRFs were significantly correlated with those of SelW at 24, 48, and 72?h. These data demonstrate that Se is involved in the differentiation of chicken embryonic myoblasts, and SelW showed correlation with MRFs.  相似文献

This study describes the effects of selenium (Se) deficiency on the messenger ribonucleic acid (mRNA) expression of 25 selenoproteins (Sels) (including glutathione peroxidases (GPx1–GPx4), thioredoxin reductases (TrxR1–TrxR3), iodothyronine deiodinases (ID1–ID3), selenophosphate synthetase 2 (SPS2), 15-kDa Sel (Sel15), SelH, SelI, SelK, SelM, Sepn1, SelO, Sepx, Selpb, SelS, SelT, SelW, Sepp1, and SelU in the adipose tissues (subcutaneous adipose, visceral adipose, and articular adipose) of chickens. One hundred and fifty 1-day-old chickens were randomly assigned to two groups of 75 each and were fed a low-Se diet (0.032 mg/kg Se) or a control diet (0.282 mg/kg Se). The expression levels of 25 Sel mRNAs were determined on days 35, 45, and 55 from three parts (subcutaneous adipose, visceral adipose, and articular adipose) of the chicken adipose tissues. The results showed that the expression levels of the 25 Sel mRNAs were significantly lower (P?<?0.05) in the low-selenium group than in the control group. In addition, the Sel mRNA expression levels in the three adipose tissues were observed to decrease in a time-dependent manner with increasing feeding time.  相似文献

Selenium (Se) is an essential trace element in many life forms due to its occurrence as selenocysteine (Sec) residue in selenoproteins. However, little is known about the expression pattern of selenoproteins in the liver of layer chicken. To investigate the effects of Se deficiency on the mRNA expressions of selenoproteins in the liver tissue of layer chickens, 1-day-old layer chickens were randomly allocated into two groups (n?=?120/group). The Se-deficient group (?Se) was fed a Se-deficient corn–soy basal diet; the Se-adequate group as control (+Se) was fed the same basal diet supplemented with Se at 0.15 mg/kg (sodium selenite). The liver tissue was collected and examined for mRNA levels of 21 selenoprotein genes at 15, 25, 35, 45, 55, and 65 days old. The data indicated that the mRNA expressions of Gpx1, Gpx2, Gpx3, Gpx4, Sepn1, Sepp1, Selo, Sepx1, Selu, Txnrd1, Txnrd2, Txnrd3, Dio1, Dio2, SPS2, Selm, SelPb, Sep15, and Sels were decreased (p?<?0.05), but not the levels of Dio3 and Seli (p?>?0.05). The results showed that the mRNA levels of 19 selenoprotein (except Seli and Dio3) genes in the layer chicken liver were regulated by diet Se level. The present study provided some compensated data about the roles of Se in the regulation of selenoproteins.  相似文献

As an essential trace element, selenium (Se) deficiency results in White Muscle Disease in livestock and Keshan disease in humans. The main objectives of this study were to clone and characterize the chicken selenoprotein W (SeW) gene and investigate SeW mRNA expression in chicken tissues. The deduced amino acid (AA) sequence of chicken SeW contains 85 AAs with UAG as the stop codon. Like all SeW genes identified in different species, chicken SeW contains one well-conserved selenocysteine (Sec) at the 13th position encoded by the UGA codon. The proposed glutathione (GSH)-binding site at the Cys37 of SeW is not conserved in the chicken, but Cys9 and Sec13, with possible GSH binding, are conserved in SeWs identified from all species. There are 23–59% and 50–61% homology in cDNA and deduced AA sequences of SeW, respectively, between the chicken and other species. The predicted secondary structure of chicken SeW mRNA indicates that the selenocysteine insertion sequence element is type II with invariant adenosines within the apical bulge. The SeW mRNA expression is high in skeletal muscle followed by brain, but extremely low in other tissues from chickens fed a commercial maize-based diet. The SeW gene is ubiquitously expressed in heart, skeletal muscle, brain, testis, spleen, kidney, lung, liver, stomach and pancreas in chickens fed a commercial diet supplemented with sodium selenite. These results indicate that dietary selenium supplementation regulates SeW gene expression in the chicken and skeletal muscle is the most responsive tissue when dietary Se content is low.  相似文献

The aim of this study was to investigate the effect of dietary supplementation with different sources of selenium and/or organic chromium on the growth performance, digestibility, lipid profile, and mineral content of hair, liver, and fore and hind limb of growing rabbits. A total of 150 weanling New Zealand White (NZW) male rabbits were randomly allotted to six dietary treatment groups: (1) basal diet (control group), (2) basal diet + 0.6 mg sodium selenite/kg diet, (3) basal diet + 0.6 mg selenium yeast/kg diet, (4) basal diet + 0.3 mg sodium selenite/kg diet + 0.3 mg selenium yeast/kg diet, (5) basal diet + 0.6 mg chromium yeast/kg diet + 0.6 mg selenium yeast/kg diet, (6) basal diet + 0.6 mg chromium yeast/kg diet. Only the combination between inorganic and organic selenium led to significant improvement in body weight, body weight gain, and feed conversion ratio. Carcass traits were not different in all groups. Selenium (Se) and chromium (Cr) were deposited in the tissues of rabbits fed diets supplemented with Se and Cr, respectively. Blood serum in both of selenium- and chromium-supplemented groups showed declined total cholesterol, triglycerides, and low-density lipoprotein (LDL). Group supplemented with organic chromium showed higher high-density lipoprotein (HDL) than the other groups. It could be concluded that using a mixture of inorganic and organic Se has a positive effect on the growth performance of growing rabbits. Both Se and Cr have hypocholesterolemic effect. Both of Se and Cr can be deposited in the meat and other tissues of rabbits and that improves meat quality which positively reflects on human acceptance. The combination between inorganic (0.3 mg sodium selenite/kg diet) and organic selenium (0.6 mg selenium yeast/kg diet) improved growth performance traits of growing rabbits.

To verify the antioxidative role of SelW in oxidant-induced chicken splenic lymphocyte, in this report, the influence of selenite supplementation and SelW gene silence on H2O2-mediated cell viability and cell apoptosis in cultured splenic lymphocyte derived from spleen of chicken were examined. The cultured cells were treated with sodium selenite and H2O2, or knocked down SelW with small interfering RNAs (siRNAs). The lymphocytes were examined for cell viability, cell apoptosis and mRNA expression levels of SelW and apoptosis-related genes (Bcl-2, Bax, Bak-1, caspase-3 and p53). The results show that the mRNA expression of SelW were effectively increased after treatment with sodium selenite, and H2O2-induced cell apoptosis was significantly decreased and cell viability was significantly increased. 20 μM H2O2 was found to induce cell apoptosis and decrease cell viability, which was alleviated obviously when cells were pretreated with sodium selenite before exposure to 20 μM H2O2. Meanwhile, H2O2 induced a significantly up-regulation of the Bax/Bcl-2 ratio, Bax, Bak-1, caspase-3 and p53 and down-regulation of Bcl-2 (P < 0.05). When lymphocytes were pretreated with Se before treated with H2O2, the Bax/Bcl-2 ratio and mRNA expression of those genes were significantly decreased, and Bcl-2 was increased (P < 0.05). SelW siRNA-transfected cells were more sensitive to the oxidative stress induced by treatment of H2O2 than control cells. Silencing of the lymphocyte SelW gene decreased their cell viability, and increased their apoptosis rate and susceptibility to H2O2. Silencing of SelW significantly up-regulated the Bax/Bcl-2 ratio, Bax, Bak-1, caspase-3 and p53 and down-regulated Bcl-2 (P < 0.05). The present study demonstrates that SelW plays an important role in protection of splenic lymphocyte of birds from oxidative stress.  相似文献

The aim of this study was to investigate the effects of xylanase supplementation on performance, characteristics of the gastrointestinal tract, blood parameters and gut microflora in broilers fed on wheat-based diets. The experimental diets consisted of a wheat-based diet supplemented with 0 or 1 g/kg enzyme preparation (xylanase activity was 1218 U/g). The diets were fed between 7 and 49 days of age. Enzyme supplementation (ES) improved (P<0.05) growth performance and feed conversion efficiency. The addition of enzyme to a wheat-based diet reduced the relative weights of the duodenum, jejunum, pancreas (P<0.05) and colon (P<0.01) in 21-day-old broiler chickens. Enzyme preparation reduced digesta viscosity in the proventriculus and jejunum of 21-day-old broiler chickens (P<0.05) and in colon of 49-day-old broiler chickens (P<0.05). The pH of the digesta in the crop, duodenum and jejunum was increased (P<0.05) in 21-day-old broiler chickens and was reduced in the caecum of 49-day-old broiler chickens (P<0.05) with enzymes. There was no significant difference between the two experimental groups in counts of lactobacillus and coliform bacteria in the caecum. Enzyme supplementation increased the concentration of blood insulin-like growth factor I (IGF-I) (P<0.01) of 21-day-old broilers, triiodothyronine (T3) and insulin (P<0.05) at 49 days. ES reduced the concentrations of blood thyroxine (T4) (P<0.01) and uric acid (P<0.05) at 49 days, but had no effect on glucose concentration (P>0.05). In conclusion, ES can improve performance and digestive parameters and can change some blood parameters in broiler chickens fed a wheat-based diet.  相似文献
